Resumo

The family Potamotrygonidae are the only species of stingrays restricted to fresh water and located exclusively in South America. The objective of this research was to analyze the morphological aspects and germ cells of the male reproductive tract of Potamotrygon amandae. The samples were fixed in 10% formalin, and then dehydrated in an ascending ethanol series (70 to 100%). To carry out light microscopy analyses, they were embedded in paraffin, cut and stained; as for scanning electron microscopy analyses, the samples were dried, glued in metallic bases and metalized. The gross morphology consisted of the following paired organs: testis, epididymis, deferent duct, Leydig gland, seminal vesicle, clasper, and the clasper gland. Microscopically, several stages of spermatogenesis were observed in the testis, occurring in spherical follicles, similar to other stingrays. The epididymis was formed by one duct subdivided in various tubules. The deferent ducts were continuous with the epididymis, and the lumen was full of spermatozoa. The Leydig glands consisted of glandular units with eosinophilic content in the lumen of some, and the deferent ducts ran parallel to the ventral portion. The seminal vesicles possessed numerous compartments to store the sperm, with a wall similar to a hive, and the lumen was full of spermatozoa. Alcian Blue (AB) and Periodic Schiff-Acid (PAS) performed in the Leydig Gland, deferens ducts and seminal vesicle was positive only in the connective tissue, the cilia were PAS+ and the nuclei stained weakly for AB. The clasper gland was composed of unit glands and was covered with striated muscle externally. It stained very well with Periodic Schiff-Acid. The morphological aspects of the male reproductive tract of Potamotrygon amandae were similar to other stingrays.(AU)

Resumo

Biliary neoplasms are uncommon in cats and affect older animals. A 12-year-old female crossbreed cat showed prostration, lethargy, apathy, and severe jaundice. Ultrasonography showed distention of the gallbladder associated with severe obstruction of the bile ducts with thickening of the biliary wall, forming amorphous masses of irregular contour and heterogeneous appearance directed to the lumen measuring up to 2 cm. Necropsy showed a gallbladder with a yellowish and soft nodule measuring 3 × 3 cm, compressing the extrahepatic bile duct, occluding the passage of bile. There were also firm, yellowish multifocal to coalescing nodules in the liver, ranging from 0.5 to 1 cm, affecting 10% of the organ, in addition to lungs with firm, yellowish multifocal nodules ranging from 0.2 to 0.5 cm, affecting 20% of the organ. Histologically, gallbladder and bile ducts had malignant epithelial neoplastic proliferation, which was organized into multiple papillary and ductal projections, separated by moderate fibrovascular stroma compatible with gallbladder adenocarcinoma. The liver and lungs also contained neoplastic structures with a ductal appearance and papilliform projections identical to those observed in the gallbladder. The immunohistochemical examination (IHC) showed intense positive staining of epithelial neoplastic cells for pan-cytokeratin (AE1/AE3) and no staining for vimentin (Clone V9). The diagnosis of gallbladder adenocarcinoma with metastasis in the liver and lungs was established based on the clinical, macroscopic, histopathological, and immunohistochemical findings.(AU)

Resumo

Termites are among the insects that consume lichens and may be potential dispersers of these symbionts. This study evaluated the vitality of photobionts and mycobionts after passing through the digestive tract of Constrictotermes cyphergaster. The percentage of live and dead algae was verified throughout the alimentary canal of 450 workers, originating from five sampled colonies in the Caatinga Dry Forest, NE, Brazil. A progressive growth in algae mortality was observed in the crop, paunch and rectum, however more than 40% of the algae found in faeces presented signs of vitality. Photobiont morphology was different between cells extracted from thallus in natura and algae present in termite faeces. The photobiont cells presented more shrunken cytoplasms after passing through the alimentary canal of C. cyphergaster. There was also an increase between the cell wall space and the cytoplasm membrane of algae found in the termite faecal pellets. Only four broken spores were found in the intestine, which made the vitality analysis unfeasible for these cells. The record of photobiont vitality in termite faecal pellets is indicative of endozoochoric dispersal, suggesting that this relationship between insects and lichens extends beyond a trophic interaction.


Os térmitas estão entre os insetos consumidores de liquens e representam um potencial dispersor desses simbiontes. Este estudo avaliou a vitalidade de fotobiontes e micobiontes depois de terem passado pelo trato digestivo de Constrictotermes cyphergaster. O percentual de algas vivas e mortas foi verificado ao longo do canal alimentar de 450 operários, oriundos de cinco colônias amostradas em Caatinga Dry Forest, NE, Brazil. Um crescimento progressivo na mortalidade das algas foi observado no sentido papo, pança e reto, porém mais de 40% das algas presentes nas fezes apresentaram sinais de vitalidade. A morfologia dos fotobiontes foi diferente entre células extraídas de talos in natura e algas presentes nas fezes dos térmitas. As células do fotobionte apresentaram seu citoplasma mais encolhido, após a passagem pelo canal alimentar de C. cyphergaster. Também houve um aumento entre o espaço da parede celular e a membrana do citoplasma das algas encontradas nas pelotas fecais dos térmitas. Apenas quatro esporos quebrados foram encontrados no intestino dos cupins, o que inviabilizou a análise de vitalidade dessas células. O registro da vitalidade de fotobiontes nas pelotas fecais dos térmitas é um indicativo de dispersão endozoocórica, remetendo que a relação entre esses insetos e os liquens pode ir além de uma interação trófica.

Onychomycosis is the most common disease affecting the nail unit and accounts for at least 50% of all nail diseases. In addition, Candida albicans is responsible for approximately 70% of onychomycoses caused by yeasts. This study investigated the antifungal effect of (R) and (S)-citronellal enantiomers, as well as its predictive mechanism of action on C. albicans from voriconazole-resistant onychomycoses. For this purpose, in vitro broth microdilution and molecular docking techniques were applied in a predictive and complementary manner to the mechanisms of action. The main results of this study indicate that C. albicans was resistant to voriconazole and sensitive to the enantiomers (R) and (S)-citronellal at a dose of 256 and 32 µg/mL respectively. In addition, there was an increase in the minimum inhibitory concentration (MIC) of the enantiomers in the presence of sorbitol and ergosterol, indicating that these molecules possibly affect the integrity of the cell wall and cell membrane of C. albicans. Molecular docking with key biosynthesis proteins and maintenance of the fungal cell wall and plasma membrane demonstrated the possibility of (R) and (S)-citronellal interacting with two important enzymes: 1,3-ß-glucan synthase and lanosterol 14α-demethylase. Therefore, the findings of this study indicate that the (R) and (S)-citronellal enantiomers are fungicidal on C. albicans from onychomycoses and probably these substances cause damage to the cell wall and cell membrane of these micro-organisms possibly by interacting with enzymes in the biosynthesis of these fungal structures.


A onicomicose é a doença mais comum que afeta a unidade ungueal e representa pelo menos 50% de todas as doenças ungueais. Além disso, a Candida albicans é responsável por aproximadamente 70% das onicomicoses causadas por leveduras. Nesse estudo, foi investigado o efeito antifúngico dos enantiômeros (R) e (S)-citronelal, bem como seu mecanismo de ação preditivo sobre C. albicans de onicomicoses resistentes ao voriconazol. Para este propósito, foram aplicadas técnicas in vitro de microdiluição em caldo e docking molecular de forma preditiva e complementar para os mecanismos de ação. Os principais resultados deste estudo indicam que C. albicans foi resistente ao voriconazol e sensível aos enantiômeros (R) e (S)-citronelal na dose de 256 e 32 µg/mL respectivamente. Além disso, houve aumento da concentração inibitória mínima (CIM) dos enantiômeros na presença do sorbitol e do ergosterol, indicando que estas moléculas possivelmente afetem a integridade da parede e da membrana celular de C. albicans. O docking molecular com proteínas chave da biossíntese e manutenção da parede celular e da membrana plasmática fúngica, demonstraram a possibilidade do (R) e (S)-citronelal interagir com duas importantes enzimas: 1,3-ß-glucan sintase e lanosterol 14α-demetilase. Portanto, os achados desse estudo indicam que os enantiômeros (R) e (S)-citronelal são fungicidas sobre C. albicans de onicomicoses e provavelmente essas substâncias causem danos a parede e a membrana celular desses microrganismos possivelmente por interagir com as enzimas da biossíntese destas estruturas fúngicas.

Background: Osteosarcoma is the most observed primary bone tumor in dogs, and may affect the appendicular and axial skeletons. In addition, it may be present in extraskeletal form, accounting for only 1% of cases. As shown by few reports in the literature, the involvement of the intestinal region by is rare. The objective of this study was to report the case of a 13-year-old Yorkshire dog, submitted to an exploratory laparotomy for suspected partial intestinal obstruction, diagnosed with extraskeletal osteosarcoma. Case: A 13-year-old dog, Yorkshire Terrier, male, presented clinical signs of gastrointestinal abnormalities. An ultrasound examination was performed and was found a mass in small intestine region with wall and lumen invasion. Then, was realized exploratory laparotomy and detected intestinal obstruction due to a mass with approximately 5.0 x 6.0 x 4.4 cm localized in duodenum. Surgical removal was performed and the sample sent to the veterinary diagnostic laboratory for histopathological examination. The sample had an irregular surface and firm consistency. In addition, when cut, the mass enveloped the intestinal layers and sometimes obstructed the lumen. Then, the sample were processed routinely for histopathology. After that, in microscopy evaluation was detected cell proliferation, affecting all layers of intestine. In detail, cells were elongated with pleomorphism marked and atypical mitosis. In addition, there was production of cartilage and bone matrix. So, due the absence of others sites, the neoplasm was considered primary of intestine. After that, to evaluate the expression of KI-67 and COX-2 was performed, and the cell proliferation index was 54.0% and the COX-2 expression was moderate in less than 10% of neoplastic cells. After the surgery, the patient was hospitalized for a week and continue the treatment in home. Afterwards, the tutor received the diagnosis, but even though he was instructed on the severity of the case, he chose not to undergo chemotherapy. After three months, the patient presented abdominal fluid and nodules in your liver, suggesting metastasis, but without diagnosis confirmation. The patient died five months after the diagnosis of extraskeletal osteosarcoma. However, no necropsy was realized, impossibility the diagnosis confirm. Discussion: The frequency of extraskeletal osteosarcoma in dogs remains unknown, with the mammary glands being the most affected site. In the present study, osteosarcoma affects the duodenal region and no reports of this neoplasm in the duodenum of dogs have been found in the literature. The clinical sign of dyschezia was important for the tutor to refer the animal to the veterinarian and perform the ultrasound in an attempt to elucidate the case, as the tumor mass is not always palpable. Histopathological examination and immunohistochemistry were necessary for the differential diagnosis and to establish the prognosis, although after the surgery the tutor chose not to perform chemotherapy. Extraskeletal osteosarcoma are usually highly metastatic, mainly affecting the lymph nodes and liver. In this case, the patient presented a liver nodule three months after the tumor removal surgery, but unfortunately, there was no diagnostic confirmation. Such neoplastic type is rarer and more aggressive than appendicular and axial osteosarcoma, with an average survival of 1 to 3 months. In this case, as a necropsy was not obtained, we cannot attribute the survival time to the disease. The survival rates of osteosarcomas in dogs are few months, but in the present case, although the patient died five months after surgery, the failure to perform a necropsy compromises the attribution of survival time to extraskeletal osteosarcoma.

Background: Caudal vena cava thrombosis (CVCT) is a serious disease that affects cattle. Due to being commonly a fatal pathology, it causes economic losses for producers and national livestock. Thus, the present study describes the epidemiological, clinical, ultrasonographic imaging and pathological findings in 4 cattle with CVCT attended at the Veterinary Hospital (HV) of the Institute of Veterinary Medicine (IMV) of the Federal University of Pará (UFPA). Cases: The animals were crossbreds of the Gir x Holstein and Jersey x Holstein breeds, aged between 4 and 8 years old, raised in semi-extensive and intensive systems. The main clinical signs were pale mucous membranes, reluctance to move, markedly positive venous pulse, engorged jugular with positive stasis test, and serous to mucopurulent nasal exudation. The auscultation of the lung fields revealed tachypnea, silent areas, wheezing, and pleural friction, in addition to coughing, expiratory dyspnea, mouth breathing, and expiratory grunts. One animal had severe hemoptysis. The ultrasound examination performed on a bovine revealed a circular and dilated caudal vena cava in cross-section. Laboratory tests in 3 cattle revealed anemia, leukocytosis with neutrophil left shift, and increased liver enzymes. At necropsy, all cattle had thrombi in the hepatic segment of the caudal vena cava. In the lung, multiple abscesses and areas of parenchymal consolidation, crateriform areas, as well as thrombi in the arteries were observed. Pleural effusion and ascites were seen in all cattle. Clotted blood was seen in the trachea, bronchi, and on rumen contents of an animal. Histopathological alterations seen in the liver were centrilobular hepatocytes with frequent intensely eosinophilic cytoplasm, and pyknotic, karyorrhexic, or absent nuclei and cell borders barely distinguishable. In the lung were nodular and random formations, with a thick wall of mature connective tissue and a central area full of cellular debris, necrotic cells, and intact and degenerated neutrophils (abscesses). Discussion: The set of diagnostic tools that include epidemiology, clinical signs and clinical examinations, ultrasound, necropsy, and histopathology were efficient in the diagnosis of CVCT. The possible causes that led the animals to develop CVCT were diffuse septic pododermatitis in the medial nail of the right pelvic limb associated with traumatic reticuloabomasitis and liver abscesses. In 1 cow, it was not possible to establish the probable cause of CVCT, but for the other cattle in the present study, the probable causes are in agreement with studies that have shown that this disease can occur as a sequel to several septic conditions such as jugular phlebitis, mastitis, hoof rot, enteritis, pneumonia, traumatic reticulopericarditis, acidosis and rumen laminitis, as well as omphalophlebitis in calves. The tachypnea, serous to purulent nasal exudation, pulmonary wheezing, pleural friction, coughing, and expiratory dyspnea, usually with open mouth breathing and expiratory grunts evidenced in the animals of this study, occurred as a result of embolic abscess pneumonia. The presence of multiple lung abscesses, areas of parenchymal consolidation, crateriform foci, and thrombi in the pulmonary arteries and chronic suppurative pneumonia, found at necropsy of the animals in the present study, are related to the development of a thrombus in the caudal vena cava that detaches and embolizes and lodges in the pulmonary arteries. The histopathological findings in 1 cow are compatible with lesions found at necropsy and draw attention to embolic pneumonia and liver lesions, which, are related to thrombi in pulmonary arteries and abscesses formed from CVCT, as well as venous stasis exerted in the return circulation.

Resumo

This study investigated the effects of adding different adsorbent substances to broilers feed naturally contaminated by mycotoxins. Two hundred and eighty male 1-day-old chicks, Cobb Slow® lineage, were distributed in a randomized block design with 4 treatments, 5 repetitions with 14 birds each. The treatments consisted of: T1-basal feed naturally contaminated with mycotoxins. T2-basal feed + Bentonite, Thistle Extract, Yeast Extract, Vitamin E and Choline. T3-basal feed + Bentonite, Thistle Extract, yeast cell wall and Silymarin. T4-basal feed + Bentonite and Algae extract. Performance (weight gain, feed intake, feed conversion) at 7, 14, 21, 28 days were evaluated. At 28 days, a portion of the jejunum was collected in two birds by replicate to study the intestinal morphology. The relative weight of the gizzard, proventricle and total intestine was evaluated. The data obtained were analyzed using the statistical program SAS (9.3). With the use of any adsorbents studied, the performance and liver weight were improved in all evaluated periods. Thus, the inclusion of adsorbents improves the performance of the broiler chickens when the feed is contaminated by mycotoxins.(AU)

Abstract High resistance to antimicrobials is associated with biofilm formation responsible for infectious microbes to withstand severe conditions. Therefore, new alternatives are necessary as biofilm inhibitors to control infections. In this study, the antimicrobial and antibiofilm activities of Fagonia indica extracts were evaluated against MDR clinical isolates. The extract exhibited its antibiofilm effect by altering adherence and disintegration of bacterial cell wall. Fagonia indica has antibacterial effect as minimum inhibitory concentration (MIC) values ranging from 125 to 500 µg mL-1 and minimum bactericidal concentration (MBC) value was 500-3000 µg mL-1 against multidrug resistant (MDR) clinical isolates. The extract exhibited its antibiofilm effect by altering adherence and disintegration of bacterial cell wall. Fagonia indica had antibacterial effect as minimum inhibitory concentration (MIC) values ranging from 125 to 500 µg mL-1 and minimum bactericidal concentration (MBC) value was 500-3000 µg mL-1 against MDR isolates. The maximum inhibitory effects of Fagonia indica chloroform extract on biofilm formation was observed on Staphylococcus aureus (71.84%) followed by Klebsiella pneumoniae (70.83%) after 48 hrs showing that inhibition is also time dependent. Our results about bacterial cell protein leakage indicated that MDR isolates treated with chloroform extract of Fagonia indica showed maximum protein leakage of K. pneumoniae (59.14 µg mL-1) followed by S. aureus (56.7 µg mL-1). Cell attachment assays indicated that chloroform extract resulted in a 43.5-53.5% inhibition of cell adherence to a polystyrene surface. Our results revealed that extracts of Fagonia indica significantly inhibited biofilm formation among MDR clinical isolates, therefore, could be applied as antimicrobial agents and cost effective biofilm inhibitor against these MDR isolates.


Resumo A alta resistência aos antimicrobianos está associada à formação de biofilme responsável por micróbios infecciosos para suportar condições severas. Portanto, novas alternativas são necessárias como inibidores de biofilme para controlar infecções. Neste estudo, as atividades antimicrobiana e antibiofilme dos extratos de Fagonia indica foram avaliadas contra isolados clínicos MDR. O extrato exibiu seu efeito antibiofilme ao alterar a aderência e a desintegração da parede celular bacteriana. Fagonia indica tem efeito antibacteriano com valores de concentração inibitória mínima (CIM) variando de 125 a 500 µg mL-1, e valor de concentração bactericida mínima (MBC) de 500-3000 µg mL-1 contra isolados clínicos multirresistentes (MDR). O extrato exibiu seu efeito antibiofilme ao alterar a aderência e a desintegração da parede celular bacteriana. Fagonia indica teve efeito antibacteriano com valores de concentração inibitória mínima (CIM) variando de 125 a 500 µg mL-1, e concentração bactericida mínima (MBC) de 500-3000 µg mL-1 contra isolados MDR. Os efeitos inibitórios máximos do extrato de clorofórmio Fagonia indica na formação de biofilme foi observada em Staphylococcus aureus (71,84%), seguido por Klebsiella pneumoniae (70,83%) após 48 horas, mostrando que a inibição também é dependente do tempo. Nossos resultados sobre extravasamento de proteínas de células bacterianas indicaram que isolados MDR tratados com extrato clorofórmico de Fagonia indica apresentaram vazamento máximo de proteínas de K. pneumoniae (59,14 µg mL-1), seguido por S. aureus (56,7 µg mL-1). Ensaios de fixação de células indicaram que o extrato de clorofórmio resultou em uma inibição de 43,5-53,5% da aderência das células a uma superfície de poliestireno. Nossos resultados revelaram que extratos de Fagonia indica inibiram significativamente a formação de biofilme entre isolados clínicos MDR, portanto, poderiam ser aplicados como agentes antimicrobianos e inibidores de biofilme de baixo custo contra esses isolados MDR.

High resistance to antimicrobials is associated with biofilm formation responsible for infectious microbes to withstand severe conditions. Therefore, new alternatives are necessary as biofilm inhibitors to control infections. In this study, the antimicrobial and antibiofilm activities of Fagonia indica extracts were evaluated against MDR clinical isolates. The extract exhibited its antibiofilm effect by altering adherence and disintegration of bacterial cell wall. Fagonia indica has antibacterial effect as minimum inhibitory concentration (MIC) values ranging from 125 to 500 µg mL-1 and minimum bactericidal concentration (MBC) value was 500-3000 µg mL-¹ against multidrug resistant (MDR) clinical isolates. The extract exhibited its antibiofilm effect by altering adherence and disintegration of bacterial cell wall. Fagonia indica had antibacterial effect as minimum inhibitory concentration (MIC) values ranging from 125 to 500 µg mL-¹ and minimum bactericidal concentration (MBC) value was 500-3000 µg mL-¹ against MDR isolates. The maximum inhibitory effects of Fagonia indica chloroform extract on biofilm formation was observed on Staphylococcus aureus (71.84%) followed by Klebsiella pneumoniae (70.83%) after 48 hrs showing that inhibition is also time dependent. Our results about bacterial cell protein leakage indicated that MDR isolates treated with chloroform extract of Fagonia indica showed maximum protein leakage of K. pneumoniae (59.14 µg mL-¹) followed by S. aureus (56.7 µg mL-1). Cell attachment assays indicated that chloroform extract resulted in a 43.5-53.5% inhibition of cell adherence to a polystyrene surface. Our results revealed that extracts of Fagonia indica significantly inhibited biofilm formation among MDR clinical isolates, therefore, could be applied as antimicrobial agents and cost effective biofilm inhibitor against these MDR isolates.(AU)

ß-Glucans (ßG) are polysaccharides widely distributed in nature with chemopreventive properties. The aim of this study was to investigate the effects of ßG and the combined treatment with doxorubicin (Dox) on cell viability and mRNA levels of genes involved in cell cycle, apoptosis and antioxidant response. ßG was not cytotoxic. The mRNA levels of CCNA2of cells exposed to ß-glucan was upregulated and the exposure to Dox decreased the expression, while the combination led to an upregulation. Modulation of mRNA levels of CASP9suggest that ßG could inhibit promotion and progression steps of carcinogenesis, eliminatingneoplastic cells. The upregulation of CCNA2gene in combined treatment could be occurred due to ability of ßG in restoring the cell cycle distribution pattern after treatment with Dox. The upregulation of SOD1suggests that ßG can enhance the intracellular antioxidant defense, reducing the levels of superoxide dismutase induced by Dox. This response could reduce oxidative damage and attenuate tissue damage during chemotherapeutic treatment. Our data suggest that the drug combination may be less effective in killing tumor cells than the treatment with Dox alone. They are constituent of the cell wall of plants (oats and barley), algae, bacteria and fungi. ß-glucans (ßG)have a common structure comprising a main chain of ß-(1,3) and/or ß-(1,4) D-glucopyranosyl unit and they differ in length and branching structures. ßG of Saccharomyces cerevisiaehave 1→6 side branches while those of bacteria have 1→4 side branches (Chan, Chan, & Sze, 2009). ßGcan prevent DNA damage induced by chemical and physical agents (Ghavami,Goliaei, Taghizadeh, & Nikoofar, 2014). Some authors showed its significant efficacy in preventing mutagenic effects caused by doxorubicin, cyclophosphamide and cisplatin (Tohamy, El-Ghor, El-Nahas, & Noshy, 2003), methyl methanesulfonate (Oliveira et al., 2007)and hydrogen peroxide (Slamenová, 2003). Moreover, some studies have related the antioxidant ability of ßGagainst reactive free radicals formed by endogenous metabolic processes or exogenous chemicals (Tsiapali et al., 2001; Slamenová,2003; Sener, Eksioglu-Demiralp, Cetiner, Ercan, & Yegen, 2006; Guerra Dore et al., 2007; Kofuji et al., 2012; Lei et al., 2015). Yeast-derived ßGhave modulating action of humoral and cellular immune responses (Vetvicka et al., 2007).This activity provides protection to the organism against infections and cancer development (Samuelsen, Schrezenmeir, & Knutsen, 2014; Roudbary, Daneshmand, Hajimorad, Roudbarmohammadip, & Hassan, 2015). Despite postulated modes of action by which ß-glucan works are lacking information about the molecular mechanisms involved in the chemopreventive activity of this polysaccharide. In addition, compounds with chemopreventive properties can contribute to reduce side effects and toxicity during the chemotherapeutic treatment. Therefore, the aim of this study was to investigate the effects of ßG and the combined treatment with doxorubicin (Dox) on the expression of genes related with apoptosis (CASP9), cell cycle control (CCNA2)and antioxidant defense (SOD1)in human breast cancer MCF-7 cells. Doxorubicin (Dox) was chosen because it is one of the most used chemotherapeutic agent for cancer treatment. The limitation on the use of Dox in cancer treatment is the lack of selectivity against cancer cells and, consequently, its toxicity to patients.(AU)

ABSTRACT: It is reported the occurrence of enzootic hematuria (EH) in buffaloes in Brazil after performing an epidemiological survey and clinicopathological analises. To date, EH caused by ingestion of Pteridium esculentum subsp. arachnoideum, a radiomimetic plant popularly known as bracken fern, has not been described in this species in Brazil. Bovine EH is responsible for high economic losses in Brazils Southeast Region not only because of the deaths it causes, but also owing to its negative effect on productivity. In São José do Barreiro County, São Paulo, some farmers in areas with a high incidence of bovine EH have been replacing cattle with buffaloes, based on the premise that the latter would be more resistant to poisoning by ingestion of Pteridium spp. However, even though initial observations indicated that buffaloes are indeed less sensitive than cattle to the toxic principle of Pteridium spp., cases of hematuria in this species have been reported. According to preliminary date, EH only occurs in buffaloes over six years of age. Macroscopic examination revealed a thickened urinary vesicle mucosa, along with multiple foci of ulcerated, exophytic, verrucous, and pedunculated lesions. In one of the buffaloes studied, the bladder wall was ruptured and exhibited marked secondary inflammation. Histologically, neoplastic and non-neoplastic changes similar to those described in cattle poisoned by Pteridium spp. were observed. The neoplasms found included papilloma, carcinoma in situ, urothelial carcinoma (low and high grade), inverted, microcystic, and trabecular variants, urothelial carcinoma with divergent differentiation (squamous and glandular), squamous cell carcinoma, lymphangioma, hemangioma, and hemangiosarcoma. There was also coexistence of epithelial and mesenchymal neoplasms. Bovine papillomavirus particles were not detected by polymerase chain reaction in the bladder samples analyzed.


RESUMO: Descreve-se, através de levantamento epidemiológico e avaliação clínico-patológica, a ocorrência de hematúria enzoótica (HE) em búfalos no Brasil. Essa condição, causada pela ingestão da planta radiomimética Pteridium esculentum subsp. arachnoideum, conhecida popularmente como samambaia ou samambaia do campo, até então não havia sido descrita nessa espécie no Brasil. Na Região Sudeste, a HE bovina é responsável por elevadas perdas econômicas, devidas não apenas aos óbitos, mas também em função da queda de produtividade. No município de São José do Barreiro/SP, alguns produtores de áreas com alta incidência de HE bovina, vêm substituindo os bovinos por búfalos, com base na premissa de que estes seriam mais resistentes à intoxicação. Embora, de acordo com observações iniciais, os búfalos realmente sejam menos sensíveis que os bovinos ao princípio tóxico de Pteridium spp., ainda assim, tem-se verificado a ocorrência de casos de hematúria nessa espécie. De acordo com o levantamento inicial, a HE só ocorre em búfalos com idade a partir de seis anos. Ao exame macroscópico, verificou-se a mucosa da bexiga espessa, com múltiplos focos de lesões ulceradas, exofíticas, papiliformes, verrucosas, pedunculadas. Histologicamente, foram observadas alterações neoplásicas e não neoplásicas semelhantes às descritas nos bovinos com HE. Entre as neoplasias foram encontrados papiloma, carcinoma in situ, carcinoma urotelial (baixo e alto grau), variantes invertida, microcística e trabecular, carcinoma urotelial com diferenciação divergente (escamosa e glandular), carcinoma de células escamosas, linfangioma, hemangioma e hemangiossarcoma. Ocorreu também coexistência entre neoplasias epiteliais e mesenquimais. Não foram detectadas partículas de papilomavírus bovino pelo teste PCR nas amostras de bexiga analisadas.

O segmento de produção de papel de fibra longa no Brasil está concentrado na utilização de duas espécies do gênero Pinus (P. taeda e P. elliottii). Assim, éfundamental avaliar novas espécies, visando ampliar as alternativas para esse segmento. Opresente estudo teve como objetivo avaliar as propriedades físicas, químicas e anatômicas da madeira de Pinus greggii visando a produção de polpa celulósica. Foram utilizadas três árvores, com seteanos de idade, das quais foram retirados discos ao longo do fuste. Estes foram utilizados para avaliar as propriedades físicas (densidade básica média e ponderada), químicas (teores de cinza, extrativos, lignina e holocelulose) e anatômicas (dimensões dos traqueídeos e indicadores anatômicos). De acordo com a caracterização físicada madeira, obteve-se um valor médio de densidade básica de 0,346 g.cm-3e ponderadode 0,343 g.cm-3, o que permite classificá-la como leve ou de baixa densidade. Em relação à composição química, a espécie apresentou elevados teores de lignina e extrativos (34,83% e 6,25%, respectivamente) e baixos teores de holocelulose (58,92%) e cinza (0,20%). Para as dimensões dos traqueídeos obtiveram-se valores médios de 2,31 mm de comprimento, 41,30 µm de largura, 34,81 µm de diâmetro do lúmen, e 3,25 µm de espessura da parede celular. Quanto aos resultados dos índices de qualidade, a espécie obteve destaque para a fração parede (15,96%), coeficiente de flexibilidade (84,31%), índice de Runkel (0,19) e índice de enfeltramento (55,15). De modo geral, a madeira de P. greggiiapresenta potencial para produção de polpa celulósica, destacando-se pelos indicadores anatômicos e pelas dimensões dos traqueídeos, que são semelhantes aos valores encontrados para P. taedana mesma idade. Entretanto, possui elevados teores de lignina e extrativos, que podem aumentar o consumo de reagentes e diminuir o rendimento do processo de polpação, o que precisa ser comprovado em estudos com esta temática.(AU)


The long fiber paper production segment in Brazil is restrict on the use of only two species of the Pinusgenus (P. taedaand P. elliottii), therefore, it is essential to evaluate new species, aiming to expand the alternatives for this segment. In this sense, the present study aimed to evaluate the physical, chemical and anatomical properties of Pinus greggiiwood aiming at the pulp production. For this, three 7-year-old trees were used, from which disks were removed along the stem. These were used to evaluate physical (average and weighted basic density), chemical (ash, extractive, lignin and holocellulose) and anatomical (tracheid dimensions and anatomical indicators) properties. According to the physical characterization, an average value of basic density of 0.346 g.cm-3and weighted of 0.343 g.cm-3was obtained, which allows classifying it as light or low density. Regarding the chemical composition, the species showed high levels of lignin and extractives (34.83% and 6.25%, respectively) and low levels of holocellulose (58.92%) and ash (0.20%). For the tracheids dimensions, mean values of 2.31 mm in length,41.30 µm in width, 34.81 µm in lumen diameter, and 3.25 µm in cell wall thickness were obtained. As for the results of the quality index, the species stood out for the wall fraction (15.96%), flexibility coefficient (84.31%), Runkel index (0.19) and felting index (55.15). In general, the P. greggiiwood presents potential for the pulp production, standing out for its anatomical indicators and for the tracheids dimensions, which are similar to the values found for P. taedaat the same age. However, it has high levels of lignin and extractives, which can increase the consumption of reagents and decrease the yield of the pulping process, which needs to be proven in studies on this topic.(AU)

The goal of this study was to evaluate the effects of successive harvests, with or without application of foliar fungicide, on the morphometric characteristics, yield and nutritional value of rye forage harvested at the hard dough grain stage, aiming at silage production. The preventive control of foliar diseases was carried out with the fungicide OrkestraTM SC®, in a single application, at early flowering, for both harvesting systems. The first harvest was conducted at the full vegetative stage and the second, at the hard dough grain stage. The system with two harvests resulted in higher cumulative yield compared with the single-harvest system (40,680 and 9,029 kg ha-1 fresh and dry biomass with two harvests, against 8,816 and 5,375 kg ha-1 fresh and dry biomass). Fungicide application promoted a reduction in neutral detergent fiber content in both systems, with values of 753.9 against 790.6 g kg-1 for the single-harvest system and 734.4 against 773.3 g kg-1 for the two-harvest system, with and without fungicide, respectively. For lignin content, the application of the fungicide reduced values (97.2 against 110.3 g kg-1) only in the two-harvest system. Rye management with a harvest at the vegetative stage increases the cumulative dry biomass yield without negatively affecting the harvest yield at the hard dough grain stage, and without drastic changes in the nutritional value of the plant. Even under adverse conditions, fungicide application positively interferes with plant cell wall componentes.(AU)


O objetivo desse estudo foi avaliar os efeitos de sistemas de cortes sucessivos, com ou sem uso de fungicida foliar, sobre as características morfométricas, o rendimento e o valor nutricional da forragem de centeio colhida em estágio de grão farináceo, visando a confecção de silagem. O controle preventivo de doenças foliares foi feito com o fungicida OrkestraTM SC®, em aplicação única, em estágio inicial de florescimento, para ambos sistemas de corte. A sucessão de cortes teve o primeiro em estágio de pleno vegetativo e o segundo em estágio de grão farináceo. O sistema com dois cortes gerou maior rendimento acumulado em comparação ao sistema de corte único (40.680 e 9.029 kg ha-1 de biomassa fresca e seca com dois cortes, contra 8.816 e 5.375 kg ha-1 de biomassa fresca e seca). O uso de fungicida promoveu redução nos teores de fibra em detergente neutro em ambos os sistemas, com valores de 753,9 contra 790,6 g kg-1 para o sistema de um corte e 734,4 contra 773,3 g kg-1 para o sistema de dois cortes, com e sem fungicida respectivamente. Para teores de lignina, apenas no sistema de dois cortes o uso de fungicida promoveu redução de valores (97,2 contra 110,3 g kg-1). O manejo do centeio com um corte no estágio vegetativo eleva o rendimento de biomassa seca acumulada sem prejudicar o rendimento para colheita em grão farináceo, e tampouco promove mudanças drásticas no valor nutricional da planta. Mesmo em condições adversas, o uso de fungicida interfere positivamente nos componentes da parede celular da planta.(AU)

Background: Cladophialophora bantiana is a dematiaceous fungus that causes phaeohyphomycosis, a generic term used to describe a variety of unusual mycoses caused by fungi that have melanin in their cell wall. C. bantiana targets the central nervous system, commonly causing localized brain infections that may result in disseminated infections. In Brazil, minimal phaeohyphomycosis data are available, and information about C. bantiana infections in animals, especially canines, is scarce. Thus, the aim of this study was to describe the clinical and pathological aspects of systemic phaeohyphomycosis caused by C. bantiana in a dog. Case: A 1-year-old female Pit Bull presented with weight loss, reduced appetite, and a history of cutaneous lesions on the right thoracic limb; however, clinical evolution was not reported. The bitch had reportedly given birth recently. Physical examination revealed thinness, pale ocular and oral mucosa, submandibular lymph nodes, and enlarged popliteal lymph nodes. The bitch died after convulsive crises during hospitalization. At necropsy, white-yellowish multifocal nodules were observed in the liver and right kidney. The brain featured left cerebral hemisphere asymmetry with blood vessel congestion in the leptomeninges and an irregular brownish focal area on the surface of the right occipital cortex. Cross-sections of the formalin-fi xed brain exhibited compression of the left lateral ventricle and the presence of grayish and friable multifocal areas in the gray matter of the left parietal and right occipital cortices. Fragments of the lesions were collected for histopathological and microbiological examination. Resumo

It is reported the occurrence of enzootic hematuria (EH) in buffaloes in Brazil after performing an epidemiological survey and clinicopathological analises. To date, EH caused by ingestion of Pteridium esculentum subsp. arachnoideum, a radiomimetic plant popularly known as "bracken fern", has not been described in this species in Brazil. Bovine EH is responsible for high economic losses in Brazil's Southeast Region not only because of the deaths it causes, but also owing to its negative effect on productivity. In São José do Barreiro County, São Paulo, some farmers in areas with a high incidence of bovine EH have been replacing cattle with buffaloes, based on the premise that the latter would be more resistant to poisoning by ingestion of Pteridium spp. However, even though initial observations indicated that buffaloes are indeed less sensitive than cattle to the toxic principle of Pteridium spp., cases of hematuria in this species have been reported. According to preliminary date, EH only occurs in buffaloes over six years of age. Macroscopic examination revealed a thickened urinary vesicle mucosa, along with multiple foci of ulcerated, exophytic, verrucous, and pedunculated lesions. In one of the buffaloes studied, the bladder wall was ruptured and exhibited marked secondary inflammation. Histologically, neoplastic and non-neoplastic changes similar to those described in cattle poisoned by Pteridium spp. were observed. The neoplasms found included papilloma, carcinoma in situ, urothelial carcinoma (low and high grade), inverted, microcystic, and trabecular variants, urothelial carcinoma with divergent differentiation (squamous and glandular), squamous cell carcinoma, lymphangioma, hemangioma, and hemangiosarcoma. There was also coexistence of epithelial and mesenchymal neoplasms. Bovine papillomavirus particles were not detected by polymerase chain reaction in the bladder samples analyzed.


Descreve-se, através de levantamento epidemiológico e avaliação clínico-patológica, a ocorrência de hematúria enzoótica (HE) em búfalos no Brasil. Essa condição, causada pela ingestão da planta radiomimética Pteridium esculentum subsp. arachnoideum, conhecida popularmente como "samambaia" ou "samambaia do campo", até então não havia sido descrita nessa espécie no Brasil. Na Região Sudeste, a HE bovina é responsável por elevadas perdas econômicas, devidas não apenas aos óbitos, mas também em função da queda de produtividade. No município de São José do Barreiro/SP, alguns produtores de áreas com alta incidência de HE bovina, vêm substituindo os bovinos por búfalos, com base na premissa de que estes seriam mais resistentes à intoxicação. Embora, de acordo com observações iniciais, os búfalos realmente sejam menos sensíveis que os bovinos ao princípio tóxico de Pteridium spp., ainda assim, tem-se verificado a ocorrência de casos de hematúria nessa espécie. De acordo com o levantamento inicial, a HE só ocorre em búfalos com idade a partir de seis anos. Ao exame macroscópico, verificou-se a mucosa da bexiga espessa, com múltiplos focos de lesões ulceradas, exofíticas, papiliformes, verrucosas, pedunculadas. Histologicamente, foram observadas alterações neoplásicas e não neoplásicas semelhantes às descritas nos bovinos com HE. Entre as neoplasias foram encontrados papiloma, carcinoma in situ, carcinoma urotelial (baixo e alto grau), variantes invertida, microcística e trabecular, carcinoma urotelial com diferenciação divergente (escamosa e glandular), carcinoma de células escamosas, linfangioma, hemangioma e hemangiossarcoma. Ocorreu também coexistência entre neoplasias epiteliais e mesenquimais. Não foram detectadas partículas de papilomavírus bovino pelo teste PCR nas amostras de bexiga analisadas.

Bovine enzootic hematuria (BEH) is a chronic disease caused by ingestion of Pteridium esculentum subsp. arachnoideum (Kaulf.) Thomson. This disease has great importance because it affects the animal wellbeing, causes economic losses, and hinders public health due to the toxic principle that is present in the milk of cows that ingest this plant. Early diagnosis assists in the decision making for disposal and replacement of animals in endemic regions. Despite ultrasonography is a useful tool for diagnosis of this disease, there is a lack of studies evaluating BEH in the state of Tocantins, Brazil, using ultrasound and epidemiological data and clinical signs; however, the histopathological examination is the method that provides the definitive diagnosis. The objective of this study was to describe the occurrence of BEH in the state of Tocantins and evaluate the efficiency of bladder ultrasound examination for diagnosis and prognosis of BEH. Ten animals that presented intermittent hematuria and hypo-colored mucous membranes were subjected to clinical and complementary exams. The urine physical analysis presented light red color, high turbidity, and positive sedimentation. The ultrasonography showed a thick, irregular mass in the bladder wall, containing an echogenic liquid of high cellularity and pedunculated papillary lesions. Macro and microscopic analyses of the animals' bladders showed areas with vascular proliferation in the sub-mucous membranes associated with mononuclear inflammatory cell infiltrate and proliferation of the epithelium. Ultrasonography is an excellent option and, combined with clinical and laboratory exams of blood and urine, enables the diagnosis and prognosis of BEH. (AU)


Hematúria enzoótica bovina (HEB) doença crônica causada pela ingestão de Pteridium esculentum subsp. arach-noideum (Kaulf.) Thomson. Essa intoxicação tem grande importância no bem-estar animal, causando prejuizos econômicos e na saúde pública, uma vez que o princípio tóxico está presente no leite de vacas que ingerem a planta. Em regiões endêmicas o diagnóstico precoce auxilia na tomada de decisões, como o descarte e a substituição dos animais. Apesar da ultrossonografia ser uma ferramenta diagnóstica muito útil, há uma carência de estudos de HEB no estado do Tocantins no que diz respeito aos dados ultrassonográficos, epidemiológicos e sinais clínicos; entretanto, o exame histopatológico é o diagnóstico definitivo. O objetivo deste estudo foi descrever a ocorrência da doença neste estado e avaliar a eficácia do exame ultrassonográfico da bexiga urinária para o diagnóstico e prognóstico de HEB. Foram 10 animais submetidos a exames clínicos e complementares, ao exame apresentavam hematúria intermitente e mucosas hipocoradas. A análise física da urina apresentou-se vermelho vivo, turva e com sedimentação positiva. A ultrassonografia revelou massa na parede da bexiga urinária, espessa e irregular, contendo líquido ecogênico de alta celularidade e com lesões papilares pedunculadas. A análise macro e microscópica da bexiga do ani-mal mostrou áreas com proliferação vascular na submucosa, associadas a infiltrado inflamatório mononuclear e proliferação do epitélio. Pode-se concluir que a ultrassonografia é uma excelente opção e, associada aos exames clínicos e laboratoriais de sangue e urina, possibilitou o diagnóstico e o prognóstico da doença.(AU)

Background: Equine colic syndrome comprises numerous conditions associated with abdominal pain in horses. Impaction, a common cause of this manifestation, is strongly related to these animals' diet. Highly fibrous diets such as sugarcane can predispose horses to colic. The clinical condition can be worsened by fermentative processes, which lead to dysbiosis, circulatory disorders and even endotoxemia. The aim of this study was to report 4 cases of colic syndrome among 8 horses that underwent an experiment to adapt them to a sugarcane-based diet, and to correlate the animals' clinical conditions to the forage they ingested. Cases: Eight male castrated Mangalarga Marchador horses, between 5.5 and 7 years old, were subjected to an experiment to test the feasibility of sugarcane as forage. Four of these horses were taken to the Large Animal Veterinary Hospital (HVGA) of the Federal Rural University of Rio de Janeiro for treatment of abdominal signs of discomfort a few days after the exclusive consumption of sugarcane, in a proportion of 1.75% of live weight in dry matter. The animals' symptoms ranged from behavioral signs indicative of pain to changes in vital parameters and structure of the feces, as well as changes revealed by transrectal palpation. Three of the 4 cases presented impaction in the small colon, and 1 of the horses also presented impaction in the right dorsal colon and rostral displacement of the pelvic flexure, with accumulation of contents in the right ventral colon and sternal flexure. Two of the cases were treated medically, while the other 2 required surgical intervention. The clinical condition of all the patients evolved favorably and they were discharged between 2 and 18 days. Discussion: Colic originating in the digestive system is a syndrome strongly associated with management, especially with respect to confinement, nutrition, and parasite control. During the experiment, 4 of the 8 horses fed with sugarcane presented with colic syndrome. The low quality of sugarcane fiber is due to the high degree of lignification of the plant cell wall, which favors accumulation of ingesta. The poor digestibility and sweet taste of this roughage favor increased consumption. Furthermore, its high sucrose content, associated with an increased rate of passage in the small intestine, alters the intestinal microbiome, and hence, the fermentation byproducts and pH of the ingesta. High intestinal content, allied to longer retention times in the colon and activation of the renin-angiotensin-aldosterone system, promote greater dryness of the ingesta, predisposing the occurrence of impactions in the most distal portion of the large intestine. Intestinal distension and mesenteric traction caused by the accumulation of contents and gases trigger pain, which can worsen due to displacement of the large colon. Small colon impaction, which is easily identified by transrectal palpation, evolves gradually and its treatment, both clinical and surgical, tends to have a favorable prognosis. The need for alternative food sources for horses is a growing demand; however, sugarcane as an exclusive roughage has been shown to be unsafe for horses. The low quality of the fiber and the high sucrose content of this forage can alter the digestive physiology of horses through changes in the passage rate, microbiome and motility of digesta, predisposing them to intestinal dysfunction, ingesta compaction and displacement of the large colon.

ABSTRACT Increasing interest in multiple strain Bacillus probiotics and parietal yeast fractions as feed ingredients for egg laying hen diets has also led to food safety questions. This study was undertaken to evaluate the ability of these products to reduce Salmonella Enteritidis colonization. Sixty Hy-Line hens aged 56 weeks were placed in individual cages and fed a mash diet containing one of the following treatments, control, Bacillus spp. probiotic, yeast cell wall, or a combination of yeast cell wall and Bacillus probiotic. At 60 weeks of age all hens were challenged orally with 7 x 107 CFU/bird of Salmonella Enteritidis. At 61 weeks of age, birds were humanely euthanized, by cervical dislocation and the ceca aseptically removed and cultured for S. Enteritidis prevalence and number by the Most Probable Number method. There was no significant difference in prevalence of Salmonella Enteritidis between the control and any treatments. The control birds had 4.37 log10 MPN/g of S. Enteritidis detected in the ceca. The Probiotic group had 2.96 MPN/g, a reduction of 1.41(p 0.05) and the yeast cell wall group had 2.89 MPN/g a reduction of 1.48 (p 0.05). The combination had 3.60 MPN/g a numerical reduction of 0.78 (p=0.14). The yeast cell wall and Bacillus probiotic groups significantly reduced the amount of Salmonella Enteritidis in the ceca of the laying hens.
