Resumo

Background: In several countries, including Brazil, the livestock industry plays a key role in the country's economy. Brazil has the second largest bovine herd in the world and the biggest commercial herd. Ticks are an ongoing problem for both large operation cattle producers and small family farmers. Rhipicephalus microplus causes expressive losses in cattle breeding, since it occurs in important beef production zones like South America, Africa, and Oceania. Some of the negative consequences of tick infestation to cattle breeding are anemia, loss in milk and beef production, and transmission of Babesia bovis and B. bigemina. Significant losses are caused by the cattle tick (R. microplus) in several regions of the world, costing around US$ 3.3 billion per year to the Brazilian livestock industry alone. The tick control methods are mainly based on synthetic acaricides. However, the improvement of current tick control requires the identification of new molecular targets in tick physiology and development of molecule compounds to target important physiology pathways. The strategies proposed to address this issue are expand the knowledge about the molecules involved in the detoxification of chemicals to enhance the efficacy of the acaricides as well as to develop new compounds for chemical control. Review: Tick control is currently based on chemical acaricides; however, effective control and prevention of tick infestation remain distant goals. In recent decades, a progressive decrease in the efficiency of acaricides due to drug resistance has been observed. Acaricide resistance is an evolutionary adaptation, which implies the existence of behavioral and physiological mechanisms that allow the survival of resistant individuals. Four resistance mechanisms are described: behavioral resistance, reduced drug penetration, target site insensitivity and increased drug detoxification. Augmented drug detoxification may be due to increased activity of enzymes or transporters due to increased gene expression or mutations in some genes. Research focus on mechanisms of acaricide resistance in ticks characterized detoxification pathways based on (1) increased activity of enzymes (cytochrome p450, esterase and GST) which play a role in biochemically altering acaricides towards decreased toxicity and, (2) enhanced excretion of the modified less toxic compounds. To bypass the current problems, a better understanding of the biology, physiology, and molecular biology of the mechanisms of resistance to acaricides is fundamental to prolong their efficiency in controlling ticks. Moreover, identifying the genes and proteins associated with resistance can support in the development of more sensitive diagnostic methods to identify acaricide resistance, as well as improving control strategies. Discussion: In the last years, many researchers have been studying resistance mechanisms and important advances have been made which showed that, in several tick species, ABC transporters, esterases, P-450 cytochromes and glutathione-S-transferases participate in acaricide resistance. The characterization of the alterations in the targets in tick physiology and identification of new drugs with potential to tick control are crucial goals to increase tick control

Xilooligossacarídeos (XOS) são reconhecidos pelo seu potencial prebiótico relevante para diversos setores industriais e foram obtidos após o pré-tratamento hidrotérmico da biomassa lignocelulósica residual de galhos de eucalipto. Subprodutos inibitórios são gerados durante o processo de solubilização dos oligossacarídeos e acabam comprometendo a utilização do licor em microrganismos. Neste trabalho, o processo de destoxificação, hidrólise enzimática e atividade estimulantes de crescimento da bactéria Staphylococcus xylosus foram estabelecidos. Os resultados mostraram que a adsorção com carvão ativado em pó removeu cerca de 55% do ácido acético e mais de 90% do ácido fórmico, compostos fenólicos, lignina solúvel, furfural e 5-hidroximetilfurfural, e que a soma dos oligossacarídeos xilobiose (X2) e xilotriose (X3) foram maximizadas de 0,57 g/L para 1,21 g/L com 110 U/gXOS da enzima endoxilanase e 6,3% do licor destoxificado na hidrólise enzimática. O consumo de cerca de 63% de X2 e de 46% de X3 pela bactéria em meio basal deficiente em fontes de carbono, mas acrescido com os oligômeros, proporcionou maior crescimento celular em relação aos meios basais com alta composição de carbono, com e sem XOS, revelando seu potencial prebiótico pelo efeito estimulante de crescimento. Xylooligosaccharides (XOS) are recognized for their prebiotic potential relevant to several industrial sectors and were obtained after hydrothermal pretreatment of residual lignocellulosic biomass from eucalyptus branches. Inhibitory by-products are generated during the solubilization process of oligosaccharides and end up compromising the utilization of the liquor in microorganisms. In this work, the detoxification process, enzymatic hydrolysis and growth stimulating activity of Staphylococcus xylosus bacteria were established. The results showed that adsorption with powdered activated carbon removed about 55% of acetic acid and more than 90% of formic acid, phenolic compounds, soluble lignin, furfural, and 5-hydroxymethyl furfural and the sum of the oligosaccharides xylobiose (X2) and xylotriose (X3) were maximized from 0.57 g/L to 1.21 g/L with 110 U/gXOS of the enzyme endoxylanase and 6.3% of the detoxified liquor in the enzymatic hydrolysis. The consumption of X2 and X3 were about 63% and 46%, respectively, by the bacteria in basal medium deficient in carbon sources, but in medium added with the oligomers, provided higher cell growth compared to basal medium with high carbon composition, with and without XOS, revealing its prebiotic potential by its growth-stimulating effect. In this study, the possibility of increasing fermentation efficiency of Saccharomyces cerevisiae on sugarcane bagasse (a type of lignocellulosic waste) was analyzed. Sugarcane bagasse was subjected to hydrothermal and acidic pre-treatment. Next, the enzymatic hydrolysis of raw biomass and each pre treated biomass was performed using CellicCtec® enzymatic complex to obtain sugarcane hydrolysate, hydrothermal hydrolysate and acidic hydrolysate. Next, these were fermented by S. cerevisiae to check if the by-products of enzymatic hydrolysis, furfural and acetic acid had an inhibitory effect on fermentation efficiency. Next, each pre-treated biomass was subjected to detoxification involving activated charcoal. Each detoxified biomass was tested for fermentation efficiency. The lignocellulosic composition for sugarcane hydrolysate, hydrothermal hydrolysate and acidic hydrolysate, varied significantly, and were found to be, for cellulose 36.7%, 27.7% and 63.7% respectively; for hemicellulose 22.2%, 4.4% and 12% respectively; and for lignin 21.2%, 27.7% and 28.7% respectively. The presence of furfural and acetic acid had a strong influence on the fermentation efficiency of S. cerevisiae, and affected the consumption of sugars in each biomass by more than 90%. Further, we found that the detoxification process increased fermentation efficiency by 12.7% for the hydrothermal hydrolysate while for the acidic hydrolysate no significant difference was observed. This study showed that fermentation with greater efficiency is viable through the combined use of hydrothermal pre-treatment and detoxification. This combination of methods also causes less pollution as compared with the method involving acid pre-treatment due to the reduced number of effluents produced.(AU)


Nesse trabalho avaliou-se a possibilidade de se aumentar a eficiência de fermentação de um hidrolisado de bagaço de cana submetido aos pré-tratamentos hidrotérmico (195 ºC, usando 200 rpm por 10 min) e ácido (0,5% (v/v) de ácido sulfúrico a 121ºC por 15 min) (carga de sólidos de 10% m/v). A hidrólise enzimática do material pré-tratado foi realizada utilizado o complexo enzimático CellicCtec® (60 FPU/gbiomassa seca, tampão citrato a 50 mM e pH 4,8) a 50ºC usando 150 rpm por 72h. Antes do processo de detoxificação, realizou-se um teste com a espécie de Saccharomyces cerevisiae para verificar se os compostos furfural (1 e 4g.L-1) e ácido acético (1 e 5% v/v) exerciam significativa inibição na espécie testada. O processo de detoxificação avaliou a concentração de carvão ativado (1, 3 e 5% m/v) e o tempo do processo (30, 45 e 60 min) a 30 ºC, 150 rpm por 24 h. A composição lignocelulosica da biomassa in natura e pré-tratada (hidrotérmico e ácido) foi para celulose (36,7, 27,7 e 63,7%), hemicelulose (22,2, 4,4 e 12%) e lignina (21,2, 27,7 e 28,7%), respectivamente e com rendimento mássico em torno de 60%. A presença de furfural e ácido acético exibiu forte influência na espécie considerada, chegando a prejudicar em mais de 90% o consumo de açúcares no meio. O processo de destoxificação aumentou 13% a eficiência de fermentação para o hidrolisado obtido hidrotermicamente, enquanto que para o ácido não houve diferença significativa. Obtendo assim uma fermentação com maior eficiência, tecnicamente viável e menos poluente.(AU)

This study was planned to examine the effect of silymarin on diminishing adverse effects of ochratoxin-A (OTA) in laying hens. A total number of 300 Inshas, local layer hens of 28 weeks of age were randomly distributed into 4 groups with 5 replicates each (15 hens). The birds were fed on the following treatments, (Control): fed a hen diet without any supplementation; (SL): fed the control diet supplemented with SL (1000 mg/kg feed); (OTA-diet): was fed the control diet contaminated with 1 ppm of OTA/kg diet, and (OTA+SL): fed the OTA-diet plus SL (1ppm OTA+1000 mg SL/kg feed). Results showed that feeding OTA at 1 ppm reduced productive performance compared with those fed the control diet. In OTA treated groups the total erythrocytes count, leukocytes count, PCV and Hb were decreased when compared to the control and SL groups. Albumin, globulin and serum total proteins in OTA treated groups were significantly lower when compared to the control and SL groups. Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were significantly increased in OTA fed groups in comparison with the control and SL groups. Creatinine and uric acid were increased in OTA treated groups but were almost normal in the SL group. The results showed that OTA had a severe effect on liver and kidney, but SL treated group had normal liver and kidneys showing its hepatoprotective effects.(AU)

The insecticidal effect of the essential oil of Piper aduncum (EOPA), and of its constituent dillapiole [1-allyl-2,3-dimethoxy-4,5- (methylenedioxy) benzene] in particular, is well documented in the literature and can be associated with its interference with the enzymatic detoxification in arthropods. However, no data exist on the range of dillapiole content associated with insecticidal activity, which is necessary to establish reliable dose-activity parameters for a formulated product. The oil composition can also change during storage after distillation, mainly due to environmental factors such as light incidence, atmospheric oxygen and temperature, which can be deleterious to oil quality. In this study, EOPA subjected to different storage conditions over four years and its rectified fractions were submitted to bioassays to evaluate their insecticidal effect by topical contact and residual contact against Spodoptera frugiperda. Our objectives were to determine the relationship between dillapiole content and the insecticidal activity of EOPA, and to evaluate its chemical and toxicological properties over time under different conditions. Our results showed that EOPA was stable with respect to the dillapiole content and the toxicological effect against S. frugiperda under different storage conditions for four years. The overall chemical composition of the EOPA did not vary significantly among storage conditions. EOPA with dillapiole content ranging between 68% and 100% showed greater insecticidal toxicity by residual and topical contact against S. frugiperda larvae.(AU)


O efeito inseticida do óleo essencial de Piper aduncum (OEPA) e, particularmente, de seu constituinte dilapiol [1-alil-2,3- dimetoxi-4,5-(metilenodioxi) benzeno], está bem documentado na literatura e pode estar associado à sua interferência na desintoxicação enzimática em artrópodes. No entanto, não existem dados sobre a amplitude de teores de dilapiol associados à atividade inseticida, o que é necessário para estabelecer parâmetros de dose-atividade confiáveis para um produto formulado. A composição do óleo também pode sofrer alterações durante seu armazenamento após a destilação, principalmente devido a fatores ambientais como incidência de luz, oxigênio atmosférico e temperatura, que podem ser deletérios à qualidade do óleo. Neste estudo, durante quatro anos, OEPA submetido a diferentes condições de armazenamento e suas frações retificadas foram submetidos a bioensaios para avaliar seu efeito inseticida por contato tópico e contato residual contra Spodoptera frugiperda. Nossos objetivos foram determinar a relação entre o teor de dilapiol e a atividade inseticida do OEPA, e avaliar suas propriedades químicas e toxicológicas ao longo do tempo sob diferentes condições. Nossos resultados mostraram que o OEPA foi estável em relação ao teor de dilapiol e o efeito toxicológico contra S. frugiperda sob diferentes condições de armazenamento durante quatro anos. A composição química do OEPA não variou significativamente entre as condições de armazenamento. OEPA com teor de dilapiol entre 68% e 100% apresentou maior toxicidade inseticida por contato residual e tópico contra larvas de S. frugiperda.(AU)

Aristolochia plants are notable from an ethnopharmacological viewpoint, but the relevance of these species for medicinal purposes has been debated because of their inherent toxicity. The convergence of these contrasting realities can be readily achieved using bioconversion methods, which have been shown to be useful tools for numerous applications, including the detoxification of biomass. In this context, methanolic extracts of leaves from Aristolochia triangularis and Aristolochia gibertii, as well as the feces of Battus polydamas larvae fed with leaves from these plants, were prepared, and their cytotoxic activities were evaluated on a human fibroblast cell line (GM07492). The leaf extracts were found to be cytotoxic, leading to reductions of 42.1 and 33.8% on cell viability, respectively, while the fecal extracts were considered inactive. In addition to evidencing the cytotoxicity of A. triangularis and A. gibertii, these findings demonstrated a potential bioconversion strategy for obtaining aristolochiaceous extracts with reduced toxicity using the larvae of a specialist phytophagous insect, thus renewing expectations in relation to the pharmacological importance of Aristolochia spp. The results were also ecologically relevant, as B. polydamas larvae were found to be able to detoxify compounds from host plants.(AU)

Herbicides play an important role in preventing crop yield losses due to both their weed interference ability and their capacity for increasing soil conservation in no-till systems. Group A herbicides or acetyl-CoA carboxylase (ACCase) are essential tools the selective management of glyphosate resistance in grass weed species. In this review, we describe important aspects of ACCase biology and herbicides targeting this enzyme, along with a discussion on stewardship programs to delay the evolution of herbicide resistance which can evolve either through target site and/or non-target site mechanisms. Sixteen-point mutations have been reported to confer resistance to ACCase inhibitors. Each mutation confers cross resistance to a different group of herbicides. Metabolic resistance can result in resistance to multiple herbicides with different mechanisms of action (MoA), and herbicide detoxification is often conferred by cytochrome P450 monooxigenases and glutathione- S -transferases. Regardless of whether resistance mechanisms are target or non-target site, using herbicides with the same MoA will result in resistance evolution. Therefore, while field surveys and resistance mechanism studies are crucial for designing reactive management strategies, integrated weed management plays a central role in both reactive and proactive mitigation of herbicide resistance evolution.(AU)

Resumo

Bortezomib, an inhibitor of 26S proteasome, is an anti-cancer therapeutic agent used in different cancer types. It leads to the arrest of the cancerous cell cycle by inhibiting angiogenesis and inducing apoptosis. Liver is the vital organ for detoxification and excretion of toxic products. The treatment with chemotherapy is a challenge, drugs are used to destroy cancer cells, but healthy cells can be affected during cancer treatment as well. The main objective of this study was to analyze the histopathological and biochemical effects of bortezomib on liver. Twenty-four female C57BL/6 mice were distributed into 4 groups, bortezomib injected treatment groups (Btz1, Btz2) and saline injected control groups (C1, C2). Bortezomib and saline were treated twice per week for 6 weeks and sacrificed at the end of one day (Btz1, C1) and 4 weeks (Btz2, C2) after the last injection. Liver samples were examined for histopathological analysis and the serum samples processed for biochemical analysis. Tissue samples were fixed, routinely processed, sectioned, and stained with Hematoxylin and Eosin (H&E). Periodic Acid-Schiff (PAS), Sudan Black staining and Masson's trichrome histochemical staining methods were performed to characterize the lesions. Histopathological analysis of the Btz1 and Btz2 groups revealed acute hepatic morphological changes such as hepatocellular swelling (cloudy swelling), necro-inflammatory reaction, and increased mononuclear polyploidy. Based on the negative staining with PAS and Sudan Black staining, hepatocellular swelling was diagnosed as hydropic degeneration. Necro-inflammatory reaction observed in the form of acute hepatitis was composed of mainly mononuclear cell infiltration accompanied by multifocal necrotic foci. Kupffer cell proliferation was observed in parallel with degenerative and necrotic changes. An Increase in hepatocellular mononuclear polyploidy visualized as hepatocytes with a single enlarged nucleus was detected in all liver sections of Btz1 and Btz2 groups. Individual cases of cholestasis (n = 1) and mild hepatic fibrosis (n = 1) were also reported. Significant elevated levels of alanine aminotransferase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP) and gamma-glutamyl transferase (GGT) were detected in bortezomib treated groups. Few clinical cases reported liver injury related to bortezomib used for cancer treatment. However, the liver was not considered as a target for bortezomib treatment. Our data suggesting that bortezomib caused liver damage and induces elevations in serum levels. The reported hepatic lesions including hepatocellular swelling, acute hepatitis and mononuclear polyploidy were mainly mild and moderate in severity. The increase of polyploidy in liver tissue of mice treated with bortezomib in this study was explained as a reaction of the liver facing the drug-induced hepatic damage. The mechanism leading to the hepatotoxicity of bortezomib treatment is not known but the production of a toxic metabolite through its metabolism in the liver can be suggested. Moreover, no recovery was also observed in histopathological and biochemical analyses suggesting that the bortezomib effect is non-reversible four weeks after the drug was withdrawn. Patients should be informed about the possibility of acute drug-induced hepatitis and hepatotoxicity of this chemotherapeutic agent after the treatment.(AU)

Objetivou-se avaliar a destoxificação da torta de mamona bruta (TMB), por meio de dois produtos alcalinos em diferentes concentrações, e seus efeitos sobre a composição química, a degradabilidade in situ da MS e o fracionamento de proteínas. Utilizou-se o hidróxido de cálcio [Ca(OH)2] e o hidróxido de sódio (NaOH) em duas concentrações (60 e 90 gramas), diluídos em quatro quantidades de água (1.000; 1.500; 2.000 e 2.500mL de água por quilo de TMB). Observou-se que, das diferentes concentrações utilizadas, somente a utilização de 90 e 60 gramas de Ca(OH)2 e NaOH, respectivamente, conseguiu destoxificar 100% da TMB, ambas diluídas em 2.000mL de água. Por outro lado, ao avaliar o tempo mínimo de contato dos reagentes com a TMB para uma máxima destoxificação, observou-se que três horas de contato é o tempo necessário para os reagentes diminuírem em 100% as proteínas citotóxicas, além de não deixar atividade hemaglutinante nesse material. A destoxificação com o NaOH proporcionou maior degradação das proteínas solúveis e da matéria seca, favorecendo a disponibilização do nitrogênio não proteico, estando sua aplicação em escala industrial na dependência de estudos sobre viabilidade operacional e econômica.(AU)


This study aimed to evaluate the detoxification of crude castor (DCC) through two alkaline products in different concentrations and their effects on the chemical composition, in situ degradability of DM and the fractionation of proteins. We used the calcium hydroxide [Ca(OH)2] and sodium hydroxide (NaOH) in two concentrations (60 and 90 grams) diluted in 4 quantities of water (1,000; 1,500; 2,000 and 2,500ml of water per kilo of DCC). It was observed that in the different concentrations used, only the use of 90 and 60 grams of Ca(OH)2 and NaOH, respectively managed to detoxify 100% of the DCC, both diluted in 2,000ml of water. On the other hand, when assessing the minimum time of contact of the reagents with the DCC for maximum detoxification, it was observed that with three hours of contact is the time required for the reagents decrease in 100% of the cytotoxic proteins, in addition to not leave haemagglutinating activity in this material. The detoxification with NaOH provided greater degradation of soluble proteins and degradation of dry matter, favoring the provision of non-protein nitrogen, while its application on an industrial scale is in the dependence of studies on operational feasibility and cost.(AU)

This study aimed to evaluate the influence of the substitution of soybean meal (SM) by detoxified castor cake (DCC) by two alkaline products on intake, performance and on the growth curve of goat's kids. 24 were used, Saanen goats (n=12) and Anglo Nubian (n=12), with initial body weight of 16.2±0.67 kg, initial age of 3 months and confined during the growth phase. The treatments consisted of three diets: a standard diet formulated with a traditional protein source, the soybean meal (SM); and test diets, which consisted of a protein source alternative, differing detoxification processes, on the basis of calcium hydroxide (Ca(OH)2) or sodium hydroxide (NaOH). The animals were distributed in a completely randomized design, in factorial 3 x 2 (diet SM, Ca(OH)2 DCC and NaOH DCC x Saanen and Anglo Nubian) totaling six sessions with four replicates each. The experimental period lasted 270 days. The non-linear models used were the Gompertz and Logistic. There was no effect (P 0.05) of breeds and interaction between both factors. The intake of DM during the phase of growth was greater for the goats fed with SM. There was no effect (P>0.05) of diets nor of the breeds in the ADG. Feed conversion was influenced (P < 0.05) by the diets, in which the goats fed diets with DCC presented better feed conversion. It has been observed that both models had good adjustments, with values numerically superior to 90%, however, the logistic model showed higher R2 and, at the same time, asymptotic index (AI). Despite the ADG be equal, the breeds presented absolute growth rate (AGR) vary over time, where the Saanen goats reached the peak first (181 days), with AGR of 0.14 kg day-¹. However, even the goats Anglo Nubian reaching the highest peak late, the AGR was higher, around 0.16 kg day-¹ higher than the Saanen. Both castor cake has the potential to replace the SM on a diet of goat's kids [...].


Objetivou-se avaliar a influência da substituição do farelo de soja (FS) pela torta de mamona destoxificada (TMD) por dois produtos alcalinos sobre o consumo, desempenho e na curva de crescimento de cabritas de aptidão leiteira. Foram utilizadas 24 cabras, Saanen (n=12) e Anglo Nubiana (n=12), com peso corporal inicial de 16,2±0,67 kg, idade inicial de 3 meses e confinadas durante a fase de crescimento. Os tratamentos consistiram de três dietas: dieta padrão formulada com uma fonte proteica tradicional, o farelo de soja (FS); e dietas testes, que consistiram de uma fonte proteica alternativa, diferindo os processos de destoxificação, à base de hidróxido de cálcio (Ca (OH)2) ou hidróxido de sódio (NaOH). Os animais foram distribuídos em delineamento inteiramente casualizado, em esquema fatorial 3 x 2 (dieta FS, TMD Ca(OH)2 e TMD NaOH x raça Saanen e Anglo Nubiana) perfazendo seis sessões com quatro repetições cada. O período experimental durou 270 dias. Os modelos não-lineares utilizados foram os de Gompertz e Logístico. Houve efeito (P 0,05) das raças e interação entre ambos os fatores. O consumo de MS durante a fase de crescimento foi maior para as cabras alimentadas com FS. Não houve efeito (P > 0,05) das dietas nem das raças no GMD. A conversão alimentar foi influenciada (P < 0,05) pelas dietas, em que as cabras que consumiram as dietas com TMD apresentaram melhor conversão alimentar. Snakebites remain a major life-threatening event worldwide. It is still difficult to make a positive identification of snake species by clinicians in both Western medicine and Chinese medicine. The main reason for this is a shortage of diagnostic biomarkers and lack of knowledge about pathways of venom-induced toxicity. In traditional Chinese medicine, snakebites are considered to be treated with wind, fire, and wind-fire toxin, but additional studies are required. Methods: Cases of snakebite seen at the Affiliated Hospital of Jiangxi University of Traditional Chinese Medicine were grouped as follows: fire toxin - including four cases of bites by Agkistrodon acutus and three bites by Trimeresurus stejnegeri - and wind-fire toxin - four cases of bites by vipers and three bites by cobras. Serum protein quantification was performed using LC-MS/MS. Differential abundance proteins (DAPs) were identified from comparison of snakebites of each snake species and healthy controls. The protein interaction network was constructed using STITCH database. Results: Principal component analysis and hierarchical clustering of 474 unique proteins exhibited protein expression profiles of wind-fire toxins that are distinct from that of fire toxins. Ninety-three DAPs were identified in each snakebite subgroup as compared with healthy control, of which 38 proteins were found to have significantly different expression levels and 55 proteins displayed no expression in one subgroup, by subgroup comparison. GO analysis revealed that the DAPs participated in bicarbonate/oxygen transport and hydrogen peroxide catabolic process, and affected carbon-oxygen lyase activity and heme binding. Thirty DAPs directly or indirectly acted on hydrogen peroxide in the interaction network of proteins and drug compounds. The network was clustered into four groups: lipid metabolism and transport; IGF-mediated growth; oxygen transport; and innate immunity. Conclusions: Our results show that the pathways of snake venom-induced toxicity may form a protein network of antioxidant defense by regulating oxidative stress through interaction with hydrogen peroxide.(AU)

GO analysis revealed that the DAPs participated in bicarbonate/oxygen transport and hydrogen peroxide catabolic process, and affected carbon-oxygen lyase activity and heme binding. Thirty DAPs directly or indirectly acted on hydrogen peroxide in the interaction network of proteins and drug compounds. The network was clustered into four groups: lipid metabolism and transport; IGF-mediated growth; oxygen transport; and innate immunity. Conclusions: Our results show that the pathways of snake venom-induced toxicity may form a protein network of antioxidant defense by regulating oxidative stress through interaction with hydrogen peroxide.(AU)

Pyrene and benzo[a]pyrene (BaP) are high molecular weight polycyclic aromatic hydrocarbons (PAHs) recalcitrant to microbial attack. Although studies related to the microbial degradation of PAHs have been carried out in the last decades, little is known about degradation of these environmental pollutants by fungi from marine origin. Therefore, this study aimed to select one PAHs degrader among three marine-derived basidiomycete fungi and to study its pyrene detoxification/degradation. Marasmiellus sp. CBMAI 1062 showed higher levels of pyrene and BaP degradation and was subjected to studies related to pyrene degradation optimization using experimental design, acute toxicity, organic carbon removal (TOC), and metabolite evaluation. The experimental design resulted in an efficient pyrene degradation, reducing the experiment time while the PAH concentration applied in the assays was increased. The selected fungus was able to degrade almost 100% of pyrene (0.08 mg mL-1) after 48 h of incubation under saline condition, without generating toxic compounds and with a TOC reduction of 17%. Intermediate metabolites of pyrene degradation were identified, suggesting that the fungus degraded the compound via the cytochrome P450 system and epoxide hydrolases. These results highlight the relevance of marine-derived fungi in the field of PAH bioremediation, adding value to the blue biotechnology.(AU)

A series of experiments were performed on one day-old chicks (Group A to F). Clinical signs of the positive control (group B fed on 400 ppb ochratoxin A contaminated feed) included depression, diarrhea, increased water intake, low body weight, high degree of genotoxicity, swollen and hemorrhagic kidneys and liver. Serum alanine amino transferase (ALT), alkaline phosphatase (ALP), creatinine and urea concentration significantly increased while albumin, globulin and total proteins were found significantly low. All these lead to increased mortality. Among adsorbents treated groups, chickens in group C (0.3% of the prepared adsorbent mixed in OTA contaminated feed) and D (0.6% of the prepared adsorbent mixed in Ochratoxin A (OTA) contaminated feed) showed very low pathological effects while group E (0.9% of the prepared adsorbent mixed in OTA contaminated feed) and F (1.2% of the prepared adsorbent mixed in OTA contaminated feed) showed normal behavior, blood biochemistry and well maintained histological structure like that in group A. As a whole, the adsorbents treated groups fed with ochratoxin A contaminated feed, supplemented with different levels of the prepared adsorbent helped to ameliorate the toxic effects of OTA and nearly showed normal clinical signs.(AU)

Aluminum (Al) toxicity in plants is seen in about 15% of the soils worldwide, restraining yields in arable land. In Brazil, acidic soils limit production of wheat (Triticum aestivum L.) and other cereals. Al is toxic for most winter cereals when its concentration increases and soil pH is below 5. One of the main concerns with acidic soil is the increase in the mobility of Al3+ions. Al binds to cell walls in roots, preventing meristematic elongation in sensitive species, causing damage to the root system and results in lower yields. Al3+ forms highly stable complexes with phosphorus (P), limiting its availability to plants, as well as reducing cell division and elongation. To deal with Al toxicity, plants have developed strategies such as organic acid (OA) exudation by roots; this mechanism of detoxification has been well-characterized. OAs, in turn, chelate ions Al3, forming non-toxic compounds that do not penetrate the root system. Some genes responsible for Al tolerance in wheat have been identified, particularly TaALMT1 and TaMATE1B that transport malate and citrate OAs, respectively. In this review, we discussed the mechanisms by which Al damages roots those by which plants are protected, primarily through two genes. We also described the interaction of the ALMT1 gene with P and iron (Fe).(AU)


A toxicidade do alumínio (Al) às plantas é observada em cerca de 15% dos solos no planeta, sendo um fator restritivo à produtividade em terras cultiváveis. No Brasil, os solos ácidos são limitantes à produção de trigo (Triticum aestivum L.) e outros cereais. O Al é tóxico para a maioria dos cereais de inverno, quando a sua concentração aumenta e o pH do solo atinge valores inferiores a 5. Uma das principais preocupações sobre o solo ácido é o aumento da mobilidade dos íons Al3+. O Al pode se ligar as paredes celulares das raízes e, como consequência, impedir o alongamento meristemático em espécies sensíveis, provocando danos ao sistema radicular, que resulta em menor desempenho agronômico das plantas. O Al3+ é também capaz de formar complexos altamente estáveis com fósforo (P), limitando sua disponibilidade para as plantas, e também reduzindo a divisão e o alongamento celular. Para lidar com a toxicidade ao Al, as plantas desenvolveram algumas estratégias como a exsudação de ácido orgânicos (AOs) pelas raízes, sendo este mecanismo de destoxificação bem caracterizado. Os AOs, por sua vez, quelam ions Al3+ formando compostos não tóxicos que não penetram no sistema radicular. Alguns genes responsáveis pela tolerância ao Al em trigo foram identificados, com ênfase para TaALMT1 e TaMATE1B, que exsudam os AOs malato e citrato, respectivamente. Nesta revisão, discutimos os mecanismos pelos quais Al danifica raízes, bem como plantas protegem-se, através de dois genes principalmente. O objetivo do presente estudo foi avaliar o valor nutricional e energético da torta de mamona destoxificada por diferentes métodos para suínos em crescimento. Foram utilizados 24 suínos machos, castrados, distribuídos em seis tratamentos, sendo uma ração referência e cinco rações testes (75% da dieta referência e 25% das tortas de mamona) contendo a torta de mamona destoxificada pelos métodos do hidróxido de cálcio, da autoclavagem, do hidróxido de cálcio+ensilagem, da autoclavagem+ensilagem e da extrusão. As tortas de mamona submetidas aos processos de destoxificação com hidróxido de cálcio e autoclavagem resultaram em maior teor de proteína digestível. Para o valor de energia metabolizável, os processos de destoxificação com hidróxido de cálcio, autoclavagem, hidróxido de cálcio+ensilagem e extrusão não diferiram entre si. Considerando-se a valoração nutricional e energética da torta de mamona por meio dos processos de destoxificação, os métodos com uso de hidróxido de cálcio e autoclavagem são os mais eficientes.(AU)


The aim of this study was to evaluate the nutritional and energy value of castor bean cake detoxified by different methods for growing pigs. A total of 24 barrows was distributed in six treatments, being in a reference diet and 5 test diets (75% of the reference diet and 25% of castor beam cake) containing castor bean cake detoxified by the method of calcium hydroxide, autoclaving, calcium hydroxide + silage, autoclaving + silage and extrusion. Castor bean cake subjected to detoxification processes with calcium hydroxide and autoclaving resulted in higher content of digestible protein. For the metabolizable energy value, the process of detoxification with calcium hydroxide, autoclaving, calcium hydroxide+silage, and extrusion did not differ. Considering the nutritional and energy valuation of castor bean cake detoxified by different procedures, the methods using calcium hydroxide and autoclaving were the most efficient.
