Resumo

A expectativa de vida dos animais de companhia vem aumentando devido as mudanças comportamentais e de cuidados dos tutores. Em analogia, com as idades cada vez mais avançadas, é comum o desenvolvimento de doenças associadas, sendo o tumor o mais comum entre elas. Todavia, as neoplasias mais comuns na clínica de pequenos animais estão associadas ao sistema tegumentar. O carcinoma de células escamosas (CCE), ou carcinoma espinocelular, é uma neoplasia de epitélio, maligno, de crescimento lento e de baixo potencial metastático. Sua etiologia ainda não é precisamente conhecida e a causa exógena mais comum e descrita pela literatura é a exposição à luz ultravioleta, gerando consequentes lesões nas estruturas genéticas e imunogênicas na pele. Além disso, essas lesões apresentam-se de forma mais comum em animais de pelagem clara, com grande exposição solar e em área anatômicas hipopigmentadas. O prognóstico varia de acordo com a localização e o estágio clínico, sendo favorável o diagnóstico realizado precocemente, e o tratamento consiste na avaliação clínica seguida do protocolo adequado O presente relato de caso tem como objetivo principal reunir e discutir informações associadas sobre o carcinoma de células escamosas em caninos e felinos, abordando aspectos clínicos e patológicos, a fim de facilitar o raciocínio sobre o CCE, desde sua abordagem inicial, até seu diagnóstico final e estadiamento.(AU)


The life expectancy of companion animals has been increasing due to behavioral and care changes of the guardians. In analogy, with the increasingly advanced ages, the development of associated diseases is common, the tumor being the most common among them. However, the most common neoplasms in the small animal clinic are associated with the integumentary system. Squamous cell carcinoma (SCC), or squamous cell carcinoma, is a malignant, slow-growing, low-potential metastatic epithelial neoplasm. Its etiology is not yet precisely known and the most common exogenous cause described by the literature is exposure to ultraviolet light, generating consequent lesions on the genetic and immunogenic structures in the skin. In addition, these lesions are more common in light-haired animals, with high exposure to the sun, and in hypopigmented anatomical areas. The prognosis varies according to the location and clinical stage, being favorable to the early diagnosis, and the treatment consists in the clinical evaluation followed by the appropriate protocol. The present case report has as its main objective to gather and discuss associated information about squamous cell carcinoma in canines and cats, addressing clinical and pathological aspects, in order to facilitate the reasoning about the CCE, from its initial approach, to its final diagnosis and staging.(AU)


La esperanza de vida de los animales de compañía ha ido en aumento debido a los cambios de comportamiento y cuidado de los guardianes. Por analogía, con las edades cada vez más avanzadas, el desarrollo de enfermedades asociadas es común, siendo el tumor el más común entre ellos. Sin embargo, las neoplasias más comunes en la clínica de animales pequeños se asocian con el sistema tegumentario. El carcinoma epidermoide (CCE), o carcinoma epidermoide, es una neoplasia epitelial metastásica maligna de crecimiento lento y bajo potencial. Su etiología aún no se conoce con precisión y la causa exógena más común descrita por la literatura es la exposición a la luz ultravioleta, generando lesiones consecuentes en las estructuras genéticas e inmunogénicas de la piel. Además, estas lesiones son más comunes en animales de pelo claro, con alta exposición al sol, y en áreas anatómicas hipopigmentadas. El pronóstico varía según la localización y el estadio clínico, siendo favorable al diagnóstico precoz, y el tratamiento consiste en la evaluación clínica seguida del protocolo adecuado. El presente reporte de caso tiene como objetivo principal reunir y discutir información asociada sobre el carcinoma epidermoide en caninos y gatos, abordando aspectos clínicos y patológicos, con el fin de facilitar el razonamiento sobre el CCE, desde su abordaje inicial, hasta su diagnóstico final y estadificación.(AU)

Resumo

Background: Squamous cell carcinoma (SCC) is a malignant neoplasm associated with prolonged exposure to ultraviolet light and lack of skin pigment in white-coated animals. The disease diagnosis is based on the animal's history, clinical signs and histopathology of the samples collected. The therapeutic alternatives are diverse. Among them, one of the most used is the metronomic chemotherapy which is based on the use of cytostatics drugs, applied in low and frequent doses for a longer period of time. One of the mechanisms of action of this type of therapy is the decrease in angiogenesis. Non-steroidal anti-inflammatory drugs (NSAIDs) are simultaneously used to maximize the angiogenic effect. The objective of this study is to report the results of a treatment accomplised with metronomic chemotherapy (cyclophosphamide with meloxicam) in a domestic feline which presented a squamous cell tumor. Case: A 7-year-old female cat, mixed breed, not neutered, weighing 3.5 kg, was assisted at the Veterinary Specialties Teaching Clinic (CEEV) of the Technical University of Machala (UTMACH), presenting bilateral ulcerative lesion in the nasal plane for 5 months. Chronic exposure to sunlight is referred to in the patient's anamnesis. Aspiration cytology performed in the patient revealed an inflammatory process with a predominance of neutrophils and macrophages, and showed absence of neoplastic cells. A treatment based on antibiotics and corticosteroids was adopted. As there were no favorable results, an excisional biopsy procedure was chosen to collect the affected tissues. The result of the histopathological analysis was a level 2 squamous cell carcinoma (SCC). The initial treatment consisted of surgical removal of the tumor and later the use of metronomic chemotherapy based on Cyclophosphamide, Meloxicam, DHA (docosahexaenoic acid) and Sucralfate for 5 months was adopted. At the beginning of the second phase of treatment, the patient developed a macula that progressively diminished. Currently, the patient does not have any recurrence of the diagnosed neoplasm. Discussion: The diagnosis of squamous cell carcinoma was based on clinical and histopathological findings. The macroscopic lesions were crusted ulcers that do not heal in the nasal plane and auricular pavilion, as reported in the literature. SCC is characterized by the infiltration of squamous cells in the dermal layer of the skin, forming epithelial islands with keratin pearls. The histopathological findings of this case present the same characteristics. The climatic conditions of Ecuador, high temperatures with greater incidence of sunlight, hypopigmented areas of the animal's body and chronic exposure to solar radiation are risk factors present in this case that foster the development of this malignant neoplasm. There are multiple treatments to be introduced in this sort of pathology. In this case, surgical removal of the tumor was adopted, followed by metronomic chemotherapy based on cyclophosphamide at 10 mg/m2 and meloxicam at a 0.05 mg/kg per day for a period of time of five months, obtaining favorable results and absence of recurrence. Despite metronomic chemotherapy being easy to administer, with fewer side effects, which includes a better acceptance of care takers who are reluctant to administer conventional chemotherapy, further studies are still necessary in what concerns certain variables such as age, carcinoma level, appropriate dosages and drugs according to each neoplasm.

Background: Although Squamous Cell Carcinoma (SCC) is most commonly seen in the epidermis, this malignant neoplasm can manifest in various other tissues. A wide range of factors may be related to the development of SCC in the cornea, with continuous exposure to ultraviolet radiation and chronic friction to the ocular surface being the most accepted theories. In addition to surgical procedures, the use of mitomycin C in the topical treatment of corneal SCC has shown good results in therapeutic management. Thus, the objective of the current work is to report the satisfactory response observed in the use of mitomycin C in a case of SCC in the cornea of a dog. Case: A 9-year-old male Pug dog was attended by the Veterinary Ophthalmology and Microsurgery Sector (SOMVET) of the University Veterinary Hospital (HVU) of the Federal University of Santa Maria (UFSM) with Squamous Cell Carcinoma (SCC) in the cornea of the left eye, which had recurred after a previous surgical intervention. Secretion and discomfort in the left eye were noted during clinical examination. In addition, both eyes presented pigmentary keratitis in the medial corner. Surgical excision of the neoplasm was performed using the surgical technique of anterior lamellar keratectomy. In the immediate postoperative period, topical therapy was started with eye drops based on mitomycin C at 0.02%, for a period of 28 consecutive days. The use of this chemotherapy drug in the topical form was intended to minimize the chances of recurrence of the clinical condition, since the patient is predisposed to this alteration. In addition, supportive therapy was implemented to improve patient comfort, consisting of the use of tobramycin-based eye drops (6 times a day, for 14 days), atropine 1% eye drops (BID, for 3 days), lubricating eye drops based on sodium hyaluronate (3 times a day, for continuous use), and immunomodulatory eye drops based on tacrolimus 0.02% (twice a day, for continuous use). The patient was evaluated 7, 14, 21, and 28 days after surgery, when good results were observed. One year after treatment, the dog was still showing no signs of recurrence of the treated clinical condition. Discussion: It is known that chronic friction on the surface of the cornea predisposes to the diagnosis of corneal SCC. Accordingly, in the clinical examination of the patient in this report, the presence of bilateral pigmentary keratitis was observed, an alteration that leads to a framework of chronic aggression to the cornea. Surgical treatment is recommended to remove the tumor mass in the ocular region. However, in cases of corneal SCC, the literature highlights that recurrence after surgical excision is a common factor. As the patient in this study arrived at the clinic with a history of recurrence, topical therapy with mitomycin C associated with surgical treatment was instituted. This chemotherapy drug has shown encouraging results in the treatment of some neoplasms, especially SCC. Its use in this case supported previous findings, pointing to a satisfactory result in which the patient had no recurrences after one year of follow-up. In addition, the concentration and frequency used of the active ingredient did not lead to adverse effects in the short or medium term.

Resumo

Squamous cell carcinoma (SCC) is a common neoplastic skin disease that is highly prevalent in tropical countries. As the skin has a variety of cells, overexposure to environmental factors, such as ultraviolet light, can affect this organ, resulting in malignancies, such as cutaneous SCC, hemangioma, and hemangiosarcoma. SCC arises from keratinocytes in the skin and is locally invasive with low metastatic rates, commonly affecting unpigmented skin in sites with high exposure to sunlight, such as ventral regions. SCC has a variable etiology that is not well understood. Therefore, literature review aimed to critically evaluate the risk factors involved in the SCC development.(AU)

Resumo

Purpose: Fluorescence spectroscopy techniques have been investigated aiming to reduce the invasiveness of methods for investigation of tissue. In transplantation procedures, it may offer the possibility of a complementary technique for the monitoring of liver grafts' conditions prior to and during the transplantation procedure stages involving cold perfusion. The objective of this study was to evaluate fluorescence spectroscopy under violet light excitation (408 nm) for the monitoring of clinical hypothermic liver transplantation procedures. Methods: Organ grafts were monitored from before the removal of the donor's body to 1 h after the implant into the receptor's body. Fluorescence spectroscopy was assessed over five stages within these transplant stages. Results: The study provided evidence of a correlation between fluorescence information collected during liver grafts transplantation and the survival of patients. Conclusions: Fluorescence spectroscopy can become a tool to monitor transplantation grafts, providing objective information for the final decision of surgeons to use organs.

Iron is a fundamental microelement for human life; however, deficiencies or excesses of these metal ions can cause severe complications and mortality. Chelators are compounds that bind and inhibit iron. Ultraviolet-visible (UV-vis) spectrophotometric methods are key analytical tools in the identification of chemical entities, with the benefits of having good precision and accuracy, and the equipment being easily available as well as quick and simple to implement. In this study, we aimed to provide an alternative, cheaper method for the quantification of iron ion chelation by substituting ferrozine for gallic acid and validating its use with UV-vis according to official ANVISA and ICH guidelines. The parameters assessed were specificity, linearity, precision, accuracy, robustness, and finally, the percentage of iron ions chelating was calculated. The results demonstrated that this method was accurate, simple, specific, selective, precise, and reproducible, and was successfully validated for the determination of iron ions chelating. The percentage of iron ions chelating, promoted by the standard chelator EDTA, was 45% and 47% for Fe2+ and Fe3+, respectively. It is concluded that this new method is beneficial in terms of its simplicity, rapidness, low cost, and the fact that it produces very low levels of dangerous residues.(AU)

Abstract Background: Acylpolyamines are one of the main non-peptide compounds present in spider venom and represent a promising alternative in the search for new molecules with antimicrobial action. Methods: The venom of Acanthoscurria natalensis spider was fractionated by reverse-phase liquid chromatography (RP-HPLC) and the antimicrobial activity of the fractions was tested using a liquid growth inhibition assay. The main antimicrobial fraction containing acylpolyamines (ApAn) was submitted to two additional chromatographic steps and analyzed by MALDI-TOF. Fractions of interest were accumulated for ultraviolet (UV) spectroscopy and ESI-MS/MS analysis and for minimum inhibitory concentration (MIC) and hemolytic activity determination. Results: Five acylpolyamines were isolated from the venom with molecular masses between 614 Da and 756 Da, being named ApAn728, ApAn614a, ApAn614b, ApAn742 and ApAn756. The analysis of UV absorption profile of each ApAn and the fragmentation pattern obtained by ESI-MS/MS suggested the presence of a tyrosyl unit as chromophore and a terminal polyamine chain consistent with structural units PA43 or PA53. ApAn presented MIC between 128 µM and 256 µM against Escherichia coli and Staphylococcus aureus, without causing hemolysis against mouse erythrocytes. Conclusion: The antimicrobial and non-hemolytic properties of the analyzed ApAn may be relevant for their application as possible therapeutic agents and the identification of an unconventional chromophore for spider acylpolyamines suggests an even greater chemical diversity.

Resumo

Purpose: The active melanocytes in the skin were affected by hormones and ultraviolet (UV) irradiation. Licorice zinc has a whitening effect, which may have a prominent potential in the treatment of pigmented skin disease. Methods: Modeling chloasma C57BL/6J mice by daily progesterone injection (15 mg/kg) and ultraviolet B (UVB) irradiation (λ = 312 nm, 2 h/day) for 30 days. Then, mice were given 0.65, 1.3, and 2.6 (g/kg) of licorice zinc and tranexamic acid 250 mg daily by oral administration for 14 days, respectively. Hematoxylin and eosin and Fontana-Masson staining, and Western blotting (WB) were performed to test the inhibitory of melanogenesis and activation of c-Jun-N-terminal (JNK)/p38 mitogen-activated protein kinases (MAPK) for licorice zinc. Melanogenesis was induced by α-melanocyte-stimulating hormone in vitro. Cell counting kit-8, melanin content determination, and WB were performed to verify the inhibitory effect of licorice zinc on melanogenesis. Results: The present study showed that licorice zinc decreased melanin formation, cutaneous tissue injury, and the phosphorylation of JNK and P38MAPK, which was caused by UVB irradiation in vivo. In vitro, licorice zinc showed opposite effects from JNK/p38 activator. Meanwhile, tyrosinase-related protein-1, tyrosinase, and microphthalmia-associated transcription factor were decreased too. Conclusions: Licorice zinc induced a decrease in melanin synthesis by inhibiting the JNK and the P38MAPK signaling pathway, suggesting licorice zinc is a potential agent of anti-chloasma.

Abstract Many pathogenic strains have acquired multidrug-resistant patterns in recent a year, which poses a major public health concern. The growing need for effective antimicrobial agents as novel therapies against multidrug-resistant pathogens has drawn scientist attention toward nanotechnology. Silver nanoparticles are considered capable of killing multidrug-resistant isolates due to their oligo-dynamic effect on microorganisms. In this research study NPs were synthesized using the gram-positive bacteria Lactobacillus bulgaricus and its activity against selected pathogenic strains. Lactobacillus bulgaricus pure cultures were isolated from raw milk and grown in De Man, Rogasa, and Sharp broth for synthesis of nanoparticles. Lactobacillus bulgaricus culture was centrifuged and Cell- free supernatant of it was employed with aqueous silvery ions and evaluated their antibacterial activities against bacterial strains i.e. Staphylococcus aureus, Staphylococcus epidermidis and Salmonella typhi using agar well diffusion assay. Antibiotic profiling against selected pathogenic strains were also conducted using disc diffusion method. The synthesis and characterization of silver nanoparticles were monitored primarily by the conversion of the pale-yellow color of the mixture into a dark-brown color and via ultraviolet-visible absorption spectroscopy and Scanning electron microscopy respectively. The result showed that that AgNPs with size (30.65-100 nm) obtained from Lactobacillus bulgaricus were found to exhibit antibacterial activities against selected bacterial strains. Taken together, these findings suggest that Lactobacillus bulgaricus has great potential for the production of AgNPs with antibacterial activities and highly effective in comparison to tested antibiotics.


Resumo Muitas cepas patogênicas adquiriram padrões multirresistentes nos últimos anos, o que representa um grande problema de saúde pública. A crescente necessidade de agentes antimicrobianos eficazes como novas terapias contra patógenos multirresistentes atraiu a atenção dos cientistas para a nanotecnologia. As nanopartículas de prata são consideradas capazes de matar isolados multirresistentes por causa de seu efeito oligodinâmico em microrganismos. Neste estudo de pesquisa, as NPs foram sintetizadas usando a bactéria Gram-positiva Lactobacillus bulgaricus e sua atividade contra cepas patogênicas selecionadas. Culturas puras de Lactobacillus bulgaricus foram isoladas do leite cru e cultivadas em caldo De Man, Rogasa e Sharp para síntese de nanopartículas. A cultura de Lactobacillus bulgaricus foi centrifugada, e o sobrenadante livre de células foi empregado com íons prateados aquosos, avaliando-se suas atividades antibacterianas contra cepas bacterianas, isto é, Staphylococcus aureus, Staphylococcus epidermidis e Salmonella typhi usando ensaio de difusão em poço de ágar. O perfil de antibióticos contra cepas patogênicas selecionadas também foi conduzido usando o método de difusão em disco. A síntese e a caracterização das nanopartículas de prata foram monitoradas principalmente pela conversão da cor amarelo-pálida da mistura em uma cor marrom-escura e por espectroscopia de absorção visível e ultravioleta e por microscopia eletrônica de varredura, respectivamente. O resultado mostrou que AgNPs com tamanho de 30,65-100 nm, obtidas de Lactobacillus bulgaricus, exibiram atividades antibacterianas contra cepas bacterianas selecionadas. Tomados em conjunto, esses achados sugerem que o Lactobacillus bulgaricus tem um grande potencial para a produção de AgNPs com atividades antibacterianas e altamente eficazes em comparação aos antibióticos testados.

El albinismo es una anomalía genética recesiva en la que la produccíon de la melanina está reducida o ausente, lo que interfiere com la  pigmentación de la piel, pelaje y los ojos de los individuos. Se considera un fenómeno natural raro, principalmente en animales salvajes. Los albinos son muy susceptibles al daño de la piel por el sol, ya que la exposición acumulada a los rayos ultravioleta (UV) puede ser dañina. Este reporte discute la ocurrencia de seborrea seca en un tapir albino (Tapirus terrestris), mantenida bajo cuidado humano. La seborrea seca es una enfermedad crónica de la piel en la que falla la queratinización de la epidermis, formándose descamaciones excesivas y alterando la oleosidad de la piel y la pelaje. El tratamiento clínico instituido se basó en la aplicación tópica de uma solución dermatológica hidratante en spray y em la modificación del recinto para reducir la incidencia de la luz solar sobre el individuo.  El resultado fue satisfactorio y tuvo como ayuda un miscroscopio digital portátil que se utilizó periódicamente hasta la recuperación del animal.


Albinism is a recessive genetic disorder in which melanin prodution is reduced or absent, interfering with pigmentation of the skin, hair and eyes of individuals. It is considered to be of rare natural occurrence, mainly in wild animals. Albino individuals are highly susceptible to skin damage caused by the sun, once cumulative exposure to ultraviolet (UV) rays can be harmful. This report discusses the occurrence of dry seborrhea in an albino lowland tapir (Tapirus terrestris), kept under human care. Dry seborrhea is a chronic skin disease in which there is a failure in the keratinization of the epidermis, forming excessive scales and altering the oiliness of the skin and hair. The clinical treatment instituted was based on topical application of a moisturizing dermatological spray solution and on changes made to the enclosure in order to reduce the incidence of sunlight on the individual. The result was satisfactory and had as na aid a portable digital microscope that was used periodically until the recovery of the animal.


O albinismo é uma desordem genética recessiva em que a produção de melanina é reduzida ou ausente, o que interfere na pigmentação da pele, pelos e olhos dos indivíduos. É considerada de rara ocorrência natural, principalmente em animais selvagens. Indivíduos albinos são altamente suscetíveis a injúrias na pele causadas pelo sol, já que a exposição aos raios ultravioletas (UV) de forma cumulativa pode ser prejudicial. Este relato discorre sobre a ocorrência de seborreia seca em uma anta albina (Tapirus terrestris), mantida sob cuidados humanos. A seborreia seca é uma enfermidade cutânea de caráter crônico em que ocorre falha na queratinização da epiderme, formando excessivamente escamas e alterando a oleosidade da pele e pelos. O tratamento clínico instituído foi baseado na aplicação tópica de solução dermatológica hidratante spray e em mudanças realizadas no recinto a fim de diminuir a incidência de luz solar no indivíduo. O resultado foi satisfatório e teve como auxílio um microscópio digital portátil que foi utilizado periodicamente até a recuperação do animal.

Purpose: Poultry by-products can contribute as an innovative natural source for the development of composites based on polymers and minerals aiming at bone regeneration. The objective of this study was the physicochemical and biological characterization of collagen-based hydrogels crosslinked with ultraviolet (UV)-riboflavin. Methods: Pure hydrogels of 100% collagen (G1) or hybrid hydrogels, 90% collagen:10% apatite (G2), 90% collagen:10% nanokeratin (G3), and 90% collagen:5% apatite:5% nanokeratin (G4) were characterized by scanning electron microscope, Fourier-transform infrared spectroscopy, differential scanning calorimetry, swelling degree and quali-quantitative histological analysis. Ectopic implantation in subcutaneous tissue in mice at one, three and nine weeks allowed to assess the inflammation (neutrophils, lymphocytes, macrophages, and giant cells) and repair (neovascularization, and connective tissue) to determine biocompatibility and the integrity of biomaterials to score their biodegradability. Histomorphometry on critical size defects in rat calvaria at one and three months evaluated the percentage of bone, connective tissue, and biomaterials in all groups. Results: The hydrogels presented porous microstructure, water absorption and physicochemical characteristics compatible with their polymeric and/or mineral composition. All materials exhibited biocompatibility, biodegradability, and low osteoconductivity. G2 showed greater density of new bone and biomaterial than the G1, G3 and G4. Conclusions: The collagen-apatite group formulation suggests potential for development as osteopromoting membrane.

Lupus erythematosus complex is an immune-mediated dermatological disease, mainly represented by the generalized and discoid forms. The last form described is milder, as it is limited to the appearance of lesions, usually on the face and in mucocutaneous regions. Its pathophysiology is considered multifactorial, however, continuous exposure to ultraviolet radiation seems to be very relevant to trigger and/or worsen clinical manifestations. DeÞ nitive diagnosis is obtained by histopathological analysis, and treatment is mainly based on immunosuppression and protection against ultraviolet radiation. The objective of this study was to report the case and clinical evolution of a mixed breed bitch, diagnosed with discoid lupus erythematosus. The bitch presented moderate desquamation, crusts and depigmentation restricted to the nasal plane. No other clinical or laboratory Þ ndings were evidenced in the screening tests. Upon conÞ rmation by histopathology, the initial therapy was started with oral prednisolone. The owners were also instructed to avoid exposure to sunlight, as well as to use topical protectors against ultraviolet radiation. The patient presented good response to therapy, showing remission of signs. Other sporadic recurrences were observed later, however, they were controlled only with topical corticosteroids, but always reinforcing the other precautions of environmental management. This work also addressed the risks, beneÞ ts and need to institute ongoing care to control discoid lupus erythematosus. Therapeutic success can vary among patients, as the intensity of the disease can be manifested in varying degrees. Therefore, in those individuals in which the condition is mild, it may be advantageous to opt for more conservative therapies in order to avoid side effects.(AU)


O complexo lúpus eritematoso é uma enfermidade dermatológica imunomediada, sendo principalmente representado pela forma generalizada e discoide. A última descrita é mais branda, pois se limita ao aparecimento de lesões geralmente em face e em regiões muco-cutâneas. Sua Þ - siopatogenia é considerada multifatorial, entretanto, a exposição contínua à radiação ultravioleta parece ser muito relevante para desencadear e/ou agravar as manifestações clínicas. O diagnóstico deÞ nitivo é obtido pela análise histopatológica, e o tratamento se baseia principalmente na imunossupressão e proteção contra a radiação ultravioleta. O objetivo deste trabalho foi relatar o caso e evolução clínica de uma cadela sem raça deÞ nida, diagnosticada com lúpus eritematoso discoide. A mesma apresentou moderada descamação, crostas e despigmentação restritas ao plano nasal. Nenhum outro achado clínico ou laboratorial foi evidenciado nos demais exames de triagem. Após conÞ rmado pela histopatologia, a terapia inicial foi instituída a partir da prednisolona por via oral. Os tutores também foram orientados a evitar exposição a luz solar, bem como, fazer a utilização de protetores tópicos contra a radiação ultravioleta. A paciente teve boa resposta à terapia, apresentando remissão dos sinais. Após este episódio, outras recidivas esporádicas foram observadas, entretanto, controladas apenas com corticoides tópicos, mas sempre reforçando os demais cuidados com o manejo ambiental. Este trabalho também abordou os riscos, benefícios e necessidade de instituir o cuidado contínuo para controle do lúpus eritematoso discoide. O sucesso terapêutico pode variar entre os pacientes, uma vez que a intensidade da doença pode ser manifestada em vários graus. Portanto, naqueles indivíduos em que o quadro é brando, pode ser vantajoso optar por terapias mais conservadoras, a Þ m de evitar seus efeitos colaterais.(AU)

The growing need for effective antimicrobial agents as novel therapies against multidrug-resistant pathogens has drawn scientist attention toward nanotechnology. Silver nanoparticles are considered capable of killing multidrug-resistant isolates due to their oligo-dynamic effect on microorganisms. In this research study NPs were synthesized using the gram-positive bacteria Lactobacillus bulgaricus and its activity against selected pathogenic strains. Lactobacillus bulgaricus pure cultures were isolated from raw milk and grown in "De Man, Rogasa, and Sharp" broth for synthesis of nanoparticles. Lactobacillus bulgaricus culture was centrifuged and Cell free supernatant of it was employed with aqueous silvery ions and evaluated their antibacterial activities against bacterial strains i.e. Staphylococcus aureus, Staphylococcus epidermidis and Salmonella typhi using agar well diffusion assay. Antibiotic profiling against selected pathogenic strains were also conducted using disc diffusion method. The synthesis and characterization of silver nanoparticles were monitored primarily by the conversion of the pale-yellow color of the mixture into a dark-brown color and via ultraviolet-visible absorption spectroscopy and Scanning electron microscopy respectively. The result showed that that AgNPs with size (30.65-100 nm) obtained from Lactobacillus bulgaricus were found to exhibit antibacterial activities against selected bacterial strains. Taken together, these findings suggest that Lactobacillus bulgaricus has great potential for the production of AgNPs with antibacterial activities and highly effective in comparison to tested antibiotics.

Background: Squamous cell carcinoma (SCC) is a common cutaneous neoplasm in horses, which mainly affects the external genitalia, oral cavity, and periocular region. The development of SCC metastases is rare in these animals, and the most common occurrence is a marked local infiltration. Exposure to ultraviolet rays and skin depigmentation are the main etiological factors of SCC. Definitive diagnosis of the neoplasm is performed through histological examination of lesions. The present report describes the clinical, anatomopathological, and immunohistochemical (IHC) aspects of a case of metastatic SCC, with the vulva as the primary site in a mare. Case: A 17-year-old mare, mixed breed, was referred to the Veterinary Hospital of the Universidade de Passo Fundo (UPF), with history of areas of depigmented skin (pinto coat), and clinical history of anorexia, frequent episodes of colic, and recumbency. During clinical evaluation, heart rate (68 bpm), rectal temperature (38.4ºC), and respiratory rate (48 mpm) were elevated, and the oral mucosa was pale. The mare also had an ulcerated tumor mass involving the vulva, which extended to the inguinal region and involved the mammary gland. A cytological aspirate of the vulvar tumor was performed, in which no neoplastic cells were found. Next, a biopsy of 2 distinct areas of the vulva was performed. The material was sent for anatomopathological examination, which showed markedly pleomorphic malignant squamous cells, with individual keratinization and high mitotic index, organized in trabeculae with rare keratin pearl-like formations. The exam allowed the diagnosis of SCC Grade II. Due to the poor prognosis and high cost of treatment, the owner consented to euthanasia and necropsy examination. During necropsy, the vulvar tumor mass was grayish, firm to hard, infiltrative, and had friable areas. Tumor foci suggestive of metastasis were also observed in inguinal, mesenteric, mediastinal and renal lymph nodes, adrenal glands, lung, pericardium, medullary canal, intercostal muscles, right 15th rib, and tissue surrounding the azygos vein. Samples from all organs were collected for anatomopathological examination. Diagnosis of metastatic vulvar SCC was confirmed through histological and IHC studies, which evaluated the expression of cytokeratins (AE1/ AE3), as well as the proliferative activity of neoplastic cells through the PCNA marker. Discussion: The diagnosis of metastatic SCC was obtained through the observed clinical, necroscopic, histological, and IHC characteristics. This neoplasm usually appears in depigmented regions exposed to ultraviolet light, and older animals are more likely to be affected. The mare in the present case had areas of depigmented skin (pinto coat). The animal was kept in a paddock outdoor and exposed to constant solar radiation. Although rare in horses, manifestation of anorexia, progressive weight loss, and frequent colic episodes and recumbency may be closely related to the multiple sites of metastasis in the present case. The main histological findings of the neoplasm were the dense proliferation of malignant squamous cells with individual keratinization, arranged in a trabecular pattern and with rare formations of keratin pearls, in line with previous studies. During IHC evaluation, the neoplastic cells showed expression of cytokeratins (AE1/AE3), as well as high proliferative activity evidenced by the PCNA marker. Given this background, the present report describes the clinical, anatomopathological, and IHC aspects of a case of metastatic SCC with a primary site in the vulva of a mare.

Pigmentary superficial keratitis (PSK) is a chronic corneal disorder with different causes, which may include immune-mediated diseases and reactions to ultraviolet rays. This study aimed to evaluate the use of automated central lamellar superficial keratectomy (ACLSK) in the treatment of chronic pigmentary superficial keratitis (CPSK) in dogs. We enrolled 24 animals with CPSK and loss of visual function even after clinical treatment with preserved post-corneal transparent media, and the potential for recovery of visual function after surgical treatment. The microkeratome was positioned on the eye surface, and the central corneal lamellae were obtained after translation. With ACSLC, 21 animals (87.5%) recovered their visual function. In 3 animals (12.5%), the formation of granuloma and posterior central leukoma in the cornea compromised the transparency of the visual axis. This study concludes that ACLSK is a viable surgical alternative for the restoration of visual function in dogs with severe PSK, thus improving their quality of life. The short execution time of this procedure reduces the costs and anesthetic risks.


A ceratite pigmentar superficial (CPS) é uma doença crônica da córnea, com diferentes causas, que podem incluir doenças imunomediadas e reação aos raios ultravioleta. Este estudo teve como objetivo avaliar a utilização da ceratectomia superficial lamelar central automatizada (CSLCA) no tratamento da ceratite superficial pigmentar crônica (CSPC) em cães. Vinte e quatro animais incluídos apresentaram CSPC e perda da função visual mesmo após o tratamento clínico, porém mantiveram os meios transparentes pós-córnea preservados e o potencial de recuperação da função visual após o tratamento cirúrgico. O microcerátomo foi posicionado na superfície do olho e, após o término de sua translação, foram obtidas as lamelas centrais da córnea. Com a CSLCA, 21 animais (87,5%) recuperaram a função visual. Em três animais (12,5%), a formação de granuloma e leucoma central posterior na córnea comprometeu a transparência do eixo visual. Este estudo conclui que a CSLCA é uma alternativa cirúrgica viável para o restabelecimento da função visual em cães com CPS grave, melhorando, assim, sua qualidade de vida. Seu curto tempo de execução proporciona redução de custos e de riscos anestésicos.

Background: Melanoma is a malignant neoplasm that arises from melanocytes and malanoblasts. It is also more frequently reported in dogs than in other species. They may arise from melanocytes in the skin, on the surfaces of the mucous membranes, and eyes. The aim of this study was to describe the epidemiological aspects, risk factors and clinicopathological findings of melanoma in dogs in the backlands, northeastern Brazil. Materials, Methods & Results: A retrospective study was carried out in all biopsy samples and necropsy examinations of dogs, from January 2003 to December 2021, at the Animal Pathology Laboratory of the Federal University of Campina Grande, Patos, Paraiba, northeastern Brazil. Epidemiological data, clinical signs, and gross lesions were reviewed from the diagnostic laboratory reports. Samples of the skin, lymph nodes, central nervous system and organs of the thoracic and abdominal cavities were fixed in 10% buffered formalin, processed routinely for histopathology, embedded in paraffin wax, cut into 4 µm sections, and stained with hematoxylin and eosin (HE). Histological sections were also submitted to immunohistochemistry with the primary antibody anti-Melan A. Of the 4717 records found, 1158 (24.5%) were diagnosed with neoplasms, of which 48 (4.14%) cases were of melanoma. Of this total, 28 (58.3%) dogs were elderly, 19 (39.6%) were adults, and 1 (2.1%) was young. Mixed breed animals were the most affected (42.6%), followed by the pinscher breed (19.1%). According to the anatomical region, the most affected site was the skin (38/53=71.7%), followed by the oral cavity (12/53=22.65%) and the eyes (3/53=5.7%). Grossly, the skin lesions were characterized by exophytic and usually blackened, sometimes irregular and firm, nodules. At cut, they had a smooth, compact and blackened surface. Lesions in the oral cavity were characterized by blackened, irregular and infiltrating nodules or masses. The ocular lesions were always unilateral and were characterized by an enlarged and diffusely blackened eyeball, with areas of ulceration and subversion of tissue architecture. In 5 animals there was more than one anatomical site affected, totaling 53 lesions. In 9 (17%) cases, metastases were identified, 8 in regional lymph nodes and 1 in the lung. Histopathology showed a densely non-encapsulated, poorly delimited, expansive and infiltrative neoplasm, composed of neoplastic cells arranged in islands or nests and supported by fibrovascular stroma, containing a variable amount of brownish pigment (melanin). Immunohistochemistry showed strong immunostaining of the neoplastic cells in brown by the anti-Melan A antibody. Discussion: The diagnosis of melanoma was established based on epidemiological, clinical, anatomopathological, and immunohistochemical findings. Gender is not a predisposing factor, and although there was no statistically significant relationship, males were more affected. Senescence is a conditioning risk factor. Elderly animals were more affected (P < 0.0001) than adult ones, with OR = 4.38; and young ones (P = 0.0051), with OR = 12.65. Some breeds, especially those with marked skin pigmentation, were more affected, however the most affected ones in this survey were pinscher and poodle. Cutaneous melanoma accounted for almost 72% of cases, contesting recent studies where oral cavity melanoma was more frequent. Therefore, it is believed that the climatic conditions of the backlands sub-region, in northeastern Brazil, associated with the individual characteristics of the dogs, are involved in the development of these neoplasms, since the climate is predominantly dry, with high temperatures throughout the year, with maximums that can reach 40ºC, favoring the exposure to high incidence of ultraviolet radiation.

Resumo

Background: Acylpolyamines are one of the main non-peptide compounds present in spider venom and represent a promising alternative in the search for new molecules with antimicrobial action. Methods: The venom of Acanthoscurria natalensis spider was fractionated by reverse-phase liquid chromatography (RP-HPLC) and the antimicrobial activity of the fractions was tested using a liquid growth inhibition assay. The main antimicrobial fraction containing acylpolyamines (ApAn) was submitted to two additional chromatographic steps and analyzed by MALDI-TOF. Fractions of interest were accumulated for ultraviolet (UV) spectroscopy and ESI-MS/MS analysis and for minimum inhibitory concentration (MIC) and hemolytic activity determination. Results: Five acylpolyamines were isolated from the venom with molecular masses between 614 Da and 756 Da, being named ApAn728, ApAn614a, ApAn614b, ApAn742 and ApAn756. The analysis of UV absorption profile of each ApAn and the fragmentation pattern obtained by ESI-MS/MS suggested the presence of a tyrosyl unit as chromophore and a terminal polyamine chain consistent with structural units PA43 or PA53. ApAn presented MIC between 128 µM and 256 µM against Escherichia coli and Staphylococcus aureus, without causing hemolysis against mouse erythrocytes. Conclusion: The antimicrobial and non-hemolytic properties of the analyzed ApAn may be relevant for their application as possible therapeutic agents and the identification of an unconventional chromophore for spider acylpolyamines suggests an even greater chemical diversity.(AU)

Resumo

O trabalho tem o objetivo de relatar um caso de Dermatose Actínica em cadela adulta, 8 anos apresentando pele eritematosa. Na avaliação clínica a cadela apresentou os parâmetros normais da espécie, porém devido às lesões observadas foram solicitados exames citológico e parasitológico de pele, que demonstraram a presença de bactérias (cocos), neutrófilos degenerados e fungos dermatófitos além de demodicidose. O tratamento consistiu do uso de inúmeras drogas como itraconazol, amoxicilina com clavulonato de potássio, predinisolona, Sarolaner e imunomoduladores, além de tratamento tópico com cetoconazol shampoo e creme. Geralmente, as piodermites profundas que atingem o folículo de cães e formam feridas ulceradas, causam uma inflamação crônica na pele, por conta de raios-ultravioletas do sol, ou predisposição familiar ou hipersensibilidade microbiana, ou por outras doenças e, normalmente, são reincidentes e não permitem a cura. Entretanto, nesse caso específico, o tratamento permitiu a melhora do animal (~80%).(AU)


The aim of this paper is to report a case of Actinic Dermatosis in an 8-year-old female adult female with erythematous skin. In the clinical evaluation, the animal presented normal parameters for the species, but, due to the lesions observed, cytological and parasitological skin tests were requested, which showed the presence of bacteria (cocci), degenerated neutrophils, and dermatophyte fungi, and demodicidosis. Treatment consisted of the use of numerous drugs such as itraconazole, amoxicillin with potassium clavulonate, predinisolone, Sarolaner, and immunomodulators; in addition to topical treatment with ketoconazole shampoo and cream. Generally, deep pyoderma that reach the follicle of dogs and form ulcerated wounds, cause a chronic inflammation of the skin, due to the sun's ultraviolet rays, or familial predisposition or microbial hypersensitivity, or due to other diseases and, usually, they are recurrent and do not allow healing. However, in this specific case, the treatment allowed the animal to improve (~80%).(AU)
