Clinical History: A 25-year-old female emu (Dromaius novaehollandiae) presented with a 1-week history of lethargy, hyporexia, and recumbency. The bird had lived its entire life at the Winnie Carter Wildlife Center in Texas and had an unremarkable medical history, apart from bilateral congenital blindness. Necropsy Findings: Within the coelomic cavity, the serosal surface of the intestines was coated with a moderate amount of tan to green, mucoid, opaque, foul-smelling fluid (fibrinoheterophilic coelomitis) (Fig. 1). The large intestine was diffusely dilated and filled with dark red fluid and strands of yellow, friable material (fibrin). The mucosa of the small intestine, and most severely the colon, was diffusely roughened, thickened, dull, and covered by a thick yellow to green, opaque, friable diphtheritic membrane (Fig. 2). The distal colon was diffusely, markedly distended and focally contained a 50x7x5 cm, semi-firm accumulation of feces coated by a 4-5 mm diameter layer of tan, friable material (fibrin) located 21 cm from the opening of the cloaca. Clostridioides (Clostridium) difficile is the main causative agent of antimicrobial-related diarrhea in humans and a major pathogen-associated enteric disorder in foals and adult horses. Moreover, studies have suggested that animals are a possible reservoir of toxigenic C. difficile strains for humans. Despite this known importance, the epidemiology of C. difficile infection (CDI) in equine is still largely unknown. Therefore, this study described six cases of equine CDI occurring in Minas Gerais, Brazil, including the characterization of the isolates. All but one equine included in this research developed CDI after antimicrobial therapy, three of which occurred during hospitalization. Coinfection with Salmonella Heidelberg and S. Infantis was detected in three cases, making the antimicrobial treatment challenging. All animals recovered after metronidazole administration. All C. difficile isolates were susceptible to metronidazole and vancomycin, while three were resistant to moxifloxacin and two were resistant to clindamycin. The isolates were classified as RT126 (n = 4), RT078 (n = 1), and RT014/020 (n = 1), all previously reported infecting humans and animals worldwide.

Apesar da importância na saúde animal e humana, a epidemiologia da infecção por C. difficile (ICD) é ainda pouco conhecida. Dessa forma, o presente estudo tem como objetivo caracterizar seis casos de diarreia por C. difficile ocorridos em Minas Gerais, Brasil. Com exceção de um animal, todos os equinos incluídos no presente estudo desenvolveram ICD após antibioticoterapia, três dos quais durante a hospitalização. Coinfecção por Salmonella Heidelberg e S. Salmonellosis is a bacterial disease that affects several domestic animal species, and is commonly diagnosed in cattle, horses, and pigs. This study aimed to describe the epidemiological and pathological findings of eleven cases of enteric salmonellosis and two cases of salmonellosis with pulmonary involvement in cattle in Rio Grande do Sul State, Brazil. Clinical signs included fever, yellow diarrhea, sometimes with blood streaks, and dyspnea, with a clinical course ranging from 1 to 30 days. Eight cases occurred as outbreaks, whereas five cases occurred individually. Risk factors included inadequate handling practices, such as overcrowded facilities and comorbidities, including anaplasmosis. The main gross finding of the enteric presentation was fibrinonecrotic enterocolitis, occasionally associated with button ulcers, mesenteric lymphadenomegaly, splenomegaly, cholecystitis and hepatomegaly. In addition, one steer with a chronic clinical progression presented severe segmental thickening of the ileum, associated with intestinal rupture and peritonitis. In the respiratory system, the main findings were reddened, non-collapsed lungs, with multifocal areas of atelectasis. The main microscopic findings were observed in the small and large intestines, and these were characterized by severe necrosis and mucosal ulceration, associated with marked inflammatory infiltrate of neutrophils and fibrin deposition intermixed by rod-shaped bacterial aggregates, and fibrosis, as well as interstitial pneumonia. Seven cases yielded positive bacterial cultures for Salmonella spp. and three serovars, namely Typhimurium, Dublin, and Panama were identified. Salmonelose é uma doença bacteriana que afeta inúmeras espécies animais, especialmente os bovinos, os equinos e os suínos. O presente estudo teve como objetivo descrever os aspectos epidemiológicos e patológicos de onze casos de salmonelose entérica e dois de salmonelose pulmonar em bovinos no estado do Rio Grande do Sul, Brasil. Os sinais clínicos incluíram febre, diarreia amarelada, por vezes com estrias de sangue, anorexia, perda de peso e dispneia, com curso clínico que variou de um a 30 dias. Em oito casos, a doença ocorreu em forma de surtos e cinco foram individuais. Identificou-se fatores de risco relacionados ao manejo inadequado com os bovinos, como alta lotação, além de comorbidades associadas, como anaplasmose. Os principais achados macroscópicos da forma entérica consistiram em enterocolite fibrinonecrótica, por vezes associada a formação de úlceras botonosas, linfonodos mesentéricos e baço aumentados, colecistite e hepatomegalia. Ainda, um bovino com quadro clínico crônico apresentou acentuado espessamento segmentar da parede do íleo associado a ruptura intestinal e peritonite. Na forma respiratória, os principais achados incluíram pulmões não colabados, avermelhados, com áreas multifocais de atelectasia. Os principais achados microscópicos foram observados no intestino delgado e grosso e foram caracterizados por acentuada necrose e ulceração da mucosa, associada a acentuado infiltrado inflamatório de neutrófilos e deposição de fibrina entremeada por agregados bacterianos cocobacilares e fibrose, além de pneumonia intersticial. Sete casos foram positivos para Salmonella sp. no cultivo bacteriano, com identificação dos sorovares Typhimurium, Dublin e Panama. Ao exame imuno-histoquímico para Salmonella sp. todos os casos apresentaram marcação positiva nos órgãos avaliados.(AU)

Clostridium perfringens is considered one of the main causative agents of superacute enterocolitis, usually fatal in the equine species, due to the action of the ß toxin, and is responsible for causing severe myonecrosis, by the action of the α toxin. The great importance of this agent in the equine economy is due to high mortality and lack of vaccines, which are the main form of prevention, which guarantee the immunization of this animal species. The aim of this study was to evaluate three different concentrations (100, 200 and 400µg) of C. perfringens α and ß recombinant toxoids in equine immunization and to compare with a group vaccinated with a commercial toxoid. The commercial vaccine was not able to stimulate an immune response and the recombinant vaccine was able to induce satisfactory humoral immune response in vaccinated horses, proving to be an alternative prophylactic for C. perfringens infection.(AU)

Purpose To evaluate the effect of N-Acetylcysteine (NAC) in newborn rats submitted to hypoxia and reoxygenation (H/R) conditions in an experimental model of necrotizing enterocolitis. Methods Eight pregnant rats and their 70 cubs were used (5 groups) and exposed to H/R conditions and received NAC at different times. The animals in the H/R groups were placed in a gas chamber (100% CO2) for 10 minutes and then reoxygenated for 10 minutes (100% O2), twice a day for the first three days of life, with a six-hour span between events. On the third day of life, the animals were anesthetized, laparotomized and the intestines were resected. Results The H/R and NAC groups showed changes in the intestinal wall in relation to the number, height and width of the villi when compared to the control group (p 0.0001), but with better preservation of structures in the NAC group. There were no differences between groups regarding the number (%) of mitoses. Conclusion The administration of NAC decreased the lesions in the intestinal wall of rats submitted to H/R, therefore suggesting that this drug can be used to prevent the development of necrotizing enterocolitis in newborns.(AU)

We evaluated 1906 anatomopathological exams of pigs at growing/finishing phases, of which the infectious diseases corresponded to 75.6% of the cases (1,441/1,906). Porcine circovirus type 2 (PCV2) infections were the most frequent, accounting for 51.3% of the cases (739/1,441) with a higher frequency from 2005 to 2007, characterizing an epidemic distribution, with a gradual decline after 2008. Infectious diseases affecting the respiratory system were the second major cause with 30.1% of the cases. Among these, necrotizing bronchiolitis caused by swine Influenza (15.1%, 218/1,441) and bacterial pneumonia (15%, 216/1,441) were the main conditions. Influenza was mostly diagnosed from 2010 to 2013, accounting for 43.1% (167/387) of the cases. After this period, both respiratory infectious diseases were endemic. Digestive system infectious diseases accounted for 10.5% of the diagnoses (151/1,441), with the following main conditions: Salmonella spp. enterocolitis (43.7%, 66/151), Lawsonia spp. proliferative enteropathy (41.7%, 63/151), and Brachyspira spp. colitis (14.6%, 22/151). The latter had a higher incidence from 2012 to 2014 with all cases detected in this period. Polyserositis and bacterial meningitis represented, respectively, 5.8% (84/1,441) and 2.3% (33/1,441) of the cases diagnosed, with a constant endemic character.(AU)

Background: This report describes the occurrence of equine neorickettsiosis (EN) in the northern region of Paraná,southern Brazil. EN is a non-contagious infectious disease caused by the Gram-negative bacterium, Neorickettsia risticii.Equine neorickettsiosis was previously known as Potomac horse fever and monocytic ehrlichiosis. The disease occurspredominantly in the USA and Canada; data relative to EN in Brazil is scarce. The aim of this study was to report the firstcase of putative EN in the state of Paraná due to a combination of IHC and molecular testing.Case: A 2-year-old Quarter Horse was referred to a Veterinary Hospital with episodes of abdominal discomfort, fever,anorexia, tachycardia, and tachypnea. The animal reportedly demonstrated episodes of blackened and fetid diarrhea afterthe ingestion of hay. A treatment was established upon arrival at the veterinary hospital, but the mare died after 12 hoursof monitoring. An autopsy examination performed soon after death revealed severe hyperemia of the mucosa of the cecum and colon, with multifocal cecal erosions and ulcerations. The principal histological lesion observed was necrotizingenterocolitis. Additional significant histopathologic lesions included widespread lymphoid depletion affecting the spleen,tonsils, and lymph nodes. An IHC assay designed to identify the antigens of N. helminthoeca (NH) in formalin fixed paraffin embedded (FFPE) tissues, identified antigens of intralesional neorickettsial organisms within macrophages of themucosa of the colon. Additionally, a PCR assay designed to amplify the 16S rRNA gene of Neorickettsia, amplified thedesired amplicon, but sequencing was frustrating.Discussion: A putative diagnosis of equine neorickettsiosis was established due to the combination of epidemiologicalevidence, pathologic findings, immunohistochemical identification of intralesional antigens of neorickettsial agents, andamplification...

The aim of this study was to report the firstcase of putative EN in the state of Paraná due to a combination of IHC and molecular testing.Case: A 2-year-old Quarter Horse was referred to a Veterinary Hospital with episodes of abdominal discomfort, fever,anorexia, tachycardia, and tachypnea. The animal reportedly demonstrated episodes of blackened and fetid diarrhea afterthe ingestion of hay. A treatment was established upon arrival at the veterinary hospital, but the mare died after 12 hoursof monitoring. An autopsy examination performed soon after death revealed severe hyperemia of the mucosa of the cecum and colon, with multifocal cecal erosions and ulcerations. The principal histological lesion observed was necrotizingenterocolitis. Additional significant histopathologic lesions included widespread lymphoid depletion affecting the spleen,tonsils, and lymph nodes. An IHC assay designed to identify the antigens of N. helminthoeca (NH) in formalin fixed paraffin embedded (FFPE) tissues, identified antigens of intralesional neorickettsial organisms within macrophages of themucosa of the colon. Purpose: To evaluate the functional and structural response of tadalafil effects in the intestinal mucosa, using an experimental model of hypoxia and reoxygenation injury in rats. Methods: The animals were divided into 4 groups: CTL, H/R, H/R+Td and M+Td. The newborn rats allocated in groups H/R, H/R+Td and M+Td were submitted twice a day, to a gas chamber with CO2 at 100% for 10 minutes and afterward reoxygenation with O2 at 98% for 10 minutes, in the three first days of life. Tadalafil dose was given to newborn of group H/R+Td and to the pregnant rat of group M+Td. Histological analysis was made with hematoxylin-eosin technique and oxidative stress through nitrite and nitrate levels and lipid peroxidation. Results: The histological analysis showed a reduction of mucosa alterations in the groups that received tadalafil. In the oxidative stress evaluation, occurred an increase of NO levels and less lipidic peroxidation in the ileum segments that received tadalafil. Conclusion: Tadalafil provides tissue protection when administered independently to both, pregnant or newborns.(AU)

Necropsy, histologic examination, bacterial culture, R. equi and Pneumocystis spp. immunohistochemistry were performed. The foals had a mean age of 60 days and presented respiratory signs (11/20), hyperthermia (10/20), articular swelling (6/20), prostration (4/20), locomotor impairment (3/20) and diarrhea (3/20), among others. The main lesions were of pyogranulomatous pneumonia, seen in 19 foals, accompanied or not by pyogranulomatous lymphadenitis (10/20) and pyogranulomatous and ulcerative enterocolitis (5/20). Pyogranulomatous osteomyelitis was seen in 3 foals, one of which did not have pulmonary involvement. There was lymphoplasmacytic (4/20), lymphoplasmacytic and neutrophilic (1/20) or pyogranulomatous arthritis (1/20), affecting multiple or singular joints. Immunohistochemistry revealed to be a valuable tool for the detection of R. equi, confirming the diagnosis in all cases. Furthermore, pulmonary immunostaining for Pneumocystis spp. demonstrates that a coinfection with R. equi and this fungal agent is a common event in foals, seen in 13 cases.

Resumo
A 10-day old foal presented with a history of acute recumbency and generalized weakness, that progressed to seizure episodes and death. Post mortem examination revealed necrotizing and purulent omphalophlebitis and fibrinopurulent meningoencephalomyelitis. Salmonella Typhimurium was isolated from the central nervous system and determined to be the cause of the meningoencephalomyelitis. Due to the lack of evidence of gastrointestinal disease, the umbilical cord was considered the most likely portal of entry of the bacteria. The isolated S. Typhimurium was resistant to ampicillin and cephalotin, and partially resistant to enrofloxacin. These drugs are commonly used in the treatment of salmonellosis. This is the first report of S. Typhimurium affecting the brain and spinal cord of a foal. Salmonellosis should be considered a differential diagnosis in foals with neurologic signs, even in the absence of enterocolitis.(AU)
Um potro de 10 dias de idade foi admitido com histórico de decúbito e fraqueza generalizada aguda, com progressão para episódios de convulsão e morte. A avaliação post mortem demonstrou onfalite necropurulenta e meningoencefalomielite fibrinopurulenta. Salmonella Typhimurium foi isolada do material purulento coletado do sistema nervoso central. A ausência de diarreia e lesões entéricas associadas à presença de onfalite sugerem que o umbigo foi possivelmente a porta de entrada do agente. O isolado de S. Typhimurium apresentou resistência a cefalotina e ampicilina e resistência intermediária a enrofloxacina, drogas utilizadas para o tratamento de salmonelose. Este é o primeiro relato de S. Typhimurium levando a alterações encefálicas e medulares em potro. A salmonelose deve ser considerada, portanto, como diagnóstico diferencial em potros neonatos com alterações neurológicas, mesmo na ausência de enterocolite.(AU)

Background: Aspergillus spp. are dimorphic fungus widely distributed in the environment, including in soil, dust and decaying vegetation. Clinical signs of aspergillosis in horses including rhinitis, pneumonia, guttural pouch mycosis, keratomycosis, endometritis, abortions and systemic involvement. In addition, horses with a history of enterocolitis may be predisposed to pulmonary or systemic mycotic infection. However, reports about systemic aspergillosis in horses are restricted to infections by A. fumigatus and A. niger. There have been no reports of systemic infection caused by A. flavus in horses or in other domestic species. Thus, the objective of this work was to describe the epidemiological, clinical and pathological aspects of systemic infection by Aspergillus flavus in a mare.Case: A 3-year-old pregnant mare of the Manga Larga, had signs of colic two days prior to admission after grazing in a landfill area to which it had free access. The owner observed remains of plastic bags in the stool. Clinically, there was dehydration, apathy, ocular mucosal congestion, oral cyanosis, reluctance to move, diarrhea, fever, drooling and tachypnea. Due to its clinical condition, the animal was referred to the surgical center for exploratory laparotomy, where compaction in the colon and cecum was verified. Enterotomy and enterolith removal were performed in the small colon region. The mare died after eight days of hospitalization, and necropsy was performed. Macroscopically disseminated lesions were observed in the small colon, stomach, kidneys, lungs, heart and brain. Fragments of tissues from organs in the abdominal and thoracic cavities, as well as from the central nervous system, were collected, fixed in 10% buffered formalin solution, and subsequently routinely processed and stained with hematoxylin and eosin and special histochemical stains to visualize the infectious agent and its morphological characteristics.[...]

Purpose: To evaluate the effect of remote ischemic preconditioning (r-IPC) administered to pregnant rats, in the ileum of newborn rats subjected to hypoxia and reoxygenation. Methods: We used three pregnant rats and their newborn rats distributed in three groups: 1) Control (C) - Newborn rats born from a pregnant rat which did not undergo any intervention; 2) Hypoxia-Reoxygenation (H/R) - Newborn rats born from a pregnant rat which did not undergo any intervention, and were subjected to hypoxia-reoxygenation; 3) Remote Ischemic Preconditioning (r-IPC) - newborn rats born from a pregnant rat which was subjected to remote ischemic preconditioning twenty-four hours before giving birth and the newborn rats were subjected to hypoxia-reoxygenation. Segments of ileum were prepared for histological analysis by HE and immunohistochemistry by the Ki67 to evaluate cell proliferation, crypt depth and villus height and evaluation of apoptosis by cleaved caspase-3. Results: The intensity of the lesions was lower in the r-IPC than in the H/R group, showing significant difference (p 0.01). The r-IPC group showed a higher proliferative activity compared to the H/R group (p 0.01), with deeper crypts (r-IPC > H/R - p 0.05), and higher villi, showing significant difference (r-IPC > H/R - (p 0.01). The occurrence of apoptosis in the H/R group was lower in comparison to groups C and r-IPC, with significant difference (H/R r-IPC; p 0.05). Conclusion: The remote ischemic preconditioning applied to the pregnant rat protected the ileum of newborn rats subjected to hypoxia and reoxygenation, with decreased intensity of the lesions in the ileum mucosa and preservation of proliferative activity, keeping the villus height and crypt depth similar to group C.(AU)

The importance of Clostridium perfringens and C. difficile for most wild animal species remains unclear. This study aimed to isolate and genotype C. perfringens and C. difficile in stool samples from free-living and captive capuchin monkeys (Sapajus flavius and Sapajus libidinosus) in Brazil. Ten free-living S. flavius and 14 captive S. libidinosus were sampled for this study. To isolate C. difficile, stool samples were inoculated on plates containing cycloserine-cefoxitin fructose agar supplemented with horse blood and sodium taurocholate. Two different protocols for C. perfringens isolation were tested: direct plating onto selective agar and enrichment in brain heart infusion (BHI) broth followed by plating onto selective agar. C. difficile was not detected in the present study. The results were identical for both protocols tested for isolation of C. perfringens. Four samples (16.7%) were positive for C. perfringens type A, including one sample from a free-living animal (4.2%) and three from captive animals (12.5%), meaning there was no significant difference between these two groups. C. perfringens isolates were negative for all additional virulence factors evaluated, including enterotoxin encoding-gene (cpe) and beta-2 encoding-gene (cpb2). These results suggested that C. perfringens type A is found in the microbiota of capuchin monkeys, although it is less frequent than previously reported in domestic animals.
A importância de Clostridium perfringens e C. difficile para a maioria das espécies silvestres ainda não está clara. O objetivo do presente estudo foi isolar e genotipar C. perfringens e C. difficile em amostras de fezes de macacos-prego (Sapajus flavius e Sapajus libidinosus) de vida livre e criados em cativeiros no Brasil. Dez S. flavius de vida livre e 14 S. libidinosus de cativeiro foram incluídos no presente estudo. Para isolamento de C. difficile, as amostras de fezes foram inoculadas em agar cicloserina-cefoxitina frutose, suplementado com sangue e taurocolato de sódico. Para isolamento de C. perfringens, foram testados dois protocolos: plaqueamento direto em ágar seletivo e enriquecimento em caldo seguido de plaqueamento em ágar seletivo. C difficile não foi detectado no presente estudo. Os resultados foram idênticos para ambos os protocolos testados para isolamento de C. perfringens, resultando em quatro animais (16,7%) positivos para C. perfringens tipo A. Destes, uma amostra era de um animal de vida livre (4,2%) e três de animais de cativeiro (12,5%), não havendo diferença entre esses dois grupos. Os isolados de C. perfringens foram negativos para todos os fatores de virulência adicionais avaliados, incluindo o gene codificador de enterotoxina (cpe) e o gene codificador beta-2 (cpb2). O presente estudo sugere C. perfringens tipo A como parte da microbiota de macacos-prego, embora esse agente seja menos frequente como comensal, do que relatado anteriormente, em animais domésticos.
