The telencephalon refers to the most highly developed and anterior part of the forebrain, consisting mainly of the cerebral hemispheres. The study determined Neuroglobin (Ngb) and Hypoxia-inducible factor (HIF-1α) expression in the telencephalon of yak and cattle, and compare the expression and distribution pattern of Ngb and HIF-1α in the two animals. Immunohistochemistry (IHC), quantitative real-time Polymerase Chain Reaction (qRT-PCR), and Western blot (WB) were employed to investigate Ngb and Hif-1α expression in the telencephalon of yak and cattle. mRNA and protein expressions of Ngb and HIF-1α showed positive in different tissues of the yak and cattle telencephalon. Ngb expression in tissues of the yak recorded higher as compare to cattle while HIF-1α expression was found higher in cattle than yak. The HIF-1α expression in some tissues of yak telencephalon was consistent with the cattle. The results documented that HIF-1α may have a direct or indirect synergistic effect on Ngb expression in the yak telencephalon to improve hypoxia adaptation. It is suggested that yak may need more Ngb expression for adaptation, but the expression of HIF-1α seems to be down-regulated during long-term adaptation, and the specific causes of this phenomenon needs to be further verified.
O telencéfalo refere-se à parte anterior e mais desenvolvida do prosencéfalo, consistindo principalmente dos hemisférios cerebrais. O estudo determinou a expressão de neuroglobina (Ngb) e fator indutível por hipóxia (HIF-1α) no telencéfalo de iaques e bovinos e comparou a expressão e o padrão de distribuição de Ngb e HIF-1α nos dois animais. Imuno-histoquímica (IHC), reação em cadeia da polimerase quantitativa em tempo real (qRT-PCR) e Western blot (WB) foram empregados para investigar a expressão de Ngb e Hif-1α no telencéfalo de iaques e bovinos. As expressões de mRNA e proteínas de Ngb e HIF-1α mostraram-se positivas em diferentes tecidos do telencéfalo de iaque e bovino. A expressão de Ngb nos tecidos do iaque foi registrada mais alta em comparação com o gado, enquanto a expressão do HIF-1α foi encontrada mais alta no gado do que no iaque. A expressão de HIF-1α em alguns tecidos do telencéfalo de iaque foi consistente com o gado. Os resultados documentaram que o HIF-1α pode ter um efeito sinérgico direto ou indireto na expressão de Ngb no telencéfalo de iaque para melhorar a adaptação à hipóxia. É sugerido que o iaque pode precisar de mais expressão de Ngb para adaptação, mas a expressão de HIF-1α parece ser regulada para baixo durante a adaptação de longo prazo, e as causas específicas desse fenômeno precisam ser verificadas.

The aim of this study was to investigate the association between chronic Anaplasma marginale and Babesia spp. infection and hematological parameters of pregnant and non-pregnant taurine heifers. Blood samples from 94 females were collected on the first day (D-10) of timed artificial insemination (TAI) protocol and on pregnancy diagnosis (D+34). Hematological parameters were determined and compared between pregnant (PG) and non-pregnant (NPG) heifers, and within group at different sampling days. Real-time PCR (qPCR) was used to determine A. marginale and Babesia bovis infection, and for absolute quantification of Babesia spp. between PG and NPG groups. Correlation analysis was performed between the number of gDNA copies (CN) of Babesia spp. and hematological parameters. On D-10, mean hemoglobin concentration was higher for NPG, and hematocrit and total plasma protein were higher on D+34 for both groups. There was no difference in Babesia spp. CN between groups. In the first qPCR, all heifers were positive for A. marginale and B. bovis. Significant correlations were found between hemoglobin and erythrocyte and between hemoglobin and hematocrit (r = 0.8082 and r = 0.3009, respectively). Low levels of A. marginale and Babesia spp. did not affect hematological parameters of chronically infected pregnant and non-pregnant taurine heifers.(AU)
O objetivo deste estudo foi investigar a associação entre infecção crônica por Anaplasma marginale e Babesia spp. e parâmetros hematológicos de novilhas taurinas prenhes e não prenhes. Sangue de 94 fêmeas foi coletado no primeiro dia (D-10) do protocolo de inseminação artificial em tempo fixo (IATF) e no diagnóstico de gestação (D+34). Parâmetros hematológicos foram comparados entre novilhas prenhes (PG) e não prenhes (NPG) e dentro dos grupos entre dias de coleta. Usando-se PCR em tempo real (qPCR), determinou-se a infecção por A. marginale e Babesia bovis e quantificação absoluta de Babesia spp. Entre os grupos PG e NPG. A análise de correlação foi realizada entre o número de cópias (CN) de Babesia spp. e parâmetros hematológicos. No D-10, a concentração de hemoglobina foi maior para NPG e hematócrito, e proteína plasmática total foram maiores em D+34 para ambos os grupos. Não houve diferença para CN de Babesia spp. entre os grupos. Na primeira qPCR, todas as novilhas foram positivas para A. marginale e B. bovis. Correlações significativas foram encontradas entre hemoglobina/eritrócito e hemoglobina/hematócrito (r=0,8082 e r=0,3009, respectivamente). Baixos níveis de A. marginale e Babesia spp. não afetaram os parâmetros hematológicos de novilhas taurinas prenhes e não prenhes cronicamente infectadas.(AU)

Background: Dermatophytes, fungi of universal distribution, invade semi or fully keratinized structures, such as skin, fur/ hair and nails. The various species of dermatophytes are classified into three genera anamorphic: Microsporum, Trichophyton and Epidermophyton. The genus Epidermophyton includes only E. floccosum, that rarely affects animals. The main species responsible for the disease in dogs and cats are Microsporum canis, M. gypseum and Trichophyton mentagrophytes, which were characterized through conventional mycological methodology (microscopic examination with KOH and culture). Molecular methodologies, such as real-time PCR, can contribute to a rapid laboratory diagnosis, helping clinicians to initiate an early antifungal treatment. This case report describes a case of canine dermatophytosis due to Trichophyton mentagrophytes detected from a clinical sample by SYBR-Green real-time PCR. Case: A 8-year-old dog, rescued from the street, was referred to a private veterinary clinic in the city of Canoas, RS, Brazil, presenting generalized lymphadenomegaly, crusted lesions all over the body, generalized alopecia, signs of excoriation and epistaxis. Initially, were administered prednisone [1 mg/kg every 48 h, BID] and cephalexin [30 mg/kg, BID]. Weekly baths with benzoyl peroxide were also given. The therapy was not clinically successful. Wood's Lamp Test was negative. As a differential diagnosis, PCR for detection of Leishmania was negative. Complete blood count and serum biochemical assay were also performed. For mycological diagnosis, hair specimen was clarified and examined microscopically using 10% potassium hydroxide (KOH) for the visualization of chains of arthroconidia (ectothrix invasion of hair). The infected hair was plated onto MycoselTM Agar, incubated at 28°C for 15 days. Microscopy of hyphae/ conidia and macroscopic colony characteristics (colors and texture) were conducted for the differentiation of the species within the genus Microsporum and Trichophyton. In addition, real-time PCR was applied for direct analysis of the fungal DNA obtained from the hair sample. Microscopic examination was negative. The dermatophyte present in the hair sample was confirmed as Trichophyton mentagrophytes by culture and qPCR (melting-point analysis). The patient was treated with systemic itraconazole [10 mg/ kg SID - 90 days]. Twice-weekly application of 2.5 % miconazole and 2% chlorhexidine shampoo until complete cure. Discussion: Dermatophytosis is often listed as self-limiting infection; however, animal dermatophytosis can spread between pets, as well as a zoonotic transmission to humans. The literature on dermatophytosis indicates that Microsporum canis is the predominant etiological agent, followed by M. gypseum. Trichophyon mentagrophytes that appear in a lower percentage of isolation. The culture of hair, even with specific medium containing chloramphenicol and cyclohexamide, may present contaminating fungi, not related to dermatophytosis, which can inhibit or override the growth of dermatophytes. The use of real-time PCR provided a faster and specific diagnosis of dermatophytosis when compared to the conventional mycological methodology for detection and identification of T. mentagrophytes, which takes around 10 to 15 days for culture. It is possible to use this technique as an alternative diagnosis for dermatophytes associated to clinical hair samples of dogs.

Porcine hemoplasmosis is characterized as a geographically cosmopolitan disease caused by Mycoplasma suis and Mycoplasma parvum. Asymptomatic pigs are considered the focus of hemoplasmosis because they are carriers and reservoirs to new infections. This study aimed to determine the molecular occurrence of porcine hemoplasmas (PH) in the production cycle of technified farrow-to-finished swine herds. For this purpose, 20 swine herds were evaluated, where 501 whole blood samples were collected for qPCR and phylogenetic analyses for hemoplasmas. The epidemiological analysis was performed for the entire population and per the growth stage. The total prevalence for PH was 31.93% (161/501); 95% (19/20) of sampled herds were positive. The occurrence of PH by swine growth stages was nursery (30.47%), growing (31.29%), finishing (26.18%), and slaughter (40.25%). The quantification cycles (Cq) ranged from 3.18-39.56 and the number of PH 16S rRNA copies per µL of DNA ranged from 5,57 x10 to 2.23 x1010 . Sequencing and phylogenetic analysis of five selected samples showed 100% identity with M. parvum strain Indiana and two M. parvum sequences from Brazil/Goiás. This is the first report on PH in technified herds in Southeastern Brazil by growth stages.
A hemoplasmose suína é uma doença geograficamente cosmopolita, causada por Mycoplasma suis e Mycoplasma parvum. Suínos assintomáticos são considerados foco de hemoplasmose por serem portadores e reservatórios de novas infecções. Este estudo teve como objetivo determinar a ocorrência molecular de hemoplasmas suínos (HP) no ciclo de produção de rebanhos suínos tecnificados. Foram avaliados 20 rebanhos suínos e coletadas 501 amostras de sangue total para qPCR e análises filogenéticas para hemoplasmas. A análise epidemiológica foi realizada pela população e por estágio de crescimento. A prevalência total de HP foi de 31,93% (161/501); 95% (19/20) dos rebanhos amostrados foram positivos. A ocorrência de HP por fases de crescimento dos suínos foi: creche (30,47%), em crescimento (31,29%), acabamento (26,18%) e abate (40,25%). Os ciclos de quantificação (Cq) variaram de 3,18-39,56 e o número de cópias de rRNA PH 16S por µL de DNA variou de 5,57 x102 a 2,23 x10¹0. O sequenciamento e a análise filogenética de cinco amostras selecionadas mostraram 100% de identidade com a cepa indiana de M. parvum e duas sequências de M. parvum do Brasil / Goiás. Este é o primeiro relato de HP em rebanhos tecnificados, na região Sudeste do Brasil, por estágios de crescimento.

ABSTRACT: This research aimed to morphologically characterize and estimate the genetic diversity of 21 Capsicum accessions belonging to the Capsicum Germplasm Active Bank at the Universidade Federal do Piauí (BAGC-UFPI) using uni- and multivariate analysis. The experiment was carried out in a greenhouse, by completely randomized experimental design with four repetitions, with one plant per plot. Analysis of variance (ANOVA) and the comparison of means for seven quantitative variables were performed, followed by clustering the averages by the Scott-Knott test (P < 0.05). The analysis of the seven quantitative and thirteen qualitative descriptors was estimated based on the Gower distance. Later, it was performed the principal component analysis and the UPGMA hierarchical cluster method. Results characterized and identified a wide intra- and interspecific genetic variability related to the fruit size, colors, and shapes among the Brazilian Capsicum genotypes belonging to the BAGC-UFPI. The descriptors used in this research were effective in the discrimination of the pepper accessions, especially the closely related C. frutescens and C. chinense species.
RESUMO: Este trabalho teve como objetivo caracterizar e estimar a diversidade genética em 21 acessos de Capsicum pertencentes ao Banco Ativo de Germoplasma da Universidade Federal do Piauí (BAGC-UFPI), por análises uni e multivariadas. O experimento foi conduzido em telado, utilizando-se o delineamento inteiramente ao acaso, com quatro repetições, sendo uma planta por parcela. Realizou-se análise de variância (ANOVA) e comparação da média para as setes variáveis quantitativas, seguidas do agrupamentos de médias pelo teste Scott-Knott (P < 0,05). A análise dos sete descritores quantitativos e treze qualitativos foi estimada com base na distância de Gower. Posteriormente, foi realizada a análise de componentes principais e o método de agrupamento hierárquico UPGMA. Os resultados caracterizaram e identificaram uma ampla variabilidade genética intra e interespecífica relacionada ao tamanho, cor e formato dos frutos entre os genótipos brasileiros de Capsicum do banco de germoplasma de pimentas BAGC-UFPI. Os descritores utilizados foram eficientes na discriminação dos acessos de pimentas, especialmente para espécies proximamente relacionadas C. frutescens e C. chinense.

The Rufous treepie (Dendrocitta vagabunda) belongs to family corvidae, order Passeriformes which includes about 100 species. The current study was conducted to gather information about the Population distribution and habitat analysis of D. vagabunda at District Abbottabad, Pakistan. The data were collected on monthly basis both morning and evening times (2018-2019). The "Point count Method" was used for population estimation and "Quadrates Method" for habitat analysis of study area. The result shows an average month-wise population density of D. vagabunda was maximum at Jhangra 0.14±0.039/ha, whereas minimum at Havelian 0.11±0.022/ha. There was no significant difference (p>0.05) among monthly population densities of D. vagabunda, however, a significant difference (p<0.05) was found between morning and evening times population of the specie. The present study revealed that importance value index (IVI) of plants species at Sherwan, Bakot, Havelian, Langra and Jhangra were 59.6±12.6, 50.1±6.9, 53.4±6.3, 66.8±10 and 60.1±7.7. Likewise, the frequency of shrubs at Sherwan, Bakot, Havelian, Langra and Jhangra were 33.3±4.2, 45±9.4, 46.7±8.2, 55.6±22.2 and 37.5±8.5. Similarly, the frequency of herbs at Sherwan, Bakot, Havelian, Langra and Jhangra were 40.4±6.0, 37.5±5.6, 53.3±7.4, 48.5±5.2 and 46.9±7.4 respectively. Our results show the study area as suitable habitat for D. vagabunda.

Abstract Croton argyrophylloides Muell. Arg., from the Euphorbiaceae family, popularly known as marmeleiro prateado or sacatinga, is a plant from the Caatinga biome commonly found in Brazils northeastern region. The present study aimed to evaluate the antioxidant activity of the species. The phytochemical study was performed through qualitative analysis of chemical constituents and quantitative determination of the total phenol content through the Folin-Ciocalteu test. The qualitative and quantitative antioxidant tests were performed using the DPPH method (2.2 diphenyl-1-picryl hydrazil) and ferric reducing antioxidant power (FRAP). The minimum inhibitory concentration (MIC) was determined by microdilution in 96-well plates. The ethanolic extract of the leaves of C. argyrophylloides manifested antioxidant action in the quantitative DPPH test with a significant bioactivity of 84.70 AAO% in 500 µg/mL, with an EC50 of 236.79. The content of total phenolic compounds was 946.06 mg of gallic acid equivalents/g of sample, and total flavonoids was 58.11 mg of quercetin equivalents/g of sample, the result obtained for FRAP was 15294.44 µM Trolox/g of sample and ABTS was 718 M Trolox of sample. The prospecting of the chemical constituents of the leaves of C. argyrophylloides revealed the presence of the main compounds that manifests the antioxidant activity and it was proven by the DPPH method that there is antioxidant activity in the analyzed sample, in addition to demonstrating a significant content of phenolic compounds and total flavonoid content in the species, which corroborates the antioxidant activity of the plant sample. Resumo Croton argyrophylloides Muell. Arg., pertencente à família Euphorbiaceae, conhecida popularmente como marmeleiro prateado e sacatinga, é um vegetal do bioma caatinga comumente encontrado no Nordeste do Brasil. O presente trabalho teve como objetivo avaliar a atividade antioxidante da espécie. O estudo fitoquímico foi realizado por meio de análise qualitativa dos constituintes químicos e determinação quantitativa do teor de fenóis totais pelo teste de Folin-Ciocalteu. Os testes antioxidantes qualitativos e quantitativos foram realizados pelo método do DPPH (2,2 difenil-1- picril-hidrazila) e redução do ferro (FRAP). A concentração inibitória mínima (CIM) foi determinada por microdiluição em placas de 96 poços. O extrato etanólico das folhas de C. argyrophylloides apresentou ação antioxidante no teste DPPH quantitativo com uma significativa bioatividade de 84.70 AAO% em 500 µg/mL, apresentando um CE50 de 236.79. O teor de compostos fenólicos totais, foi de 946,06 mg equivalentes de ácido gálico/g de amostra, e de flavonoides totais de 58,11 mg equivalentes de quercetina/g da amostra, o valor encontrado para FRAP foi de 15294,44 µM Trolox/g da amostra e de ABTS foi 718 M Trolox da amostra. A prospecção dos constituintes químicos das folhas de C. argyrophylloides revelou a presença dos principais compostos que caracterizam a atividade antioxidante e foi possível comprovar pelo método de DPPH que há atividade antioxidante na amostra analisada, além de demonstrar um resultado significativo de teor de compostos fenólicos e teor de flavonoides totais na espécie e o que corrobora com a atividade antioxidante da amostra vegetal. Os extratos das folhas apresentaram halos de inibição de crescimento de 10 e 12mm frente a Staphylococcus aureus ATCC 25923.
Resumo
Regression analysis is highly relevant to agricultural sciences since many of the factors studied are quantitative. Researchers have generally used polynomial models to explain their experimental results, mainly because much of the existing software perform this analysis and a lack of knowledge of other models. On the other hand, many of the natural phenomena do not present such behavior; nevertheless, the use of non-linear models is costly and requires advanced knowledge of language programming such as R. Thus, this work presents several regression models found in scientific studies, implementing them in the form of an R package called AgroReg. The package comprises 44 analysis functions with 66 regression models such as polynomial, non-parametric (loess), segmented, logistic, exponential, and logarithmic, among others. The functions provide the coefficient of determination (R2), model coefficients and the respective p-values from the t-test, root mean square error (RMSE), Akaike's information criterion (AIC), Bayesian information criterion (BIC), maximum and minimum predicted values, and the regression plot. Furthermore, other measures of model quality and graphical analysis of residuals are also included. The package can be downloaded from the CRAN repository using the command: install.packages("AgroReg"). AgroReg is a promising analysis tool in agricultural research on account of its user-friendly and straightforward functions that allow for fast and efficient data processing with greater reliability and relevant information.

The prospecting of the chemical constituents of the leaves of C. argyrophylloides revealed the presence of the main compounds that manifests the antioxidant activity and it was proven by the DPPH method that there is antioxidant activity in the analyzed sample, in addition to demonstrating a significant content of phenolic compounds and total flavonoid content in the species, which corroborates the antioxidant activity of the plant sample. Single nucleotide polymorphisms (SNPs, rs12979860 e rs8099917) in the Interferon Lambda 4 gene (IFNL4, formerly IFNL3and/or IL28B) has been associated with failure in the innate immune response, sustained virological response in hepatitis C, and HTLV-1-associated myelopathy (HAM) development. To search for these polymorphisms several methodologies can be employed, such as sequencing, real-time or quantitative polymerase chain reaction (qPCR), restriction fragment length polymorphism analysis in PCR products (PCR-RFLP), and tetra-primer PCR. The present study compared the performance of the tetra-primer PCR in relation to the PCR-RFLP, both optimized in the Research HTLV Laboratory of the Center of Immunology of Instituto Adolfo Lutz in São Paulo. One hundred DNA samples obtained from patients of STD/Aids Reference Centre in São Paulo, previously analyzed for IL28B SNPs by PCR-RFLP were selected for analysis, after confirming that they represent all IL28B SNPs patterns described in the literature. The results obtained showed concordance between the PCR-RFLP and the tetra-primer PCR SNPs results, and because of the low cost, easy to perform, and minor employment of biological specimen and reagents, the tetra-primer PCR is of choice to be used in routine. Polimorfismos de nucleotídeos únicos (single nucleotide polymorphisms, SNPs rs12979860 e rs8099917) no gene que codifica o Interferon Lambda 4 (IFNL4, antigamente IFNL3 e/ou IL28B) têm sido associados às falhas na resposta imune inata e resposta virológica sustentada na hepatite C, e a mielopatia associada ao HTLV-1 (HTLV-1-associated myelopathy, HAM). A pesquisa destes polimorfismos pode empregar diversas metodologias: sequenciamento, reação em cadeia da polimerase em tempo real ou quantitativa (quantitative polymerase chain reaction, qPCR), análise de fragmentos de restrição enzimática em produtos de PCR (restriction fragment length polymorphism in PCR products, PCR-RFLP) e a tetra-primer PCR. Este estudo comparou o desempenho da tetra-primer PCR em relação a PCR-RFLP, ambas otimizadas no Laboratório de Pesquisa em HTLV do Centro de Imunologia do Instituto Adolfo Lutz de São Paulo. Foram selecionadas 100 amostras de DNA obtidas de pacientes do Centro de Referência e Treinamento em DST/Aids de São Paulo cujos SNPs na IL28B foram anteriormente determinados por PCR-RFLP e representaram todos os perfis descritos em literatura. Os resultados obtidos mostraram concordância entre elas, e pelo fato da tetra-primer PCR ter menor custo, ser de fácil execução, empregar menos tempo, insumos e material biológico, é a técnica de escolha para uso em rotina. This research aimed to morphologically characterize and estimate the genetic diversity of 21 Capsicum accessions belonging to the Capsicum Germplasm Active Bank at the Universidade Federal do Piauí (BAGC-UFPI) using uni- and multivariate analysis. The experiment was carried out in a greenhouse, by completely randomized experimental design with four repetitions, with one plant per plot. Analysis of variance (ANOVA) and the comparison of means for seven quantitative variables were performed, followed by clustering the averages by the Scott-Knott test (P < 0.05). The analysis of the seven quantitative and thirteen qualitative descriptors was estimated based on the Gower distance. Later, it was performed the principal component analysis and the UPGMA hierarchical cluster method. Results characterized and identified a wide intra- and interspecific genetic variability related to the fruit size, colors, and shapes among the Brazilian Capsicum genotypes belonging to the BAGC-UFPI. The descriptors used in this research were effective in the discrimination of the pepper accessions, especially the closely related C. frutescens and C. chinense species.
Este trabalho teve como objetivo caracterizar e estimar a diversidade genética em 21 acessos de Capsicum pertencentes ao Banco Ativo de Germoplasma da Universidade Federal do Piauí (BAGC-UFPI), por análises uni e multivariadas. O experimento foi conduzido em telado, utilizando-se o delineamento inteiramente ao acaso, com quatro repetições, sendo uma planta por parcela. Realizou-se análise de variância (ANOVA) e comparação da média para as setes variáveis quantitativas, seguidas do agrupamentos de médias pelo teste Scott-Knott (P < 0,05). A análise dos sete descritores quantitativos e treze qualitativos foi estimada com base na distância de Gower. Posteriormente, foi realizada a análise de componentes principais e o método de agrupamento hierárquico UPGMA. Os resultados caracterizaram e identificaram uma ampla variabilidade genética intra e interespecífica relacionada ao tamanho, cor e formato dos frutos entre os genótipos brasileiros de Capsicum do banco de germoplasma de pimentas BAGC-UFPI. Os descritores utilizados foram eficientes na discriminação dos acessos de pimentas, especialmente para espécies proximamente relacionadas C. frutescens e C. chinense.

Nucleotide-binding oligomerization domain receptors (NOD-like receptors, NLRs) have critical effects on interfaces of the immune and reproductive systems, and the spleen plays a key role in both innate and adaptive immune functions. It is hypothesized that NLR family participates in maternal splenic immune regulation during early pregnancy in sheep. In this study, maternal spleens were collected on day 16 of the estrous cycle, and days 13, 16 and 25 of gestation (n = 6 for each group) in ewes. Expression of NLR family, including NOD1, NOD2, class II transactivator (CIITA), NLR family apoptosis inhibitory protein (NAIP), nucleotide-binding oligomerization domain, Leucine rich repeat and Pyrin domain containing 1 (NLRP1), NLRP3 and NLRP7, was analyzed using quantitative real-time PCR, Western blot and immunohistochemistry analysis. The results revealed that expression levels of NOD1, NOD2, CIITA and NLRP3 were downregulated at days 13 and 16 of pregnancy, but expression of NLRP3 was increased at day 25 of pregnancy. In addition, expression values of NAIP and NLRP7 mRNA and proteins were improved at days 16 and 25 of pregnancy, and NLRP1 was peaked at days 13 and 16 of pregnancy in the maternal spleen. Furthermore, NOD2 and NLRP7 proteins were limited to the capsule, trabeculae and splenic cords. In summary, early pregnancy changes expression of NLR family in the maternal spleen, which may be related with the maternal splenic immunomodulation during early pregnancy in sheep.(AU)

Purpose: Low-intensity pulsed ultrasound (LIPUS) has been used to stimulate the healing of the fresh fracture, delayed union, and non-union in both animal and clinical studies. Besides, biphasic calcium phosphate ceramic (BCP) is a promising biomaterial for bone repair as it shows favorable biocompatibility, osteoinduction, and osteoconduction. However, scarcity is known about the combined effect of LIPUS and BCP on bone formation. Methods: The combined effect of LIPUS and BCP was studied in a beagle model. Twelve dogs were used. BCP granules without any additions were implanted into bilateral erector spinae muscles. One side is the BCP group, while the counterlateral side is LIPUS + BCP group. Histological and histomorphometric analyses, and quantitative real-time polymerase chain reaction were evaluated. Results: Compared with BCP alone, the LIPUS + BCP showed no advantages in early bone formation. Furthermore, the Notch signaling pathway-related mRNA has no significant difference between the two groups. Conclusions: The preliminary results showed that the BCP, which has intrinsic osteoinduction nature, was an effective and promising material. However, LIPUS has no enhanced effect in BCP induced ectopic bone formation. Furthermore, LIPUS has no effect on the Notch signaling pathway. Whether costly LIPUS could be used in combination with BCP should be a rethink.

Objetivou-se avaliar métodos alternativos de restrição alimentar qualitativa e quantitativa em comparação à técnica convencional de muda forçada em codornas japonesas para o segundo ciclo de produção. O período de muda forçada teve duração de 10 dias e dois períodos de 21 dias de pós-muda. Foram utilizadas 72 codornas japonesas fêmeas, emfinal de produção com 72 semanas deidade. As aves foram distribuídas em delineamento inteiramente casualizado com três tratamentos e quatro repetições cada, com seiscodornas por repetição. Os tratamentos experimentais foram:T1 aves submetidas a três dias de jejum com posterior fornecimento de ração de postura à vontade por setedias; T2 aves que receberam alimentação controlada (10 g/ave/dia) por 10 dias; T3 aves que receberam farelo de arroz à vontade por 10 dias. Durante todo o experimento todasas aves foram submetidas a iluminação natural com média de 12 horas de luz diária e água a vontade e após os 10 dias de muda foi fornecido ração de postura a vontade. A utilização do T2 (10 g/ave/dia de ração de postura) e de farelo de arroz como alimentoalternativo no programa de muda forçada foi viável. Mesmo a perda de peso sendo abaixo do recomentado, as aves que receberam 10 g de ração ave/dia e farelo de arroz à vontade apresentaram resultados de desempenho e qualidade do ovo pós muda semelhantes àsaves que ficaram em jejum, além de possibilitar melhores condições de bem-estar animal.(AU)
The objective was to evaluate alternative methods of qualitative and quantitative feed restriction compared to theconventional technique of forced moulting in Japanese quails for the second production cycle. The forced moulting period lasted 10 days and two 21-day post-molt periods. 72 female Japanese quails were used, at the end of production at 72 weeks of age. Thebirds were distributed in a completely randomized design with three treatments and four replications each, with sixquails per replicate. The experimental treatments were: T1 birds submitted to three days of fasting with subsequent provision of layingfeed ad libitum for sevendays; T2 birds that received controlled feeding (10 g/bird/day) for 10 days; T3 birds that received rice bran ad libitum for 10 days. Throughout the experiment, all birds were submitted tonatural lighting with an average of12hours of daily light and water ad libitum and after 10 days of molting, laying feed was provided ad libitum. The use of T2 (10 g/bird/day of laying ration) and ricebran as an alternativefood in the forced molt program was feasible. Even the weight loss being below the recommended, the birds that received 10 g of poultry feed/day and rice bran ad libitum showed similar post-moult performance and egg quality results to birds that were fasted, in addition to enabling better conditions. of animal welfare.(AU)

The growth differentiation factor 9 (GDF9) gene plays a vital role in the growth and maturation of ovarian follicles in laying hens. However, its messenger ribonucleic acid (mRNA) expression levels in preovulatory ovarian follicles of indigenous chickens remain poorly understood. The study aimed to identify the association between egg weight and egg quality traits, ovarian follicles morphology, and mRNA expression levels of the GDF9 gene in pre-ovulatory ovarian follicles of the South African Potchefstroom Koekoek chicken breed. The correlation results showed that egg weight (EW) had a positively high significant correlation (p<0.01) with egg width (EWD), yolk weight (YW), shell surface area (SSA), albumen weight (AW), albumen ratio (AR) and egg volume (EV), and a positive significant correlation (p<0.05) with egg length (EL). The Student's T-test results revealed that the numbers of large yellow follicles were significantly lower (p<0.05) than those of small yellow follicles. ANOVA findings showed that there was a significant difference (p<0.05) in the average weight of the large yellow follicles. The quantitative Reverse Transcription Polymerase Chain Reaction (RT-qPCR) findings indicated that there were significant differences (p<0.05) in the mRNA expression levels of the GDF9 gene in preovulatory ovarian follicles of the Potchefstroom Koekoek chicken breed. The mRNA expression was more abundant in F1 and F4 than in other ovarian follicles.(AU)

Background: Isoliquiritigenin (ISL) presents antitumor effects against melanoma cells. It is known that various circular RNAs (circRNAs) are involved in the development of melanoma. Therefore, the present study aims to investigate the molecular mechanisms of ISL and circ_0002860. Methods: Circ_0002860, microRNA-431-5p (miR-431-5p) and member RAS oncogene family (RAB9A) were detected through reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assay. Cell viability was examined via cell counting kit-8 assay. The proliferation ability was assessed using colony formation assay. Cell apoptosis and cell cycle were determined by flow cytometry. Transwell assay was used for detection of migration and invasion. Western blot was conducted for protein analysis. Target binding was confirmed via dual-luciferase reporter assay and RNA immunoprecipitation (RIP) assay. In vivo research was performed through xenograft tumor assay. Results: Circ_0002860 was downregulated by ISL in melanoma cells. ISL-induced inhibitory effects on cell proliferation, cell cycle progression, migration and invasion were alleviated by circ_0002860 overexpression. MiR-431-5p was a target of circ_0002860. Circ_0002860 eliminated the ISL-induced tumor inhibition via sponging miR-431-5p in melanoma cells. Circ_0002860 elevated the RAB9A level by targeting miR-431-5p. The function of ISL was related to miR-431-5p/RAB9A axis in melanoma progression. Tumor growth was reduced by ISL in vivo through downregulating circ_0002860 to regulate miR-431-5p and RAB9A levels. Conclusion: The current data indicates that ISL suppressed cell malignant progression of melanoma via targeting the circ_0002860/miR-431-5p/RAB9A pathway.(AU)

This purpose of this study was to compare the efficiency of the McMaster and Mini-FLOTAC quantitative techniques in the investigation of helminths in feces of pigs. An analysis was made of 74 fecal samples from pigs raised on family farms located in Rio de Janeiro, Brazil. These were analyzed by the Mini-FLOTAC and McMaster techniques in a solution of 1,200g/mL NaCl. This investigation revealed a superiority in the frequency of all helminths detected by Mini-FLOTAC, including Ascaris suum, Trichuris suis, strongyles and Strongyloides ransomi. The Kappa index revealed substantial agreement in all comparisons made in relation to the frequency of positive samples. However, significant statistical differences in the comparison of EPGs between McMaster and Mini-FLOTAC were observed for all nematodes (p ≤0.05). Higher values of Pearson's linear correlation coefficient (r), between the techniques in relation to EPG were observed for A. suum and T. suis, differently from what was observed for strongyles and S. ransomi. Mini-FLOTAC proved to be a more satisfactory and reliable technique both for the diagnosis of parasites and for the determination of EPG in pig feces due to the larger size of its counting chambers, thus increasing the helminth egg recovery rates.(AU)
Este estudo objetivou comparar a eficiência das técnicas quantitativas de McMaster e Mini-FLOTAC para a pesquisa de helmintos em fezes de suínos. Foram analisadas 74 amostras fecais de suínos mantidos em propriedades familiares, localizadas no Rio de Janeiro, Brasil. Estas foram analisadas pelas técnicas de Mini-FLOTAC e McMaster com solução de NaCl 1.200g/mL. Pode-se verificar uma superioridade na frequência de todos os helmintos detectados por meio do Mini-FLOTAC, incluindo-se Ascaris suum, Trichuris suis, estrongilídeos e Strongyloides ransomi. O índice Kappa mostrou uma concordância substancial em todas as comparações realizadas, em relação à frequência de amostras positivas. Entretanto, diferenças estatísticas significativas na comparação do OPG entre McMaster e Mini-FLOTAC foram observadas para todos os nematoides (p ≤0.05). Elevados valores do coeficiente de correlação de Pearson foram observados entre as técnicas, em relação ao OPG para A. suum e T. suis, diferentemente do evidenciado para estrôngilos e S. ransomi. Mini-FLOTAC mostrou-se uma técnica mais satisfatória e confiável, tanto para o diagnóstico de parasitos quanto para a determinação do OPG em fezes de suínos, devido ao maior tamanho das suas câmaras de contagem, situação que acaba aumentando a eficiência da recuperação dos ovos dos helmintos.(AU)
