Resumo

O controle da qualidade seminal produzido por centrais e unidades de disseminação genética permite a maximização da produção de doses inseminantes por ejaculado, bem como organiza o acompanhamento da produtividade de cada macho. Este acompanhamento é crítico tanto quando o reprodutor jovem é introduzido na rotina de coleta de sêmen quanto na determinação do momento do descarte e reposição. Essencialmente, a avaliação do sêmen em suínos é a mesma há décadas, porém novas metodologias de análise individual da célula como a citometria de fluxo e de constituintes de tecidos e fluídos como a proteômica e metabolômica já estão trazendo avanços na andrologia animal. Na presente revisão, são abordados os aspectos técnicos, vantagens e limitações destas três análises avançadas aplicadas ao sêmen suíno, discutindo a sua implementação e impactos no manejo reprodutivo da espécie.(AU)


The quality control of the semen doses produced by artificial insemination centers and allows the maximization of the production of inseminating doses per ejaculate, as well as organizes the monitoring of the productivity of each male. This follow-up is critical when the young male is initiated in routine semen collection and in determining the moment of male replacement. Essentially, semen evaluation in pigs has been the same for decades, but new methodologies for individual cell analysis such as flow cytometry and tissue and fluid constituents such as proteomics and metabolomics are already bringing advances in animal andrology. In this review, the technical aspects, advantages, and limitations of these three advanced analyzes applied to swine semen are addressed, discussing their implementation and impacts on the reproductive management of the species.(AU)

Resumo

O conhecimento sobre a reprodução de aves silvestres está em constante evolução com o avanço de novas pesquisas. O número gigantesco de espécies de aves e a dificuldade de acesso aos seus gametas são aspectos complicadores para o estabelecimento de técnicas de reprodução assistida. Aspectos muitos básicos como métodos de coleta de sêmen e as características seminais da grande maioria das aves silvestres são desconhecidos. Essas informações são fundamentais para entender a biologia reprodutiva e planejar programas de multiplicação com o uso de inseminação artificial. Por isso o levantamento das principais particularidades da reprodução de aves silvestres norteará ações para o aprofundamento de pesquisas já realizadas e estímulo ao desenvolvimento de novas metodologias de coleta de sêmen e avaliação seminal em aves silvestres até o momento não estudadas.(AU)


The knowledge about the reproduction of wild birds is constantly evolving with the advancement of new research. The large number of bird species and the difficult access to their gametes are complicating aspects for the establishment of assisted reproduction techniques. Very basic aspects such as semen collection methods and the seminal characteristics of the vast majority of wild birds are unknown. This information is fundamental for understanding reproductive biology and planning multiplication programs using artificial insemination. Therefore, the survey of the main particularities of the reproduction of wild birds will guide actions for the deepening of research already carried out and encourage the development of new methodologies for semen collection and seminal evaluation in wild birds that have not been studied so far.(AU)

Resumo

A espermiogênese é o processo final da espermatogênese no qual a espermátide se transforma em espermatozoide. Durante esse processo podem ocorrer alterações espermáticas, especialmente no acrossoma, na morfologia da cabeça, na condensação da cromatina e na formação dos vacúolos nucleares. A diferenciação entre os quadros clínicos é feita com repetições dos exames andrológicos. A avaliação morfológica indica a situação da espermiogênese nas 4 semanas anteriores a coleta do sêmen. Com os resultados do exame andrológico é possível identificar a qualidade seminal naquele período. O presente artigo é o relato de caso de um touro jovem doador de sêmen com 26 meses de idade, no início do regime de coletas de sêmen em um Centro de Coleta e Processamento de Sêmen no Sul do Brasil. A produção espermática deste animal foi avaliada durante 12 meses de coleta, apresentando sempre morfologia espermática muito alterada e motilidade média de 54% no exame imediato. Em 13 coletas seminais, o ejaculado deste animal apresentou em média 86% de defeitos maiores, 10% de defeitos menores e 96% de defeitos totais. Os defeitos maiores, que são os que possuem maior efeito na fertilidade, estão diretamente ligados a espermiogênese e os defeitos menores, que possuem menor efeito na fertilidade, são ocasionados principalmente durante o trânsito pelo epidídimo. O quadro clínico desse animal demonstrou a importância do exame morfológico para avaliar a espermatogênese e principalmente o processo de diferenciação final da espermátide em espermatozoide. Casos como este devem ser descritos como espermiogênese alterada e o reprodutor deve ser afastado da reprodução, após descartar alterações morfológicas devido a degeneração testicular grave.(AU)


Spermiogenesis is the final process of spermatogenesis in which the spermatid transforms into sperm. During this process, sperm alterations may occur, especially in the acrosome, head morphology, chromatin condensation and formation of nuclear vacuoles. Differentiation between clinical conditions is made with repetitions of andrological exams. The morphological evaluation indicates the status of spermiogenesis in the 4 weeks prior to semen collection. With the results of the breeding soundness examination, it is possible to identify the seminal quality in that period. The present article is a case report of a young 26-month-old semen donor bull, that started semen collection routines at a Semen Collection and Processing Center in southern Brazil. The sperm production of this animal was evaluated during 12 months of collection, always showing very altered sperm morphology and average motility of 54% in the immediate examination. In 13 seminal collections, the ejaculate of this animal presented an average of 86% of major defects, 10% of minor defects and 96% of total defects. Major defects, which have the greatest impact on fertility, are directly linked to spermiogenesis and minor defects, which have less effect on fertility, are mainly caused during transit through the epididymis. The clinical conditions of this animal demonstrated the importance of the morphological examination to evaluate spermatogenesis and especially the process of final differentiation of the spermatid into spermatozoa. Cases like this should be described as altered spermiogenesis and the bull should be withdrawn from breeding, after ruling out morphological changes due to severe testicular degeneration.(AU)

The objective of this study was to evaluate characteristics of the testicular parenchyma and vascular parameters of the pampiniform plexus obtained by ultrasound, semen quality parameters, and sperm freezability in Nellore bulls classified based on residual feed intake (RFI). Twenty-seven bulls (21.82±0.88 months of age) evaluated for feed efficiency were sampled for the study, including 15 with low RFI (−0.592±0.09 kg dry matter/day) and 12 with high RFI (0.792±0.10 kg dry matter/day). In ultrasound and Doppler assessment, the most efficient animals (low RFI) showed higher pulsatility and resistive indexes, as well as a tendency towards greater heterogeneity of the testicular parenchyma (0.625±0.032 vs. 0.508±0.032, 1.012±0.072 vs. 0.802±0.072, and 12.9±0.96 vs. 10.2±0.96, respectively, for low vs. high RFI). However, these animals tended to have lower peak diastolic velocity (5.19±0.50 for low RFI vs. 6.54±0.50 for high RFI). Analysis of fresh semen showed a lower percentage of minor defects in low RFI animals (2.67±1.19%) compared with high RFI animals (8.10±1.19%), without differences in the other parameters in fresh or thawed semen and after thermoresistance testing. Evaluation of flow cytometry parameters showed a higher quality of mitochondrial respiration in semen samples of low RFI animals (22.04±2.50%) compared with high RFI animals (12.29±2.71%). Therefore, although RFI exerts an effect on the Doppler parameters of the pampiniform plexus, it is not sufficient to affect the quality of fresh or thawed semen.(AU)

This study was aimed to assess the efficiency of coconut water extender with addition of soy lecithin and sucrose as nonpermeable cryoprotectants for canine semen vitrification, using a simple method that yields a high survival rate of spermatozoa for clinical use. Twelve ejaculates from 12 adult normozoospermic dogs were collected separately by digital manipulation and only the second semen fraction was used in this study. After evaluation of volume, concentration, viability, total and progressive motility, velocity parameters and morphology, semen was diluted with a coconut water extender (50% (v/v(volume per volume)) coconut water, 25% (v/v) distilled water and 25% (v/v) 5% anhydrous monosodium citrate solution) with addition of soy lecithin and fructose at 1% and 0.25M sucrose until final concentration of 100x106 spermatozoa/ml. After equilibration at 5ºC for 60 minutes, semen was vitrified by "direct dropping method" into liquid nitrogen in spheres with a volume of 30 µl. After a week of storage the spheres were devitrified as three of them were dropped into 0.5 mL of CaniPlus AI medium (Minitüb, Germany), which was previously warmed in a water bath at 42ºC for 2 minutes and evaluated about the above mentioned parameters. It was found that vitrification resulted in a lower percentage of viable sperms, normal morphology, total and progressive motilities (p0.05) compared to fresh semen samples. In conclusion, our results demonstrate that vitrification with coconut water extender with addition of 1% soy lecithin and 0.25M sucrose as cryoprotectants, has an excellent potential for routine canine sperm cryopreservation.(AU)

At least 30-40% of stallions in commercial breeding programs are moderately fertile and 8-12% are subfertile (0.5-3% with severe subfertility). From the total reported cases of the subfertility, in 2-20% of the stallions the cause is unknown or was not established. The objective of this work is to present the concept of subfertile stallion based on the current state of knowledge and advanced molecular diagnostic technologies. Low pregnancy rates have been reported in stallions with normal semen quality after conventional evaluation. Acrosome reaction (AR) is necessary for natural fertilization and impaired acrosome reaction (IAR) leads to subfertility or infertility in horses, however, AR test is not included in routine semen analysis. Genome-wide association study identified FKBP6 as a strong candidate gene responsible for this failure. The gene encodes for FK506 binding protein 6 (FKBP6) which is involved in sperm development and functions. We could conclude that the evaluation of the acrosomal status is essential in cases of stallions with good motility, concentration, morphology and viability but unexplained (idiopathic) subfertility or infertility. It is important to highlight the recent increase in reports of fertility problems in stallions related to disorders of genetic origin.(AU)

Resumo

A qualidade do sêmen criopreservado, utilizado na inseminação artificial em tempo fixo (IATF) em bovinos, é um dos principais fatores que impactam sobre a fertilidade, e está relacionada à capacidade de produção espermática dos touros, à criotolerância dos espermatozoides e aos critérios técnicos do processo de criopreservação adotados. Neste sentido, devemos destacar a importância do controle de qualidade das partidas de sêmen antes de serem liberadas para uso na IATF. Nos últimos anos várias técnicas vêm sendo desenvolvidas para avaliar com mais acurácia as partidas de sêmen e evitar o uso daquelas que possam resultar em prejuízos na fertilidade, mas mesmo assim, tem se notado alta variabilidade na taxa de prenhez entre touros e entre partidas de sêmen. Esta divergência se deve ao fato da habilidade fértil do espermatozoide ser multifatorial, ou seja, dependente de diversas características estruturais, morfofuncionais e moleculares. Ademais, os eventos que permitem os espermatozoides passarem por capacitação espermática, um pré-requisito para a fertilidade, têm intrigado os pesquisadores em vista da complexidade dos processos envolvidos e dos efeitos deletérios da criopreservação espermática. Esta revisão tem por objetivo compilar estudos que mostrem a relação entre a capacitação espermática e a fertilidade do sêmen bovino criopreservado, levando em consideração as técnicas de avaliação e os resultados até o momento.(AU)


The quality of cryopreserved semen, used in fixed-time artificial insemination (FTAI) in cattle, is one of the main factors that impact fertility and is related to the sperm production capacity of bulls, sperm cryotolerance, and the technical criteria for cryopreservation. In this sense, we must highlight the importance of quality control of semen batches before commercialization for the FTAI. In recent years, several techniques have progressed to more accurately evaluate semen batches and avoid using those that may result in impaired fertility. However, we still notice a high variability in the pregnancy rate between bulls and between semen batches. This divergence is due to the multifactorial sperm-fertilizing ability, i.e., it depends on several structural, morphofunctional, and molecular characteristics. In addition, the events that allow sperm to undergo sperm capacitation, a prerequisite for fertility, have intrigued researchers due to the complexity of the processes involved and the adverse effects of sperm cryopreservation. This review aims to compile studies that show the relationship between sperm capacitation and fertility in cryopreserved bovine semen, considering the evaluation techniques and results to date.(AU)

Background: The artificial insemination has become a well-established method in the breeding of bitches, and evaluation of the factors that may potentially affect pregnancy success is essential. For this reason, it is essential to evaluate the factors that may affect fertility of the bitch when artificial insemination is performed. Serum progesterone concentrations and vaginal cytology have been used to determine the time of ovulation and stage of the estrus cycle. This study aimed to evaluate the artificial insemination method, the serum progesterone concentration, the breed size, age, the whelping number, vaginal cytology parameters, and their interactions on pregnancy success in bitches. Materials, Methods & Results: A total of 607 bitches that had undergone reproductive consultation with the Mexican Canine Federation from January to December 2016 were enrolled in the present study and assigned to one of 2 artificial insemination methods (intravaginal and transcervical) using fresh semen. Determination of the estrus cycle phase and the time of Artificial insemination was based on vaginal cytology and serum progesterone concentrations. Bitches inseminated by the transcervical technique had a higher pregnancy rate with respect to females inseminated by the intravaginal technique (P < 0.05). Moreover, females with a serum progesterone concentration of 5-10 ng/mL had a greater probability (> 4 times) of getting pregnant than animals with lower or higher progesterone concentrations (P < 0.05). Bitches inseminated by the intravaginal technique and with serum progesterone concentrations >10 ng/mL had a considerable reduction in pregnancy (P < 0.05) compared with females with < 10 ng/mL serum progesterone or with bitches inseminated by the transcervical technique. Discusion: Serum progesterone concentration, the artificial insemination method, and superficial cells without a nucleus modified the pregnancy rate in bitches. Females inseminated by transcervical semen deposition had a higher pregnancy rate than females inseminated by the intravaginal technique. Using fresh or frozen-thawed semen produced a higher pregnancy rate in bitches inseminated by transcervical semen deposition than females inseminated by the intravaginal technique. Differences in the pregnancy rate between transcervical and intravaginal insemination could be associated with the correct semen disposition, the distance that the sperm must travel to reach the oocyte, as well as the number of sperm that reach the oviduct ampulla. Exist evidences that after ovulation, as progesterone rises, the cervix is closed, which may compromise the passage of the sperm deposited into the vagina. Therefore, it is likely that in females with a serum progesterone concentration > 10 ng/mL, the cervix was closed, compromising the ability of the sperm to access the oviduct. Thus, the use of intravaginal insemination should be done in bitches with a serum progesterone concentrations < 11 ng/mL to reduce the possibility of cervical closure and to increase the odds of pregnancy. It is well documented that the serum progesterone concentration and vaginal cytology parameters have a great influence on pregnancy success, and the results confirm these findings. In the present study, 96% of the bitches inseminated with a serum progesterone concentration of 5-10 ng/mL got pregnant and had higher odds of pregnancy than bitches with lower or higher serum progesterone concentrations.

Resumo

O melhoramento genético possibilitou a evolução da eficiência e taxas de desfrute da pecuária nacional. A biotécnica da IATF (Inseminação Artificial por Tempo Fixo) consolidou-se como a principal ferramenta de disseminação de genética melhorada em rebanhos de leite e de corte. A avaliação espermática laboratorial tem o objetivo de analisar o potencial de reprodutores/partidas de sêmen e buscar a desafiadora tarefa de determinar e estabelecer características confiáveis que possam ser utilizadas como indicadores de fertilidade. Nesse contexto emerge o sistema CASA (Computer-Assisted Sperm Analysis) como ferramenta promissora para obtenção de parâmetros indicativos de fertilidade seminal.(AU)


Genetic improvement enabled the evolution and efficiencies of national livestock activity. The IATF (Fixed Time Artificial Insemination) biotechnology, has established itself as the main tool for the dissemination of improved genetics in dairy and beef herds. Laboratory sperm evaluation aims to analyze the potential of a sires/semen batches and seek the challenging task of determining and establishing reliable characteristics that can be used as fertility indicators. In this context, the computerized system of sperm motility (CASA) emerges as a promising tool for obtaining parameters indicative of seminal fertility.(AU)

Resumo

A grande demanda por sêmen congelado para inseminação artificial no Brasil está ocasionando que criadores, programas genéticos, e centros de coleta e processamento de sêmen produzam touros cada vez mais jovens para doação de sêmen. A avaliação genômica é uma nova ferramenta para a seleção de touros jovens para os centros de coleta, porém, junto vieram os problemas na seleção de bons doadores de sêmen que muitas vezes não atingem a puberdade na idade desejada, principalmente na raça Nelore que é a mais importante para o mercado de inseminação no país. A maioria dos problemas com a produção de sêmen do touro jovem genômico podem ser resolvidos com o tempo, mas ainda ocasiona vários problemas não resolvidos. Os touros jovens genômicos da raça Nelore atingem a puberdade entre 13 a 16 meses e os touros jovens das raças Holandesa e Angus entre 10 a 12 meses de idade sendo que a maturidade sexual no geral é alcançada entre 18 a 24 meses quando passam a produzir sêmen rotineiramente e produzem grandes quantidades de unidades de sêmen criopreservado para uso na inseminação artificial. Este cenário ocasionará um problema para o mercado da inseminação artificial, com a demanda de mais touros com maturidade sexual.(AU)


The great demand for frozen semen in the artificial insemination in Brazil is causing breeders, genetic programs, and semen collection and processing centers to produce ever younger bulls for semen donation. The genomic evaluation is a new tool for the selection of young bulls for the collection centers, however, along came the problems in the selection of good semen donors that often do not reach puberty at the desired age, especially in the Nellore breed, the most important for the insemination market in the country. Most problems with genomic young bull semen production can be resolved over time, but there are still several unresolved problems. Young genomic Nellore bulls reach puberty between 13 to 16 months and young Holstein and Angus bulls between 10 to 12 months of age and sexual maturity is generally reached between 18 to 24 months when they start to produce semen routinely, so producing a large quantities of cryopreserved semen units for use in artificial insemination. This will cause a problem for the artificial insemination business where more sexually mature bulls will be needed.(AU)

Morphological sperm evaluation supported by the morphometry can be used in the determination of the seminal quality and in the investigation of potential extenders. Although there are studies comparing TRIS and ACP extenders, there are no comparative studies between them for the computerized assisted semen analysis (CASA), sperm viability, membrane functionality and sperm morphometry parameters of cryopreserved canine semen. Hence, we aimed to evaluate the effects of ACP-106c and TRIS on post-freezing canine sperm quality. Five dogs were submitted to semen collection twice with one-week interval. The semen was evaluated within the parameters: total motility, vigor, concentration, viability, plasma membrane functionality, morphology and morphometry. In the morphometric evaluation, the morphologically normal sperm was measured as: length, width, area and perimeter of the head and the midpiece, tail length and total length. The parameters of ellipticity, elongation, regularity and roughness were determined. Then, the semen was divided into two aliquots that were diluted in TRIS or ACP-106c, with the addition of egg yolk and glycerol. The diluted semen was refrigerated and frozen. The thawed samples were evaluated. Total motility, viability, sperm membrane functionality and normal morphology reduced after thawing in both extenders (morphology reduced from 89.60 ± 1.3% to 84.40 ± 1.8 and 84.60 ± 1.1% in TRIS and ACP-106c, respectively). However, it did not differ between TRIS and ACP-106c. In the ACP106c the sperm head defects in cryopreserved semen were higher compared to fresh semen (P < 0.05). For all the morphometric parameters evaluated, there were no differences between fresh and cryopreserved samples (3.70 ± 0.4% vs. 2.30 ± 0.5%). In kinetics, with an interval of one week statistical differences between the extenders were found only in the parameters ALH and LIN (P < 0.05). Regardless of the extender, there were no changes in the morphometric parameters of sperm after thawing.(AU)

Resumo

A manutenção do sêmen suíno sob condições de refrigeração tem se destacado como uma técnica eficiente para a difusão do material genético, através dos programas de inseminação artificial. Neste sentido, a adição de várias substâncias, como antioxidantes, ao diluente vem sendo adotada na conservação do sêmen de diversas espécies domésticas. Assim, o objetivo deste estudo foi investigar a ação do ácido fítico adicionado ao diluente do sêmen conservado sob refrigeração, através da avaliação dos resultados de fertilidade de fêmeas suínas inseminadas. Foram utilizados quatro reprodutores (Agroceres) e setenta fêmeas para formação de lotes no decorrer de sete semanas. Os lotes de inseminação foram formados com o sêmen diluído em BTS (35 fêmeas) e BTS adicionado de ácido fítico na concentração de 525µM/100mL (35 fêmeas). Na avaliação dos resultados, considerou-se os seguintes aspectos: 1) taxa de fertilidade; 2) taxa de parição; 3) número total de leitões/parto; 4) número de leitões nascidos vivos/parto. Observou-se que as taxas de fertilidade e parição, assim como a prolificidade das porcas inseminadas com BTS e BTS adicionado de ácido fítico não diferiram estatisticamente (p>0,05). A adição do ácido fítico ao diluente BTS não influenciou os resultados reprodutivos das fêmeas suínas. Dessa forma, estudos futuros devem ser realizados a fim de avaliar a eficácia de diferentes concentrações do ácido fítico sobre a fertilidade e prolificidade de fêmeas suínas.


The maintenance of boar semen under refrigerated conditions has been highlighted as an effective technique for the diffusion of genetic material through artificial insemination programs. In this sense, the addition of several substances, such as antioxidants, to the diluente has been used in the conservation semen from various domestic species. Thus, this study aimed to investigate the action of phytic acid added to the semen extender, conserved under refrigeration by evaluating the fertility results obtained from inseminated swine females. Four boars (Agroceres), which were used for artificial insemination as semen donors, and seventy females were used to compose the lots during the seven weeks of the experiment. The insemination lots were formed with the semen diluted in BTS (35 females) and BTS added with phytic acid at 525µM/100mL (35 females). For results evaluation, the following aspects were considered: 1) fertility rate; 2) farrowing rate; 3) total number of piglets/delivery; 4) number of live birth piglets/delivery. It was observed that the fertility and farrowing rates, as well as the prolificacy of sows inseminated with BTS and BTS added with phytic acid did not differ statistically (p>0.05). The addition of phytic acid to the BTS diluent did not influence the reproductive results of swine. Therefore, further studies must be performed to evaluate the efficacy of different phytic acid concentrations on the fertility and prolificacy of sows.

The spermatic membrane is rich in polyunsaturated fatty acids, which makes it sensitive to the action of reative species of oxygen, which can damage the seminal quality of the scraps. The purpose of this study was to evaluate the effect of the supplementation of two selenium sources at different doses. Third five scraps were allocated in four groups: (INOR30) 0.30 ppm sodium selenite; (COMP30) 0.30 ppm selenium metal-amino acid; (MIXED15+15) 0.15 ppm sodium selenite + 0.15 ppm selenium metal-amino acid and (COMP15) 0.15 ppm selenium metal-amino acid. The ejaculates of the scraps were evaluated over 22 weeks, resulting in 210 samples evaluated for volume, motility, pH, presence of agglutination and morphological changes, and 140 samples for spermatic concentration. The data was analyzed with repeated measures in time in a mixed model with type of selenium supplementation, periods of evaluation (one period of two weeks + five periods of four weeks) and their interactions as fixed effects, and animal and the worker that collected the ejaculates as random effects. Results showed no difference in selenium supplementation with the sources and doses used. In this way, it was possible to verify that the metal amino acid of selenium at the dose of 0.15 ppm promotes the same effect as the diets formulated with 0.30 ppm of sodium selenite.(AU)


A membrana espermática é rica em ácidos graxos poliinsaturados, o que a torna sensível à ação de espécies reativas de oxigênio, que podem prejudicar a qualidade seminal dos cachaços. O objetivo do presente estudo foi avaliar o efeito da suplementação de duas fontes de selênio em diferentes doses. Trinta e cinco cachaços foram distribuídos em quatro grupos: (INOR30) 0,30 ppm de selenito de sódio; (COMP30) 0,30 ppm de metal-aminoácido de selênio; (MISTO15+15) 0,15 ppm de selenito de sódio + 0,15 ppm de metal-aminoácido de selênio e (COMP15) 0,15 ppm de metal-aminoácido de selênio. Os ejaculados dos cachaços foram avaliados durante 22 semanas, resultando em 210 amostras avaliadas para volume, motilidade, pH, presença de aglutinação e alterações morfológicas, e 140 amostras para concentração espermática. Os dados foram analisados com medidas repetidas no tempo em modelo misto, em que o tipo de suplementação de selênio, os períodos de avaliação (um período de duas semanas + cinco períodos de quatro semanas) e suas interações foram os efeitos fixos, e o animal e o funcionário que coletou os ejaculados foram os efeitos aleatórios. Os resultados obtidos demonstraram não haver diferença na suplementação de selênio com as fontes e doses utilizadas. Com isso, foi possível verificar que o metal-aminoácido de selênio na dose de 0,15 ppm promove o mesmo efeito das dietas formuladas com 0,30 ppm de selenito de sódio.(AU)

SexedULTRA-4M™ is made using an improved method of sex-sorting sperm in a less damaging environment for better retaining sperm integrity throughout the sorting process. The objective of this research was to compare conventional (CONV) and SexedULTRA-4M™ (ULTRA-4M) semen for bovine IVP using four Angus bulls. Matured slaughterhouse oocytes (n = 4000) were divided into the CONV group and the ULTRA-4M group (2000 COCs for each semen type). The IVF process was implemented with CONV and ULTRA-4M semen from the same bull. The cleavage rates, eight cell embryos and blastocysts on day 7 of culture were evaluated for each semen type and each bull. The statistical analysis was carried out with the ANOVA procedure SAS software. The results were 54.45% ± 1.03 and 58.10% ± 1.07; 35% ± 1.57 and 39.15% ± 1.62; 22.8% ± 1.09 and 27.15% ± 1.12 for CONV and ULTRA-4M, respectively, for cleavage rate, eight cell embryos and blastocysts on day 7 for the average of all bulls, comparing only the semen type. Concerning only the semen type, ULTRA-4M was significantly superior to CONV for cleavage rates (P = 0.01) and blastocysts on day 7 (P = 0.009). There were no significant differences between the CONV and ULTRA-4M groups (P>0.05) for all variables analyzed for Bull 1 and Bull 4, however, for Bull 2 ULTRA-4M was significantly superior to CONV for cleavage rates and blastocysts on day 7 (P< 0.05). In Bull 3, ULTRA-4M was significantly higher (P< 0.05) for blastocysts on day 7 compared to CONV. In conclusion, under the conditions of this research the ULTRA-4M and CONV semen produced similar bovine IVP results overall.(AU)

In recent years, there has been a great interest of purebred cat breeders in the possibilities and benefits of implementing of consistent veterinary care on reproduction. This includes tools available in the field of Assisted Reproductive Technologies (ART). As in dogs before, there is a tendency to reduce the reproductive potential of purebred cats, which is partly due to the nature of the selection of animals for reproduction on the basis of phenotypic features, omitting the control of their fertility. It is worthy to note hazardous specificity of this selection where inbreeding is commonly used. If all this is imposed on the frequent occurrence of infectious diseases in a domestic cat, then the population of purebred cats appears as a group of animals, which is a kind of challenge for a specialist of reproduction. This is a paradoxical phenomenon, because regarding non-purebred cats, the main challenge for the veterinary community is to limit their population. Some of the most important barriers to the implementation of advanced control of reproduction of purebred cats over the years have been: 1) difficulties with sperm collection, 2) technically troublesome artificial insemination, 3) numerous and difficult to recognize and treat forms of fertility disorders. In recent years, progress has been observed within each of these areas. It is possible to implement into practice extremely valuable tools that are helpful in monitoring the reproductive status of these animals. The breakthrough was the development of effective methods of semen collection, its evaluation and preservation and the development of increasingly effective methods of artificial insemination. Therefore, there is a real prospect of organizing cat reproduction centers operating within veterinary facilities dealing with small animals. The time is coming to carry out veterinary care for cat reproduction at such an advanced level as it has been developed in recent decades for dogs. Development in this area is facilitated by progress in research on the field of the use of ART in programs of rescue of endangered feline species where domestic cat is treated as model animal.(AU)

In the industry of bull semen freezing centers, one-step and two-step semen dilution protocols are two standard and well-known methods in semen freezing process. As the freezing/thawing processes cause detrimental effects on sperm function, the addition of antioxidants can improve sperm characteristics. Hesperidin (Hesp) is an antioxidant used as the male reproductive protective agent. Therefore, the aim of this study was to investigate two different dilution methods, as well as to evaluate Hesp supplementation influence on sperm characteristics in fresh and frozen thawed semen. Semen samples were collected from 12 Simmental bulls. Two separate examinations were conducted in, with and without Hesp supplementation groups. Statistical analysis was performed by an independent t-test, Mann Whitny test, MANOVA and ANOVA tests. In comparison to the one and two-step dilution protocols without Hesp supplementation, the two-step dilution showed greater cryoprotective potential. In the Hesp supplemented group, each semen sample was divided into six equal parts for experimental groups (dilution step method/µM of Hesp). In the both one and two step dilution protocols, significant improvements were detected in semen motility parameters by Hesp administration. Also, oxidative stress status was reduced in seminal plasma of Hesp treatment groups. Interestingly, in comparison with Hesp dosage, 1µM was shown to have greater semen cryoprotective potential. Abstract Semen motility is the most widely recognized semen quality parameter used by Artificial Insemination (AI) centers. With the increasing worldwide export of semen between AI centers there is an increasing need for standardized motility assessment methods. Computer-Assisted Sperm Analysis (CASA) technology is thought to provide an objective motility evaluation; however, results can still vary between laboratories. The aim of present study was to verify the impact of different setting values of the CASA IVOS II on motility, concentration, and morphology of bovine semen samples frozen in an extender with or without egg yolk and then decide on optimal settings for a further validation step across AI centers. Semen straws from 30 different bulls were analyzed using IVOS II with twelve modified settings. No significant changes were observed in semen concentration, percentage of motile sperm or kinetic results for either extender type. However, increasing settings for both STR and VAP progressive (%) from Low, Medium, and High cut-off values significantly (p<0.05) reduced the percentage of detected progressive spermatozoa, in egg yolk extender from 49.5±15.2, 37.2±11.9 to 11.9±5.3%, and in clear extender from 51.9±9.1, 35.8±7.3 to 10.0±2.4%, respectively. In clear extender only, the modification of droplet proximal head length significantly affected the detection of normal sperm percentages (88.0± 4.7 to 95.0±0.6 and 96.0±0.6%) and of the percentage of detected proximal droplets (12.2±4.7, 2.5±2.7 to 0.6±0.2%) for Low, Medium and High values respectively (p<0.05). The identification of sensitivity within the CASA system to changes in set parameters then led to the determination of an optimal IVOS II setting. The existing variability among centers for these phenotypes was reduced when the standardized settings were applied across different CASA units. The results clearly show the importance of applied settings for the final CASA results and emphasize the need for standardized settings to obtain comparable data.

A morfologia espermática é uma característica seminal que está associada à fertilidade e é, portanto, um componente importante na rotina do controle de qualidade em Centros de Coleta e Processamento de Sêmen (CCPS). É utilizada para caracterizar e quantificar os vários tipos de anormalidades nas células espermáticas do touro, usando preparação em montagem úmida para microscopia de Contraste de Interferência Diferencial (DIC), Contraste de Fase (CF) e Lâmina Corada (LC). O perfil da morfologia espermática permite a tomada de decisão sobre o aproveitamento ou descarte de um ejaculado para criopreservação e predizer a fertilidade de uma amostra de sêmen. Alterações no espermograma, com evidências do histórico e exame físico do touro, levam a interpretação da função testicular anormal, e consequentemente, ao diagnóstico, tratamento e prognóstico, se for o caso. As causas mais comuns de uma espermatogênese anormal nos touros incluem: termorregulação testicular anormal, desequilíbrios hormonais, estresse de manejo, ambiental e nutricional, efeitos de toxinas ou expressão de genes deletérios, além de defeitos de origem genética. Conhecendo-se a estrutura do espermatozoide, torna-se mais fácil a identificação das anormalidades espermáticas bem como a descrição das mesmas e uma melhor compreensão de sua origem.(AU)


Sperm morphology is a seminal trait that is associated with fertility and is therefore an important component of routine quality control in Semen Collection and Processing Centers (SCPC). It is used to characterize and quantify the various types of abnormalities in bull sperm cells, using wet mount preparation for Differential Interference Contrast (DIC), Phase Contrast (PC) and Slides Smear (SS) microscopy. The sperm morphology profile allows decision making on the use or disposal of an ejaculate for cryopreservation and predicts the fertility of a semen sample. Changes in sperm morphology may indicate that fertility may be impacted and therefore it is important to routinely monitor ejaculates. An abnormal spermogram with evidence from a history and physical examination of the bull can lead to the interpretation of abnormal testicular function, and consequently, to the diagnosis, treatment and prognosis, if any. The most common causes of abnormal spermatogenesis in bulls include: abnormal testicular thermoregulation, hormonal imbalances, management, environmental and nutritional stress, effects of toxins or expression of deleterious genes, in addition to defects of genetic origin. Knowledge in the structure of the sperm makes easier the identification of sperm abnormalities, as well as their description and provides a better understanding of their origin.(AU)

The aim of the study was to evaluate whether supplementation with different levels of selenium (Se) can change the biochemical and electrolytic components of semen, causing an improvement in seminal quality in rams. Thirty animals were kept in an intensive pen system, fed with hay and commercial ration, allocated into five groups (six animals/pen) and supplemented with a mineral mixture containing Omg (G1), 5mg (G2), 10mg (G3), 15mg (G4) and 20mg (G5) of Se/kg. Each group received a different treatment every 56 days and treatments were rotated between groups following a dynamic sequence. Semen samples were collected by electroejaculation after the end of each treatment to evaluate the levels of fructose, citric acid, potassium (K), sodium (Na), calcium (Ca), Se, zinc (Zn), manganese (Mn), sulfur (S) and lead (Pb). The statistical design was a 5x5 Latin square. The different levels of Se supplementation evaluated maintained the concentrations of electrolytes and minerals in the semen at the required levels and did not change the sperm quality, concluding that higher intakes of Se do not cause antagonistic effects on the absorption and subsequent action of other essential minerals supplied to the animals and still maintains electrolyte balance.


O objetivo do estudo foi avaliar se a suplementação com diferentes níveis de selênio (Se) pode alterar os componentes bioquímicos e eletrolíticos do sêmen, ocasionando, por conseguinte, uma melhoria na qualidade seminal em carneiros. Foram utilizados 30 animais, mantidos em sistema intensivo de baias, alimentados com feno e ração comercial, sendo alocados em cinco grupos (seis animais/baia) e suplementados com uma mistura mineral contendo 0mg (G1), 5mg (G2), 10mg (G3), 15mg (G4) e 20mg (G5) de Se/kg. Cada grupo recebeu um tratamento diferente a cada 56 dias, e foi realizado um rodízio de tratamentos entre os grupos seguindo uma sequência dinâmica. Amostras de sêmen foram colhidas por eletroejaculação após o fim de cada tratamento, a fim de se avaliarem os níveis de frutose, ácido cítrico, potássio (K), sódio (Na), cálcio (Ca), Se, zinco (Zn), manganês (Mn), enxofre (S) e chumbo (Pb). O desenho estatístico foi um quadrado latino 5x5. Os diferentes níveis de suplementação de Se avaliados mantiveram as concentrações de eletrólitos e de minerais do sêmen nos níveis exigidos e não modificaram a qualidade espermática, concluindo-se que maiores ingestões de Se não causam efeitos antagônicos na absorção e subsequente ação de outros minerais essenciais fornecidos aos animais e ainda mantêm o equilíbrio eletrolítico.
