Resumo

Purpose: To investigate the role and mechanism of ß1,3-N-acetylglucosaminyltransferase-3 gene (B3GNT3) in esophageal cancer (ESCA). Methods: The starBase database was used to evaluate the expression of B3GNT3. B3GNT3 function was measured using KYSE-30 and KYSE-410 cells of esophageal squamous cell carcinoma (ESCC) cell lines. The mRNA levels were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Cell counting kit-8, clone formation assay and transwell assay were used to detect the changes of proliferation, invasion and migration. Results: B3GNT3 expression was higher in ESCA tissues than in normal tissues. The overall survival rate of ESCA patients with high B3GNT3 expression was lower than that of ESCA patients with low B3GNT3 expression. In vitro functional experiments showed that the proliferation ability, migration and invasion ability of KYSE-30 and KYSE-410 cells with B3GNT3 interference were lower than those of the control, and the overexpression of B3GNT3 had the opposite effect. After silencing B3GNT3 expression in ESCC cell lines, the growth of both cell lines was inhibited and the invasiveness was decreased. Knockdown of B3GNT3 reduced the growth rate and Ki-67 expression level. Conclusion: B3GNT3, as an oncogene, may promote the growth, invasion and migration of ESCC cell.

Background: Thyroid tumor is a common endocrine tumor that accounts for up to 3.8% of all tumors in dogs. Most of them are malignant and usually nonfunctional in dogs. 18F-fluorodeoxyglucose positron emission tomography (FDG-PET) is an imaging modality that detects intracellular accumulation of radioactive deoxyglucose administered in the body and is used in combination with computed tomography to provide functional information with exact anatomical localization. It is used in human medicine to detect residual or recurrent head and neck neoplasm after treatments, such as surgical resection. This report describes the first case of diagnosing recurrent thyroid carcinoma (TC) through FDG-PET in a dog. Case: A 9-year-old castrated male Maltese dog presented with a palpable mobile mass in the right ventral cervical region. Radiography and ultrasonography (US) showed a radiopaque mass adjacent to the trachea, and the right thyroid gland was enlarged on computed tomography. The surgically excised mass was encapsulated and measured to be 2.3 × 1.0 × 3.4 cm (width x length x height) in size. Histopathologically, the mass was diagnosed as differentiated follicular TC, and gross and vascular invasions were observed. To prevent recurrence, postoperative carboplatin chemotherapy was performed for 5 months. Two months after completion of chemotherapy, a nodule of approximately 7 mm in diameter was detected in the thyroidectomy bed by US. FDG-PET scanning was performed as an effective means of evaluating the malignancy, local recurrence, and metastasis of differentiated follicular TC. The nodule had the dimensions of 2.8 × 5.9 × 8.6 mm, a maximum standardized uptake value (SUV) of 8.49, and a mean SUV of 5.6. The results of FDG-PET suggested the recurrence of TC; therefore, the second chemotherapy protocol using toceranib was applied for 16 months. After initiation of the 2nd chemotherapy, follow-up examinations were conducted approximately every 4 months. On the 134th day, although the nodule was not palpated, its size was observed to have increased to 5.0 × 3.8 × 13.6 mm on cervical US on the 232nd day, showing heterogeneous and hypoechoic parenchyma. On the 405th day, the tumor was enlarged to a size of 13.4 × 12.9 × 22 mm and identified as a lobular, amorphous shape, and its heterogeneity was increased. Moreover, 2 pulmonary nodules with well-defined margins were found on radiography in the left caudal lung lobe (9 × 10 mm and 12 × 12 mm [width × length]); thus, lung metastasis was suspected. On the 536th day, anorexia and lethargy occurred, and the dog was lost to follow-up. Discussion: In the present case, local recurrence of TC was suspected based on cervical US. Although US was useful as a screening tool, additional examinations were necessary for evaluating local invasiveness, malignancy, and nodal/distant metastasis. FDG-PET can detect recurrence at an early stage because it can sense increased tumor metabolism through physiologic absorption of FDG, even before the beginning of anatomic change in the lesion. Therefore, FDG-PET can assist in treatment planning and provide better prognosis. In humans, focal FDG uptake and a high maximum SUV in the thyroid gland on FDG-PET were associated with a higher risk of cancer. Because there was no evidence of neoplasia except the thyroid lesion during the FDG-PET examination, the tumor showed an increasingly malignant pattern of the thyroid gland on US during the follow-up period, and the metastatic pulmonary nodules were identified on the 650th day after the thyroidectomy, the present case was diagnosed as recurrent TC. This report describes the use of FDG-PET for diagnosing local recurrence of TC, pointing to FDG-PET as a potential strategy to evaluate loco-regional recurrence and distant metastasis of TC.

Abstract Fluoroquinolones are important antimicrobial agents for the treatment of Pseudomonas infections. A total of 11 isolates of P. aeruginosa were collected from different clinical samples from different medical centers in the North West Bank-Palestine during 2017. In this study, resistance to fluoroquinolones and secretions of -lactamases were detected by phenotypic methods, while presence of -lactamase gene sequences and other virulence factors were detected by PCR technique. PCR product for gyrA, parC and parE genes were sequenced for further analyses. The phylogenetic analyses, population diversity indices and haplotypes determination were conducted using computer programs MEGA version 6, DnaSP 5.1001 and median-joining algorithm in the program Network 5, respectively. Results of this study showed that the MIC for ciprofloxacin and norfloxacin had a range of 32-256 µg/ml. In addition, all isolates carried either exoT or exoT and exoY genes, different -lactamase genes and 82% of these isolates harbored class 1 integrons. Analyses of the gyrA, parC and parE sequences were found to be polymorphic, had high haplotype diversity (0.945-0.982), low nucleotide diversity (0.01225-0.02001) and number of haplotypes were 9 for each gyrA and parE genes and 10 haplotypes for parC gene. The founder haplotypes being Hap-1 (18%), Hap-2 (27.3%) and Hap-6 (9.1%) for gyrA, parC and parE genes, respectively. Two of ParE haplotypes were detected as indel haplotypes. The Median-joining- (MJ) networks constructed from haplotypes of these genes showed a star-like expansion. The neutrality tests (Tajimas D test and Fus Fs test) for these genes showed negative values. Palestinian fluoroquinolone resistant P. aeruginosa strains showed high MIC level for fluoroquinolones, -lactamase producers, carried type III secretion exotoxin-encoding genes, most of them had integrase I gene and had high level of mutations in QRDR regions in gyrA, parC and parE genes. All these factors may play an important role in the invasiveness of these strains and make them difficult to treat. Isolation of these strains from different medical centers, indicate the need for a strict application of infection control measures in Medical centers in the North West Bank-Palestine that aim to reduce expense and damage caused by P. aeruginosa infections. Molecular analyses showed that Palestinian fluoroquinolone resistant P. aeruginosa haplotypes are not genetically differentiated; however, more mutations may exist in these strains.


Resumo Fluoroquinolonas são agentes antimicrobianos importantes para o tratamento de infecções por Pseudomonas. Um total de 11 bacilos isolados de P. aeruginosa foram coletados de diferentes amostras clínicas provenientes de diferentes centros médicos na Cisjordânia-Palestina durante o ano de 2017. Neste estudo, resistência a fluoroquinolonas e secreções de -lactamases foram detectadas por métodos fenotípicos, enquanto a presença de sequências do gene -lactamase e outros fatores de virulência foram detectados pela técnica de PCR (Proteína C-reativa). O produto de PCR para os genes gyrA, parC e parE foram sequenciados para análises posteriores. As análises filogenéticas, os índices de diversidade populacional e a determinação de haplótipos foram realizados utilizando os softwares MEGA versão 6, DnaSP 5.1001 e o algoritmo de junção de mediana do programa Network 5, respectivamente. Os resultados deste estudo mostraram que a MIC para ciprofloxacina e norfloxacina tinha um intervalo de 32-256 µg/ml. Além disso, todos os bacilos isolados carregavam genes exoT ou exoT e exoY, genes de -lactamase diferentes e 82% desses isolados continham integrons de classe 1. As análises das sequências gyrA, parC e parE foram consideradas polimórficas, com alta diversidade de haplótipos (0,945-0,982), baixa diversidade de nucleotídeos (0,01225-0,02001) e o número de haplótipos foi de 9 para cada gene de gyrA e parE e 10 haplótipos para o gene parC. Os haplótipos fundadores são Hap-1 (18%), Hap-2 (27,3%) e Hap-6 (9,1%) para os genes gyrA, parC e parE, respectivamente. Dois dos haplótipos parE foram detectados como haplótipos InDel. As redes Median-joining (MJ) construídas a partir de haplótipos desses genes mostraram uma expansão semelhante à de uma estrela. Os testes de neutralidade (teste D de Tajima e teste Fs de Fu) para esses genes apresentaram valores negativos. As cepas palestinas de P. aeruginosa resistentes a fluoroquinolonas mostraram alto nível de MIC para fluoroquinolonas, produtores de -lactamase, genes codificadores de exotoxina de secreção tipo III, a maioria deles tinha o gene integrase I e tinha alto nível de mutações nas regiões QRDR nos genes gyrA, parC e parE. Todos esses fatores podem desempenhar um papel importante na invasão dessas cepas e torná-las difíceis de tratar. O isolamento dessas cepas em diferentes centros médicos, indica a necessidade de uma aplicação estrita de medidas de controle de infecção em centros médicos da Cisjordânia-Palestina que visam reduzir despesas e danos causados por infecções por P. aeruginosa. As análises moleculares mostraram que os haplótipos de P. aeruginosa resistentes à fluoroquinolona palestina não são geneticamente diferenciados; no entanto, mais mutações podem existir nessas cepas.

Resumo

Purpose: Fluorescence spectroscopy techniques have been investigated aiming to reduce the invasiveness of methods for investigation of tissue. In transplantation procedures, it may offer the possibility of a complementary technique for the monitoring of liver grafts' conditions prior to and during the transplantation procedure stages involving cold perfusion. The objective of this study was to evaluate fluorescence spectroscopy under violet light excitation (408 nm) for the monitoring of clinical hypothermic liver transplantation procedures. Methods: Organ grafts were monitored from before the removal of the donor's body to 1 h after the implant into the receptor's body. Fluorescence spectroscopy was assessed over five stages within these transplant stages. Results: The study provided evidence of a correlation between fluorescence information collected during liver grafts transplantation and the survival of patients. Conclusions: Fluorescence spectroscopy can become a tool to monitor transplantation grafts, providing objective information for the final decision of surgeons to use organs.

Background: Lacrimal gland adenocarcinoma is an extremely rare malignant neoplasm, with few descriptions in the literature. Therefore, its etiology and treatment are not well understood. The present study aims to report the case of an equine histologically diagnosed with lacrimal gland adenocarcinoma in the lower eyelid region and third eyelid treated by surgical excision and intralesional chemotherapy. Case: A 17-year-old male mixed-breed equine weighing 300 kg was treated in the large animal clinic and surgery sector of the Federal University of Santa Catarina in the city of Curitibanos, Santa Catarina, Brazil. The equine revealed an alert and docile temperament without considerable physiological changes in the physical examination. In the evaluation of the right eye, a tumor mass was observed in the region of the lacrimal gland adhered to the third eyelid and lower eyelid, with a light red ulcerated appearance approximately 7 cm in diameter. Therefore, the surgical excision of the adhered tumor mass was performed, with the subsequent intralesional application of 2 mL of Vincristine Sulfate. In the histopathological evaluation following the surgical excision of the tumor mass, yellowish-white fragments of irregular nodules were observed measuring from 3.5×2.0×1.7 cm to 2.0×0.5×0.3 cm, in addition to microscopy focus clusters of weakly basophilic neoplastic glandular cells, some with a randomly arranged lacy aspect, forming disorganized acinar structures and others showed marking islands of the cells organized in a palisade shape by vascular delicate stroma. The neoplastic cells presented anisocytosis, anisokaryosis, prominent nucleoli sometimes binucleated and with basophilic intracytoplasmic secretory material, delimited by scarce connective tissue. In some areas, there were small foci of infiltrated lymphocytes and plasmocytes and areas of necrosis. The mass was surrounded by connective tissue, where red blood vessels were found outside the vessels (hemorrhage), and mitoses were observed 2 per field at high magnification (40x). After the surgical procedure, it was not possible to follow up and reassess of the patient, so there is no clarification on the possibility of tumor recurrence. Discussion: The case reported is uncommon, being only the second report of adenocarcinoma in the equine lacrimal gland. Therefore, there are few descriptions in the literature about its defined etiology and the best treatment method, although, surgical excision is the method of choice because it is a tumor of high recurrence and invasiveness. In this case, we opted for the exeresis of the tumor mass and later application of chemotherapy, a treatment that was relatively effective, to avoid recurrence and the failure of the technique. In these cases, monitoring the animal after the procedure is recommended, evaluating whether there was tumor recurrence, although this was not possible in the present possible. The third eyelid, lacrimal, and zygomatic salivary glands may be sources of intraorbital neoplasms. These neoplastic processes may have similar histological and behavioral characteristics, and their differentiation is, therefore, problematic. These structures may be differentiated based on the anatomical location, however, clinical appearance and symptoms are identical in most cases. Adenocarcinoma in the equine lacrimal gland is a neoplasm considered rare, so it is important to emphasize an accurate diagnosis through histopathological analyses to differentiate it from other frequent orbital neoplasms in horses, allowing more information about this tumor and establishing different treatment methods.

Background: Sarcoids are the most frequent skin tumors among horses, causing serious lesions due to their different shapes, sizes, degree of invasiveness and distribution on the body. The pathogenesis of sarcoids is multifactorial, with genetic, viral and environmental involvement, making their treatment complex. The aggressiveness and high rate of recurrence of sarcoids makes it difficult to use an effective treatment, which is why there are several therapeutic routes described in the literature. Aiming to describe and expand sarcoid treatments, this paper reports on the use of acyclovir in the treatment of this type of tumor. Cases: Four horses sent to the Large Animal Veterinary Hospital of the Federal University of Lavras - UFLA were diagnosed and treated for sarcoids. Case 1. Lesions on the right ear, region of the masseter muscle of the right side of the face, neck, vulva and medial aspect of the left pelvic limb. Case 2. Lesion in the left groin region. Case 3. Lesions on the face, masseter muscle region on the left side, mandibular region and right ear pinna. Case 4. Lesion in the lateral region of the left pelvic limb, close to the tarsometatarsal joint. All horses had a diagnosis of sarcoid, which was confirmed by histopathological examination of material collected after surgical excision. Macroscopically, the neoplastic lesions were classified as fibroblastic, verrucous and nodular. The tumors exhibited irregular surfaces, keratinization, and a firm consistency. Their surfaces were alopecic, slightly rough, some of them were ulcerated, and their color ranged from greyish to rosy and reddish. All the tumor masses were surgically excised from the 4 horses, and one sarcoid was treated by thermal cauterization with liquid nitrogen due to the lesion's depth and size. In most cases, the sarcoid removal sites were closed with sutures. Only 2 lesions were not sutured due to the impossibility of bringing the edges of tissue close together, or due to proximity to the tarsometatarsal joint. The surgical wounds were cleaned twice a day with sterile saline solution followed by the topical application of Acyclovir. One of the animals received complementary therapy with Cimetidine. Discussion: The equines recovered completely from their sarcoid treatment, and no recurrences were reported a year later. Thus, the combination of treatments employed for the extirpation of sarcoids proved to have greater chances of success. First, all the tumors were surgically removed with the largest possible margins of safety in order to ensure the elimination of neoplastic cells. In the postoperative period, all the animals received topical treatment of the lesions with acyclovir after surgical resection of the tumors. The drug aided the complete healing of post-surgical wounds, and healing time varied according to the size and depth of the lesion. One tumor was treated with liquid nitrogen after surgical excision of the sarcoid. Another horse was treated with cimetidine over a 3-month period after surgical excision of the neoplasm in order to reduce the sarcoid and prevent its evolution. Surgical excision of the sarcoid associated with topical application of acyclovir ointment showed satisfactory results. Moreover, the combination of surgical excision and administration of liquid nitrogen on the lesion and topical application of acyclovir ointment in the postoperative period also provided good results. Given the complexity of sarcoid treatment and the high recurrence rate of these tumors, the use of combined treatments should be taken into account.

ABSTRACT: The aim of this study was to verify the applicability and accuracy of B-mode ultrasonography in detecting malignancy in dog cutaneous neoplasms. Forty-two neoplasms (12 benign and 30 malignant) of mesenchymal, round cells, epithelial and melanocytic origins from 24 dogs of different breeds and ages were included. The ultrasound evaluation was performed with a linear multi-frequency transducer (7.0 to 12MHz), with frequency dependent on the mass dimension. Ultrasonographic characteristics of echogenicity (hypo/hyperechogenic), echotexture (homogeneous/heterogeneous), regularity, invasiveness in adjacent tissues were classified. Dimensions were also measured to calculate the depth/width ratio. Neoplasms were classified as malignant or benign after cytological and/or histopathological analysis and the results were associated with ultrasound characteristics. Resumo

O lipoma infiltrativo é uma neoplasia benigna rara que acomete os adipócitos. Apresenta etiologia desconhecida, sem raça predisponente. É altamente agressivo, não metastático e com índices de recidivas significativos. Um cão da raça Shih Tzu, macho, castrado e com 7 anos e 7 meses foi atendido na clínica veterinária da Universidade Salvador, com queixa de tumoração em membro torácico esquerdo, presente já há 5 anos. Exames de imagem, histopatológico e cirurgia confirmaram o diagnóstico de lipoma infiltrativo em tórax e membro torácico esquerdo. O tratamento preconizado foi amputação do membro devido a magnitude da infiltração. Para o pós-operatório foi realizada fisioterapia e acompanhamento. A recidiva, em região dorsal, se deu após 82 dias de cirurgia e novamente se optou pelo procedimento cirúrgico.


Infiltrative lipoma is a rare benign neoplasm that affects adipocytes. It has unknown etiology, with no predisposing race. It is highly aggressive, non-metastatic and with significant relapse rates. A Shih Tzu dog, male, neutered, 7 years and 7 months old, was seen at the veterinary clinic of Salvador University, complaining of tumor in the left thoracic limb. Imaging, surgery and histopathological exams to confirm the diagnosis of infiltrative lipoma in the chest and left thoracic limb presente for 5 years. The recommended treatment was amputation of the limb due to the magnitude of the infiltration. In the postoperative period, physiotherapy and follow-up were performed, recurrence in the dorsal region occurred 82 days after the surgery and the surgical procedure was again chosen.

Background: Acute lymphoblastic leukemia (ALL) is a malignant neoplasia in which there is proliferation of lymphoid progenitor cells in the bone marrow, blood, and extramedullary sites. This disorder has a fast and progressive development; in dogs, cases of infiltration of ALL cells in the central nervous system (CNS) are uncommon and rare. Diagnosis can be achieved with the help of the clinical history and physical, radiographic, hematological, myelographic, and cerebrospinal fluid (CSF) tests in patients with or without neurological clinical signs. The present report aims to describe a case of ALL and the presence of lymphoblasts in the CSF of a dog with neurological clinical signs. Case: An 8-year-old Lhasa Apso dog was examined at the Veterinary Hospital of Universidade Federal do Paraná, Curitiba campus. At the physical examination, the animal exhibited apathy and paralysis of pelvic limbs, which progressed to tetraplegia. Abdominal palpation revealed presence of hepatosplenomegaly and absence of lymphadenomegaly. No alterations were observed in radiographs of the cervical, thoracic or lumbar spine. A complete blood count revealed presence of non-regenerative anemia (hematocrit = 22%), extreme lymphocytosis (185,229 cells/µL), lymphoblasts at a level of 72% (133,364 cells/µL), and thrombocytopenia (66,000 platelets/µL). The biochemical tests revealed increased alkaline phosphatase (859 IU/L). The levels of alanine aminotransferase, creatinine, urea, total protein, albumin, and globulin were normal. The diagnosis of ALL was achieved with the help of a myelogram. The myelogram findings included 39% of mature lymphocytes and 59% of lymphoblasts exhibiting large size, spherical shape, poorly delimited borders, with a high nucleus/cytoplasm ratio, marked cytoplasmic basophilia, and 2 to 3 evident nucleoli; metarubricytes (1%) and promyelocytes (0.6%) were also observed. The CSF contained an increased number of nucleated cells (27 cells/µL) comprising lymphocytes (43%), macrophages (33%), and segmented neutrophils (24%). Of the 11.6 lymphocytes per µL of CSF, 8.1 were lymphoblasts, which indicates infiltration of ALL cells in the CNS. The animal died one day after collection of bone barrow and CSF. Discussion: Relevant alterations observed in this case included the neurological signs caused by the infiltration of neoplastic cells in the CNS, severe leukocytosis and lymphocytosis, with large amounts of lymphoblasts in the blood and predominance of lymphoblasts in the bone marrow, which are alterations typically found in ALL. The animal also exhibited non-regenerative anemia and thrombocytopenia, which were secondary to infiltration of leukemic cells in the bone marrow. The CSF exhibited pleocytosis (27 cells/ µL), and 30% of the cells observed were lymphoblasts. Lymphoblast infiltration in the CNS of leukemic dogs is rare, and other studies have reported absence of neurological signs or neurological signs different from those observed in the present study. CSF analysis in indicated in cases of leukemia to assess leukemic cell infiltration in the CNS. In the case reported here, the plasma level of alkaline phosphatase was increased (859 IU/L) as a consequence of hepatomegaly and hepatic cholestasis. ALL is a very aggressive, proliferative neoplasia, and the resulting lymphoblasts infiltrated the CNS of the animal. In cases of ALL, performing complete blood count, myelogram, and CSF analysis is indicated whether the patients exhibit neurological signs or not.(AU)

Videolaparoscopic procedures have gained prominence due to their low invasiveness, causing less surgical trauma and better post-surgical recovery. However, the increase in intra-abdominal pressure due to the institution of pneumoperitoneum can alter the patient's homeostasis. Therefore, volume-controlled ventilation, associated with positive end-expiratory pressure (PEEP), improves arterial oxygenation and prevents pulmonary collapse, but it can lead to important hemodynamic changes. The aim of this study was to evaluate, comparatively, the effects of positive end expiratorypressure (PEEP) on hemodynamic variables of pigs submitted to volume-controlled ventilation, during pneumoperitoneum and maintained in head-down tilt and determine which PEEP value promotes greater stability on hemodynamic variables. Twenty-four pigs were used, between 55 and 65-day-old, weighing between 15 and 25 kg, randomly divided into 3 distinct groups differentiated by positive end-expiratory pressure: PEEP 0 (volume-controlled ventilation and PEEP of 0 cmH2O), PEEP 5 (volume-controlled ventilation and PEEP of 5 cmH2O) and PEEP 10 (volume-controlled ventilation and PEEP of 10 cmH2O). Volume-controlled ventilation was adjusted to 8 mL/kg of tidal volume and a respiratory rate of 25 movements per min. Anesthesia was maintained with continuous infusion of propofol (0.2 mg/kg/min) and midazolam (1 mg/kg/h). Pneumoperitoneum was performed with carbon dioxide (CO2), keeping the intraabdominal pressure at 15 mmHg and the animals were positioned on a 30° head-down tilt. The evaluations of hemodynamic variables started 30 min after induction of anesthesia (M0), followed by measurements at 15-min intervals (from M15 to M90), completing a total of 7 evaluations. The variables of interest were collected over 90 min and submitted to analysis of variance followed by Tukey's post-hoc test, with P < 0.05. The PEEP 10 group had higher values of CVP and mCPP, while the PEEP 5 group, mPAP and PVR were higher. The PEEP 0 group, on the other hand, had higher means of CI. Regarding the moments, there were differences in HR, SAP, DAP, MAP, CO, IC and TPR. According to the literature, important hemodynamic effects due to pneumoperitoneum are reported, which can be caused by the pressure used in abdominal insufflation, CO2 accumulation, duration of the surgical procedure, hydration status and patient positioning. Mechanical ventilation associated with PEEP can also cause an increase in intrathoracic pressure and, therefore, reduce cardiac output. Cardiovascular changes are proportional to the PEEP used. Central venous pressure (PVC) measure the patient's preload, and intrathoracic pressure can interfere with this parameter. The peak pressure values in the PEEP 10 group were higher than the other groups, demonstrating that the increase in intrathoracic pressure results in higher PVC values. Regarding PAPm and PCPm, these variables can be influenced according to the PEEP values and the patient's position. In relation to CI, the increase in PEEP may reflect on intrathoracic pressure, resulting in greater compression of the heart, with a consequent reduction in cardiac output and cardiac index. Therefore, it is concluded that the PEEP effects of 0 cmH2O and 5 cmH2O on hemodynamics are discrete, under the proposed conditions.(AU)

ABSTRACT: Extraintestinal pathogenic Escherichia coli (ExPEC) is a highly diverse pathotype of E. coli which colonizes the intestine, and it is considered an important etiological agent associated with bacteremia and other systemic infections, among them urinary tract infection. Retrospective studies evaluating morbidity and mortality of nondomestic felids have demonstrated that urinary tract diseases are among the main causes of death for geriatric animals. Also, mesenchymal neoplasms of the uterus are common in wild felids, and they possess variable morphologic characteristics related to invasiveness and malignancy. This report describes a case of bilateral pyelonephritis due to extraintestinal uropathogenic E. coli infection in a captive jaguar (Panthera onca). The diagnosis was confirmed through pathological, bacterial and immunohistochemical findings. According to molecular analysis, this E. coli strain was classified in the phylogroup F, possessing the following virulence-associated genes: usp, cnf-1, hlyA, papC and sfa. Additionally, this E. coli was highly resistant to -lactams and first-generation cephalosporin. This jaguar also presented a uterine leiomyoma with distinct distribution, and severe degenerative articular disease, both of them described as frequently seen lesions in geriatric animals from the Panthera genus.


RESUMO: Escherichia coli extraintestinal patogênica (ExPEC) é um patotipo altamente diverso de E. coli que coloniza o intestino e é considerada um agente etiológico importante, associado com bacteremia e outras infecções sistêmicas, dentre elas infecções do trato urinário. Estudos retrospectivos avaliando morbidade e mortalidade de felídeos não domésticos demostram que doenças do trato urinário estão entre as principais causas de morte de animais geriátricos. Ainda, neoplasias mesenquimais uterinas são comuns em felídeos de cativeiro e possuem características morfológicas variáveis relacionadas a invasividade e malignidade. Neste relato é descrito um caso de pielonefrite bilateral por E. coli extraintestinal uropatogênica em uma onça-pintada de cativeiro (Panthera onca). O diagnóstico foi confirmado através dos achados patológicos, bacteriológicos e imuno-histoquímicos. A partir da análise molecular, esta cepa de E. coli foi classificada no filogrupo F, possuindo os seguintes genes associados a virulência: usp, cnf-1, hlyA, papC and sfa. Adicionalmente, a bactéria isolada foi altamente resistente a -lactâmicos e cefalosporinas de primeira geração. Foi observado ainda um leiomioma uterino com distribuição distinta e doença articular degenerativa severa, ambas descritas na literatura como comumente observadas em animais geriátricos do gênero Panthera.

Paragangliomas are tumors originating from paraganglia cells which represent a considerable constituent of the dispersed neuroendocrine system. Rarely these tumors are found within the orbital region. This report documents a case of primary extra-adrenal paraganglioma in a 15-year-old female quarter horse, diagnosed on the basis of clinical signs, gross anatomopathological, histopathological and immunohistochemical investigations. Clinical diagnostics led to full appreciation of the severe tumor invasiveness and to an informed decision by the owner for euthanasia. Gross inspection confirmed the previous clinical assessments of exophthalmos consequential to pressure from the orbital neoplasia, revealing a mass infiltrating into surrounding tissues. Histopathology and immunohistochemistry allowed for the final diagnosis of paraganglioma. Histology sections showed a partially encapsulated infiltrative mass with multifocal angiotropic growth, and composed of polygonal cells organized in nests, packets, and bundles supported by a fibrovascular stroma. The lobules were lined peripherally by spindle cells. The tumor was highly vascular with blood-filled lacunae and multifocal to coalescing areas of liquefactive necrosis. The polygonal cells were characterized by indistinct borders, lightly eosinophilic cytoplasm with moderate amount of granules, round or oval central nuclei with stippled chromatin (“salt and pepper” appearance) and inconspicuous nucleoli. Immunohistochemistry aided in the diagnostic discrimination of paraganglioma from neuroendocrine carcinoma and in the assessment of tumor differentiation. Orbital paraganglioma holds low likelihood for animal survival, but may have good prognosis when timely addressed.

Resumo

Paragangliomas are tumors originating from paraganglia cells which represent a considerable constituent of the dispersed neuroendocrine system. Rarely these tumors are found within the orbital region. This report documents a case of primary extra-adrenal paraganglioma in a 15-year-old female quarter horse, diagnosed on the basis of clinical signs, gross anatomopathological, histopathological and immunohistochemical investigations. Clinical diagnostics led to full appreciation of the severe tumor invasiveness and to an informed decision by the owner for euthanasia. Gross inspection confirmed the previous clinical assessments of exophthalmos consequential to pressure from the orbital neoplasia, revealing a mass infiltrating into surrounding tissues. Histopathology and immunohistochemistry allowed for the final diagnosis of paraganglioma. Histology sections showed a partially encapsulated infiltrative mass with multifocal angiotropic growth, and composed of polygonal cells organized in nests, packets, and bundles supported by a fibrovascular stroma. The lobules were lined peripherally by spindle cells. The tumor was highly vascular with blood-filled lacunae and multifocal to coalescing areas of liquefactive necrosis. The polygonal cells were characterized by indistinct borders, lightly eosinophilic cytoplasm with moderate amount of granules, round or oval central nuclei with stippled chromatin (“salt and pepper” appearance) and inconspicuous nucleoli. Immunohistochemistry aided in the diagnostic discrimination of paraganglioma from neuroendocrine carcinoma and in the assessment of tumor differentiation. Orbital paraganglioma holds low likelihood for animal survival, but may have good prognosis when timely addressed.(AU)

Background: Anaplastic carcinoma of the mammary gland is considered a highly invasive and malignant neoplasia, whichusually leads to neoplastic thrombosis and epidermal ulceration due to invasiveness of lymphatic vessels by tumor emboli.Loss of epidermal barrier predisposes to secondary bacterial infection, mainly by commensal bacteria of the skin, andsepticemia can be observed in cases of systemic dissemination. Neoplastic emboli and septic thrombi can lead to vascularobstruction, which can predispose to infarction in multiple organs. The aim of this case report is to describe a case ofhemorrhagic stroke in a dog with anaplastic carcinoma of the mammary gland.Case: A 12-year-old, mixed breed, female spayed canine presented with mammary tumor with plaque formation, associatedwith skin ulceration which extended to the medial region of pelvic limbs. The animal also displayed neurologic clinicalsigns characterized by stupor, opisthotonos and apathy. Due to poor prognosis, the canine was euthanized and submittedto necropsy. During external examination, the animal was in a good body condition, had moderately pale mucosae, andit was noted a nodular plaque in the abdominal ventral area extending from thoracic to inguinal mammary gland. Themass was firm and white, extensively ulcerated, and extending from the thoracic to inguinal mammary gland. There wasa well circumscribed area of infarction in the spleen and kidney. In the cerebral cortex it was observed an extensive areaof hemorrhage with adhered blood clot from frontal to temporal lobe. There were multiple well circumscribed, firm, andwhite areas in the myocardium. Histologically, the neoplastic proliferation was composed by epithelial cells with high pleomorphism, features of malignancy, and no delimitations, leading to the diagnosis of anaplastic carcinoma of the mammarygland. Resumo

Previous studies have evaluated the effects of different reproductive procedures on discomfort markers in sheep and cattle. Such studies may help stimulate the adoption of techniques that are more beneficial for animal welfare. However, markers that are commonly used to evaluate discomfort are highly influenced by external factors. To overcome this, several systemic markers can be evaluated to more precisely identify stress, pain, and inflammation. Such markers include cortisol, acute phase proteins, bradykinin, and substance P. We aimed to review the potential markers of stress, pain, and inflammation, and discuss how and when they are regulated after different stimuli related to reproductive procedures in cattle and sheep. Furthermore, we aimed to review how reproductive procedures with different degrees of invasiveness cause stress and provide information that may help develop strategies to limit animal discomfort.(AU)


Estudos anteriores avaliaram o efeito de diferentes procedimentos reprodutivos sobre marcadores de desconforto em bovinos e ovinos. Tais estudos podem estimular a adoção de técnicas que preservem o bem-estar animal. Entretanto, os marcadores comumente utilizados apresentam alta influência de fatores externos. Para contornar isso, a avaliação conjunta de diferentes parâmetros sistêmicos pode ser utilizada para determinar com maior precisão a presença de estresse, dor ou inflamação, como cortisol, proteínas de fase aguda, a bradicinina e a substância P. O objetivo desta revisão é relacionar potenciais marcadores de inflamação e estresse, discutindo como e quando são regulados frente aos estímulos em bovinos e ovinos. Ainda, pretende-se revisar de que forma procedimentos reprodutivos com diferentes graus de invasividade acarretam em desconforto, fornecendo informações para a elaboração de estratégias que possibilitem minimizá-lo.(AU)
