Resumo

Background: In the literature, there are a few descriptions of epididymis neoplasia in domestic animals, especially considering primary tumors. In the few reports found in literature, the lesions were a consequence of the invasion of testicular or paratesticular neoplasia, as a papillar carcinoma in a dog's and a bull's epididymis, and mesenchymal tumors - fibrome/ fibrosarcoma, leiomyoma/leiosarcome. On the other hand, mast cell tumors are the second most prevalent neoplasia in dogs in Brazil, affecting especially the skin. The aim of this report is to describe for the first time a low malignancy mast cell tumor in a mixed-breed dog's epididymis, without metastasis or recurrence in a 2-year follow-up period. Case: A 10-year-old male mixed-breed dog was presented for pre-surgical evaluation for elective orchiectomy. In the physical examination, an increase in the volume of approximately 2 cm with an irregular appearance was identified on palpation in the cranial pole of the left testis. In the trans surgical period, an increase in testicular volume (4 cm long x 2 cm wide) was observed, with a firm consistency in the region of the vas deferens with macroscopic changes in the region. The testis was sectioned, and the fragments were sent for histopathological evaluation in 10% buffered formaldehyde. There was a fairly cellular circumscribed neoplastic infiltrate, distributed in a sheet and separated by fibrovascular stroma, and rounded neoplastic cells with a moderate amount of basophilic cytoplasmic granulation, and discrete anisocytosis and anisokaryosis. The nuclei were rounded with vesicular chromatin with 1 or 2 distinct nucleoli. No mitosis figures were observed in 10 high power fields (400x). Few eosinophils were distributed throughout the neoplastic cell population. Immunohistochemistry demonstrated immunostaining for KIT protein with perimembranous staining in 95% of neoplastic mast cells, giving a KIT 1 pattern. There was no positive nuclear staining for Ki67 in any cell of the histological sections examined. A grade II mast cell tumor (low grade of malignancy) was diagnosed. After diagnosis, the animal underwent radiographic evaluation of the chest and abdominal ultrasound, and a new physical inspection in search of nodules, plaques, skin lesions, or subcutaneous masses. There were no metastases in the thorax and abdominal cavity, nor physical alterations, and it can be inferred that the epididymis was the primary site of the mast cell tumor. After 2 years of orchiectomy, there were no recurrences, and no chemotherapy treatment was performed. Discussion: Extracutaneous mast cell tumors are uncommon in animals, but have been reported in oral and nasal mucosa, nasopharynx, larynx, trachea, intestine, visceral lymph nodes, spleen, liver, spinal cord, intestine, ureter, conjunctiva, lung and more recently in tear gland of the third eyelid. However, in the authors' assessment, this is the first description of mast cell tumor in the epididymis in dogs. The diagnosis was established by histopathological examination, which revealed a grade II epididymal mast cell tumor and immunohistochemical evaluation (KIT and Ki-67) as being of low aggressiveness. The diagnosis of a primary tumor was confirmed since the staging was established after the histopathological diagnosis, involving chest radiography, abdominal ultrasound, cutaneous evaluation in search of nodules, plaques, cutaneous and subcutaneous lesions, and did not reveal other abnormalities or metastases not identified in the preoperative evaluation. In addition, immunostaining with KIT and Ki-67 reaffirmed the low degree of malignancy and the potential for metastases, which can be observed by the asymptomatic follow-up of the patient 2 years after the surgical excision.

Resumo

The family Potamotrygonidae are the only species of stingrays restricted to fresh water and located exclusively in South America. The objective of this research was to analyze the morphological aspects and germ cells of the male reproductive tract of Potamotrygon amandae. The samples were fixed in 10% formalin, and then dehydrated in an ascending ethanol series (70 to 100%). To carry out light microscopy analyses, they were embedded in paraffin, cut and stained; as for scanning electron microscopy analyses, the samples were dried, glued in metallic bases and metalized. The gross morphology consisted of the following paired organs: testis, epididymis, deferent duct, Leydig gland, seminal vesicle, clasper, and the clasper gland. Microscopically, several stages of spermatogenesis were observed in the testis, occurring in spherical follicles, similar to other stingrays. The epididymis was formed by one duct subdivided in various tubules. The deferent ducts were continuous with the epididymis, and the lumen was full of spermatozoa. The Leydig glands consisted of glandular units with eosinophilic content in the lumen of some, and the deferent ducts ran parallel to the ventral portion. The seminal vesicles possessed numerous compartments to store the sperm, with a wall similar to a hive, and the lumen was full of spermatozoa. Alcian Blue (AB) and Periodic Schiff-Acid (PAS) performed in the Leydig Gland, deferens ducts and seminal vesicle was positive only in the connective tissue, the cilia were PAS+ and the nuclei stained weakly for AB. The clasper gland was composed of unit glands and was covered with striated muscle externally. It stained very well with Periodic Schiff-Acid. The morphological aspects of the male reproductive tract of Potamotrygon amandae were similar to other stingrays.(AU)

The mule is a sterile hybrid domestic animal that results from the breeding of a male donkey with a female horse, understanding the reproductive biology of these species is very critical. The goal of this paper was to perform a comparative and more accurate histomorphometric of the testicles in Barb horse, donkeys and mules. Microscopic examinations and histological description were carried on genital tract of horses, donkeys and mules healthy and mature; this study was conducted during April-May 2018. The histological and the morphological results shows a similarity between the two equine species and the infertile hybrid for the testicles, the epididymis and the vas deferens. However, the difference was presented on the morphometric data; vas deferens was more voluminous in the horse and donkey than a mule. Moreover, the differences were significantly higher for the surface of the seminiferous tubules and for the epididymis. The lumen of the seminiferous tubules in mule was significantly higher than in the horse and donkey. Absence of gametes in the epididymal cavity and lower number of gametes in the mule. Furthermore, we have noted the presence of spermatozoa in one mule 16.67%. Therefore, the mule could complete development of spermatogenesis.(AU)

Paternal programming is the concept that the environmental signals from the sire's experiences leading up to mating can alter semen and ultimately affect the phenotype of resulting offspring. Potential mechanisms carrying the paternal effects to offspring can be associated with epigenetic signatures (DNA methylation, histone modification and non-coding RNAs), oxidative stress, cytokines, and the seminal microbiome. Several opportunities exist for sperm/semen to be influenced during development; these opportunities are within the testicle, the epididymis, or accessory sex glands. Epigenetic signatures of sperm can be impacted during the pre-natal and pre-pubertal periods, during sexual maturity and with advancing sire age. Sperm are susceptible to alterations as dictated by their developmental stage at the time of the perturbation, and sperm and seminal plasma likely have both dependent and independent effects on offspring. Research using rodent models has revealed that many factors including over/under nutrition, dietary fat, protein, and ingredient composition (e.g., macro- or micronutrients), stress, exercise, and exposure to drugs, alcohol, and endocrine disruptors all elicit paternal programming responses that are evident in offspring phenotype. Research using livestock species has also revealed that sire age, fertility level, plane of nutrition, and heat stress can induce alterations in the epigenetic, oxidative stress, cytokine, and microbiome profiles of sperm and/or seminal plasma. In addition, recent findings in pigs, sheep, and cattle have indicated programming effects in blastocysts post-fertilization with some continuing into post-natal life of the offspring. Our research group is focused on understanding the effects of common management scenarios of plane of nutrition and growth rates in bulls and rams on mechanisms resulting in paternal programming and subsequent offspring outcomes. Understanding the implication of paternal programming is imperative as short-term feeding and management decisions have the potential to impact productivity and profitability of our herds for generations to come.(AU)

A espermiogênese é o processo final da espermatogênese no qual a espermátide se transforma em espermatozoide. Durante esse processo podem ocorrer alterações espermáticas, especialmente no acrossoma, na morfologia da cabeça, na condensação da cromatina e na formação dos vacúolos nucleares. A diferenciação entre os quadros clínicos é feita com repetições dos exames andrológicos. A avaliação morfológica indica a situação da espermiogênese nas 4 semanas anteriores a coleta do sêmen. Com os resultados do exame andrológico é possível identificar a qualidade seminal naquele período. O presente artigo é o relato de caso de um touro jovem doador de sêmen com 26 meses de idade, no início do regime de coletas de sêmen em um Centro de Coleta e Processamento de Sêmen no Sul do Brasil. A produção espermática deste animal foi avaliada durante 12 meses de coleta, apresentando sempre morfologia espermática muito alterada e motilidade média de 54% no exame imediato. Em 13 coletas seminais, o ejaculado deste animal apresentou em média 86% de defeitos maiores, 10% de defeitos menores e 96% de defeitos totais. Os defeitos maiores, que são os que possuem maior efeito na fertilidade, estão diretamente ligados a espermiogênese e os defeitos menores, que possuem menor efeito na fertilidade, são ocasionados principalmente durante o trânsito pelo epidídimo. O quadro clínico desse animal demonstrou a importância do exame morfológico para avaliar a espermatogênese e principalmente o processo de diferenciação final da espermátide em espermatozoide. Casos como este devem ser descritos como espermiogênese alterada e o reprodutor deve ser afastado da reprodução, após descartar alterações morfológicas devido a degeneração testicular grave.(AU)


Spermiogenesis is the final process of spermatogenesis in which the spermatid transforms into sperm. During this process, sperm alterations may occur, especially in the acrosome, head morphology, chromatin condensation and formation of nuclear vacuoles. Differentiation between clinical conditions is made with repetitions of andrological exams. The morphological evaluation indicates the status of spermiogenesis in the 4 weeks prior to semen collection. With the results of the breeding soundness examination, it is possible to identify the seminal quality in that period. The present article is a case report of a young 26-month-old semen donor bull, that started semen collection routines at a Semen Collection and Processing Center in southern Brazil. The sperm production of this animal was evaluated during 12 months of collection, always showing very altered sperm morphology and average motility of 54% in the immediate examination. In 13 seminal collections, the ejaculate of this animal presented an average of 86% of major defects, 10% of minor defects and 96% of total defects. Major defects, which have the greatest impact on fertility, are directly linked to spermiogenesis and minor defects, which have less effect on fertility, are mainly caused during transit through the epididymis. The clinical conditions of this animal demonstrated the importance of the morphological examination to evaluate spermatogenesis and especially the process of final differentiation of the spermatid into spermatozoa. Cases like this should be described as altered spermiogenesis and the bull should be withdrawn from breeding, after ruling out morphological changes due to severe testicular degeneration.(AU)

Resumo

We report a case of urethral obstruction due to seminal vesiculitis in a Dorper sheep, with symptoms of anuria, rectal prolapse, orchitis/epididymitis, and uroperitoneum and biochemical tests indicating severe azotemia. The animal died due to advanced azotemia, and necropsy revealed kidneys with cortical and medullary necrosis, pyelonephritis of renal calyces, hydronephrosis, ruptured and necrotic bladder, and vesicular, bulbourethral, and ampoule accessory sex glands. There was prostate hyperplasia that revealed a large amount of pus in the cross section, which was also observed bilaterally in the epididymis and right testis. Morphotintorial and biochemical analyses of bacteria obtained from microbiological culture revealed Corynebacterium sp. and Escherichia coli. Infection, hyperplasia, and abscessation of accessory sex glands caused urethral compression, resulting in an obstructive condition, similar to urolithiasis, in addition to bacteremia. Hyperplastic seminal vesiculitis, although rare, must be included among the differential diagnoses of obstructive processes in the urinary tract of sheep.


Relata-se um caso de obstrução uretral decorrente de vesiculite seminal em carneiro da raça Dorper, com quadro de anúria, prolapso retal, orquite/epididimite e uroperitôneo, com exames bioquímicos indicativos de severa azotemia. Devido ao quadro avançado de azotemia, o animal faleceu e, na necropsia, foi observado:: rins com necrose de cortical e medular, pielonefrite dos cálices renais e hidronefrose, bexiga rompida e necrosada, assim como glândulas sexuais acessórias vesiculares, bulbouretrais e ampola. Havia hiperplasia de próstata que, ao corte transversal, revelou grande quantidade de pus, que também foi observado bilateralmente no epidídimo e no testículo direito. As análises morfotintoriais e bioquímicas das bactérias obtidas a partir do cultivo microbiológico foram compatíveis com Corynebacterium sp. e Escherichia coli. A infecção, a hiperplasia e a abscedação das glândulas sexuais acessória, causaram compressão uretral, levando a um quadro obstrutivo, semelhante à urolitíase, somado à bacteremia. A vesiculite seminal hiperplásica, apesar de rara, deve ser incluída entre os diagnósticos diferenciais dos processos obstrutivos do trato urinário de carneiros.

This study was carried out to investigate the morphological and histological structures of the testis, epididymis and vas deferens of the Al-Ahsa native rooster (ANR). There were two types of ANR; the brown feather one with light yellow shank and the black feather one with grey or dark grey shank. Their body weight was 1840.88± 92.13 g and 1555.66± 82.83g, respectively. The morphology of the testes showed that the black rooster has larger testes than the brown rooster and there was asymmetry in size between the right and left testis in both. They were grey yellowish in color and oval-shaped, situated in the abdomen dorsal to the proventriculus, the liver and the gizzard, cranial to the lungs, caudal to the abdominal sac and ventral to the kidneys. The histology of the testes revealed the capsule, the different cells of the lining epithelium of the seminiferous tubules and the interstitial tissue. The morphology of the epididymis was revealed pseudostratified columnar epithelium, light brown in color with c to L-shaped, located cranial to the testis and extended caudally to continue with vas deferens. The latter has columnar epithelium, light grey in color, run caudally medial to the kidneys and opened in the cloaca.(AU)

Abstract Cadmium (Cd) is one of the major toxicants, which affects human health through occupational and environmental exposure. In the current study, we evaluated the protective effects of morel mushrooms against Cd-induced reproductive damages in rats. For this purpose, 30 male rats were divided into 6 groups (n=5/group), the first group served as the control group, second group was treated with an intraperitoneal (i.p) injection of 1 mg/kg/day of Cd. Third and fourth groups were co-treated with 1 mg/kg/day of Cd (i.p) and 10 and 20 mg/kg/day of morel mushroom extract (orally) respectively. The final 2 groups received oral gavage of 10 and 20 mg/kg/day of morel mushroom extract alone. After treatment for 17 days, the animals were euthanized, and testes and epididymis were dissected out. One testis and epididymis of each animal were processed for histology, while the other testis and epididymis were used for daily sperm production (DSP) and comet assay. Our results showed that Cd and morel mushrooms have no effect on animal weight, but Cd significantly decreases the DSP count and damages the heritable DNA which is reversed in co-treatment groups. Similarly, the histopathological results of testes and epididymis show that morel mushrooms control the damage to these tissues. Whereas the morel mushroom extract alone could enhance the production of testosterone. These results conclude that morel mushrooms not only control the damage done by Cd, but it could also be used as a protection mechanism for heritable DNA damage.


Resumo O cádmio (Cd) é um dos principais tóxicos, que afeta a saúde humana por meio da exposição ocupacional e ambiental. No presente estudo, avaliamos os efeitos protetores dos cogumelos morel contra os danos reprodutivos induzidos pelo Cd em ratos. Para tanto, 30 ratos machos foram divididos em 6 grupos (n = 5 / grupo); o primeiro grupo serviu de controle, o segundo grupo foi tratado com injeção intraperitoneal (i.p) de 1 mg / kg / dia de Cd. O terceiro e o quarto grupos foram cotratados com 1 mg / kg / dia de Cd (i.p) e 10 e 20 mg / kg / dia de extrato de cogumelo morel (por via oral), respectivamente. Os dois grupos finais receberam gavagem oral de 10 e 20 mg / kg / dia de extrato de cogumelo morel sozinho. Após o tratamento por 17 dias, os animais foram sacrificados e os testículos e o epidídimo foram dissecados. Um testículo e epidídimo de cada animal foram processados para histologia, enquanto o outro testículo e epidídimo foram usados para produção diária de esperma (DSP) e ensaio cometa. Nossos resultados mostraram que os cogumelos Cd e morel não têm efeito sobre o peso do animal, mas o Cd diminui significativamente a contagem de DSP e danifica o DNA hereditário, que é revertido em grupos de cotratamento. Da mesma forma, os resultados histopatológicos dos testículos e do epidídimo mostram que os cogumelos morel controlam os danos a esses tecidos. Considerando que o extrato de cogumelo morel sozinho pode aumentar a produção de testosterona. Esses resultados concluem que os cogumelos morel não apenas controlam os danos causados pelo Cd, mas também podem ser usados como um mecanismo de proteção para danos hereditários ao DNA.

Arsenic exposure is a global health concern. This toxic metalloid is ubiquitous in the environment and contaminates food and drinking water. Once ingested, it undergoes a complex metabolic process within the body, which contributes to its accumulation and reactivity. Arsenic toxicity stems from the induction of oxidative stress, inhibition of thiol-containing proteins, and mimicry of inorganic phosphates. Arsenic poisoning is associated with the development of reproductive disorders. In males, arsenic causes a reduction in testicular weight and alterations in steroidogenesis and spermatogenesis. Moreover, it reduces the number and quality of spermatozoa harvested from the cauda epididymis. The mitochondria are targets of arsenic toxicity because of the production of free radicals and their high content of cysteine-rich proteins and fatty acids. Mitochondrial dysfunction may contribute to reproductive disorders because this organelle is crucial for controlling testicular and epididymal events related to sperm production and maturation. All of these alterations mediated by arsenic exposure contribute to the failure of male reproductive competence by reducing gamete viability. This review describes the potential mechanisms of arsenic toxicity, its detrimental effects on male reproductive organs, and consequences on sperm fertility.(AU)

The present study investigated the toxic effect of a mixture of three pesticides (cypermethrin, mancozeb, and metalaxyl) on reproduction and oxidative stress parameters in male Wistar rats. Animals were treated at doses 1/60, 1/30, and 1/10 LD50 of each pesticide daily in the diet for 08 weeks. At the end of the treatment period, animals were sacrificed by decapitation. The results indicate a decrease in the absolute weight of testes and epididymis, the serum of testosterone hormone, and cholesterol levels. These parameters were significant reduced in males exposed to the mixed pesticides. A reduction in sperm concentration, motility, and viability also was observed. Besides, the ingestion of mixed pesticides at all three concentrations caused a significant decrease in GSH, GPx levels and an increase in MDA levels compared to the control group. This was accompanied by histopathological changes in testis and epididymis of rats such as seminiferous tubules degeneration, decreasing number of spermatogenic cells, edema, expansion of interstitial spaces, cell necrosis, and reducing the diameter of the epididymal tube compared to the control group. Thus, we strongly suggest that the mixture of pesticides causes damages to the male reproductive system.(AU)

Considerando que o extrato de cogumelo morel sozinho pode aumentar a produção de testosterona. Esses resultados concluem que os cogumelos morel não apenas controlam os danos causados pelo Cd, mas também podem ser usados como um mecanismo de proteção para danos hereditários ao DNA.

The present study investigated the toxic effect of a mixture of three pesticides (cypermethrin, mancozeb, and metalaxyl) on reproduction and oxidative stress parameters in male Wistar rats. Animals were treated at doses 1/60, 1/30, and 1/10 LD50 of each pesticide daily in the diet for 08 weeks. At the end of the treatment period, animals were sacrificed by decapitation. The results indicate a decrease in the absolute weight of testes and epididymis, the serum of testosterone hormone, and cholesterol levels. These parameters were significant reduced in males exposed to the mixed pesticides. A reduction in sperm concentration, motility, and viability also was observed. Besides, the ingestion of mixed pesticides at all three concentrations caused a significant decrease in GSH, GPx levels and an increase in MDA levels compared to the control group. This was accompanied by histopathological changes in testis and epididymis of rats such as seminiferous tubules degeneration, decreasing number of spermatogenic cells, edema, expansion of interstitial spaces, cell necrosis, and reducing the diameter of the epididymal tube compared to the control group. Thus, we strongly suggest that the mixture of pesticides causes damages to the male reproductive system.

Cadmium (Cd) is one of the major toxicants, which affects human health through occupational and environmental exposure. In the current study, we evaluated the protective effects of morel mushrooms against Cd-induced reproductive damages in rats. For this purpose, 30 male rats were divided into 6 groups (n=5/group), the first group served as the control group, second group was treated with an intraperitoneal (i.p) injection of 1 mg/kg/day of Cd. Third and fourth groups were co-treated with 1 mg/kg/day of Cd (i.p) and 10 and 20 mg/kg/day of morel mushroom extract (orally) respectively. The final 2 groups received oral gavage of 10 and 20 mg/kg/day of morel mushroom extract alone. After treatment for 17 days, the animals were euthanized, and testes and epididymis were dissected out. One testis and epididymis of each animal were processed for histology, while the other testis and epididymis were used for daily sperm production (DSP) and comet assay. Our results showed that Cd and morel mushrooms have no effect on animal weight, but Cd significantly decreases the DSP count and damages the heritable DNA which is reversed in co-treatment groups. Similarly, the histopathological results of testes and epididymis show that morel mushrooms control the damage to these tissues. Whereas the morel mushroom extract alone could enhance the production of testosterone. These results conclude that morel mushrooms not only control the damage done by Cd, but it could also be used as a protection mechanism for heritable DNA damage.


O cádmio (Cd) é um dos principais tóxicos, que afeta a saúde humana por meio da exposição ocupacional e ambiental. No presente estudo, avaliamos os efeitos protetores dos cogumelos morel contra os danos reprodutivos induzidos pelo Cd em ratos. Para tanto, 30 ratos machos foram divididos em 6 grupos (n = 5 / grupo); o primeiro grupo serviu de controle, o segundo grupo foi tratado com injeção intraperitoneal (i.p) de 1 mg / kg / dia de Cd. O terceiro e o quarto grupos foram cotratados com 1 mg / kg / dia de Cd (i.p) e 10 e 20 mg / kg / dia de extrato de cogumelo morel (por via oral), respectivamente. Os dois grupos finais receberam gavagem oral de 10 e 20 mg / kg / dia de extrato de cogumelo morel sozinho. Após o tratamento por 17 dias, os animais foram sacrificados e os testículos e o epidídimo foram dissecados. Um testículo e epidídimo de cada animal foram processados para histologia, enquanto o outro testículo e epidídimo foram usados para produção diária de esperma (DSP) e ensaio cometa. Nossos resultados mostraram que os cogumelos Cd e morel não têm efeito sobre o peso do animal, mas o Cd diminui significativamente a contagem de DSP e danifica o DNA hereditário, que é revertido em grupos de cotratamento. Da mesma forma, os resultados histopatológicos dos testículos e do epidídimo mostram que os cogumelos morel controlam os danos a esses tecidos. Os hermafroditas verdadeiros são animais com intersexualidade, nos quais as gônadas masculinas e femininas se desenvolvem simultaneamente no mesmo indivíduo. Este trabalho relata um caso de hermafrodita na espécie suína após descarte de uma fêmea por diagnóstico de anestro. Tratava-se de uma fêmea com 222 dias de idade e 150 kg a qual havia sido selecionada para reprodução e permanecia em anestro. O indivíduo foi descartado e enviado para o abate em frigorífico sob inspeção federal. Após o abate foi observado no sistema genital aspectos anatômicos sugestivos de hermafroditismo. O sistema genital foi coletado, inspecionado macroscopicamente e conservado em formol a 10% para avaliação microscópica. Durante a análise macroscópica foi verificada a presença de vulva e vagina, contendo um pênis rudimentar. O útero, a cérvix, os cornos uterinos e oviduto estavam presentes e com ausência de anormalidade. Havia um ovário esquerdo com aspectos morfológicos de ciclicidade e folículos contendo oócitos de alta qualidade. À direita, entretanto, foi constatada a presença de um testículo, epidídimo e plexo pampiniforme. A avaliação histopatológica comprovou os achados macroscópicos, demonstrando um ovário contendo folículos, corpos lúteos e cistos foliculares; plexo pampiniforme; testículo e epidídimo com aspecto histológico normal, porém, com ausência de células espermatogênicas. Assim, com base nos achados macro e microscópicos descritos nesse relato de caso, o indivíduo foi considerado como hermafrodita verdadeiro.(AU)


True hermaphrodites are intersexual animals, in which both male and female gonads develop simultaneously in the same individual. This paper reports a case of a true hermaphrodite in the swine species after its culling for anestrus. The specimen was a female of 222 days of age and 150 kg of body weight, that was selected for reproduction and was permanently in anestrus. This individual was culled and sent to an abattoir under federal inspection. After slaughtering, it was observed in the reproductive tract anatomical aspects that suggested hermaphroditism. The reproductive tract was collected, evaluated macroscopically, and preserved in formaldehyde at 10 % to be evaluated by light microscopy. During the macroscopic analysis, the presence of a vulva and a vagina with a rudimentary penis was detected. The uterus, the cervix, uterine horns, and the oviduct were present and abnormalities were not detected. The left ovary exhibited morphological signs of cyclicity and contained follicles with high quality oocytes. On the right side, however, it was observed a testicle, epididymis, and a pampiniform plexus. The histopathological assessment corroborated the macroscopic findings, which demonstrated that the ovary had follicles, corpus luteum, and follicular cysts, and pampiniform plexus, testicle and epididymis were histologically normal, though with the absence of spermatogenic cells. Thus, based on the macro and microscopic findings described herein, the individual was considered a true hermaphrodite.(AU)

O estudo das lesões testiculares em felinos é essencial no diagnóstico das doenças reprodutivas, uma vez que as gônadas podem apresentar alterações importantes, mesmo quando macroscopicamente normais. Assim, este trabalho foi desenvolvido com o objetivo de avaliar as principais lesões testiculares em gatos adultos submetidos à orquiectomia eletiva, determinando as lesões de maior frequência. Para isso, foram utilizados 25 gatos machos, adultos, com idade entre um e dois anos. Todos os animais eram provenientes do Programa de Controle Populacional de Animais de Estimação Pelo Método de Esterilização Cirúrgica, do Hospital Veterinário da Universidade Federal de Uberlândia (HOVET-UFU). Após a orquiectomia, os testículos foram coletados e enviados ao Setor de Patologia Animal do HOVET-UFU, onde foram separados dos seus respectivos epidídimos. Foram obtidas as dimensões de comprimento, altura, largura, peso e volume de cada testículo. Esses foram então fixados em formalina 10%, por 24 horas, e clivados nas regiões cranial, média e caudal. Os fragmentos foram submetidos ao processamento para inclusão em parafina e coloração em hematoxilina e eosina. Mesmo sem alterações macroscópicas significativas, a maioria dos gatos avaliados no estudo exibiu alguma lesão testicular, sendo a degeneração testicular e a fibrose intersticial as mais comuns. Desta forma, destaca-se a importância da histopatologia no diagnóstico de alterações testiculares em animais considerados saudáveis.


The study of testicular lesions in felines is essential in the diagnosis of reproductive diseases, since, the gonads can present important alterations, even when they are macroscopically normal. Thus, the present work aimed to evaluate the main testicular injuries in adult cats submitted to elective orchiectomy, determining the most frequent ones. For this, 25 adult male cats, aged between one and two years, were used. All animals came from the Pet Population Control Program through the Surgical Sterilization Method of the Veterinary Hospital of the Federal University of Uberlândia (HOVET-UFU). After the orchiectomy, the testicles were collected and sent to the Animal Pathology Sector of HOVET-UFU, where they were separated from their respective epididymis. The dimensions of length, height, width, weight, and volume of each testicle were obtained. These were then fixed in 10% formalin for 24 hours, and cleaved in the cranial, middle, and caudal regions. The fragments were submitted to processing for inclusion in paraffin and staining by hematoxylin and eosin. Even without significant macroscopic changes, most cats evaluated in the present study showed some testicular damage, with testicular degeneration and interstitial fibrosis being the most common. Therefore, the importance of histopathology in the diagnosis of testicular alterations in animals considered healthy is highlighted.

O gato doméstico é a única espécie da família Felídea sem risco ou iminência de extinção, diferente da maior parte dos felinos selvagens. Desta forma, o desenvolvimento e aprimoramento de diferentes biotécnicas reprodutivas, são essenciais para a manutenção da qualidade reprodutiva, tendo em vista a preservação de espécies mais vulneráveis. Além disso, as biotécnicas do sêmen são para as tecnologias reprodutivas, como a inseminação artificial (IA) e a fertilização in vitro (FIV). Sendo assim, o objetivo deste compilado bibliográfico foi abordar as principais técnicas de colheita, análise e preservação de sêmen/espermatozoides felino, assim como o uso dessas células em IA e FIV. Para a colheita do sêmen felino, diferentes métodos têm sido aplicados: ejaculação farmacológica, eletroejaculação e vagina artificial. Em caso de óbito do reprodutor, os espermatozoides recuperados do epidídimo também apresentam viabilidade reprodutiva. Ademais, a cinética espermática avaliada pelo sistema CASA, a morfologia e a morfometria são as principais análises que demonstram a qualidade espermática e refletem na fertilidade do ejaculado. O sistema CASA também avalia a trajetória individual de cada espermatozoide, que ao se agrupar em clusters, demonstra a heterogeneidade do ejaculado nas subpopulações. Contudo, os diluentes para a conservação e refrigeração dos espermatozoides felinos e as curvas de congelação ainda não estão totalmente estabelecidos e influenciam diretamente a viabilidade dos espermatozoides criopreservados. Diante disso, os resultados da utilização do sêmen felino após criopreservação são inconsistentes, sendo necessários mais estudos para elucidar melhores curvas de congelação e meios de diluentes para viabilizar a preservação do material genético dos gatos.


The domestic cat is the only species of the Felidea family without risk or imminence of extinction, unlike most wild cats. Therefore, the development and improvement of different reproductive biotechnologies are essential for the maintenance of reproductive quality for the preservation of the most vulnerable species. Furthermore, semen biotechnologies are the basis for reproductive technologies such as artificial insemination (AI) and in vitro fertilization (IVF). Thus, the objective of this bibliographic compilation was to approach the main techniques of collection, analysis, and preservation of feline semen/sperm, as well as the use of these cells in AI and IVF. For feline semen collection, different methods have been applied: pharmacological ejaculation, electroejaculation, and artificial vagina. In case of death of the sire, sperm recovered from the epididymis also show reproductive viability. Moreover, the sperm kinetics evaluated by the CASA system, the morphology, and the morphometry are the main analyzes that demonstrate sperm quality and reflect on ejaculate fertility. The CASA system also evaluates the individual path of each sperm, which, when grouped into clusters, demonstrates the heterogeneity of the ejaculate in the subpopulations. However, diluents for the conservation and refrigeration of feline sperm and freezing curves are not yet fully established and directly influence the viability of cryopreserved sperm. Therefore, the results of using feline semen after cryopreservation are inconsistent, and further studies are needed to elucidate better freezing curves and diluents to enable the preservation of the genetic material of cats.

Abstract ADAM2 (fertilin β) is a sperm surface protein reported in several mammalian species. However, the presence of ADAM2 in the male reproductive system and sperm of the camel is not well known. The present study was to clarify the localization and expression of ADAM2 in the dromedary camel testis, epididymis and spermatozoa during rutting season using immunohistochemistry (IHC) and the quantitative real-time polymerase chain reaction (qPCR). Tissue samples were obtained from the testis (proximal and distal) and epididymis (caput, corpus, and cauda) from eight mature male camels. Epididymal and ejaculated sperms were collected from four other fertile camels. IHC analysis clearly showed the localization of ADAM2 protein in the spermatocytes and the round and elongated spermatids of the testis, in the epithelial cells along the epididymis tract, on the posterior head of the sperm within the cauda epididymis, and on the acrosomal cap of both the epididymal and ejaculated sperm. The expression of camel ADAM2 mRNA was significantly higher (P < 0.05) in the testis when compared with the epididymis. These findings may suggest an important role of ADAM2 in the fertility of male dromedary camels.

Abstract This study investigated the morphology and immunoexpression of aquaporins (AQPs) 1 and 9 in the rete testis, efferent ducts, epididymis, and vas deferens in the Azara's agouti (Dasyprocta azarae). For this purpose, ten adult sexually mature animals were used in histologic and immunohistochemical analyses. The Azara's agouti rete testis was labyrinthine and lined with simple cubic epithelium. Ciliated and non-ciliated cells were observed in the epithelium of the efferent ducts. The epididymal cellular population was composed of principal, basal, apical, clear, narrow, and halo cells. The epithelium lining of vas deferens was composed of the principal and basal cells. AQPs 1 and 9 were not expressed in the rete testis. Positive reaction to AQP1 was observed at the luminal border of non-ciliated cells of the efferent ducts, and in the peritubular stroma and blood vessels in the epididymis, and vas deferens. AQP9 was immunolocalized in the epithelial cells in the efferent ducts, epididymis and vas deferens. The morphology of Azara's agouti testis excurrent ducts is similar to that reported for other rodents such as Cuniculus paca. The immunolocalization results of the AQPs suggest that the expression of AQPs is species-specific due to differences in localization and expression when compared to studies in other mammals species. The knowledge about the expression of AQPs in Azara's agouti testis excurrent ducts is essential to support future reproductive studies on this animal, since previous studies show that AQPs may be biomarkers of male fertility and infertility.

ABSTRACT Description and seasonal variation in epididymal histomorphometry of Dermanuracinerea (Chiroptera: Phyllostomidae) in a fragment of the Atlantic Forest in the Northeastern Brazil. This study aimed to evaluate seasonal patterns in the histomorphometry of the epididymis of Dermanura cinerea (Gervais, 1856) in a fragment of the Atlantic Forest in the Northeastern Brazil. Eighteen adult male specimens captured by mist net were used. The field-work occurred monthly over 18 months, during two consecutive nights. Meteorological data (precipitation) were obtained from the National Institute of Meteorology. After euthanasia, specimens had the epididymis collected, which were fixed and processed. The histological slides produced were stained by Hematoxylin - Eosin and analyzed by optical microscopy. The morphometric parameters analyzed were the tubular, lumen and epithelium areas, of the regions of the initial segment, caput, corpus and cauda of the epididymis. The histomorphometric data were submitted to the Mann-Whitney U test analyzes. The results showed that D. cinerea presented spermatozoa in all regions of the epididymis, except in the initial segment. The highest averages of the tubular, lumen and epithelial areas in the four regions were observed during the dry months. Therefore, D cinerea presented greater sensibility in the region of the cauda of the epididymis, during the months with low rainfall indices. This indicates that environmental conditions have considerable influence on the epididymal morphophysiology of this species, especially in relation to the storage of sperm in the tail of this organ, in area of Atlantic forest in northeastern Brazil.


RESUMO Esse estudo objetivou avaliar sazonalmente a histomorfometria do epidídimo de Dermanura cinerea (Gervais, 1856) em um fragmento de Mata Atlântica no nordeste do Brasil. Foram utilizados 18 espécimes machos adultos capturados por redes de neblina. As coletas ocorreram mensalmente ao longo de dezoito meses, durante duas noites consecutivas e os dados meteorológicos foram fornecidos pelo Instituto Nacional de Meteorologia. Depois de eutanasiados, os espécimes tiveram os epidídimos coletados e esses órgãos foram fixados e processados. As lâminas histológicas foram coradas por Hematoxilina - Eosina e analisadas em microscopia óptica. Os parâmetros morfométricos analisados foram as áreas do túbulo, do lúmen e do epitélio das regiões do segmento inicial, cabeça, corpo e cauda do epidídimo. Os dados histomorfométricos obtidos foram submetidos às análises no teste U de Mann-Whitney. Os resultados revelaram que D. cinerea apresentou espermatozoides em todas as regiões do epidídimo, exceto no segmento inicial. As maiores médias das áreas tubular, do lúmen e do epitélio nas quatro regiões, foram constatadas durante os meses secos. Portanto, D cinerea apresentou maior sensibilidade na região da cauda do epidídimo, ao longo dos meses com baixos índices pluviométricos. Isso indica que as condições ambientais apresentam considerável influência sobre a morfofisiologia epidídimária dessa espécie, sobretudo, em relação ao armazenamento de espermatozoides na cauda desse órgão, em área de Mata atlântica do nordeste brasileiro.
