Resumo

Thai Mucuna pruriens (L.) DC. var pruriens (T-MP) seed containing levodopa (L-DOPA) and antioxidant capacity has been shown to improve sexual behavior and male reproductive parameters in rats treated with ethanol (Eth). However, its protective effect on testicular apoptotic germ cells has never been reported. This study aimed to investigate the potential effects of T-MP seed extract on expressions of caspase, proliferating cell nuclear antigen (PCNA), and dopamine D2 receptor (D2R) proteins in Eth rats. Thirty-six male Wistar rats were divided into four groups (9 animals/group), including control, Eth, T-MP150+Eth, and T-MP300+Eth, respectively. Control rats received distilled water, and Eth rats received Eth (3g/kg BW; 40%v/v). The T-MP groups were treated with T-MP seed extract at a dose of 150 or 300 mg/kg before Eth administration for 56 consecutive days. The results showed that the seminiferous tubule diameter and epithelial height were significantly increased in both T-MP treated groups compared to the Eth group. Additionally, the caspase-9 and -3, and PCNA expressions were decreased, but D2R expression was markedly increased in T-MP groups. It was concluded that T-MP seed extract could protect testicular apoptosis induced by Eth via changes in caspase, PCNA, and D2R protein expressions.

DC. var pruriens (T-MP) contendo levodopa (L-DOPA) e capacidade antioxidante demonstrou melhorar o comportamento sexual e os parâmetros reprodutivos masculinos em ratos tratados com etanol (Eth). No entanto, seu efeito protetor sobre células germinativas apoptóticas testiculares nunca foi relatado. Este estudo teve como objetivo investigar os efeitos potenciais do extrato de semente de T-MP na expressão de proteínas de caspase, antígeno nuclear de proliferação celular (PCNA) e receptor de dopamina D2 (D2R) em ratos Eth. Trinta e seis ratos Wistar machos foram divididos em quatro grupos (9 animais/grupo), incluindo controle, Eth, T-MP150+Eth e T-MP300+Eth, respectivamente. Ratos controle receberam água destilada e ratos Eth receberam Eth (3g/kg PC; 40%v/v). Os grupos T-MP foram tratados com extrato de semente de T-MP na dose de 150 ou 300 mg/kg antes da administração de Eth por 56 dias consecutivos. Os resultados mostraram que o diâmetro dos túbulos seminíferos e a altura epitelial foram significativamente aumentados em ambos os grupos tratados com T-MP em comparação com o grupo Eth. Além disso, as expressões de caspase-9 e -3 e de PCNA diminuíram, mas a expressão de D2R aumentou acentuadamente nos grupos T-MP. Concluiu-se que o extrato de semente de T-MP pode proteger a apoptose testicular induzida por Eth através de alterações na expressão de proteínas caspase, PCNA e D2R.

Intramuscular fat (IMF) content is a crucial parameter for estimating meat quality. Growing evidence indicates that gene regulation plays an important role in IMF deposition. This study aimed to determine the function of Mfsd2a in chicken intramuscular preadipocytes. In the present study, high Mfsd2a mRNA levels were observed in the liver and adipose tissues of broilers. Subsequently, we synthesized small interfering RNAs to silence the expression of Mfsd2a in chicken intramuscular preadipocytes. The following results suggested that CDK2, PCNA, CCND1, CCND2 and MKI67 were inhibited, with CCK-8 and EdU assays revealing that cell proliferation was inhibited. Scratch test showed that cell migration ratios were declined. We also found that Mfsd2a silencing decreased the mRNA levels of PPARγ, RXRG and their target genes. The similar results were found in some key genes that contribute to lipid synthesis, including C/EBPα, C/EBPß, FABP4, FASN, ACACA and ACSL1. Finally, Oil red O staining showed that IMF accumulation was blocked after Mfsd2a silencing. In conclusion, our results implied that Mfsd2a promotes the proliferation and migration of chicken intramuscular preadipocytes, as well as the differentiation and adipogenesis through PPARγ signaling pathway, which may provide a potential target to improve chicken meat quality.(AU)

Resumo

Bone marrow-derived mesenchymal stromal cells (BMSCs) have been used for treating inflammatory disorders. Due to the large size of BMSCs compared to nanoparticles, BMSCs cannot be loaded into the nanoparticles. It is hypothesized that BMSCs lysate loading into the nanocarriers will effectively deliver cellular contents and regulatory elements of BMSCs at the injury site. This study aimed to investigate nanostructured lipid carriers (NLC) loading with BMSCs lysate through basic characterization and morphological analysis. Moreover, this study was mainly designed to investigate the role of NLC loaded BMSCs lysate in reducing inflammation via in-vitro and in-vivoassays. The in-vitro study involves cell viability assays, p53, annexin V and VEGF expression through ELISA and immunocytochemistry, real-time BAX, caspase-3, IL-6, IL-8, TOP2A, PCNA, and Ki-67 gene expression analysis. Additionally, to evaluate in-vivo anti-inflammatory activity, the carrageenan-induced rat paw oedema model was used. In-vitro results showed that NLC loaded BMSCs lysate increased cell viability, decreased apoptosis and pro-inflammatory genes expression and up-regulated angiogenesis and proliferation in H2O2 pre-stimulated cells. Findings of the in-vivo assay also indicated a reduction in rat's paw oedema volume in NLC-loaded BMSCs lysate, and downregulation of BAX, Caspase-3, IL-6, and IL-8 was observed. Enhanced expressions of TOP2A, PCNA, and Ki-67 were obtained. Concluding the results of this study, NLC-loaded BMSCs lysate could reduce inflammation and possibly regenerate damaged tissue mainly via increasing cell viability, angiogenesis and proliferation, and reducing apoptosis and pro-inflammatory cytokines.


Células estromais mesenquimais derivadas da medula óssea (BMSCs) têm sido utilizadas para o tratamento de distúrbios inflamatórios. Devido ao grande tamanho das BMSCs em comparação com as nanopartículas, as BMSCs não podem ser carregadas nas nanopartículas. Supõe-se que o carregamento de lisado de BMSCs no nanocarriers será eficaz na entrega de conteúdos celulares e elementos reguladores de BMSCs no local da lesão. Este estudo teve como objetivo investigar a carga de carreador lipídico nanoestruturado (NLC) com lisado de BMSCs através de caracterização básica e análise morfológica. Além disso, este trabalho foi projetado, principalmente, para investigar o papel do lisado de BMSCs carregado com NLC na redução da inflamação por meio de ensaios anti-inflamatórios in vitro e in vivo. O estudo in vitro envolve ensaios de viabilidade celular, expressão de p53, anexina V e VEGF por ELISA e imunocitoquímica e expressão gênica em tempo real de BAX, caspase-3, IL-6, IL-8, TOP2A, PCNA e Ki-67 . Além disso, para avaliar a atividade anti-inflamatória in vivo,o modelo de edema de pata de rato induzido por carragenina foi utilizado. Os resultados in vitro mostraram que o lisado de BMSCs carregadas com NLC aumentou a viabilidade celular, diminuiu a apoptose e a expressão de genes pró-inflamatórios e aumentou a angiogênese e proliferação em células pré-estimuladas com H2O2. Os achados do ensaio in vivo também indicaram uma redução no volume do edema da pata de rato no lisado de BMSCs carregado com NLC, entretando, foi observada a regulação negativa de BAX, Caspase-3, IL-6 e IL-8. Expressões aumentadas de TOP2A, PCNA e Ki-67 foram obtidas. Assim, concluindo os resultados do estudo, é possível afirmar que o lisado de BMSCs carregado com NLC pode reduzir a inflamação e possivelmente regenerar o tecido danificado principalmente por meio do aumento da viabilidade celular, angiogênese e proliferação e redução da apoptose e citocinas pró-inflamatórias.

Purpose: To evaluate the effect of preoperative intravenous chemotherapy with 5-fluorouracil on liver regeneration in an experimental model of major hepatectomy in rats. Methods: Wistar rats were divided into two groups of 20 animals each and submitted to 70% hepatectomy 24 h after intravenous injection of 5-fluorouracil 20 mg/kg (fluorouracil group, FG) or 0.9% saline (control group, CG). After hepatectomy, each group was subdivided into two subgroups of 10 animals each according to the day of sacrifice (24 h or 7 days). Liver weight during regeneration, liver regeneration rate using Kwon formula, and the immunohistochemical markers proliferating cell nuclear antigen (PCNA) and Ki-67 were used to assess liver regeneration. Results: At early phase (24 h after hepatectomy) it was demonstrated the negative effect of 5-fluorouracil on liver regeneration when assessed by Kwon formula (p < 0.0001), PCNA analysis (p = 0.02). With regeneration process complete (7 days), it was possible to demonstrate the sustained impairment of chemotherapy with 5-fluorouracil on hepatocytes regeneration phenomenon when measured by Kwon formula (p = 0.009), PCNA analysis (p = 0.0001) and Ki-67 analysis (0.001). Conclusions: Preoperative chemotherapy with intravenous 5-fluorouracil negatively affected the mechanisms of liver regeneration after major hepatectomy in rats.

Background: Squamous cell carcinoma (SCC) is a common cutaneous neoplasm in horses, which mainly affects the external genitalia, oral cavity, and periocular region. The development of SCC metastases is rare in these animals, and the most common occurrence is a marked local infiltration. Exposure to ultraviolet rays and skin depigmentation are the main etiological factors of SCC. Definitive diagnosis of the neoplasm is performed through histological examination of lesions. The present report describes the clinical, anatomopathological, and immunohistochemical (IHC) aspects of a case of metastatic SCC, with the vulva as the primary site in a mare. Case: A 17-year-old mare, mixed breed, was referred to the Veterinary Hospital of the Universidade de Passo Fundo (UPF), with history of areas of depigmented skin (pinto coat), and clinical history of anorexia, frequent episodes of colic, and recumbency. During clinical evaluation, heart rate (68 bpm), rectal temperature (38.4ºC), and respiratory rate (48 mpm) were elevated, and the oral mucosa was pale. The mare also had an ulcerated tumor mass involving the vulva, which extended to the inguinal region and involved the mammary gland. A cytological aspirate of the vulvar tumor was performed, in which no neoplastic cells were found. Next, a biopsy of 2 distinct areas of the vulva was performed. The material was sent for anatomopathological examination, which showed markedly pleomorphic malignant squamous cells, with individual keratinization and high mitotic index, organized in trabeculae with rare keratin pearl-like formations. The exam allowed the diagnosis of SCC Grade II. Due to the poor prognosis and high cost of treatment, the owner consented to euthanasia and necropsy examination. During necropsy, the vulvar tumor mass was grayish, firm to hard, infiltrative, and had friable areas. Tumor foci suggestive of metastasis were also observed in inguinal, mesenteric, mediastinal and renal lymph nodes, adrenal glands, lung, pericardium, medullary canal, intercostal muscles, right 15th rib, and tissue surrounding the azygos vein. Samples from all organs were collected for anatomopathological examination. Diagnosis of metastatic vulvar SCC was confirmed through histological and IHC studies, which evaluated the expression of cytokeratins (AE1/ AE3), as well as the proliferative activity of neoplastic cells through the PCNA marker. Discussion: The diagnosis of metastatic SCC was obtained through the observed clinical, necroscopic, histological, and IHC characteristics. This neoplasm usually appears in depigmented regions exposed to ultraviolet light, and older animals are more likely to be affected. The mare in the present case had areas of depigmented skin (pinto coat). The animal was kept in a paddock outdoor and exposed to constant solar radiation. Although rare in horses, manifestation of anorexia, progressive weight loss, and frequent colic episodes and recumbency may be closely related to the multiple sites of metastasis in the present case. The main histological findings of the neoplasm were the dense proliferation of malignant squamous cells with individual keratinization, arranged in a trabecular pattern and with rare formations of keratin pearls, in line with previous studies. During IHC evaluation, the neoplastic cells showed expression of cytokeratins (AE1/AE3), as well as high proliferative activity evidenced by the PCNA marker. Given this background, the present report describes the clinical, anatomopathological, and IHC aspects of a case of metastatic SCC with a primary site in the vulva of a mare.

Sebaceous tumors are common in dogs and include benign and malignant lesions. The increase in immunohistochemical evaluation of sebaceous tumors aggregates information regarding the origin and degree of malignancy of the lesions. The objective of this study was to evaluate PCNA immunohistochemical expression of  sebaceous tumors in dogs. Sixty-one samples include normal skin and sebaceous tumors were selected from dogs of various breeds and ages, no gender predilection, sent to the Veterinary Pathology Service of Universidade Federal Fluminense, Niterói/RJ, Brazil. Samples passed through histological processing, routine staining and immunostaining from PCNA. Descriptive statistical analysis, Wilcoxon-Mann-Whitney and Fishers exact test were performed. The mean age of the affected animals was 10.56 years. There was no sexual predilection. Breeds more frequently affected were: Poodles, mixed-breed dogs and Cocker Spaniels. Most of the tumors were neoplasms (67.27%), and 75.67% of those were benign. Sebaceous adenoma was the most frequent neoplasm (56.75%). PCNA immunoexpression was present in hyperplasia, benign and malignant tumors. No previous studies were found using PCNA antibody in sebaceous tumors of dogs. Thus, more studies are needed to provide greater clarity regarding the role of these markers on normal skin and sebaceous tumors of dogs, as well as their use as prognos(AU)

Foram realizadas análises estatísticas descritivas além dos testes de Wilcoxon-Mann-Whitney para comparar a distribuição da marcação de anti-PCNA entre grupos de variáveis. Para variáveis com mais de dois grupos, aplicou-se a Análise de Variância (ANOVA). A idade média dos animais afetados foi de 10.56 anos. As raças mais afetadas foram Caniches e Cocker Spaniel, e ainda animais sem raça definida. Houve imunomarcação de PCNA em tumores benignos, malignos, e ainda em lesões hiperplásicas com intensidade variada. Sebaceous tumors are common in dogs. These tumors include both benign and malignant lesions. Immunohistochemical evaluation of these tumors can aggregate information regarding the origin and degree of malignancy of the lesions. Focusing on this matter, sixty-one samples including normal skin and sebaceous tumors were selected from dogs of various breeds and ages, with no predilection for sex, from the archive of Veterinary Pathology Service of Federal Fluminense University, Niterói/RJ, Brazil. The samples underwent to histological processing, routine staining and immunohistochemistry with anti-PCNA (proliferating cell nuclear antigen). Descriptive statistical analysis was performed, the Wilcoxon-Mann-Whitney test was used to compare the distribution of anti-PCNA labelling in different groups of variables. In case there were more than two groups, the Analysis of Variance (ANOVA) test was performed. The mean age of the affected animals was 10.56 years. The most affected breeds were Caniches and Cocker Spaniels, as well as mixed breed animals. There was immunostaining of PCNA in both benign and malignant tumors, as well as in hyperplasic lesions with varying intensity. Most of the tumors were neoplasms which represented 67.27% of the total sample; within these, 75.00% were benign. The most frequent neoplasm was sebaceous adenoma (37.74%). Results indicated no statistical difference in the distribution of anti-PCNA labelling between the groups of sex, age, reproductive status, localization, size of tumor, and histopathological diagnosis. Although there are not many studies analyzing anti-PCNA labelling in sebaceous tumors, several of them pointed out to the predictive value in other neoplasms. With this matter in mind, we intended to evaluate the expression of anti-PCNA in canine sebaceous tumor and a possible association with the malignancy of the lesions.

Purpose: To investigate the effects of pine needle extract (PNE) on the expression of proliferating cell nuclear antigen (PCNA) and Ki-67 during liver regeneration induced by 70% partial hepatectomy (PH) in rat. Methods: Forty-eight male rats (SD, 7 weeks) had surgery (70% PH). They were randomly divided into two groups. PH + PNE group was only provided PNE diluted in water (10%) for drinking and PH group was provided water from 5 days before surgery to the time of sacrifice. PNE was made by pressing and filtering. Animals were sacrificed at 12h, 24h, 36h, 60h, 84h, 168h after PH, respectively. The expressions of PCNA and Ki-67 were determined as proliferation indices. Results: Immunohistochemistry turned out to increase the expression of PCNA and Ki-67. PCNA expression of PH+PNE group increased up to twice of that of PH group. Western blot also seemed to increase the PCNA expression. These results indicated the promotion of cell proliferation in liver tissue and hepatic regeneration. Conclusions: Pine needle extract stimulates the expression of some mitotic proteins during liver regeneration induced by 70% PH in rats. It suggests that administration of pine needle extract could accelerate the liver regeneration after partial hepatectomy.(AU)

Resumo

A case of congenital hepatic fibrosis in a 10-month-old calf is described. The calf had been raised in a feed lot for the past 173 days and exhibited loss of body condition and depression, followed by death. The most remarkable macroscopic lesions observed during necropsy were: jaundice, hepatomegaly with a multilobulated liver surface and diffuse pale brown colour of the parenchyma. Microscopically, the portal tracts were thickened and connected to each other by abundant fibrous tissue, delimiting irregular lobules of hepatocytes and occasional fibrosis of the central veins (perisinusoidal scars) and hyperplasia of embryonal bile ducts. Immunohistochemistry staining of the cytoplasm of the bile ducts was positive to AE1/AE3 and cell proliferation nuclear antigen (PCNA). The findings at necropsy, together with the results of the histopathological and immunohistochemistry studies, confirmed the diagnosis of congenital hepatic fibrosis.

Resumo

Purpose: To evaluate liver regeneration after selective ligation of portal vein and hepatic artery by 3D Computed Tomography in an experimental model. Methods: Sixteen Wistar rats were randomized into four equal groups: Group I- control (sham), Group II- isolated selective ligation of the hepatic artery, Group III- isolated selective ligation of the portal vein and Group IV- combined ligation of portal vein and hepatic artery. Before procedure and five days after a 3D CT Scan was performed to analyze the hypertrophy, weight and function of the remnant liver. Results: The largest regeneration rate and increase of weight in the hypertrophied lobe was detected in group IV, the first with an average of 3.99 (p=0.006) and the last varying from 6.10g to 9.64g (p=0.01). However, total liver weight and the R1 ratio (Hypertrophied Lobe Weight/Total Liver Weight) was higher in group III (P 0.001) when compared with groups I, II and IV and showed no difference between them. The immunohistochemical examination with PCNA also found higher percentages with statistical significance differences in rats of groups III and IV. It was possible to confirm a strong correlation between hypertrophied lobe weight and its imaging volumetric study. Liver function tests only showed a significant difference in serum gamma-glutamyltransferase and phosphorous. Conclusion: There is a largest liver regeneration after combined ligation of portal vein and hepatic artery and this evidence may improve the knowledge of surgical treatment of liver injuries, with a translational impact in anima nobile.(AU)

We conclude that oocytes in multi-oocyte follicles seem to be in different stages of the cell cycle.


O objetivo deste trabalho foi caracterizar a atividade proliferativa de folículos multi-oócitos com antígeno marcador de proliferação celular (PCNA). Ovários (n = 12) de novilhas foram processados para histologia. A partir de 789 folículos multi-oócitos identificados, somente 11 foram considerados adequados para imunohistoquímica por apresentarem todos os núcleos claramente visíveis. Todos os folículos multi-oócitos foram positivos para PCNA, mas alguns oócitos não apresentaram marcação. Concluímos que oócitos presentes em folículos multi-oócitos parecem estar em diferentes fases do ciclo celular.

The aim of this study was to compare the cell proliferation rate of follicles from Bostaurus indicusfemales with different antral follicle counts (AFCs). Thirty pairs of ovaries were classified as having low(≤ 31 follicles), intermediate (≥ 46 and ≤ 76 follicles) or high AFC (≥ 91 follicles). The ovaries were cut into 1 x 1 x 0.3 cm fragments, fixed in 10% buffered formalin solution and subjected to immunohistochemical analysis for proliferating cell nuclear antigen (PCNA). PCNA detection was detected in all stages of follicular development, being 1,388 (82.23%) primordial, 197 (11.67%) primary, 29 (1.72%) secondary and 74 (4.83%) antral follicles. The PCNA detection rate was higher (P ≤ 0.05) for antral and secondary follicles in ovaries with intermediate and low AFCs. This study showed that higher PCNA at early stages of follicular growth is detected in animals with high AFC, whereas a higher PCNA is seen in secondary and antral follicles of animals with low AFC.

Purpose: To determine the effects of propofol and ketamine anesthesia on liver regeneration in rats after partial hepatectomy (PHT). Methods: Male Wistar albino rats were assigned randomly to four groups of 10. Anesthesia was induced and maintained with propofol in groups 1 and 2, and with ketamine in groups 3 and 4. PHT was undertaken in groups 1 and 3. Rats in groups 2 and 4 (control groups) underwent an identical surgical procedure, but without PHT. At postoperative day-5, rats were killed. Regenerated liver was removed, weighed, and evaluated (by immunohistochemical means) for expression of inducible nitric oxide synthase (iNOS), endothelial NOS (eNOS), apoptosis protease-activating factor (APAF)-1, and proliferating cell nuclear antigen (PCNA). Also, blood samples were collected for measurement of levels of tumor necrosis factor (TNF)- and interleukin (IL)-6. Results: Between groups 2 and 4, there were no differences in tissue levels of iNOS, eNOS, and APAF-1 or plasma levels of TNF- and IL-6. eNOS expression was similar in group 1 and group 3. Expression of iNOS and APAF-1 was mild-to-moderate in group 1, but significantly higher in group 3. Groups 1 and 3 showed an increase in PCNA expression, but expression in both groups was comparable. Plasma levels of TNF- and IL-6 increased to a lesser degree in group 1 than in group 3. Conclusion: Propofol, as an anesthetic agent, may attenuate cytokine-mediated upregulation of iNOS expression and apoptosis in an animal model of liver regeneration after partial hepatectomy.(AU)

Purpose: To evaluate the effect of aqueous extract of Baccharis trimera leaves on the proliferative capacity of the liver after partial hepatectomy (PH) in rats. Methods: Twenty Wistar rats weighing between 300 and 450g were divided into two groups: control (HP) and test (HP100-rats that received the aqueous extract of Baccharis trimera for four days at a dose of 100 mg / kg / day). On the fifth day, animals from both groups underwent resection of 70% of the liver. Twenty-four hours later, they were sacrificed and the remnant liver was removed and prepared for studied through PCNA immunohistochemistry. Data analysis for comparison between the two groups was made through the non-parametric statistical test Mann-Whitney test. Results: In all the animals studied was found most abundant nuclear immunostaining positive hepatocytes interlobular located in regions of the liver. Quantitative analysis of PCNA-positive cells revealed positivity rate significantly higher mean (p = 0.02) in HP100 group (77.1 ± 13.6) compared to the HP group (45.8 ± 12.9). Conclusion: DAdministration of aqueous extract of the leaves of Baccharis trimera 100 mg/kg of animal has a significant positive effect on liver regeneration in rats, 24 hours after hepatectomy (70%).(AU)

The extract of Auxemma oncocalyx (A. oncocalyx) and its main component, oncocalyxone A (onco A), possess anti-tumoral activity that may affect fertility. There is limited literature on the action of these substances regarding caprine folliculogenesis. In this study, we evaluated the effect of A. oncocaly and onco A on the in vitro culture of isolated secondary follicles (Experiment 1) and on the in vitro maturation (IVM) of oocytes from caprine antral follicles grown in vivo (Experiment 2). Isolated secondary follicles were distributed in six groups: the non-cultured control was immediately fixed in 4% paraformaldehyde. The remaining follicles were cultured for 7 days in α-minimal essential medium (α-MEM+ ) alone (control) or with dimethyl sulfoxide (DMSO), doxorubicin (DXR), A. oncocalyx, or onco A. After culture, the follicles were evaluated for antrum formation, growth rate, apoptosis (TUNEL), cellular proliferation (PCNA), as well as the expression of BCL2 and BAX. In addition, cumulus oocyte complexes (COCs) were aspirated and allocated into five treatments for IVM: control, cultured only in maturation base medium (TCM 199+ ); or supplemented with DMSO; DXR; A. oncocalyx or onco A. After IVM, oocyte chromatin configuration and viability were assessed. After 7 days of culture, a reduction was noted in the percent of morphologically intact follicles, antrum formation, growth rate, and numbers of PCNApositive granulosa cells (P < 0.05). After culture, the DXR treatment showed a higher percent of TUNELpositive follicles and relative BAX:BCL2 mRNA ratio’s (P < 0.05). After IVM of the COCs, DXR, A. oncocalyx, and onco A treatments showed a greater percent (P < 0.05) of abnormal oocytes and a lower percent of viable oocytes as compared with the control group (P < 0.05)...

Entretanto, estudos sobre a ação dessas substâncias em relação à foliculogênese caprina são desconhecidos. O objetivo desse estudo foi avaliar o efeito da A. oncocalyx e onco A no cultivo in vitro de folículos secundários isolados (Experimento 1) e na maturação in vitro (MIV) de oócitos de folículos antrais caprinos crescidos in vivo (Experimento 2).
