Purpose: This study aimed to evaluate the effects of ozone therapy applied topically and/or by bagging on the healing of clean wounds induced in rat's skin. Methods: One hundred and twenty male rats of about 16 weeks old was divided into five groups: G1) saline solution (0.9%); G2) sunflower oil; G3) ozonated sunflower oil; G4) ozone bagging; G5) association of ozonated sunflower oil and ozone bagging. The wounds were evaluated through macroscopic, morphometric, histopathologic, and tensile strength analyses. Results: Analysis among groups showed a lower percentage of wound contraction in G1 compared to G4 only in M7D. The tensile strength of the wounds showed differences among groups in the seventh (M7D) and the 14th (M14D) postoperative day, and among time points in G1 (M14D > M7D). The elongation of the wounds showed differences in G3 (M7D > M14D). Histological evaluation of the wounds showed significant change in bleeding, mixed to mononuclear infiltrate, congestion, and tissue disorganization for tissue organization between groups and time points. Conclusions: Ozone therapy applied topically and/or by bagging was not deleterious to the healing of clean wounds induced in rat's skin, but ozone bagging showed the best contribution to the healing process.

Background: Paracoccidioidomycosis (PCM) is a severe granulomatous disease. The hallmark of this mycosis is fibrin degradation and granuloma formation as a result of a wound-healing process in the context of excessive inflammation. Therefore, as the content of collagen can be assessed by the methodology described in this manuscript, we propose that the content of hydroxyproline (HYP) be employed as a new and efficient measurement of granulomatous lesions developed. To estimate the level of HYP the major byproduct of the degradation process, we hypothesized that this simple and efficient technique could serve as a marker of disease severity. Methods: Five B10.A female mice were infected with P. brasiliensis and, after 15 days, the omentum was removed, subjected to histopathological analysis or processed (i.e. deproteinized and derivatized), and further analyzed on a reverse phase HPLC using a C-18 column. The omentum of five uninfected controls was also collected and similarly analyzed. Results: Infected mice showed numerous, disseminated paracoccidioidomycotic lesions, as well as marked collagen deposits, as observed in histopathologic analysis, and high levels of HYP. Normal uninfected mice showed no granulomas, little or no deposits of collagen fibers, and very low levels of HYP, as evaluated by HPLC. Our results show that the disease intensity as evaluated number and the morphology of the granulomatous lesions were correlated to the HYP levels using small tissue samples from the omentum, the main target organ of P. brasiliensis. Conclusions: Here we propose an alternative methodology to follow disease evolution and, to some extent, fungal load in experimental P. brasiliensis infection and suggest its usefulness to other diseases with pronounced fibrin degradation.(AU)

As neoplasias cutâneas são as mais comuns em cães e são diversas as suas classificações. O objetivo deste estudo foi avaliar a acurácia da agulha de corte Super Core II® (14-ga x 9cm) no diagnóstico histopatológico de nódulos cutâneos de origem epitelial em cães, quando comparada aos resultados histopatológicos das peças cirúrgicas analisadas. Foram analisados histologicamente 32 nódulos cutâneos. Os espécimes de biópsia e nódulos cutâneos foram classificados segundo a World Health Organization (WHO) e 71,87% dos nódulos foram identificados como neoplasias. A core biopsy mostrou sensibilidade de 85,2%, especificidade de 100%, valor preditivo positivo de 100% e valor preditivo negativo de 55,55% para o diagnóstico de neoplasias. O teste exato de Fisher revelou p<0,0001 para a identificação de neoplasias e de tumores malignos, confirmando que houve concordância dos resultados obtidos pelos dois métodos na classificação dos tumores. Sendo assim, esta biópsia é um método eficaz para a classificação de tumores cutâneos de origem epitelial.


Skin tumors are the most common neoplasms in dogs and there are several classifications of them. This study aimed to evaluate the accuracy of cut needle Super Core II (14-ga x 9cm) in the histopathologic diagnosis of cutaneous nodules of epithelial origin in dogs when compared with the histopathologic findings of surgical specimens analyzed. A total of 32 skin nodes were histologically analyzed. The biopsy specimens and skin nodules were classified according to the World Health Organization (WHO) and there were 71,87% of nodes identified as neoplasms. To diagnose neoplasms, the core biopsy showed 85.2% sensitivity, 100% specificity, a positive predictive value of 100%, and a negative predictive value of 55.55%. The Fisher exact test showed p<0.0001 for the identification of neoplasms and malignant tumors, which confirms the concordance between the results obtained by both methods in the classification of tumors. Therefore, the biopsy is an effective method for the classification of cutaneous tumors of epithelial origin.

ABSTRACT: Thoracoscopy is replacing open lung biopsies because it is less invasive, usually the technique is done using three portals and intracorporeal suture technique. This study described the feasibility and efficacy of a novel pre tied loop ligature and to propose a thoracoscopic access strategy with two portals to perform lung biopsy in patients under 5 kg. Ten rabbits were positioned in dorsal recumbency. Total thoracoscopic lung biopsy was performed using a combined transdiaphragmatic approach and a right intercostal approach. A pre tied loop ligature was placed to perform a caudal lung lobe biopsy. Insufflation of the thoracic cavity was not performed. The total surgery time was 41.4 ± 14.5 min. The procedure was carried out free of complications that prevented slippage or tightening the knot or that made it come loose after the lung biopsy; there was no serious complication during the surgical procedure. The samples obtained averaged 1x0.64x0.45 cm (Length, Width, Depth) and were considered satisfactory according to the histopathologic evaluation. Thorax radiographs taken before and after the surgeries were compared and showed no pneumothorax or hemothorax. Necropsy confirmed no knot failure occurred at the biopsy site. The use of the novel pre tied loop ligature is a safe and effective technique, avoiding problems with the limited size of the thoracic cavity in small patients.


RESUMO: A toracoscopia está substituindo as biópsias pulmonares abertas por ser menos invasiva. Usualmente a técnica é feita utilizando três portais e ligaduras feitas intra corporalmente. O objetivo deste estudo foi descrever a viabilidade e eficácia de uma nova ligadura com alça pré-amarrada e propor uma estratégia de acesso toracoscópico com dois portais para realizar biópsia pulmonar em pacientes com menos de 5kg. Dez coelhos foram posicionados em decúbito dorsal. A biópsia pulmonar por toracoscopia total foi feita pela abordagem combinada transdiafragmática e abordagem intercostal direita. Uma nova ligadura pré-amarrada foi colocada para realizar uma biópsia do lobo pulmonar caudal. Não foi realizada insuflação da cavidade torácica. O tempo total de cirurgia foi de 41,4 ± 14,5 min. Os procedimentos foram realizados sem complicações que impedissem o escorregamento ou aperto do nó ou que o soltassem após a biópsia pulmonar; não houve nenhuma intercorrência grave durante o procedimento cirúrgico. As amostras coletadas tinham tamanho médio 1x0,64x0,45 cm e foram consideradas satisfatórias de acordo com a avaliação histopatológica. As radiografias de tórax feitas antes e depois das cirurgias foram comparadas e não mostraram pneumotórax ou hemotórax. A necropsia confirmou que não ocorreu falha do nó no local da biópsia. Conclui-se que uso da nova ligadura com alça pré-amarrada é uma técnica segura e eficaz, evitando problemas com o tamanho limitado da cavidade torácica em pacientes pequenos.

Resumo

The article describes the case of a 3 year 6 months old Guinea Pig (Cavia porcellus) that was assisted at the Wildlife and Exotic Animal Practice of the Faculty of Veterinary Sciences, National University of Asunción. The patient presented a hard mass in the right hind limb extending from the proximal portion of the femur, projecting caudally to the proximal tuberosity of the calcaneus, with an increase in temperature, and a marked increase in sensitivity. Radiographic diagnosis refers to a mass with amorphous areas of mineral density. Fine needle aspiration puncture revealed an undifferentiated malignant proliferative process, requiring histopathological analysis to characterize the process. Disarticulation was performed and subsequent histopathologic diagnosis under hematoxylin-eosin and Masson's trichrome stain revealed fibroblastic osteosarcoma.(AU)

Bovine gammaherpesvirus 6 (BoGHV6), formerly known as bovine lymphotropic virus, belongs to the genus Macavirus, which includes the porcine lymphotropic herpesviruses (PLHV) and members collectively referred to as malignant catarrhal fever virus (MVFV) group. This study investigated the possible participation of BoGHV6 in the development of intestinal and pulmonary diseases of buffaloes. Intestinal and pulmonary fragments were obtained from 19 buffaloes from Goiás, Central-western Brazil. Histopathology revealed interstitial pneumonia in all pulmonary fragments evaluated, while two of these had suppurative bronchopneumonia. Furthermore, all intestinal fragments demonstrated atrophic enteritis. BoGHV6 DNA was amplified from 31.6% (6/19) of the buffaloes with interstitial pneumonia and in 26.3% (5/19) with atrophic enteritis. The phylogenetic evaluation revealed that the strain of BoGHV6 amplified from these buffaloes had 100% nucleotide (nt) sequence identity with the reference strains of BoGHV6, but only 69.5 to 73.7% and 65.8 ­ 69.5% nt sequence identity with members of the MCFV and the PLHV, respectively. Additionally, molecular assays to detect important pathogens of ruminants did not amplify the respective nucleic acids in the tissues evaluated. These results demonstrated that BoGHV6 was amplified from tissues of buffaloes with histopathologic diagnoses of interstitial pneumonia and atrophic enteritis, suggesting the possible participation of this virus in the development of the intestinal and pulmonary lesions herein identified.(AU)


Gamaherpesvírus bovino 6 (BoGHV6), anteriormente conhecido como vírus linfotrópico bovino, pertence ao gênero Macavirus, que inclui o herpesvírus linfotrópico dos suínos (PLHV) e membros coletivamente referidos como vírus da febre catarral maligna (VMVF). Este estudo investigou a possível participação do BoGHV6 no desenvolvimento de doenças intestinais e pulmonares em búfalos. Fragmentos intestinais e pulmonares foram obtidos de 19 búfalos de Goiás, Centro-Oeste do Brasil. A histopatologia revelou pneumonia intersticial em todos os fragmentos pulmonares avaliados, sendo que dois deles apresentavam broncopneumonia supurativa. Além disso, todos os fragmentos intestinais demonstraram enterite atrófica. O DNA do BoGHV6 foi amplificado em 31,6% (6/19) dos búfalos com pneumonia intersticial e em 26,3% (5/19) com enterite atrófica. A avaliação filogenética revelou que a cepa de BoGHV6 amplificada desses búfalos tinha 100% de identidade de sequência de nucleotídeos (nt) com as cepas de referência de BoGHV6, mas apenas 69,5 a 73,7% e 65,8 ­ 69,5% de identidade de sequência nt com membros do VMCF e o PVHS, respectivamente. Adicionalmente, ensaios moleculares para detectar importantes patógenos de ruminantes não amplificaram os respectivos ácidos nucléicos nos tecidos avaliados. Esses resultados demonstraram que BoGHV6 foi amplificado a partir de tecidos de búfalos com diagnóstico histopatológico de pneumonia intersticial e enterite atrófica, sugerindo a possível participação desse vírus no desenvolvimento das lesões intestinais e pulmonares aqui identificadas.(AU)

Background: Pancreatic cysts are liquid accumulations, rich in enzymes or blood, surrounded by epithelial tissue that develop from a variety of causes, including injury, neoplasm, or congenital disease. In most cases, its diagnosis is incidental, as patients present with non-specific symptoms or no symptoms at all. Imaging exams can be used to elucidate the structure located along the topography of the pancreas and, based on this, to plan the patient's treatment. Treatment consists of surgical removal, drainage, monitoring, and/or symptom control. The aim of this paper was to report a case of pancreatic cyst in a 9 months old female cat, to show the diagnostic methods and the success of the surgical therapeutic planning based on the clinical assessment and complementary tests. Case: A 9 months old feline patient, with no defined breed, presenting with sporadic episodes of emesis, was seen at the Harmonia Veterinary Hospital, located in the city of Recife/Pernambuco. During the anamnesis, it was reported that the patient had been experiencing bouts of emesis and abdominal discomfort for four months, but that her behavior and appetite remained normal. As the patient had been adopted and was of unknown origin, it is worth pointing out that there is no way of knowing whether her parents had any similar alterations. Clinical evaluation revealed discomfort on palpation and a firm and mobile structure in the right upper quadrant of the abdomen. The ultrasound scan showed the presence of a structure in the right lobe of the pancreas, characterizing a pancreatic cyst associated with pancreatic enlargement, without clear involvement of adjacent structures, which needed to be complemented by a CT scan. In this case, a single cystic lesion was found with no evidence of adjacent organ involvement, and no peripancreatic inflammation was noted. Laboratory tests, including those for the feline retroviruses FIV and FeLV, showed no significant changes. In view of the results, the patient was referred for exploratory laparotomy to evaluate the adjacent tissues and excise the cyst for histopathologic analysis, as well as to evaluate the other organs in their macroscopic aspects for possible changes not seen on imaging studies. After surgery, the patient remained in the intensive care unit and was monitored for 120 days with no clinical changes. Discussion: The diagnosis of feline pancreatic cyst was based on clinical signs, which although non-specific were the most common among the reported cases, evaluation by abdominal ultrasound and computed tomography, and definitively by exploratory laparotomy surgery and histopathology showing simple columnar epithelial tissue that may have formed from obstruction due to anomalous formation of one of the pancreatic ducts leading to neoformation with fluid content. Other forms of injury such as inflammation or trauma, congenital diseases such as feline polycystic disease, or neoplasms were ruled out by complementary examinations and patient history. The adopted therapy, consisting of symptomatic treatment with analgesics, antiemetics and surgical excision, showed satisfactory and curative results after one hundred and twenty days, with careful follow-up, with periodic evaluations and without any clinical alterations or complications after the procedure. Histopathology showed no evidence of malignancy, only the presence of simple columnar tissue associated with the epithelial tissue of the pancreatic parenchyma.

Porcine enzootic pneumonia (PES), mainly caused by the bacteria Mycoplasma hyopneumoniae, is the main cause of respiratory problems in pigs. Infection by M. hyopneumoniae leads to production losses and the predisposition of affected animals to secondary infections, which may result in the condemnation of carcasses and organs due to lung lesions at the time of slaughter. The objective of the research was to evaluate the infection by M. hyopneumoniae in pigs submitted to slaughter in São Luís Island/MA, using molecular and histopathological diagnostic methods. One hundred fifty lung samples were collected from inspected (n=65) and non-inspected (n=85) slaughter pigs on São Luís Island, Maranhão, from July 2019 to August 2021. Of the 150 DNA samples collected, 121 showed an amplified product for Cyt B in the PCR assay. Thus, 121 samples were submitted to qPCR of M. hyopneumoniae, of which 44 (36.36%) showed positive results. The mean amount of bacterial load ranged from 1.20 × 101 to 7.20 × 104, with a mean of 1.73 × 104 copies. Of the reagent samples, 81.81% (36 samples) were obtained from non-inspected slaughter, while 18.18% (8 samples) were obtained from slaughterhouses. In the histopathological analysis, 44 positive qPCR samples were evaluated, of which 28 (63.63%) presented results compatible with the main inflammatory process associated with the presence of M. hyopneumoniae, that is, bronchial-associated lymphoid tissue hyperplasia (BALT). Three samples that showed the highest bacterial load (qPCR: 5.63 × 10³, 2.19 × 104 and 7.23 × 104) showed more evident lesions in this study. The microscopic findings associated with the quantifications indicated a relationship between the amount of bacterial load and the presence of microscopic lesions; higher bacterial load in lung tissue is associated with increased histopathologic staining for BALT hyperplasia. In conclusion, the results point to the circulation of the etiological agent in the sampled animals and the need for preventive measures on pig farms in Maranhão with the involvement of producers, sanitary defense and inspection agencies.


A pneumonia enzoótica suína (PES), causada principalmente pela bactéria Mycoplasma hyopneumoniae, é a principal causa de problemas respiratórios em suínos. A infecção por M. hyopneumoniae leva a perdas produtivas e a predisposição dos animais acometidos a infecções secundárias, o que pode resultar em condenação de carcaças e órgãos por lesões pulmonares no momento do abate. O objetivo da pesquisa foi avaliar a infecção por M. hyopneumoniae em suínos submetidos ao abate na Ilha de São Luís, por meio de métodos diagnósticos moleculares e histopatológicos. Para isso, foram coletadas 150 amostras de pulmão de suínos de abate inspecionado (n=65) e não inspecionado (n=85) na Ilha de São Luís/Maranhão, no período de julho de 2019 a agosto de 2021. Das 150 amostras de DNA coletadas, 121 apresentaram produto amplificado para Cyt B no ensaio de PCR. Assim, 121 amostras foram submetidas à qPCR de M. hyopneumoniae, das quais 44 (36,36%) apresentaram resultados positivos. A quantidade média de carga bacteriana variou de 1,20 × 101 a 7,20 × 104, com média de 1,73 × 104 cópias. Das amostras reagentes, 81,81% (36 amostras) foram obtidas de abate não inspecionado, enquanto 18,18% (8 amostras) foram obtidas em abatedouro. Na análise histopatológica, foram avaliadas 44 amostras positivas para qPCR, das quais 28 (63,63%) apresentaram resultados compatíveis com o principal processo inflamatório associado à presença de M. hyopneumoniae, ou seja, hiperplasia do tecido linfóide associado ao brônquio (BALT). Três amostras que apresentaram maior carga bacteriana (qPCR: 5,63 × 10³, 2,19 × 104 e 7,23 × 104) foram mais evidentes neste estudo. Os achados microscópicos associados às quantificações indicaram uma relação entre a quantidade de carga bacteriana e a presença de lesão microscópica; a maior carga bacteriana no tecido pulmonar está associada a maior alteração histopatológica para hiperplasia BALT. Em conclusão, os resultados obtidos sinalizam para a circulação do agente etiológico nos animais amostrados e a necessidade de medidas preventivas nas criações de suínos do estado do Maranhão com envolvimento dos produtores, órgãos de defesa sanitária e inspeção.

Resumo

Clinical History: Multiple fragments of various organs from the necropsy of an adult sloth (Choloepus hoffmani) were received. The animal, prior to its death, had presented decay and poor general body condition, which is why it was euthanized. Necropsy Findings: According to the referring veterinarian, at necropsy, irregular and locally extensive hard yellowish lesions, were observed in the subcutaneous tissue of the thorax and abdomen (Figs. 1 and 2). Similar lesions were observed in the tunica media of the aorta (Fig. 3). Microscopically, the kidney had the aspect shown in Fig. 4. Resumo

Porcine respiratory disease complex is a major health concern for the porcine industry, causing significant economic loss. In this study, a total of 156 samples from pigs referred to a diagnostic laboratory in Brazil for 15 months were analyzed by histopathology, bacterial isolation, PCR, and immunohistochemistry. Multiple infections were common, so 42.3% of the pigs had more than one pathogen detected in the lungs. Swine influenza virus was detected in 25.0% of the cases. Porcine circovirus type 2 was detected in 7.1% of the pigs, which was often associated with Pasteurella multocida. In addition, one case of porcine circovirus type 3 infection associated with granulomatous pneumonia was diagnosed. Bacteria were isolated in 125 cases, namely Pasteurella multocida (34.0%), Glaesserella (Haemophilus) parasuis (35.2%), Streptococcus suis (13.5%), and Actinobacillus pleuropneumoniae (7.7%). Mycoplasma hyopneumoniae was identified in 7.0% of the cases, and 18.6% of pigs carried Salmonella sp. The most common patterns of pulmonary inflammation were broncopneumonia, bronchointerstitial pneumonia, and pleuritis, in that order. This study demonstrated that histopathology is an efficient tool along with other laboratorial diagnostic tests for establishing an etiologic diagnosis in cases of porcine respiratory disease complex.


O complexo de doenças respiratórias de suínos é um dos principais problemas sanitários na suinocultura, causando perdas econômicas significativas. O presente estudo incluiu amostras de 156 suínos, que foram encaminhados a um laboratório de diagnóstico no Brasil, durante um período de 15 meses, sendo realizados histopatologia, isolamento bacteriano, PCR e imuno-histoquímica. Coinfecções por múltiplos patógenos foram comuns, correspondendo a 42,3% dos animais, que tiveram mais de um agente identificado nos pulmões. O vírus da influenza suína foi detectado em 25,0%. O circovírus suíno tipo 2 foi detectado em 7,1% dos animais, frequentemente associado à Pasteurella multocida. Além disso, foi diagnosticado um caso de circovírus suíno tipo 3 associado à pneumonia granulomatosa. Foram isoladas bactérias em 125 casos, a saber: Pasteurella multocida (34,0%), Glaesserella (Haemophilus) parasuis (35,2%), Streptococcus suis (13,5%) e Actinobacillus pleuropneumoniae (7,7%). Mycoplasma hyopneumoniae foi identificado em 7,0%, e 18,6% dos animais tiveram isolamento de Salmonella sp. Os padrões mais frequentes de inflamação pulmonar foram: broncopneumonia, pneumonia broncointersticial e pleurite, nesta ordem. Este estudo demonstrou que a histopatologia é uma ferramenta eficiente, juntamente a outras técnicas laboratoriais de diagnóstico, para o estabelecimento de diagnóstico etiológico em casos do complexo de doenças respiratórias de suínos.

Background: Hemangiosarcoma is a malignant vascular tumor commonly described in veterinary ophthalmology as emerging at the margin of the third eyelid and bulbar conjunctiva. The primary corneal attachment of the tumor is considered rare, as the cornea is an avascular structure, but there are sparse reports of primary occurrences in the cornea of dogs, cats, and horses. No reports were found in wild animals. The Tapirus terrestris, commonly known as Tapir, is a perissodactyl mammal that inhabits South America regions, being considered the largest terrestrial mammal of Brazil and one of the most popular animals in zoos in various countries. This paper aims to report the first case of primary corneal hemangiosarcoma in a 25-year-old female Tapir (Tapirus terrestris) held in captivity. Case: A 25-year-old Tapirus terrestris female presented an irregular mass in the right cornea, mucopurulent secretion, and inconclusive previous cytological evaluation. It also had a history of a milky cornea for many years, and in the last 12 months, a tissue growth was observed, as well as the occurrence of mucopurulent secretion. The menace response in the right eye was negative. In the ophthalmologic examination, a red-colored, lobed, hemorrhagic, and ulcerated mass occupying approximately 90% of the cornea was observed, and in the peripheral cornea, there was an intense discoloration. Due to the extension of the mass and visual loss, the transpalpebral enucleation was performed under general inhalation anesthesia. The histopathological analysis evidenced a vascular tumor restricted to the central peripheral cornea, composed of blood vessels of small and large caliber, showing polygonal endothelial cells with moderate atypia, anisocytosis, and anisocariasis, in addition to dense stroma and inflammatory cells. The mass occupied the anterior corneal stroma, and there was no involvement of bulbar conjunctiva, sclera, or intraocular structures. The immunohistochemical study revealed the positivity of neoplastic cells for CD31, and the proliferative index of the lesion was evaluated by the Ki-67 as inferior to 10%. Discussion: The histopathologic and immunohistochemical examinations evidenced a hypercellular lesion, and the diagnosis of primary corneal hemangiosarcoma (HSA) was possible due to the significant cellular atypia identified in the lesion. To the authors' knowledge, this is the first case of corneal hemangiosarcoma described in Tapirus terrestris. The HAS is a malignant neoplasm originating in the vascular endothelial cells. There is a significant correlation between the HSA and prolonged exposure to solar radiation. The tapir of this report lived in a high-altitude region with exposure to ultraviolet rays; therefore, it might be an important predisposing factor for the HSA emergence. The chosen treatment for vascular tumors according to the visual prognostic is the lamellar keratectomy; however, the transpalpebral enucleation was the elected treatment since the eye was not visual, the neoplasm had a great extension, the animal was in advanced age and lived in captivity, and it was difficult to handle the animal without sedation to perform adjuvant therapies. Since the mass corresponded to 90% of the cornea, total removal with free edges would not be possible as described in the literature, and, therefore, there was an increased risk of relapse. In the follow-up after 3 years of surgical excision, the animal remained free of metastasis, reinforcing the diagnosis of primary corneal hemangiosarcoma.

Background: Being the major cause of bovine abortion in the world, Neosporosis is considered to be a very important protozoal infection in dairy cattle. Vertical transplacental transmission is the major route of the infection causing either abortion or birth of calves with persistent infection. As the seropositivity in individual cows and in fetal serology only indicate exposure to the protozoa, the diagnosis of the infection has to be based on histopathology of aborted fetuses. Additional techniques such as immunohistochemistry (IHC) and PCR are required for the detection of the etiological agent. The purpose of the current study was to diagnose Neospora caninum infection in aborted bovine fetuses in Trakya Region of Turkey. For this purpose, serological, histopathological, IHC, and PCR methods were used. Materials, Methods & Results: The blood samples and the fetuses of 55 aborted dairy cattle from various farms located in 3 provinces of Trakya, Turkey constituted the material of the present study. The sera obtained from the blood samples were tested using a Neospora caninum Antibody Test Kit cELISA and anti-N. caninum antibodies were detected in the sera of the dams of the 8 aborted fetuses (8/55; 14.54%). Following the necropsy, samples from the brain, heart, liver, lung, kidney, spleen, and placenta of 55 fetuses were routinely processed for histopathological examination and evaluated under a light microscope. Nonsuppurative encephalitis (15/55; 27.27%), necrosis (5/55; 9%) and gliosis (1/55; 1.8%) in the brain, mild to severe nonsuppurative myocarditis and epicarditis (14/55; 25.45%), and portal to mid-zonal nonsuppurative hepatitis (13/55; 23.63%) were the relevant findings. PCR analysis was performed on fresh frozen fetal tissues. Nested PCR detected N. caninum DNA in the brain, heart, liver, lung, and kidney tissues of 6 fetuses (6/55; 10.9%). IHC was performed on the brain, heart, and liver tissues of all the fetuses using avidin-biotin-complex peroxidase method. Immunoreactivity was observed in the brain of 1 fetus (1/55; 1.8%). Discussion: In the present study, histopathological, immunohistochemical and PCR analyses were performed to detect N. caninum in 55 spontenously aborted bovine fetuses in Trakya Region, Turkey. Histopathologic hallmark of the study was nonsuppurative inflammation found mostly in the brain, heart and liver followed by kidneys and lungs. No protozoa was observed in the microscopic examination supporting the fact that definitive diagnosis of N. caninum infection requires ancillary techniques such as IHC and PCR. Nested PCR detected N. caninum DNA in the tissues of 6 fetuses (6/55; 10.9%). Brain was the most reliable organ for detection by PCR (6/6; 100%), compatible with the previous reports. IHC diagnosis revealed only 1.8% positivity in the present study which was remarkably lower than found in the previous studies. Even though histopathology in conjunction with IHC are accepted as the "gold standard" methods to detect N. caninum infection in aborted bovine fetuses, there are studies claiming that IHC is relatively insensitive in the diagnosis of neosporosis as parasite numbers can be low and thus, false negative results can be obtained. Other factors affecting the sensitivity of the technique are thoroughly discussed by many authors. Supportively, the findings of the current study showed that using both IHC and PCR as complementary techniques, increases the success of detection of N. caninum as recommended in previous studies. In conclusion, the present study demonstrated the first molecular diagnosis of Neospora caninum infection in bovine aborted fetuses in Trakya Region of Turkey which has a critical geographical location bordering Europe.

The aim of the present study was to evaluate the post-vaccinal reaction to two lentogenic vaccine strains of Newcatle disease virus (NDV) and a recombinant turkey herpesvirus (rHVT) vaccine expressing the fusion glycoprotein of NDV in broiler chickens through histomorphometric and histopathologic analyses of the trachea. The experiment involved 245 chicks housed in randomized blocks with three different enclosures under controlled conditions of temperature, light and ventilation. Each enclosure represented a vaccine strain and was divided into groups according to the administration route. Each block also had its own control group composed of unvaccinated birds. The vaccine strains PHY.LMV.42 (PL42) and La Sota (LS) were selected according to the Intracerebral Pathogenicity Index (ICPI) and the rHVT-NDV Serotype 3 strain (ST3) was selected for representing non-NDV infection. At two, four, seven, 14 and 21 days post vaccination, fragments from the middle third of the trachea were collected and submitted to routine histological processing. For the histomorphometric analysis, the slides were photographed, and the thickness of the tracheal mucosa was measured. Statistical analysis involved two-way ANOVA and Tukey's post-hoc test with a 5% significance level. For the histopathological evaluation, lesions were described as to the degree of intensity and distribution. At four and 14 days post vaccination with the LS strain administered by the ocular route, the means of thickening of the tracheal mucosa (20.85±7.31µm and 26.97±5.50µm, respectively) were significantly higher (p<0.05) than for all other strains, which was related to the severe histopathological lesions found in this group, characterized by hyperemia, hyperplasia of the mucous glands, moderate deciliation and multifocal lymphohistiocytic inflammatory infiltrate. At 21 days, broiler chickens vaccinated with the ST3 strain showed more discrete lesions and less thickening of the tracheal mucosa (23.23±7.62µm; p<0.05) in comparison with other studied strains. The lesions found in this group were only hemorrhage, deciliation and mild focal lymphocytic inflammatory infiltrate. The results of the histomorphometry and histopathology of the trachea indicated that vaccination with rHVT-NDV Serotype 3 strain induced lower degree post-vaccine tracheal lesions compared to other vaccine strains analyzed in this study.


Objetivou-se avaliar a reação pós-vacinal de duas estirpes lentogênicas do vírus da doença de Newcastle (VDN) e uma vacina recombinante de herpesvirus de perus (rHVT) que expressa a glicoproteína de fusão de VDN em frangos de corte por meio da histomorfometria e histopatologia da traqueia. Foram utilizados 245 pintos alojados em blocos ao acaso, sendo três galpões distintos em condições controladas de temperatura, luz e ventilação. Cada galpão representou uma cepa vacinal, onde foram divididos por grupos de acordo com a via de administração. Todos os blocos possuíam um grupo controle composto por aves não vacinadas. As cepas vacinais PHY.LMV.42 (PL42) e La Sota (LS) utilizadas foram selecionadas de acordo com o Índice de Patogenicidade Intracerebral (IPIC) e a cepa Sorotipo 3 (ST3), da vacina rHVT-VDN foi selecionada por não representar infecção do VDN. Aos dois, quarto, sete, 14 e 21 dias pós-vacinação, fragmentos do terço médio da traqueia foram coletados e posteriormente processados conforme rotina histológica. Para análise histomorfométrica da mucosa traqueal, as lâminas foram fotografadas e realizadas as mensurações da espessura da mucosa traqueal sendo aplicado teste de análise de variância a dois critérios (ANOVA) e utilizando o post-hoc de Tukey com nível de significância de 5%. Para a avaliação histopatológica foram observadas a presença de lesões microscópicas e estas foram descritas quanto ao grau de intensidade e distribuição. Aos quatro e quatorze dias pós-vacinação com a cepa LS administrada por via ocular, as médias do espessamento da mucosa traqueal (20,85±7,31µm e 26,97±5,50µm, respectivamente) foram significativamente maiores (p<0,05) quando comparada a todas as demais cepas utilizadas, isto se deve às severas lesões histopatológicas encontrados neste grupo, caracterizadas por hiperemia, hiperplasia das glândulas mucosas, deciliação moderada e infiltrado inflamatório linfohistiocitário multifocal moderado. Já aos 21 dias as aves vacinadas com a cepa ST3 apresentaram lesões mais discretas e menor espessamento da mucosa da traqueia (23,23±7,62µm; p<0,05) em comparação às demais cepas estudadas. As lesões encontradas neste grupo foram apenas hemorragia, deciliação e infiltrado inflamatório linfocitário focal discreto. Os resultados da histomorfometria e da histopatologia da traqueia indicou que a vacinação com a rHVT-NDV, cepa Sorotipo 3 induziu menor grau de lesões pós-vacinais na traqueia comparada a outras cepas vacinais analisadas nesse estudo.

ABSTRACT: This study assessed microscopic morphology of protozoan and metazoan parasites, as well as parasite-associated histopathologic changes in five Brazilian free-ranging armadillos. Three armadillos had intra sarcolemmal cysts of Sarcocystis sp. in skeletal muscles without microscopic changes. One Dasypus novemcinctus was found parasitized with a nematode morphologically compatible with an oxyurid in the small intestine. One Dasypus sp. had neutrophilic enteritis associated with adult and larval stages of Strongyloides sp. and one D. novemcinctus had multiple embryonated eggs free in the lumen of the small intestine with mild neutrophilic enteritis. These findings represent a contribution for expanding our knowledge on parasitic diseases of armadillos.


RESUMO: Este estudo avaliou a morfologia microscópica de parasitos protozoários e metazoários, bem como lesões associadas ao parasitismo em cinco tatus de vida livre no Brasil. Três tatus tinham cistos de Sarcocystis sp. Intra-sarcolemal em músculos esqueléticos sem alterações microscópicas. Um Dasypus novemcinctus estava parasitado com um nematodo morfologicamente compatível com oxiurideo no intestino delgado. Um Dasypus sp. apresentou enterite neutrofílica associada com estágios larvais de Strongyloides sp. e um D. novemcinctus apresentou múltiplos ovos embrionados livres no lúmen do intestino delgado, associado a enterite neutrofílica discreta. Estes achados representam uma contribuição para a expansão do conhecimento sobre doenças parasitárias de tatus.

ABSTRACT: A high prevalence of pneumonic lesions has been reported to affect slaughtered pigs in southern Brazil. In order to identify which microorganisms have been causing those lesions, 30 pig lungs presenting pneumonic gross lesions were collected from five different slaughterhouses, totaling 150 lungs. Samples for bacterial isolation, molecular, histopathologic and immunohistochemistry (IHC) evaluation were taken from each lung. The pneumonic lesion scoring ranged from 1.53 to 2.83. The most frequent histopathological lesions found was the concomitant Influenza A virus (IAV) and Mycoplasma hyopneumoniae infection, corresponding to 55.3% (83/150), and Pasteurella multocida type A was isolated in 54.2% (45/83) of these cases. In 102 samples (68%), there was histopathologic suggestion of involvement of more than one infectious agent. M. hyopneumoniae was the most frequent agent associated with pneumonic lesions, being present in 92.1% (94/102) of the lungs with coinfections, followed by IAV in 89.2% (91/102). Besides the coinfections, IAV lesions were observed also in six samples without another pathogenic microorganism detected. A total of 46 samples with acute and subacute IAV suspected lesions in histopathological examination were assessed for IHC and real time RT-PCR for IAV. A total of 35% (16/46) of them were positive by IHC and 13% (6/46) by real time RT-PCR. Regarding M. hyopneumoniae, 79.3% (119/150) of samples were positive by qPCR and 84.9% (101/119) of them also presented M. hyopneumoniae suspected lesions in the histopathological examination. The results of this study suggest the importance of IAV in respiratory diseases in finishing pigs, even though this virus is more frequently reported in the nursery phase. In addition, our results emphasize the importance of lung coinfections in finishing pigs.


RESUMO: Lesões sugestivas de pneumonia são frequentemente encontradas em altas prevalências em suínos abatidos no sul do Brasil. Para identificar quais microrganismos causam essas lesões, foram coletados 30 pulmões de suínos com lesão macroscópica sugestiva de pneumonia em cinco frigoríficos diferentes, totalizando 150 pulmões. Amostras para isolamento bacteriano, avaliação molecular, histopatológica e imuno-histoquímica (IHC) foram coletadas de cada pulmão. O escore de lesão pulmonar variou entre 1,53 a 2,83. O achado histopatológico mais observado foi a lesão sugestiva de infecção concomitante pelo vírus Influenza A (IAV) e Mycoplasma (M.) hyopneumoniae, correspondendo a 55,3% (83/150), e em 54,2% (45/83) desses casos Pasteurella (P.) multocida tipo A foi isolado. Em 102 amostras (68%), houve lesão histopatológica sugestiva do envolvimento de mais de um agente infeccioso. M. hyopneumoniae foi o microrganismo mais frequente associado a lesões de pneumonia, estando presente em 92,1% (94/102) dos pulmões com coinfecções, seguido de IAV, que foi encontrado em 89,2% (91/102). Além das coinfecções, lesões de IAV foram observadas em mais seis amostras que não aparentavam envolvimento de outro agente infeccioso. Um total de 46 amostras com suspeita de lesão aguda e subaguda de IAV no exame histopatológico foram avaliadas para IHC e RT-PCR em tempo real para IAV e 35% (16/46) delas foram positivas por IHC e 13% (6/46) foram positivas por RT-PCR em tempo real. Com relação a M. hyopneumoniae, 79,3% (119/150) das amostras foram positivas por qPCR e 84,9% (101/119) delas também apresentaram lesões suspeitas de M. hyopneumoniae no exame histopatológico. Os resultados deste trabalho sugerem a importância do IAV como agente causador de pneumonias em suínos de terminação, embora esse vírus seja mais frequentemente relatado na fase de creche. Além disso, os achados deste trabalho demonstram a presença frequente de coinfecções pulmonares em suínos de terminação.

ABSTRACT: Histopathology is an old science that is still currently utilized for disease diagnosis and research. The routinely processed histologic slides stained with hematoxylin and eosin are still used worldwide in most if not every histopathology laboratory. The technique is inexpensive, quick to perform, and allows the diagnosis of a fantastic variety of tissue changes and diseases. Skills in description and interpretation in histopathology are a craft that can be learned by repeatedly and systematically observing simple rules. In this article, we offer a few advices to help trainees in veterinary pathology at the start of their careers. Those advices are drawn from our experience in the diagnostic pathology routine and from the veterinary pathology literature. To enhance the understanding of these important steps in the histopathologic description of tissues, we decided to illustrate most concepts expressed here. We hope that our effort can add a bit to the development of future pathologists. Just like Alice, let us follow the White Rabbit into his burrow for this challenging experience!


RESUMO: A histopatologia é uma ciência antiga, mas ainda usada para diagnosticar e investigar a patogênese de doenças. As lâminas histológicas processadas rotineiramente e coradas por hematoxilina e eosina ainda são utilizadas em virtualmente todos os laboratórios de histopatologia do mundo. A técnica não é cara, é de execução rápida e permite o diagnóstico de uma fantástica variedade de doenças. As habilidades em descrever e interpretar os achados histopatológicos é um ofício que pode ser aprendido pela observação repetida e sistemática de regras simples. Para ajudar os estudantes de patologia no início de suas carreiras, abordamos aqui algumas dessas regras, extraídas tanto de nossa experiência quanto da literatura relacionada à patologia veterinária. Para aumentar a compreensão dos tópicos, decidimos ilustrar praticamente todos os conceitos expressados neste manuscrito. Esperamos que nosso esforço possa contribuir um pouco para o desenvolvimento de aspirantes a patologistas. Assim como Alice no País das Maravilhas, vamos seguir o coelho branco até a sua toca para essa aventura desafiadora.

Resumo

The transmissible venereal tumour (TVT) is one of the most frequent neoplasias in dogs. This tumour has specific characteristics, and it is exclusively of canines. Its transmission occurs through viable neoplastic cell transplantation when in contact with mucosa or unhealthy skin and rarely metastasise. This paper aims to report a rare presentation of pulmonary metastasis of widespread transmissible venereal tumours in a Blue Heeler dog. The patient was cachectic, dyspnoeic, and dehydrated and had multiple skin and pharynx nodulations. The cytology of all cutaneous nodulations showed round vacuolated cells with large eccentric nuclei and loose chromatin, which is compatible with TVT’s microscopic characteristics. Owing to the clinical evolution and reserved prognosis, the patient was euthanized. Necroscopy revealed a mass in the right pulmonary caudal lobe. The mass showed the same histopathologic characteristic of the others: not encapsulated infiltrative neoplastic proliferation of round vacuolated cells. The atypical manifestation of cutaneous metastasis and mainly pulmonary metastasis, in this case, denote the importance of TVT inclusion as a differential in cutaneous neoplasia, even if they show distant organ metastasis. Therefore, it emphasised the importance of cytology and histology in the diagnosis of nodular affections.


O tumor venéreo transmissível (TVT) é uma das neoplasias mais frequentes em cães. Com características específi-cas, esse tumor é exclusivo da espécie canina. Sua transmição acontece através da transplantação de células neoplásicas viáveis quando em contato com mucosas ou pele não íntegra, raramente ocorrendo metástases. Este trabalho visa relatar a apresen-tação rara de metástase pulmonar de tumor venéreo transmissível generalizado em um cão Blue Heeler. O paciente apresen-tava-se caquético, dispneico, desidratado e com diversas nodulações cutâneas e em faringe. A citologia de todas as nodulações cutâneas evidenciaram células redondas vacuolizadas, com núcleo excêntrico grande e cromatina frouxa, compatível com as características microscópicas de TVT. Em decorrência da evolução clínica e prognóstico reservado, o paciente foi eutanasiado. No exame macroscópico foi evidenciado massa em lobo caudal do pulmão direito com as mesmas características histopatoló-gicas dos demais nódulos, proliferação neoplásica de células redondas vacuolizadas infiltrativas não encapsuladas. A manifes-tação atípica de metástases cutâneas e principalmente pulmonar neste caso denota a importância da inclusão do TVT como diferencial de neoplasias cutâneas mesmo com metástase em órgãos. Com isso ressalta-sea importância de exames de citopa-tologia e histopatologia no diagnóstico de afecções nodulares.

Resumo

Histopathology is an old science that is still currently utilized for disease diagnosis and research. The routinely processed histologic slides stained with hematoxylin and eosin are still used worldwide in most if not every histopathology laboratory. The technique is inexpensive, quick to perform, and allows the diagnosis of a fantastic variety of tissue changes and diseases. Skills in description and interpretation in histopathology are a craft that can be learned by repeatedly and systematically observing simple rules. In this article, we offer a few advices to help trainees in veterinary pathology at the start of their careers. Those advices are drawn from our experience in the diagnostic pathology routine and from the veterinary pathology literature. To enhance the understanding of these important steps in the histopathologic description of tissues, we decided to illustrate most concepts expressed here. We hope that our effort can add a bit to the development of future pathologists. Just like Alice, let us follow the White Rabbit into his burrow for this challenging experience!(AU)


A histopatologia é uma ciência antiga, mas ainda usada para diagnosticar e investigar a patogênese de doenças. As lâminas histológicas processadas rotineiramente e coradas por hematoxilina e eosina ainda são utilizadas em virtualmente todos os laboratórios de histopatologia do mundo. A técnica não é cara, é de execução rápida e permite o diagnóstico de uma fantástica variedade de doenças. As habilidades em descrever e interpretar os achados histopatológicos é um ofício que pode ser aprendido pela observação repetida e sistemática de regras simples. Para ajudar os estudantes de patologia no início de suas carreiras, abordamos aqui algumas dessas regras, extraídas tanto de nossa experiência quanto da literatura relacionada à patologia veterinária. Para aumentar a compreensão dos tópicos, decidimos ilustrar praticamente todos os conceitos expressados neste manuscrito. Esperamos que nosso esforço possa contribuir um pouco para o desenvolvimento de aspirantes a patologistas. Assim como Alice no País das Maravilhas, vamos seguir o coelho branco até a sua toca para essa aventura desafiadora.(AU)
