Streptomyces sampsonii is a kind of biocontrol bacterium with antifungal effects, and chitinase is one of the main antifungal substances. To improve and further study the structure and function of the chitinase gene of S. sampsonii, we amplified the target fragment by PCR, ligated the fragment to the expression vector pET-32a, introduced the resulting plasmid into Escherichia coli BL21 (DE3) and induced expression of the chitinase. Then, the recombinant chitinase was purified by is-labelled protein micro purification kit. A chitinase gene, Sschi61, was cloned from the genome and expressed in a prokaryote. The antifungal effect of the recombinant protein was also studied. Finally, the chitinase gene Sschi61 with a length of 1755 bp was obtained, and the expression of the 82 kDa recombinant chitinase was induced in E. coli by IPTG. The recombinant chitinase could inhibit the black spot pathogen of Eucommia ulmoides (Pestalotiopsis trachicarpicola). After the hyphae of the pathogen of black spot of Eucommia ulmoides (Pestalotiopsis trachicarpicola) were soaked with recombinant chitinase, the hyphae cells expanded, broke, and dissolved.
Streptomyces sampsonii é uma espécie de bactéria de biocontrole com efeitos antifúngicos, sendo a quitinase uma das principais substâncias desse tipo. Para melhorar e estudar mais a estrutura e função do gene da quitinase de S. sampsonii, amplificamos o fragmento alvo por PCR, ligamos o fragmento ao vetor de expressão pET-32a, introduzimos o plasmídeo resultante em Escherichia coli BL21 (DE3) e induzimos expressão da quitinase. Em seguida, a quitinase recombinante foi purificada pelo kit de micropurificação de proteína marcada. Um gene da quitinase, Sschi61, foi clonado do genoma e expresso em um procarioto. O efeito antifúngico da proteína recombinante também foi estudado. Finalmente, o gene da quitinase Sschi61 foi obtido, contando comprimento de 1755 pb, e a expressão da quitinase recombinante de 82 kDa foi induzida em E. coli por IPTG. A quitinase recombinante pode inibir o patógeno da mancha preta de Eucommia ulmoides (Pestalotiopsis trachicarpicola). Após as hifas do patógeno da mancha preta de Eucommia ulmoides (Pestalotiopsis trachicarpicola) serem embebidas com quitinase recombinante, as células das hifas se expandiram, quebraram e se dissolveram.

Biliary neoplasms are uncommon in cats and affect older animals. A 12-year-old female crossbreed cat showed prostration, lethargy, apathy, and severe jaundice. Ultrasonography showed distention of the gallbladder associated with severe obstruction of the bile ducts with thickening of the biliary wall, forming amorphous masses of irregular contour and heterogeneous appearance directed to the lumen measuring up to 2 cm. Necropsy showed a gallbladder with a yellowish and soft nodule measuring 3 × 3 cm, compressing the extrahepatic bile duct, occluding the passage of bile. There were also firm, yellowish multifocal to coalescing nodules in the liver, ranging from 0.5 to 1 cm, affecting 10% of the organ, in addition to lungs with firm, yellowish multifocal nodules ranging from 0.2 to 0.5 cm, affecting 20% of the organ. Histologically, gallbladder and bile ducts had malignant epithelial neoplastic proliferation, which was organized into multiple papillary and ductal projections, separated by moderate fibrovascular stroma compatible with gallbladder adenocarcinoma. The liver and lungs also contained neoplastic structures with a ductal appearance and papilliform projections identical to those observed in the gallbladder. The immunohistochemical examination (IHC) showed intense positive staining of epithelial neoplastic cells for pan-cytokeratin (AE1/AE3) and no staining for vimentin (Clone V9). The diagnosis of gallbladder adenocarcinoma with metastasis in the liver and lungs was established based on the clinical, macroscopic, histopathological, and immunohistochemical findings.(AU)

Purpose: To investigate the role and mechanism of ß1,3-N-acetylglucosaminyltransferase-3 gene (B3GNT3) in esophageal cancer (ESCA). Methods: The starBase database was used to evaluate the expression of B3GNT3. B3GNT3 function was measured using KYSE-30 and KYSE-410 cells of esophageal squamous cell carcinoma (ESCC) cell lines. The mRNA levels were detected by quantitative real-time polymerase chain reaction (qRT-PCR). Cell counting kit-8, clone formation assay and transwell assay were used to detect the changes of proliferation, invasion and migration. Results: B3GNT3 expression was higher in ESCA tissues than in normal tissues. The overall survival rate of ESCA patients with high B3GNT3 expression was lower than that of ESCA patients with low B3GNT3 expression. In vitro functional experiments showed that the proliferation ability, migration and invasion ability of KYSE-30 and KYSE-410 cells with B3GNT3 interference were lower than those of the control, and the overexpression of B3GNT3 had the opposite effect. After silencing B3GNT3 expression in ESCC cell lines, the growth of both cell lines was inhibited and the invasiveness was decreased. Knockdown of B3GNT3 reduced the growth rate and Ki-67 expression level. Conclusion: B3GNT3, as an oncogene, may promote the growth, invasion and migration of ESCC cell.

The cultivation of cocoa is of great socio-economic importance worldwide. Cocoa beans are the essential raw material for chocolate production. The variability of cacao studied presents only a small fraction of the existing genetic diversity, mainly in the Amazon region. Furthermore, just a small part of this variability has been exploited in cocoa breeding. Thus, the present study processed the genetic evaluation and selection of cocoa clones, based on morpho-agronomical traits. For this, we evaluated 145 clones, during 37 harvests from, four consecutive years. The following traits were evaluated: total number of fruits collected (TNFC), total number of healthy fruits (TNHF), weight of wet seeds from healthy fruits (WWSHF), average weight of wet seeds per healthy fruit (AWWSHF), weight of dry seeds from healthy fruit (WDSHF), average weight of dry seeds per healthy fruit (AWDSHF), percentage of fruits with witches' broom disease (PFWB), percentage of fruits with borer (PFBR), percentage of fruits with germinated seeds (PFGS), number of branches with witches' broom disease (NBWB), and number of inflorescence with witches' broom disease (NIWB). Significant differences (P < 0.05) among the clones were observed for all traits, which reveal an expressive variability and possibility of gains with selection. The highest significant correlations (P < 0.05) occurred between traits TNFC and TNHF (0.94), TNHF and AWWSHF (0.86), and TNHF and AWDSHF (0.86). Based on the selection index, the clones POUND 12 and CAB 12, 228, 253, 257, 258, and 422 were the most suitable for selection.
O cultivo do cacau tem grande importância socioeconômica mundial. Os amêndoas de cacau são a matéria-prima essencial para a produção de chocolate. A variabilidade do cacaueiro avaliada representa apenas uma pequena fração da diversidade genética existente, principalmente na região amazônica. Além disso, apenas uma pequena parte dessa variabilidade foi explorada no melhoramento de cacau. Dessa forma, o presente estudo teve como objetivo realizar a avaliação genética e seleção de clones de cacau, com base em características morfo-agronômicas. Para isso, foram avaliados 145 clones, durante 37 colheitas, em quatro anos consecutivos. As seguintes características de frutos e sementes foram avaliadas: número total de frutos colhidos (NTFC), número total de frutos sadios (NTFS), peso de sementes úmidas dos frutos sadios (PSUFS), peso médio de sementes úmidas dos frutos sadios (PMSUFS), peso de sementes secas dos frutos sadios (PSSFS), peso médio de sementes secas dos frutos sadios (PMSSFS), porcentagem de frutos com vassoura-de-bruxa (PFVB), porcentagem de frutos com broca (PFBR), porcentagem de frutos com sementes germinadas (PFSG), número de ramos com vassoura-de-bruxa (NRVB) e número de almofadas florais com vassoura-de-bruxa (NAFVB). Diferenças significativas (P < 0,05) entre os clones foram observadas para todas as características, o que revela uma expressiva variabilidade e possibilidade de ganhos com a seleção. As maiores correlações significativas (P < 0,05) ocorreram entre os caracteres NTFC e NTFS (0,94), NTFS e PMSUFS (0,86) e, NTFS e PMSSFS (0,86). Com base no índice de seleção, os clones POUND 12 e CAB 12, 228, 253, 257, 258 e 422 foram considerados os mais adequados para a seleção.

Date fruit is known to be the staple food in the Arab countries. It provides a lot of potential health benefits and can be the essential source of nutrients. The majority of Moroccan varieties are not characterized for their chemical, biochemical and quality properties. The aim of this work was to assess the chemical composition of 17 varieties of Moroccan date fruits (Phoenix dactylifera L.) and to determine their nutritive components. The analysis showed that the dates are rich in sugars (51.80-87.98%), they contain low concentration of proteins (1.09-2.80%) and lipids (0.16-0.39%). The predominant mineral is potassium (1055.26-1604.10 mg/100 g DW). Moreover, they contain high concentrations of malic acid (69.48-495.58 mg/100 g (DW)), oxalic acid (18.47-233.35 mg/100 g DW) and tartaric acid (115.70-484.168 mg/100 g DW). These results suggest that the date fruit are nutritious and can be an excellent source for human nutrition and health benefits.
A fruta da tâmara é conhecida por ser o alimento básico nos países árabes. Oferece muitos benefícios potenciais à saúde e pode ser a fonte essencial de nutrientes. A maioria das variedades marroquinas não se caracteriza por suas propriedades químicas, bioquímicas nem de qualidade. O objetivo deste trabalho foi avaliar a composição química de 17 variedades de frutos de tâmara marroquina (Phoenix dactylifera L.) e determinar seu valor nutritivo. A análise mostrou que as tâmaras são ricas em açúcares (51,80-87,98%) e contêm baixa concentração de proteínas (1,09-2,80%) e lipídios (0,16-0,39%). O mineral predominante é o potássio (1.055,26-1.604,10 mg/100 g DW). Além disso, contêm altas concentrações de ácido málico (69,48-495,58 mg/100 g DW), ácido oxálico (18,47-233,35 mg/100 g DW) e ácido tartárico (115,70-484,168 mg/100 g DW). Esses resultados sugerem que o fruto da tamareira é nutritivo e pode ser uma excelente fonte de nutrição humana e conferir benefícios à saúde.

Potato (Solanum tuberosum L.) is one of the main crops in the Andean region and due to environmental aspects, the use of biocontrol agents is considered a safe way to produce potato seed tubers.The objective of the study was to evaluate potato inoculation with Trichoderma sp. as a matrix and rhizobacteria Bacillus simplex and Azotobacter sp. on the growth of potato seedlings from in vitro culture, for the production of seed tubers in a greenhouse. The inoculation of microorganisms was carried out in pots, using five potato genotypes for processing. The inoculation treatments were: control, Trichoderma sp., Trichoderma sp. + Azotobacter sp., Trichoderma sp. + Bacillus simplex, Trichoderma sp. + B. simplex + Azotobacter sp. The potato genotypes were cv. Unica (CIP392797.22), cv. Bicentenaria, the advanced clones CIP 396311.1, CIP 399101.1, and the experimental clone UH-09 from the Universidad Nacional José Faustino Sánchez Carrión. A completely randomized design was used, under a factorial arrangement and comparisons between treatments were made at p<0.05. All inoculant treatments exceeded the control in number and weight of tubers per plant as well as in tuber size. Inoculations of Trichoderma sp. alone or with Azotobacter sp. increased plant height, number of leaves per plant and vegetative uniformity; inoculations with the Trichoderma sp. + B. simplex + Azotobacter sp. consortium, improved the dry weight of the foliage, number of shoots per plant and vegetative vigor. There were significant interactions between potato genotypes and inoculant treatments for plant uniformity, vegetative vigor, and the foliage's dry weight. Coinoculation with Trichoderma sp. and some bacterial strains promote the growth of in vitro potato seedlings, increasing the size and weight of the seed tubers and plant biomass, indicating an interrelation between fungi and bacteria that influence the production of potatoes in a greenhouse.(AU)
A batata (Solanum tuberosum L.) é uma das principais culturas da região andina e devido aos aspectos ambientais, o uso de agentes de biocontrole é considerado uma forma segura de produzir tubérculos de batata-semente. O objetivo do trabalho foi avaliar a inoculação de batata com Trichoderma sp. como matriz e rizobactérias Bacillus simplex e Azotobacter sp. sobre o crescimento de mudas de batata provenientes de cultivo in vitro, para produção de tubérculos-semente em casa de vegetação. A inoculação dos microrganismos foi realizada em vasos, utilizando-se cinco genótipos de batata para processamento. Os tratamentos de inoculação foram: testemunha, Trichoderma sp., Trichoderma sp. + Azotobacter sp., Trichoderma sp. + B.simplex, Trichoderma sp. + B.simplex + Azotobacter sp. Os genótipos de batata foram cv. Única (CIP392797.22), cv. Bicentenaria, os clones avançados CIP396311.1, CIP399101.1, e o clone experimental UH-9 da Universidade Nacional José Faustino Sánchez Carrión. O delineamento foi inteiramente casualizado, em esquema fatorial e as comparações entre os tratamentos foram feitas com p<0,05. Todos os tratamentos com inoculantes excederam o controle em número e peso de tubérculos por planta, bem como em tamanho de tubérculo. Inoculações de Trichoderma sp. sozinho ou com Azotobactersp. aumento da altura das plantas, número de folhas por planta e uniformidade vegetativa; inoculações com o Trichoderma sp. + B.simplex + Azotobacter sp. consorcio, melhorou a massa seca da folhagem, número de brotações por planta e vigor vegetativo. Houve interações significativas entre genótipos de batata e tratamentos inoculantes para uniformidade e vigor vegetativo, e para a massa seca da folhagem. Coinoculação com Trichoderma sp. e algumas cepas bacterianas promovem o crescimento de mudas de batata in vitro, aumentando o tamanho e o peso dos tubérculos-semente e da biomassa vegetal, indicando que existe inter-relação entre fungos e bactérias que influenciam a produção de batata em casa de vegetação.(AU)

O presente estudo teve como objetivo avaliar a qualidade microbiológica de sorvetes comercializados na cidade de Pouso Alegre MG. Foram adquiridas 2 amostras de sorvete de 18 sorveterias em um intervalo de 30 dias totalizando 36 amostras. As amostras de sorvetes foram submetidas às análises para enumeração de coliformes totais, coliformes termotolerantes, Estafilococos coagulase positiva e Salmonella spp. As análises foram realizadas segundo os protocolos da American Public Health Association descrita no Compendium of methods for the microbiological examination of foods. Neste estudo não foram encontradas espécies de Salmonella spp. entretanto, houve amostras contaminadas por E. coli e Estafilococos coagulase positiva.13,89% das amostras analisadas estavam fora dos padrões estabelecidos. Verificou-se ainda a presença de coliformes totais em 77,78% e de Staphylococcus sp. em 36,11% e, embora a legislação vigente não mencione limites de contaminação para estes microrganismos, pode-se dizer que a matéria-prima estava contaminada, que as condições higiênicas, no processamento e armazenamento estavam inadequados para este produto. As amostras analisadas obtidas de sorveterias da região central foram as mais contaminadas, onde foi encontrada a presença de E. coli e Estafilococos coagulase positiva. A presença desses microrganismos sugere falhas em algum momento do processamento, armazenamento ou da venda destes produtos tornando-os inadequados, podendo não só causar uma deterioração mais rápida do sorvete, mas também colocando em risco a saúde do consumidor.(AU)
This study aimed to evaluate the microbiological quality of ice cream sold in the city of Pouso Alegre MG. Two ice cream samples were acquired from 18 ice cream parlors in a 30-day interval, totaling 36 samples. The ice cream samples were subjected to analysis for enumeration of total coliforms, thermotolerant coliforms, coagulase positive Staphylococci and Salmonella spp. The analyzes were performed according to the protocols of the American Public Health Association described in the Compendium of Methods for the Microbiological Examination of Foods. In this study no species of Salmonella spp. were found, however, there were samples contaminated by E. coli and coagulase positive Staphylococci. 13.89% of the analyzed samples were outside the established standards. It was also verified the presence of total coliforms in 77.78% and Staphylococcus sp. in 36.11% and, although the current legislation does not mention contamination limits for these microorganisms, it can be said that the raw material was contaminated, that the hygiene, processing and storage conditions were inadequate for this product. The analyzed samples obtained from ice cream parlors in the central region were the most contaminated, where the presence of E. coli and coagulase positive Staphylococci was found. The presence of these microorganisms suggests failures at some point in the processing, storage or sale of these products, making them unsuitable and not only causing a faster deterioration of the ice cream, but also putting the consumer's health at risk.(AU)

A pesquisa teve o objetivo de avaliar as condições higiênicas e sanitárias de comercialização do queijo de coalho por vendedores ambulantes na praia de Copacabana, por meio de inspeção visual e análises microbiológicas. Para o estudo foram coletadas quinze amostras de cinco diferentes ambulantes e estas foram encaminhadas para o Centro Estadual de Pesquisa em Qualidade de Alimentos da Empresa de Pesquisa Agropecuária do Estado do Rio de Janeiro (PESAGRO-RIO/CEPQA) onde foram submetidas às análises microbiológicas. O resultado da avaliação visual das condições higiênico-sanitárias demonstrou 100% de não conformidades em relação à adoção das Boas Práticas de Fabricação por parte dos manipuladores. Os resultados das contagens de Estafilococos coagulase positiva e de Coliformes termotolerantes se apresentaram acima do limite preconizado pela legislação vigente em 75% e 6,7% das amostras respectivamente. Em 25% das amostras foi verificada a presença de Salmonella spp. O estudo demonstrou que o queijo de coalho vendido por ambulantes da praia de Copacabana estava impróprio para o consumo podendo representar risco à saúde dos consumidores.(AU)
The research aimed to evaluate the hygienic and sanitary commercialized conditions of coalho cheese by street vendors on Copacabana beach, through visual inspection and microbiological analysis. For conducting the study, fifteen samples were collected from five different street vendors and these were analyzed at the State Center for Research in Food Quality of the Agriculture Research Company of the State of Rio de Janeiro (PESAGRO-RIO/CEPQA). The result of the visual assessment of hygienic-sanitary conditions demonstrated 100% of non-conformities in relation to the adoption of Good Manufacturing Practices by the manipulators. In counts of coagulase positive Staphylococcus and Thermotolerant Coliforms were above the limit recommended by the current legislation in 75% and 6.7% of the samples, respectively. Salmonella spp. was present in 25% of the analyses.The study showed that the coalho cheese sold by street vendors on Copacabana beach was unfit for consumption and could pose a risk to consumer health.(AU)

Staphylococcus is one of the most frequent etiological agents of bovine mastitis, causing economic losses to milk farming. This study aimed to describe the milk production and management practices in 15 herds located in three microregions of the State of Rondônia and to identify the antimicrobial resistance profile of 97 isolates of the genus Staphylococcus from these herds. The isolates were subjected to antimicrobial susceptibility tests using the agar diffusion method. Twenty-nine S. aureus isolates were selected to determine their minimum inhibitory concentration (MIC). In the farms studied, the breeding system was semi-intensive, with the Girolando breed being predominant. Milking was performed predominantly using a mechanical milking system (86.7%), twice a day (66.7%), in the presence of the calf (53.8%). The average number of lactating cows in the farms was 24, with an average milk production of 204.9 L d-1 and a milk productivity of 10.2 L animal-1 d-1. The use of antimicrobials for the treatment and prevention of bovine mastitis was reported for all properties, and therapy for dry cows was adopted in 80% of the herds. The percent susceptibility to antimicrobials ranged from 85.5% to 100% for S. aureus, 22.2% to 88.9% for coagulase-positive staphylococci (CPS), and 69.2% to 100% for coagulase-negative staphylococci (CNS), with the highest resistance frequencies for penicillin, ampicillin, and tetracycline. Among the S. aureus isolates, 100% susceptibility to oxacillin, cefalexin, and gentamicin was observed, and among the CNS isolates, 100% susceptibility to gentamicin was observed. None of the antimicrobials tested showed 100% in vitro effectiveness for CPS. CPS and CNS presented lower percentages of susceptibility to penicillin, ampicillin, and tetracycline than the S. aureus isolates. Twelve resistance patterns were detected, six of which were multiresistance patterns. The most prevalent resistance patterns were penicillin and ampicillin (PEN-AMP) and penicillin, ampicillin, and tetracycline (PEN-AMP-TET). Results of the MIC assay revealed that all 29 S. aureus isolates were susceptible to cephalothin, cefoxitin, tetracycline, and erythromycin, whereas 25 (86.2%) were susceptible to penicillin.(AU)
O gênero Staphylococcus destaca-se como um dos agentes etiológicos mais frequentes da mastite bovina, causando prejuízos econômicos à pecuária de leite. Este estudo teve o objetivo de descrever a produção de leite e práticas de manejo de 15 rebanhos localizados em três microrregiões do Estado de Rondônia, e identificar o perfil de resistência aos antimicrobianos de 97 isolados do gênero Staphylococcus provenientes destes rebanhos. Os isolados foram submetidos a testes de suscetibilidade a antimicrobianos, utilizandose a técnica de difusão em ágar. Foram selecionados 29 isolados de Staphylococcus aureus (S. aureus) para determinação da concentração inibitória mínima (CIM). Nas propriedades estudadas, o sistema de criação era semi-intensivo, com predomínio da raça Girolando. A ordenha era realizada predominantemente em sistema de ordenha mecânica (86,7%), duas vezes ao dia (66,7%) e com a presença do bezerro (53,8%). A média do número de vacas em lactação das propriedades era de 24 animais, com média de produção de leite de 204,9 L d-1 e produtividade de leite de 10,2 L animal-1 d-1. O uso de antimicrobianos para o tratamento e prevenção da mastite bovina foi relatado em todas as propriedades, sendo adotada a terapia da vaca seca em 80% dos rebanhos. A percentagem de susceptibilidade aos antimicrobianos variou de 85,5 a 100% para S. aureus, 22,2 a 88,9% para Staphylococcus coagulase positivos (SCP) e 69,2 a 100% para Staphylococcus coagulase negativos (SCN), sendo as maiores frequências de resistência para penicilina, ampicilina e tetraciclina. Entre os isolados de S. aureus foi observada 100% de susceptibilidade aos antimicrobianos oxacilina, cefalexina e gentamicina, e entre os isolados de SCN, 100% de susceptibilidade à gentamicina. Nenhum dos antimicrobianos testados apresentou 100% de efetividade in vitro para SCP. SCP e SCN apresentaram menores percentagens de susceptibilidade à penicilina, ampicilina e tetraciclina do que os isolados de S. aureus. Foram observados 12 padrões de resistência, sendo seis padrões de multirresistência. Os padrões de resistência mais prevalentes foram resistência à penicilina e ampicilina (PEN-AMP) e à penicilina, ampicilina e tetraciclina (PEN-AMP-TET). A determinação da CIM dos 29 isolados de S. aureus mostraram que todos foram susceptíveis à cefalotina, cefoxitina, tetraciclina e eritromicina, e 25 (86,2%) foram susceptíveis à penicilina.(AU)

Background: Acute myeloid leukemia (AML) is a group of non-lymphoid hematological tumors characterized by aberrant proliferation and/or decreased apoptosis of a clone of non-mature cells, resulting in the accumulation of immature blast cells in the bone marrow and peripheral blood. It is considered rare, as it represents 10% of neoplasms of hematopoietic origin. However, it is known that felines seroreactive for FIV and FeLV are more predisposed and reports of this type of leukemia in cats in the literature are scarce. Thus, the objective of this study was to evaluate the blood and bone marrow of a cat seroreactive for FeLV that presented with myelodysplastic syndrome that progressed to acute myeloid leukemia. Case: A 6-year-old male mixed-breed cat, neutered, seroreactive for FeLV, showed apathy, weight loss, and pale mucous membranes. Initial peripheral blood smear evaluation revealed hypochromic normocytic anemia, leukopenia, neutropenia, lymphopenia, and thrombocytosis with many macropackets and giant platelets. Based on this blood picture, a long-spectrum antimicrobial therapy with amoxicillin and clavulanate [Clavulin® BD - 25 mg/kg, every 12 h] was started. Granulocyte colony stimulating factor used filgrastim (rHu G-CSF) [Fiprina® - 5 µg/kg, SC, every 48 h] and appetite stimulant mirtazapine [Mirtz® - 2 mg/cat, orally, every 48 h] were used to correct leukopenia and nutritional status, respectively. Follow-up blood smear evaluation on the 30th day showed persistence of the hematological changes noticed earlier. A bone marrow puncture was performed, and immunosuppressive therapy with prednisolone [Predsim® - 4 mg/kg, orally, every 24 h] was initiated. The aspirated material showed increased cellularity for age, decreased myeloid:erythroid ratio, and 39.8% of blasts of myeloid origin. An average of 17.7 megakaryocytes were observed per field (10x magnification). Bone marrow cytological evaluation suggested acute myeloid leukemia with dysmegakaryocytopoiesis. After the diagnosis, the examinations were repeated monthly, and there was still intense leukopenia. However, in view of the stable clinical status and leukopenia with neutropenia, treatment for leukemia was not instituted and only supportive treatment was administered when necessary. Eight months after the diagnosis, clinical status had worsened, and unlike the earlier hemograms, global leukocyte count had increased with predominant lymphocytosis (95% of the total leukocytes) with atypical lymphocytes. The cat died a few days later. Discussion: Bone marrow evaluation is indicated when peripheral blood cell abnormalities are present and cannot be explained in the context of the clinical history. In the present report, the bone marrow aspirate was hypercellular (cellularity above 75%); however, intense leukopenia was observed in the peripheral blood. In myelodysplastic syndromes, it is common for the bone marrow to be normal to hypercellular, which occurs when there is a greater production of myeloid or erythroid cell lines in response to the loss, destruction, or consumption of cells. Despite this, cytopenias may be present in the peripheral blood, since the defective cells undergo apoptosis and die before being released into the circulation, characterizing inefficient hematopoiesis. The diagnosis of acute leukemia comprises a variety of hematopoietic neoplasms that are complex and unique. Each acute leukemia subtype has defining characteristics that affect the prognosis and treatment of each animal.

For chipping, in the subtropical region, no clone had adequate quality, but in tropical F177-11-20 presented similar processing traits and higher or similar yield to 'Atlantic' in both systems, and also ORG7785 in organic. Comparing with Asterix, for French fry, in the subtropical region, superior or similar to the standard this cultiva rwere F129-10-06 and F163-11-01; while in the tropical region and conventional system, were the clones ORG7785, F129-10-06, F163-11-01, F177-11-20, ORG6464, ORG 6377, F05-11-03, Odone 80-02 and F174-11-01; and in organic system, the best clones were F54-11-03, ORG7785, F129-10-06, F163-11-01, F177-11-20 and ORG6464.(AU)
Neste trabalho foi avaliado o desempenho de clones de batata para caracteres agronômicos e de qualidade de processamento, em regiões subtropical e tropical de altitude, sob sistema convencional e orgânico de produção. Os experimentos foram conduzidos na primavera de 2017 e 2018 em Canoinhas-SC (subtropical), sobre sistema convencional, e nos invernos de 2018 e 2019 em Brasília-DF (tropical), Brasil, sob sistemas convencional e orgânico. Isso porque maior rendimento e maiores tamanhos de tubérculos foram associados com ciclo vegetativo mais longo, maior peso específico de tubérculos, porém cor mais escura de fritura. Os melhores clones diferiram com a região e sistemas de produção. Para chips, n aregião subtropical, nenhum clone apresentou bom desempenho, porém no ecossistema tropical, F177-11-20 apresentou similares qualidades industriais e maior ou superior produtividade em comparação com 'Atlantic' em ambos os sistemas de cultivo, também o clone ORG7785 no sistema orgânico. This paper aimed to evaluate crop yield and physical and chemical characteristics of acerola (Malpighia emarginata Sessé & Mociño ex DC) clones grown in the Alta Paulista region, Brazil. The evaluation comprised 7 clones, namely: Olivier, BRS 238-Frutacor, BRS 236-Cereja, BRS 235-Apodi, BRS 237-Roxinha, Okinawa, and Waldy-CATI 30, studiedfrom October 2018 to April 2019. Crop yield was analyzed based on the accumulated production of 12 harvests, with fresh mass expressed in kg fruit·plant-1. Considering the capacity of30 harvests per plant within the complete cropping period, total fruit mass (in kg fruit·plant-1) and crop yield (in tha-1) were estimated. Samples were collected from 4 of the 12 harvests in order to determine average fruit mass (g); average fruit size, fruit height/diameter ratio, and pulp yield (%). Regarding fruit pulp, the analysis consisted of quantifying the soluble solids, expressed in degrees Brix, and the technological index (i.e. pulp yield x °Brix/100). The experiment was conducted in a randomized block design, with 7 treatments and 4 repetitions. The obtained data were statistically processed by analysis of variance using the F test, and the means were compared using the Tukeystest at the 0.05 significance level. The study concluded that, given the soil and weather conditions of the Alta Paulista region, Olivier, Apodi and Frutacor are the clones mainly recommended for the agro-industrial market, being the Olivier cultivar the first option among the three and appearing in a greater proportion in the area of the commercial orchard studied. Regarding the acerola production for fresh consumption, Olivier and Roxinha are the most suitable clones. Olivier stands out for its good fruit yield, adequate physical and chemical characteristics, and easy manual harvesting.

Na polpa foram quantificados os sólidos solúveis expressos em ° Brix e o índice tecnológico (rendimento de polpa x °Brix/100). Como delineamento experimental utilizou-se blocos ao acaso, com 7 tratamentos e 4 repetições. Os dados foram submetidos à análise de variância para o Teste F e as médias comparadas pelo Teste Tukey ao nível de 5% de significância. Concluiu-se que nas condições de clima e solo da região Alta Paulista recomenda-se o plantio dos clones Olivier, Apodi e Frutacor principalmente visando o mercado agroindustrial, sendo o clone Olivier a primeira opção e em maior proporção na área de implantação do pomar comercial. No cultivo de acerola voltado para consumo in natura indicam-se clones Olivier e Roxinha. O clone Olivier se destaca pela boa produtividade, características físico-químicas adequadas e facilidade de colheita manual dos frutos.

Sweet potato (Ipomoea batatasL.) is a crop with high market demands because of its well-known health benefits. Limited use of improved clones reduces the crops income potential. Plant breeding programs seek to selectgenotypes with good results in a variety of environments by evaluating the interaction effects of genotypes and environments. This study aimed to assess Clone × Growing season effects on morphological and productive parameters of sweet potato clones grown in the municipality of Curitibanos, Santa Catarina, Brazil, in 2016/2017 and 2017/2018. The following clones were evaluated: Brazlândia Rosada, BRS Amélia, Beauregard, BrazlândiaRoxa, and BRS Rubissol. A randomized block design with three replications was used to investigate root length, root diameter, mean root weight, total root number, number of marketable roots, total yield, and marketable yield. Data were subjected to one-way and two-way analysis of variance. Beauregard afforded the largest number of marketable roots and was stable in both growing seasons. Beauregard, Brazlândia Roxa, and Brazlândia Rosada differed in total number of roots between growing seasons. There were no significant differences between growing seasons in mean root weight; BRS Amélia afforded a higher mean root weight than Brazlândia Rosada. Beauregard did not differ in total or marketable yield between growing seasons and provided higher yields than the other clones. BRS Rubissol and Beauregard were found to be the most promising for sweet potato cultivation under the environmental conditions of Curitibanos.
A batata-doce (Ipomoea batatas L.) é uma cultura de alta demanda pelomercado consumidor, devido aos estudos que comprovamos seus benefícios à saúde. Contudo, o baixo uso de clones melhorados reduz a lucratividade potencial da cultura. O melhoramento de plantas busca selecionar genótipos com bons resultados em uma gama de ambientes atravésda avaliação da presença de interação genótipos x ambientes. A partir disso, objetivou-se avaliar a presença de interação clones x safras em caracteres morfológicos e produtivos de clones de batata-doce, no município de Curitibanos-SC, nas safras de 2016/2017 e 2017/2018. Os clones avaliados foram Brazlândia Rosada, BRS Amélia, Beauregard, Brazlândia Roxa e BRS Rubissol. O delineamento experimental utilizado foi de blocos casualizados com três repetições. Foram realizadas as avaliações dasraízes tuberosas: comprimento (CR), diâmetro (DR), massa média (MMR), número total (NTR), número de raízes comerciais (NRC), produção total (PT) e produção comercial (PC). Os dados foram submetidos à análise de variância individual e conjunta. O clone Beauregard se destacou por apresentar o maior número de raízes comerciais e por ser estável em diferentes ambientes. Beauregard, Brazlândia Roxa e Brazlândia Rosada apresentaram diferença entre safras para o número total de raízes. Não houve diferenças significativas entre safras para a massa média de raízes, notando a superioridade de BRS Amélia sobre Brazlândia Rosada.O clone Beauregard não apresentou comportamento diferencial entre safras tanto para a produtividade comercial e total, e foi destaque em comparação com os outros clones. Os clones BRS Rubissol e o Beauregard foram considerados os mais promissores para o cultivo de batata-doce nas condições de Curitibanos/SC.

Background: Extraskeletal osteosarcoma (EOS), a rare variant of osteosarcoma (OS), is a malignant neoplasm that develops insoft tissues without primary bone involvement. This study aims to describe a case of EOS with a mediastinal location in a canine.Case: A 10-year-old male Uruguayan Cimarron dog, was presented to the Laboratório Regional de Diagnóstico, Faculdadede Veterinária, Universidade Federal de Pelotas (LRD/FV/UFPel) for necropsy. The dog had a history of submandibularswelling, progressive hind limb paralysis, muscle atrophy, and breathing difficulties. During necropsy, in the thoracic cavity,approximately 900 mL of serosanguinous exudate and a reddish-brown, bossed mediastinal mass measuring 15.0 cm in thelongest axis were also noted. The lung exhibited multifocal to coalescent, white, firm nodules extending from the pleurato the parenchyma and measuring up to 4.5 cm in diameter. In the parietal and occipital region of the brain, a matte winemass measuring 2.3 cm in the longest axis was observed. Fragments of the neoplastic mass, organs of the abdominal andthoracic cavities, and the brain were harvested and fixed in 10% buffered formalin. After 48 h, the samples were routinelyprocessed, incorporated in paraffin, cut into 3 µm-thick sections, and stained using Hematoxylin and Eosin (HE). Selectedsections of the neoplasm, the lung, and the brain were subjected to Von Kossa staining and immunohistochemical (IHC)analysis. Background: Extraskeletal osteosarcoma (EOS), a rare variant of osteosarcoma (OS), is a malignant neoplasm that develops insoft tissues without primary bone involvement. This study aims to describe a case of EOS with a mediastinal location in a canine.Case: A 10-year-old male Uruguayan Cimarron dog, was presented to the Laboratório Regional de Diagnóstico, Faculdadede Veterinária, Universidade Federal de Pelotas (LRD/FV/UFPel) for necropsy. The dog had a history of submandibularswelling, progressive hind limb paralysis, muscle atrophy, and breathing difficulties. During necropsy, in the thoracic cavity,approximately 900 mL of serosanguinous exudate and a reddish-brown, bossed mediastinal mass measuring 15.0 cm in thelongest axis were also noted. The lung exhibited multifocal to coalescent, white, firm nodules extending from the pleurato the parenchyma and measuring up to 4.5 cm in diameter. In the parietal and occipital region of the brain, a matte winemass measuring 2.3 cm in the longest axis was observed. Fragments of the neoplastic mass, organs of the abdominal andthoracic cavities, and the brain were harvested and fixed in 10% buffered formalin. After 48 h, the samples were routinelyprocessed, incorporated in paraffin, cut into 3 µm-thick sections, and stained using Hematoxylin and Eosin (HE). Selectedsections of the neoplasm, the lung, and the brain were subjected to Von Kossa staining and immunohistochemical (IHC)analysis. The biotrophic fungus Hemileia vastatrix causes coffee leaf rust (CLR), one of the most devastating diseases in Coffea arabica. Coffee, like other plants, has developed effective mechanisms to recognize and respond to infections caused by pathogens. Plant resistance gene analogs (RGAs) have been identified in certain plants as candidates for resistance (R) genes or membrane receptors that activate the R genes. The RGAs identified in different plants possess conserved domains that play specific roles in the fight against pathogens. Despite the importance of RGAs, in coffee plants these genes and other molecular mechanisms of disease resistance are still unknown. This study aimed to sequence and characterize candidate genes from coffee plants with the potential for involvement in resistance to H. vastatrix . Sequencing was performed based on a library of bacterial artificial chromosomes (BAC) of the coffee clone Híbrido de Timor (HdT) CIFC 832/2 and screened using a functional marker. Two RGAs, HdT_LRR_RLK1 and HdT_LRR_RLK2, containing the motif of leucine-rich repeat-like kinase (LRR-RLK) were identified. Based on the presence or absence of the HdT_LRR_RLK2 RGA in a number of differential coffee clones containing different combinations of the rust resistance gene, these RGAs did not correspond to any resistance gene already characterized (SH1-9). These genes were also analyzed using qPCR and demonstrated a major expression peak at 24 h after inoculation in both the compatible and incompatible interactions between coffee and H. vastatrix . These results are valuable information for breeding programs aimed at developing CLR-resistant cultivars, in addition to enabling a better understanding of the interactions between coffee and H. vastatrix .(AU)
