ABSTRACT: We described a case of rhinitis case caused by Exserohilum rostratum in a 3-year-old, mixed breed, afemale goat, presenting with decreased appetite, dyspnea, and face deformity, with evolution of approximately 2 months. Necropsy revealed an extensive brownish yellow, irregular, ulcerated, and friable focal lesion in the nasal cavity. Purulent secretion drained from the lesion, affecting the nasal vestibule and extending the caudal portion of the dorsal concha, associated with nasal meatus obstruction, destruction of the nasal septum, and nasal bone palatine and vomer. Histologically, the lesion was characterized as rhinitis and pyogranulomatous and necrotizing osteomyelitis (multifocal to coalescent, sharp, and chronic) and associated with a myriad of fungal structures. Morphologically, the hyphae were characterized by thin, slightly tortuous, and rarely septate walls (2-6 μm diameter), multiple chains of individual conidia or in groups (8-10 μm diameter), and pigmented chlamydoconidia (7.5-15 μm diameter). They showed positive results for the special histochemical techniques such as Fontana Masson, methenamine silver nitrate by Grocott, and periodic acid by Schiff. E. rostratum was identified as the causative agent using microbiological isolation associated with molecular identification. Pyrogranulomatous rhinitis caused by E. rostratum in goats is unprecedented, being the first isolation of the agent in animals in Brazil and the first case of nasal phaeohyphomycosis in the goats.

Cryopreservation is the best method of storing semen indefinitely, providing high flexibility of use. In addition, it is the primary means for forming a germplasm bank. However, freezing boar semen is still a challenge to be faced because, in addition to the characteristics of the spermatic membrane that cause deleterious effects to the cells during the freezing and thawing processes, it is still necessary to deal with the significant variability of results, whether between individuals, ejaculates from the same individual or even fractions of an even ejaculated. Because of this, different strategies have been investigated to obtain better results and thus increase the applicability of frozen boar semen at a commercial level. Furthermore, we believe it is possible to relate the cryotolerance capacity of a given sperm to its greater or lesser capacity to fertilize the oocyte.(AU)

The olive crop has a large diversity in cultivars around the world, whose identification have been based on leaf, stem or fruit traits. However, these traits may be influenced by the environment, which has led to the misidentification of cultivars. The variability of the pistil and pollen structure under electron scanning and light microscopy were studied using the cultivars 'Arbequina', 'Arbosana', 'Frantoio', 'Koroneiki', 'Manzanilla de Sevilla' and 'Picual', cultivated in Southern Brazil. The aim was to find unique patterns for their identification. This type of study has not been carried out in the conditions of Southern Brazil, where the identification of commercial cultivars sometimes is complex and confusing. There were significant differences in the variables, showing uniformity within each cultivar in different locations. The principal component analysis indicated a separation of cultivars according to similarities or micromorphological differences in the pollen wall, area of the lumen and the structural elements of the tectum. For the analysis of the pistil, the separation was even clearer, each cultivar individually forming a cluster, using as characters the length of the style in contrast to the length, width and area of the stigma. This evidenced little variation among individuals within the same cultivar. The information provided can contribute to the individual identification of very close olive tree cultivars in the conditions of Southern Brazil.(AU)

In pig farming, measurements of production parameters play a fundamental role in the success of the activity. Minimal differences in fertility between breeders can lead to less reproductive efficiency and, less productivity. However, assessing the fertility of each male and the early identification of subfertile males is a difficult task to be performed. Thus, the aim of this study was to evaluate the use of in vitro and in vivo parameters in the identification of subfertile males of the Landrace breed, aiming to collaborate with genetic improvement programs, routine optimization in the Genetic Diffusion Units (GDUs) and the results of performance. In experiment 1, an approach to identify males with subfertility was evaluated based on retrospective data. For this, the results (averages of birth rates, number of total births and average percentages of female and male piglets per litter) were evaluated for a total of 996 matings and 847 parturitions. The inseminations came from ejaculates of 32 males, who had at least 19 females inseminated with homospermic doses in the concentration of 2.5 x 109 total sperm from the same male. As for the birth rate (BR), an average of 85.47% ± 6.05 was observed with a group of median males, seven males that stood out and one individual (M32) with a performance of 58.06% ± 9.0. For the total number of piglets born (PB) the average was 13.41 ± 0.56, with three males with better performance and one (M32) with very poor performance (8.62 ± 0.59). In experiment 2, it was verified whether evaluations of inseminating doses (ID) of semen in vitro (motility and sperm morphology) after 96 hours of storage had correlations with fertility in vivo, which can be used to identify subfertile males. The evaluations were performed on 30 ejaculates regarding the means of BR and PB, considering only those who had at least 7 females inseminated. There were no correlations between the motility assessments and semen morphological changes and the reproductive parameters evaluated. The results obtained in vivo, referring to BR and PB, demonstrated that it was possible to identify differences between males, the individual (M32) had the worst results for the percentages of BR and PB. It is concluded that there are males of high and low fertility and that only the in vitro analyzes carried out in this study are not enough to categorize them, however, the evaluation of retrospective data was efficient for this purpose.(AU)

Prediction of bull fertility is critical for the sustainability of both dairy and beef cattle production. Even though bulls produce ample amounts of sperm with normal parameters, some bulls may still suffer from subpar fertility. This causes major economic losses in the cattle industry because using artificial insemination, semen from one single bull can be used to inseminate hundreds of thousands of cows. Although there are several traditional methods to estimate bull fertility, such methods are not sufficient to explain and accurately predict the subfertility of individual bulls. Since fertility is a complex trait influenced by a number of factors including genetics, epigenetics, and environment, there is an urgent need for a comprehensive methodological approach to clarify uncertainty in male subfertility. The present review focuses on molecular and functional signatures of bull sperm associated with fertility. Potential roles of functional genomics (proteome, small noncoding RNAs, lipidome, metabolome) on determining male fertility and its potential as a fertility biomarker are discussed. This review provides a better understanding of the molecular signatures of viable and fertile sperm cells and their potential to be used as fertility biomarkers. This information will help uncover the underlying reasons for idiopathic subfertility.(AU)

Background: Actinobacillus pleuropneumoniae (A. pleuropneumoniae) is one of the most important bacterial respiratory pathogens. It is the only etiological agent of porcine pleuropneumonia (PPP) or it appears as a secondary bacterial infection in the swine respiratory disease complex (PRDC). In Serbia, apart from the identification of serotype 2, no tests have been performed to establish the presence of other A. pleuropneumoniae serotypes in the pig population. The aim of this study was to perform genotyping of A. pleuropneumoniae isolates originating from pig farms in Serbia by apx genes and using multiplex polymerase chain reaction (mPCR). Materials, Methods & Results: Isolates of A. pleuropneumoniae examined in this study were obtained from lungs with macroscopically visible alterations characteristic of a A. pleuropneumoniae. A total of 46 isolates were examined. They were extracted from the lung tissue samples of pig carcasses from 9 farms across different parts of Serbia. Genotyping of isolates was performed in the previously described manner. Briefly, 5 pairs of oligonucleotide primers were used for amplification of 4 different apx genes which encode synthesis of exotoxins (ApxI , ApxII , ApxIII i ApxIV) characteristic for all A. pleuropneumoniae serotypes and biovars. Amplification of appropriate genome parts was performed with a reaction chain polymerase (PCR) in multiplex (m) format using appropriate diagnostic kits to extract DNA from bacteria and perform mPCR reaction. The results of genotyping of 46 isolates of A. pleuropneumoniae indicate the existence of a large number of different serotypes of A. pleuropneumoniae on the studied farms or that different serotypes of this microorganism circulate in the pig population in Serbia. In addition to the detection of dominant serotype 2, which was established on 7 farms, of which in 4 farms it was the only detected serotype, in the examined pig population the presence of serotypes 3, 5, 6, 7 and 9 was also found. Furthermore, the presence of 2 different serotypes of A. Pleuropneumoniae was also detected on 3 farms; on the first farm serotypes 2 and 3, on the second farm serotypes 2 and 6, and on the third farm serotypes 2 and 7. Discussion: Although the research was done with a relatively small number of isolates of A. pleuropneumoniae, comparing the obtained results with the results on the presence and prevalence of appropriate serotypes from other countries, we concluded that there is significant diversity of this pathogen in the pig population in farms of Serbia. Detection of different serotypes of A. pleuropneumoniae in the pig population and the presence of several different serotypes on 1 farm was established for the very first time in Serbia. All isolates from our study can be characterized as highly virulent, considering that the clinical symptoms, pathological findings and the results of bacteriological examination indicated A. pleuropneumoniae to be the cause of animal death. Like in the neighbouring countries, the strongly pathogenic serotype 9 and the less pathogenic serotype 2 are the most frequently identified causative agents of porcine pleuropneumonia in the Autonomous Province of Vojvodina, Republic of Serbia. The necessity to establish the presence of all A. pleuropneumoniae serotypes in the pig population, and in particular to determine the presence of different serotypes on individual farms, is crucial for several reasons: making a definitive e diagnosis; development of prophylactic strategies for medicines; implementation of immunoprophylactic vaccination.

Distemper is a disease or the disease by the CDV virus, Distemper Virus. This disease mainly affects dogs, but also affects other domestic and wild species. The animal's immunity is related to the degree to which it will reach the individual's organism. It affects the animal's breathing, can cause vomiting, diarrhea, convulsions, and can lead to death. The aim of the present work test was to standardize an indirect ELISA for distemper diagnosis in experiments using a surface antigen. For the study of technical identification, it was necessary to specify the antigen for the best concentration of plaque sensitization. The test was initially applied with different dilutions of the antigen to detect the best performance of the antigen. This was tested in a serum bank of 45 animals proven positive in the commercial ELISA test and in the serum of 45 animals proven negative in the commercial ELISA test, later it was tested on the cut-off point, specificity and sensitivity of the test. The indirect ELISA test proved to be excellent as a diagnostic test for canine distemper, with an optical density cut-off of 0.229, sensitivity of 95.5% and specificity of 84.4% being obtained.

This research assessed the fumigant activity of the essential oil from Piper sancti-felicis Trel and five of its components on the Tribolium castaneum (Herbst) biological model. Hydrodistillation was used for extraction of the essential oil, with separation and identification of the compounds through gas chromatography coupled to mass spectrometry (GC-MS). The fumigant was evaluated through gas dispersion on the T. castaneum. The majority compounds found in the EO were b-nerolidol (15.4%), 3-carene (14.9%), p-cymene (9.1%), spathulenol (8.2%), a-cubebene (6.2%) and calamenene (5.2%). Piper sancti-felicis displayed fumigant activity with a LC50 = 108.5 & 956;g L-1 air, and other individual monoterpenes tested such as & 945;-terpinolene (LC50 = 110.1 & 956;g L-1 air), p-cymene (LC50 = 120.3 & 956;g L-1 air), 3-carene (LC50 = 130.6 & 956;g L-1 air), (R) -limonene (CL50 = 189.6 & 956;g L-1 air), and a-pinene (LC50 = 213.1 & 956;g L-1 air), were significantly less toxic than methyl pyrimiphos used as a positive control, CL50 = 87.4 & 956;g L-1 air. The essential oil of P. sancti-felicis can be considered as a natural source of biocides.

Brazil has a vast territory and favorable climatic conditions that allow the cultivation of freshwater fish. The intensification of the productive system can cause an imbalance in the aquatic environment as a result of poor water quality, nutritional deficiencies and infectious or parasitic diseases. The laboratory diagnosis and the determination of the prevalence of the main lesions, which occur in a certain region, help to guide towards the etiological diagnosis. This study aimed to describe the main parasitic lesions in fish in the routine at the Veterinary Pathology Laboratory of the Universidade de Brasília (UnB). All records of fish with parasitic lesions were recovered. Those cases in which there was an intralesional parasite and which presented lesions compatible with the parasite were included. The screening of ectoparasites was done by scraping the superficial mucus from the gills and skin. Organ sections were routinely processed for histopathologyand stained with hematoxylin-eosin (HE). In some records, parasitological identification was carried. The information was divided into the species of the affected fish, epidemiology of the outbreak (water quality, temperature, type of breeding), lesion distribution, etiology and macroscopic and microscopic changes. The resulting data was organized in absolute frequency and percentage. In this study, 22 cases were counted, between individual deaths and outbreaks, totaling 83 necropsied teleost fish. Inflammatory changes of parasitic origin were seen in 13/22 (59%) of the cases had lesions of parasitic origin. Skin lesions and gills were the most relevant. Macroscopically, red areas or spots of hyperemia or hemorrhage on the body surface were the most prevalent findings. Under microscopy, proliferative gill inflammation was the most relevant diagnosis. Pscinoodinium pilullare (Dinoflagellida), Ichthyophthirius multifiliis (Ciliophora), and monogenetic worms (Monogenea) were the main parasites found. Trichodina sp. (Ciliophora), Ichtyobodo sp. (Kinetoplastida), Amoebas, and Chilodonella sp. (Ciliophora), were seen in fewer numbers. An unusual case suggestive of parasitism by Eustrongylides sp. (Nematoda), in a pirá-brasília (Simpsonichthys boitonei), specimen has been recorded. The diagnoses were based on epidemiology, anatomopathological and parasitological findings. The most frequent and significantly lethal lesion in the study was proliferative and / or hyperplastic branchitis. Proliferative branchitis with lamellar epithelial hyperplasia (LEH) is a response to some type of chemical or mechanical injury to the gill epithelium in order to protect the capillaries from further damage or microbial penetration. However, it also increases the diffusion distance between capillaries and the environment and, therefore, hinders breathing, excretory and osmoregulatory functions. Protozoan infections and monogenetic worms in general generated LEH and skin lesions of mechanical origin. Secondary bacterial infection, were observed in this parasitosis determining the cause of death of the fish. Its pathogenicity comes from the lesions caused by the colonization and histophagy of the epithelial surfaces, mainly gills and skin, causing epithelial proliferation, lamellar cell fusion, epithelial cell degeneration and necrosis forming several ulcers in the epithelium after the release of mature trophies. The pathogenesis of parasitism by Eustrongylides spp. is considerable when there is a large quantity of these larvae that can cause intestinal obstruction, rupture and compression of viscera, of greater importance in small fish. The main parasites of necropsied fish were protozoa and monogenetic worms, which mainly cause branquitis and dermatitis in varying grades.(AU)

The essential oil of P. sancti-felicis can be considered as a natural source of biocides.(AU)

ABSTRACT: Sausages are highly susceptible to microbial spoilage. Lactic acid bacteria (LAB) is the main group of spoilage bacteria in vacuum packed cooked sausages. To control microbial growth natural antimicrobials have been used as food preservatives. The aim of this study was to identify strains of lactic acid bacteria isolated from spoiled commercial Calabresa sausages and use them in an in vitro challenge with the natural antimicrobials, nisin (NI) and -poly-L-lysine (-PL). Mass spectrometry identification of LAB isolated from sausages using MALDI-TOF revealed a predominance of L. plantarum in the LAB population. RAPD-PCR of L. plantarum strains showed four different genetic profiles. Minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) of NI and -PL, alone and in combination, against a pool of different profiles L. plantarum were determined. MIC of NI and -PL were 0.468 mg/ L and 75 mg/ L; respectively, whereas MBC of NI and -PL were 12.48 mg/L and 150 mg/L, respectively. The combined effect of NI and -PL was determined using concentrations at 1/4 and 1/8 of individual MICs. Synergistic effect was confirmed at both concentrations showing a fractional inhibitory concentration index of 0.5 and 0.2, respectively. The combination of NI and -PL at a small concentration of 0.05 mg/L and 9.375 mg/L, respectively, showed inhibitory effect towards spoilage L. plantarum Results show the potential of the combined use of NI and -PL to control sausage spoilage-associated with lactobacilli.

A PCR-RAPD revelou quatro perfis genéticos diferentes.

Bovine mastitis is widely distributed among dairy herds and causes large economic losses to farmers. The objective of this study was to identify the prevalence of clinical and subclinical mastitis in dairy herds in the plateau of Santa Catarina. Forty-four dairy farms were visited and the main information about the lactating female herd was recorded. The test of the mug of black background was performed for the identification of clinical mastitis, while CMT (California Mastitis Test) was performed for subclinical mastitis. The data was processed with the statistical program R, where the prevalence was calculated. Chi-square test at 5% significance level were used to the variables: milking maneuvers, individual animal data (number and stage of lactation) and Udder Cleaning Score (ELU). The production losses resulting from subclinical mastitis were estimated through the CMT result. The mean prevalence of clinical and subclinical mastitis was 4.29% and 78,92%, respectively, for the animals evaluated. There was no correlation between the prevalence found and the management of milking, number and stage of lactation and ELU. Production losses were significant, with more than 15.000 liters of milk being stopped every month, due to the presence of subclinical mastitis in the herd. Thus, the prevalence found is worrisome, given the importance of the activity as a source of income for these producers. Public bodies should invest in awareness and in measures of control and prevention of the disease so that the profitability of these producers improves and the activity grows in the region.

Leaf-cutting ants of the genus Acromyrmex cause serious damage to cultivated forest areas. Some flies of the family Phoridae are known to parasitize them, and the result of this interaction can provide a promising basis for the management of these ants. This study was developed to record the parasitism and behavioral responses of Acromyrmex spp. leaf-cutting ants to parasitoid phorids in the municipality of Londrina-PR, Brazil. Visual examinations and collections of leaf-cutting ants and associated phorids were carried out between October/2019 and April/2020 in selected anthills and their trails, located on the campus of the State University of Londrina (UEL) and at the Botanical Garden of Londrina. The collected insects were preserved in 70% alcohol, for later identification of the species. The behavioral response of the ants to the parasitoids was recorded individually and collectively, following protocols cited in the literature. The frequencies of the different response behaviors exhibited by the attacked ants were compared using analysis of variance. The leaf-cutting ant species Acromyrmex coronatus and Acromyrmex crassispinus were found at the two evaluated sites. Myrmosicarius catharinensis was the only species of parasitoid phorid found, which attacked both ant species. Although the occurrence of M. catharinensis has been reported in other Brazilian states and its association with A. crassispinus recorded in Buenos Aires, Argentina, this is the first report of the occurrence of the phorid in the state of Paraná with a new association for the species A. coronatus. Under foraging action by the parasitoid, 63.5% of the ants exhibited only one behavioral response, while 36.5% showed two. Accelerated walking, attempting to "bite" the parasitoid, and adopting a 'C' posture were the most frequent behavioral responses to avoid parasitism [...].

The Amazonian basin harbours some of the most bat-diverse ecosystems worldwide. Yet, information on elusive, high-flying bat species such as Molossidae is scarce or virtually missing in the literature, which hampers conservation efforts both locally and globally. The recent advent of new technologies specifically designed to survey bats, such as passive ultrasound detectors and acoustic lures, has significantly increased understanding of bat ecology and distribution, and has allowed researchers to gather new and valuable information which was impossible to collect in the past. We undertook a rapid bat diversity assessment in French Guiana using acoustic lures to aid in capturing high-flying insectivorous bat species. Here we report the second and third capture record of Promops centralis (Chiroptera, Molossidae) for French Guiana, captured after 28 years since the first and only captures so far in the county. One individual was a post-lactating female and represents the first record of breeding P. centralis in French Guiana. We provide (i) morphometric and acoustic data (including the species distress calls) as well as detail photography to aid in species identification; and (ii) COI and CytB sequences of the two individuals (first mitochondrial sequences for French Guiana).(AU)

This study aimed to compare method-based and newly developed sample-based methods for Mycoplasma gallisepticum (MG) detection in different samples of breeder flocks suffering from respiratory disease problems by using culture, real-time PCR (rPCR) and ELISA from chicks and embryonated eggs. Overall, 450 samples of 19-day-old chicken embryos trachea, 450 samples of 8-day-old chicken tracheal swabs and 900 blood samples of 20-, 27-, 34-, 40- and 46-week-old breeder chickens from 5 flocks were sampled for 26 weeks, and were all tested for MG by culture, MG-rPCR and MG-ELISA. Culturing assays and rPCR were applied to 450 mixture samples from 19-day-old chicken embryos trachea and 450 tracheal swab samples (each pooled into groups of 3) from 8-day-old chicks from the same flocks. Also, 900 blood samples from the same 5 breeder flocks suffering from respiratory disease problems were tested by MG-ELISA. In individual sample-based analyses, 55 (18.3%) of the 300 pooled swab samples were positive for MG using culture methods, and 106 (35.3%) of the same samples were found positive by rPCR (sensitivity, specificity). The ELISAs indicated that 252 (28%) of the 900 breeding blood samples were MG seropositive. Using age-based analyses, the most positive period was 46 weeks, followed by 40 weeks, 34 weeks, 27 weeks and at least 20 weeks, in order of decreasing seropositivity. When comparing the culture and rPCR results of the two different sampling methods, chicken embryos trachea yielded more positive results than did tracheal swabs from the same flocks. In conclusion, rPCR is a highly specific, sensitive and reliable method for MG identification.(AU)
