Resumo

Bovine spongiform encephalopathy (BSE) is a transmissible progressive neurodegenerative disease characterized by the accumulation of a pathological isoform (PrpSC) of the cellular prion protein (PrpC) in the brain of cattle. Two insertion/deletion polymorphisms in the PRNP gene (23bp in the promoter and 12bp in intron 1) have been associated with resistance or susceptibility to the disease. The aim of this study was to analyze the distribution of these polymorphisms in 214 healthy bovines belonging to four different breed groups (Aberdeen Angus, Aberdeen Angus x Hereford, Holstein Friesian and Uruguayan Creole cattle). DNA samples were amplified by end-point PCR. A high frequency of the alleles and haplotype associated with susceptibility to BSE (del12 and del23, and del12-del23, respectively) were found in the Aberdeen Angus, Aberdeen Angus x Hereford and Holstein Friesian animals. At the same time, the Uruguayan Creole cattle presented a higher frequency of the alleles and haplotype associated with resistance to BSE (ins12 and ins23, and ins12-ins23, respectively). These data could indicate a greater genetic resistance of the Uruguayan Creole cattle to BSE compared to other analyzed breeds, reinforcing its value as a zoogenetic resource.


A encefalopatia espongiforme bovina (EEB) é uma doença neurodegenerativa progressiva transmissível dos bovinos, caracterizada pelo acúmulo no cérebro de uma isoforma patológica (PrpSC) da proteína priônica celular (PrpC). Dois polimorfismos de inserção/deleção no gene PRNP (23bp no promotor e 12bp no íntron 1) foram associados à resistência ou suscetibilidade à doença. O objetivo deste trabalho foi analisar a distribuição desses polimorfismos em 214 bovinos sadios, pertencentes a quatro diferentes grupos raciais (Aberdeen Angus, Aberdeen Angus x Hereford, Holstein Friesian e Crioulo Uruguaio). As amostras de DNA foram amplificadas por PCR de tempo final. Uma alta frequência dos alelos e haplótipos associados à suscetibilidade à BSE (del12 e del23 e del12-del23, respectivamente) foram encontrados nos animais Aberdeen Angus, Aberdeen Angus x Hereford e Holstein Friesian, enquanto o gado Crioulo Uruguaio apresentou maior frequência dos alelos e haplótipos associados à resistência à BSE (ins12 e ins23 e ins12-ins23, respectivamente). Esses dados podem indicar uma maior resistência genética do gado Crioulo Uruguaio à BSE em comparação com as outras raças analisadas, reforçando seu valor como recurso zoogenético.

The objective of this study was to analyze the immune responses of bucks to small ruminant lentivirus (SRLV) with a focus on the reproductive system of males with recent and chronic infection. A total of 12 bucks were selected, six seronegative and six seropositive with chronic natural infection for more than 18 months (chronic infection group). After selecting the animals, the six seronegative males were intravenously inoculated with caprine arthritis-encephalitis virus (CAEV)-Co viral strain at a titer of 10-5,6 TCID50/mL. After viral inoculation, this group was called the recent infection group and was monitored weekly with the chronically infected group for 180 days with blood serum and seminal plasma Western Blot (WB) analysis. Of the animals with chronic SRLV infection, 18.94% (50/264) showed anti-SRLV antibodies in at least one of the samples, and 81.06% (214/264) were negative. Anti-SRLV antibodies were detected in 27.27% (36/132) of the blood serum samples from this group, while 10.60% (14/132) were reactive in the seminal plasma WB test. The animals inoculated with CAEV-Co became seropositive after the third week of viral inoculation. In this group, 31.06% (41/132) of seminal plasma samples had anti-SRLV antibodies, and of these, 70.73% (29/41) coincided with blood serum results. Of the remaining 29.27% (12/41), the seminal plasma sample of only three animals (RIA2, RIA3, and RIA5) had anti-SRLV antibodies. One of the animals with a recent infection presented anti-SRLV antibodies only in seminal plasma samples, possibly due to virus compartmentalization. Intermittent viral shedding was observed in both biological samples, regardless of the infection stage. The immune response in bucks with recent SRLV infection is more significant than in chronically infected animals. Regardless of the stage of infection, there is a fluctuation in antibody levels, therefore, this creates a risk of false-negative samples when performing the diagnosis.


O objetivo desse estudo foi analisar a resposta imunológica aos lentivírus de pequenos ruminantes (LVPR) com enfoque no sistema reprodutor de machos com infecção recente e crônica. Para isso, foram selecionados 12 reprodutores caprinos, sendo seis soronegativos e seis soropositivos com infecção natural crônica há mais de 18 meses (grupo com infecção crônica). Após seleção dos animais, os seis machos soronegativos foram inoculados com a cepa viral do vírus da artrite encefalite caprina (CAEV)-Co, título 10-5,6 TCID50/mL, por via intravenosa. A partir da inoculação viral este agrupamento passou a ser denominado de grupo com infecção recente e juntamente com o grupo com infecção crônica foram acompanhados, semanalmente por 180 dias, com análise dos testes de Western Blot (WB) no soro sanguíneo e plasma seminal. Nos animais com infecção crônica para LVPR, 18,94% (50/264) apresentaram anticorpos anti-LVPR em pelo menos uma das distintas amostras, e 81,06% (214/264) tiveram resultados negativos. Das amostras de soro sanguíneo do referido grupo, em 27,27% (36/132) detectou-se anticorpos anti-LVPR, enquanto que no plasma seminal 10,60% (14/132) foram reagentes no teste de WB. Nos animais inoculados com o CAEV-Co, ocorreu a soroconversão após a terceira semana da inoculação viral. Nesse grupo, 31,06% (41/132) das amostras de plasma seminal tiveram anticorpos anti-LVPR, sendo que dessas 41, 70,73% (29/41) coincidiram com resultado das amostras de soro sanguíneo. Nos 29,27% (12/41) restante, houve a detecção somente no plasma seminal e eram amostras provenientes de três animais (AIR2, AIR3 e AIR5). Em um dos animais com infecção recente, só foi identificado anticorpos anti-LVPR em amostras de plasma seminal, possivelmente em função da compartimentalização do vírus. Intermitência viral foi observada em ambas as amostras biológicas, independentemente do estágio de infecção. Conclui-se que a resposta imunológica em reprodutores com infecção recente LVPR é mais acentuada do que em animais cronicamente infectados. E, independentemente do estágio da infecção há uma flutuação nos níveis de anticorpos, sendo, portanto, um fator de risco, em virtude da existência de amostras falso-negativo ao realizar o diagnóstico.

The present objective was to investigate the presence of anti-equine viral encephalomyelitis (EVE) antibodies and the possible risk factors for its dissemination in horses raised in the East and West Potiguar mesoregions of the state of Rio Grande do Norte, Brazil. Serological diagnosis for neutralizing antibodies against Eastern (EEEV), Western (WEEV) and Venezuelan (VEEV). Equine viral encephalomyelitis was performed using a seroneutralization technique on 811 blood samples from horses from ninety properties and sixteen municipalities between July 2018 and February 2019. Factors associated with EVE were evaluated using an investigative epidemiological questionnaire, and the data were statistically analyzed using the Epi Info 3.5.2 software with a confidence level of 95%. The seroprevalence of anti-EVE antibodies was 14.2% (115), with 10.36% (84) for EEEV, 6.9% (56) for WEEV, and null for EVE. When analyzing risk factors, it can be concluded that horses raised in properties that do not clean installations and/or rent out their pasture are more likely to have anti-EVE antibodies. These results show evidence that horses raised in the East and West Potiguar mesoregions were exposed to EEEV and WEEV, thus reinforcing the importance of vaccination and serological survey of nonvaccinated horses as a means of monitoring the disease.

Background: The West Nile virus (WNV) antibodies were reported in Brazil in the serum samples taken from horses andbirds in the Midwest region and Paraíba state in 2008 and 2013, respectively. In 2014, the first human case was confirmedin a rural worker in the state of Piauí and, in 2018, the virus was isolated from the central nervous system of a horse withnervous symptoms in the state of Espírito Santo. The virus is a member of the Flaviviridae family of the genus Flavivirus(neurotropic), infecting several mammalian species, with humans and horses being the most susceptible. Approximately35% of horses develop clinical signs, thus they are considered the best sentinels for this disease. The aim of this case reportis to describe the first confirmed cases of West Nile Fever (WNF) in two horses in the state of São Paulo.Cases: Two horses with neurological symptoms were treated at the Veterinary Hospital of Cruzeiro do Sul University (SãoPaulo, SP), in 2019. Both horses came from neighboring regions that have a large Atlantic Forest preservation area and arealso routes for migratory birds, known to be competent hosts for transmitting the West Nile Fever virus, such as the swallow,the falcon and the hawk. The first one had symptoms, such as weakness and sporadic seizures; however, after recovering,it was hospitalized a few days later due to kidney failure and laminitis. The second one showed incoordination, pelviclimb weakness, and was walking in circles, evolving to seizures. Resumo

Lagoquilascariasis is a zoonotic disease caused by a nematode parasite of the genus Lagochilascaris (Family Ascarididae), with the ingestion of contaminated rodents being the main source of infection. The main clinical signs are dysphagia and the presence of ulcerated nodules in the cervical region. The objective was to describe a case of lagaroquilascariasis in a domestic cat, in the city of Caxias do Sul / RS. A young adult male feline, without defined breed presented lethargy, dehydration, bilateral fistulas in the oropharynx cavity, right eyeball with exophthalmos, corneal opacity, third eyelid prolapse and periocular ulcerated skin lesion in the upper medial region. The diagnosis was made through the visualization and microscopic identification of the parasite Lagochilascaris minor and coproparasitological examination with the presence of eggs. Treatment was based on the use of doramectin and sanitary measures. According to the current literature, there are few reports of lagoquilascariasis in felines with clinical presentation similar to this case, since the most common is the formation of an abscess in the neck region. Morover, it is the first case described in the municipality, thus contributing to the epidemiology of the disease.


A lagoquilascaríase é uma doença zoonótica causada por um parasita nematoide do gênero Lagochilascaris (família Ascarididae), sendo a ingestão de roedores contaminados a principal fonte de infecção. Os principais sinais clínicos são disfagia e presença de nódulos ulcerados em região cervical. O objetivo deste trabalho foi descrever um caso de lagoquilascaríase em um felino doméstico, na cidade de Caxias do Sul/RS. Um felino, macho, sem raça definida, adulto jovem, apresentou letargia, desidratação, fístulas bilaterais em cavidade orofaringe, globo ocular direito com exoftalmia, opacidade de córnea, prolapso de terceira pálpebra e lesão cutânea ulcerada periocular em região superior medial. O diagnóstico foi realizado por meio da visualização e identificação microscópica do parasita Lagochilascaris minor e de exame coproparasitológico com presença de ovos. O tratamento baseou-se no uso de doramectina e de medidas sanitárias. De acordo com a literatura atual, existem poucos relatos de lagoquilascaríase em felinos com apresentação clínica semelhante a este caso, visto que o mais comum consiste na formação de abscesso na região de pescoço. Além disso, é o primeiro caso descrito no município, contribuindo, assim, para a epidemiologia da doença.

Resumo

Macrocyclic lactones are widely used as endectocides in farm animals. Intoxications occur in situations of overdose and/or malnutrition, in young animals, and in genetically sensitive breeds. We describe the intoxication by doramectin in malnourished 15-month-old cattle that received 1.6 times the recommended dose. The animals presented salivation, ataxia, motor incoordination, reluctance to move, and sternal recumbency. Two animals recovered spontaneously; one died and was necropsied. No gross or microscopic changes were observed. This study suggests that doramectin may cause intoxication when administered to malnourished cattle in doses higher than those recommended and that knowing the history is essential to establish a diagnosis.


As lactonas macrocíciclas são amplamente utilizadas como endectocidas em animais de produção. Casos de intoxicação ocorrem em situações de sobredosagem e/ou desnutrição, em animais jovens ou em raças geneticamente suscetíveis. Descreve-se a intoxicação por doramectina em bovinos desnutridos, com 15 meses de idade, que receberam uma dose 1,6 vezes maior que a dose recomendada. Os animais apresentaram salivação, ataxia, incoordenação motora, relutância em se movimentar e decúbito esternal. Dois animais se recuperaram espontaneamente; um morreu e foi necropsiado. Não foram observadas alterações macro e microscópicas. Esse relato sugere que a doramectina pode causar intoxicação em bovinos desnutridos quando administrada em doses maiores que as recomendadas e que o histórico é fundamental para estabelecer o diagnóstico.

As zoonoses representam 75% das doenças emergentes. Estas doenças são uma ameaça permanente à saúde e bem-estar humanos, e têm o potencial de se tornar cada vez mais frequentes devido à degradação de habitats, alteração de utilização das terras, e aumento da mobilidade global de pessoas, animais e produtos animais. O objetivo deste estudo foi investigar o impacto de dez zoonoses (brucelose, cisticercose, equinococose, leishmaniose, leptospirose, doença de Lyme, raiva, toxoplasmose, triquinelose e febre do Nilo Ocidental) nos internamentos em hospitais do Serviço Nacional de Saúde português entre 2002 e 2016.(AU)

Listeria monocytogenes is a bacterium that infect humans and animals and causes a zoonotic disease characterized by encephalitis, septicemia or abortion. In addition, listeriosis leads to significant economic losses due to animal death and sacrifice. This research compared the technique of immunofluorescence (IF) and immunohistochemistry (IHC) for the diagnosis of L. monocytogenes in formalin-fixed and paraffin-embedded (FFPE) tissues. A total of 30 tissue blocks from 15 animals with history and/or lesions compatible with listeriosis were selected. For both IHC and IF, the same diluted (1:200) polyclonal primary antibody was used against L. monocytogenes serotypes 1 and 4. For IHC, a polymer secondary antibody conjugated to peroxidase (HRP) was used. For IF, samples were incubated with a fluorescein-labeled anti-rabbit IgG secondary antibody. Each sample was classified according to the presence and percentage of immunolabeling area. From 30 samples, 10 were positive at least for one technique, whereas eight samples were positive for both IHC and IF with similar score. There was strong immunolabeling in tissue samples from bovines experimentally infected with L. monocytogenes ATCC 7644, as well as in nervous tissues from naturally infected ruminants. Additionally, IF did not show any difference in sensitivity when compared to IHC. Using processed biological materials for IF, instead of fresh tissues, is a quite unique technique, since there are few protocols described. Therefore, this study demonstrated that both techniques are efficient to detect L. monocytogenes in FFPE tissues.


Listeria monocytogenes é uma bactéria que infecta humanos e animais, podendo causar uma doença zoonótica caracterizada por encefalite, septicemia e abortos. Além disso, a listeriose resulta em perdas econômicas significativas pela morte e sacrifício de animais. O objetivo deste trabalho foi comparar a técnica de imunofluorescência (IF) e imuno-histoquímica (IHQ) para detecção de L. monocytogenes em tecidos fixados e parafinizados. Foram selecionados 30 blocos de tecidos de 15 animais com histórico e/ou lesões compatíveis com listeriose. Para ambas as técnicas, foi utilizado o mesmo anticorpo primário policlonal diluído (1:200) para detecção de L. monocytogenes sorotipos 1 e 4. Para a IHQ foi utilizado anticorpo secundário em polímero acoplado a peroxidase (HRP). Para a IF, as amostras foram incubadas com anticorpo secundário anti-IgG de coelho marcado com fluoresceína. Cada amostra foi classificada quanto à presença de imunomarcação e porcentagem de área positiva. Das 30 amostras, 10 foram positivas em pelo menos uma das técnicas, sendo oito amostras positivas em ambas IHQ e IF com o mesmo escore. Houve forte imunomarcação tanto em amostras de bovino experimentalmente infectado com L. monocytogenes ATCC 7644, como em amostras de tecido nervoso de ruminantes naturalmente infectados. Além disso, a IF não apresentou diferença na sensibilidade quando comparada com a IHQ. O uso de materiais biológicos processados para IF, ao invés de tecidos frescos, é algo inovador, uma vez que existem poucos protocolos descritos. Portanto, este estudo demonstrou que as duas técnicas foram eficientes para detectar L. monocytogenes em tecidos fixados em formol e parafinizados.

Background: Polioencephalomalacia (PEM) is a neurological disease in ruminants, which is characterized by malacia of brain gray matter. Thiamine deficiency and sulfur intoxication are the most common causes of PEM in sheep. Affected animals present signs of cerebrocortical syndrome, including amaurosis, ataxia, head pressing, mental depression, seizures, and opisthotonus. The neurological examination aims to determine the neurolocalization of the lesions and advanced imaging techniques are useful for confirming the affected area(s) in the central nervous system. The aim of this study is to describe clinical features and ante-mortem diagnosis using magnetic resonance imaging (MRI) in a sheep with PEM. Case: A 18-month-old male Dorper sheep from a flock started receiving concentrate 7 days before. According to the owner, no clinical signs of abnormality were observed on the previous morning. However, in the afternoon, the animal became selfisolated and did not follow the flock to the sheepfold. The following day, he was found in recumbency. Physical examination revealed lateral recumbency, rectal temperature 39.5ºC, 52 bpm, 120 bpm, congested mucous membranes, capillary refill time 1 s, ruminal (4/5 min) and intestinal hypomotility. The assessment of the central nervous system revealed a decreased level of consciousness, focal seizures, opisthotonus, and absence of menace response. The following differential diagnoses were listed: PEM, head trauma, focal symmetrical encephalomalacia, bacterial encephalitis, and rabies. Treatment was composed of dexamethasone [0.2 mg/kg - i.v., SID (1st-3rd day), 0.1 mg/kg, i.v., SID (4th-6th day), and 0.05 mg/kg, i.v., SID (7th-9th day)]; mannitol [1 g/kg - i.v. and diazepam 0.4 mg/kg, i.v. single dose at admission]; vitamin B1 [10 mg/kg - i.m., SID], furosemide [1 mg/kg - i.v., SID for 3 days] and sulfadoxine/trimethoprim [30 mg/kg - i.m., SID for 10 days]. After the initial treatment, the patient showed mild clinical improvement; however, the amaurosis was still present. Magnetic resonance imaging of the brain was performed on the 2nd day of hospitalization, showing a symmetrical hypersignal in the parietal and occipital cortices, in the axial and sagittal sequences weighted in T2 and FLAIR. Discussion: This study aimed to describe the clinical signs and MRI findings in a sheep with PEM. In this case, the sudden change to the feed composition probably led to ruminal dysbiosis, inhibition of thiamine-producing microorganisms and proliferation of bacteria that synthesize thiaminase. Thiamine therapy proved to be effective and capable of reverting the clinical signs. The decrease in the level of consciousness, cortical blindness, and opisthotonus are due to alterations in the parietal cortex, in the occipital cortex, and in the cerebellum, respectively, which were demonstrated by hypersignal areas in the MRI. Therefore, the neurolocalization of the lesion based on neurologic examination and the MRI findings were related. The physicochemical and cytological evaluations of the cerebrospinal fluid, and dosage of thiamine and the concentration of hydrogen sulphide in the rumen were not performed. However, the response to thiamine treatment associated with the neurologic examination and MRI findings helped in determining the diagnosis. Additionally, MRI can be used as a useful tool for the ante mortem diagnosis of PEM.

Background: Hypothyroidism is characterized by hypofunction of the thyroid gland. It results in deficient production of thyroid hormones. Neurological disorders resulting from hypothyroidism are rare, which highlights the importance of this study. This study reports a case of hypothyroidism in a dog with neurological clinical signs, that was treated at the Universidade Estadual de Santa Cruz's Veterinary Hospital (HV-UESC). Case: A 4-year-old male intact Dogo Argentino breed dog, weighing 64 kg, presenting obesity, anorexia, prostration, walking in circles, and chronic dermatopathy was presented at HV-UESC. Upon physical examination, the animal presented a deficit of proprioception in the 4 limbs, with preserved superficial and deep pain. No alteration was observed in the ears, that could explained the clinical signs. In terms of dermatopathy, the animal presented symmetrical alopecia in the lateral region of the thighs and tail. Blood samples were collected for a complete blood count and biochemical tests of urea, creatinine, ALT, AST and cholesterol. Imaging radiography and ultrasonography were performed, which ruled out thoracic and abdominal changes that could be related to the case. Prior to receiving the blood test results, idiopathic encephalitis was suspected and enrofloxacin and prednisone were prescribed for 7 days. During the medication period, previous exams were provided, which indicated only increased cholesterol (500 mg/dL). The animal showed no improvement with the prescribed medication. In view of the clinical signs presented by the patient and the results of the additional tests, hormonal disease was suspected, compatible with hypothyroidism. Thus, hormonal tests of total T4, free T4, and TSH were requested, leading to verification of reduced total T4 (0.3 ng/dL) and free T4 (0.15 ng/dL) levels, and confirming the dysfunction of the thyroid gland. The previous treatment was suspended and thyroid hormone replacement was initiated. After 3 days of treatment, the neurological signs regressed and the animal became more active; after 30 days, the areas of alopecia decreased. Although the patient did not receive the recommended clinical follow-up for such cases, it was possible to establish the ideal levothyroxine dosage for the dog after appropriate adjustments, which permitted thyroid hormone levels to return to normal. Discussion: This report refers to a case of hypothyroidism in a giant dog breed. The dog in the report showed clinical signs of a dermatological, metabolic, and neurological nature, which is consistent with a lack of thyroid hormone. The main signs presented by the animal were neurological, such as walking in circles and a deficit of proprioception in the four limbs. These clinical signs are rarely mentioned in the literature associated with hypothyroidism. Laboratory abnormalities are correlated with the severity and chronicity of the disease. The animal showed a decrease in total T4 and free T4, which is to be expected in a hypothyroid animal. As a result, levothyroxine replacement treatment was initiated. The dose used for the dog, which is the recommended dose in the literature, greatly increased its total T4 levels. As a result, the dose was readjusted after a new clinical evaluation. The rate of metabolism and absorption of levothyroxine varies widely and is independent of weight. The patient showed quickly improvement in neurological signs, activity level, and serum cholesterol rate. Regarding dermatological signs and body condition, there was a more gradual improvement. This corroborates what is mentioned in the literature, which indicates that improvements can take many months

Felines play a leading role in the epidemiology of Toxoplasma gondii infection, but there is scarce information about the epidemiology of Neospora caninum, particularly in feline immunodeficiency virus (FIV)-infected cats. Cats seropositive to T. gondii do not usually show symptoms unless they are immunosuppressed, such as FIV-infected cats. The same relationship remains poorly known for N. caninum, although it has been associated with neurological disorders in HIV-infected people. Since FIV-infected cats are prone to develop encephalitis of unknown etiology, this study aimed to evaluate the presence of specific antibodies to T. gondii and N. caninum in a shelter for stray cats naturally infected with FIV. A total of 104 serum samples from cats living in a shelter, located in São Paulo city (Brazil), was assessed for T. gondii and N. caninum specific antibody by indirect fluorescent-antibody test (IFAT). Of the 104 cats, 25 (24%) were infected with FIV and, aside from these, 8 (32%) had antibodies against T. gondii (titers from 16 to 128). Only 1 (4%) of the FIV-infected cats had antibodies against N. caninum, which was the first record of coinfection. Among the FIV-naïve cats, 11 (14%) were positive for T. gondii(titers from 16 to 256) and only 1 (1.2%) had antibodies against N. caninum. Serologically positive reactions to T. gondii and N. caninum were not correlated with age or sex (p>0.05), and there was no correlation between FIV and the occurrence of anti-T. gondii or anti-N. caninum antibodies (p>0.05). Further studies encompassing larger cat populations from different origins and locations are essential to clarify the prevalence of T. gondii and N. caninum antibodies in FIV-positive cats.(AU)


Os felinos têm um papel importante na epidemiologia da infecção por Toxoplasma gondii, mas pouco se sabe sobre a epidemiologia da infecção por Neospora caninum em gatos, particularmente em gatos infectados com o vírus da imunodeficiência felina (FIV). Gatos soropositivos para Toxoplasma gondii geralmente não apresentam sintomas a não ser que estejam imunossuprimidos, como gatos infectados com FIV. A mesma relação ainda é pouco conhecida para N. caninum, embora tenha sido associada a distúrbios neurológicos em pessoas infectadas pelo HIV. Considerando que gatos infectados com FIV são propensos a desenvolver encefalite de etiologia desconhecida, o presente estudo teve como objetivo avaliar a presença de anticorpos específicos para T. gondii e N. caninum em gatos infectados com FIV. Um total de 104 amostras de soro de gatos residentes em um abrigo na cidade de São Paulo, Brasil, foram avaliadas para a presença de anticorpos contra T. gondii e N. caninum pelo teste de imunofluorescência indireta (RIFI). Dos 104 gatos, 25 (24%) estavam infectados com FIV e destes 8, (32%) tinham anticorpos contra T. gondii (titulação entre 16 e 128). Apenas 1 (4%) dos gatos infectados com FIV apresentava anticorpos contra N. caninum, sendo este o primeiro registro dessa coinfecção. Entre os gatos não infectados com FIV, 11 (14%) foram positivos para T. gondii (titulação entre 16 e 256) e apenas 1 (1,2%) tinha anticorpos contra N. caninum. Background: Being the major cause of bovine abortion in the world, Neosporosis is considered to be a very important protozoal infection in dairy cattle. Vertical transplacental transmission is the major route of the infection causing either abortion or birth of calves with persistent infection. As the seropositivity in individual cows and in fetal serology only indicate exposure to the protozoa, the diagnosis of the infection has to be based on histopathology of aborted fetuses. Additional techniques such as immunohistochemistry (IHC) and PCR are required for the detection of the etiological agent. The purpose of the current study was to diagnose Neospora caninum infection in aborted bovine fetuses in Trakya Region of Turkey. For this purpose, serological, histopathological, IHC, and PCR methods were used. Materials, Methods & Results: The blood samples and the fetuses of 55 aborted dairy cattle from various farms located in 3 provinces of Trakya, Turkey constituted the material of the present study. The sera obtained from the blood samples were tested using a Neospora caninum Antibody Test Kit cELISA and anti-N. caninum antibodies were detected in the sera of the dams of the 8 aborted fetuses (8/55; 14.54%). Following the necropsy, samples from the brain, heart, liver, lung, kidney, spleen, and placenta of 55 fetuses were routinely processed for histopathological examination and evaluated under a light microscope. Nonsuppurative encephalitis (15/55; 27.27%), necrosis (5/55; 9%) and gliosis (1/55; 1.8%) in the brain, mild to severe nonsuppurative myocarditis and epicarditis (14/55; 25.45%), and portal to mid-zonal nonsuppurative hepatitis (13/55; 23.63%) were the relevant findings. PCR analysis was performed on fresh frozen fetal tissues. Nested PCR detected N. caninum DNA in the brain, heart, liver, lung, and kidney tissues of 6 fetuses (6/55; 10.9%). IHC was performed on the brain, heart, and liver tissues of all the fetuses using avidin-biotin-complex peroxidase method. Immunoreactivity was observed in the brain of 1 fetus (1/55; 1.8%). Discussion: In the present study, histopathological, immunohistochemical and PCR analyses were performed to detect N. caninum in 55 spontenously aborted bovine fetuses in Trakya Region, Turkey. Histopathologic hallmark of the study was nonsuppurative inflammation found mostly in the brain, heart and liver followed by kidneys and lungs. No protozoa was observed in the microscopic examination supporting the fact that definitive diagnosis of N. caninum infection requires ancillary techniques such as IHC and PCR. Nested PCR detected N. caninum DNA in the tissues of 6 fetuses (6/55; 10.9%). Brain was the most reliable organ for detection by PCR (6/6; 100%), compatible with the previous reports. IHC diagnosis revealed only 1.8% positivity in the present study which was remarkably lower than found in the previous studies. Even though histopathology in conjunction with IHC are accepted as the "gold standard" methods to detect N. caninum infection in aborted bovine fetuses, there are studies claiming that IHC is relatively insensitive in the diagnosis of neosporosis as parasite numbers can be low and thus, false negative results can be obtained. Other factors affecting the sensitivity of the technique are thoroughly discussed by many authors. Supportively, the findings of the current study showed that using both IHC and PCR as complementary techniques, increases the success of detection of N. caninum as recommended in previous studies. In conclusion, the present study demonstrated the first molecular diagnosis of Neospora caninum infection in bovine aborted fetuses in Trakya Region of Turkey which has a critical geographical location bordering Europe.

Dairy activity in the semi-arid region has gained prominence due to its profit potential; however, there are still sanitary problems that result in significant economic impact. Thus, this study aimed to assess the economic impact and to conduct an epidemiological study of caprine arthritis-encephalitis (CAE) and subclinical mastitis in dairy goat herds of the semi-arid northeastern region of Brazil. The study proceeded with the collection of blood (n = 351) and milk samples (n = 321) from 15 rural properties with up to 50 animals, risk factor assessment, economic profile analysis, and georeferencing of the properties. The blood serum obtained was subjected to the AGID test. The milk samples were cultured on blood agar, and the reading was performed 24 hours after incubation. Overall, 3.4% of the animals were seropositive for CAE, and 15.8% were diagnosed with subclinical mastitis. 71.95% of the positive animals belonged to properties with a semi-intensive rearing system, weekly cleaning of the facilities, and no pre- or post-dipping. Subclinical mastitis caused by the main agent Staphylococcus spp. was associated with the breed type of the animals, highlighting two risk factors: purebred animals (OR = 12.52) and the milking performed close the soil (OR = 18.07). A reduction of up to 35.34% was observed in the income of dairy properties, constituting a considerable economic impact. Due to these sanitary problems, the economic impact demonstrates the need for a sanitary management program focused on these infirmities.

Assim, objetivou-se dimensionar o impacto econômico e realizar o estudo epidemiológico da artrite encefalite caprina (CAE) e mastite subclínica, nos rebanhos caprinos leiteiros do semiárido nordestino. A pesquisa seguiu-se com a coleta das amostras de sangue (n=351) e leite (n= 321), ava-liação dos fatores de risco, análise do perfil econômico das propriedades e georreferenciamento. O soro sanguíneo obtido foi submetido ao teste de IDGA. As amostras de leite foram semeadas em ágar sangue e a leitura realizada 24 horas após incuba-ção. No total, 3,4%, dos animais foram soropositivos para CAE e 15,8% diagnosticados com mastite subclínica. 71,95% dos animais positivos pertenciam a propriedades com sistema de criação semi-intensivo, limpeza das instalações semanalmente e sem realização de pré e pós dipping. A mastite subclínica causada pelo principal agente Staphylococcus spp. mostrou-se asso-ciada ao padrão racial dos animais, sendo observado dois fatores de risco: padrão racial puro (OR = 12,52) e ordenha no solo (OR = 18,07). A redução nas receitas das propriedades leiteiras foi de até 35,34%, portanto, um relevante impacto econômico. Pode-se constatar a circulação do vírus da CAE nas propriedades rurais visitadas, além da presença de patógenos causadores da mastite com importância na área da saúde pública. O impacto econômico gerado em função da presença desses proble-mas sanitários demonstra a necessidade da implantação de um programa de manejo sanitário com foco nessas enfermidades.

Forty 1-2-y-old water buffaloes were simultaneously treated with trichlorfon and chlorpyrifos products in the recommended dose for cattle. After a week, 19 animals started presenting clinical signs characterized by apathy, diarrhea, aggressiveness, dehydration, and motor incoordination, followed by flaccid paralysis and permanent lateral recumbency. All affected buffaloes died after a clinical course of 1-4 days. Reduction of serum cholinesterase activity in three cases was indicative of significant exposure to organophosphorus compounds (OPs). Pathological examination of three buffaloes revealed no gross and histological lesions. By thin layer chromatography, chlorpyrifos residues and trace of trichlorfon residues were detected in fresh tissue samples. The epidemiological, clinical, pathological, and toxicological findings were highly compatible with OPs-induced delayed neurotoxicity, a neurological manifestation rarely described in domestic animals.


Quarenta búfalos foram simultaneamente tratados com clorpirifós e triclorfom na dose recomendada para bovinos. Após uma semana, 19 animais apresentaram sinais clínicos caracterizados por apatia, diarreia, agressividade, desidratação e incoordenação motora, seguidos por paralisia flácida e decúbito lateral permanente. Todos os búfalos afetados morreram após um curso clínico de 1-4 dias. Redução da atividade da colinesterase sérica em três casos foi indicativa de exposição significativa a organofosforados (OPs). O exame patológico de três búfalos não revelou lesões macroscópicas e histológicas. Por cromatografia em camada delgada, resíduos de clorpirifós e traços de resíduos de triclorfon foram detectados em amostras de tecidos frescos. Os achados epidemiológicos, clínicos, patológicos e toxicológicos foram compatíveis com neuropatia tardia induzida por OPs, uma manifestação neurológica raramente descrita em animais domésticos.

O vírus do Nilo Ocidental (VNO) é um arbovírus transmitido principalmente por mosquitos do gênero Culex e responsável pela doença Febre do Nilo Ocidental (FNO). Foi identificado no Brasil pela primeira vez em 2009, através de um estudo soro-epidemiológico em equídeos e, desde então, a presença de anticorpos contra o VNO e/ou ácido nucleico viral tem sido identificado em seres humanos, equídeos e aves. Por causar infecções neurológicas em animais e humanos, é considerado um problema global de saúde pública. O controle da FNO está intimamente relacionado à atuação dos médicos-veterinários na vigilância, principalmente, de casos em equídeos, animais considerados sentinelas na identificação da doença.(AU)


The West Nile virus is an arbovirus transmitted mainly by Culex mosquitoes and responsible for West Nile Fever (FNO) disease. It was identified in Brazil for the first time in 2009, through a seroepidemiological surveillance in equids and, since then, the presence of antibodies against the virus and/or viral nucleic acid has been identified in humans, horses and birds. The virus is responsible for neurological infections in animals and humans, and is considered a global public health problem. The control of WNF is closely related to the role of veterinarians, mainly in the surveillance of cases in equines, animals considered sentinels in the identification of the disease.(AU)

O controle da FNO está intimamente relacionado à atuação dos médicos-veterinários na vigilância, principalmente, de casos em equídeos, animais considerados sentinelas na identificação da doença.


The West Nile virus is an arbovirus transmitted mainly by Culex mosquitoes and responsible for West Nile Fever (FNO) disease. It was identified in Brazil for the first time in 2009, through a seroepidemiological surveillance in equids and, since then, the presence of antibodies against the virus and/or viral nucleic acid has been identified in humans, horses and birds. The virus is responsible for neurological infections in animals and humans, and is considered a global public health problem. The control of WNF is closely related to the role of veterinarians, mainly in the surveillance of cases in equines, animals considered sentinels in the identification of the disease.

Resumo

Objetivou-se descrever a ocorrência do Bovine alphaherpesvirus 5 (BoHV5) como causa de meningoencefalite não supurativa em bovinos do estado de Pernambuco, Brasil. Para tanto, 32 amostras de sistema nervoso embebidas em parafina foram obtidas de animais acometidos por doenças neurológicas atendidos na Clínica de Bovinos de Garanhuns da Universidade Federal Rural de Pernambuco (CBG-UFRPE), entre 2012 e 2016. As amostras foram analisadas quanto à presença do gene da glicoproteína C do BoHV5 por reação em cadeia da polimerase (PCR). Dois animais (6,25%) tiveram resultado positivo à PCR, e sua análise de sequenciamento indicou 100% de similaridade para o BoHV5. Os resultados histopatológicos desses dois animais revelaram lesões multifocais de meningoencefalite não supurativa associada à polioencefalomalácia, presença de corpúsculos de inclusão basofílicos, infiltração de células de Gitter e presença de manguitos perivasculares. A PCR se mostra uma importante ferramenta para diferenciação das infecções por BoHV5 de outras enfermidades neurológicas de bovinos, especialmente a raiva.(AU)
