Nutritional Secondary Hyperparathyroidism (NSH) is a metabolic disorder that mainly affects young animals. This disease causes imbalance between the levels of calcium and phosphorus, culminating in bone changes. Wild animals raised in captivity can develop the disease when they are given an inadequate diet. The aim of this study is to report the clinical findings, the complementary exams and the therapeutics utilized in a Leopardus geoffroyi (Geoffroy's cat) cub originated from captivity with NSH. The animal went through complementary imaging exams and biochemical evaluation that respectively revealed generalized osteopenia and pathological fracture of the right femur, reduction of serum calcium level and elevation of serum phosphorus level. The diagnosis of NSH was established by associating the history, clinical signs, and findings of ancillary exams. The treatment was based on diet correction, mineral and vitaminic supplementation and sun exposure. The NSH diagnosis was stablished by associating history, clinical signs, imaging exams findings and serum biochemicals. The early identification of this disease is of paramount importance, because in this way corrections can be made in food management, in order to provide successful clinical recovery and promote quality of life for wild captive animals.(AU)
O hiperparatireoidismo secundário nutricional (HSN) é uma desordem metabólica que acomete principalmente animais jovens. Essa doença cursa com desequilíbrio entre os níveis de cálcio e fósforo, culminando com alterações ósseas. Animais silvestres quando criados em cativeiro podem desenvolver a doença quando recebem uma dieta inadequada. O obje-tivo deste estudo é relatar os achados clínicos, os exames complementares e a terapêutica utilizada em um filhote de Leopardus geoffroyi (gato-do-mato-grande)proveniente de cativeiro com HSN. O animal passou por exames complementares de imagem e avaliação bioquímica que revelaram respectivamente osteopenia generalizada e fratura patológica de fêmur direito, redução dos níveis séricos de cálcio e elevação do fósforo. O diagnóstico de HSN foi estabelecido associando o histórico, sinais clínicos e achados dos exames complementares. O tratamento foi baseado na correção da dieta, suplementação mineral e vitamínica e exposição solar. A identificação precoce dessa doença é de suma importância, pois dessa forma podem ser feitas correções no manejo alimentar, afim de proporcionar a recuperação clínica com êxito e promover qualidade de vida para os animais selva-gens de cativeiro.(AU)

Immunomediated thrombocytopenia is a systemic metabolic disorder in which the platelet count falls below reference values, as the patient's immune system destroys them. The main clinical signs in thrombocytopenia are petechial, hemorrhages, ecchymoses and suffusions. Hematomas can also occur in coagulation disorders. The diagnosis is based on clinical findings and hematological examinations. The treatment consists of the use of corticosteroids and immunosuppressants, delaying cell destruction, and may last for months, not always obtaining a cure for the disease. The present work reports the use of therapy with allogeneic mesenchymal stem cells, derived from the adipose tissue of dogs, for the treatment of chronic immunomediated thrombocytopenia, with an evolution of more than one year, in a Pinscher dog. The alternative treatment showed a good evolution, keeping platelets within the reference values during the treatment, giving the patient quality of life and removing the need for continuous medication for homeostasis after treatment.
A trombocitopenia imunomediada é uma desordem metabólica sistêmica, na qual a contagem plaquetária fica abaixo dos valores de referência, pois o sistema imunológico do paciente a destrói. O principal sinal clínico na trombocitopenia são hemorragias, petequiais, equimoses e sufusões. Hematomas podem ocorrer também em alterações da coagulação. O diagnóstico baseia-se nos achados clínicos e nos exames hematológicos. O tratamento consiste na utilização de corticosteroides e imunossupressores, o que retarda a destruição celular, mas pode se prolongar por meses, nem sempre obtendo cura da doença. O presente trabalho relata a utilização da terapia com células-tronco mesenquimais alogênicas, oriundas do tecido adiposo de cães, para tratamento de trombocitopenia imunomediada crônica, com evolução de mais de um ano, em um cão da raça Pinscher. O tratamento alternativo revelou boa evolução, pois manteve as plaquetas dentro dos valores de referência durante o tratamento, o que proporcionou qualidade de vida ao paciente e tornou desnecessárias medicações de uso contínuo para a homeostase após o tratamento.

In order to establish a bone loss induction model in rabbits to study osteoporosis, 18 rabbits underwent ovariectomy and received methylprednisolone 1mg/kg intramuscularly on alternate days for two weeks. Immediately before ovariectomy up to 42 days after methylprednisolone administration, radiographs of the right olecranon were taken in the mediolateral position to evaluate the radiographic optical density. Before the induction of osteoporosis, rabbits presented mean values of radiographic density of 2.305mm Al and at 42 days of 1.575mm Al. The values obtained were submitted to ANOVA for repeated measures that revealed a significant drop (P< 0.001) in the density over time, proving that the induction was able to trigger bone loss of these animals. With this, it can be affirmed that the adopted protocol was enough to provoke a significant bone loss, characterizing a valid model for the study of new treatments for osteoporosis.(AU)

Background: Calcium is essential for the healthy development of animals, especially regarding bone formation. Diversefactors influence calcium metabolism, and failure in any of these points may result in metabolic bone diseases. Ricketsis an uncommon disease of growing bones occurring exclusively in young animals, it is characterized by the failure ofosteoid to calcify and can be a result of lack of vitamin D, imbalance of calcium and phosphorus or hereditary conditions.Nutritional origin is more frequent in veterinary medicine, but it has become rarer after the availability of balanced commercial rations. This report describes the diagnosis and treatment of rickets in a crab-eating fox puppy.Case: A 3-month-old male crab-eating fox was presented with ataxia and angular deviation of the tibia and radius/ulnawith suspected osteometabolic disease. It was part of a group of three orphan siblings hand-raised. The other two siblingsdid not present any clinical signs. All three animals presented pale mucous membranes and were infected by ancylostoma,coccidia, toxocara, and sarcocystis. Radiographic findings of the affected animal showed generalized osteopenia of theentire skeleton associated with widening radiolucent distal physeal plates and cupping of the metaphysis, changes classical of rickets. The other two siblings presented no skeletal alterations and complete blood count and biochemistry of allthree animals were taken for comparison, including serum vitamin D and PTH. Background: Rickets is a deficiency pathology that occurs in young and growing animals, leading to deficient bone mineralization. Rickets has been reported in several species producing numerous economic losses. The disease is caused by nutritional imbalance of calcium (Ca), phosphorus (P) and vitamin D. The aim of this work was to report two outbreaks of rickets in commercial weaning farms in the Seara city, in the western region of Santa Catarina State (SC), Brazil. Case: In August 2016, the Veterinary Pathology Laboratory (LPV) at the Concórdia Campus of the Catarinense Federal Institute (IFC) diagnosed two outbreaks of rickets in pigs in the weaning phase in Seara, SC. The clinical history was obtained by interviewing the field veterinarian and the farmers. In the anamnesis, both pig farmers stated having used a feed premix product from the same company, starting three months ago, and after that the pigs presented clinical signs of posterior paralysis and progressive weight loss. Six animals were submitted to necropsy and organs were collected from the abdominal and thoracic cavities; central nervous system and bones; fixed in 10% buffered formalin, routinely processed, paraffin embedded and stained with hematoxylin and eosin (HE) for histopathological analysis. Bone specimens were decalcified in nitric acid working solution 20 times their volume, during 5 days. In addition, samples of the premix product containing minerals and vitamins were sent to a specialized laboratory to analyze macroelements levels through the atomic absorption methodology. Two young opossums were necropsied and diagnosed with rickets. This study aims to describe the clinical-pathological aspects of rickets in Didelphis albiventris. Macroscopically, the opossums presented kyphosis and scoliosis, lateral deviation of the limbs in varus, locomotion difficulty, and enlargement with softening of costochondral junctions (rickety rosary). Samples of bones and joints were processed for hematoxylin and eosin staining and Masson's trichrome. Microscopically, we observed thickening of the epiphyseal plate, characterized by irregular and multifocal proliferation of serialized and hypertrophic cartilage zones, which formed circular groups of large, dysplastic chondrocytes towards the spongy zone, often surrounded by non-mineralized osteoid tissue. In the cortical bone, there were pale eosinophilic zones around the Havers channels consistent with non-mineralized osteoid. The staining of Masson's trichrome evidenced the accumulation of osteoid tissue in cortical and trabecular bones. It is possible that a mixed cause of absorption deficiency of vitamin D3 associated with an unbalanced Ca:P diet based on lactose-free milk and fruits may have triggered the disease.(AU)

The amount of research on metabolic bone diseases in commercial birds is considerable, however, there is a large gap in the knowledge of these diseases in wild birds. Aspects related to diet are the main factors that cause these diseases, such as: vitamin D deficiency, insufficient calcium intake, or imbalance of calcium and phosphorus in the diet, and problems in the absorption and metabolism of these compounds. This study reports two cases of bone diseases in birds of the order Accipitriformes, which are wild in the state of Paraíba, a crab-eating hawk with rickets, and a caboclo hawk with fibrous osteodystrophy. The diagnosis in both cases was based on macroscopic and microscopic findings. These reports are subsidies for the bird clinic of the order Accipitriformes and for the maintenance of these birds in nurseries and zoos. This study demonstrates the need for further studies to assess whether such cases can be used as indicators of environmental imbalance.

ABSTRACT: Computerized microtomography is the gold standard examination for the evaluation of the three-dimensional bone structure. This experiment was developed to evaluate the structure and bone quality of Caiman yacare with metabolic bone disease using high resolution computerized microtomography (CT). The animals were distributed into four groups: G1 - hyperphosphatemic diet with sun exposure deprivation (n=4), G2 - hyperphosphatemic diet with sun exposure (n=4), G3 - balanced diet with sun exposure deprivation (n=4), and G4 - balanced diet with exposure to sunlight (n=4). The parameters for the trabecular bone (Trabecular Number, Trabecular Thickness, Trabecular Separation, Bone Pattern Factor, Fractal Dimension, Euler Number, Structural Model Index, Degree of Anisotropy, Eigenvalues 1, 2 and 3, and Centroides X, Y and Z), and cortical bone (Number of Closed Pores, Volume of Closed Pores, Surface of Closed Pores, Closed Porosity, Volume of Open Pores, Open Porosity and Total Porosity). The overall results showed that the structure and bone quality of group G3 and G4 were better than those of groups G1 and G2, and that the diet factor influenced more than the sun exposure factor. The computerized microtomography allowed to evaluate the quality of the cortical and trabecular bones of the Pantanal alligator tibia with osteometabolic disease. The diet and sun exposure factors influenced individually the results of the CT parameters between the groups, demonstrating the functional and structural complexity. Thus, these parameters can contribute to the interpretation of the mechanical behavior of bones and correlate them with the risk of lesions and fractures associated with osteometabolic diseases.
RESUMO: Microtomografia computadorizada é o exame padrão-ouro para a avaliação da estrutura tridimensional do osso. Este estudo experimental foi desenvolvido para avaliar a estrutura e a qualidade óssea de jacarés-do-pantanal (Caiman yacare) com doença óssea metabólica utilizando a microtomografia computadorizada (CT) de Alta Resolução. Os animais foram distribuídos em quatro grupos, G1 - dieta hiperfosfatêmica com privação de luz solar (n=4), G2 - dieta hiperfosfatêmica com exposição à luz solar (n=4), G3 - dieta balanceada com privação de luz solar (n=4) e G4 - dieta balanceada com exposição à luz solar (n=4). Avaliaram-se os parâmetros para o osso trabecular (Número de Trabéculas, Espessura Trabecular, Separação Trabecular, Fator do Padrão Ósseo, Dimensão Fractal, Número de Euler, Índice do Modelo Estrutural, Grau de Anisotropia, Autovalores 1, 2 e 3 e Centroides X, Y e Z) e osso cortical (Número de Poros Fechados, Volume dos Poros Fechados, Superfície de Poros Fechados, Porosidade Fechada, Volume de Poros Abertos, Porosidade Aberta e Porosidade Total). Os resultados gerais evidenciaram que a estrutura e a qualidade óssea dos grupos G3 e G4 foram superiores aos dos grupos G1 e G2, sendo que o fator dieta influenciou mais do que o fator exposição solar. A Microtomografia Computadorizada permitiu avaliar a qualidade dos ossos cortical e trabecular da tíbia de jacarés do pantanal com doença osteometabólica. Os fatores dieta e exposição solar influenciaram individualmente no resultado dos parâmetros do CT entre os grupos, demonstrando a complexidade funcional e estrutural. Assim, esses parâmetros podem contribuir na interpretação do comportamento mecânico dos ossos e correlacioná-los com o risco de lesões e fraturas associadas às doenças osteometabólicas.

Computerized microtomography is the gold standard examination for the evaluation of the three-dimensional bone structure. This experiment was developed to evaluate the structure and bone quality of Caiman yacare with metabolic bone disease using high resolution computerized microtomography (μCT). The animals were distributed into four groups: G1 - hyperphosphatemic diet with sun exposure deprivation (n=4), G2 - hyperphosphatemic diet with sun exposure (n=4), G3 - balanced diet with sun exposure deprivation (n=4), and G4 - balanced diet with exposure to sunlight (n=4). The parameters for the trabecular bone (Trabecular Number, Trabecular Thickness, Trabecular Separation, Bone Pattern Factor, Fractal Dimension, Euler Number, Structural Model Index, Degree of Anisotropy, Eigenvalues 1, 2 and 3, and Centroides X, Y and Z), and cortical bone (Number of Closed Pores, Volume of Closed Pores, Surface of Closed Pores, Closed Porosity, Volume of Open Pores, Open Porosity and Total Porosity). The overall results showed that the structure and bone quality of group G3 and G4 were better than those of groups G1 and G2, and that the diet factor influenced more than the sun exposure factor. The computerized microtomography allowed to evaluate the quality of the cortical and trabecular bones of the Pantanal alligator tibia with osteometabolic disease. The diet and sun exposure factors influenced individually the results of the μCT parameters between the groups, demonstrating the functional and structural complexity. Thus, these parameters can contribute to the interpretation of the mechanical behavior of bones and correlate them with the risk of lesions and fractures associated with osteometabolic diseases.(AU)

Computerized microtomography is the gold standard examination for the evaluation of the three-dimensional bone structure. This experiment was developed to evaluate the structure and bone quality of Caiman yacare with metabolic bone disease using high resolution computerized microtomography (μCT). The animals were distributed into four groups: G1 - hyperphosphatemic diet with sun exposure deprivation (n=4), G2 - hyperphosphatemic diet with sun exposure (n=4), G3 - balanced diet with sun exposure deprivation (n=4), and G4 - balanced diet with exposure to sunlight (n=4). Locomotion problems in broiler chickens can cause economic losses of up to 40% of the total revenues due to carcass condemnation and downgrading at processing. Leg disorders, such as femoral lesions, tibial dyschondroplasia, and spondylolisthesis, substantially impact the welfare of chickens as these disorders can prevent birds from reaching the feeders and drinkers, thus reducing feed and water intake. The most important issues related to broiler welfare reported in the last two decades are their growing sensitivity to metabolic and locomotion problems due to the fast growth rates and inactivity. Traditional methods for the determination of gait score include the manual scoring of animal behavior in the broiler house. Recorded video images can also be used for manual scoring of chicken gait score. However, scoring of some animal-based information by human experts and manual methods remain difficult, time consuming and expensive when implemented at farm level. In an effort to objectively detect leg disorders, this study aimed at validating the photogrammetry technique as a non-invasive method for identifying locomotion problems in broilers. Photogrammetry allows determining the geometric properties of broilers from digital photos that are processed and analyzed using a computer software. Results obtained using photogrammetry were tested for their correlation with those obtained by accepted methodologies, including gait score and macroscopic examination of femoral degeneration and tibial dyschondroplasia. The photogrammetry results agreed with the results of the afore mentioned accepted methods.

A osteoporose é uma doença metabólica que acomete os ossos promovendo fragilidade. É um problema mundial de saúde que afeta uma em cada três mulheres e um em cada cinco homens. Os Bifosfonatos são o grupo farmacológico mais utilizado no tratamento desta doença, mas após um longo período de utilização sua ação é revertida, deixando os ossos susceptíveis a fratura. Desta forma faz se necessário pesquisas que elucidem a fragilidade óssea devido a osteoporose bem como após longos tempo de tratamento. Sendo assim, objetivamos desenvolver novas estratégias para estudar ossos osteoporóticos através de modelos de osteoporose em camundongos e ratos, sendo uma nova técnica para avaliar a qualidade óssea em escala nanométrica in vivo, estratégias para realizar análises biomecânicas, um modelo de osteopenia e um modelo experimental de camundongos para estudar o efeito deletério do Bifosfonato a longo prazo. Foram realizados quatro estudos. Primeiro, o osso osteoporótico foi avaliado através da Espectroscopia Raman in vivo aonde os camundongos controle (N = 7) foram comparados com os ovariectomizados (n = 7) para avaliação da qualidade óssea. Segundo, para avaliar a fragilidade óssea através de parâmetros biomecânicos em modelos de osteoporose induzida em roedores utilizou-se um grupo de camundongos controle (n = 7) e outro ovariectomizados (n = 7); e um grupo de ratos controle (n = 5) e ovariectomizados (n = 6) que tiveram os fêmures dissecados após eutanásia para que o colo do fêmur recebesse um teste de carga até a deformação permanente, de maneira fisiológica, restrita ou irrestrita. E a influência do ganho de peso nos resultados foi discutida através de duas maneiras de normalizar os mesmo. Terceiro, foi realizado um modelo de osteopenia por desuso para avaliar como os compartimento da tíbia (cortical, trabécula e geometria) se comportam após 5 (n = 5), 35 (n = 4), 65 (n = 5) e 95 (n = 3) dias de desuso por neurectomia do ciático. Por último, buscando o efeito deletério do Bifosfonato ovariectomizados (n = 7) e ovariectomizados seguida da administração de Ibandronato. Após eutanásia os fêmures foram coletados para testes biomecânicos no colo femoral a as tíbias foram utilizadas para análise de arquitetura, massa e geometria. Conclusões: Primeira, a espectroscopia Raman mostrou uma diminuição na qualidade óssea dos animais osteoporóticos através da diminuição da quantidade de cálcio, glicosaminoglicanos e aumento do conteudo de lipídios. Segunda, é possível realizar testes de carga em fêmures de camundongos e ratos de maneira fisiológica para encontrar a fragilidade promovida pela osteoporose, porem isto depende da sensibilidade do teste (restrita e irrestrita) e é necessária uma normalização dos resultados através da massa corporal. Terceira, os ossos osteopenicos reabsorveram cada compartimento de uma maneira diferente, aonde a trabécula se deteriora primeiro, mas exige mais tempo para atingir um platô do que a cortical. E por último, a ovariectomia combinada com Bifosfonato a longo prazo em um grupo de camundongos controle (n = 7) e outro três grupos camundongos foram formados: controle (n = 7),camundongos promove um padrão diferente de fratura no colo do fêmur e um efeito deletério na geometria da tíbia.
Osteoporosis is a metabolic bone disease that promotes bone weakens. It is a world health problem affecting one in three women and one in five men. Bisphosphonates are the drug most used to treat this disease, although after a long period the action reverses prompting bones to fracture. There is research required to solve this osteoporosis and long-term treatment fragility. On this basis we aim to develop a new strategy to study osteoporotic bones through murine osteoporosis models, in a new technique to evaluate bone quality in nanoscale, strategies to perform biomechanical analyses, an osteopenia model and a mice model to study the deleterious effect of bisphosphonate delivered for long-term. It was performed in four studies. First, a group of sham-operated (N=7) mice was compared to ovariectomized mice (n=7) to evaluate osteoporotic bone through Raman Spectroscopic in vivo. Secondly, to find bone fragility through biomechanical parameters in murine models ex vivo a group of mice sham-operated (n=7) and ovariectomized (n=7); rats sham-operated (n=5) and ovariectomized (n=6) had the femurs loaded to fail in a physiological approach and in a constrained and unconstrained manner. The influence of the weight gain on results were discussed through two ways to normalize it. Third, an osteopenia model through disuse was performed to evaluate how the tibia compartments (cortical, trabecula and shape) behave after 5 (n=5), 35 (n=4), 65 (n=5) and 95 (n=3) days of disuse due to sciatic neurectomy. Finally, seeking for the deleterious effect of Bisphosphonate, three groups of mice were formed: control (n = 7), ovariectomized (n = 7) and ovariectomized add to the administration of Ibandronate. After euthanasia, the femurs were collected for biomechanical tests on the femoral neck and the tibiae were used for analysis of architecture, mass and geometry. In conclusion, first, Raman Spectroscopic showed a diminishment in bone quality of osteoporotic bones as a result of decrease calcium and glycosaminoglycans and increase lipids content. Secondly, it is possible to load to fail murine bones to find osteoporosis weakens, but it depends on test sensibility (constrained and unconstrained) and a normalization of the results through body mass is required. Third, bones with osteopenia reabsorbed each compartment in a different way where trabecula starts first but required more time to reach a plateau than cortical. And finally, ovariectomy combined with bisphosphonate for long term promote a different fracture padrone on femora neck and a deleterious effect on tibia shape.

Este trabalho objetivou verificar se o ultrassom de baixa potência (US) previne a ocorrência de osteopenia em tíbias de ratos sob ausência de carga. Foram utilizados 45 Rattus novergicus albinus, Wistar adultos, machos, distribuídos em cinco grupos iguais: C - animais-controle livres em gaiolas por 21 dias; S - animais suspensos pela cauda por 21 dias; ST - suspensos pela cauda por 21 dias e concomitantemente tratados com US; S→C - suspensos por 21 dias e depois permanecendo livres em gaiolas por mais 21 dias; S→CT - suspensos por 21 dias e depois permanecendo livres em gaiolas por mais 21 dias e concomitantemente tratados com US. O tratamento foi realizado com US de 1,5MHz, ciclo de trabalho 1:4, 30mW/cm², na tíbia direita, por 15 sessões de 20 minutos cada, cinco sessões por semana. Ainda vivos, os animais foram submetidos a exame de densitometria óssea para verificação da densidade mineral óssea (DMO) e do conteúdo mineral ósseo (CMO). Após a eutanásia dos animais, as tíbias foram desarticuladas, dissecadas e submetidas a ensaio mecânico destrutivo para análise da força máxima (Fmáx) e da rigidez (R). Foram avaliados também o comprimento (L) e o diâmetro (D) no ponto médio da tíbia. O grupo S apresentou valores de DMO, CMO, Fmáx, R, L e D menores em relação ao grupo C, demonstrando que a suspensão pela cauda é prejudicial a estas variáveis. O tratamento dos animais suspensos com o US, grupo ST, elevou os valores de CMO e DMO em relação aos do grupo S, igualando-os aos do grupo C. A Fmáx, R e L do grupo ST aumentou em relação ao grupo S e também em relação ao grupo C (p<0,05). Nenhuma diferença significativa foi encontrada entre as variáveis analisadas para os grupos S→CT e S→C (p>0,05). Os resultados obtidos neste estudo permitem concluir que o US de baixa potência contribuiu na prevenção e reversão da ocorrência da osteopenia nos animais submetidos à suspensão pela cauda, demonstrando que a ausência do estímulo mecânico causada pela impossibilidade da deambulação pode ser minimizada pela ação mecânica deste.(AU)
This work intended to verify if the low power ultrasound (US) prevents the occurrence of osteopenia on the tibia of rats under the absence of load. 45 adult male Wistar Rattus novergicus albinus were used, separated in five equal groups: C - control free animals in cages for 21 days; S - animals suspended by the tail for 21 days; ST - animals suspended by the tail for 21 days and concurrently treated with US; S→C - animals suspended for 21 days and then free in cages for another 21 days; S→CT - animals suspended for 21 days and then free in cages for another 21 days and concurrently treated with US. The treatment was performed with US with 1.5 MHz, work cycle 1:4, 30 mW/cm², on tibia, for 15 sessions of 20 minutes each, 5 sessions per week. While still alive, the animals underwent bone densitometry examination to verify the Bone Mineral Density (BMD) and the Bone Mineral Content (BMC). After the euthanasia of the animals, the tibias were disjointed, dissected and underwent destructive mechanical testing for the analysis of the maximum force (Fmax) and Rigidity (R). The length (L) and the diameter (D) of the middle point of the tibia were also measured. In group S, BMD, BMC, Fmax, R, L and D were smaller than in group C, demonstrating that the suspension by the tail is detrimental to these variables. The treatment of the suspended animals with the US, in group ST, increased the BMD and BMC compared to group S, leveling them to group C. The Fmax, R and L of group ST increased compared to group S and compared to group C as well (p<0.05). No significant difference was found between the measured variables for the groups S→CT and S→C (p>0.05). The results obtained in this study lead to the conclusion that the low power US contributes to the prevention of the occurrence of osteopenia on animals under suspension by the tail, showing that the absence of the mechanical stimulation caused by the impossibility of deambulation can be minimized by its action.(AU)

Introdução: A osteoporose é a doença metabólica do tecido ósseo, caracterizada pela perda gradual de massa óssea e deterioração da microarquitetura do tecido ósseo em humanos, tendo como consequência a fragilidade óssea e risco de fraturas. O comprometimento ósseo acarreta aumento de custo na carga econômica nos sistemas de saúde com consultas médicas, hospitalizações e colocações em lares de idosos relacionados a fraturas osteoporóticas, sendo considerado um grave problema de saúde pública. Objetivo: Esta revisão sistemática com meta-análise examinou ensaios pré-clínicos para elucidar os efeitos dos alimentos prebióticos na osteoporose induzida em ratas ovariectomizadas. Material e métodos: Os dados e artigos em inglês utilizados na meta-análise foram obtidos através de buscas nos bancos eletrônicos de dados científicos disponíveis na internet: Pub Med, ScienceDirect, Scielo , LILACS, Cochrane e Bireme publicados antes de novembro de 2018. Encontrado 813 artigos completos, resumos ou capítulos de livros que citavam as palavras chaves utilizadas na pesquisa bibliográfica, após triagem de títulos e resumos, seis estudos atendiam os critérios de inclusão. Ao todo foram utilizados 116 animais, divididos randomicamente entre grupos controle e tratamento. Foram selecionados trabalhos completos de ensaios experimentais e que atendiam os seguintes critérios: (i) População: murinos (ratas/camundongos) ovariectomizadas; (ii) Intervenção: suplementação na dieta com prebióticos; (iii) Controle: avaliação dos efeitos nos grupos suplementados e não suplementados; (iv) Desfechos: avaliação de parâmetros indicadores de osteoporose. Usamos o software Review Mananger 5 para a realização dos cálculos da meta-análise e avaliação do risco de viés. Resultados: Os dados da densidade mineral óssea (DMO), conteúdo mineral ósseo (CMO) e biomecânica óssea dos seis estudos incluídos demonstraram que os prebióticos são capazes de melhorar significativamente estes parâmetros nos animais estudados. Conclusão: Esta meta-análise forneceu evidências de que a atividade prebiótica esta envolvida nos mecanismos anti-osteoporóticos e sugere que novos estudos em humanos devam ser realizados usando prebióticos como uma alternativa terapêutica ou terapia complementar aos tratamentos convencionais da osteoporose pós-menopausa em humanos.
Introduction: Osteoporosis is a metabolic disease of bone tissue, characterized by the gradual loss of bone mass and the microarchitectural deterioration of bone tissue in men, resulting in bone fragility and a risk of fractures. Bone impairment leads to an increase in the cost of health services with medical consultations, hospitalizations and hopices related to osteoporotic fractures, being a serious public health problem. Objective: This systematic review with meta-analysis aimed to elucidate the effects of prebiotics on induced osteoporosis in ovariectomized rats. Material and methods: The data and methods used in the statistical analysis were obtained through searches in the scientific databases available on the Internet: "Pub Med", "ScienceDirect", "SciELO", "LILACS", "Cochrane" and "bireme "2018 before. found 813 complete articles, abstracts or book chapters that cited the key words in the bibliographic research, after screening of titles and abstracts, six studies met the inclusion criteria, in total 116 animals were used, divided randomly between control groups and treatment. The following experimental works were selected: (i) Population: murine (rats / mice) ovariectomized; (ii) Intervention: dietary supplementation with prebiotics; (iii) Control: dosage of supplements and not supplemented; (iv) Outcomes: evaluation of osteoporosis indicators. We used the Risk Analysis software 5 to measure the meta-analysis calculations and bias risk assessment. Results: Bone mineral density (BMD), bone mineral content (BMD) and bone biomechanical data from the six included studies demonstrated that serum levels are able to register from them. Conclusion: This meta-analysis provided evidence that prebiotic activity is involved in anti-osteoporotic mechanisms and suggests that further studies in humans should be performed using prebiotics as a therapeutic alternative or therapy complementary to conventional postmenopausal osteoporosis treatments in humans.
