The advancements in the cell culture studies have led to the development of regenerative medicine concept. The aim of this study is to compare the effectiveness of some washing solutions, including phosphate buffered saline (PBS), sodium chloride (NaCl), and ringer's lactate (RL) on the rate of detachment and confluency in fibroblast and osteoblast cell culture. Baby Hamster Kidney 21 clone 13 (BHK21/C13) fibroblast cells and 7F2 osteoblast were cultured on T25 flasks for 3-4 days. Three treatment groups were classified on the basis of different washing solutions used in the moment before trypsinization: PBS, 0.9% NaCl, and RL. Each group was measured for the detachment rate and cell confluence. The measurement was done in 2 passage numbers. The use of PBS, NaCl, and RL washing solution showed that detachment time was less than 5 minutes for the fibroblasts and 3 minutes for the osteoblasts. There was a significant difference in the rate of fibroblast cell detachment (p=0.006) and osteoblast (p=0.016). The capability of fibroblasts and osteoblasts to achieve a confluence of 106 cells/well on the first and second measurements was almost the same between the washing solution groups. The use of physiological 0.9% NaCl solution as a washing solution in fibroblast and osteoblast cell culture has almost the same effectiveness as PBS to help accelerate cell detachment in less than 5 minutes without influencing the capability of cells to proliferate.
Os avanços nos estudos de cultura de células levaram ao desenvolvimento do conceito de medicina regenerativa. O objetivo deste estudo é comparar a eficácia de algumas soluções de lavagem, incluindo solução salina tamponada com fosfato (PBS), cloreto de sódio (NaCl) e lactato de ringer (RL) na taxa de desprendimento e confluência em cultura de células de fibroblastos e osteoblastos. Células de fibroblastos Baby Hamster Kidney 21 clone 13 (BHK21/C13) e osteoblastos 7F2 foram cultivadas em frascos T25 por 3-4 dias. Três grupos de tratamento foram classificados com base nas diferentes soluções de lavagem utilizadas no momento anterior à tripsinização: PBS, NaCl 0,9% e RL. Cada grupo foi medido para a taxa de desprendimento e confluência celular. A medição foi feita em 2 números de passagem. O uso de solução de lavagem PBS, NaCl e RL mostrou que o tempo de descolamento foi inferior a 5 minutos para os fibroblastos e 3 minutos para os osteoblastos. Houve uma diferença significativa na taxa de desprendimento de células fibroblásticas (p=0.006) e osteoblasto (p=0.016). A capacidade de fibroblastos e osteoblastos de atingir uma confluência de 106 células/well na primeira e segunda medições foi quase a mesma entre os grupos de solução de lavagem. O uso de solução fisiológica de NaCl (0.9%) como solução de lavagem em cultura de células de fibroblastos e osteoblastos tem quase a mesma eficácia que o PBS para ajudar a acelerar o desprendimento celular em menos de 5 minutos sem influenciar a capacidade das células de proliferar.

The present study presents a comparative evaluation of the transit kinetics of straw briquette in response to the dietary addition of non-protein nitrogen sources in the form of a mineral supplement. Four rumen-cannulated, castrated Holstein-Gir crossbred cattle, weighing an average of 380±22.64 kg, were distributed into a 4 × 4 Latin square design (four supplements with non-protein nitrogen sources × four experimental periods). The following non-protein nitrogen sources were studied: conventional urea, slow-release urea, extruded urea, and monoammonium phosphate. During the experiment, the animals were housed in individual stalls with concrete floors where they received a basal diet consisting of straw briquette, potato starch, and the mineral supplement, the latter whose variation was only in the non-protein nitrogen source, which characterized the treatments. The different non-protein nitrogen sources did not affect the parameters of transit or degradation kinetics of straw fiber briquette. These results can be associated with the low nitrogen content limited by the types of supplements and the particle size of straw briquette, which is smaller due to processing.(AU)

An outbreak of photosensitization in Holstein and Jersey cattle occurred after the passage of an extratropical cyclone in Southern Brazil. On that occasion, several trees fell, including some of the species Myrocarpus frondosus (Fabaceae family) popularly known as "gabreúva", "cabriúna" or "gabriúna". Five heifers consumed the leaves of this plant, and all of them became ill. The main clinical signs were decreased appetite, permanence in the water, decreased ruminal movements, dry stools, congested and icteric sclera and conjunctivae, dark urine and restlessness when exposed to the sun. One heifer showed circling, incoordination, restlessness and death six days after the onset of clinical signs. From the seventh day, three heifers showed improvement; one remained apathetic, with jaundiced mucous membranes, ulceration, and scaling on the tongue, increased respiratory rate, and fever, and died 18 days after ingestion of the plant. At necropsy, photosensitivity lesions were observed on the depigmented skin, snout, teats, and ear tip, characterized by discontinuity of the skin with the formation of crusts, inflammatory exudate associated with redness of the skin, in addition to ulceration in the mouth, snout and ventral portion of the tongue and jaundice and edema in the submandibular region. The liver was enlarged, with an orange color, full gallbladder, and wall edema. Through microscopy, liver lesions were characterized by moderate and diffuse necrosis of hepatocytes, vacuolar and centrilobular degeneration, bilirubin retention and proliferation of biliary epithelium. The literature shows no reports of M. frondosus toxicity in cattle. Experiments were carried out in cattle to clarify the possible clinical-pathological picture produced by this plant. A survey of the epidemiological data of the diagnosed spontaneous outbreak was carried out, and subsequently, three cattle received the plant's green leaves, and a fourth received the dried leaves. After administering the plant, blood samples were collected daily for blood count, measurement of liver enzymes (gamma glutamyl transferase, alkaline phosphatase, alanine aminotransferase, aspartate transferase), bilirubin, measurement of vital parameters and liver biopsy from the onset of signs. In case of death, a necropsy was performed with collection of viscera samples for macro and microscopic evaluation. The experimental part was developed at the Laboratory of Animal Pathology, CAV-UDESC. The single dose of 33g/kg of green leaves caused severe clinical signs, while the administration of 22g/kg and 11g/kg caused mild to moderate signs of the disease. The supply of desiccated leaves in a single dose did not cause clinical manifestations. M. frondosus can be blamed as a cause of spontaneous hepatogenous photosensitization in cattle. Experimentally, the ingestion of green leaves of this plant, in doses greater than 11g/kg can lead to clinical changes, with subsequent recovery, or progress to death with severe liver damage.
Um surto de fotossensibilização em bovinos da raça Holandesa e Jersey foi observado após a passagem de um ciclone extratropical no Sul do Brasil, quando houve queda de diversas árvores, dentre elas Myrocarpus frondosus (família Fabaceae), popularmente conhecida como "gabreúva, cabriúna ou gabriúna". Cinco novilhas consumiram folhas dessa planta, e todas adoeceram. Os principais sinais clínicos foram diminuição do apetite, permanência dentro da água, diminuição dos movimentos ruminais, fezes ressecadas, esclera e conjuntivas congestas e ictéricas, urina escura e inquietação quando expostas ao sol. Uma novilha apresentou andar em círculos, incoordenação e inquietação e morte seis dias após início dos sinais clínicos. A partir do sétimo dia, três novilhas apresentaram melhora e uma permaneceu apática, com as mucosas ictéricas, ulceração e descamação na língua, aumento da frequência respiratória, febre e morreu 18 dias após ingestão da planta. Na necropsia foram observadas lesões de fotossensibilização na pele despigmentada, focinho, tetos, ponta da orelha, caracterizadas por descontinuidade da pele com formação de crostas, exsudato inflamatório associado a vermelhidão da pele além de ulceração na boca, focinho e porção ventral da língua e icterícia acentuada e edema, na região submandibular. O fígado estava aumentado de volume, de cor laranja, vesícula biliar repleta e com edema de parede. Através da microscopia foram observadas, lesões hepáticas, caracterizadas por necrose de hepatócitos, moderada e difusa, degeneração vacuolar, centrolobular, retenção de bilirrubina e proliferação de epitélio biliar. Na literatura, não há relatos da toxicidade de M. frondosus para bovinos. Com o objetivo de esclarecer o possível quadro clínico-patológico produzido por essa planta foram conduzidos experimentos em bovinos. Foi realizado um levantamento dos dados epidemiológicos do surto espontâneo diagnosticado e, posteriormente, três bovinos receberam as folhas verdes da planta e um quarto recebeu as folhas dessecadas. Após a administração da planta, foram coletadas amostras de sangue diariamente para hemograma, dosagem de enzimas hepáticas (gama glutamil transferase, fosfatase alcalina, alanina aminotransferase, aspartato transferase), bilirrubina, aferição dos parâmetros vitais e biópsia hepática a partir do surgimento dos sinais clínicos. Em caso de morte, foi realizada necropsia com coleta de amostras de vísceras para avaliação macro e microscópica. A parte experimental foi desenvolvida nas dependências do Laboratório de Patologia Animal, CAV-UDESC. A dose única de 33g/kg de folhas verdes causou graves sinais clínicos, enquanto a administração de 22g/kg e 11g/kg causou sinais leves a moderados da doença. O fornecimento de folhas dessecadas em dose única não causou manifestações clínicas. M. frondosus pode ser responsabilizada como causa de fotossensibilização hepatógena espontânea em bovinos. A ingestão de folhas verdes dessa planta em doses superiores a 11g/kg pode levar a alterações clínicas, com posterior recuperação, ou evoluir para morte com lesões hepáticas graves.

The objective of this study was to characterize the dynamics of food passage in the gastrointestinal tract (GIT) of Japanese quail. A randomized design was used with four treatments and four experimental units with one bird each, nine of which were measured in time. The ingredients used were albumin, starch, cellulose, and soybean oil. Suspensions containing barium sulfate and saline were prepared, except for soybean oil. For each bird, 2.5 mL of the suspension was administered directly to the crop. Dual-energy absorptiometry (DEXA) was used to quantify the passage of food. The birds were sedated and maintained under inhalation anesthesia during the scan. Measurements were taken at intervals of 0.5, 1, 2, 4, 6, 8, 10, 12, and 24 h. The variables identified were passage time, first quality, and the average time of each variable. Based on these results, the ingredients may show differences in dynamic passing on the Japanese quail GIT. The duration of the first attempt was 32 min, ranging from 21 to 44 min. The average time value of food choice was close to 10.8 h and varied according to the ingredient from 8.45-12.16 h. Among the variables, soybean oil presented values ââthat denote a fast passage in the GIT, while albumin presented values ââthat denote a slower passage. The dynamics of food passage in the GIT of Japanese quails varies according to the chemical composition of the ingredients.(AU)

Background: The artificial insemination has become a well-established method in the breeding of bitches, and evaluation of the factors that may potentially affect pregnancy success is essential. For this reason, it is essential to evaluate the factors that may affect fertility of the bitch when artificial insemination is performed. Serum progesterone concentrations and vaginal cytology have been used to determine the time of ovulation and stage of the estrus cycle. This study aimed to evaluate the artificial insemination method, the serum progesterone concentration, the breed size, age, the whelping number, vaginal cytology parameters, and their interactions on pregnancy success in bitches. Materials, Methods & Results: A total of 607 bitches that had undergone reproductive consultation with the Mexican Canine Federation from January to December 2016 were enrolled in the present study and assigned to one of 2 artificial insemination methods (intravaginal and transcervical) using fresh semen. Determination of the estrus cycle phase and the time of Artificial insemination was based on vaginal cytology and serum progesterone concentrations. Bitches inseminated by the transcervical technique had a higher pregnancy rate with respect to females inseminated by the intravaginal technique (P < 0.05). Moreover, females with a serum progesterone concentration of 5-10 ng/mL had a greater probability (> 4 times) of getting pregnant than animals with lower or higher progesterone concentrations (P < 0.05). Bitches inseminated by the intravaginal technique and with serum progesterone concentrations >10 ng/mL had a considerable reduction in pregnancy (P < 0.05) compared with females with < 10 ng/mL serum progesterone or with bitches inseminated by the transcervical technique. Discusion: Serum progesterone concentration, the artificial insemination method, and superficial cells without a nucleus modified the pregnancy rate in bitches. Females inseminated by transcervical semen deposition had a higher pregnancy rate than females inseminated by the intravaginal technique. Using fresh or frozen-thawed semen produced a higher pregnancy rate in bitches inseminated by transcervical semen deposition than females inseminated by the intravaginal technique. Differences in the pregnancy rate between transcervical and intravaginal insemination could be associated with the correct semen disposition, the distance that the sperm must travel to reach the oocyte, as well as the number of sperm that reach the oviduct ampulla. Exist evidences that after ovulation, as progesterone rises, the cervix is closed, which may compromise the passage of the sperm deposited into the vagina. Therefore, it is likely that in females with a serum progesterone concentration > 10 ng/mL, the cervix was closed, compromising the ability of the sperm to access the oviduct. Thus, the use of intravaginal insemination should be done in bitches with a serum progesterone concentrations < 11 ng/mL to reduce the possibility of cervical closure and to increase the odds of pregnancy. It is well documented that the serum progesterone concentration and vaginal cytology parameters have a great influence on pregnancy success, and the results confirm these findings. In the present study, 96% of the bitches inseminated with a serum progesterone concentration of 5-10 ng/mL got pregnant and had higher odds of pregnancy than bitches with lower or higher serum progesterone concentrations.

It is important to evaluate nutritional value of new grass species, which can be better characterized through rumen fermentation and degradation techniques. The aim was to evaluate the effects of Nitrogen (N) fertilization on the kinetics of fermentation and ruminal degradation of the Mulato II grass. Two distinct sources of N (common and protected urea) were used during two periods of the year (summer - I and autumn - II). A completely randomized experimental design was used, in a 2x4 factorial arrangement with three replicates. Fraction A changed based on N dosage. The passage rate (K) and effective degradability (ED) were influenced by sources and doses of N, while Fraction B and Lag Time were not influenced during period I. Fractions A and B and EDs were all influenced by N sources and doses during period II. Lag time and potential degradability were influenced by N sources only. Among the kinetics parameters of dry matter ruminal fermentation obtained during period I, only K1 was influenced by the interaction between N doses x sources, while the remaining parameters were only influenced by the N doses applied. As for period II, with the exception of K2, all remaining parameters were influenced by the interaction of N doses and sources. Protected urea improved fermentation parameters during period I. The dose equivalent to applying 150kg/ha positively influenced ruminal fermentation parameters of the Mulato II grass mainly in the summer. The use of protected urea as a Nitrogen source improved fermentation parameters in the summer.
É importante avaliar o valor nutricional de novas espécies forrageiras, parâmetro que pode ser melhor caracterizado por meio de técnicas de fermentação e degradação ruminal. Objetivou-se avaliar os efeitos da adubação nitrogenada (N) sobre a cinética de fermentação e degradação ruminal do capim Mulato II. Duas fontes de N (ureia comum e protegida) foram utilizadas em dois períodos do ano (verão - I e outono - II). O delineamento experimental utilizado foi inteiramente casualizado, em esquema fatorial 2x4 com três repetições. A fração A mudou com base na dosagem de N. A taxa de passagem (K) e a degradabilidade efetiva (DE) foram influenciadas pelas fontes e doses de N, enquanto a Fração B e Lag Time não foram influenciadas durante o período I. As frações A e B e DE foram influenciadas pelas fontes e doses de N durante o período II. Lag time e a degradabilidade potencial foram influenciadas apenas pelas fontes de N. Dentre os parâmetros cinéticos da fermentação ruminal da matéria seca obtidos no período I, apenas K1 foi influenciado pela interação entre doses de N x fontes, enquanto os demais parâmetros foram influenciados apenas pelas doses de N aplicadas. Já para o período II, com exceção do K2, todos os demais parâmetros foram influenciados pela interação das doses e fontes de N. A ureia protegida trouxe melhorias para os parâmetros fermentativos no período I. A dose equivalente à aplicação de 150kg/ha influenciou positivamente os parâmetros fermentativos ruminais do capim Mulato II, principalmente no verão. O uso de ureia protegida como fonte de nitrogênio melhorou os parâmetros de fermentação no verão.

Background: Gastroesophageal foreign bodies (GFD) are commonly diagnosed in dogs and are considered an endoscopic emergency that, although not resulting in serious clinical sequelae or mortality, can compromise the health and well-being of the patient. The use of the digestive endoscopy for the diagnosis and treatment of GFD can be a valuable and viable alternative. There are cases of GFD in dogs for which the indicated treatment is surgery, which can be performed using minimally invasive or conventional techniques, associated or not with flexible endoscopy. The objective of this work is to describe 16 cases of GFD removal in dogs demonstrating the efficiency of upper digestive endoscopy. Cases: Of the 16 GFD cases, 63% (10/16) were male and 37% (6/16) female. Most aged under 1 year (63%), puppies (5/16) and juveniles (5/16). The patient with the lowest body weight was a miniature pinscher weighing 0.8 kg (Case 14) and the heaviest was an American Pit Bull Terrier weighing 28 kg (Case 11), the mean body weight of patients diagnosed with GFD was 10.2 ± 6.7 kg. Small and medium breeds were more affected, 44.7% (7/16) and 44.7% (7/16), respectively, and large breeds (Golden Retrievier and Bull Terrier), from cases 1 and 4, the least affected, 12.6% (2/16) of the cases. The 16 patients underwent a 12 h food fast and a 4 h water fast, as gastrointestinal emptying in these cases of GFD can be influenced by these foreign bodies. All underwent general inhalation anesthesia with monitoring of physiological parameters (temperature, heart rate, respiratory rate, oxygen saturation and blood pressure) before, during and after EGD, being positioned in left lateral decubitus. The 16 canine patients with suspected GFD underwent EGD for diagnostic confirmation and removal of foreign bodies. Five esophageal FB were diagnosed, 31% (5/16), and 11 gastric FB, 69% (11/16). The most frequently diagnosed foreign bodies were bone and tissue, 37.5% (6/16) and 31% (5/16). Other foreign bodies were materials such as plastics, metals, rubber, foam and stone. Of the 16 cases of GFD, EGD efficiently treated 88% (14/16) without the need for hospitalization, with only supportive treatment for the remission of complications caused by the presence of foreign bodies in the gastroesophageal tract. The main complications related to the presence of GFD were esophagitis in 25% (4/16) of cases, gastritis in 38% (6/16) and both alterations in 13% (2/16). Discussion: In this work, we can observe that more than a third of the clinical cases of treated dogs were diagnosed with GFD, demonstrating that these cases are common in the veterinary clinic. Most of these animals were males less than 1 year old. The improvement of learning in this category can lead these animals to exacerbated oral exploration of new objects. Most FBs were found in the stomach because they were of adequate size, consistency and shape for their passage through the esophagus, whereas esophageal FBs were all bone fragments of rigid consistency with diameters and sizes larger than the esophageal lumen. The interval between the ingestion of the object and the veterinary care can be decisive for the removal of the FB in the esophagus or stomach. Most gastric FBs removed were fabrics and plastics, flexible objects that can pass through the esophageal lumen more easily. Removal of GFD by endoscopy was performed with a high success rate, with only 2 cases being resolved by esophagostomy and gastrotomy. Flexible endoscopy proved to be an efficient technique for removing treated GFD, which can help remove FB during esophagotomy and be associated with rigid endoscopy. Patients recovered quickly and without complications, but it is important to emphasize that inadequate maneuvers and conducts can determine other outcomes. The use of endoscopy for GFD removal needs to be more popularized, as it can ensure better results for dogs treated with GFD.

Extracellular vesicles (EVs) derived from stem cells (SCs) have regenerative potential and the possibility of being used in treating chronic diseases. EVs present lower risk of tumorigenicity and easily to isolation and storage. Therefore, this research aims to compare the morphological characteristics of the EVs (up to 150nm) derived from stem cells obtained from canine amniotic membranes in different passages during the in vitro culture. For this, cells from the amniotic membranes were isolated, cultured, and characterized. In order to answer our aim, the number of cells was normalized at each passage to generate conditioned media for EVs separation. The cells were differentiated into adipogenic, chondrogenic, and osteogenic tissue, to characterize these cells as mesenchymal stem cells (MSC). Moreover, flow cytometry analysis was performed and showed that the MSC were positive for CD90, CD105 and negative for CD34, CD45, mesenchymal and hematopoietic markers, respectively. For EVs analysis, MSC in different passages (P0-P2) were culture until 80% of confluence, then the medium was replaced by EVs depleted medium. After 48h, culture medium was collected and centrifuged to separate EVs, followed by nanoparticle tracking analysis. The EVs were also characterized by western blot and transmission electron microscopy (TEM). EVs were positive for Alix and negative for Cytochrome C as well as presented the traditional cup-shape by transmission electronic microscopy. Our results demonstrated that the concentration in the different passages was increased in P0 compared to P1 and P2 (p<0.05). No differences were found in EVs size (P0=132nm, P1=130nm and P2=120nm). Together, these results demonstrate that P0 of MSC is enriched of EVs when compared to later passages, suggesting that this passage would be the best to be applied in pre-clinical tests. Despite that, more studies are necessary to identify the EVs content and how the cells will respond to treatment with them.(AU)

Abstract Three experiments were done with different particle sizes of corn feed on its zootechnical performance, passing rate and apparent digestibility of juvenile tambaqui (Colossoma macropomum). In the first, 200 juveniles were used and distributed in 20 tanks (220 L), 10 fish per unit (four replicates). The experimental system used to record passage time was composed of five incubators with 200 L. In the second, 75 juveniles were used per 55 days (three replications). In the third, 75 juveniles were used and distributed in five incubators of 200 L. All experiments were performed randomly. Different linear behavior treatments were observed for apparent digestibility of crude protein; and the smaller particle size (150 m) had better results digestibility. smaller particle size of the corn had better results, affected the growth performance of tambaqui and the apparent digestibility of crude protein and ether extract. Thus, is recommended that a particle size of 150 m of corn be used for tambaqui.
Resumo Três experimentos foram realizados com diferentes tamanhos de partícula de ração para o desempenho zootécnico, taxa de aprovação e digestibilidade aparente de tambaqui juvenil (Colossoma macropomum). No primeiro, foram utilizados 200 juvenis e distribuídos em 20 tanques (220 L), 10 peixes por unidade (quatro repetições). O sistema experimental utilizado para registrar o tempo de passagem foi composto por cinco incubadoras com 200 L. No segundo, 75 juvenis foram utilizados por 55 dias (três repetições). No terceiro, 75 juvenis foram utilizados e distribuídos em cinco incubadoras de 200 L. Todos os experimentos foram realizados aleatoriamente. Diferentes tratamentos de comportamento linear foram observados para digestibilidade aparente da proteína bruta; e o menor tamanho de partícula (150 m) apresentou melhor digestibilidade dos resultados. menor tamanho de partícula do milho apresentou melhores resultados, afetou o desempenho de crescimento de tambaquis e a digestibilidade aparente da proteína bruta e extrato etéreo. Portanto, recomenda-se o uso de um tamanho de partícula de 150 m de milho para o tambaqui.

Chicken coccidiosis is a common and severe parasitic disease caused by infection from Eimeria spp., which affects the integrity of the intestinal mucosa. TGF-ß has been shown to play an important role in the healing of intestinal mucosas, immunity, and the maintenance of bowel mucosa integrity. Very little is known about the presence of the components of TGF-ß/Smads signaling pathway of chicken at different times following coccidian infection. In the present study, we measured the levels of TGF-ß2, 3, 4, receptor TßRI, II, down-stream Smad 2, 3, 7 in cecum and spleen of chicken at different times after inoculation with Eimeria tenella using quantitative real-time PCR. The results showed that the TGF-ß/Smads signaling pathway was not activated in cecum in the early stage of infection. However, on the 8th day, the expression of TGF-ß2, 4, down-stream protein Smad 2, 7 were significant up-regulated in the spleen, which indicated that the TGF-ß/Smads signaling was changed in the E. tenella infection and was differentially expressed in various tissues in the early stages of infection.(AU)

Three experiments were done with different particle sizes of corn feed on its zootechnical performance, passing rate and apparent digestibility of juvenile tambaqui (Colossoma macropomum). In the first, 200 juveniles were used and distributed in 20 tanks (220 L), 10 fish per unit (four replicates). The experimental system used to record passage time was composed of five incubators with 200 L. In the second, 75 juveniles were used per 55 days (three replications). In the third, 75 juveniles were used and distributed in five incubators of 200 L. All experiments were performed randomly. Different linear behavior treatments were observed for apparent digestibility of crude protein; and the smaller particle size (150 µm) had better results digestibility. smaller particle size of the corn had better results, affected the growth performance of tambaqui and the apparent digestibility of crude protein and ether extract. Thus, is recommended that a particle size of 150 µm of corn be used for tambaqui.

Background: Equine colic syndrome comprises numerous conditions associated with abdominal pain in horses. Impaction, a common cause of this manifestation, is strongly related to these animals' diet. Highly fibrous diets such as sugarcane can predispose horses to colic. The clinical condition can be worsened by fermentative processes, which lead to dysbiosis, circulatory disorders and even endotoxemia. The aim of this study was to report 4 cases of colic syndrome among 8 horses that underwent an experiment to adapt them to a sugarcane-based diet, and to correlate the animals' clinical conditions to the forage they ingested. Cases: Eight male castrated Mangalarga Marchador horses, between 5.5 and 7 years old, were subjected to an experiment to test the feasibility of sugarcane as forage. Four of these horses were taken to the Large Animal Veterinary Hospital (HVGA) of the Federal Rural University of Rio de Janeiro for treatment of abdominal signs of discomfort a few days after the exclusive consumption of sugarcane, in a proportion of 1.75% of live weight in dry matter. The animals' symptoms ranged from behavioral signs indicative of pain to changes in vital parameters and structure of the feces, as well as changes revealed by transrectal palpation. Three of the 4 cases presented impaction in the small colon, and 1 of the horses also presented impaction in the right dorsal colon and rostral displacement of the pelvic flexure, with accumulation of contents in the right ventral colon and sternal flexure. Two of the cases were treated medically, while the other 2 required surgical intervention. The clinical condition of all the patients evolved favorably and they were discharged between 2 and 18 days. Discussion: Colic originating in the digestive system is a syndrome strongly associated with management, especially with respect to confinement, nutrition, and parasite control. During the experiment, 4 of the 8 horses fed with sugarcane presented with colic syndrome. The low quality of sugarcane fiber is due to the high degree of lignification of the plant cell wall, which favors accumulation of ingesta. The poor digestibility and sweet taste of this roughage favor increased consumption. Furthermore, its high sucrose content, associated with an increased rate of passage in the small intestine, alters the intestinal microbiome, and hence, the fermentation byproducts and pH of the ingesta. High intestinal content, allied to longer retention times in the colon and activation of the renin-angiotensin-aldosterone system, promote greater dryness of the ingesta, predisposing the occurrence of impactions in the most distal portion of the large intestine. Intestinal distension and mesenteric traction caused by the accumulation of contents and gases trigger pain, which can worsen due to displacement of the large colon. Small colon impaction, which is easily identified by transrectal palpation, evolves gradually and its treatment, both clinical and surgical, tends to have a favorable prognosis. The need for alternative food sources for horses is a growing demand; however, sugarcane as an exclusive roughage has been shown to be unsafe for horses. The low quality of the fiber and the high sucrose content of this forage can alter the digestive physiology of horses through changes in the passage rate, microbiome and motility of digesta, predisposing them to intestinal dysfunction, ingesta compaction and displacement of the large colon.

ABSTRACT: The application of the intrauterine artificial insemination (IUAI) technique allows optimization of a swine production system due to the reductions in volume and number of sperm cells in the insemination dose, and by reducing the time taken to perform the insemination. However, IUAI is not recommended for gilts due to the difficulty of intrauterine cannula passage through the cervix. This difficulty is associated mainly with the fact that the reproductive tract is smaller in gilts than in pluriparous females. However, few studies have evaluated the application of IUAI in gilts. In these studies, there are variations in approach concerning the definition of the success rate for cannula passage through the cervix, the type of cannula and the body characteristics of the gilts used, making it difficult to extrapolate the recommendation for the use of IUAI in gilts. Considering the evidence that such characteristics influence or even determine the success of the application of IUAI, there is a necessity for an understanding of the influence of these factors in the improvement and later application of the technique. Gilts represent about 15-20% of the breeding group, and the use of IUAI could optimize the processes of insemination on farms. The approach used in this review highlights the aspects that could aid in structuring further studies for improving IUAI in gilts, allowing its use on commercial farms.
RESUMO: A aplicação da técnica de inseminação artificial intrauterina (IAU) permitiu uma otimização do sistema de produção de suínos por possibilitar a redução do volume e número de células espermáticas na dose inseminante e, também, por diminuir o tempo de execução da inseminação. Porém, a IAU não tem sido recomendada para leitoas devido à dificuldade de passagem do cateter intrauterino através da cérvix. Essa dificuldade é associada principalmente ao menor tamanho do trato reprodutivo de leitoas se comparado ao das fêmeas pluríparas. Entretanto, ainda são poucos os estudos que avaliaram a aplicação de IAU em leitoas. Nesses estudos, existem variações quanto à definição da taxa de sucesso na inserção do cateter através da cérvix, ao tipo de cateter e, também, quanto às características corporais das matrizes utilizadas, dificultando extrapolações de recomendação do uso da IAU em leitoas. Considerando os indícios de que tais características podem influenciar ou, até mesmo, determinar o sucesso de aplicação da técnica, ainda há necessidade de compreender a influência desses fatores para que a técnica possa ser aprimorada e posteriormente aplicada. Leitoas representam cerca de 15 a 20% do grupo de cobertura e viabilizar a IAU nessa categoria pode otimizar os processos de inseminação nas granjas. A abordagem realizada nessa revisão traz aspectos que podem auxiliar na estruturação de futuros estudos para aprimorar a IAU em leitoas e permitir seu emprego em granjas comerciais.
Resumo
To evaluate the bromatological composition and ruminal degradability of dry matter (DM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF) of Xaraés palisade grass (Urochloa brizantha Xaraes syn Brachiaria brizantha) under grazing in integrated crop, livestock (ICL), and forest (ICLF) systems, we conducted an in situ degradability trial in randomized blocks with three non-lactating 3/4 Gyr × 1/4 Holstein cows, provided with ruminal cannula. The management of Xaraés palisade grass was similar in both systems, differing only regarding shading in the ICLF system provided by eucalyptus trees (average 65% crown cover). Grass samples were incubated for 0, 3, 6, 9, 12, 24, 36, 48, 72, and 96 hours. Considering the passage rate 2% h-1, the Xaraés palisade grass of ICL system had greater NDF effective degradability in relation to ICLF (46.38% vs 44.98%). However, the palisade grass CP potential degradability was greater in the ICLF than in the ICL system (68.92% vs. 65.40%). The presence of trees in the pasture has effect on nutritional traits of the Xaraés palisade grass, increasing its protein content and degradability and reducing its fiber degradability.

Arsenic (As) is a naturally occurring chemical element considered toxic and carcinogenic by health and environmental protection agencies. Studies of As adsorption/desorption behavior in soils are important to predictions of As potential mobility in natural systems. The aim of this study was to assess the adsorption of As(V) in soils from Minas Gerais, Brazil, and determine its immobilization rate in order to identify soils with characteristics more favorable to its deployment as an As geochemical barrier. The adsorption experiment was performed using different As concentrations and the data pertaining to the maximum adsorption capacity of As(V) (MACAs) were determined by Langmuir and Freundlich isoterms. The Oxisols, due to their more oxidic mineralogy, especially more gibbsitic, and clayey texture, showed the highest MACAs, followed by Ultisols, Inceptisols, and Entisols. In terms of the desorption of As the Inceptisols were the soils that showed the most As desorption. Both As desorption and mobility was lower in the more oxidic and clayey soils. In all soils, the total amount of As was desorbed in due course, but the As release ratio tended to decrease with the passage of time. In general, soils with higher MACAs did not necessarily show less As desorption. For use as a geochemical barrier, as important as a high adsorption capacity of As by the soil is a low As desorption rate. The increase in As mobility may increase the risks of contaminating the supplies of water. To be a good As geochemical barrier the soil has to be a clayey Oxisol, with relatively high amounts of Fe and Al oxides, especially gibbsite.

This study aimed to evaluate the effect of substituting corn with wheat middling on the in-situ degradability of foods and ruminal parameters of cattle. The experiment was conducted at the Ruminant Studies Unit and the Laboratory of Food Analysis and Animal Nutrition of Londrina State University. Five rumen-fistulated cattle with an average weight of 521 ± 38.22 kg were housed in individual stalls. The experimental design used was a Latin square with five treatments (100% C, 75% C 25% WM, 50% C 50% WM, 25% C 75% WM, and 100% WM) and five periods of 25 days each. The pH and ammoniacal nitrogen (N-NH3) concentration in ruminal fluid was measured before feeding and 2, 4, 6, and 8 h after feeding in subdivided parcels. The in-situ degradability of dry matter (DM), organic matter (OM), crude protein (CP), neutral detergent fiber (NDF), and acid detergent fiber (ADF) were determined by incubating silage samples and measuring concentrates in the rumen of cattle. The levels of substitution of corn by wheat middling did not alter the pH and N-NH3 values but was influenced by the collection time. The N-NH3 presented quadratic behavior with a point of maximum rumen concentration (5.31 mgdL-¹) reached 1.12 h after feeding. The pH presented quadratic behavior with a point of minimum (6.04) reached 4.32 h after feeding. The effective degradability (ED) of DM with 2, 5 and 8%h-¹ of passage rate was influenced by the substitution of corn by wheat middling, with increasing linear behavior. The ED 2%h-¹ varied from 46.19% to 50.24%, the ED 5%h-¹ varied from 33.76% to 38.68%, and the ED 8%h-¹ values ranged from 28.16% to 32.92% for the 100% C and 100% WM treatments. Higher values, for treatments with higher levels of substitution of corn by wheat middling, occurred owing to the lower fiber content and to the largest soluble fractions and degradation rate of DM in rations with wheat middling. Objetivou-se com este estudo avaliar o efeito da substituição do milho por triguilho sobre a degradabilidade in situ das rações e os parâmetros ruminais de bovinos. O experimento foi conduzido na Unidade de Estudos de Ruminantes (UNER) e no Laboratório de Análises de Alimentos e Nutrição Animal (LANA) da Universidade Estadual de Londrina. Foram utilizados cinco bovinos fistulados no rúmen, com peso médio de 521 ± 38,22 kg, alojados em baias individuais. O delineamento experimental utilizado foi o quadrado latino, com cinco tratamentos, sendo as proporções de milho e triguilho (100% M, 75% M 25% T, 50% M, 50% T, 25% M 75% T e 100% T) e cinco períodos de 25 dias cada. O pH e nitrogênio amoniacal (N-NH3) do líquido ruminal foram mensurados antes da alimentação e 2, 4, 6 e 8 horas após alimentação, em parcelas subdivididas. A degradabilidade in situ da matéria seca (MS), matéria orgânica (MO), proteína bruta (PB), fibra em detergente neutro (FDN) e fibra em detergente ácido (FDA) foram determinadas com a incubação das amostras de silagem e concentrados no rúmen dos animais. Os teores de substituição do milho pelo triguilho não influenciaram os valores de N-NH3 e pH ruminal, mas os mesmos foram influenciados pelos tempos de coleta. O N-NH3 apresentou comportamento quadrático com ponto de máxima concentração no rúmen (5,31 mgdL-¹) 1h12 após a alimentação. O pH apresentou comportamento quadrático com ponto de mínima (6,04) 4h32 após a alimentação dos animais. A degradabilidade efetiva (DE) da MS com 2, 5 e 8%h-¹ de taxa de passagem foram influenciadas pela substituição do milho por triguilho, observando comportamento linear crescente. A DE 2%h-¹ apresentou variação de 46,19% a 50,24%, a DE 5%h-1 variou de 33,76% a 38,68% e a DE 8%h-¹ apresentou valores de 28,16% a 32,92% para os tratamentos 100% M e 100% T. The objective of this study was to evaluate the ruminal degradability of dry matter and crude protein of high moisture triticale silage ensiled with different chemical and biological additives. Urea, sodium benzoate and an enzyme-bacterial inoculant were used as treatments. Four samples from each treatment were incubated in rumen on four sheep. Effective degradability was estimated for ruminal passage rateof 2%, 5% and 8% hour-¹. Bayesian procedures were used to estimate potential degradation parameters in situ. The high moisture triticale silage with urea showed highest value for the soluble fraction (70.46%) and the best effective dry matter degradability, with a passing rate of 2% h-¹ (90.63%), of control silage at other rates of passage. In relation to control silage, the addition of sodium benzoate and enzyme-bacterial inoculant decreased the effective degradability of dry matter, regardless of rate passage evaluated. Due to high solubility of urea, the silage added with this additive had the highest soluble fraction of crude protein (76.42%). The addition of enzyme-bacterial inoculant accelerated the ruminal passage rate of dry matter and protein to 0.26 and 0.20% h-¹, respectively, providing less potential degradability of both in relation to other silages. As enzyme-bacterial inoculation reduces rumen degradability of crude protein, it tends to increase the availability of amino acids for intestinal absorption. The addition of urea to high moisture triticale silage may be recommended for sheep feeding at a low level of consumption, as it improves the effective dry matter degradability.
O objetivo do trabalho foi avaliar a degradabilidade ruminal da matéria seca e da proteína bruta de silagens de grãos úmidos de triticale ensilados com diferentes aditivos químicos e biológico. Ureia, benzoato de sódio e um inoculante enzimo-bacteriano foram utilizados como tratamentos. Quatro amostras de cada tratamento foram incubadas in situ no rúmen de quatro ovinos. A degradabilidade efetiva foi estimada para taxa de passagem ruminal de 2%, 5% e 8% hora-¹. Foi utilizado procedimento Bayesiano para estimar parâmetros de degradação potencial in situ. A silagem de grãos úmidos de triticale com ureia apresentou o maior valor para a fração solúvel (70,46%) e melhor degradabilidade efetiva da matéria seca, em taxa de passagem de 2% h-¹ (90,63%), não diferindo da silagem controle nas demais taxas de passagens. Em relação a silagem controle, a adição de benzoato de sódio e de inoculante enzimo-bacteriano diminuíram a degradabilidade efetiva da matéria seca, independente da taxa de passagem avaliada. Devido à alta solubilidade da ureia, a silagem acrescentada com esse aditivo, apresentou a maior fração solúvel da proteína bruta (76,42%). A adição de inoculante enzimo-bacteriano acelerou a taxa de passagem ruminal da matéria seca e da proteína para 0,26 e 0,20% h-1,respectivamente, proporcionando menor degradabilidade potencial de ambos em relação às demais silagens. Como a inoculação enzimo-bacteriana reduz a degradabilidade ruminal da proteína bruta, tende-se a aumentar a disponibilidade de aminoácidos para absorção intestinal. A adição de ureia em silagens de grãos úmidos de triticale pode ser recomendada para alimentação de ovinos em baixo nível de consumo, por melhorar a degradabilidade efetiva da matéria seca.

The aim of this study was to evaluate the ruminal fermentation and degradation parameters, the kinetic flow of digesta in the gastrointestinal tract, and the milk fatty acid (FA) profile in Holstein x Gyr cows fed 59% sugarcane silage-based diets with 0% (control), 5%, 10% and 15% whole cottonseed (WCS) on a dry matter (DM) basis. Four rumen-cannulated cows with an average milk yield of 14.4±3.3 kg day-1 and 85±25 days in milk were allocated in a 4 x 4 Latin square design. There was no effect of dietary WCS levels on the intake of organic matter (OM), crude protein (CP), neutral detergent fiber (NDF) and nonfibrous carbohydrates, but there was a linear increase in the intake of ether extract (EE). There was no treatment effect on the apparent digestibility of DM, OM, CP, EE and NDF or on the ruminal degradability parameters for DM and NDF of sugarcane silage. Also, no treatment effects were observed on rumen pH, ruminal and postruminal particulate passage rates or on the rate of passage of the fluids in the rumen. WCS promoted linear reductions in the milk fat contents of lauric, myristic and palmitic acids and linear increases in the milk fat contents of stearic, oleic, trans-10 C18:1 and elaidic acids. The milk fat contents of vaccenic, rumenic, linoleic and α-linolenic acids were unaffected by WCS supplementation. The inclusion of up to 15% WCS in sugarcane silage-based diets did not alter the digestibility of nutrients, the rumen degradability of the fiber, or the kinetic flow of digesta in the gastrointestinal tract of Holstein x Gyr cows but improved the nutritional quality of milk fat through an increase in the content of oleic acid, which is beneficial to human health, and decreased levels of hypercholesterolemic lauric, myristic and palmitic acids.(AU)
O objetivo deste estudo foi avaliar os parâmetros de fermentação e degradação ruminal, a cinética de fluxo da digesta no trato gastrintestinal, e o perfil de ácidos graxos do leite de vacas Holandês x Gir alimentadas com dietas à base de 59% de silagem de cana de açúcar com 0% (controle), 5%, 10% e 15% de caroço de algodão na base de matéria seca (MS). Quatro vacas canuladas no rúmen, com produção de leite de 14,4±3,3 kg dia-1 e 85±25 dias em lactação foram alocadas em delineamento quadrado latino 4 x 4. Não houve efeito da inclusão do caroço de algodão sobre os consumos de matéria orgânica (MO), proteína bruta (PB), fibra em detergente neutro (FDN) e de carboidratos não fibrosos, mas houve aumento linear no consumo de extrato etéreo (EE). Não houve efeito dos tratamentos sobre a digestibilidade da MS, MO, PB, EE e FDN nem sobre os parâmetros de degradabilidade ruminal da MS e FDN da silagem de cana de açúcar. Não houve efeito dos tratamentos sobre o pH ruminal, taxas de passagem de fluidos e partículas no rúmen, nem sobre a taxa de passagem pós-ruminal de partículas. Houve reduções lineares nos teores dos ácidos láurico, mirístico e palmítico, e aumentos lineares nos teores dos ácidos esteárico, oleico, C18:1 trans-10 e elaídico no leite. A inclusão de caroço de algodão não alterou os teores dos ácidos vacênico, rumênico, linoleico e α-linolênico no leite. A inclusão de até 15% de caroço de algodão em dietas à base de silagem de cana de açúcar não alterou a digestibilidade dos nutrientes, a degradabilidade ruminal da fibra, nem a cinética de fluxo da digesta no trato gastrintestinal de vacas Holandês x Gir, mas melhorou a qualidade nutricional da gordura do leite, com aumento no teor de ácido oleico, benéfico para a saúde humana, e redução nos teores dos ácidos hipercolesterolêmicos láurico, mirístico e palmítico.(AU)

The objective was to evaluate the rumen degradability of dry matter, crude protein and fiber fraction of some oilseed byproducts. Oilseed byproducts evaluated were: Sunflower Meal, Cotton Cake and Castor Bean Cake. The rumen degradability test was performed following a split plot randomized complete block design with three treatments (plots) and seven incubation times (subplots) and six blocks (animals). The readily soluble fraction of the dry matter of sunflower meal was 35.0% higher than the same fraction of cotton cake and castor bean cake (mean 24.46%, P < 0.01). The highest potential degradability of crude protein (P < 0.01) was verified in castor bean cake (96.90%, 5% rate of passage). Sunflower meal had the lowest insoluble fraction of the fiber fraction, 34.4%, compared to cotton cake and castor bean cake, with 64.4% and 39.4%, respectively (P < 0.01). Sunflower meal had the highest fiber effective degradability (25.2%). Sunflower meal presented better values of ruminal parameters of dry matter, crude protein and neutral detergent fiber in relation to the other byproducts.
Objetivou-se avaliar a degradabilidade ruminal da matéria seca, proteína bruta e fração fibrosa de alguns coprodutos de oleaginosas. Os coprodutos de oleaginosas avaliados foram: Farelo de Girassol, Torta de Algodão e Torta de Mamona. O ensaio de degradabilidade ruminal foi conduzido seguindo o delineamento em blocos casualizados em esquema de parcelas subdivididas com três tratamentos (parcelas) e sete tempos de incubações (subparcelas) e seis blocos (animais. A fração prontamente solúvel da matéria seca do farelo de girassol foi 35,0 % superior em relação à mesma fração dos coprodutos torta de algodão e torta de mamona (média 24,46%; P < 0,01). A maior degradabilidade potencial da proteína bruta (P < 0,01) foi verificada na torta de mamona (96,90%; taxa de passagem de 5%). O farelo de girassol apresentou menor fração insolúvel potencialmente degradável da fração fibrosa, 34,4%, em relação à torta de algodão e torta de mamona que apresentaram média de 64,4% e 39,4%, respectivamente (P < 0,01). O farelo de girassol apresentou maior degradabilidade efetiva da fibra (25,2%). O farelo de girassol apresentou melhores valores dos parâmetros ruminais da matéria seca, proteína bruta e fibra em detergente neutro em relação aos demais coprodutos.
