Background: Anaplasmosis, also called gall sickness or tropical bovine ehrlichiosis, is an infectious disease caused by species belonging to the genus Anaplasma in domestic and wild animals in tropical and subtropical regions. Anaplasma ovis and A. phagocytophilum are important pathogens of sheep. A. ovis is considered the most common species affecting sheep. The infection is usually subclinical and progresses with high fever, anaemia, icterus, weight loss and abortions. This study aimed to investigate changes in cardiac damage markers, oxidative stress and antioxidant status, cytokines, and acute phase proteins in sheep naturally infected with A. ovis. Materials, Methods & Results: For this purpose, a total of 40 animals, including 20 healthy sheep and 20 sheep infected with anaplasmosis, were used. A. ovis was diagnosed based on clinical findings and peripheral blood smear. Blood smears were prepared from the ear vein. The smears were stained with Giemsa and examined for the presence of Anaplasma spp. Infection was also confirmed by polymerase chain reaction (PCR) analysis. The genomic DNA was isolated from blood, and the MSP-4 gene region was amplified as A. ovis specific target gene. Twenty clinically healthy sheep of the same age group, reared under the same conditions and testing negative in the molecular assessment were used as controls. Blood samples were collected from the cephalic vein and and centrifuged to obtain serum. The serum stored at -20°C until the analysis stage. Serum samples were used for the analysis of cardiac damage markers [troponin I (cTnI), creatine kinase MB (CK-MB), lactate dehydrogenase (LDH) and aspartate transaminase (AST)], oxidative stress parameters [malondialdehyde (MDA), total antioxidant status (TAS), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx)], cytokines [interleukins IL-6, IL-1ß and IL-10, tumour necrosis factor α (TNF-α), and interferon-γ (IFN-γ)] and acute phase proteins [C-reactive protein (CRP), serum amyloid A (SAA) and haptoglobin (Hp)]. cTnI and CK-MB levels were measured using a chemiluminescent immunoassay. MDA, TAS, SOD, CAT, GPx, TNF-α, IL-1ß, IL-6, IL-10, IFN-γ, SAA and Hp levels were measured by an ELISA reader. LDH, AST and CRP levels were measured in an autoanalyzer. cTnI and LDH levels were significantly increased in the infected animals compared to the healthy ones (P < 0.05). The concentration of AST was decreased in infected animals. MDA, TAS, SOD, CAT and GPx levels were significantly increased in the infected animals compared to the healthy ones (P < 0.05). The levels of the inflammatory parameters such as TNF-α, IL-1ß, IL-10 and IFN-γ were significantly increased in the infected animals compared to the healthy ones (P < 0.05). Hp level were significantly increased in the infected group compared with the control group (P < 0.05). However, there was no significant change in CK-MB, SAA and CRP concentrations in the infected animals (P > 0.05). Discussion: Ovine anaplasmosis is an obligate intracellular arthropod disease that causes widespread changes in haematobiochemical, immune response and oxidative stress parameters. Cardiac damage is often overlooked in field conditions due to the lack of adequate knowledge about the pathophysiology of the disease. Our results showed that A. ovis infection leads to significant changes in cardiac biomarkers and that the parasite can cause cardiac dysfunction. This is the first report on cardiac damage markers in Anaplasma-infected sheep. Additionally, the levels of proinflammatory and oxidative stress markers that may cause functional disorders were also found to be increased. Thus, measuring markers of cardiac function, oxidative stress and inflammation can be a useful tool in the early diagnosis of ovine anaplasmosis.

Background: Prostaglandin F2 alpha (PGF2 α) binds to the specific receptor (PTGFR) on the corpus luteum (CL) in mammals, inducing regression of the CL structure (luteolysis) and initiating a new cycle. While PGF2 α is effective only on mature CL, the immature CL structure (early luteal phase) resists PGF2 α. In this study, sildenafil citrate, which is used to increase blood flow in the genital organs for treating specific pregnancy issues in women, was administered during the early luteal phase in a rabbit model to test the hypothesis of enhancing blood flow to the CL, thereby promoting earlier maturation and enabling a response to PGF2 α. Materials, Methods & Results: The study was conducted in 2 sub-studies: clinical and molecular. A large number of rabbits were initially included in the sub-studies to ensure a sufficient number of pseudo-pregnant rabbits. Ovulation in rabbits was induced with buserelin acetate and was considered as day 0 of the study. The sub-studies were continued with rabbits whose pseudo-pregnancies were confirmed according to progesterone (P4 ) results. As a result, the studies were continued with a total of 41 pseudo-pregnant New Zealand female rabbits, 21 of which were included in the clinical sub-study and 20 in the molecular sub-study. In both sub-studies, on day 3 of the luteal period, rabbits in the treatment group received 5 mg/kg sildenafil citrate and all rabbits received a single dose of exogenous PGF2 α on day 4 to induce luteolysis. In the clinical sub-study, echotexture and intraovarian blood flow changes in the ovaries were determined by ultrasonography (USG) examination. In the molecular sub-study, the expression changes of Hypoxia Inducible Factor 1 Alpha (HIF1A) and Vascular Endothelial Growth Factor (VEGF) related to angiogenesis, Steroidogenic Acute Regulatory Protein (StAR) related to P4 metabolism, Prostaglandin-Endoperoxide Synthase 2 (PTGS2) related to prostaglandin (PG) mechanism and 15-Hydroxyprostaglandin Dehydrogenase (HPGD) genes at mRNA level were determined using Real Time Polymerase Chain Reaction (RT-PCR) in CL tissues obtained with ovariohysterectomy (OVH) at 1 and 12 h after PGF2 α injection. In addition, blood samples were collected for determine P4 levels from all rabbits. In the clinical sub-study; there was no difference between the groups in mean gray values (MGV), whereas there was a significant decrease in both pulsatile index (PI) and resistance index (RI) values at 40 min after PGF2 α injection (P < 0.05). In the molecular sub-study, it was determined that sildenafil citrate had no significant effect (P > 0.05) on the expression levels 1 and 12 h after PGF2 α injection. According to the results of the molecular sub-study, no significant effect of sildenafil citrate on the mRNA expression levels in the investigated genes was detected (P > 0.05). However, within each group, differences were found according to OVH time after PGF2 α injection. It was observed that PTGS2 and HPGD mRNA expressions decreased at the 12th h compared to the 1st h, while HIF1A expression increased (P < 0.05). Discussion: According to the results obtained from clinical and molecular sub-studies, it was determined that a single dose of sildenafil citrate (5 mg/kg) applied on the 3rd day of the luteal period did not contribute to the maturation process of the CL, did not increase blood flow, and was insufficient to break the resistance of the CL against PGF2 α applied on the 4th day of the luteal period. However, a significant decrease in the PI value at the 40th min after PGF2 α injection suggests that sildenafil citrate has a supportive effect, and that this decrease is also seen in the RI value, suggesting that its effect is insufficient against the vasoconstrictive effect of PGF2 α.

Studies on the germination and establishment of plants are key pieces to understanding the reproductive success of plants. This work aimed to describe in vitro germination and reserve mobilization in the bromeliad Vriesea friburgensis through morphological, histochemical, and biochemical analysis. The conditions used in this study for the in vitro germination are adequate. From the third day of in vitro inoculation, a uniform germination of 98% was obtained, exhibiting a high physiological quality of the seeds and a high potential to produce seedlings (94%). There is early reserve mobilization, which began in the imbibition phase. The accumulated reserves in the endosperm cytoplasm are degraded by hydrolytic enzymes provided by the aleurone layer. It is possible that compounds in the cell walls of the endosperm contribute to a lesser extent in mobilization. Additionally, it was observed that starch accumulation in the cotyledon increases when the seedling has formed. Results from this study provide insights for future studies on ecology, seed technology, and conservation in this species. This study contributes to the limited knowledge of the dynamics of reserves during germination and seedling establishment in Bromeliaceae. To the best of our knowledge, this is the first study with this approach in the genus Vriesea.
Estudos sobre germinação e estabelecimento de plantas são peças-chave para entender o sucesso reprodutivo das plantas. Este trabalho teve como objetivo descrever a germinação in vitro e a mobilização de reservas na bromélia Vriesea friburgensis por meio de análises morfológicas, histoquímicas e bioquímicas. As condições utilizadas neste estudo para a germinação in vitro são adequadas. A partir do terceiro dia de inoculação in vitro, obteve-se germinação uniforme de 98%, apresentando alta qualidade fisiológica das sementes e alto potencial de produção de plântulas (94%). Há uma mobilização precoce de reservas, iniciada na fase de embebição. As reservas acumuladas no citoplasma do endosperma são degradadas por enzimas hidrolíticas fornecidas pela camada de aleurona. É possível que compostos nas paredes celulares do endosperma contribuam em menor grau na mobilização. Além disso, observou-se que o acúmulo de amido no cotilédone aumenta com a formação da plântula. Os resultados deste estudo fornecem informações para estudos futuros sobre ecologia, tecnologia de sementes e conservação desta espécie. Este estudo contribui para o conhecimento limitado da dinâmica das reservas durante a germinação e estabelecimento de plântulas em Bromeliaceae. Até onde sabemos, este é o primeiro estudo com esta abordagem no gênero Vriesea.

The desiccation time of high-biomass pasture and nitrogen (N) fertilization of pasture and soybean can influence the soybean sowing, establishment and growth. The objective of this study was to evaluate how the time of desiccation of the preceding pasture of Urochloa brizantha cv. BRS Piatã, cultivated at three levels of N, and by the soybean N fertilization affect soybean growth and N accumulation. Three N rates (0; 150 and 300 kg ha-1), broadcast as urea on the U. brizantha pasture were evaluated separately in each N level, every one considered as one experiment. In each experiment, five times of pasture desiccation were evaluated (60; 45; 30; 15, and 1 day before soybean sowing) and two levels of soybean N fertilization: 30 kg ha-1 (urea) broadcast at sowing or without N fertilization. A randomized complete block design with five replications was used. Early desiccation of U. brizantha pasture favors the establishment of soybean and promotes an increase in biomass and N accumulation in the vegetative stages, however these differences are not observed during the grain filling, regardless the soybean N fertilization. The soybean yield was not influenced by the desiccation time. N fertilization with 30 kg ha-1 at sowing intensifies soybean growth at the vegetative phase, but after full flowering, there were no effects on biomass and grain yield, independently of the desiccation time.
A época da dessecação da pastagem de braquiária com alta quantidade de biomassa e a adubação nitrogenada na pastagem e na soja podem influenciar a qualidade da semeadura e as condições para o estabelecimento e o crescimento das plantas de soja. O objetivo do trabalho foi avaliar o efeito de épocas de dessecação de pastagem de Urochloa brizantha cv. BRS Piatã, cultivada em três doses de nitrogênio, e da adubação nitrogenada na soja, sobre o crescimento e acúmulo de N pela cultura. Foram estudadas três doses de N na pastagem de U. brizantha cv. BRS Piatã: 0; 150 e 300 kg ha-1 de N, aplicados na forma de ureia, a lanço, constituindo três experimentos, que foram analisados separadamente. Em cada experimento foram avaliadas cinco épocas de dessecação da pastagem: 60; 45; 30; 15 e 1 dias antes da semeadura da soja e dois níveis de adubação nitrogenada na soja, com 30 kg N ha-1 aplicados a lanço no dia da semeadura (ureia) e sem adubação nitrogenada. Foi utilizado o delineamento de blocos completos casualizados, com cinco repetições. Os resultados evidenciaram que a dessecação antecipada da Urochloa brizantha cv. BRS Piatã favoreceu o estabelecimento de plantas de soja e promoveu incremento na massa seca e acúmulo de N nos estádios vegetativos da cultura, entretanto essas diferenças não são observadas no enchimento e na produtividade de grãos, independentemente da adubação nitrogenada na soja. A produtividade de grãos não é influenciada pelas épocas de dessecação. A adubação nitrogenada na implantação da soja com 30 kg ha-1 de N proporciona maior crescimento das plantas de soja na fase vegetativa, mas, após o pleno florescimento, não interfere na biomassa e na produtividade de grãos, independentemente da época de dessecação.

Pyrrhulina brevis is an endemic fish from the Amazon basin that is valued in the ornamental fish market. In larviculture, fish are very sensitive to stressors and have a high mortality rate. Salinized water may provide more energy to overcome stress and feeding frequency is an important strategy to improve productive performance. Therefore, the aim of this study was to evaluate the best feeding frequency and the best level of water salinization for larval development of P. brevis. A total of 360 post-larvae of P. brevis (5.26 ± 1.65 mg, 5.57 ± 0.68 mm) were randomly distributed in 36 aquariums (1 L) in a 3x4 factorial scheme, to evaluate three concentrations of salt in water (0, 1 and 2 g L-1) and four feeding frequencies (once, twice, three and four times a day). We offered 150 Artemia nauplii per day and per post-larvae during 15 days. The fish kept at 1 g L-1 salinized water, and those fed 3 and 4 times a day presented significantly greater final length and weight, weight gain, and specific growth rate for length and weight. An interaction between salinized water and feeding frequency was observed for survival rate. We conclude that, during the initial rearing phase of P. brevis, feeding frequency of three times a day and salinized water at 1 g L-1 is recommended.(AU)
Pyrrhulina brevis é um peixe endêmico da bacia amazônica que apresenta bons valores no mercado de peixes ornamentais. Na larvicultura, os peixes são muito sensíveis aos estressores e apresentam alta taxa de mortalidade. A água salinizada pode fornecer mais energia para superar o estresse e a frequência alimentar é uma estratégia importante para melhorar o desempenho produtivo. Portanto, o objetivo deste trabalho foi avaliar a melhor frequência alimentar e o melhor nível de salinização da água para o desenvolvimento larval de P. brevis. Um total de 360 pós-larvas de P. brevis (5,26 ± 1,65 mg, 5,57 ± 0,68 mm) foram distribuídos aleatoriamente em 36 aquários (1 L) em esquema fatorial 3x4, para avaliar três concentrações de sal na água (0, 1 e 2 g L-1) e quatro frequências de alimentação (uma, duas, três e quatro vezes ao dia). Foram oferecidas 150 náuplios de Artemia por dia e por pós-larva, durante 15 dias. Os peixes mantidos com 1 g L-1 de água salinizada e os alimentados 3 e 4 vezes ao dia apresentaram comprimento e peso finais, ganho de peso e taxa de crescimento específico para comprimento e peso significativamente maiores. Uma interação entre água salinizada e frequência alimentar foi observada para a taxa de sobrevivência. Concluímos que, durante a fase inicial de criação de P. brevis, recomenda-se uma frequência alimentar de três vezes ao dia e água salinizada a 1 g L-1.(AU)

O coração é composto por duas importantes valvas átrio-ventriculares, a tricúspide e a bicúspide ou mitral. As válvulas cardíacas asseguram o sentido do fluxo sanguíneo dos átrios para os ventrículos e destes para as artérias. A endocardiose é um processo degenerativo crônico progressivo das válvulas, que acomete especialmente os cães, denominada endocardiose ou "doença do cão velho". Embora qualquer das válvulas possa ser afetada, é a válvula mitral que é habitualmente envolvida, isoladamente ou em combinação com a válvula tricúspide. A sua prevalência tem sido associada a parâmetros como idade, sexo e raça, afetando especialmente animais adultos de raças de pequeno porte. A etiologia da endocardiose é desconhecida, mas parece haver uma base hereditária. As lesões macroscópicas iniciais consistem em pequenos nódulos nas margens livres da valva, ficando distorcidas e com suas dimensões reduzidas, mas significativamente espessas e de coloração opaca, branca ou reluzente. Microscopicamente, a endocardiose é vista como um processo degenerativo do tecido valvular com discreta infiltração de tecido conjuntivo fibroso. Em estágios iniciais a doença apresenta, à auscultação, murmúrios cardíacos de baixa intensidade, sem sinais de descompensação e é geralmente achado incidental durante a rotina de exame clínico. As cúspides fundem-se em sua inserção, mas separam-se em direção ao centro da abertura, ajustando-se estreitamente ao mesmo tempo quando a valva se fecha e suas bordas livres são espessas e irregulares, especialmente durante uma fase tardia da vida. Com isso, o objetivo do presente trabalho foi averiguar as principais lesões, a epidemiologia e o grau de prevalência da endocardiose em cães na região noroeste do estado do Rio Grande do Sul, o que se verificou que não há casos frequentes na rotina clínica de pequenos animais de endocardiose. Não sendo possível verificar se há um sexo mais predisposto a apresentar a endocardiose. As lesões observadas na macroscopia eram referentes a nódulos brilhantes, e na microscopia presença de colágeno causando o espessamento da cúspide, além de achados inflamatórios.(AU)
The heart is composed of two important atrio-ventricular valves, the tricuspid and the bicuspid or mitral valve. The heart valves ensure the direction of blood flow from the atria to the ventricles and from the ventricles to the arteries. Endocardiosis is a chronic progressive degenerative process of the valves that affects dogs in particular, called endocardiosis or "old dog's disease". Although any of the valves can be affected, it is the mitral valve that is usually involved, either alone or in combination with the tricuspid valve. Its prevalence has been associated with parameters such as age, sex, and breed, affecting especially adult animals of small breeds. The etiology of endocardiosis is unknown, but there appears to be a hereditary basis. The initial macroscopic lesions consist of small nodules on the free margins of the valve, becoming distorted and reduced in size, but significantly thickened and opaque, white or shiny in color. Microscopically, endocardiosis is seen as a degenerative process of the valvular tissue with a discrete infiltration of fibrous connective tissue. In early stages the disease presents, on auscultation, with low-intensity cardiac murmurs without signs of decompensation and is usually an incidental finding during routine clinical examination. The leaflets fuse at their insertion, but separate towards the center of the opening, fitting closely together when the valve closes, and their free edges are thick and irregular, especially during a late phase of life. Thus, the aim of this study was to investigate the main lesions, the epidemiology and the degree of prevalence of endocardiosis in dogs in the northwest region of the state of Rio Grande do Sul, which was found that there are no frequent cases in the clinical routine of small animals of endocardiosis. It was not possible to verify whether there is a sex more predisposed to present endocardiosis. The lesions observed at macroscopy were referring to bright nodules, and at microscopy the presence of collagen causing thickening of the leaflet, besides inflammatory findings.(AU)
El corazón está compuesto por dos importantes válvulas aurículo- ventriculares, la tricúspide y la bicúspide o válvula mitral. Las válvulas cardíacas aseguran la dirección del flujo sanguíneo de las aurículas a los ventrículos y de los ventrículos a las arterias. La endocardiosis es un proceso degenerativo crónico y progresivo de las válvulas que afecta sobre todo a los perros, denominado endocardiosis o "enfermedad del perro viejo". Aunque cualquiera de las válvulas puede verse afectada, es la válvula mitral la que suele estar implicada, ya sea sola o en combinación con la válvula tricúspide. Su prevalencia se ha asociado a parámetros como la edad, el sexo y la raza, afectando especialmente a animales adultos de razas pequeñas. La etiología de la endocardiosis es desconocida, pero parece tener una base hereditaria. Las lesiones macroscópicas iniciales consisten en pequeños nódulos en los márgenes libres de la válvula, que se distorsionan y reducen de tamaño, pero se engrosan significativamente y adquieren un color opaco, blanco o brillante. Microscópicamente, la endocardiosis se observa como un proceso degenerativo del tejido valvular con una discreta infiltración de tejido conectivo fibroso. En fases tempranas, la enfermedad se presenta, en la auscultación, con soplos cardíacos de baja intensidad sin signos de descompensación y suele ser un hallazgo incidental durante el examen clínico rutinario. Las valvas se fusionan en su inserción, pero se separan hacia el centro de la apertura, encajando estrechamente cuando la válvula se cierra, y sus bordes libres son gruesos e irregulares, especialmente durante una fase tardía de la vida. Por lo tanto, el objetivo de este estudio fue investigar las principales lesiones, la epidemiología y el grado de prevalencia de endocardiosis en perros de la región noroeste del estado de Rio Grande do Sul, que se encontró que no hay casos frecuentes en la rutina clínica de pequeños animales de endocardiosis. No fue posible verificar si existe un sexo más predispuesto a presentar endocardiosis. Las lesiones observadas en la macroscopia fueron referentes a nódulos brillantes, y en la microscopia la presencia de colágeno causando engrosamiento de la valva, además de hallazgos inflamatorios.(AU)

Proteiongram analysis is useful for the early diagnosis of intramammary infections during the period of colostrogenesis. This study aimed to evaluate the profile of total proteins, immunoglobulins, lactoferrin, and gamma-glutamyl transferase (GGT) in the colostrum of dairy goats with intramammary infections. Animals were divided in groups: GI (n=12) of goats without mammary gland infections, and GII (n=8) of goats with mammary gland infections. Intramammary infections were diagnosed using microbiological isolations and somatic cell counts Cs). Total protein was evaluated in the samples using SDS-PAGE shortly after parturition, and 24 and 48 hours after that event. Non-aureus Staphylococcus (NAS) were detected all isolates. At 48 h, GII had high IgG levels and a SCC of 1660.25 x 103/mL. Levels of total protein were high in this group at 24 and 48 h. Albumin levels were high in goats with mastitis at 24 h. Overall, the IgG, lactoferrin, and albumin levels differed between animals with and without intramammary infections at M0. GGT activity was not influenced by the intramammary infection. The results of this study reinforce the importance of the proteinogram as an auxiliary tool in the diagnosis of mastitis in dairy goats.
O proteinograma do colostro/leite pode ser útil para diagnóstico precoce de infecções intramamárias, assim como para a avaliação da intensidade da resposta inflamatória. Este estudo teve como objetivo avaliar o perfil de proteínas, imunoglobulinas, lactoferrina e gamaglutamiltransferase no colostro de cabras leiteiras portadoras s de infecções intramamárias. Os animais foram distribuídos em dois grupos, GI (n 12), composto por cabras sem isolamento microbiológico das glândulas mamárias, e GII (n 8), composto por cabras com resultados positivos na cultura do leite de pelo menos uma das glândulas mamárias. O diagnóstico de infecção intramamária foi realizado logo após o parto, por meio de isolamento microbiológico e de contagem de células somáticas (CCS). Os níveis de proteína total, imunoglobulina A (IgA), imunoglobulina (G) (IgG), lactoferrina, albumina e atividade da gamaglutamiltransferase (GGT) foram avaliados em amostras de colostro/leite de transição, usando-se SDS-PAGE logo após o parto bem como às 24 e às 48 horas após esse evento. Staphylococcus não aureus (NAS) foram encontrados em todos os isolamentos. A concentração de IgG de 186 foi superior no GII apenas às 48h, ao mesmo tempo em que o CCS foi de 1660,25 x 10/mL. No entanto, a proteína total foi maior neste grupo às 24 e às 48h. A albumina foi maior nas cabras com mastite às 24h, e a lactoferrina no momento 0h (M0) e às 48h. Em geral, os valores de IgG, lactoferrina e albumina diferiram entre os animais com e sem infecções intramamárias no M0. A atividade da gamaglutamiltransferase não foi influenciada pela infecção intramamária. Os resultados deste estudo reforçam a importância do proteinograma como uma ferramenta auxiliar no diagnóstico da mastite em cabras leiteiras.

The objective of this work was to evaluate the early and late immunological modulation of an experimental infection of T. canis larvae in mice. Mice were infected with 100 infective larvae and euthanized at different period: 24, 48 hours post infection (HPI), 15- and 30 days post infection (DPI). The humoral response was evaluated by indirect ELISA. Quantitative RT-PCR (qPCR) was used to quantify the mRNA transcription of cytokines IL4, IL10, IL12 and Ym1 in the early and late infection periods. Infection with T. canis was able to generate specific total IgG at 15- and 30- DPI. Analyzing the IgG isotype revealed a significant differentiation for IgG1 compared with IgG2a, IgG2b and IgG3, characterizing a Th-2 response. Evaluating the gene transcription at the early phase of infection, higher transcription levels of IL10, IL4 and Ym1 and a downregulation of IL12 were observed. By the late phase, increased transcription levels of IL4, Ym1 and IL12 were observed, and downregulation of IL-10 transcription was observed. The data obtained suggest that during experimental infection with T. canis, the participation of the IL4, IL10, IL12 cytokines and Ym1 can play an important role in T. canis immunomodulation.(AU)
O objetivo deste trabalho foi avaliar a modulação imunológica precoce e tardia da infecção experimental de larvas de T. canis em camundongos. Estes foram infectados com 100 larvas infectantes e eutanasiados em diferentes períodos: 24 e 48 horas pós infecção (HPI), 15 e 30 dias após a infecção (DPI). A resposta humoral foi avaliada por ELISA indireto. RT-PCR quantitativo (qPCR) foi usado para quantificar a transcrição de mRNA das citocinas IL4, IL10, IL12 e Ym1 nos períodos de infecção precoce e tardia. A infecção por T. canis foi capaz de gerar IgG total específico aos 15 e 30 DPI. A análise do isótipo IgG revelou uma diferenciação significativa para IgG1 em comparação com IgG2a, IgG2b e IgG3, caracterizando uma resposta Th-2. Avaliando-se a transcrição gênica na fase inicial da infecção, foram observados níveis mais elevados de transcrição de IL10, IL4 e Ym1 e a regulação negativa de IL12. Na fase tardia, foram observados níveis aumentados de transcrição de IL4, Ym1 e IL12, e foi observada regulação negativa da transcrição de IL-10. Os dados obtidos sugerem, que durante a infecção experimental com T. canis, a participação das citocinas IL4, IL10, IL12 e Ym1 podem desempenhar um papel importante na imunomodulação de T. canis.(AU)

Purpose: To evaluate the effect of preoperative intravenous chemotherapy with 5-fluorouracil on liver regeneration in an experimental model of major hepatectomy in rats. Methods: Wistar rats were divided into two groups of 20 animals each and submitted to 70% hepatectomy 24 h after intravenous injection of 5-fluorouracil 20 mg/kg (fluorouracil group, FG) or 0.9% saline (control group, CG). After hepatectomy, each group was subdivided into two subgroups of 10 animals each according to the day of sacrifice (24 h or 7 days). Liver weight during regeneration, liver regeneration rate using Kwon formula, and the immunohistochemical markers proliferating cell nuclear antigen (PCNA) and Ki-67 were used to assess liver regeneration. Results: At early phase (24 h after hepatectomy) it was demonstrated the negative effect of 5-fluorouracil on liver regeneration when assessed by Kwon formula (p < 0.0001), PCNA analysis (p = 0.02). With regeneration process complete (7 days), it was possible to demonstrate the sustained impairment of chemotherapy with 5-fluorouracil on hepatocytes regeneration phenomenon when measured by Kwon formula (p = 0.009), PCNA analysis (p = 0.0001) and Ki-67 analysis (0.001). Conclusions: Preoperative chemotherapy with intravenous 5-fluorouracil negatively affected the mechanisms of liver regeneration after major hepatectomy in rats.

Abstract Cinclodes pabsti is an endemic passerine restricted to the highland areas in southern Brazil. The aim of this study was to provide information on its breeding biology. The nesting cavities along road cuts were monitored from May 2008 to March 2011. The survey was carried out monthly from May to July 2008, February to July 2009, 2010 (non-breeding season), weekly from August 2008 to January 2009, 2010 (breeding season), and on a 2 to 4-days basis from August 2010 to January 2011. The geographic location, physical characteristics, and soil/substrate type in which the nesting cavities were situated were recorded. The total number of cavities used in the three breeding seasons was 136, resulting in 295 nests. The distance of a nest to its nearest neighbor ranged from 24-2,368 m, with a higher number of nests (n = 34; 59.7%) in the distance interval of 24-500 m. There was a greater usage of cavities located in Inceptisols, and the distances of nesting cavity entrances to the ground and to the top of road cuts were 1.6 ± 0.9 m and 0.8 ± 0.62 m, respectively. The breeding season lasted 148 days from mid-August to early January. Clutch size (n = 256) varied from 2 to 3 eggs, and the eggs (n = 155) had a total length of 27.2 ± 1.3 mm, breadth of 20.9 ± 0.8 mm, and mass of 6.2 ± 0.7 g. The incubation phase lasted 17.3 ± 0.8 days and the nestling phase for 18.3 ± 1.5 days. The body mass of the chicks was 6.0 ± 1.0 g just after hatching and reached a maximum of 59.6 ± 2.4 g at 16 days of age. Our results can contribute to filling the gaps in knowledge of C. pabsti ecology, because its habitat is under high anthropic pressures and the information on its life history is yet limited.

Resumo
For the past years, there is an increased interest for maternal behavior in humans best friend the dog. Indeed, studies show that the quality of mothering is not only important for the survival of the puppies during the first weeks of life, for the bonding process but recently it shows that it might have a long-lasting impact on the puppies future performance. However, how to define and measure maternal behavior? From the early studies to nowadays, the display of acts from the bitch right prior to parturition (preparation phase), and then continuously toward the puppies until weaning are the repertoire of actions that define maternal behavior. For the scientific community, maternal care is quantified as a principal component assessing attitudes that are easy to measure from the dam to the puppies such as: the time spent in contact, time and/or frequency of licking/grooming and time and position of nursing. Though, there is still a need to elucidate different aspects of maternal behavior in the dog, for example the effect of genetics, the impact of the emotional state of the bitch and the quality of maternal behavior, the role of hormones, the possibility to improve maternal behavior, there is some information to share. The idea is to give an overview of maternal behavior that permits veterinarians, breeders to identify and prevent problems to optimize the dog welfare.

This study was aimed at determining the concentration of pyrethroid in the surface water, sediment, Weyonia acuminata and Synodontis clarias fish in Lekki lagoon, Lagos, Nigeria. In-situ physicochemical analysis of the surface water was conducted using a calibrated handheld multi-parameter probe (Horiba Checker Model U-10). Intestinal samples from infected and uninfected fish were analyzed for pyrethroid concentrations, microbial colonization, proteins (PRO), superoxide dismutase (SOD), catalase (CAT), reduced glutathione (GSH), malonaldehyde (MDA) and glutathione peroxidase (GPx). The histopathology of infected and uninfected intestinal tissues were analyzed using Haematoxylin and Eosin (H&E) stains and examined under a binocular light microscope (Model 230485). A total of 39 out of 98 S. clarias fish were infected with cestode parasite, Wenyonia acuminate, amounting to 39% parasite prevalence. Among the tested pyrethroids, Cyfluthrin and Alpha cypermethrin had significant sorption of 1.62 and 3.27 respectively from the aqueous phase to the bottom sediment of the lagoon. Pyrethroid concentration was in the order of sediment > parasite> water > intestine> liver. The cholesterol, triglycerides, and LDL (low density lipids) in the parasite were higher than in the host fish (p 0.05). On the other hand, the fish hepatic protein, high density lipids (HDL) and glucose were higher than the levels in the parasites (p 0.05). There was a high prevalence of gut microbes (30 - 40%), which include Salmonella sp, Escherichia coli, Pseudomonas sp and Bacillus sp among individuals infected with gut Cestodes, Wenyonia sp compared with uninfected individual which had higher gut Staphylococcus sp., Klebsiella sp., and Proteus sp. (10- 40%). among the congeners of pyrethroids analyzed, Cyfluthrin > Alpha-Cyermethrin had significant sorption on the sediment, however unlike a benthopelagic fish, no pyrethroid was accumulated in the S. clarias from the sediment. The fish however accumulated Alpha-Cypermethrin from the aqueous phase. The parasite on the other hand accumulated Bathroid significantly from the surface water and bottom sediment which may be linked to the higher stress levels observed in the parasite than the host fish. The parasite in turn inflicted histological alterations on the host intestine, marked by moderate inflammation of mucosa, alteration of the villi microstructure, moderate stunting of the villous structure and moderate fibrosis of villous structure. The study demonstrated the efficiency of histopathological and microbial analysis in biomonitoring studies enteric parasites and early detection of pyrethroid toxicity respectively compared to bioaccumulation analysis.(AU)
Este estudo teve como objetivo determinar a concentração de piretroide nos peixes de águas superficiais, sedimentos, Wenyonia acuminata e Synodontis clarias na lagoa de Lekki, Lagos, Nigéria. A análise físico-química in situ da água de superfície foi realizada usando uma sonda multiparâmetro manual calibrada (Horiba Checker Modelo U-10). Amostras intestinais de peixes infectados e não infectados foram analisadas quanto a concentrações de piretroides, colonização microbiana, proteínas (PRO), superóxido dismutase (SOD), catalase (CAT), glutationa reduzida (GSH), malonaldeído (MDA) e glutationa peroxidase (GPx). A histopatologia dos tecidos intestinais infectados e não infectados foi analisada usando manchas de Hematoxilina e Eosina (H&E) e examinada sob um microscópio de luz binocular (Modelo 230485). Um total de 39 dos 98 peixes de S. clarias foi infectado com o parasita cestoide Wenyonia acuminata, totalizando 39% de prevalência do parasita. Entre os piretroides testados, ciflutrina e alfa-cipermetrina tiveram sorção significativa de 1,62 e 3,27, respectivamente, da fase aquosa ao sedimento de fundo da lagoa. A concentração de piretroides estava na ordem de sedimentos > parasita > água > intestino > fígado. O colesterol, os triglicerídeos e o LDL (lipídios de baixa densidade) no parasita foram maiores que no peixe hospedeiro (p 0,05). Por outro lado, a proteína hepática dos peixes, os lipídios de alta densidade (HDL) e a glicose foram superiores aos níveis nos parasitas (p 0,05). Houve uma alta prevalência de micróbios intestinais (30-40%), incluindo Salmonella sp., Escherichia coli, Pseudomonas sp. e Bacillus sp. entre indivíduos infectados no intestino com o parasita cestoide Wenyonia sp. em comparação com indivíduos não infectados com Staphylococcus sp., Klebsiella intestinal sp. e Proteus sp. (10-40%). Entre os congêneres dos piretroides analisados, ciflutrina > alfa-cipermetrina teve sorção significativa no sedimento, porém, diferentemente de um peixe bentopelágico, nenhum piretroide foi acumulado no S. clarias do sedimento. No entanto, os peixes acumularam alfa-cipermetrina a partir da fase aquosa. O parasita, por outro lado, acumulou Bathroid significativamente da água superficial e do sedimento do fundo, o que pode estar relacionado aos níveis mais altos de estresse observados no parasita do que no peixe hospedeiro. O parasita, por sua vez, causou alterações histológicas no intestino hospedeiro, marcadas por inflamação moderada da mucosa, alteração da microestrutura das vilosidades, atrofia moderada da estrutura das vilosidades e fibrose moderada da estrutura das vilosidades. O estudo demonstrou a eficiência da análise histopatológica e microbiana nos estudos de biomonitoramento de parasitas entéricos e detecção precoce da toxicidade dos piretroides, respectivamente, em comparação à análise de bioacumulação.(AU)

Purpose To analyze the plasma lipid spectrum between healthy control and patients with pancreatic cancer and to select differentially expressed tumor markers for early diagnosis. Methods In total, 20 patents were divided into case group and healthy control group according to surgical pathology. Of almost 1206 plasma lipid molecules harvested from 20 patients were measured by HILIC using the normal phase LC/MS. Heat map presented the relative levels of metabolites and lipids in the healthy control group and patients with pancreatic cancer. The PCA model was constructed to find out the difference in lipid metabolites. The principal components were drawn in a score plot and any clustering tendency could be observed. PLS-DA were performed to distinguish the healthy control group and pancreatic cancer according to the identified lipid profiling datasets. The volcano plot was used to visualize all variables with VIP>1 and presented the important variables with P 0.01 and ,FC,>2. Results The upregulated lipid metabolites in patients with pancreatic cancer contained 9 lipids; however, the downregulated lipid metabolites contained 79 lipids. Conclusion There were lipid metabolomic differences in patients with pancreatic cancer, which could serve as potential tumor markers for pancreatic cancer.(AU)

The effect of basal zone defoliation on vines influences yield and ripening depending on the phenological stage and its intensity. Early basal defoliation (EBD) was carried out at flowering on the autochthonous Vitis vinifera (L.) cultivar Nerello mascalese grown on the eastern slopes of Mount Etna, Sicily. The effects were evaluated over a two-year period. In the first year, the canopy retained 67 % of its original leaf area after EBD and in the second year, 58 %. Compared with control vines, mid-day leaf water potentials in the EBD vines enjoyed higher water status throughout the growing seasons in both years. EBD had no significant negative effects on yield but did have significant positive effects on the levels of total polyphenols, total flavonols and total anthocyanins. Yield and its components are conditioned by a number of factors mainly linked to climate and to the vines general condition during berry growth and ripening. Even after two consecutive years of treatment, EBD does not have any negative effects on yield. However, EBD does have strong positive effects on berry quality attributes as a consequence of the following: greater exposure of the cluster to sunlight, higher vine water status and avoidance of high temperature stresses in the last phase of ripening during the analysis of anthocyanin.

ABSTRACT: The present study evaluated the volume and function of the left atrium by two-dimensional echocardiographic feature-tracking imaging (2D-FTI) and Simpsons monoplanar modeling in dogs with asymptomatic degenerative mitral valve disease (DMVD). The study consisted of 80 dogs that were divided into the following three groups: Group 1, 21 dogs (A); Group 2, 30 dogs (B1) and Group 3, 29 dogs (B2). The variable strain (contraction phase) was significantly lower in Group 3 than in Group 1 (12.92±4.54 x 16.69±5.74, p=0.014), and significant differences in the contraction strain index (CSI) were observed between all of the groups that were evaluated (1 = 46.82±8.10, 2 = 39.88±8.03, 3 = 35.25±5.64, p 0.0001). The atrial diastolic volume index (AdVi) that was measured by 2D-FTI was significantly higher in Group 3 than in Group 1 (1.31±0.95 x 0.96±0.31, p=0.038), and the atrial cardiac index (ACI) was also higher in Group 3 than in Group 1 (102.38±80.18 x 78.19±33.38, p=0.030). Atrial function was assessed by Simpsons monoplanar method, which demonstrated an increase in the left atrial systolic volume, while the contractile function decreased with an increasing disease severity (Group 1 0.21±0.06; Group 2 0.25±0.06; Group 3 0.32±0.08, p 0.0001). The intraobserver and interobserver assessments showed low to moderate variability; most of the values for the coefficient of variation for the variables that were analysed with each method were below 25%. Thus, DMVD was determined to cause an alteration in atrial function, especially in the contraction phase, and even in asymptomatic animals, and the methods of 2D-FTI echocardiography and Simpsons monoplanar evaluation are sensitive and early methods for the detection of left atrial dysfunction.
RESUMO: O presente estudo avaliou o volume e a função atrial esquerda obtidos por meio da ecocardiografia bidimensional feature tracking (2D-FTI) e pelo método monoplanar de Simpson em cães saudáveis e cães com DMVD assintomáticos. Foram avaliados 80 cães distribuídos em três grupos: Grupo 1, 21 cães (classe A); Grupo 2, 30 cães (classe B1) e Grupo 3, 29 cães (classe B2). A variável strain (fase de contração) foi significativamente menor no Grupo 3 que no Grupo 1 (12,92±4,54 x 16,69±5,74, p=0,014) e para a variável índice de strain de contração (CSI), houve diferença estatística entre todos os grupos avaliados (1 = 46,82±8,10; 2 = 39,88±8,03; 3 = 35,25±5,64, p 0,0001). O índice de volume diastólico atrial (iVdA) mensurado por meio do 2D-FTI foi significativamente maior no Grupo 3 que no Grupo 1 (1,31±0,95 x 0,96±0,31, p=0,038), assim como para o índice cardíaco atrial (iCA) também foi maior no Grupo 3 (102,38±80,18 x 78,19±33,38, p=0,030). A função atrial avaliada pelo método monoplanar de Simpson demonstrou um aumento do volume atrial esquerdo e do volume sistólico do átrio esquerdo, enquanto que a função contrátil diminuiu com o aumento da gravidade da doença (Grupo 1 0,21±0,06; Grupo 2 0,25±0,06; Grupo 3 0,32±0,08; p 0,0001). A avaliação intraobservador e interobservador, demonstrou variabilidade baixa a moderada, uma vez que a maioria dos valores de coeficiente de variação se concentraram abaixo de 25% para as variáveis analisadas em ambos os métodos. Dessa forma, conclui-se que a DMVD causa alteração na função atrial, principalmente na fase de contração, mesmo em animais assintomáticos e que a ecocardiografia 2D-FTI e o método monoplanar de Simpson são métodos sensíveis e precoces na detecção da disfunção atrial esquerda.
Resumo
Fifty-seven primiparous Angus, Hereford, and crossbred cows were used to study the effect of strategic supplementation on metabolic, productive, and reproductive responses. The experiment had two periods including four phases: prepartum supplementation (phase I; 52 days), early postpartum (phase II; 43 days), pre-mating supplementation (phase III; 21 days), and the last phase including mating, gestation, and lactation until weaning (phase IV; 103 days). Phases I and II were considered as period 1, and phases III and IV were considered as period 2. During phase I (−52±2 days before calving to birth), half of the cows received a supplement (S-), and the rest only grazed native swards (C-). For phase III (59±2 days postpartum [DPP] until mating), cows from the previous treatments (C- or S-) were sorted in two levels of pre-mating supplementation, supplemented (-S) or not supplemented (-C), resulting in four treatment combinations (CC, CS, SC, and SS). The supplement was 1 kg dry matter/cow per day of whole rice bran and 550 mL/cow per day of crude glycerin. In period 1, cows receiving prepartum supplementation increased concentration of cholesterol, glucose, and albumin and decreased concentration of non-esterified fatty acids, beta-hydroxybutyrate, and urea. This improvement in energy balance was reflected in a higher body condition score at calving. Alternatively, in period 2, pre-mating supplementation only increased cholesterol concentration. None of the supplementation periods affected the weaning weight of calves. Prepartum, but not pre-mating, supplementation increased total pregnancy rate. A short prepartum supplementation improves pregnancy rate of primiparous cows managed under extensive production systems. However, there is no additional benefit of supplementation during the pre-mating period.(AU)

Breeding success plays a crucial role in the dynamics of bird populations and yet is the least-studied avian life-stage. Habitat quality during breeding phase may have important implications for survival and conservation, particularly for declining populations in landscapes that have undergone wide-scale human modification. The European Turtle dove is a widespread but fast declining species both in breeding and wintering zones. Reduced food availability isthought to influence breeding success of this game species, but it is not known how agriculture practices could influence breeding Doves, in its high altitude breeding zones. Here, wemonitored Turtle dove nests in apple orchards from early Marsh to the end of October 2015. Nest-tree support, breeding success, and predation were determined and analysed depending on agricultural practices. Compared to prune, cherry and other plantation, apple orchards had the highest overall Turtle doves' nests (85%).
