Clinical History: A euthanized, 14-year-old, Warmblood gelding that had participated in an equine show was presented for necropsy and diagnostic workup to the San Bernardino laboratory of the California Animal Health and Food Safety Laboratory System. The animal had a 4-day history of mildly swollen limbs and hyperthermia, and developed neurologic signs shortly before euthanasia. Necropsy Findings: The urinary bladder had multifocal to coalescing hemorrhages in the mucosa and approximately 5 ml of turbid urine with sandy sludge (Figure 1). The entire spinal cord was removed and cross-sectioned serially after fixation in 10% neutral-buffered formalin during 48h. Multifocally, in multiple sections of the cervical, thoracic, and lumbar segments, there were uni- or bilateral and asymmetrical, wedge-shaped areas of gray discoloration and hemorrhage (Figures 2-4). In addition, there were extensive hemorrhages around the nerve roots of the cauda equina. Follow-up questions: Five differential diagnoses for the gross lesions in the spinal cord (Fig. 1B-1D) Microscopic description for the lesions in the spinal cord (Fig. 2A-2D) Most likely cause based on clinical history and gross and microscopic findings.

The neuropathology of canid alphaherpesvirus 1 (CaHV-1) infection in dogs has been reported, but the importance of the cerebellar lesions in the routine diagnosis remains relatively understated in the veterinary literature. Here we characterize the neuropathology of natural CaHV-1 infection in 25 dogs. The disease affected predominantly male dogs (22 cases). Clinical signs in 15 cases varied and sudden death was reported in 10 cases. All dogs had the typical areas of necrosis and hemorrhage in multiple organs with eosinophilic intranuclear viral inclusions in epithelial and/or inflammatory cells in multiple organs in 21 cases. Cerebral swelling and reddening were reported in 3 cases. Neurohistologic changes consisted of mild lymphoplasmacytic meningoencephalitis with occasional vasculitis and widespread glial nodules. Rare intranuclear viral inclusions (4 cases) were observed in endothelial cells or inflammatory cells in glial nodules. Cerebellum was examined in 11 cases and had extensive segmental necrosis of Purkinje cell layer (8 cases) and granule neurons in the internal (9 cases) or external (2 cases) granular cell layer. Intranuclear viral inclusions were observed in 8 cases and occurred in necrotic granule neurons in the inner granular cell layer (7 cases), outer granular cell layer (2 cases), Purkinje neurons (1 case), or endothelial cells (1 case). Diagnostic confirmation in 23 cases was based on routine histology and fluorescent antibody testing for CaHV-1 on fresh or frozen tissue samples. A routine diagnosis was based solely on the visualization of intranuclear viral inclusions in 2 cases.(AU)

The herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) occur worldwide. Infections caused by these viruses have great public health importance due to the growing resistance to the first-choice drug, acyclovir, especially in immunosuppressed patients. Alkaloids derived from species of Annonaceae have been reported as antiviral agents against HSV and others viruses. Within this context, we evaluated the antiviral activity of the total alkaloid fraction (TAF) extracted from the branches of Fusaea longifolia (Aubl.) Saff. (Annonaceae), a species native to the Amazon region, against the HSV-1 and HSV-2 viruses. The antiviral activity was evaluated through the plate reduction assay and the mode of action was investigated by a set of other assays. The TAF was active against the HSV-2 strain 333 and against the HSV-1 strains KOS and 29R (acyclovir resistant), with selectivity index values (SI = 50% cytotoxic concentration/50% effective concentration) of 5, 4 and 3, respectively. In the preliminary study of the anti-HSV-2 mode of action, TAF showed viral inhibitory effects if added up to 12 h post-infection, had virucidal activity and did not present viral inhibition in pre-treatment. Our results showed that the TAF exhibited anti-HSV activity. Regarding HSV-2, TAF acted after the viral infection and had virucidal activity. A mass spectrometry analysis revealed the presence of nine alkaloids in the TAF that had previously been reported for Annonaceae, including liriodenine, lysicamine and isoboldine, which have been described as potential anti-HSV-1 agents.(AU)
Os vírus herpes simplex tipo 1 (HSV-1) e tipo 2 (HSV-2) têm ampla ocorrência global. As infecções causadas por esses vírus têm grande importância em saúde pública devido à crescente resistência ao fármaco de primeira linha, aciclovir, principalmente em pacientes imunossuprimidos. Alcaloides derivados de espécies de Annonaceae têm sido relatados como agentes antivirais contra o HSV e outros vírus. Neste contexto, nós avaliamos a atividade antiviral da fração alcaloide total (TAF) dos ramos de Fusaea longifolia (Aubl.) Saff. (Annonaceae), uma espécie nativa da região amazônica, contra os vírus HSV-1 e HSV-2. A atividade antiviral foi avaliada através do ensaio de redução em placa e o modo de ação foi investigado por um conjunto de ensaios. O TAF foi ativo contra a cepa HSV-2 333 e contra as cepas HSV-1 KOS e 29R (resistente ao aciclovir), com valores de índice de seletividade (IS = 50% concentração citotóxica/50% concentração efetiva) de 5, 4 e 3, respectivamente. No estudo preliminar do modo de ação da atividade anti-HSV-2, o TAF inibiu a replicação viral quando adicionados até 12 h pós-infecção, apresentou atividade virucida e não apresentou inibição viral no pré-tratamento. Nossos resultados mostraram que o TAF exibiu atividade anti-HSV. Em relação ao HSV-2, o TAF atuou após a infecção viral e apresentou atividade virucida. Uma análise do TAF por espectrometria de massas identificou a presença de nove alcaloides, incluindo liriodenina, lisicamina e isoboldina, que já foram descritos como potenciais agentes anti-HSV-1.(AU)

Objetivou- se com o presente estudo avaliar e discutir as ocorrências da "Doença de Pacheco" em psitaciformes descritas no Brasil e em outros países, pretendendo-se discernir sobre as causas da infecção, discorrendo sobre as causas de contágio e disseminação, descrevendo brevemente a sintomatologia, possíveis lesões, diagnósticos, profilaxia e tratamento, a fim de evitar o contágio, minimizando a morbidade e mortalidade das aves. Trata-se de uma revisão bibliográfica, a qual foi realizada por meio de consultas à periódicos e livros mais recentes. Os principais sinais clínicos são anorexia, sonolência, letargia, penas eriçadas, diarreia amarelada, regurgitação, inatividade e, às vezes, sinais nervosos, chegando, por fim, à morte súbita e rápida. Na necropsia, podem ser achados hepatomegalia, esplenomegalia e necrose. A profilaxia se concentra no controle da superpopulação e protocolo adequado de quarentena das aves. O tratamento indicado para o herpesvírus é o uso de nucleosídeo sintético, com atividade inibitória, o aciclovir, que tem apresentado bons resultados na redução das taxas de mortalidade.(AU)
The herpesvirus of the species Psittacid alphaherpesvirus 1 (PsHV-1), is responsible for the highly infectious and acute avian disease, described as "Pacheco's Disease" (PD). Several species of parrots (Amazon parrots, followed by African gray parrots, common parrots, macaws, cockatoos and some species of parakeets) are susceptible to the disease, especially those originating from breeding sites that have entered rehabilitation centers in any region. geographic. The aim of the present study was to evaluate and discuss the occurrences of "Pacheco's Disease" in parrots described in Brazil and in other countries, intending to discern the causes of the infection, discussing the causes of contagion and dissemination, briefly describing the symptomatology, possible lesions, diagnosis, prophylaxis and treatment, in order to avoid contagion, minimizing the morbidity and mortality of the birds. This is a bibliographic review, which was carried out through consultations with the most recent journals and books. At necropsy, hepatomegaly, splenomegaly and necrosis may be found. Prophylaxis focuses on overpopulation control and proper bird quarantine protocol. The treatment indicated for herpesvirus is the use of a synthetic nucleoside, with inhibitory activity, acyclovir, which has shown good results in reducing mortality rates.(AU)
El herpesvirus de la especie Psittacid alphaherpesvirus 1 (PsHV-1), es responsable de la enfermedad aviar aguda y altamente infecciosa, descrita como "Enfermedad de Pacheco" (EP). Varias especies de loros (loros amazónicos, seguidos de loros grises africanos, loros comunes, guacamayos, cacatúas y algunas especies de periquitos) son susceptibles a la enfermedad, en especial los que se originan en criaderos que han ingresado a centros de rehabilitación en cualquier región geográfica. El objetivo del presente estudio fue evaluar y discutir las ocurrencias de la "Enfermedad de Pacheco" en loros descritas en Brasil y en otros países, con la intención de discernir las causas de la infección, discutiendo las causas de contagio y diseminación, describiendo brevemente la sintomatología, posibles lesiones, diagnóstico, profilaxis y tratamiento, con el fin de evitar el contagio, minimizando la morbimortalidad de las aves. Se trata de una revisión bibliográfica, que se realizó mediante consultas a las revistas y libros más recientes. Background: Enzootic bovine leukosis (EBL) is a widespread infectious disease caused by the bovine leukemia virus (BLV), which results in immune system dysfunction. The resulting immunosuppression may lead to an increased prevalence of other diseases. Dairy cows infected have altered immune function associated with decreased milk production and shortened lifespan and decreased immune response to immunization. BLV infection, however, is often asymptomatic, so any connection between subclinical infection and common reproductive diseases remains unknown. This study aimed to describe the relationship between naturally occurring subclinical BLV and infectious reproductive diseases seroconversion in the field. Materials, Methods & Results: The diseases investigated included Bovine viral diarrhoea (BVD), Bovine alfaherpesvirus 1 (BoHV-1), Bovine gammaherpesvirus 4 (BoHV-4), Chlamydiosis, Leptospirosis, Brucellosis and Neosporosis in dairy cattle. Six hundred fifty-five sera samples from the northern and south-central regions of Uruguay, from asymptomatic female Holstein and Holstein crosses without a history of vaccination against reproductive diseases were processed using reference diagnostic methods (Seronautalization, ELISA, MAT, Rose Bengal Plate test). The seroprevalence of BLV was 20.0%. Seroprevalence of reproductive diseases BVD, BoHV-1 and BoHV-4 were 99.3%, 41.2% and 27.3% of the populations, respectively, and the total seroprevalence of Leptospirosis, bovine Neospora caninum and Chlamydiosis were 19.8%, 29.8% and 33.0% respectively. The results revealed positive associations between naturally contracted BLV and the presence of antibodies against BoHV-1 (P = 0.002), as well as between naturally contracted BLV and presence of antibodies against Leptospira spp. (P = 0.028). Discussion: BLV infection can impact innate and adaptive immune system cells and alter the proper functioning of uninfected cells. BLV infection may also induce changes in the complex balance of cytokine expression, cell proliferation, and programmed cell death in T- and B-lymphocytes, which is critical for immune competence and effective response to infectious challenges. The progression of BLV infection has a substantial effect on host defense mechanisms. Indeed, lowmagnitude serologic responses to a commercial foot-and-mouth disease vaccine and a J5 Escherichia coli vaccine have been observed. These results are supported by recent trial studies showing a reduced immune response to vaccination against BoHV-1 and Leptospira spp. in asymptomatic animals infected with BLV. These are 2 of the most prevalent infectious reproductive diseases in cattle worldwide, and our results provide evidence that a link between BLV and susceptibility to these diseases may exist. Although there is evidence of the co-occurrence of these diseases, it remains unknown whether there is a direct or indirect effect of BLV on infertility, embryonic loss, or abortion. Another possibility is that natural infection with these reproductive pathogens (BoHV-1, Leptospira, or others) promotes BLV expression, negatively affecting the farms where these pathogens are endemic. Considering the high seroprevalence of BLV in dairy herds in North and South America where the infection is endemic, it was explored BLV's role as an immunosuppressant by quantifying its co-occurrence with diseases that affect reproductive performance in breeding herds. Future work should clarify the role of BLV and the co-occurring pathogens in causing infertility or abortions.

The effectiveness of vectored recombinant vaccines to control infectious laryngotracheitis (ILT) in chickens from a region (State of Minas Gerais, Brazil) with ~10 million layers was evaluated under field conditions from 2014-2018. During this period, only recombinant turkey herpesvirus (rHVT) or fowl poxvirus (rFPV) vaccines that express antigens of infectious laryngotracheitis virus (Gallid herpesvirus-1; GaHV-1) were used. Layer chickens (n=1,283), from eight different egg-producing companies, were individually sampled and examined (active surveillance), and in instances when government poultry health veterinarians were notified due to respiratory disease (passive surveillance). Clinical, macroscopic, and histopathology examinations were performed to diagnose ILT as well as molecular techniques for the detection and characterization of the GaHV-1 DNA from the trachea and trigeminal ganglia (TG). The layer hens sampled and examined belonged to flocks and farms that used different vaccination protocols (non-vaccinated, single dose vaccination, and prime/ boost vaccination). This is the first long-term field study of the effectiveness of ILT vectored vaccines in a high-density multiple age layer hen region. Using various diagnostic methods, the occurrence of GaHV-1 infection and ILT clinical disease in layer hens vaccinated with vectored recombinant vaccines in one quarantined region of Brazil were investigated. The number of ILTV positive chickens by PCR and ILT clinical disease cases was lower in farms when all chickens were vaccinated with at least one vaccine. However, the difference in the detection rates of GaHV-1 infection was significant only when compared farms with prime/ boost and farms using single dose of HTV-LT.
A efetividade das vacinas recombinantes vetorizadas para o controle da laringotraqueíte infecciosa (LTI) nas aves de uma região (Minas Gerais, Brasil) com aproximadamente 10 milhões de poedeiras foi avaliada em condições de campo, no período de 2014 a 2018. Durante este período, somente as vacinas recombinantes "turkey herpesvirus" (rHVT) ou "fowl poxvirus" (rFPV), que expressam antígenos do vírus da laringotraqueíte (Gallid herpesvirus-1; GaHV-1) foram utilizadas. Galinhas poedeiras (n=1.283), de oito diferentes granjas produtoras de ovos, foram individualmente amostradas e examinadas por monitoramento ativo e, na ocorrência de notificação de doença respiratória aos veterinários do serviço oficial, por monitoramento passivo. Exames clínicos, macroscópicos e histopatológicos foram realizados para o diagnóstico de LTI, bem como técnicas moleculares para a detecção e caracterização do DNA de GaHV-1 da traqueia e gânglio trigêmeo. As galinhas poedeiras pertenciam a lotes e granjas que usavam diferentes protocolos de vacinação (não vacinadas, uma dose ou tipo de vacina e duas doses ou tipos de vacina). Este é o primeiro longo estudo a campo sobre a efetividade das vacinas vetorizadas em uma região com população elevada de poedeiras de múltiplas idades. Utilizando vários métodos de diagnóstico, a ocorrência da infecção por GaHV-1 e a LTI clínica em poedeiras de uma região interditada do Brasil foi investigada. O número de galinhas positivas para o vírus GaHV-1 e para casos clínicos de LTI nas granjas foi menor quando todas as aves estavam vacinadas com, pelo menos, um tipo ou dose de vacina. Entretanto, a diferença na taxa de detecção da infecção por GaHV-1 foi significativa somente quando a comparação foi realizada entre granjas com aves vacinadas com duas doses e aves de granjas vacinadas com uma única dose de HVT-LT.

One of the limiting factors of productive efficiency in cattle and buffalo herds is related to the high prevalence of infectious diseases which affect reproduction. The aim of this study was to determine the prevalence of anti-Leptospiraantibodies for bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BoHV-1) in bovine and buffalo herds in Colombia. Blood serum samples were collected from 1100 buffaloes and 1000 cattle. The ELISA technique was used to detect antibodies against BVDV and BoHV-1, and the microscopic agglutination technique to detect anti-Leptospiraantibodies. The prevalence of anti-Leptospiraantibodies and of BVDV and BoHV-1 in bovine samples was observed in 16, 39.7, and 65% of animals, respectively, while the positivity in samples for the same antibodies in buffalos was detected in 18.7, 27.5 and 51.5%, respectively. Exposure of cattle and buffaloes to BoHV-1 was positively associated with age, higher prevalence rates were observed in older ages. Seropositivity of cattle for BVDV and BoHV-1 was higher in male animals. Activities such as embryo transfer, milking, and needle reuses, as well as the presence of cats and rodents are factors which favor positivity of the herd for BVDV and BoHV-1.(AU)

Background: Marek's disease (MD) is a transmissible disease in chickens caused by Gallid alphaherpesvirus 2 (GaHV-2). The infection is characterized by lymphocyte cellular infiltrates in peripheral nerves and other organs and tissues, including the skin; which can lead to dysfunction causing progressive asymmetric paresis and complete spastic paralysis of body extremities. Dermatitis and cardiac myositis caused by GaHV-2 in free-range chickens has rarely been described in Brazil. This reports the occurrence of the disease with a confirmatory molecular diagnosis in free-range poultry showing signs of dermatitis, poor performance, and cachexia and no mortality in the semi-arid Potiguar region. Cases: Twenty roosters of the Shamo lineage, among a brood of 42 birds, had a history of progressive weight loss and skin lesions. Two birds with poor body condition, erythema, and scaling of the skin in the head and cervical regions were sent for clinical care. All birds were between 12 and 18 months of age and were vaccinated against Newcastle disease and Fowlpox with only a few receiving vaccines against MD and Gumboro disease. According to the owner's report, some birds were previously kept outdoors, and when they were transferred to a small shed with little air circulation, they began to develop clinical signs after approximately 15 days. The first signs of the disease were also reported to have appeared 2.5 months before clinical care and, in the meantime, several treatments were instituted without success. Owing to the general condition of the animals and inconclusive clinical suspicion, the birds were subjected to euthanasia and necropsy. Tissue samples were collected for histopathological and polymerase chain reaction analyses to search for the GaHV-2 DNA meq gene. The main clinicopathological findings were erythema (47%, 20/42) and desquamation of skin and mild, prominent white multifocal areas in the heart. Histopathology revealed infiltration of pleomorphic lymphoblastic cells in the skin, heart, and sciatic nerve. The amplification of the L-meq and meq oncoprotein genes in these organs and in the liver, confirmed the infection by GaHV-2, consistent with that of a field strain. Discussion: MD was confirmed based on the macroscopic and histological lesions, and with the detection of GaHV-2 DNA in the affected tissues. The unusual clinical presentation represented an initial challenge for diagnosis. The clinical history was important to lead to the suspicion of MD, as roosters initiated clinical signs 15 days after they were transferred to a small shed with poor air circulation. This probably favored the high viral concentration and disease transmission among susceptible birds in the brood because the feather follicle is the primary site of viral replication for transmission; and desquamation of infected epithelial cells favor airborne horizontal transmission to susceptible chickens. The roosters had not been vaccinated against MD, which probably favored the infection, as vaccination is known to be a fundamental approach for MD control for effective growth of the poultry industry. Clinical findings and lesions, together with viral molecular detection, were fundamental for the diagnosis, a premise for the application of adequate prevention and control measures for the disease in breeding. This is the first report of MD with a confirmatory molecular diagnosis in northeastern Brazil.

Felid alphaherpesvirus type 1 (FHV-1) is an important cause of respiratory and ocular diseases in cats worldwide. Mice have been widely used to study the pathogenesis of several human and animal viruses, especially herpesviruses. This study aimed to verify whether BALB/c mice are susceptible to FHV-1 infection. The animals were intranasally inoculated with FHV-1 and their clinical signs were observed from 3 days postinfection (dpi). At 10 dpi, the animals were euthanized and the lungs, liver, spleen, and kidneys were collected for histopathological examination and quantitative polymerase chain reaction. The results showed that mice were infected with FHV-1 and reproduced several features of the disease observed in its natural host. Histological lesions and viral DNA were found in all sampled tissues, with a higher frequency of FHV-1 DNA copies detected in the lungs. All mice were seroconverted to FHV-1 at 7 dpi. To our knowledge, this is the first report of experimental infection of BALB/c mice with FHV-1. Our findings demonstrate that this murine model can contribute to understanding of FHV-1 pathogenesis and may be useful for trials against this virus.(AU)
O herpesvírus felino tipo 1 (FHV-1) é um importante agente causador de doença respiratória e ocular em gatos em todo o mundo. Camundongos têm sido amplamente utilizados para estudar a patogenia de diversos vírus humanos e animais, especialmente os herpesvírus. O objetivo deste estudo foi verificar se camundongos BALB/c poderiam ser suscetíveis a infecção pelo FHV-1. Os animais foram inoculados com FHV-1 intranasalmente e sinais clínicos foram observados a partir do dia 3 após a infecção (dpi). Após dez dias da inoculação, os animais foram eutanasiados e os pulmões, fígado, baço e rins foram coletados para exame histopatológico e PCR quantitativo (qPCR). Os resultados mostraram que os camundongos foram infectados com o FHV-1 e reproduziram várias características da doença que são observadas em seu hospedeiro natural. Lesões histológicas e DNA viral foram detectadas em todos os tecidos amostrados, com maior frequência de DNA do FHV-1 nos pulmões. Todos os camundongos soroconverteram para FHV-1 aos 7 dpi. Para nosso conhecimento, este estudo é o primeiro relato de infecção experimental de camundongos BALB/c com FHV-1. Nossos resultados demonstraram que esse camundongo pode contribuir para o entendimento da patogenia do FHV-1 e pode ser um modelo útil para ensaios contra esse vírus.(AU)

Fibropapillomatosis (FP) is an infectious disease caused by Chelonid alphaherpesvirus 5 (ChHV5). Nevertheless, its clinical manifestations are considered multifactorial. Due to its relevance, FP is currently monitored in sea turtle populations in the United States, Australia, Caribbean, and Brazil. Between 2000 and 2020, the TAMAR Project/ TAMAR Project Foundation analyzed the prevalence of FP in nine states and oceanic islands along the Brazilian coast, including Fernando de Noronha Archipelago (FNA), a historically FP-free area. A total of 4,435 green sea turtles (Chelonia mydas) were monitored from 2010 to 2016. Additionally, in 2012 and 2014, 43 FP-free skin samples were analyzed for ChHV5 using a qualitative PCR for the UL30 polymerase (pol) sequence. In 2015, a bilateral ocular nodule characterized as an FP tumor was reported in one of the monitored individuals undergoing rehabilitation. Tissue samples were collected following surgical removal of the tumor. Characterization of a 454 bp UL30 polymerase gene revealed a ChHV5 sequence previously reported in other areas of the Atlantic Brazilian coast. In the years following this finding from January 2017 to March 2020, a total of 360 C. mydas were monitored in the same area and no FP tumors were detected. This is the first report of FP and the first detection of ChHV5 in FNA, a finding of great concern considering this site's historical absence of FP occurrence. This study highlights the importance of monitoring this disease in historically FP-free areas of the Brazilian Atlantic coast.(AU)

Esta doença é monitorada atualmente em populações de tartarugas marinhas nos EUA, Austrália, Caribe e Brasil. Desde 2000, o Projeto TAMAR/Fundação Projeto TAMAR analisa a presença de FP em nove estados da costa brasileira e ilhas oceânicas, incluindo o arquipélago de Fernando de Noronha, uma área historicamente livre de FP. Um total de 4.435 indivíduos de Chelonia mydas foram monitorados de 2010 a 2016 e 43 amostras de pele foram analisadas para detectar ChHV5 em 2012 e 2014 com o objetivo de avaliar a presença do vírus em tecidos sem FP, usando uma PCR qualitativa para detecção de sequências do gene da UL30 polimerase. Em 2015, uma tartaruga verde (C. mydas) foi relatada com um nódulo ocular bilateral caracterizado como FP. Amostras de tecido foram coletadas durante sua reabilitação e procedimento cirúrgico para remover o tumor. A caracterização parcial de uma sequência de 454 bp do gene UL30 polimerase detectou ChHV5 anteriormente relatado em outras áreas da costa atlântica brasileira. Após estes achados, de janeiro de 2017 a março de 2020, um total de 360 indivíduos de C. mydas foram monitorados e nenhum caso de FP foi registrado. Este é o primeiro relato de FP e a primeira caracterização de ChHV5 no arquipélago de Fernando de Noronha, uma questão preocupante e que ressalta a importância do monitoramento desta doença em áreas historicamente livres de FP na costa atlântica brasileira.(AU)

The aim of the present study was to evaluate the post-vaccinal reaction to two lentogenic vaccine strains of Newcatle disease virus (NDV) and a recombinant turkey herpesvirus (rHVT) vaccine expressing the fusion glycoprotein of NDV in broiler chickens through histomorphometric and histopathologic analyses of the trachea. The experiment involved 245 chicks housed in randomized blocks with three different enclosures under controlled conditions of temperature, light and ventilation. Each enclosure represented a vaccine strain and was divided into groups according to the administration route. Each block also had its own control group composed of unvaccinated birds. The vaccine strains PHY.LMV.42 (PL42) and La Sota (LS) were selected according to the Intracerebral Pathogenicity Index (ICPI) and the rHVT-NDV Serotype 3 strain (ST3) was selected for representing non-NDV infection. At two, four, seven, 14 and 21 days post vaccination, fragments from the middle third of the trachea were collected and submitted to routine histological processing. For the histomorphometric analysis, the slides were photographed, and the thickness of the tracheal mucosa was measured. Statistical analysis involved two-way ANOVA and Tukey's post-hoc test with a 5% significance level. For the histopathological evaluation, lesions were described as to the degree of intensity and distribution. At four and 14 days post vaccination with the LS strain administered by the ocular route, the means of thickening of the tracheal mucosa (20.85±7.31µm and 26.97±5.50µm, respectively) were significantly higher (p<0.05) than for all other strains, which was related to the severe histopathological lesions found in this group, characterized by hyperemia, hyperplasia of the mucous glands, moderate deciliation and multifocal lymphohistiocytic inflammatory infiltrate. At 21 days, broiler chickens vaccinated with the ST3 strain showed more discrete lesions and less thickening of the tracheal mucosa (23.23±7.62µm; p<0.05) in comparison with other studied strains. The lesions found in this group were only hemorrhage, deciliation and mild focal lymphocytic inflammatory infiltrate. The results of the histomorphometry and histopathology of the trachea indicated that vaccination with rHVT-NDV Serotype 3 strain induced lower degree post-vaccine tracheal lesions compared to other vaccine strains analyzed in this study.
Objetivou-se avaliar a reação pós-vacinal de duas estirpes lentogênicas do vírus da doença de Newcastle (VDN) e uma vacina recombinante de herpesvirus de perus (rHVT) que expressa a glicoproteína de fusão de VDN em frangos de corte por meio da histomorfometria e histopatologia da traqueia. Foram utilizados 245 pintos alojados em blocos ao acaso, sendo três galpões distintos em condições controladas de temperatura, luz e ventilação. Cada galpão representou uma cepa vacinal, onde foram divididos por grupos de acordo com a via de administração. Todos os blocos possuíam um grupo controle composto por aves não vacinadas. As cepas vacinais PHY.LMV.42 (PL42) e La Sota (LS) utilizadas foram selecionadas de acordo com o Índice de Patogenicidade Intracerebral (IPIC) e a cepa Sorotipo 3 (ST3), da vacina rHVT-VDN foi selecionada por não representar infecção do VDN. Aos dois, quarto, sete, 14 e 21 dias pós-vacinação, fragmentos do terço médio da traqueia foram coletados e posteriormente processados conforme rotina histológica. Para análise histomorfométrica da mucosa traqueal, as lâminas foram fotografadas e realizadas as mensurações da espessura da mucosa traqueal sendo aplicado teste de anaÌlise de variaÌncia a dois criteÌrios (ANOVA) e utilizando o post-hoc de Tukey com niÌvel de significaÌncia de 5%. Para a avaliação histopatológica foram observadas a presença de lesões microscópicas e estas foram descritas quanto ao grau de intensidade e distribuição. Aos quatro e quatorze dias pós-vacinação com a cepa LS administrada por via ocular, as médias do espessamento da mucosa traqueal (20,85±7,31µm e 26,97±5,50µm, respectivamente) foram significativamente maiores (p<0,05) quando comparada a todas as demais cepas utilizadas, isto se deve às severas lesões histopatológicas encontrados neste grupo, caracterizadas por hiperemia, hiperplasia das glândulas mucosas, deciliação moderada e infiltrado inflamatório linfohistiocitário multifocal moderado. Já aos 21 dias as aves vacinadas com a cepa ST3 apresentaram lesões mais discretas e menor espessamento da mucosa da traqueia (23,23±7,62µm; p<0,05) em comparação às demais cepas estudadas. As lesões encontradas neste grupo foram apenas hemorragia, deciliação e infiltrado inflamatório linfocitário focal discreto. Os resultados da histomorfometria e da histopatologia da traqueia indicou que a vacinação com a rHVT-NDV, cepa Sorotipo 3 induziu menor grau de lesões pós-vacinais na traqueia comparada a outras cepas vacinais analisadas nesse estudo.

Infectious laryngotracheitis (ILT) is a severe respiratory disease, which causes high morbidity and mortality in affected birds. In our study, ILT were reported in 42 farms from nine governates over two years (20182020) that showed clinical signs of ILT including dyspnea, blood expectoration of, excessive lacrimation, rattling, conjunctivitis. The disease affected different chicken breeds and age groups despite vaccination with licensed and commonly used vaccines. Samples of larynx, trachea, lungs and air sacs were examined and collected for histopathological, ultrastructural, immunohistochemical examination and molecular detection. Gross examination of laryngeal and tracheal lumen revealed different types of exudate varied from catarrhal to fibrinonecotric, also pneumonia and airsacculitis were detected. Histopathological examination showed different alternation in larynx, trachea, lung and air sac as characteristic syncytial cells containing intranuclear inclusion body hanged in fibrinoheterphilic exudate that precent in laryngeal, tracheal, bronchial and parabronchial lumen and air sacs. Tracheal lesion scoring system was used to categorize the severity of lesion in different governates. Tracheal lesion score showed that 6.02%, 26.5%, 43.3% of the birds exhibited mild, moderate, and severe changes, respectively, while 24.18% of the birds exhibited very severe changes. Furthermore, severe cases were related to the Qalyubia , Fayoum then Sharkia Governorate. Moreover, immunohistochemistry was used to detect viral particles in syncytial cells, inflammatory cells beside epithelium of trachea and lung. Transmission electron microscopy enabled the detection of virus particles and demonstrated that heterophils could be infected. PCR targeting a region in the thymidine kinase gene and glycoprotein gJ gene confirmed the presence of infectious laryngotracheitis ILT virus-specific DNA. In conclusion, anatomopathological, immunohistochemical, molecular and ultrastructural findings showed increased of ILTV severity in Egypt. Larynx, trachea, lungs and air sac should be collected and examined that aid in diagnosis. Importance of good biosecurity level to be considered.(AU)

The serumsamples were collected in sterile tubes without anticoagulant in stored isothermal boxes, with serum separation and storedat -20ºC. The samples were sent to the Animal Virology Laboratory of Universidade Estadual de Londrina, UEL for serological analysis. Serological diagnosis using the virus neutralization (VN) technique was performed according the OIEmanual of diagnostic tests and vaccines for terrestrial animals, using the BoHV-1 Los Angeles strain. Serum samples from 719 dairy cattle of reproductive age, from 47 farms in a settlement in the municipality of Andradina, São Paulo, were analyzed for the presence of antibodies against the Nebraska strain of BoHV-1 and Singer type 1 of BVDV-1, using the virus neutralization technique (VN). Regarding BoHV-1, 72.0% of the samples analyzed (518/719) were positive, with a geometric mean of antibody titers of 318. For BVDV-1, 44.8% of the samples were reactive, only one farm had no reactive animals, and the majority of antibody titers found in the animals' sera were low. The results of this study revealed that BoHV-1 was present in all of the studied herds, with moderate titration in the majority of cases, with the presence of some high individual titers among the tested animals. Also, even with the reactivation or circulation of BoHV-1 in herds, the clinical disease was not observed, an epidemiological finding that also applied to BVDV-1. In view of the results obtained, the circulation of diseases in family farm herds in the studied region was evident, suggesting the need to study local risk factors and improve health education policies as a way to prevent or reduce the prevalence of these diseases.

Em relação ao BoHV-1, 72,0% (518/719) das amostras foram positivas, sendo 318 a média geométrica dos títulos de anticorpos (GMT).Para o BVDV-1, 44,8% das amostras foram reagentes; apenas uma propriedade não teve animais reagentes, sendo a maioria dos títulos de anticorpos encontrados nos soros baixa. Observou-se que o BoHV-1 está presente em todos os rebanhos estudados com titulação moderada, com presença de alguns títulos individuais altos entre animais e com GMT alta em um dos rebanhos. Mesmo havendo a reativação ou mesmo a circulação do BoHV-1 nos rebanhos, não foi observado a doença clínica, fato epidemiológico que também ocorreu com o BVDV-1. Diante dos resultados obtidos, ficou evidente a circulação das doenças nos rebanhos da agricultura familiar na região estudada, sugerindo o estudo de fatores de risco local e melhorias de políticas de educação em saúde como forma de prevenir e ou reduzir a prevalência das enfermidades.

This study aimed to conduct a systematic review to estimate the economic impact of Bovine alphaherpesvirus 1 (BoAHV1) infection in Brazil using epidemiological indicators through a meta-analysis. Specific descriptors were used to retrieve studies from the Scopus, PubMed, Biblioteca Digital Brasileira de Teses e Dissertações, and Catálogo de Teses e Dissertações da Capes databases, selecting those that met the inclusion criteria established between the years 2000 and 2020. The selected studies were subjected to descriptive statistical analysis using prevalence data as the primary outcome with a 95% confidence interval (CI) with a meta-analysis of random effects and measures of heterogeneity, significance, magnitude of the effect, and measurement of publication bias. Abortion costs and estimates were calculated based on the prevalence of BoAHV1 infection in Brazil and the characteristics of the agent as viral subtypes that cause abortion, period of occurrence, average pregnancy rate, and morbidity applied to susceptible animals and animal replacement values. The results were obtained from 49 studies included for meta-analysis where a prevalence of BoAHV1 infection of 54.12% (95% CI: 49.07% - 59.26%) in the bovine population and 88.53% (95% CI: 82.97%92.43%) was present. From the structured formula, it is estimated that 258,779 bovine abortions occur, which causes a total loss, based only on the occurrence of abortion, of US $ 48,402,244.00 to the country. It is noteworthy that because of the losses caused, strict control and eradication measures need to be implemented based on the elaboration of normative instruction that includes health education measures, vaccination, tests for traffic, and animal trade so that BoAHV1 infections do not continue to negatively impact national producers economically, productively, and socially.(AU)
Objetivou-se com o presente estudo realizar uma revisão sistemática para estimar os impactos econômicos da infecção por Alfaherpesvirus Bovino 1 no Brasil a partir dos indicadores epidemiológicos por meio de uma metanálise. Foram usados descritores específicos para a recuperação de estudos a partir das bases Scopus, Pubmed, Biblioteca Digital Brasileira de Teses e Dissertações e do Catálogo de Teses e Dissertações da Capes, selecionando-se aqueles que se enquadraram nos critérios de inclusão estabelecidos entre os anos 2000 e 2020. Os estudos selecionados foram submetidos à análise estatística descritiva utilizando-se os dados de prevalência como desfecho primário com intervalo de confiança de 95%, sendo realizado metanálise de efeito randômico e avaliadas as medidas de heterogeneidade, significância, magnitude do efeito e aferição do viés de publicação. Os custos e estimativas de aborto foram calculados com base na prevalência encontrada para BoAHV1 no Brasil e as características do agente como subtipos virais que provocam o aborto, período de ocorrência, taxa média de prenhez e morbidade aplicados aos animais suscetíveis e aos valores de reposição animal. Os resultados foram obtidos a partir de 49 estudos incluídos para metanálise, onde se verificou uma prevalência de BoAHV1 de 54,12% (IC95%: 49,07% 59,26%) na população bovina e 88,53% (IC95%: 82,97% 92,43%) para os rebanhos. A partir da fórmula estruturada, se estima que ocorram 258.779 abortos bovinos, que totalizam um prejuízo, apenas baseado na ocorrência do aborto, de US$ 48.402.244,00 ao país. Destaca-se que em virtude dos prejuízos ocasionados, medidas de controle e erradicação mais rigorosas precisam ser implementadas a partir da elaboração de uma instrução normativa que contemple medidas de educação sanitária, vacinação, realização de exames para o trânsito e comércio de animais para que as infecções pelo BoAHV1 não continuem a impactar negativamente os produtores nacionais de forma econômica, produtiva e socialmente.(AU)

Objetivou-se descrever a ocorrência do Bovine alphaherpesvirus 5 (BoHV5) como causa de meningoencefalite não supurativa em bovinos do estado de Pernambuco, Brasil. Para tanto, 32 amostras de sistema nervoso embebidas em parafina foram obtidas de animais acometidos por doenças neurológicas atendidos na Clínica de Bovinos de Garanhuns da Universidade Federal Rural de Pernambuco (CBG-UFRPE), entre 2012 e 2016. As amostras foram analisadas quanto à presença do gene da glicoproteína C do BoHV5 por reação em cadeia da polimerase (PCR). Dois animais (6,25%) tiveram resultado positivo à PCR, e sua análise de sequenciamento indicou 100% de similaridade para o BoHV5. Os resultados histopatológicos desses dois animais revelaram lesões multifocais de meningoencefalite não supurativa associada à polioencefalomalácia, presença de corpúsculos de inclusão basofílicos, infiltração de células de Gitter e presença de manguitos perivasculares. A PCR se mostra uma importante ferramenta para diferenciação das infecções por BoHV5 de outras enfermidades neurológicas de bovinos, especialmente a raiva.(AU)

ABSTRACT: Avian influenza viruses (AIVs), Newcastle disease virus (NDV), West Nile virus (WNV), adenovirus (AV) and herpesvirus (HV) play an important role in the health of human and animal populations. However, knowledge of the prevalence of these viruses in wild birds is restricted to some groups (e.g. shorebirds) or regions worldwide. Information on grassland birds of South America, which is essential for their conservation, is scarce. The objectives of the present study were to evaluate occurrences of AIV, NDV, WNV, AV and HV for the first time in a bird community of a unique protected area in southern Brazil, which is home for the critically endangered yellow cardinal (Gubernatrix cristata), and captive yellow cardinals from fauna maintainers of the Brazilian Captive Program of the Yellow Cardinal. Passerine species of wild life were caught, identified and samples (swabs) were collected from the oropharynx and cloaca of 64 passerines of 26 species (including 3 yellow cardinals) and 30 yellow cardinals of captive, for molecular diagnosis. The samples were subjected to RNA and DNA extraction and the real-time polymerase chain reaction (RT-PCR) for AIV, NDV and WNV and nested PCR for AV and HV. One yellow cardinal of captive presented a positive result for AV, this result is important for planning, managing natural attributes and making decisions in relation to integrated conservation of threatened species. This is the first report of AV in yellow cardinal and epidemiological investigation of viruses in wild passerines of the Pampa biome, in Rio Grande do Sul, Brazil.
RESUMO: Os vírus da gripe aviária (VGA), vírus da doença de Newcastle (VDN), vírus do Nilo Ocidental (VNO), adenovírus (AV) e herpesvírus (HV) desempenham um papel importante na saúde das populações humana e animal. No entanto, o conhecimento da prevalência desses vírus em aves selvagens é restrito a alguns grupos (por exemplo, aves limícolas) ou regiões em todo o mundo. As informações sobre as aves campestres da América do Sul, essenciais para a sua conservação, são escassas. Os objetivos do presente estudo foram avaliar a ocorrência de VGA, VDN, VNO, AV e HV pela primeira vez em uma comunidade de aves de uma área única protegida no Sul do Brasil, que abriga o cardeal-amarelo (Gubernatrix cristata) criticamente ameaçado de extinção e em cardeais-amarelos de cativeiro dos mantenedores de fauna do Programa Brasileiro de Cativeiro do Cardeal-amarelo. Espécies de passeriformes silvestres foram capturadas, identificadas e amostras (swabs) foram coletadas da orofaringe e cloaca de 64 passeriformes de 26 espécies (incluindo 3 cardeais-amarelos) e 30 cardeais-amarelos de cativeiro, para diagnóstico molecular. As amostras foram submetidas à extração de RNA e DNA e à reação em cadeia da polimerase em tempo real (RT-PCR) para VGA, VDN e VNO e nested PCR para AV e HV. Um cardeal-amarelo de cativeiro apresentou resultado positivo para AV, este resultado é importante para o planejamento, manejo dos atributos naturais e tomada de decisões em relação à conservação integrada de espécies ameaçadas. Este é o primeiro relato de AV em cardeal-amarelo e de investigação epidemiológica de vírus em passeriformes silvestres do bioma Pampa, no Rio Grande do Sul, Brasil.
