Resumo

Degenerative diseases, such as osteoporosis, could be treated by stem cells. The aim of this study was to identify the gene expression of bone marrow mesenchymal stem cells (BM-MSC) derived from Sprague Dawley rats and to assess the effect of Cissus quadrangularis Salisb. extract on their maturation into bone cells. The BM-MSC were divided into three groups: (a) BM-MSCs + osteoblast cell growth basal medium as the positive control; (b) BM-MSCs + Dulbecco's modified eagle's medium (DMEM) + 0.3 mg/mL methanol extract of C. quadrangularis as methanol group; and (c) BM-MSC + DMEM + 0.3 mg/mL ethyl acetate extract of C. quadrangularis as ethyl acetate group. A relative quantification approach using was used to analyze the expression of the alp (alkaline phosphatase) gene, with the beta-actin gene was used to normalize the expression of the alp gene. The intra-assay variation was calculated to validate the RT-qPCR data. Our study found that the intra-assay variation value was acceptable, with most of the coefficients of variability (CV) value <5. Ethyl acetate solvent outperformed methanol solvent in extracting the active compound C. quadrangularis. In the ethyl acetate extract group, the expression of the alp gene increased three times compared to the positive control. In methanol extract group, the expression of alp gene was lower six times compared to positive control. This study suggests that C. quadrangularis extracts using ethyl acetate could induce the maturation of BM-MSCs. However, further studies are warrant to confirm this effect using different indicators.


Doenças degenerativas, como a osteoporose, podem ser tratadas por células-tronco. O objetivo deste estudo foi identificar a expressão gênica de células-tronco mesenquimais da medula óssea (BM-MSCs) derivadas de ratos Sprague Dawley, bem como sua proliferação e diferenciação em células ósseas afetadas pelo extrato de Cissus quadrangularis Salisb. As BM-MSCs foram divididas em três grupos: (1) BM-MSCs + meio basal de crescimento de osteoblastos como controle positivo; (2) BM-MSCs + meio de Eagle modificado por Dulbecco (DMEM) + extrato de metanol; e (3) BM-MSCs + DMEM + extrato de acetato de etila de C. quadrangularis. Uma abordagem de quantificação relativa foi usada para analisar a expressão do gene alp (fosfatase alcalina), com o gene da beta-actina sendo usado para normalizar a expressão do gene alp em cada tratamento. A variação intraensaio foi calculada para validar os dados de RT-qPCR. Os resultados mostraram que o valor da variação intraensaio é aceitável, com a maioria dos valores dos coeficientes de variabilidade (CV) < 5. Além disso, no tratamento T2, a expressão do gene alp aumentou três vezes em relação ao controle positivo. No tratamento T1, o gene alp apresentou nível de expressão seis vezes menor em comparação com o controle positivo. Além disso, o solvente acetato de etila superou o solvente metanol na extração do composto ativo C. quadrangularis Salisb. Este estudo demonstra que extratos de C. quadrangularis Salisb., particularmente com acetato de etila como solvente, podem induzir a proliferação e diferenciação de BM-MSCs. Com base na expressão do gene alp, C. quadrangularis tem o potencial de corrigir a perda óssea.

Background: Propolis exhibits huge potential in the pharmaceutical industry. In the present study, its effects were investigated on dendritic cells (DCs) stimulated with a tumor antigen (MAGE-1) and retinoic acid (RA) and on T lymphocytes to observe a possible differential activation of T lymphocytes, driving preferentially to Th1 or Treg cells. Methods: Cell viability, lymphocyte proliferation, gene expression (T-bet and FoxP3), and cytokine production by DCs (TNF-α, IL-10, IL-6 and IL-1ß) and lymphocytes (IFN-γ and TGF-ß) were analyzed. Results: MAGE-1 and RA alone or in combination with propolis inhibited TNF-α production and induced a higher lymphoproliferation compared to control, while MAGE1 + propolis induced IL-6 production. Propolis in combination with RA induced FoxP3 expression. MAGE-1 induced IFN-γ production while propolis inhibited it, returning to basal levels. RA inhibited TGF-ß production, what was counteracted by propolis. Conclusion: Propolis affected immunological parameters inhibiting pro-inflammatory cytokines and favoring the regulatory profile, opening perspectives for the control of inflammatory conditions.(AU)

Apocrine neoplasms are rare in goats. This report describes an apocrine adenocarcinoma in a 5-year-old female Boer goat with an ulcerated, exophytic, multilobulated mass on the left dorsolateral cervical area. Histologically, the dermis was effaced and expanded by a poorly demarcated epithelial neoplasm with desmoplasia. Neoplastic cells were arranged in acini with papillary projections into the lumen and had cuboidal or columnar, eosinophilic, occasionally vacuolated cytoplasm. Nuclei were round and had finely stippled chromatin with one nucleolus. Neoplastic cells contained periodic acid-Schiff (PAS)­positive and diastase­resistant, alcian blue­positive cytoplasmic granules. There were 6 mitoses in 2.37 mm2 (equivalent to 10 FN22/40X fields). Histologic and histochemical features in the current case were consistent with a cutaneous apocrine gland adenocarcinoma.(AU)

Resumo

Biliary neoplasms are uncommon in cats and affect older animals. A 12-year-old female crossbreed cat showed prostration, lethargy, apathy, and severe jaundice. Ultrasonography showed distention of the gallbladder associated with severe obstruction of the bile ducts with thickening of the biliary wall, forming amorphous masses of irregular contour and heterogeneous appearance directed to the lumen measuring up to 2 cm. Necropsy showed a gallbladder with a yellowish and soft nodule measuring 3 × 3 cm, compressing the extrahepatic bile duct, occluding the passage of bile. There were also firm, yellowish multifocal to coalescing nodules in the liver, ranging from 0.5 to 1 cm, affecting 10% of the organ, in addition to lungs with firm, yellowish multifocal nodules ranging from 0.2 to 0.5 cm, affecting 20% of the organ. Histologically, gallbladder and bile ducts had malignant epithelial neoplastic proliferation, which was organized into multiple papillary and ductal projections, separated by moderate fibrovascular stroma compatible with gallbladder adenocarcinoma. The liver and lungs also contained neoplastic structures with a ductal appearance and papilliform projections identical to those observed in the gallbladder. The immunohistochemical examination (IHC) showed intense positive staining of epithelial neoplastic cells for pan-cytokeratin (AE1/AE3) and no staining for vimentin (Clone V9). The diagnosis of gallbladder adenocarcinoma with metastasis in the liver and lungs was established based on the clinical, macroscopic, histopathological, and immunohistochemical findings.(AU)

Litter size is one of the crucial factors in livestock production and is of high economic value, which is affected by ovulation rate, hormones, and growth factors.Growth factors play a multifaceted role in reproductive physiology. This review aims to investigate the association of bone morphogenetic protein 15 (BMP15) and growth differentiation factor 9 (GDF9)with litter size in livestock.The transforming growth factor ß(TGF-ß) superfamily includes more than 34 members; GDF9 and BMP15 are among the most significantfactors for regulating fertility and litter size in most livestock species. Ovarian follicles release BMP15 and GDF9 that are involved in the maturation of primary follicles into the basal form, proliferation of granulosa and theca cells, steroidogenesis, ovulation, and formation of the corpus luteum. Besides, these factors are highly expressed in oocytes and are necessary for female fertility and multiple ovulation in several livestock species. Animals with two inactive copies of these factors are sterile, while those with one inactive copy are fertile. Thus, the present reviewprovides valuable information on the association of BMP15 and GDF9with litter size in livestock that can be used as biological markers of multiple ovulation or for improving fertility in livestock.(AU)

Resumo

The objective of this study was to describe the histology and histochemistry of the integument covering the back and stinger of the freshwater stingray Potamotrygon rex, endemic to the Middle Upper Tocantins River. The species has a dark back and yellowish circular spots that extend to the tail, which has one to two stings located in the median portion of the tail. Through histological study it was observed that the epithelia of the back and stinger are composed of non-keratinized stratified pavement epithelial tissue, and are organized in three layers: basal, intermediate and superficial. The differences between the tissues are related to the cell types that compose them. The back is organized with epithelial cells, mucus cells, granulocyte cells and chromatophores. The mucus cells are distributed in different layers along the animal's back, influencing the thickness of the tissue. The tissue that covers the stinger is composed of epithelial cells, chromatophores and specialized cells in protein synthesis. In the histochemistry, the stinger epithelial cells were stained with Bromophenol Blue, especially those of the intermediate layer, which were called specialized cells. In the back the epithelial cells were stained with Bromophenol Blue, Alcian Blue and PAS, and the mucous cells with PAS. In both tissues the presence of protein reserves was detected, areas so called because they are stained strongly with Bromophenol Blue. The results show that the stinger presents activity directed to the production of proteins, and that the back is organized to produce different components, which constitute the cuticle that covers the animal's body.


O objetivo deste estudo foi descrever a histologia e histoquímica do tegumento que reveste o dorso e o ferrão da arraia de água doce Potamotrygon rex, endêmica do Médio Alto Tocantins. A espécie possui o dorso escuro e manchas circulares amareladas que se estendem até a cauda, que possui de um a dois ferrões localizados na porção mediana da cauda. Através do estudo histológico observou-se que os epitélios do dorso e do ferrão são compostos por tecido epitelial pavimentoso estratificado não queratinizado, e estão organizados em três camadas: basal, intermediária e superficial. As diferenças entre os tecidos estão relacionadas aos tipos de células que os compõem. O dorso é organizado com células epiteliais, células mucosas, células de granulócitos e cromatóforos. As células mucosas estão distribuídas em diferentes camadas ao longo do dorso do animal, influenciando na espessura do tecido. O tecido que reveste o ferrão é composto por células epiteliais, cromatóforos e células proteicas. Na histoquímica, as células epiteliais do ferrão foram coradas com Azul de Bromofenol, principalmente as da camada intermediária, que foram denominadas células proteicas. No dorso as células epiteliais foram coradas com Azul de Bromofenol, Azul Alcian e PAS, e as células mucosas com PAS. Em ambos os tecidos também foi detectada a presença de reservatórios de proteínas, áreas assim denominadas por estarem fortemente coradas com Azul de Bromofenol. Os resultados mostram que o ferrão apresenta atividade direcionada à produção de proteínas, e que o dorso se organiza para produzir diferentes componentes que constituem a cutícula que reveste o corpo do animal.

The treatments consisted of 0, 3, 6, 9, and 12% of inclusion of soy molasses in the dry matter for a basal diet of corn silage, which met the nutritional needs of the sheep in maintenance, distributed in a 5 × Latin square design. 5. The energy metabolites were evaluated: cholesterol, triglycerides, very-low-density lipoproteins (VLDL), and fructosamine. The hematological parameters were red blood cells, hemoglobin, hematocrits, and platelets, in addition to leukocytes, rods, neutrophils, eosinophils, monocytes, and lymphocytes. Supplementation with soy molasses did not significantly alter (P> 0.05) the serum concentrations of cholesterol, triglycerides, VLDL, and fructosamine. The basal nuclei are well-defined bodies of neurons with specific functions, located inside the white medullary center of the brain, directly involved with the motor system, participating greatly in the planning and control processes of movements. Studies on these nuclei in non-human primates are small and in the Alouatta belzebul species, nonexistent. The aim of the present study was to describe the morphology of the nuclei at the base of the brain of Alouatta belzebul. Ten male and female Alouatta belzebul brains were used, where after removal and coronal cut of the brain, the Mayland technique was performed to show the basal nuclei. There was the presence of the caudate nucleus, lentiform nucleus (this formed by the putamen, medial globus pallidus and lateral globus pallidus), claustrum and substantia nigra, which, functionally, are related to motor control. The substantia nigra is part of the midbrain and is also related to learning resulting from the effects of dopamine, responsible for activating the reward and addiction system in the telbrain and is also related to the red nucleus, which is also a midbrain nucleus. In Alouatta belzebul the red nucleus is present. It was found in the literature that degeneration of substantia nigra cells can cause Parkinson's disease in Macaca fasciculares, and because Alouatta belzebul has the same anatomical structures in the basal nuclei of the base of Macaca fasciculares, it is suggested that studies of functional evaluation of these structures should be carried out to verify whether Alouatta belzebul can be used as an experimental model for Parkinson's disease.


Os núcleos da base são corpos de neurônios, bem delimitados e com funções específicas, localizados no interior do centro medular branco do cérebro, envolvidos diretamente com o sistema motor, através de uma função moduladora dos movimentos, participando sobremaneira nos processos de planejamento e controle dos movimentos. Os estudos sobre estes núcleos em primatas são reduzidos e na espécie Alouatta belzebul, inexistente. O objetivo do presente estudo foi descrever a morfologia dos núcleos da base do encéfalo de Alouatta belzebul. Para tanto, foram utilizados dez encéfalos de Alouatta belzebul, machos e fêmeas, onde após a remoção e corte coronal do cérebro, realizou-se à técnica de Mayland para evidenciar os núcleos da base. Verificou-se a presença do núcleo caudado, núcleo lentiforme (este formado pelo putâmen, globo pálido medial e globo pálido lateral), claustro e substância negra, que, funcionalmente, estão relacionados com o controle motor. A substância negra faz parte do mesencéfalo e está ainda relacionada com a aprendizagem decorrentes dos efeitos da dopamina, responsável por ativar o sistema de recompensa e vício no telencéfalo e tem ainda, relação com o núcleo rubro que também é um núcleo do mesencéfalo. Em Alouatta belzebul o núcleo rubro está presente. Verificou-se na literatura que a degeneração de células da substância negra pode ocasionar a doença de Parkinson em Macaca fasciculares, e pelo fato do Alouatta belzebul apresentar as mesmas estruturas anatômicas dos núcleos da base do mesencéfalo de Macaca fasciculares, poderia ser utilizado como modelo experimental em estudos clínicos para a doença de Parkinson.

Background: Lacrimal gland adenocarcinoma is an extremely rare malignant neoplasm, with few descriptions in the literature. Therefore, its etiology and treatment are not well understood. The present study aims to report the case of an equine histologically diagnosed with lacrimal gland adenocarcinoma in the lower eyelid region and third eyelid treated by surgical excision and intralesional chemotherapy. Case: A 17-year-old male mixed-breed equine weighing 300 kg was treated in the large animal clinic and surgery sector of the Federal University of Santa Catarina in the city of Curitibanos, Santa Catarina, Brazil. The equine revealed an alert and docile temperament without considerable physiological changes in the physical examination. In the evaluation of the right eye, a tumor mass was observed in the region of the lacrimal gland adhered to the third eyelid and lower eyelid, with a light red ulcerated appearance approximately 7 cm in diameter. Therefore, the surgical excision of the adhered tumor mass was performed, with the subsequent intralesional application of 2 mL of Vincristine Sulfate. In the histopathological evaluation following the surgical excision of the tumor mass, yellowish-white fragments of irregular nodules were observed measuring from 3.5×2.0×1.7 cm to 2.0×0.5×0.3 cm, in addition to microscopy focus clusters of weakly basophilic neoplastic glandular cells, some with a randomly arranged lacy aspect, forming disorganized acinar structures and others showed marking islands of the cells organized in a palisade shape by vascular delicate stroma. The neoplastic cells presented anisocytosis, anisokaryosis, prominent nucleoli sometimes binucleated and with basophilic intracytoplasmic secretory material, delimited by scarce connective tissue. In some areas, there were small foci of infiltrated lymphocytes and plasmocytes and areas of necrosis. The mass was surrounded by connective tissue, where red blood vessels were found outside the vessels (hemorrhage), and mitoses were observed 2 per field at high magnification (40x). After the surgical procedure, it was not possible to follow up and reassess of the patient, so there is no clarification on the possibility of tumor recurrence. Discussion: The case reported is uncommon, being only the second report of adenocarcinoma in the equine lacrimal gland. Therefore, there are few descriptions in the literature about its defined etiology and the best treatment method, although, surgical excision is the method of choice because it is a tumor of high recurrence and invasiveness. In this case, we opted for the exeresis of the tumor mass and later application of chemotherapy, a treatment that was relatively effective, to avoid recurrence and the failure of the technique. In these cases, monitoring the animal after the procedure is recommended, evaluating whether there was tumor recurrence, although this was not possible in the present possible. The third eyelid, lacrimal, and zygomatic salivary glands may be sources of intraorbital neoplasms. These neoplastic processes may have similar histological and behavioral characteristics, and their differentiation is, therefore, problematic. These structures may be differentiated based on the anatomical location, however, clinical appearance and symptoms are identical in most cases. Adenocarcinoma in the equine lacrimal gland is a neoplasm considered rare, so it is important to emphasize an accurate diagnosis through histopathological analyses to differentiate it from other frequent orbital neoplasms in horses, allowing more information about this tumor and establishing different treatment methods.

The aim of this article is to describe a case of adrenocortical neoplasm with manifestation of hyperadrenocorticism. Adrenocortical tumors originate from different types of cells and present varied clinical manifestations, which can be functional or non-functional. Adenocarcinomas are autonomous and functional in most cases, leading to excessive secretion of glucocorticoids, regardless of pituitary control. They corroborate the occurrence of hyperadrenocorticism (HAC) due to interference in the synthesis of cortisol. Clinical signs can be observed, as polyuria, compensatory polydipsia, polyphagia, blood pressure alterations, cardiac, renal and endocrine dysfunctions, among others. Diagnosis can be made in various ways, such as urinary cortisol measurements, adrenocorticotropic hormone stimulation, low-dose dexamethasone suppression tests, and imaging tests and histopathology. This article reports the case of a 13-year-old mixed-breed female dog with signs of polyuria and polydpsia. After discarding the initial diagnoses (diabetes mellitus and/or renal alterations), HAC was suspected, with further tests being performed. The test results showed an increase in the adrenal region, and adrenalectomy and hormone replacement with trilostane were recommended. The diagnosis of HAC was confirmed by histopathology as HAC secondary to adrenal gland adenocarcinoma. The patient also developed diabetes mellitus during postoperative treatment with prednisone, which made it necessary to discontinue the medication.


O objetivo deste artigo é descrever um caso de neoplasia adrenocortical com manifestação de hiperadrenocorticismo. Tumores adrenocorticais são originados de diversos tipos de células e apresentam manifestação clínica variada, podendo ser funcionais ou não funcionais. Os adenocarcinomas são autônomos e funciona na maioria dos casos, levando a secreção excessiva de glicocorticóides, independente do controle da hipófise. Eles corroboram com a ocorrência de hiperadrenocorticismo (HAC) por interferência na síntese de cortisol. Os sinais clínicos observados podem ser poliúria, polidipsia compensatória, polifagia, alterações pressóricas, disfunções cardíacas, renais e endócrinas, entre outros. Estas manifestações clínicas podem se apresentar de forma isolada ou associada. O diagnóstico pode ser obtido de diversas formas, como dosagens de cortisol urinário, estimulação de hormônio adrenocorticotrópico, testes de supressão com baixa dose de dexametasona e por testes de imagem. No entanto, o diagnóstico definitivo baseia-se no uso da histopatologia. Este artigo relata o caso de uma fêmea sem raça definida, de 13 anos de idade, com sinais de poliúria e polidpsia. Após o descarte dos diagnósticos iniciais (diabetes mellitus e/ou alterações renais). suspeitou-se de HAC, com a realização de novos exames. Os resultados dos exames evidenciaram aumento da região adrenal, sendo recomendada a adrenalectomia e reposição hormonal com trilostano. O diagnóstico de HAC foi confirmado pela histopatologia como sendo HAC secundário a adenocarcinoma de glândula adrenal. A paciente desenvolveu ainda um quadro de diabetes mellitus durante o tratamento com prednisona no pós-operatório, sendo necessário interromper a medicação.


El objetivo de este artículo es describir un caso de neoplasia adrenocortical con manifestación de hiperadrenocorticismo. Los tumores adrenocorticales se originan a partir de diferentes tipos de células y presentan una variada manifestación clínica, que puede ser funcionante o no funcionante. Los adenocarcinomas son autónomos y funcionales en la mayoría de los casos, lo que lleva a una secreción excesiva de glucocorticoides, independientemente del control pituitario. Corroboran la aparición de hiperadrenocorticismo (HAC) al interferir con la síntesis de cortisol. Los signos clínicos observados pueden ser poliuria, polidipsia compensatoria, polifagia, alteraciones de la presión arterial, disfunciones cardíacas, renales y endocrinas, entre otras. Estas manifestaciones clínicas pueden presentarse de forma aislada o en asociación. El diagnóstico se puede obtener de varias maneras, como mediciones de cortisol urinario, estimulación con hormona adrenocorticotrópica, pruebas de supresión con dosis bajas de dexametasona y pruebas de imagen. Sin embargo, el diagnóstico definitivo se basa en el uso de la histopatología. Este artículo reporta el caso de una perra mestiza de 13 años con signos de poliuria y polidipsia. Tras descartar los diagnósticos iniciales (diabetes mellitus y/o alteraciones renales), se sospechó HAC, realizándose nuevas pruebas. Los resultados de los exámenes mostraron un aumento en la región suprarrenal, por lo que se recomendó adrenalectomía y reemplazo hormonal con trilostano. El diagnóstico de CAH se confirmó por histopatología como CAH secundaria a adenocarcinoma de la glándula suprarrenal. La paciente también desarrolló diabetes mellitus durante el tratamiento con prednisona en el postoperatorio, siendo necesario suspender la medicación.

Soy molasses is a by-product of the soy industry, with low commercial cost and rich in carbohydrates and proteins. Despite the great variety of by-products and being potentially beneÞ cial, it is not fully understood how the interaction between the nutrients in this product and the physiological changes in sheep occurs. Thus, the objective was to evaluate the effects of supplementation levels with soy molasses on the energy metabolites and hematological parameters of sheep. The treatments consisted of 0, 3, 6, 9, and 12% of inclusion of soy molasses in dry matter basis for a basal diet of corn silage, which met the nutritional needs of the sheep in maintenance, distributed in a 5 × 5 Latin square design. The energy metabolites evaluated were: cholesterol, triglycerides, very-low-density lipoproteins (VLDL), and fructosamine. The hematological parameters were: red blood cells, hemoglobin, hematocrits, and platelets, in addition to leukocytes, rods, neutrophils, eosinophils, monocytes, and lymphocytes. Supplementation with soy molasses did not signiÞ cantly alter (P> 0.05) the serum concentrations of cholesterol, triglycerides, VLDL, and fructosamine. There was no signiÞ cant difference in the blood cell, hemoglobin, hematocrit, and platelet concentrations in sheep receiving soy molasses supplementation (P> 0.05), which may indicate that the level of inclusion was sufÞ cient to meet the nutritional needs of the animals, without damage. Leukocyte, rod, neutrophil, eosinophil, monocyte, and lymphocyte concentrations did not change (P> 0.05) with the inclusion of soy molasses in the sheep's diet, indicating the absence of intoxication, diseases, and infections. It can be concluded that supplementation with soy molasses does not alter the energy metabolites and hematological parameters of sheep in maintenance.(AU)


O melaço de soja é um subproduto da indústria da soja, com baixo custo comercial e rico em carboidratos e proteínas. Apesar da grande variedade de subprodutos e de serem potencialmente benéÞ cos, ainda não está totalmente elucidado como ocorre a interação entre os nutrientes deste produto e as mudanças Þ siológicas em ovinos. Dessa forma, objetivou-se avaliar os efeitos de níveis de suplementação com melaço de soja sob os metabólitos energéticos e parâmetros hematológicos de ovinos. Os tratamentos consistiram em 0, 3, 6, 9 e 12% de inclusão de melaço de soja na base da matéria seca para uma dieta basal de silagem de milho, que atendia as necessidades nutricionais das ovelhas em mantença, distribuídas em delineamento em quadrado latino 5 × 5. Foram avaliados os metabolitos energéticos: colesterol, triglicérides, lipoproteínas de muito baixa densidade (VLDL) e frutosamina. Já os parâmetros hematológicos foram: hemácias, hemoglobinas, hematócritos e plaquetas, além de leucócitos, bastonetes, neutróÞ los, eosinóÞ los, monócitos e linfócitos. A suplementação com melaço de soja não alterou signiÞ cativamente (P>0,05) as concentrações séricas de colesterol, triglicérides, VLDL e frutosamina. Não houve diferença signiÞ cativa nas concentrações de hemácias, hemoglobinas, hematócritos e plaquetas das ovelhas com suplementação de melaço de soja (P>0,05), podendo ser indício que o nível de inclusão foi suÞ ciente para atender as necessidades nutricionais dos animais, sem causar balanço energético positivo. As concentrações de leucócitos, bastonetes, neutróÞ los, eosinóÞ los, monócitos e linfócitos não alteraram (P>0,05) com a inclusão de melaço de soja na dieta das ovelhas, não indicando a presença de intoxicação, doenças e infecções. Conclui-se que a suplementação com melaço de soja não altera os metabólitos energéticos e parâmetros hematológicos de ovinos em mantença.(AU)

O procedimento de imunocastração vem sendo utilizado como técnica alternativa amplamente favorável ao bem-estar animal, por ser indolor, pouco invasiva e com eficácia semelhante à da castração cirúrgica, que causa maior estresse aos animais, principalmente quando realizada de forma inadequada. A imunocastração estimula a produção de anticorpos contra o Hormônio Liberador de Gonadotrofina (GnRH), bloqueando temporariamente a produção da testosterona pelas gônadas masculinas. Face a escassez de informações sobre este procedimento em touros bubalinos, o estudo teve como objetivo analisar os possíveis efeitos da imunocastração no parênquima testicular de búfalos. Vinte touros bubalinos, com idade entre 2 a 3 anos, oriundos da ilha do Marajó ­ PA, foram avaliados e divididos aleatoriamente em dois grupos composto por dez animais no grupo controle (GC) e dez do grupo imunocastrado (GIM). O produto utilizado foi a vacina anti-GnRH Bopriva ® (Zoetis, SP, Brasil). O GIM recebeu duas doses de 1,0ml contendo 400µg da vacina Bopriva ® com intervalo de 8 semanas entre as aplicações e o grupo controle recebeu 1,0 ml de solução fisiológica. Após 14 dias da última dose, os animais foram abatidos e coletado os 20 pares de testículos para analises de parâmetros macroscópicos como: comprimento, largura, circunferência e peso. Além disso, foi realizada a retirada de fragmentos do parênquima testicular para confecção de lâminas histológicas. Para comparação dos dados foi aplicado o teste t de Student (Nonparametric Test) considerando a significância com p<0,05 e intervalo de confiança de 95%. Foi observado diferença significativa entre os dois grupos para grupos para peso (direto p<0001; esquerdo p≤ 0,0001), circunferência (direita p< <0,0001), 1; esquerda p<0,0001), ), largura (somente lado direito p<0,0372) e comprimento (direita p≤ 0,0013; esquerda p<0,0437). Na avaliação microscópica, os animais do GC não apresenteram alterações. Nos animais do GIM houve degeneração em todas as amostras, sendo visualizado descamação, tortuosidade e espessamento da membrana basal do túbulo seminífero, assim como vacuolização e atrofia das células de Sertoli. Foi observado também uma redução do número de células de Leydig, fibrose intertubular pronunciada, núcleos picnóticos, azoospermia e células multinucleadas no interior dos túbulos. Conclui-se a vacina anti-GnRH mostrou-se eficaz em provocar lesões no parênquima testicular, comprometendo significativamente a espermatogênese a partir da possível supressão de testosterona.(AU)


The immunocastration procedure has been used as an alternative technique widely favorable to animal welfare, as it is painless, minimally invasive and with similar efficacy to surgical castration, which causes greater stress to animals, especially when performed improperly. Immunocastration stimulates the production of antibodies against Gonadotropin Releasing Hormone (GnRH), temporarily blocking the production of testosterone by the male gonads. Due to the scarcity of information about this procedure in buffalo bulls, the study aimed to analyze the possible effects of immunocastration on the testicular parenchyma of buffaloes. Twenty buffalo bulls, aged between 2 and 3 years, from the island of Marajó - PA, were evaluated and randomly divided into two groups consisting of ten animals in the control group (CG) and ten from the immunocastrated group (GIM). The product used was the anti-GnRH vaccine Bopriva ® (Zoetis, SP, Brazil). The GIM received two doses of 1.0ml containing 400µg of Bopriva ® vaccine with an interval of 8 weeks between applications and the control group received 1.0 ml of saline solution. After 14 days of the last dose, the animals were slaughtered and the 20 pairs of testes were collected for analysis of macroscopic parameters such as: length, width, circumference and weight. In addition, fragments of the testicular parenchyma were removed for the preparation of histological slides. To compare the data, Student's t test (Nonparametric Test) was applied, considering the significance with p< 0.0372) and length (right p≤ 0.0013; left p<0.0437). In the microscopic evaluation, the animals of the CG showed no alterations. In the GIM animals there was degeneration in all samples, with desquamation, tortuosity and thickening of the seminiferous tubule basement membrane, as well as vacuolization and atrophy of Sertoli cells. A reduction in the number of Leydig cells, pronounced intertubular fibrosis, pyknotic nuclei, azoospermia and multinucleated cells within the tubules were also observed. In conclusion, the anti-GnRH vaccine proved to be effective in causing lesions in the testicular parenchyma, significantly compromising spermatogenesis from the possible suppression of testosterone.(AU)

A pele é um órgão complexo constituído por diferentes tipos celulares e possui exposição direta ao meio ambiente, representando uma barreira física do organismo. Devido a tais fatores pode ocorrer o aparecimento de diversos tipos tumorais. Além disso, a pele possui alta capacidade de renovação celular, o que aumenta as chances de mutações em comparação com outros tecidos, o que a torna um lugar favorável para o desenvolvimento de neoplasias. É comum ocorrer nos animais domésticos o desenvolvimento de neoplasias cutâneas, sendo que sua prevalência depende de fatores como genética, idade, raça, relação hormonal, nutrição, entre outros. O tricoblastoma é uma neoplasia benigna cutânea comum em cães e gatos adultos, animais com idade entre seis e nove anos são os mais acometidos, e não existe predisposição em machos e fêmeas. É derivado do folículo piloso primitivo e possui componentes epiteliais e mesenquimais. O tricoblastoma não é considerado agressivo, porém, existem relatos na literatura com características de neoplasia maligna. O tratamento de eleição do tricoblastoma é a exérese cirúrgica total do mesmo, com margens de segurança entre 1 e 2cm. O presente estudo tem como objetivo relatar o caso de um animal atendido no Centro Médico Veterinário do Centro Universitário do Sul de Minas, o qual foi diagnosticado com tricoblastoma.


The skin is a complex organ composed of different cell types and has direct exposure to the environment, representing an organism's physical barrier. Due to such factors, the appearance of several tumor types may occur. In addition, the skin has a high capacity to perform cell renewal, which increases the chances of mutations compared to other tissues, turning it a favorable place for the development of neoplasms. The development of skin neoplasms in domestic animals is a common fact, and their prevalence depends on factors such as genetics, age, race, hormonal relationship, nutrition, among others. Trichoblastoma is a benign cutaneous neoplasm common in adult dogs and cats, animals aged between six and nine years are the most affected, and there is no predisposition in males and females. It is derived from the primitive hair follicle and has epithelial and mesenchymal components. Trichoblastoma is not considered aggressive; however, there are reports in the literature with characteristics of malignant neoplasm. The treatment of choice for trichoblastoma is the total surgical excision, with safety margins between 1 and 2cm. The present study aims to report the case of an animal treated at the Veterinary Medical Center of the University Center of Southern Minas Gerais, which was diagnosed with trichoblastoma.

Abstract This study investigated the morphology and immunoexpression of aquaporins (AQPs) 1 and 9 in the rete testis, efferent ducts, epididymis, and vas deferens in the Azara's agouti (Dasyprocta azarae). For this purpose, ten adult sexually mature animals were used in histologic and immunohistochemical analyses. The Azara's agouti rete testis was labyrinthine and lined with simple cubic epithelium. Ciliated and non-ciliated cells were observed in the epithelium of the efferent ducts. The epididymal cellular population was composed of principal, basal, apical, clear, narrow, and halo cells. The epithelium lining of vas deferens was composed of the principal and basal cells. AQPs 1 and 9 were not expressed in the rete testis. Positive reaction to AQP1 was observed at the luminal border of non-ciliated cells of the efferent ducts, and in the peritubular stroma and blood vessels in the epididymis, and vas deferens. AQP9 was immunolocalized in the epithelial cells in the efferent ducts, epididymis and vas deferens. The morphology of Azara's agouti testis excurrent ducts is similar to that reported for other rodents such as Cuniculus paca. The immunolocalization results of the AQPs suggest that the expression of AQPs is species-specific due to differences in localization and expression when compared to studies in other mammals species. The knowledge about the expression of AQPs in Azara's agouti testis excurrent ducts is essential to support future reproductive studies on this animal, since previous studies show that AQPs may be biomarkers of male fertility and infertility.

The chickens were fed a basal diet under no stress (NS group), a basal diet under heat stress (HT group), or a basal diet under heat stress with the addition of either 0.5 % or 1.0 % Gln. The results showed that the broilers in the HT group exhibited fewer proliferating peripheral lymphocytes, a lower growth performance, phagocytic rate and index of neutrophils, fewer goblet cells in whole intestine and intraepithelial lymphocyte (IEL) cells in the ileum, a lower sIgA content in the duodenum and the jejunum, a lower immunoglobulin content of serum and intestinal mucosa, than those of the NS group (p<0.05). Diets supplemented with Gln increased growth performance, the number of proliferating peripheral lymphocytes, the phagocytic rate and phagocytic index of neutrophils, the number of whole intestine goblet cells and ileum IEL cells, the sIgA contents of the duodenum and the jejunum, and the immunoglobulin contents of serum and intestinal mucosa (p<0.05) in broilers exposed to HT. In conclusion, Gln can enhance intestinal immune function in broiler chickens by stimulating T and B lymphocyte proliferation, increasing the number of goblet cells and IEL cells, as well as increasing the content of sIgA and immunoglobulin secretion.(AU)

Resumo

We describe a case of hypertrophic gastropathy (Ménétriers like disease) with metastatic gastric adenocarcinoma in a seven-year-old intact female Labrador Retriever dog. The animal suddenly presented with emesis and died. Gross lesions included a marked diffuse thickening of the gastric mucosa and an ulcerated transmural neoplastic mass in the gastric body. Gastric body and fundus were affected by foveolar hyperplasia with loss of chief and parietal cells replaced by mucous cells and marked dilatation of gastric glands. An area of gastric adenocarcinoma with submucosal lymphatic vessels invasion was also present and metastases were observed in the gastric lymph nodes, small intestine, pancreas, lung and liver. Due to its similarity with other gastric proliferative disorders, including this condition in the list of differentials is a necessary step in the diagnostic investigation of canine gastropathies.(AU)

Resumo

A senile male captive bush dog (Speothos venaticus) presented a small perianal cutaneous nodule. Histologically, there was an ulcerated round cell tumor composed of well differentiated mast cells with abundant intracytoplasmic purple Giemsa-positive granules, with a diffuse eosinophilic infiltrate. Immunohistochemistry revealed that 30% of the neoplastic cells were positive for Kit in the cytoplasm and cell membrane, and all neoplastic cells were negative for MAC and CD3. Less than 10% of the neoplastic cells were positive for Ki67. At necropsy other primary tumors were identified in this animal, including an intestinal adenoma, an adrenal cortex adenoma and a testicular interstitial cell tumor.(AU)


Um cachorro-vinagre (Speothos venaticus) apresentou um nódulo cutâneo pequeno na região perianal. Histologicamente havia neoplasia cutânea de células redondas e ulcerada, constituída por mastócitos bem diferenciados, com abundantes grânulos citoplasmáticos metacromáticos na coloração de Giemsa e infiltrado eosinofílico difuso. A imuno-histoquímica demonstrou que 30% das células neoplásicas eram positivas para a proteína Kit no citoplasma e na membrana celular. As células foram negativas para MAC e CD3. Menos de 10% das células neoplásicas foram positivas para Ki67. Durante a necropsia, foram identificados outros tumores primários, como adenoma intestinal, adenoma cortical da adrenal e tumor de células intersticiais do testículo.(AU)

Skin and cartilage have been the main source for the recovery of somatic cells to be used in conservation strategies in wild mammals. In this sense, an important step for the cryopreservation of these samples is to recognize the properties of the skin and cartilage. Thus, knowing that the skin may differ among species and aiming to contribute to the establishment of cryobanks, the study examined the differences in the ear skin and cartilage of wild rodents from South America, agouti (Dasyprocta leporina) and spix's yellow-toothed cavy (Galea spixii). Ultrastructural and quantitative methods were used to measure skin and cartilage thickness, density of collagen and elastic fibers, cell type number and distribution, and proliferative activity. Although ultrastructural analysis revealed a similar pattern between species, morphometric analysis of the skin and cartilage showed differences between agoutis and cavies regarding thickness of epidermis layers (corneum: 5.3±2.5μm vs. 3.9±0.6μm; intermediate: 16.4±6.2μm vs. 23.4±8.1μm; basal: 9.9±2.1μm vs. 4.8±0.5μm), dermis (183.1±44.0μm vs. 258.2±22.9μm), total skin (211.8±46.0μm vs. 290.3±23.7μm) and perichondrium (27.6±6.1μm vs. 10.5±1.8μm). A greater number of epidermal cells (61.7±15.2 vs. 24.8±7.6) and chondrocytes (32.7±9.0 vs. 27.5±4.7) were observed in agouti, while the cavy presented a greater number of melanocytes (12.6±4.7 vs. 29.9±6.2), keratinocytes (14.7±4.2 vs. 29.8±7.6), and fibroblasts (103.6±24.7 vs. 112.2±11.3). Moreover, a higher percentage of collagen fibers and proliferative activity was observed in the skin of cavies, when compared to the skin of agoutis. Therefore, there are differences between agouti and cavy for ear skin and cartilage, requiring the establishment of species-specific cryopreservation protocols.(AU)


A pele e cartilagem têm sido uma importante fonte de recuperação de células somáticas a serem utilizadas em estratégias de conservação em mamíferos silvestres. Nesse contexto, uma importante etapa para criopreservação é conhecer, inicialmente, as propriedades que compõem a pele e cartilagem. Sabendo, então, que a pele pode diferir-se entre espécies e com o objetivo de contribuir para o estabelecimento de criobancos, o estudo evidenciou as diferenças da pele e da cartilagem do pavilhão auricular apical de cutias (Dasyprocta leporina) e preás (Galea spixii) que são roedores silvestres presentes na América do Sul. Para tanto, métodos ultraestruturais e quantitativos foram utilizados para mensurar a espessura da pele e da cartilagem, densidade de fibras colágenas e elásticas, número e distribuição dos tipos celulares e atividade proliferativa. Embora as propriedades ultraestruturais em cutias e preás tenham se mostrado semelhantes, avaliações acerca da morfometria da pele e da cartilagem demonstrou diferenças, especialmente nas camadas epidérmicas (córnea: 5,3±2,5μm vs. 3,9±0,6μm; espinhosa: 16,4±6,2μm vs. 23,4±8,1μm; basal: 9,9±2,1μm vs. 4,8±0,5μm), derme (183,1±44,0μm vs. 258,2±22,9μm), pele total (211,8±46,0μm vs. 290,3±23,7μm) e pericôndrio (27,6±6,1μm vs. 10,5±1,8μm). Além disso, um número maior de células epidérmicas (61,7±15,2 vs. 24,8±7,6) e condrócitos (32,7±9,0 vs. 27,5±4,7) foram observados em cutias, enquanto em preás um maior número de melanócitos (12,6±4,7 vs. 29,9±6,2), queratinócitos (14,7±4,2 vs. 29,8±7,6) e fibroblastos (103,6±24,7 vs. 112,2±11,3) foram evidenciados. Ainda, em preás, uma maior porcentagem de fibras colágenas e da atividade proliferativa foram observadas quando comparadas a pele de cutias. Portanto, existem diferenças entre cutias e preás para pele e cartilagem do pavilhão auricular, exigindo desta forma um estabelecimento de protocolos de criopreservação específica para cada uma destas espécies.(AU)
