Wooden breast myopathy (WB) strongly affects the poultry industry mainly in terms of consumers rejection and economical loses, due to morphological changes in broiler muscle tissue and consequently low meat quality. The aim of this study is to evaluate the histomorphometry of muscle fibers of breast fillets of broilers with severity levels of WB myopathy. The histological evaluation considered 30 samples of the pectoralis major muscle and the level of WB myopathy (ten normal fillets, ten moderate fillets, and ten severe fillets). Fillets with a severe level of WB present low average fiber number, high average fiber diameter, low percentage of fibers with diameter of less than 20 µm, low percentage of fibers with diameter between 20 and 40 µm, and high percentage of fibers with diameter between 40 and 70 µm. Fiber cross-sectional area is greater in fillets affected by moderate and severe WB. Thus, fillets with a severe level of WB damage the muscle fiber structure and contribute more severely to the degenerative processes of breast meat.(AU)

The liver was divided into three lobes with the gallbladder close to the right lobe and its color varied from light red to dark red, not varying in relation to other fish species. The hepatosomatic relationship followed body growth until the PII group stage and then declined, demonstrating the behavior of its development in Brycon amazonicus. The organ consists predominantly of hepatocytes, followed by sinusoidal vessels and capillaries, with histological morphology similar to that of many species of fish. Melanomacrophage centers were found only in the most developed animals, but in small quantities, prompting the development of new research on this cell, in this species. In this way, research of this nature allows the characterization of fish species, helping to improve breeding methods, understanding pathological processes caused by diseases, and obtaining better productive capacity, serving an increasingly demanding and prosperous market.

Purpose: To compare the effect of vein conduit filled with adipose tissue stem cells (ASC) on peripheral nerve injury regeneration. Methods: We analyzed 30 male Wistar rats surgically submitted to a 5-mm gap on the sciatic nerve. Then, the animals were divided into three groups: nerve autografting (AG, n=10), autogenous inverted glycerol-conserved vein (VG, n=10), and autogenous inverted glycerol-conserved vein + ASC (VASCG, n=10). The study endpoints were neuromotor functional analysis, gastrocnemius muscle weight, and sciatic nerve graft histomorphometry analysis. In the histologic analysis, we added a control group (naïve nerve). Results: Regarding functional analysis (Walking tract- score), the findings at week 3 showed a difference between the AG and the VG (-96.6 vs. -59.6, p=0.01, respectively) and between the VG and the inverted vein + VASCG (-59.9 vs. -88.92, p=0.02). At week 12, this study showed a difference between the AG and the VG (-64.8 vs. -47.3, p=0.004, respectively), and also a difference between the VG and the VASCG (-47.3 vs. -57.4, p=0.02, respectively). There was no difference in the histomorphometry analysis (nerve diameter, Schwann cells counting). The gastrocnemius muscles on the intervention side were more atrophic when compared to the gastrocnemius muscles on the control side. Conclusions: Our results suggested better functional recovery in the inverted vein group when compared to control group, and inverted vein + ASC group.

The tongue in its dorsal, lateral and ventral surfaces is covered with a non-keratinized multilayered stratified squamous epithelium. Lamina propria is present beneath the epithelial layers. Morphometrically, thickness of the apex tongue epithelium is more than that in the tongue body. Thickness of the ventral surface of the tongue is less than that in the dorsal one. Thickness of the lateral surface of the tongue was thicker than that in the ventral one and tongue body. Large and small conical papillae appeared on the posterior dorsal surface of the lingual body. There are lingual glands in certain areas of tongue body with numerous openings through the dorsal surface.

In bridging the knowledge gap on stress physiology of Nigerian indigenous chickens, this study investigated the effect of exogenous corticosterone (eCORT) as stress inducing agent on the testicular function and mating behavior of Nigerian indigenous cocks. Twenty-four (24) cocks and one hundred and forty four (144) hens (mating ratio of 1 cock: 6 hens) were grouped into four and assigned to each of the four eCORT treatments (0, 2, 4 and 6 mgeCORT/KgBW) daily for 14 days. Semen samples were collected on days 0, 7 and 14 and analyzed for semen volume (SV), progressive sperm motility (PSM), membrane integrity (MI) and sperm abnormality (SA). Mating behaviors were monitored on days 3, 5 and 8. Blood samples, for hormonal (Luteinizing Hormone (LH), Follicle Stimulating Hormone (FSH) Testosterone (TEST) and stress analysis (heterophil/lymphocyte ratio, H/L) were collected from brachial vein on days 7 and 14. On day 15, cocks were euthanized and testes harvested for histomorphometry. Data were analyzed using multivariate analysis, oneway ANOVA and Kruskal-Wallis tests all in SPSS 23. Administration of 4 mgeCORT/KgBW declined (P0.05) influence on the mating behaviors, H/L ratio, FSH and TEST. However, 2 mgeCORT/KgBW enhanced LH levels. Administration of eCORT did not affect the testicular epithelial height and seminiferous tubular diameter. In conclusion, optimal stress induced by eCORT impaired semen quality but with less impact on reproductive hormones, H/L and mating behaviors of intensively raised Nigerian indigenous cocks.(AU)

Purpose: Poultry by-products can contribute as an innovative natural source for the development of composites based on polymers and minerals aiming at bone regeneration. The objective of this study was the physicochemical and biological characterization of collagen-based hydrogels crosslinked with ultraviolet (UV)-riboflavin. Methods: Pure hydrogels of 100% collagen (G1) or hybrid hydrogels, 90% collagen:10% apatite (G2), 90% collagen:10% nanokeratin (G3), and 90% collagen:5% apatite:5% nanokeratin (G4) were characterized by scanning electron microscope, Fourier-transform infrared spectroscopy, differential scanning calorimetry, swelling degree and quali-quantitative histological analysis. Ectopic implantation in subcutaneous tissue in mice at one, three and nine weeks allowed to assess the inflammation (neutrophils, lymphocytes, macrophages, and giant cells) and repair (neovascularization, and connective tissue) to determine biocompatibility and the integrity of biomaterials to score their biodegradability. Histomorphometry on critical size defects in rat calvaria at one and three months evaluated the percentage of bone, connective tissue, and biomaterials in all groups. Results: The hydrogels presented porous microstructure, water absorption and physicochemical characteristics compatible with their polymeric and/or mineral composition. All materials exhibited biocompatibility, biodegradability, and low osteoconductivity. G2 showed greater density of new bone and biomaterial than the G1, G3 and G4. Conclusions: The collagen-apatite group formulation suggests potential for development as osteopromoting membrane.

This experiment was conducted to evaluate the supplemental effects of a novel protease produced from Bacillus subtilis in low crude protein (CP) corn distiller dried grain with solubles (cDDGS) based diets on growth performance, carcass attributes, nutrients digestibility, blood chemistry, and intestinal histomorphometry of broiler chickens. One hundred and sixty, one-day-old chicks were randomly allotted to one of 4 dietary treatments. Each dietary treatment had four replicates, with 10 birds in each replicate. Two basal diets were formulated for both starter (1-21d) and finisher (22-35d) phase; (PC) a corn soybean meal based diet as per standard recommendations of Ross 308; (NC) 5% cDGGS with 5% reduction in CP with concomitant reduction in essential amino acids (EAAs) compared with PC. The negative control diet was further subdivided into 3 parts. One part was without enzyme supplementation, while the other two parts were supplemented with a novel protease (PROT1) and a commercial protease (PROT2), respectively. The same procedure was adopted for finisher diets. A digestibility assay (32-35d) was carried out using acid insoluble ash (AIA), an external digestibility marker. At the end of 35d, ileal digesta were collected from four birds per experimental unit for nutrient digestibility measurement. Tissue samples of duodenum, jejunum, and ileum were collected for villus height, villus width, crypt depth, and crypt width. Body weight gain (BWG) and feed:gain were improved (p<0.05) with protease supplementation. No effect was observed on carcass parameters. However, CP digestibility, apparent digestibility coefficient for nitrogen (ADCn), nitrogen retention (Nret ), and apparent ileal amino acid digestibility (AIAAD) were improved (p<0.05). However, there was no effect on apparent metabolizable energy (AME) and apparent metabolizable energy corrected for nitrogen (AMEn), blood glucose, total protein and cholesterol (p>0.05) and intestinal integrity (p>0.05). It was concluded that protease enzyme can improve nitrogen and CP digestibility, resulting in improved amino acids availability in low protein diets.(AU)

The aim of the present study was to evaluate the post-vaccinal reaction to two lentogenic vaccine strains of Newcatle disease virus (NDV) and a recombinant turkey herpesvirus (rHVT) vaccine expressing the fusion glycoprotein of NDV in broiler chickens through histomorphometric and histopathologic analyses of the trachea. The experiment involved 245 chicks housed in randomized blocks with three different enclosures under controlled conditions of temperature, light and ventilation. Each enclosure represented a vaccine strain and was divided into groups according to the administration route. Each block also had its own control group composed of unvaccinated birds. The vaccine strains PHY.LMV.42 (PL42) and La Sota (LS) were selected according to the Intracerebral Pathogenicity Index (ICPI) and the rHVT-NDV Serotype 3 strain (ST3) was selected for representing non-NDV infection. At two, four, seven, 14 and 21 days post vaccination, fragments from the middle third of the trachea were collected and submitted to routine histological processing. For the histomorphometric analysis, the slides were photographed, and the thickness of the tracheal mucosa was measured. Statistical analysis involved two-way ANOVA and Tukey's post-hoc test with a 5% significance level. For the histopathological evaluation, lesions were described as to the degree of intensity and distribution. At four and 14 days post vaccination with the LS strain administered by the ocular route, the means of thickening of the tracheal mucosa (20.85±7.31µm and 26.97±5.50µm, respectively) were significantly higher (p<0.05) than for all other strains, which was related to the severe histopathological lesions found in this group, characterized by hyperemia, hyperplasia of the mucous glands, moderate deciliation and multifocal lymphohistiocytic inflammatory infiltrate. At 21 days, broiler chickens vaccinated with the ST3 strain showed more discrete lesions and less thickening of the tracheal mucosa (23.23±7.62µm; p<0.05) in comparison with other studied strains. The lesions found in this group were only hemorrhage, deciliation and mild focal lymphocytic inflammatory infiltrate. The results of the histomorphometry and histopathology of the trachea indicated that vaccination with rHVT-NDV Serotype 3 strain induced lower degree post-vaccine tracheal lesions compared to other vaccine strains analyzed in this study.
Objetivou-se avaliar a reação pós-vacinal de duas estirpes lentogênicas do vírus da doença de Newcastle (VDN) e uma vacina recombinante de herpesvirus de perus (rHVT) que expressa a glicoproteína de fusão de VDN em frangos de corte por meio da histomorfometria e histopatologia da traqueia. Foram utilizados 245 pintos alojados em blocos ao acaso, sendo três galpões distintos em condições controladas de temperatura, luz e ventilação. Cada galpão representou uma cepa vacinal, onde foram divididos por grupos de acordo com a via de administração. Todos os blocos possuíam um grupo controle composto por aves não vacinadas. As cepas vacinais PHY.LMV.42 (PL42) e La Sota (LS) utilizadas foram selecionadas de acordo com o Índice de Patogenicidade Intracerebral (IPIC) e a cepa Sorotipo 3 (ST3), da vacina rHVT-VDN foi selecionada por não representar infecção do VDN. Aos dois, quarto, sete, 14 e 21 dias pós-vacinação, fragmentos do terço médio da traqueia foram coletados e posteriormente processados conforme rotina histológica. Para análise histomorfométrica da mucosa traqueal, as lâminas foram fotografadas e realizadas as mensurações da espessura da mucosa traqueal sendo aplicado teste de anaÌlise de variaÌncia a dois criteÌrios (ANOVA) e utilizando o post-hoc de Tukey com niÌvel de significaÌncia de 5%. Para a avaliação histopatológica foram observadas a presença de lesões microscópicas e estas foram descritas quanto ao grau de intensidade e distribuição. Aos quatro e quatorze dias pós-vacinação com a cepa LS administrada por via ocular, as médias do espessamento da mucosa traqueal (20,85±7,31µm e 26,97±5,50µm, respectivamente) foram significativamente maiores (p<0,05) quando comparada a todas as demais cepas utilizadas, isto se deve às severas lesões histopatológicas encontrados neste grupo, caracterizadas por hiperemia, hiperplasia das glândulas mucosas, deciliação moderada e infiltrado inflamatório linfohistiocitário multifocal moderado. Já aos 21 dias as aves vacinadas com a cepa ST3 apresentaram lesões mais discretas e menor espessamento da mucosa da traqueia (23,23±7,62µm; p<0,05) em comparação às demais cepas estudadas. As lesões encontradas neste grupo foram apenas hemorragia, deciliação e infiltrado inflamatório linfocitário focal discreto. Os resultados da histomorfometria e da histopatologia da traqueia indicou que a vacinação com a rHVT-NDV, cepa Sorotipo 3 induziu menor grau de lesões pós-vacinais na traqueia comparada a outras cepas vacinais analisadas nesse estudo.

The objective of this study was to evaluate the effects of guava extract standardized in phenolic compounds (SGE) on broiler performance, nutrient digestibility, and intestinal histomorphometry. A total of 300 one-day-old male Cobb-500® broiler chicks were distributed in a completely randomized design with five treatments (basal diet supplemented with 120 mg vitamin E/kg and basal diet supplemented with 0, 600, 800, or 1,000 mg SGE/kg) and six replicates of ten birds each. Performance was evaluated at seven and 21 days of age; digestibility of nutrients was determined by total excreta collection from 18 to 21 days of age; and histomorphometry of the small intestine was assessed at 21 days of age. Broilers fed diets supplemented with SGE or vitamin E had higher body weight and weight gain and better feed conversion than those fed unsupplemented diet at seven days of age. At the same age, there was a quadratic effect of SGE levels on body weight and weight gain, with better weights for 715 and 716 mg SGE/kg, respectively; and a decreasing linear effect for feed conversion. At 21 days of age, body weight and weight gain increased linearly with the inclusion of SGE in diet. Digestibility of feed nutrients was not influenced by treatments. Broilers fed diet supplemented with 800 or 1,000 mg SGE/kg had greater villus height and villus:crypt ratio of the duodenum than those fed unsupplemented diet. Villus height in the jejunum of broilers fed diet supplemented with 600 mg SGE/kg was lower than that of broilers that received vitamin E. Guava extract standardized in phenolic compounds can be used in diets for broilers in the starter phase, considering that the extract increases weight gain, reduces feed conversion, and helps in the development of the intestinal mucosa.

Abstract Glutamine is often used to treat metabolic changes associated with anorexia-cachexia syndrome in patients with malignant neoplasms. Walker 256 tumor is an excellent model for studying these changes associated with cancer in different organs, including injuries in testicular functions. However, the effects of supplementing glutamine on testicular morphometry in this model have not yet been investigated. Thus, the objective of this study was to evaluate the effect of L-glutamine supplementation on testicular morphometry in rats transplanted with Walker 256 tumor cells. Forty puberty Wistar rats were divided into four groups: control without L-glutamine (C); control supplemented with L-glutamine (CG); inoculated with Walker 256 tumor cells (WT) and inoculated with Walker 256 tumor cells and supplemented with L-glutamine (WTG). The testicles were removed, weighed, fixed in Bouin, and included in paraffin for histomorphometric analysis. Walker 256 tumor caused quantitative changes in the tubular and intertubular compartments and tunica albuginea, with reductions in the percentages of lumen and tunica albuginea, number of Sertoli cells per gram of testis; number of Leydig cells; percentage of blood vessels and connective tissue in intertubule. However, glutamine supplementation prevented part of these changes caused by the tumor, presenting mainly a protective effect on the tunica albuginea and percentage of blood and lymph vessels in the intertubule. These results indicate the potential of L-glutamine was able to recover for testicular dysfunction associated with cancer.

Post-hatch delayed placement damages, physical and physiological development of broiler chicks. The objective of this study was to ascertain adequate levels of glutamine inclusion in post-hatch and pre-starter feed, in order to minimize the negative effects of post-hatch delayed placement on broiler chicks. The birds were distributed in a completely randomized design, with four treatments and five replicates of ten birds each. Four levels of glutamine supplementation (0, 1, 2 and 3%) were used in the pre-starter feed, which was given to the chicks in the transportation box and during the pre-starter phase. After 24 hours of access to this feed, and at 7 days of age, the chicks performance, yolk sac retraction, plasma glucose concentration, weight and small-intestine histomorphometry were evaluated. From seven to ten days of age, a metabolizability assay was performed. Glutamine supplementation tended to increase the use of the yolk sac, the concentration of plasma glucose (p 0.10) and the depth of the crypt in the ileum after 24 hours of life (p 0.05). There was no statistical difference between the glutamine supplementation levels, in relation to glucose, histomorphometry or metabolizability in the pre-starter phase. Glutamine concentrations did not differ in relation to performance, histomorphometry of the small intestine or metabolizability of nutrients in the pre-starter phase.(AU)

ABSTRACT The objective of this study was to evaluate the effects of dietary inclusion of protected sodium butyrate (PSB) on the intestinal development and feed nutrient metabolizability of commercial laying hens. The birds started to receive the treatment rations at 58 weeks of age. At 76 weeks of age the laying hens were distributed in a randomized block design to four treatments (0, 105, 210 and 300 g t-1/PSB), six replicates, and two birds/replicate. The nitrogen balance (NB), ether extract balance (EEB), dry matter metabolizability coefficient, nitrogen, ether extract, ash, apparent metabolizable energy (AME), and AME corrected by nitrogen (AMEn) were evaluated. For assessment of intestinal development, we evaluated the duodenum, jejunum, ileum, cecum, and colorectal lengths, the relative intestine weight and villus height, crypt depth, and villus:crypt ratio of the duodenum, jejunum, and ileum. A decreasing linear effect was observed in the duodenum length, while an increasing linear effect was observed in the height of the duodenum and jejunum villi. A quadratic effect was found in the jejunum crypt depth. A linear increasing effect was found on the villus:crypt ratio of the duodenum and jejunum, and a quadratic effect was observed in the ileum. Quadratic and increasing linear effects were observed in the NB and EEB, respectively. Additionally, increasing linear effects were observed in the AME and AMEn. The dietary addition of 300 g t-1 of PSB improved intestinal development and energy metabolizability of the diet.

ABSTRACT Description and seasonal variation in epididymal histomorphometry of Dermanuracinerea (Chiroptera: Phyllostomidae) in a fragment of the Atlantic Forest in the Northeastern Brazil. This study aimed to evaluate seasonal patterns in the histomorphometry of the epididymis of Dermanura cinerea (Gervais, 1856) in a fragment of the Atlantic Forest in the Northeastern Brazil. Eighteen adult male specimens captured by mist net were used. The field-work occurred monthly over 18 months, during two consecutive nights. Meteorological data (precipitation) were obtained from the National Institute of Meteorology. After euthanasia, specimens had the epididymis collected, which were fixed and processed. The histological slides produced were stained by Hematoxylin - Eosin and analyzed by optical microscopy. The morphometric parameters analyzed were the tubular, lumen and epithelium areas, of the regions of the initial segment, caput, corpus and cauda of the epididymis. The histomorphometric data were submitted to the Mann-Whitney U test analyzes. The results showed that D. cinerea presented spermatozoa in all regions of the epididymis, except in the initial segment. The highest averages of the tubular, lumen and epithelial areas in the four regions were observed during the dry months. Therefore, D cinerea presented greater sensibility in the region of the cauda of the epididymis, during the months with low rainfall indices. This indicates that environmental conditions have considerable influence on the epididymal morphophysiology of this species, especially in relation to the storage of sperm in the tail of this organ, in area of Atlantic forest in northeastern Brazil.
RESUMO Esse estudo objetivou avaliar sazonalmente a histomorfometria do epidídimo de Dermanura cinerea (Gervais, 1856) em um fragmento de Mata Atlântica no nordeste do Brasil. Foram utilizados 18 espécimes machos adultos capturados por redes de neblina. As coletas ocorreram mensalmente ao longo de dezoito meses, durante duas noites consecutivas e os dados meteorológicos foram fornecidos pelo Instituto Nacional de Meteorologia. Depois de eutanasiados, os espécimes tiveram os epidídimos coletados e esses órgãos foram fixados e processados. As lâminas histológicas foram coradas por Hematoxilina - Eosina e analisadas em microscopia óptica. Os parâmetros morfométricos analisados foram as áreas do túbulo, do lúmen e do epitélio das regiões do segmento inicial, cabeça, corpo e cauda do epidídimo. Os dados histomorfométricos obtidos foram submetidos às análises no teste U de Mann-Whitney. Os resultados revelaram que D. cinerea apresentou espermatozoides em todas as regiões do epidídimo, exceto no segmento inicial. As maiores médias das áreas tubular, do lúmen e do epitélio nas quatro regiões, foram constatadas durante os meses secos. Portanto, D cinerea apresentou maior sensibilidade na região da cauda do epidídimo, ao longo dos meses com baixos índices pluviométricos. Isso indica que as condições ambientais apresentam considerável influência sobre a morfofisiologia epidídimária dessa espécie, sobretudo, em relação ao armazenamento de espermatozoides na cauda desse órgão, em área de Mata atlântica do nordeste brasileiro.
