Slash-and-burn is a traditional agricultural system still widely used in Brazil. The variation in temperature during fires results in different levels of physical, chemical, and biological changes in the soil, which makes it necessarily better to understand the dynamics of fire severity in this system. The aim of this study was to make an analytical comparison of the variation in the temperature reached in the soil during fires in slash-and-burn agriculture. Temperature data were measured in burnt areas with similar fallow times at the following soil depths 0; 2.5 and 5 cm and compared with secondary data from other studies in the same slash-and-burn system in southern Brazil. The peak temperature showed more significant variations in the surface (0 cm) of different soils and different types of regeneration vegetation in each area. Overall, the peak temperatures ranged from 32 to 673 ºC across the three depths. The peak temperature reduction in the 5 cm layer was 88 % compared to the surface and 30 % compared to the 2.5 cm layer. The sandier soils showed greater thermal conductivity at depth. The surface litter seems to exert a more significant influence on the burning severity than the biomass load cut for burning, suggesting that the characteristics of the litter layer on the surface should be better characterized in future studies on fire severity.(AU)

The neuropathology of canid alphaherpesvirus 1 (CaHV-1) infection in dogs has been reported, but the importance of the cerebellar lesions in the routine diagnosis remains relatively understated in the veterinary literature. Here we characterize the neuropathology of natural CaHV-1 infection in 25 dogs. The disease affected predominantly male dogs (22 cases). Clinical signs in 15 cases varied and sudden death was reported in 10 cases. All dogs had the typical areas of necrosis and hemorrhage in multiple organs with eosinophilic intranuclear viral inclusions in epithelial and/or inflammatory cells in multiple organs in 21 cases. Cerebral swelling and reddening were reported in 3 cases. Neurohistologic changes consisted of mild lymphoplasmacytic meningoencephalitis with occasional vasculitis and widespread glial nodules. Rare intranuclear viral inclusions (4 cases) were observed in endothelial cells or inflammatory cells in glial nodules. Cerebellum was examined in 11 cases and had extensive segmental necrosis of Purkinje cell layer (8 cases) and granule neurons in the internal (9 cases) or external (2 cases) granular cell layer. Intranuclear viral inclusions were observed in 8 cases and occurred in necrotic granule neurons in the inner granular cell layer (7 cases), outer granular cell layer (2 cases), Purkinje neurons (1 case), or endothelial cells (1 case). Diagnostic confirmation in 23 cases was based on routine histology and fluorescent antibody testing for CaHV-1 on fresh or frozen tissue samples. A routine diagnosis was based solely on the visualization of intranuclear viral inclusions in 2 cases.(AU)

Soybean seeds (Glycine max) were dried under real scale conditions to different final moisture content (m.c.) (9.1, 9.7, 10.9 %, and control with 16.2 %) and processed through extruding-expelling. Results indicated that soybean seed m.c. affected the composition of the soybean expeller and, thus, the oil extraction efficiency (OEE), which increased as the seed m.c. decreased. A polynomic model was proposed for predicting OEE as a function of soybean m.c., indicating that drying soybean to 10 % resulted in an OEE of approximately 65 %. A thin layer drying experiment of soybean seeds indicated that the protein dispersibility index (PDI) was not affected as regards drying air temperatures up to approximately 69 °C, and a bi-linear model with a non-pre-established break point was fitted. The real scale drying treatment in a rack type dryer (mixed flow) did not show any effect (p > 0.05) on the PDI at 80 °C, while at 115 °C a reduction (p < 0.05) was observed (PDI reduction was 0.8 and 2.1 percentage points, respectively).(AU)

The current experiment aimed to evaluate the effects of Pancreatin supplementation at different levels on ileal and fecal digestibility in layer-type cockerels. A total of 480-day-old silver brown Hy-Line male chicks were randomly allocated into 5 treatments, 6 replicates (16 birds per pen) arranged in a completely randomized design. Pancreatin enzyme was supplemented on a basal corn-soybean meal-based diet at 0, 250, 500, 750, and 1000 mg/kg and was fed in two growth phases (starter and grower). The results indicated that at the end of the starter stage, except for 1000 mg/kg, dietary Pancreatin supplementation levels increased (p<0.05) the ileal crude protein (CP). Similarly, addition of Pancreatin increased (p< 0.05) apparent ileal amino acids (AA) digestibility (AIAAD) total means of AA (MTAA), means of indispensable AA (MIAA) and dispensable AA (MDAA) with the optimum performance on 250 mg/kg and 500 mg/kg. However, except for histidine and alanine which were negatively affected (p<0.05), and MIAA, MDAA, MTAA which were also positively affected, the addition level at 1000 mg/kg did not affect most of the AIAAD compared to the non-supplemented. Further, Pancreatin supplementation had no effect (p>0.05) on nitrogen digestibility (ND), nitrogen retention (NR), digestible energy (DE), apparent metabolizable energy (AME), dry matter digestibility (DMD), dry matter retention (DMR), and apparent metabolizable energy corrected for nitrogen (AME-n) on fed starter diet. On the other hand, at the end of the grower stage, dietary Pancreatin enzyme supplementation reduced (p<0.05) the ileal CP, MIAA, MDAA, MTAA, AIAAD, AME, AME-n, ND, NR, DE, DMD, and DMR in a dose-dependent manner. The rate of reduction was more marked on Pancreatin addition level 1000 mg/kg. In conclusion, Pancreatin supplementation at 250 mg/kg, 500 mg/kg, and 750 mg/kg improved AIAAD and ileal CP, especially at the young age. The rate of pancreatin enzyme effect was dependent on enzyme supplement level to the ileal CP and individual amino acid.(AU)

Abstract The poultry sector in Pakistan is contributing mainly in bridging gap between demand and supply for protein. Mycoplasma gallisepticum is an emerging bacterium causing serious problems in poultry industry of Pakistan. A cross-sectional study was conducted to evaluate the M. gallisepticum load in poultry populated regions of Pakistan. Total 600 serum and 600 swab samples were collected, 200 from each broiler, layers and breeders poultry in Rawalpindi and Abbottabad districts. Serum samples were analyzed through ELISA for seroprevalence. Swabs were cultured on Freys medium followed by PCR and partial mgc2 gene sequencing. Results of seroprevalence of M. gallisepticum showed that layers (75%, n=150) are more positive as compared to breeders (70%, n=140) and broilers (50%, n=100). Typical colonies of the M. gallisepticum were observed in breeder (26.5%), followed by layer (21%) and broilers (9%). A total of 37.1% (n=42) samples were identified positive through PCR out of total 113 cultured based positive samples. A total of six M. gallisepticum isolates of current study showed 98-99 percent similarity with previously reported isolates on the basis of mgc2 gene partial sequencing. The M. gallisepticum was found highly prevalent in different poultry breads. Results of this study would add into basic data and provide a direction for livestock sector to strengthen a control strategy for mycoplasmosis in poultry farms.
Resumo O setor avícola do Paquistão está contribuindo principalmente para preencher a lacuna entre a demanda e a oferta de proteína. Mycoplasma gallisepticum é uma bactéria emergente que causa sérios problemas na indústria avícola do Paquistão. Um estudo transversal foi conduzido para avaliar a carga de M. gallisepticum em regiões de avicultura do Paquistão. Um total de 600 amostras de soro e 600 amostras de esfregaço foi coletado, 200 de cada frango de corte, poedeiras e aves reprodutoras nos distritos de Rawalpindi e Abbottabad. Amostras de soro foram analisadas por ELISA para soroprevalência. As zaragatoas foram cultivadas em meio Frey, seguido de PCR e sequenciação parcial do gene mgc2. Os resultados da soroprevalência de M. gallisepticum mostraram que as poedeiras (75%, n = 150) são mais positivas em comparação com matrizes (70%, n = 140) e frangos de corte (50%, n = 100). Colônias típicas de M. gallisepticum foram observadas em reprodutoras (26,5%), seguidas de poedeiras (21%) e frangos de corte (9%). Um total de 37,1% (n = 42) das amostras foi identificado como positivas por PCR de um total de 113 amostras positivas baseadas em cultura. Um total de seis isolados de M. gallisepticum do estudo atual mostrou 98-99% de similaridade com isolados relatados anteriormente com base no sequenciamento parcial do gene mgc2. O M. gallisepticum foi encontrado com alta prevalência em diferentes pães de aves. Os resultados deste estudo acrescentariam dados básicos e forneceriam orientação para o setor pecuário fortalecer uma estratégia de controle da micoplasmose em granjas avícolas.

Three hundred hatching eggs from two layer breeder flocks (Hy-Line Brown and hy-Line W-36; 150 each) were obtained at three different periods of laying cycle (early, middle and late stages). The hatching eggs were randomly taken to assess the impact of chicken strain, age, and hatching time on some eggshell measurements and structures. Results indicate that egg weight, shell percentage and shell thickness were significantly affected by strain, breeder age, and incubation time. Increasing rate of calcium absorption and utilization by the embryos during the embryonic stage clearly reflects the decrease occurred in eggshell strength. The differences of eggshell breaking strength during incubation time periods were 0.41 and 0.55 kg/cm2, the differences due to the incubation time may be due to the higher demand for calcium utilization in the late incubation period (18 days), observed on the10th day of incubation. Brown eggs recorded significant higher calcium percentage in eggshell compared to the white ones. The brown breeder hens recorded significant higher total pores per egg compared to the white egg. Concerning eggshell ultrastructure, total thickness of the brown eggshell was significantly higher than those of the white eggshell by about 3.7%. All parameters, except for total thickness, were significantly affected by the interaction between strain and breeder age. Finally, we conclude that some traits including egg weight, shell percentage, shell thickness, calcium percentages in eggshell, and total pores per egg were significantly affected by strain, breeder age, and incubation time. Eggshell ultrastructure of brown egg was better compared to the white ones.(AU)

The control of Salmonella in the poultry production chain combined with biosecurity measures is an important tool to maintain and guarantee the sanitary status of Brazilian flocks. The aim of this work was to compare official laboratory data on molecular typification of Salmonella isolates from poultry breeding flocks in different Brazilian states between 2016 and 2018 and identify the production category with the most positive flocks, in light of current legislation. Surveillance data of positive samples from the official Brazilian Salmonella Control Programme sent to Federal Agricultural Defence Laboratory of São Paulo (LFDA-SP) after molecular characterization were analysed. These data were subject to an exploratory study, undergoing a descriptive statistical analysis followed by the use of frequency and non-parametric hypothesis tests. Overall, 49 serovars were detected in poultry broiler-breeder and layer-breeder flocks. Salmonella ser. Heidelberg, Salmonella ser. Anatum, Salmonella ser. Newport, Salmonella ser. Schwarzengrund and Salmonella ser. Mbandaka were the five most common isolated serovars. The data shows that there is an opportunity to improve biosecurity measures in parent breeder flocks. A total of 16 serovars were identified in turkey-breeders. Salmonella ser. Anatum, Salmonella ser. Newport, Salmonella ser. Brandenburg, Salmonella ser. Litchfield, and Salmonella ser. Livingstone were the most common ones. The four official controlled serovars represented a small part of the isolated strains. These data demonstrate the importance of an official program in Brazil for Salmonella surveillance in breeder flocks combined with biosecurity measures.(AU)

Starch processing industries use amylases, accounting for approximately 30% of the world's enzyme market. Previously, an amylase-producing strain of Epicoccum nigrumwas isolated from maize grains. Although E. nigrumamylase production is already reported in the literature, no published data on production optimization or characterization of the produced enzyme exists. The objectives of this work were to improve the amylase production by theE. nigrumPG 16 strain and to purify and characterize the produced enzyme. The E. nigrumPG 16 amylase production best conditions in submerged culture were: inoculum of 4% (v v-1) of a five-days-old stationary culture homogenate, agitation at 100 rpm, 25°C, natural light, 72 hours of incubation, starch as thecarbon source, and an initial medium pH of 7.0. A molecular exclusion chromatography profile has shown the production of only one amylase, which was partially purified with ammonium precipitation and dialysis. The enzyme optima pH and temperature are 6.0 and 50°C, respectively. The partially purified enzyme lostits activity when incubated for 30 min in temperatures above 40°C, presenting a T50of 46.25°C. The KMand Vmaxof the partially purified enzyme are 1.72 mg mL-1of starch and 0.15 mgmin-1of degraded starch, respectively. The ion Ca2+slightly activated the studied enzyme. The ions Cu2+, Zn2+, and Fe3+and the detergents SDS and Tween 80 acted as inhibitors of the studied enzyme. The partially purified enzyme released glucose from p-nitrophenyl α-D-glucopyranoside (p-NPG). Glucose was the enzyme's main product from starch hydrolysis, as evidenced by thin-layer chromatography. The E. nigrumPG 16 studied enzyme is a glucoamylase and represents an alternative for enzymatic starch hydrolysis.(AU)

Abstract Species of the genus Cordia have shown biological activities, such as anti-inflammatory, analgesic, antioxidant, antiviral, and antifungal activities. The species Cordia glabrata (MART) A.DC. Has no information concerning its phytochemical profile and possible biological activities. Thus, this study aimed to evaluate this profile in ethanolic extracts of young, adult and senescent leaves, as well as their antioxidant, photoprotective, antimicrobial, and virucidal potentials. Phytochemical analysis was performed by TLC (thin-layer chromatography) and showed the presence of flavonoids, tannins, and terpenes. The evaluation by UPLC-MS/MS (Ultra performance liquid chromatography - tandem mass spectrometer) evidenced the presence of caffeic (3.89 mgL-1), p-cumaric (6.13 mgL-1), and ferulic (0.58 mgL-1) acids, whilst, in GC/MS (Gas chromatographymass spectrometry) analysis there was a greater amount of palmitic (51.17%), stearic (20.34%), linoleic (9.62%), and miristic (8.16%) fatty acids. The DPPH (2,2-Diphenyl-1-picrylhydrazyl) and ABTS+ (2-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)) radicals were used to verify the potential antioxidant activity, observing a better activity for the leaf extract in the adult phenological stage: 54.63 ± 1.06 µgmL-1 (DPPH) and 44.21 ± 1.69 mM (ABTS). The potential photoprotective activity of the extracts was determined by spectrophotometry and the in vitro values of SPF (Sun Protection Factor) in young and adult leaves (5.47 and 5.41, respectively) showed values close to the minimum SPF of 6.0 required by ANVISA (Brazilian Health Regulatory Agency). It was not observed an antimicrobial activity for Staphylococcus aureus with a minimum inhibitory concentration of 2000 gmL-1, however the anti-herpetic assay against the Herpes simplex virus type 2 (HSV-2) showed a potent virucidal activity at the tested concentrations with CV50 value 0.195 gmL-1 and a Selectivity Index (SI = CC50 / CV50) greater than 448. The results obtained in this study suggest that extracts of leaves of C. glabrata in their adult phenological stage have potential antioxidant, photoprotective and virucidal activity, considering in vitro test results.
Resumo Espécies do gênero Cordia apresentam atividades biológicas, como anti-inflamatória, analgésica, antioxidante, antiviral e antifúngica. Para a espécie Cordia glabrata (MART) A.DC., ainda não existem informações sobre seu perfil fitoquímico e possíveis atividades biológicas, deste modo, o presente estudo teve como objetivo avaliar este perfil em extratos etanólicos de folhas jovens, adultas e senescentes, bem como o potencial antioxidante, fotoprotetor, antimicrobiano e virucida. A análise fitoquímica foi realizada por CCD (Cromatografia em Camada Delgada), mostrando a presença de flavonóides, taninos e terpenos. Na avaliação por CLAE EM/EM (Cromatografia Líquida de Ultra Eficiência acoplada a Espectrometria de Massas) foi evidenciado a presença dos ácidos caféico (3,89 mgL-1), p-cumárico (6,13 mgL-1) e ferúlico (0,58 mgL-1), paralelamente, na CG/EM (Cromatografia Gasosa acoplada a Espectrometria de Massas) verificou-se maior quantidade dos ácidos graxos palmítico (51,17%), esteárico (20,34%), linoléico (9,62%) e mirístico (8,16%). Os radicais DPPH (2,2-Difenil-1-picrilhidrazil) e ABTS+ (2-Azino-bis (ácido 3-etilbenzotiazolina-6-sulfônico)) foram utilizados para verificar o potencial antioxidante, observando-se uma atividade superior para o extrato da folha em sua fase fenológica adulta: 54,63 ± 1,06 µgmL-1 (DPPH) e 44,21 ± 1,69 mM (ABTS+). A potencial atividade fotoprotetora dos extratos foi determinada espectrofotometricamente e os valores in vitro de FPS (Fator de Proteção Solar) em folhas jovens e adultas (5,47 e 5,41 respectivamente) apresentaram valores próximos ao FPS mínimo de 6,0 exigido pela ANVISA (Agência Nacional de Vigilância Sanitária). Não foi observada atividade antimicrobiana para Staphylococcus aureus sendo a concentração inibitória mínima de 2000 gmL-1, no entanto o ensaio anti-herpético contra o vírus Herpes simplex tipo 2 (HSV-2) mostrou uma potente atividade virucida nas concentrações testadas com um valor de CV50 0,195 gmL-1 e um Índice de Seletividade (IS = CC50 / CV50) maior que 448. Os resultados obtidos neste estudo sugerem que extratos de folhas de C. glabrata em seu estágio fenológico adulto apresentam potencial antioxidante, fotoprotetora e virucida, considerando os resultados de testes in vitro.

Clinical History: A 436 kg, 2-year-old, pregnant Angus cross cow was presented for necropsy with a history of sudden death during calving season. Two other heifers on the property recently had late term abortions with the calves having to be pulled out from the birth canal. Gross Findings: The subcutaneous tissue, omentum, mesentery, perirenal adipose tissue, and intima of the major vessels were diffusely and markedly yellow (Figure 1). The right liver lobe had a large (15x11x7 cm) area of pale tan discoloration covered by a thin layer of fibrin. On the cut surface, this area was pale red and dry (Figure 2). Throughout the remaining hepatic parenchyma were irregularly shaped, serpiginous, dark green to black tracks. Some of these tracks contained adult Fasciola hepatica flukes in the lumen. The gallbladder was distended and filled with large amounts of dark green, thick bile. The kidneys were diffusely dark red with disseminated pinpoint, dark red foci in the cortex (Figure 3). The urinary bladder contained approximately 300 mL of dark red urine (Figure 4). The brain had a slight yellow discoloration. Approximately 300 mL of red-tinged fluid was in the thorax. Large numbers of fibrous adhesions were present between the visceral pleura, the parietal pleura, and the pericardium. Approximately 200 mL of red-tinged fluid was in the pericardial sac. There was extensive hemorrhage on the epicardium.

Studies on the germination and establishment of plants are key pieces to understanding the reproductive success of plants. This work aimed to describe in vitro germination and reserve mobilization in the bromeliad Vriesea friburgensis through morphological, histochemical, and biochemical analysis. The conditions used in this study for the in vitro germination are adequate. From the third day of in vitro inoculation, a uniform germination of 98% was obtained, exhibiting a high physiological quality of the seeds and a high potential to produce seedlings (94%). There is early reserve mobilization, which began in the imbibition phase. The accumulated reserves in the endosperm cytoplasm are degraded by hydrolytic enzymes provided by the aleurone layer. It is possible that compounds in the cell walls of the endosperm contribute to a lesser extent in mobilization. Additionally, it was observed that starch accumulation in the cotyledon increases when the seedling has formed. Results from this study provide insights for future studies on ecology, seed technology, and conservation in this species. This study contributes to the limited knowledge of the dynamics of reserves during germination and seedling establishment in Bromeliaceae. To the best of our knowledge, this is the first study with this approach in the genus Vriesea.
Estudos sobre germinação e estabelecimento de plantas são peças-chave para entender o sucesso reprodutivo das plantas. Este trabalho teve como objetivo descrever a germinação in vitro e a mobilização de reservas na bromélia Vriesea friburgensis por meio de análises morfológicas, histoquímicas e bioquímicas. As condições utilizadas neste estudo para a germinação in vitro são adequadas. A partir do terceiro dia de inoculação in vitro, obteve-se germinação uniforme de 98%, apresentando alta qualidade fisiológica das sementes e alto potencial de produção de plântulas (94%). Há uma mobilização precoce de reservas, iniciada na fase de embebição. As reservas acumuladas no citoplasma do endosperma são degradadas por enzimas hidrolíticas fornecidas pela camada de aleurona. É possível que compostos nas paredes celulares do endosperma contribuam em menor grau na mobilização. Além disso, observou-se que o acúmulo de amido no cotilédone aumenta com a formação da plântula. Os resultados deste estudo fornecem informações para estudos futuros sobre ecologia, tecnologia de sementes e conservação desta espécie. Este estudo contribui para o conhecimento limitado da dinâmica das reservas durante a germinação e estabelecimento de plântulas em Bromeliaceae. Até onde sabemos, este é o primeiro estudo com esta abordagem no gênero Vriesea.

This study evaluated the levels of heavy metals in the soil and castor bean plant after fertilization with sewage sludge (SS) subjected to different stabilization processes. The study was conducted in a Haplic Cambisol with the following treatments: control (CO), fertilization with solarized sewage sludge (SSS), composted sewage sludge (CSS), vermi composted sewage sludge (VSS), limed sewage sludge (LSS), and mineral fertilizer (MF). The experimental design included a subdivided plot scheme with fertilization being the primary factor and soil layers or leaf parts being the secondary factor. The data obtained were analyzed using analysis of variance and the Scott-Knott test with 5% significance. Fertilization with SSS, CSS, and LSS led to the highest increase in Zn in the soil, mainly in the surface layer. In the leaf tissues, Zn and Cu levels were higher with the CO and SSS treatments, whereas the Ni level was higher with SSS, CSS, VSS, and MF treatments. In general, Zn and Cu levels were higher in the leaf limb than in the petiole, whereas Ni, Pb, and Cr levels were higher in the petiole than in the leaf limb. In any case, no symptoms of toxicity in the plants and no risk of soil contamination were observed with any of the fertilization treatments.
O objetivo deste trabalho foi avaliar os teores de metais pesados no solo e na planta de mamoneira em resposta à adubação com lodo de esgoto (LE) submetido a diferentes processos de estabilização. O experimento foi desenvolvido em Cambissolo Háplico com os tratamentos: testemunha, adubação com LE solarizado, LE compostado, LE vermicompostado, LE caleado e adubação mineral. O delineamento experimental foi em esquema de parcelas subdivididas, sendo as adubações o fator primário e as camadas do solo ou as partes da folha o fator secundário. Os dados obtidos foram submetidos à análise de variância e teste de Scott-Knott a 5% de probabilidade. No solo, as adubações com LE solarizado, compostado e caleado promoveram os maiores incrementos dos teores de Zn, principalmente na camada superficial. No tecido foliar, os teores de Zn e Cu foram mais elevados na testemunha e no LE solarizado, enquanto o Ni foi mais elevado nos LE solarizado, compostado, vermicompostado e na adubação mineral. De modo geral, os maiores teores de Zn e Cu ocorreram no limbo foliar em comparação ao pecíolo, enquanto os teores de Ni, Pb e Cr foram mais elevados no pecíolo do que no limbo foliar. Contudo, não foram constatados sintomas de toxidez nas plantas e nenhum risco de contaminação do solo com os diferentes tipos de adubações realizadas.

This study described changes in the serum biochemistry, morphology of genital organs, long bone, and eggshell during the daily egg formation cycle in Japanese quails. Sixty quails (18-wk) were distributed in 6 groups according to hours post-oviposition (POV): 0 hr POV (16h00), 2 hrs POV (egg in magnum), and 4, 8, 14, and 20 hrs POV (egg in uterus). The magnum had higher relative weight before the next ovulation (20 and 0 hr POV), and its tubular glands showed functional variation through periods: abundant eosinophilic, PAS+, and negative Alcian blue secretion at 0 and 2 hrs, empty glands aspect at 4 hrs, and filled again at 20 hrs POV. Serum albumin and total Ca had the highest value in the 2 hrs group, and the lowest in 8 and 14 hrs groups. Egg-cycle period affected the Ca% of the medullar bone of the femur and tibiotarsus, with the lowest mean at 14 hrs POV (06h00), and the highest mean after oviposition (0 hr POV), showing the recovery of Ca stores in long bones for the next egg cycle. Analysis of the eggshell using scanning electron microscopy evidenced that palisade layer formation starts during the night (814 hrs POV), and most parts are secreted during the day period. In conclusion, eggshell secretion in light periods, high magnum activity and medullary bone Ca deposition during midday and afternoon, as well as the ovulation/oviposition in the afternoon, are the main characteristics of the distinct physiological aspects of the egg cycle in quails.(AU)

Soil microbiota has a key role in the dynamics of natural and agro-ecosystems and is sensitive to changes in these environments. This study evaluated changes in the microbiological properties of soils under an organic production system of banana 'BRS Princesa' (Musa spp.). The experimental design consisted of completely randomized blocks, with four replications. Treatments consisted of 1) soil cover with green manure and agricultural gypsum at a dose of 2,820 kg ha−1, 2) soil cover with green manure without gypsum application, 3) soil cover with weeds and agricultural gypsum at a dose of 2,820 kg ha−1, 4) soil cover with spontaneous plants without gypsum application, and two controls: 5) soil under native Caatinga and 6) soil under regenerating forest (capoeira). The evaluated properties were ß-glucosidase, arylsulfatase, acid phosphatase, fluorescein diacetate hydrolysis activities (FDA), carbon and phosphorus contents in microbial biomass, basal soil respiration, microbial and metabolic quotients, and arbuscular mycorrhizal fungi spore density. Soil samples were collected from the 00.20m depth layer in two seasons. No parameter could distinguish the treatments. Spontaneous plants provided conditions equivalent to those under green manure. Agricultural gypsum application also did not influence the microbial biomass and microbiota activity, in the analyzed soil depth. However, ß-glucosidase and arylsulfatase activities, the carbon content in microbial biomass, and metabolic and microbial quotients were sensitive to land-use changes and could distinguish areas under organic cultivation from those under native vegetation. Therefore, these properties can be considered good indicators for monitoring the quality of these soils. Furthermore, microbial communities of soils under organic cultivation responded with arylsulfatase activity corresponding to that found in soils under regenerating forest, which may indicate that organic management tends to provide the microbiota with a condition similar to that found under situations that are little disturbing to edaphic living.(AU)

The encroachment of agricultural activities on the remnants of the Atlantic Forest in the high altitudes of the Brazilian Northeast led to the formation of forest fragments that need to be better understood. To this end, research was conducted in a forest fragment of Mata do Pau-Ferro in a highland humid enclave, locally known as "Brejo Paraibano", to characterize the soil through chemical, physical, mineralogical, and geochemical attributes, to evaluate the potential ofthe soil in the provisionof services environment. Ferralsols samples were collected and analyzed in a layer from 0 to 20 cm deep. The results showed that soil properties have a direct influence on the provision of ecosystemservices, especially support, regulation, provision, andcultural services. Among the properties, essential nutrient content, carbon stock, minerals that can provide nutrients in the long term, texture, density, and organic matter content stand out, favoring water storage and adequate development of plant species.(AU)
O avanço das atividades agropecuárias sobre os remanescentes de Mata Atlântica de Altitude do Nordeste acarretou na formação de fragmentos de floresta que precisam ser melhorentendidos. Para tanto, a pesquisa foi realizada em um fragmento florestal da Mata do Pau-Ferro no Brejo Paraibano com o objetivo de caracterizar o solo por meio de atributos químicos, físicos, mineralógicos e geoquímicos a fim de avaliar a potencialidade do solo na prestação de serviços ambientais. Foram coletadas amostras de um Latossolo Amarelo Distrófico húmico na camada de 0 a 20 cm de profundidade e carreadas as análises. Os resultados demonstraram que as propriedades do solo influenciam diretamente a oferta de serviços ecossistêmicosespecialmente os de suporte, regulação, provisão e culturais. Dentre as propriedades destacam-se os teores de nutrientes essenciais, estoque de carbono, minerais que podem fornecer nutrientes no longo prazo, textura, densidade e teores de matéria orgânica que favorecem armazenamento de água e adequado desenvolvimento de espécies vegetais.(AU)

The poultry sector in Pakistan is contributing mainly in bridging gap between demand and supply for protein. Mycoplasma gallisepticum is an emerging bacterium causing serious problems in poultry industry of Pakistan. A cross-sectional study was conducted to evaluate the M. gallisepticum load in poultry populated regions of Pakistan. Total 600 serum and 600 swab samples were collected, 200 from each broiler, layers and breeders poultry in Rawalpindi and Abbottabad districts. Serum samples were analyzed through ELISA for seroprevalence. Swabs were cultured on Freys medium followed by PCR and partial mgc2 gene sequencing. Results of seroprevalence of M. gallisepticum showed that layers (75%, n=150) are more positive as compared to breeders (70%, n=140) and broilers (50%, n=100). Typical colonies of the M. gallisepticum were observed in breeder (26.5%), followed by layer (21%) and broilers (9%). A total of 37.1% (n=42) samples were identified positive through PCR out of total 113 cultured based positive samples. A total of six M. gallisepticum isolates of current study showed 98-99 percent similarity with previously reported isolates on the basis of mgc2 gene partial sequencing. The M. gallisepticum was found highly prevalent in different poultry breads. Results of this study would add into basic data and provide a direction for livestock sector to strengthen a control strategy for mycoplasmosis in poultry farms.
O setor avícola do Paquistão está contribuindo principalmente para preencher a lacuna entre a demanda e a oferta de proteína. Mycoplasma gallisepticum é uma bactéria emergente que causa sérios problemas na indústria avícola do Paquistão. Um estudo transversal foi conduzido para avaliar a carga de M. gallisepticum em regiões de avicultura do Paquistão. Um total de 600 amostras de soro e 600 amostras de esfregaço foi coletado, 200 de cada frango de corte, poedeiras e aves reprodutoras nos distritos de Rawalpindi e Abbottabad. Amostras de soro foram analisadas por ELISA para soroprevalência. As zaragatoas foram cultivadas em meio Frey, seguido de PCR e sequenciação parcial do gene mgc2. Os resultados da soroprevalência de M. gallisepticum mostraram que as poedeiras (75%, n = 150) são mais positivas em comparação com matrizes (70%, n = 140) e frangos de corte (50%, n = 100). Colônias típicas de M. gallisepticum foram observadas em reprodutoras (26,5%), seguidas de poedeiras (21%) e frangos de corte (9%). Um total de 37,1% (n = 42) das amostras foi identificado como positivas por PCR de um total de 113 amostras positivas baseadas em cultura. Um total de seis isolados de M. gallisepticum do estudo atual mostrou 98-99% de similaridade com isolados relatados anteriormente com base no sequenciamento parcial do gene mgc2. O M. gallisepticum foi encontrado com alta prevalência em diferentes pães de aves. Os resultados deste estudo acrescentariam dados básicos e forneceriam orientação para o setor pecuário fortalecer uma estratégia de controle da micoplasmose em granjas avícolas.

Rhodnius neglectus is a wild triatomine, vector of the protozoan Trypanosoma cruzi, which causes Chagas' disease, and feeds on the blood of small mammals, being essential for its growth and reproduction. Accessory glands of the female reproductive tract are important in insect reproduction, but their anatomy and histology in R. neglectus are poorly studied. The aim of this work was to describe the histology and histochemistry of the accessory gland of the female reproductive tract of R. neglectus. The reproductive tract of five females of R. neglectus was dissected and the accessory glands transferred to Zamboni's fixative solution, dehydrated in a crescent series of ethanol, embedded in historesin, sectioned at 2 µm thick, stained with toluidine blue for histological analysis or mercury bromophenol blue for detection of total proteins. The accessory gland R. neglectus is tubular, without branches, opening in the dorsal region of the vagina and differing along its length in proximal and distal regions. In the proximal region, the gland is lined by the cuticle with a layer of columnar cells associated with muscle fibers. In the distal region of the gland, the epithelium has spherical secretory cells with terminal apparatus and conducting canaliculi opening in the lumen through pores in the cuticle. Proteins were identified in the gland lumen, terminal apparatus, nucleus and cytoplasm of secretory cells. The histology of the R. neglectus gland is similar to that found in other species of this genus, but with variations in the shape and size of its distal region.
Rhodnius neglectus é um triatomíneo silvestre, vetor do protozoário Trypanosoma cruzi, causador da doença de Chagas. Este inseto se alimenta do sangue de pequenos mamíferos, que é essencial para o seu crescimento e reprodução. As glândulas acessórias do sistema reprodutor feminino são importantes na reprodução de insetos, mas sua anatomia e histologia em R. neglectus são pouco conhecidas. O objetivo deste trabalho foi descrever a histologia e histoquímica da glândula acessória do aparelho reprodutor feminino de R. neglectus. O sistema reprodutor de cinco fêmeas de R. neglectus foi dissecado e as glândulas acessórias transferidas para solução fixadora de Zamboni, desidratadas em série crescente de etanol, embebidas em historesina, seccionadas com 2 µm de espessura, coradas com azul de toluidina para análise histológica ou submetidas ao teste de mercúrio-bromofenol para detecção de proteínas totais. O sistema reprodutor de R. neglectus tem uma glândula acessória tubular, sem ramificações, abrindo-se na região dorsal da vagina, sendo diferenciada em regiões proximal e distal. Na região proximal, a glândula é revestida internamente pela cutícula com uma camada de células colunares associadas a fibras musculares. Na região distal ocorrem células secretoras esféricas com aparato terminal e canalículos condutores que se abrem no lúmen da glândula através de poros na cutícula. O teste histoquímico revelou a presença de proteínas no lúmen da glândula e no aparato terminal, núcleo e citoplasma das células secretoras. A histologia da glândula de R. neglectus é semelhante à das espécies desse gênero, mas com variações na forma e no tamanho de sua região distal.

Vitamin A is an essential micronutrient to reproduction, development, normal growth, and function of the nervous system and vision, especially in growing animals. Hypovitaminosis A can affect any of these functions. The aim of this study was to describe the epidemiology, clinical signs, pathological aspects, and diagnostic approach in an outbreak of blindness in feedlot calves fed only grain. Five of 57 ten-month-old calves became blind after eight months at the feedlot. Clinical signs were mydriasis, absent pupillary and menace reflexes, bilaterally. Hypovitaminosis A was suspected and subsequently confirmed by low serum levels of vitamin A. One calf was euthanized and necropsied, and no gross findings were present. Histopathological lesions were restricted to the retina and characterized by degeneration and atrophy of the photoreceptor layer and some retinal scars. The calves were treated with intramuscular vitamin A injection at a dose of 5,000 international units (IU) per kilogram of body weight (kg BW) every 60 days until slaughter. Affected calves did not recover visual capacity, but no other cases occurred.
Vitamina A é um micronutriente essencial para reprodução, desenvolvimento, crescimento normal, além de participar do funcionamento do sistema nervoso central e visão, especialmente em animais em crescimento. Hipovitaminose A pode afetar qualquer uma dessas funções. O objetivo desse artigo é descrever a epidemiologia, sinais clínicos, aspectos patológicos e abordagem diagnóstica em um surto de cegueira em bezerros confinados alimentados apenas com grãos. De 57 bezerros de dez meses de idade, cinco ficaram cegos após oito meses no confinamento. Os sinais clínicos eram midríase, ausência de reflexo pupilar a luz e de ameaça, bilateralmente. Suspeitou-se de hipovitaminose A, que foi subsequentemente confirmada pelos baixos níveis séricos de vitamina A. Um bezerro foi eutanasiado e necropsiado, e alterações macroscópicas não foram encontradas. Lesões histopatológicas estavam restritas a retina e caracterizavam-se por degeneração e atrofia da camada de fotorreceptores, além de cicatrizes na retina. Os bezerros foram tratados com injeção intramuscular de vitamina A, na dose de 5.000 unidades internacionais (UI) por quilograma de peso vivo (Kg PV) a cada 60 dias, até o dia do abate. Os bezerros afetados não recuperaram a capacidade visual, mas novos casos não ocorreram.
