Resumo

Three hundred hatching eggs from two layer breeder flocks (Hy-Line Brown and hy-Line W-36; 150 each) were obtained at three different periods of laying cycle (early, middle and late stages). The hatching eggs were randomly taken to assess the impact of chicken strain, age, and hatching time on some eggshell measurements and structures. Results indicate that egg weight, shell percentage and shell thickness were significantly affected by strain, breeder age, and incubation time. Increasing rate of calcium absorption and utilization by the embryos during the embryonic stage clearly reflects the decrease occurred in eggshell strength. The differences of eggshell breaking strength during incubation time periods were 0.41 and 0.55 kg/cm2, the differences due to the incubation time may be due to the higher demand for calcium utilization in the late incubation period (18 days), observed on the10th day of incubation. Brown eggs recorded significant higher calcium percentage in eggshell compared to the white ones. The brown breeder hens recorded significant higher total pores per egg compared to the white egg. Concerning eggshell ultrastructure, total thickness of the brown eggshell was significantly higher than those of the white eggshell by about 3.7%. All parameters, except for total thickness, were significantly affected by the interaction between strain and breeder age. Finally, we conclude that some traits including egg weight, shell percentage, shell thickness, calcium percentages in eggshell, and total pores per egg were significantly affected by strain, breeder age, and incubation time. Eggshell ultrastructure of brown egg was better compared to the white ones.(AU)

Resumo

Bacillus cereus is considered the most potent bacterial strain in terms of the increment in induced proteins during thermal treatment at 52 °C for 90 min. Protein production in food-born microorganism (Bacillus cereus) recovered from contaminated food was investigated in response to heat shock treatment. Bacterial tolerance towards pH, salinity, and temperature at various levels was also investigated. Heat-shock proteins (HSPs) produced when exposed to 52 °C for up to 60 minutes led to significant differences (30%) above the untreated control (37 °C), and the maximum difference was recorded at 52°C at 90 minutes. ISSR detected a higher number of bands/primer than RAPD (13.7 vs. 12.7, respectively), and more polymorphic bands (10.7 vs. 8.4 bands/primer, respectively). The untreated bacterial strain did not grow at pH levels lower than 3, whereas the thermally treated strain grew significantly at pH two. A consistent increase in HSPs was observed, with a gradual increase in salinity of less than 16%. Surprisingly, the gradual increase in temperature did not induce tolerance against higher temperatures. However, a significant growth rate was noticed in response to heat-shocked treatments. The untreated Bacillus cereus demonstrated antibiotic resistance to gentamycin and clindamycin (1.54 and 1.65 cm, respectively), much lower than the corresponding inhibition areas with preheat-treated test pathogen which were recorded (2.37 and 2.49 cm, respectively).


Bacillus cereus é considerada a cepa bacteriana mais potente em termos de incremento de proteínas induzidas durante o tratamento térmico a 52 °C por 90 min. A produção de proteínas em microorganismos de origem alimentar (Bacillus cereus) recuperados de alimentos contaminados foi investigada em resposta ao tratamento de choque térmico. A tolerância bacteriana ao pH, salinidade e temperatura em vários níveis também foram investigadas. Proteínas de choque térmico (HSPs) produzidas quando expostas a 52 °C por até 60 minutos levaram a diferenças significativas (30%) acima do controle não tratado (37 °C), e a diferença máxima foi registrada a 52 °C em 90 minutos . O ISSR detectou um maior número de bandas/iniciador do que o RAPD (13,7 vs. 12,7, respectivamente) e mais bandas polimórficas (10,7 vs. 8,4 bandas/iniciador, respectivamente). A cepa bacteriana não tratada não cresceu em níveis de pH abaixo de 3, enquanto a cepa tratada termicamente cresceu significativamente em pH dois. Observou-se aumento consistente de HSPs, com aumento gradual da salinidade inferior a 16%. Surpreendentemente, o aumento gradual da temperatura não induziu tolerância a temperaturas mais altas. No entanto, uma taxa de crescimento significativa foi observada em resposta aos tratamentos de choque térmico. O Bacillus cereus não tratado demonstrou resistência antibiótica à gentamicina e clindamicina (1,54 e 1,65 cm, respectivamente), muito menor do que as áreas de inibição correspondentes com patógeno de teste pré-tratado que foram registradas (2,37 e 2,49 cm, respectivamente).

Background: Staphylococcus spp. are the most frequently isolated microorganisms in mastitis cases of small ruminants. The virulence factors of Staphylococcus spp. are critical in the treatment. Therefore, preventive medicine and mastitis control programs, especially herd vaccinations are of great importance in the prevention of mastitis. However, it is not always easy to obtain these vaccines under field conditions. This study, it was aimed to compare the effects of different commercial Staphylococcus spp. vaccines licensed for bovines and species-specific mastitis vaccines on mastitis rates and somatic cell count (SCC) on Saanen goats on field conditions. Materials, Methods & Results: The animal material consisted of 115 (230 udder halves) nulliparous Saanen goats. Goats were randomly grouped as bovine vaccine 1 (BV1 , n = 58), bovine vaccine 2 (BV2 , n = 58), small ruminant vaccine (SRV, n = 56), and control (n = 56). Vaccines were administered to goats in 2 doses according to the label regimen. First milk samples were collected between 0-5 days in milk (DIM) for microbiological analysis and 25-35 DIM for SCC. The other milk samples were collected at 25-35 (1st month) DIM, 60-65 (2nd month) DIM, 85-95 (3rd month) DIM, 115-125 (4th month) DIM, 145-155 (5th month) DIM for microbiological analysis and SCC. Non-aureus staphylococci (NAS) and Staphylococcus aureus were the most frequently isolated microorganism. It was found that the total mastitis rate decreased in vaccine groups compared to the control group. A significant difference was found only in the BV2 and SRV groups. The significant difference in S. aureus infection was found only in the SRV group. Mastitis vaccines used in this study decreased the NAS mastitis rate, but no significant difference was observed. It was found that the clinical mastitis incidence decreased in all vaccine groups compared to the control group, and a significant difference was found between the BV2 and SRV groups compared to the control group (P < 0.05). Somatic cell count was lower in the SRV and BV2 groups compared to the control group (P < 0.05). Discussion: In this study, compatible with the previous reports NAS and S. aureus were the most frequently isolated microorganism. The diversity of virulence factors of Staphylococcus spp. also plays an important role in its high incidence. In some countries, mastitis vaccines used in cows are also administered to small ruminants for reducing infection rates. Similarly, in this study, it was found that the mastitis rate decreased in all vaccine groups compared to the control group. A significant difference was found only in the BV2 and SRV groups. It is thought that the reason for the statistical difference may be due to the biofilm antigen in the BV2 and SRV. In addition, J5 strain in the BV2 is estimated to be effective in reducing the prevalence of gram-negative mastitis. It was observed that the infection rates decreased in the vaccine groups, especially due to S. aureus and NAS. Spontaneous treatment rates were very close to each other between the groups. The reason for the high rate of spontaneous treatment in this study can be explained by the fact that the animals were young and in their 1st lactation. SCC was lower in all vaccine groups compared to the control group. This situation is associated with the decrease in infection rates related to the use of vaccines. It was observed that SCC was lower in the vaccine groups. In addition, SCC was found to be lower in this study compared to similar studies. However, it is evident that the use of species-specific vaccines in the SRV group significantly reduced the rates of total S. aureus mastitis, subclinical NAS mastitis, and new infections by NAS compared to other vaccines. Furthermore, the species-specific vaccine significantly increased the rate of spontaneous treatment for S. aureus mastitis.

Abstract Rotavirus is the main infective agent of acute gastroenteritis (AGE) in children under the age of five years and causing significant morbidity as well as mortality throughout the world. The study was carried out to detect the prevalence rate, genotypes strain and risk factors of Rotavirus among the children of rural and urban areas of district Bannu Khyber Pakhtunkhwa Pakistan. A total of 180 stool samples were collected from children under the age of 5 years from two major hospitals of Bannu from January to December (2015). The samples were analyzed by Reverse-transcriptase Polymerase Chain Reaction (RT-PCR) for the detection of Rotavirus, positive samples were further processed for genotyping (G and P type) through specific PCR. Of the total, 41 (23%) samples were positive for Rotavirus. The most prevalent G genotypes found were: G3, G8, G9 (each 29%), followed by G10 (15%), and G11 (10%). Whereas the prevalent P genotypes were: P-8 (25%), P-4 and P-10 (each 20%), P-9 (15%), followed by P-6 and P-11 (each 10%). Moreover, Rotavirus infection was more prevalent in summer (23.73%) and winter (22.7%) than spring (20%) and autumn (21.4%). Rotavirus infection exhibited high frequency in June (14%), October (8%) and November (6%). It is concluded that Rotavirus is more prevalent in children and various genotypes (G and P) of Rotavirus are present in the study area. Lack of studies, awareness and rarer testing of Rotavirus are the principal reasons of virus prevalence in district Bannu, Pakistan.


Resumo O rotavírus é o principal agente infeccioso da gastroenterite aguda (AGE) em crianças menores de 5 anos e causa de morbidade e mortalidade significativas em todo o mundo. O estudo foi realizado para detectar a taxa de prevalência, cepa de genótipos e fatores de risco de rotavírus entre as crianças de áreas rurais e urbanas do distrito de Bannu Khyber Pakhtunkhwa, Paquistão. Um total de 180 amostras de fezes foi coletada de crianças menores de 5 anos de dois grandes hospitais de Bannu de janeiro a dezembro (2015). As amostras foram analisadas por reação em cadeia da polimerase transcriptase reversa (RT-PCR) para detecção de rotavírus; as amostras positivas foram posteriormente processadas para genotipagem (tipo G e P) através de PCR específica. Do total, 41 (23%) amostras foram positivas para rotavírus. Os genótipos G mais prevalentes encontrados foram: G3, G8, G9 (cada 29%), seguidos de G10 (15%) e G11 (10%). Considerando que os genótipos P prevalentes foram: P-8 (25%), P-4 e P-10 (cada 20%), P-9 (15%), seguido por P-6 e P-11 (cada 10%). Além disso, a infecção por rotavírus foi mais prevalente no verão (23,73%) e inverno (22,7%) do que na primavera (20%) e no outono (21,4%). A infecção por rotavírus apresentou alta frequência em junho (14%), outubro (8%) e novembro (6%). Conclui-se que o rotavírus é mais prevalente em crianças e vários genótipos (G e P) do rotavírus estão presentes na área de estudo. A falta de estudos, conhecimento e testes mais raros de rotavírus são as principais razões da prevalência do vírus no distrito de Bannu, Paquistão.

ABSTRACT: This study evaluated the genetic diversity and antimicrobial susceptibility of Staphylococcus aureus isolates from dairy cows in Minas Gerais, Brazil. Thirty-seven isolates from five municipalities (8 herds) were genotyped using multi-locus sequence typing (MLST) and susceptibility to 12 antimicrobial agents was tested using the disk diffusion method. High resistance rates for penicillin [75.68% (28/37)], ampicillin [70.27% (26/37)], and tetracycline [70.27% (26/37)] were detected. Multidrug resistance was observed in seven [18.92% (7/37)] isolates, and two were suggestive of Methicillin-resistant Staphylococcus aureus (MRSA). Among the 37 isolates, 33 novel sequence types (ST) and two known STs (ST126 and ST746) were identified in MLST. The clonal complexes more frequently observed were: CC97 [78.38%; (29/37)], CC1 [8.11%; (3/37)] and CC5 [5.40%; (2/37)]. Minimum‐spanning tree (MST) analysis according to data from municipalities, herds, and resistance patterns for all isolates did not show any clustering pattern. However, the MST comparing all Brazilian S. aureus isolates deposited in the PubMLST database and from this study depicted an association between the genotype and strain origin (clinical sample). Isolates from this study that belong to CC97 were close to database isolates from milk and dairy products, while those that belong to CC1 and CC5 were close to database isolates from human sources and the environment of dairy farms or industries. In conclusion, our results showed a high rate of resistance to penicillins and tetracyclines and great genetic diversity among the S. aureus isolates from bovine mastitis genotyped in the present study.


RESUMO: Este estudo teve como objetivo avaliar a diversidade genética e a suscetibilidade a antimicrobianos de cepas de Staphylococcus aureus isoladas de vacas leiteiras em Minas Gerais, Brasil. Trinta e sete cepas provenientes de cinco municípios (oito rebanhos) foram genotipadas usando a técnica multi-locus sequence typing (MLST) e a suscetibilidade a 12 antimicrobianos foi avaliada pelo método de difusão em disco. Foram detectadas altas taxas de resistência para penicilina [75,68% (28/37)], ampicilina [70,27% (26/37)] e tetraciclina [70,27% (26/37)] entre os isolados. A multirresistência foi observada em sete [18,92% (7/37)] isolados e dois foram classificados como sugestivos de Staphylococcus aureus resistente à meticilina (MRSA). Entre os 37 isolados, 33 novos sequence types (ST) e dois STs conhecidos (ST126 e ST746) foram identificados pelo MLST. Os complexos clonais mais frequentemente observados foram: CC97 [78,38%; (29/37)], CC1 [8,11%; (3/37)] e CC5 [5,40%; (2/37)]. Foi construída uma minimum‐spanning tree (MSTs) com todos isolados estudados e esta não mostrou padrão de agrupamento quando comparada com dados epidemiológicos como municípios e rebanho, do qual foi isolado, e perfis de resistência a antimicrobianos. Uma segunda MST foi construída comparando os isolados deste estudo e todas as cepas de S. aureus depositadas no banco de dados PubMLST provenientes do Brasil, que mostrou associação entre o genótipo, STs e a origem da cepa. Foi possível observar entre os isolados do estudo que, aqueles que pertenciam ao CC97, eram geneticamente mais próximos das cepas depositadas no PubMLST isoladas de leite e de produtos lácteos, enquanto aqueles que pertenciam aos CC1 e CC5 estavam mais próximos a cepas isoladas de humanos ou do ambiente de fazendas e indústrias de laticínios. Em conclusão, nossos resultados mostraram um alto índice de resistência às penicilinas e tetraciclinas e grande diversidade genética entre as cepas de S. aureus isoladas de casos de mastite bovina.

Pullorum disease is described worldwide and is caused by Salmonella enterica subspecies enterica serovar Gallinarum biovar Pullorum (S. Pullorum). S. Pullorum infection is important in commercial poultry, provoking a systemic disease with high mortality rates. Its occurrence requires notification, and when it is diagnosed in commercial breeding flocks, its eradication is demanded. The aim of this study was to report a severe outbreak of Pullorum disease in young Guinea fowl (Numida meleagris), resulting in 100% mortality of keets (n=290) within the first two weeks of age. All examined keets had enlarged liver, kidneys and spleen (5/5), and the affected tissues were submitted to histological and bacteriological examination. On histopathology, random paratyphoid nodules characterized by areas of necrosis with fibrin and a moderate infiltrate of macrophages and heterophils were observed in the liver. In kidneys, discrete areas of necrosis associated with moderate multifocal infiltrates of lymphocytes, and plasma cells were observed. In the spleen, a moderate infiltrate of macrophages was noticed. Isolation of colonies suggestive of S. Pullorum from liver and spleen was performed in selective agars and, after biochemical tests, confirmed by specific duplex-PCR. The antimicrobial susceptibility test of the isolated strain revealed resistance to only sulfamethoxazole + trimethoprim among the tested antimicrobials. The S. Pullorum isolate recovered in the present study was highly pathogenic to N. meleagris and may represent a risk to other avian species, including industrial poultry.


A pulorose é descrita mundialmente e é causada por Salmonella enterica subespécie enterica sorovar Gallinarum biovar Pullorum (S. Pullorum). A infecção por S. Pullorum é importante em aves comerciais, provocando doença sistêmica com altas taxas de mortalidade. Sua ocorrência requer notificação e quando diagnosticada em aves de criação comercial resulta na erradicação do plantel. O objetivo deste estudo foi relatar um surto grave de pulorose em filhotes de galinhas-d'Angola (Numida meleagris), resultando em 100% de mortalidade das aves (n=290) nas primeiras duas semanas de idade. Os pintinhos recebidos tinham hepato, espleno e nefromegalia (5/5). Os tecidos dos cinco indivíduos recebidos foram submetidos a exame histológico e bacteriológico. Na histopatologia, foram observados nódulos paratifoides aleatórios caracterizados por áreas de necrose com fibrina e infiltrado moderado de macrófagos e heterófilos no fígado. Nos rins, foram observadas áreas discretas de necrose associadas a infiltrados multifocais moderados de linfócitos e plasmócitos. No baço, foi observado infiltrado moderado de macrófagos. O isolamento de colônias sugestivas de S. Pullorum de fígados e baços foi realizado em ágares seletivos e, após testes bioquímicos, confirmado por duplex-PCR específico. A susceptibilidade antimicrobiana da cepa isolada revelou resistência apenas ao sulfametoxazol + trimetoprim entre os antimicrobianos testados. O isolado de S. Pullorum recuperado no presente estudo foi altamente patogênico para N. meleagris e pode representar um risco para outras espécies de aves, incluindo aves industriais.

An experiment was carried out to study the effect of different eubiotics on productive characteristics, intestinal integrity, as well as the content of enterobacteria in the cecum of broiler chickens. A completely randomized design with five treatments and 8 replicates of 25 birds each was used. In total 1000 mixed broiler chickens from Ross308 strain, one day old were obtained from a commercial hatchery. The birds were housed on concrete floors in a conventional house. A sorghum+soybean meal control diet was used, to which the additives under study were added. The treatments were distributed as follows: T1 = Control diet without antibiotic or eubiotic; T2 = T1 + bacteriophages; T3 = T1 + antibiotic; T4 = T1 + probiotic; T5 = T1 + symbiotic. The results obtained at 49 days of age for weight gain and feed conversion rate improved (p<0.05) with the addition of the antibiotic and eubiotics. A lower (p<0.05) intestinal density was observed with the probiotic. The height, width, and area of villi in duodenum was higher (p<0.05) when antibiotic and eubiotics were included. In the histological score, in duodenum, the antibiotic and eubiotics resulted with a higher score (p<0.05), associated to a physiological and controlled inflammation response that allowed improving productivity. Finally, the relative expression of enterobacteria, such as Lactobacillus salivarius, allowed associating positive changes in the microbiome and better productive parameters when including the symbiotic, with comparable results to the antibiotic when including the eubiotics.(AU)

Whereas the prevalent P genotypes were: P-8 (25%), P-4 and P-10 (each 20%), P-9 (15%), followed by P-6 and P-11 (each 10%). Moreover, Rotavirus infection was more prevalent in summer (23.73%) and winter (22.7%) than spring (20%) and autumn (21.4%). Rotavirus infection exhibited high frequency in June (14%), October (8%) and November (6%). It is concluded that Rotavirus is more prevalent in children and various genotypes (G and P) of Rotavirus are present in the study area. Lack of studies, awareness and rarer testing of Rotavirus are the principal reasons of virus prevalence in district Bannu, Pakistan.(AU)


O rotavírus é o principal agente infeccioso da gastroenterite aguda (AGE) em crianças menores de 5 anos e causa de morbidade e mortalidade significativas em todo o mundo. O estudo foi realizado para detectar a taxa de prevalência, cepa de genótipos e fatores de risco de rotavírus entre as crianças de áreas rurais e urbanas do distrito de Bannu Khyber Pakhtunkhwa, Paquistão. Um total de 180 amostras de fezes foi coletada de crianças menores de 5 anos de dois grandes hospitais de Bannu de janeiro a dezembro (2015). As amostras foram analisadas por reação em cadeia da polimerase transcriptase reversa (RT-PCR) para detecção de rotavírus; as amostras positivas foram posteriormente processadas para genotipagem (tipo G e P) através de PCR específica. Do total, 41 (23%) amostras foram positivas para rotavírus. Os genótipos G mais prevalentes encontrados foram: G3, G8, G9 (cada 29%), seguidos de G10 (15%) e G11 (10%). Considerando que os genótipos P prevalentes foram: P-8 (25%), P-4 e P-10 (cada 20%), P-9 (15%), seguido por P-6 e P-11 (cada 10%). Além disso, a infecção por rotavírus foi mais prevalente no verão (23,73%) e inverno (22,7%) do que na primavera (20%) e no outono (21,4%). A infecção por rotavírus apresentou alta frequência em junho (14%), outubro (8%) e novembro (6%). Conclui-se que o rotavírus é mais prevalente em crianças e vários genótipos (G e P) do rotavírus estão presentes na área de estudo. A falta de estudos, conhecimento e testes mais raros de rotavírus são as principais razões da prevalência do vírus no distrito de Bannu, Paquistão.(AU)

This study evaluated the genetic diversity and antimicrobial susceptibility of Staphylococcus aureus isolates from dairy cows in Minas Gerais, Brazil. Thirty-seven isolates from five municipalities (8 herds) were genotyped using multi-locus sequence typing (MLST) and susceptibility to 12 antimicrobial agents was tested using the disk diffusion method. High resistance rates for penicillin [75.68% (28/37)], ampicillin [70.27% (26/37)], and tetracycline [70.27% (26/37)] were detected. Multidrug resistance was observed in seven [18.92% (7/37)] isolates, and two were suggestive of Methicillin-resistant Staphylococcus aureus (MRSA). Among the 37 isolates, 33 novel sequence types (ST) and two known STs (ST126 and ST746) were identified in MLST. The clonal complexes more frequently observed were: CC97 [78.38%; (29/37)], CC1 [8.11%; (3/37)] and CC5 [5.40%; (2/37)]. Minimum­spanning tree (MST) analysis according to data from municipalities, herds, and resistance patterns for all isolates did not show any clustering pattern. However, the MST comparing all Brazilian S. aureus isolates deposited in the PubMLST database and from this study depicted an association between the genotype and strain origin (clinical sample). Isolates from this study that belong to CC97 were close to database isolates from milk and dairy products, while those that belong to CC1 and CC5 were close to database isolates from human sources and the environment of dairy farms or industries. In conclusion, our results showed a high rate of resistance to penicillins and tetracyclines and great genetic diversity among the S. aureus isolates from bovine mastitis genotyped in the present study.


Este estudo teve como objetivo avaliar a diversidade genética e a suscetibilidade a antimicrobianos de cepas de Staphylococcus aureus isoladas de vacas leiteiras em Minas Gerais, Brasil. Trinta e sete cepas provenientes de cinco municípios (oito rebanhos) foram genotipadas usando a técnica multi-locus sequence typing (MLST) e a suscetibilidade a 12 antimicrobianos foi avaliada pelo método de difusão em disco. Foram detectadas altas taxas de resistência para penicilina [75,68% (28/37)], ampicilina [70,27% (26/37)] e tetraciclina [70,27% (26/37)] entre os isolados. A multirresistência foi observada em sete [18,92% (7/37)] isolados e dois foram classificados como sugestivos de Staphylococcus aureus resistente à meticilina (MRSA). Entre os 37 isolados, 33 novos sequence types (ST) e dois STs conhecidos (ST126 e ST746) foram identificados pelo MLST. Os complexos clonais mais frequentemente observados foram: CC97 [78,38%; (29/37)], CC1 [8,11%; (3/37)] e CC5 [5,40%; (2/37)]. Foi construída uma minimum­spanning tree (MSTs) com todos isolados estudados e esta não mostrou padrão de agrupamento quando comparada com dados epidemiológicos como municípios e rebanho, do qual foi isolado, e perfis de resistência a antimicrobianos. Uma segunda MST foi construída comparando os isolados deste estudo e todas as cepas de S. aureus depositadas no banco de dados PubMLST provenientes do Brasil, que mostrou associação entre o genótipo, STs e a origem da cepa. Foi possível observar entre os isolados do estudo que, aqueles que pertenciam ao CC97, eram geneticamente mais próximos das cepas depositadas no PubMLST isoladas de leite e de produtos lácteos, enquanto aqueles que pertenciam aos CC1 e CC5 estavam mais próximos a cepas isoladas de humanos ou do ambiente de fazendas e indústrias de laticínios. Em conclusão, nossos resultados mostraram um alto índice de resistência às penicilinas e tetraciclinas e grande diversidade genética entre as cepas de S. aureus isoladas de casos de mastite bovina.

Studies on aggressiveness of parasitoids, as assessed by their parasitism against pests, used in biological-control programs are highly important to select the most suitable species and/or strain to control insect pests. The present study investigated whether the egg parasitoid Trichogramma galloi Zucchi, an efficient control agent for sugarcane borer Diatraea saccharalis (Fabricius) in Brazil, could be replaced by Trichogramma atopovirilia Oatman & Platner, a parasitoid easier to mass-produce, since it has been found parasitizing D. saccharalis eggs in the warmest region of Brazil and Argentina. Three strains of the genus Trichogramma were compared: T. atopovirilia (ATP strain) reared on a factitious host Anagasta kuehniella (Zeller); T. atopovirilia isoline ATP-I, reared on D. saccharalis eggs for six generations; and T. galloi, reared on A. kuehniella eggs. We measured parasitism of each strain for 72 h and for the entire life span, parasitism rate per cluster of D. saccharalis eggs, number of parasitoids emerged (parasitism viability), and parasitoid life span. The results confirmed that T. galloi is the best species for D. saccharalis control, showing higher control potential, since parasitism and emergence rate were higher for this species. Although T. atopovirilia ATP-I performed reliably in all parameters, T. galloi exceeded and was the most indicated for mass-rearing in control programs for sugarcane borer.

The use of fungi to obtain chitin and chitosan has advantages such as handling, extraction, and production control, thus allowing to generate high quality chitosan. This study aimed to isolate and identify strains of Cunninghamella spp., and assess the production of biomass, chitin, and chitosan. We determined the macroscopic and microscopic phenotypic aspects of the superiors, mycelial growth rate index, consumption of glucose and nitrogen, as well as the pH variation, and the production of biomass, chitin, and chitosan. After assessing the mycelial growth speed index, considered fast, we found that the UFT-Ce08 and UFT-Ce09 isolates showed a higher growth speed. Therefore, we assessed the sporulation of the isolates, and all of them reached the concentration of 106spores mL-1in the period of 96 hours. The YPD was considered ideal for biomass production because it promotes an efficient consumption and synthesis of organic compounds by microorganisms. The assessment of the biomass production and the chitin and chitosan yield of nine isolates and the reference strain showed that the UFT-Ce08 isolate had the highest amount of biomass, the UFT-Ce11 isolate had the highest chitin yield, and despite having the second smallest biomass, the UFT-Ce09 isolate had thehighest chitosan yield. Seven isolates showed higher chitosan yield than the reference strain. Therefore, chitosan production from this fungus can be further optimized to improve yield on a large-scale production. Chitosan production from this fungus can be further optimized to improve yield in large-scale production.(AU)

The aim of this work was to verify the efficiency of different isolates of Trichoderma spp.on the control of Sclerotiniasclerotiorum, Sclerotiumrolfsii and Sclerotiumcepivorum, and the influence they pose on the conidia production of Trichoderma spp.For mycelial growth, discs with inoculum of phytopathogens were placed on the center of the Petri dishes followed by the addition of two Trichodermasp. discs on the opposite sides of the plate after 24hours. Every 12hoursdata were collected from colonies diameters and used for the analyses of Mycelial Growth Index (MGI) and Area Under the Curve of Mycelial Growth (AUCMG). The analyses were performed by a completely randomized design with two controls, a negative one without Trichodermasp. and one with acommercial strain of Trichodermaharzianum. Spore solution for evaluation of conidia production were made by adding 10 mL of distilled water and scratching the surface of the colonies. For S. cepivorum, all Trichoderma spp. strains reduced both indexes tested. However, while for MGI S. sclerotiorum also presented some reduction on the growth rate, the total area of this fungus was not affected. Sclerotiumrolfsii strains of Trichodermasp. from Lages and Curitibanosshowed an effect on the reduction of AUCMG of this fungus, although none of the Trichodermaaffected the growth rate of this phytopathogen. On the presence of S. sclerotiorumand S. cepivorum, none of the Trichodermaspp. showed any difference on conidia production when compared among themselves, nonetheless we did notice that on the presence of S. cepivorum, the strain from Rio do Sul retained its reproductive ability compared to control. Results obtained from this research can demonstrate the importance of biocontrol agents against different plant pathogens since it might have a specific antagonist-pathogen relation.(AU)

This study analyzed the Sardo Negro breed pedigree (41,521 animals registered from 1958 to 2019) to determine its structure, evolution, and genetic variability (GV). The population genetic parameters evaluated were effective number of founders (fe) and ancestors (fa), pedigree integrity, additive genetic relationship (AGR); number of complete generations (NCG), maximum generations traced (NMGT), and equivalent complete generations (NECG); effective population size (Ne), inbreeding coefficient (F), and generation interval (GI). The average GI was 7.45 years. A total of 7,804 founders and 4,856 ancestors were identified for a fe of 185 and a fa of 97. The average and maximum values of NCG, NECG, and NMGT were 1.6 and 5.0, 2.5 and 6.5, 4.3 and 12, with Ne estimates of 15.9, 25.9, and 69.0, respectively. The increase in F, linked to Ne, ranged from 0.72% to 3.1% per generation. The average values for F and AGR were 3.6% and 1.0%, respectively. The proportion of inbred individuals was 32.0%, with F values ranging from 0.01 to 62.2% and an average of 11.3%. The rate of inbred population was 1.3% per year. The annual rate of AGR was 0.04%. For the continuity and projection of the breed, the evolution of F as a function of Ne and the possible implications of the selection schemes must be considered. The genetic variability sustained over time results from the Ne.


Os objetivos deste estudo foram analisar o pedigree (41.521 registros de 1958 a 2019) da raça Sardo Negro para avaliar a estrutura, evolução e variabilidade genética (VG) da população. Os parâmetros genéticos populacionais utilizados foram: número efetivo de fundadores (fe) e ancestrais (fa); integridade do pedigree; relação genética aditiva (RGA); número de gerações completas (NGC), máximo plotado (NGT) e equivalentes (NGE); tamanho efetivo (Ne); consanguinidade (F); intervalo geracional (IG). O IG médio foi de 7,45 anos. Foram identificados 7.804 fundadores e 4.856 ancestrais, para fe 185 e 97 na fa. As médias e máximas para NGC, NGE e NGT foram 1,6 e 5,0, 2,5 e 6,5, 4,3 e 12, com estimativas de Ne 15,9, 25,9 e 69,0, respectivamente. O aumento de F, vinculado ao Ne, ficou na faixa de 0,72% a 3,1% por geração. A média para F 3,6% e 1,0% em RGA; a proporção de consanguíneos foi de 32,0%, com F na faixa de 0,01 a 62,2% e média de 11,3%. A taxa da população consanguínea foi de 1,3% ao ano. No RGA, a taxa ao ano era de 0,04%. Para a continuidade e projeção da raça, deve-se considerar a evolução de F em função de Ne e as possíveis implicações dos esquemas de seleção. A variabilidade genética sustentada ao longo do tempo resulta do Ne.

The growth of generations of the Genomar Supreme Tilapia (GST) strain, specifically G20 and G25, was evaluated. Tilapias 8 g were reared in a recirculating aquaculture system with 0.25 m³ tanks, 80 fish m-³, with four replicates. During growth, eight fish from each tank were weighed and measured at day 1, 30, 60, 90, 120, 150, 180 and 210. Survival, weight gain, feed conversion and batch homogeneity were determined. Weight-age data were fit to Gompertz model. In addition, absolute and relative growth rates and weight and age at inflection were determined. Final weight showed 26.7% higher in G25 when compared to G20 (920.05 and 725.87 g, respectively). The feed conversion and homogeneity indexes were better in G25 than G20. The estimate of asymptotic weight was higher in G25 (1202.0 g) when compared to G20 (912.7 g). G20 presented smaller weight (335.76 g), age (108.87 days) and absolute growth rate (4.87 g day-1) when compared with G25 (442.19 g, 113.77 days and 6.41 g day-1). Carcass characteristics were similar, but G25 presented about 25% higher fillet weight than G20. After five years in the GST breeding program, results indicated that a sustained improvement of harvest weight was achieved, as well as 6 to 10% gain in performance by generation.(AU)

Increasing resistance to antimicrobials including methicillin has become an important public concern. One hundred and eight (108) S. aureus strains isolated from a total of 572 clinical and animal products samples, were investigated for their biofilm capability, methicillin resistance, enterotoxin genes, and genetic diversity. Although only one strain isolated from raw retail was found as a strong biofilm producer, the percentage of antimicrobial resistance pattern was relatively higher. 17.59% of S. aureus strains tested in this study were resistant to cefoxitin and identified as methicillin-resistant S. aureus (MRSA) isolates. mecA and mecC harboring S. aureus strains were detected at a rate of 2.79% and 0.93%, respectively. In addition, staphylococcal enterotoxin genes including Sea, Seb, Sec, and Sed genes were found to be 18.5%, 32.4%, 6.5% and 3.7%, respectively. The phylogenetic relationship among the isolates showed relationship between joint calf and cow milk isolates. Multi locus sequence typing (MLST) revealed three different sequence types (STs) including ST84, ST829, and ST6238. These findings highlight the development and spread of MRSA strains with zoonotic potential in animals and the food chain throughout the world.

Cyanobacterial blooms can cause severe ecological and health problems in drinking water reservoirs. To alleviate this problem, allelopathically active submerged macrophytes can be used to reduce cyanobacterial growth. Accordingly, this study aimed to evaluate the sensitivity of strains of the Microcystis aeruginosacomplex isolated from reservoirs with the presence and absence of submerged macrophytes to the allelochemicals of Ceratophyllum demersum.A coexistence experiment was carried out between the submerged macrophyte C. demersum and four Microcystisstrains, with two treatments for each strain, one in coexistence with the submerged macrophyte (7 g L-1) and control (in the absence of the macrophyte). Two strains of M. aeruginosa(BMIUFRPE-06 and BMIUFRPE-07) and two of M. panniformis(BMIUFRPE-08 and BMIUFRPE-09) were used, which were isolated from Cajueiro (with submerged macrophytes) and Tapacurá (without submerged macrophytes) reservoirs, respectively. The biomass of Microcystisstrains from the reservoir without macrophytes (BMIUFRPE-08 and BMIUFRPE-09) was significantly inhibited in 96% (T-test: p 0.05; growth rate -ANOVA: p > 0.05). These results suggest that strains isolated from environments with submerged macrophytes are less sensitive to allelochemicals of these plants,as these strains may be adapted to the coexistence with submerged macrophytes.

The objective of this study was to compare the antimicrobial activity of macrophages and serum in laying hen (MM, CC, and CCc) and broiler chicken lineages (TT and LL). Macrophages were evaluated for phagocytic and antimicrobial activity. Microbicidal serum activity was evaluated by the resistance test for serum and the agar test. The results showed that phagocytic activity was higher in males of the MM strain, with 13% of macrophages presenting phagocytosis, while the other lineages studied, and even female MM, presented a rate of 6% of phagocytic cells. However, antimicrobial activity in macrophages from males of CCc lineage and females of TT lineage were higher, eliminating more than 30% of the Salmonella enterica inoculum, while in the other strains, the results were similar, with inoculum reduction below 30%. In the serum resistance assay, female laying lines presented higher antibacterial activity than female broiler lines. In the trials to evaluate the microbicide activity of the serum, females of both broiler and laying lineages presented higher performance when compared with males of the same lineage. Females of laying hen lines (MM and CC) present a greater antibacterium activity than males. These results can contribute to a better understanding of the immune response in broiler chicken and laying hen lineages, to aid development of lineages of birds more resistant to pathogens.

Resumo

Cyanobacterial blooms can cause severe ecological and health problems in drinking water reservoirs. To alleviate this problem, allelopathically active submerged macrophytes can be used to reduce cyanobacterial growth. Accordingly, this study aimed to evaluate the sensitivity of strains of the Microcystis aeruginosacomplex isolated from reservoirs with the presence and absence of submerged macrophytes to the allelochemicals of Ceratophyllum demersum.A coexistence experiment was carried out between the submerged macrophyte C. demersum and four Microcystisstrains, with two treatments for each strain, one in coexistence with the submerged macrophyte (7 g L-1) and control (in the absence of the macrophyte). Two strains of M. aeruginosa(BMIUFRPE-06 and BMIUFRPE-07) and two of M. panniformis(BMIUFRPE-08 and BMIUFRPE-09) were used, which were isolated from Cajueiro (with submerged macrophytes) and Tapacurá (without submerged macrophytes) reservoirs, respectively. The biomass of Microcystisstrains from the reservoir without macrophytes (BMIUFRPE-08 and BMIUFRPE-09) was significantly inhibited in 96% (T-test: p < 0.01) and 74% (T-test: p< 0.05), when compared to the control, respectively, with lower values of growth rates (ANOVA: p < 0.05). The strains isolated from the reservoir with macrophytes (BMIUFRPE-06 and BMIUFRPE-07) showed a growth delay (biomass reductions of 44 and 58%, respectively) in the coexistence treatment but without significant difference from the control on the sixth day of the experiment (biomass -T-test: p > 0.05; growth rate -ANOVA: p > 0.05). These results suggest that strains isolated from environments with submerged macrophytes are less sensitive to allelochemicals of these plants,as these strains may be adapted to the coexistence with submerged macrophytes.(AU)
