ABSTRACT: Canine mammary neoplasms with malignant mesenchymal components, such as carcinosarcomas and sarcomas, belong to an uncommon and histologically heterogeneous group. Little is known about the biological behavior of these histogenic variants. This study aimed to compare the clinicopathological characteristics and the COX-2 immunohistochemical expression of different histologic subtypes of carcinosarcomas and sarcomas. Samples of 23 carcinosarcomas and 15 sarcomas from the mammary glands of female dogs were studied. Medical records were reviewed to obtain clinical data. Subsequently, histology microscope slides were analyzed to assess for mesenchymal subtypes, necrosis, vascular invasion, histologic grades, and lymph node metastasis. Immunohistochemistry was used to assess the COX-2 expression. The malignant mesenchymal proliferation was categorized into osteosarcomas (23/40), fibrosarcomas (5/40), liposarcomas (6/40) and chondrosarcomas (4/40). The osteosarcomatous differentiation was the most predominant type among the sarcomas and carcinosarcomas and was associated with vascular invasion (P=0.010) and lymph node metastases (P=0.014). High COX-2 expression was detected in 14.3% of the carcinosarcomas (carcinoma and/or sarcoma cells) and 27.3% of the sarcomas. The carcinosarcomas and sarcomas had similar clinical and pathological characteristics and developed as large tumors, with intratumoral necrosis and a predominance of high histologic grades, although the frequency of vascular invasion and lymph node metastasis was low. Osteosarcoma subtypes presented more aggressive characteristics than non-osteosarcoma subtypes.


RESUMO: Neoplasias mamárias caninas com componentes mesenquimais malignos, como carcinossarcomas e sarcomas, são um grupo de neoplasias pouco frequentes e histologicamente heterogêneas e pouco se sabe sobre o comportamento biológico das variantes histogênicas. O objetivo desse estudo é comparar as características anatomopatológicas e a expressão imunoistoquímica de COX-2 de diferentes subtipos histológicos de carcinossarcomas e sarcomas. Foram estudados 23 carcinosarcomas e 17 sarcomas da glândula mamária de cadelas. Os prontuários médicos foram revisados para obtenção de dados clínicos. Posteriormente, as lâminas histológicas foram avaliadas para acessar os subtipos mesenquimais, necrose, invasão vascular, grau histológico, metástase linfonodal. A imunoistoquímica foi realizada para avaliar a expressão de COX-2. Os tipos encontrados de proliferação mesenquimal maligna foram osteossarcoma (23/40), fibrossarcoma (7/40), lipossarcoma (6/40) e condrossarcoma (4/40). A diferenciação osteossarcomatosa foi predominante entre os sarcomas e carcinossarcomas e foi associado com invasão vascular (P=0,006) e metástase linfonodal (P=0,014). Uma expressão alta de COX-2 foi detectada em 14,3% dos carcinossarcomas (células carcinomatosas e/ou sarcomatosas) e 27,3% dos sarcomas. Os carcinossarcomas e sarcomas apresentaram características clínicas e patológicas semelhantes e se desenvolveram como tumores grandes, com necrose intratumoral e predomínio de alto grau histológico, mas com baixa frequência de invasão vascular e metástase distante. Os subtipos osteossarcomatosos apresentaram características mais agressivas quando comparados com subtipos não osteossarcomatosos.

Resumo

We described the clinical and anatomopathological findings observed in a case of nasal leiomyosarcoma in a five-year-old male Quarter Mile horse, whose main complaints were decreased sports performance and bilateral purulent nasal discharge. The nodule was observed in the nasal cavity, obstructing the left nostril and associated with purulent drainage. The nodule was of irregular shape and yellow color, measuring 19.4 cm × 6.9 cm × 4.3 cm in size, with coalescent multifocal areas that were brownish, friable, opaque, and fetid. When cut, the surface was compact, grayish-white, and smooth with yellow, friable, irregular multifocal areas, measuring 1-3.2 cm in diameter. Histopathological examination showed spindle-shaped neoplastic cells, which was negative on Masson's trichromic stain. A diagnosis of leiomyosarcoma was established based on the morphotintorial aspects of neoplastic cells and confirmed through immunohistochemistry, with positive immunostaining for antibodies 1A4, HHF35, desmin, and S100. Leiomyosarcoma primarily affects the nasal cavity of horses and should be included in the differential diagnosis of diseases that affect the nasal cavity and cause nasal obstruction associated with dyspnea.


Descreve-se os achados clínicos e anatomopatológicos observados em um caso de leiomiossarcoma nasal em um equino, Quarto de Milha, macho, de cinco anos de idade, com queixa principal de diminuição do rendimento esportivo e secreção nasal purulenta bilateral. Na cavidade nasal, observou-se um nódulo obstruindo a narina esquerda associada à secreção purulenta. O nódulo era de superfície irregular, amarelada, brilhante, fibroelástica, medindo 19,4 x 6,9 x 4,3cm de tamanho, com áreas multifocais a coalescente acastanhadas, friáveis, opacas e fétidas. Ao corte, a superfície era compacta, branco-acinzentada e lisa com áreas multifocais amarelas, friáveis e irregulares, medindo 1-3,2 cm de diâmetro. No exame histopatológico foi observado proliferação de células neoplásicas fusiformes, que foram negativas na coloração de Tricrômico de Masson. O diagnóstico de leiomiossarcoma foi estabelecido com base nos aspectos morfotintoriais das células neoplásicas e confirmado através da imuno-histoquímica, no qual houve imunomarcação positiva para os anticorpos 1A4, HHF35, Desmina e S100. O leiomiossarcoma pode afetar primariamente a cavidade nasal de equinos, devendo ser incluído no diagnóstico diferencial de doenças que afetam a cavidade nasal e que provocam quadros de obstrução nasal associado a dispneia.

Os linfócitos são células de defesa do organismo que funcionam como barreira contra infecções e células cancerígenas, elas circulam pelo sistema linfático e estão presentes por todo o organismo do animal, podem se proliferar de forma maligna, caracterizando o linfoma. Acometem em sua maioria, cães de raças de grande porte, animais de meia idade e idosos. Sendo uma doença de etiologia desconhecida, vários fatores podem contribuir para sua evolução, como deficiências autoimunes, bem como hábitos alimentares ao longo da vida do animal, ou até por predisposição genética. O presente relato de caso, tem o objetivo de mostrar a evolução gradual da doença, quais sinais clínicos o animal poderá apresentar, e como os exames laboratoriais podem nos auxiliar em seu diagnóstico.(AU)


The lymphocytes are defense cells of the body that act as a barrier against infection and cancer cells, they circulate through the lymphatic system and are present throughout the animal's body, and can proliferate in a malignant way, characterizing the lymphoma. They mostly affect large breed dogs, middle-aged and elderly animals. Being a disease of unknown etiology, several factors may contribute to its evolution, such as autoimmune deficiencies, as well as food habits throughout the animal's life, or even genetic predisposition. The present case report has the objective of showing the gradual evolution of the disease, which clinical signs the animal may present, and how laboratory tests can help us in its diagnosis.(AU)


Los linfocitos son células de defensa del organismo que actúan como barrera contra infecciones y células cancerígenas, circulan por el sistema linfático y están presentes en todo el organismo del animal, pudiendo proliferar de forma maligna, caracterizando el linfoma. Afectan sobre todo a perros de razas grandes, animales de mediana edad y ancianos. Siendo una enfermedad de etiología desconocida, varios factores pueden contribuir a su evolución, como deficiencias autoinmunes, así como hábitos alimentarios a lo largo de la vida del animal, o incluso predisposición genética. El presente caso clínico tiene como objetivo mostrar la evolución gradual de la enfermedad, qué signos clínicos puede presentar el animal y cómo las pruebas de laboratorio pueden ayudarnos en su diagnóstico.(AU)

Resumo

The article describes the case of a 3 year 6 months old Guinea Pig (Cavia porcellus) that was assisted at the Wildlife and Exotic Animal Practice of the Faculty of Veterinary Sciences, National University of Asunción. The patient presented a hard mass in the right hind limb extending from the proximal portion of the femur, projecting caudally to the proximal tuberosity of the calcaneus, with an increase in temperature, and a marked increase in sensitivity. Radiographic diagnosis refers to a mass with amorphous areas of mineral density. Fine needle aspiration puncture revealed an undifferentiated malignant proliferative process, requiring histopathological analysis to characterize the process. Disarticulation was performed and subsequent histopathologic diagnosis under hematoxylin-eosin and Masson's trichrome stain revealed fibroblastic osteosarcoma.(AU)

Histiocytic sarcoma (HS) is uncommon malignant neoplasia of round cells with marked predilection in Rottweiler and Bernese Mountain. The disseminated form, which mainly affects the spleen, lungs, lymph nodes, bone marrow, skin, and subcutis, presents a quick and aggressive clinical behavior. Hemangiosarcoma (HAS) is a malignant neoplasm of endothelial vessel cells commonly reported to affect the right atrium of dogs' hearts. A male Rottweiler, five years old, presented flaccid paraplegia and progressive muscular atrophy in the temporal, masseter, and limbs muscles; Due to the clinical stage of the animal, euthanasia was conducted. During the necroscopic examination, it was noticed that several masses presented different sizes; some were whitish, and others were reddish and spread in multiple organs (lungs, heart, spleen, stomach, kidneys, brain, medulla, skeletal muscle, and pre-scapular lymph node). Microscopically, in some organs such as the stomach, right ventricle, lungs, and medulla, it was noticed a proliferation of myeloid cells, highly cellular, with poor demarcation, no encapsulation, and with the infiltrative growth pattern of cells with high pleomorphism. Numerous tumoral emboli were observed in the spleen, brain, skeletal muscle, and lymph node. These cells were submitted for immunohistochemistry and were positive for CD18 (HS antibody). In the right atrium, liver, and kidney it was observed malignant and infiltrative endothelial proliferation (HSA) and emboli in the medulla. Therefore, we conclude that both neoplasms (HS and HSA) cause the animal's paraplegia due to their embolism and metastasis to the spinal cord and skeletal muscle.(AU)

Resumo

Background: Osteosarcoma is the most observed primary bone tumor in dogs, and may affect the appendicular and axial skeletons. In addition, it may be present in extraskeletal form, accounting for only 1% of cases. As shown by few reports in the literature, the involvement of the intestinal region by is rare. The objective of this study was to report the case of a 13-year-old Yorkshire dog, submitted to an exploratory laparotomy for suspected partial intestinal obstruction, diagnosed with extraskeletal osteosarcoma. Case: A 13-year-old dog, Yorkshire Terrier, male, presented clinical signs of gastrointestinal abnormalities. An ultrasound examination was performed and was found a mass in small intestine region with wall and lumen invasion. Then, was realized exploratory laparotomy and detected intestinal obstruction due to a mass with approximately 5.0 x 6.0 x 4.4 cm localized in duodenum. Surgical removal was performed and the sample sent to the veterinary diagnostic laboratory for histopathological examination. The sample had an irregular surface and firm consistency. In addition, when cut, the mass enveloped the intestinal layers and sometimes obstructed the lumen. Then, the sample were processed routinely for histopathology. After that, in microscopy evaluation was detected cell proliferation, affecting all layers of intestine. In detail, cells were elongated with pleomorphism marked and atypical mitosis. In addition, there was production of cartilage and bone matrix. So, due the absence of others sites, the neoplasm was considered primary of intestine. After that, to evaluate the expression of KI-67 and COX-2 was performed, and the cell proliferation index was 54.0% and the COX-2 expression was moderate in less than 10% of neoplastic cells. After the surgery, the patient was hospitalized for a week and continue the treatment in home. Afterwards, the tutor received the diagnosis, but even though he was instructed on the severity of the case, he chose not to undergo chemotherapy. After three months, the patient presented abdominal fluid and nodules in your liver, suggesting metastasis, but without diagnosis confirmation. The patient died five months after the diagnosis of extraskeletal osteosarcoma. However, no necropsy was realized, impossibility the diagnosis confirm. Discussion: The frequency of extraskeletal osteosarcoma in dogs remains unknown, with the mammary glands being the most affected site. In the present study, osteosarcoma affects the duodenal region and no reports of this neoplasm in the duodenum of dogs have been found in the literature. The clinical sign of dyschezia was important for the tutor to refer the animal to the veterinarian and perform the ultrasound in an attempt to elucidate the case, as the tumor mass is not always palpable. Histopathological examination and immunohistochemistry were necessary for the differential diagnosis and to establish the prognosis, although after the surgery the tutor chose not to perform chemotherapy. Extraskeletal osteosarcoma are usually highly metastatic, mainly affecting the lymph nodes and liver. In this case, the patient presented a liver nodule three months after the tumor removal surgery, but unfortunately, there was no diagnostic confirmation. Such neoplastic type is rarer and more aggressive than appendicular and axial osteosarcoma, with an average survival of 1 to 3 months. In this case, as a necropsy was not obtained, we cannot attribute the survival time to the disease. The survival rates of osteosarcomas in dogs are few months, but in the present case, although the patient died five months after surgery, the failure to perform a necropsy compromises the attribution of survival time to extraskeletal osteosarcoma.

Resumo

There are no reports of what kind of spleen tumors have developed within a certain period of time in Uruguay. Therefore, we investigated spleen tumors that were surgically resected and brought into our laboratory for a year (2019-2020). As a result, 9 splenic tumors were observed. Hemangiosarcomas occurred at the highest incidence in 6 of 9 cases. In addition, 1 case each of lymphoma, fibrosarcoma, and perivascular wall tumor was observed. Furthermore, the perivascular wall tumor, which has not been reported so far, was observed in a seven years-old female Cocker Spaniel.(AU)

In this retrospective and prospective study, histopathological and immunohistochemical analyses of 62 cases of lymphomas in cats were performed to classify the anatomic forms and subtypes, according to the WHO guidelines, and correlate it to FeLV proviral DNA detected using PCR. The most common anatomical form was gastrointestinal (40.3%, 25/62), followed by multicentric (29%, 18/62), mediastinal (17.7%, 11/62) and extranodal (12,9%, 8/62). Among the lymphoma subtypes, diffuse large B-cell lymphoma (DLBCL) (30.6%, 19/62) was the most commonly diagnosed followed by peripheral T-cell lymphoma (PTCL) (29%, 18/62) and enteropathy associated T-cell lymphoma type 2 (14.5%, 9/62). DNA extraction from paraffin-embedded neoplastic tissue was obtained in 28 cases and FeLV proviral DNA was detected by PCR, in 23 of these. Of the cases presenting with FeLV proviral DNA, nine (32%) were of the multicentric form, five (22%) of the mediastinal and extranodal forms and four (17%) of the gastrointestinal form. The most frequent subtypes with FeLV proviral DNA, independent of the anatomical form, were DLBCL (39.1%, 9/23) and PTCL (34.7%, 8/23). The presence of the FeLV proviral DNA in 23 cats of this study, probably had association with the multicentric form of lymphoma and higher occurrence in the DLBCL and PTCL subtypes.


Neste estudo retrospectivo e prospectivo, análises histopatológicas e imuno-histoquímicas de 62 casos de linfomas em gatos foram realizadas para classificar as formas anatômicas o e subtipos do linfoma, de acordo com as diretrizes da OMS. Além disso, foi realizada a extração de DNA dos tumores incluídos na parafina para obtenção de DNA pró-viral do FeLV por PCR, e relacionada com os exames anteriores. A forma anatômica mais comum foi a gastrointestinal (40.3%, 25/62), seguida pela multicêntrica (29%, 18/62), mediastinal (17,7%, 11/62) e extranodal (12,9%, 8/62). Entre os subtipos de linfoma, o linfoma difuso de grandes células B (DLBCL) (30.6%, 19/62) foi o mais comumente diagnosticado, seguido por linfoma de células T periférico (PTCL) (29%, 18/62) e o linfoma de células T associado a enteropatia tipo 2 (14.5%, 9/62). A extração de DNA de tecido neoplásico emblocado em parafina foi obtida em 28 casos e o DNA pró-viral de FeLV foi detectado por PCR, em 23 deles. Dos casos com DNA pró-viral do FeLV, nove (32%) eram da forma multicêntrica, cinco (22%) das formas mediastinal e extranodal e quatro (17%) da forma gastrointestinal. Os subtipos mais frequentes com DNA pró-viral do FeLV, independente da forma anatômica, foram DLBCL (39.1%, 9/23) e PTCL (34.7%, 8/23). A presença do DNA pró-viral do FeLV em 23 gatos deste estudo, provavelmente teve associação com a forma multicêntrica do linfoma e maior ocorrência nos subtipos DLBCL e PTCL.

This study aimed to evaluate the serum concentration of vitamin D (25-Hydroxyvitamin D) and acute phase proteins (APPs; alpha-1 acid glycoprotein, haptoglobin, transferrin, ceruloplasmin, albumin, IgA, IgG and alpha-1 - antitrypsin) as potential biomarkers for prognostic and therapy response in dogs with multicentric lymphoma submitted to the CHOP (Cyclophosphamide, Doxorubicin, Vincristine and Prednisone) chemotherapy protocol. Thirteen dogs with multicentric lymphoma classified as high grade by cytology were included in the treatment group (GL), while ten healthy dogs were included in the control group (GC). Serum was collected in the weeks T0, T5 and T10 of CHOP chemotherapy protocol, for the GL group, and in a single collection, for the GC group. All the collected samples were evaluated for the APPs and vitamin D concentrations through electrophoresis and chemiluminescence methods, respectively. Diagnostic and staging tests were performed for all the dogs in the GL group, and included cytopathology, histopathology and immunohistochemistry of the affected lymph node. Of these dogs, 9 achieved a complete response and 4 a partial response to the treatment. Data analysis was performed with the R software. The results demonstrated that serum concentrations of IgA, haptoglobin and α1-acid glycoprotein were significantly different between the groups and also between the different chemotherapy times analyzed (p<0.05), indicating that these proteins can be considered as sensitive biomarkers for lymphoma in dogs. Furthermore, the α1-acid glycoprotein showed prognostic value for the disease, with 63% specificity. However, vitamin D concentration was not correlated with prognosis of the dogs with lymphoma.


Objetivou-se caracterizar a concentração sérica da vitamina D e das PFAs (Proteínas de Fase Aguda) (alfa-1 glicoproteína ácida, haptoglobina, transferrina, ceruloplasmina, albumina, IgA, IgG e alfa-1 ­ antitripsina) em cães com linfoma multicêntrico, submetidos ao tratamento quimioterápico com protocolo CHOP (Ciclofosfamida, Doxorrubicina, Vincristina e Prednisona), determinando o valor prognóstico desses marcadores para a doença. Foram avaliadas as concentrações séricas das PFAs, através do método da eletroforese e as concentrações da vitamina D, através da quimioluminescência em dois grupos experimentais, um grupo de 13 cães com linfoma multicêntrico classificados como alto grau pela citologia (GL) durante as semanas T0, T5 e T10 do tratamento com protocolo quimioterápico antineoplásico e em um grupo de 10 animais saudáveis para compor o grupo controle (GC), em coleta única. Para isso, foi realizado o diagnóstico, estadiamento e avaliação de resposta terapêutica dos 13 pacientes com linfoma multicêntrico através de técnicas de citopatologia, histopatologia, imuno-histoquímica do linfonodo periférico acometido. Foi observado que 9 pacientes tiveram resposta completa e 4 pacientes tiveram resposta parcial ao tratamento. Os dados foram analisados através do software R. Os resultados indicam que as diferenças entre as variáveis IgA, haptoglobina e α1-glicoproteína ácida foram significativas entre os grupos, e entre os diferentes momentos da quimioterapia (p< 0,05), indicando que podem ser marcadores sensíveis ao linfoma em cães. A α1-glicoproteína ácida apresentou valor prognóstico para o linfoma, com 63% de especificidade. Porém a vitamina D não apresentou valor prognóstico para o linfoma multicêntrico em cães.

Background: Lymphoma is a malignant lymphoid tumor originating in the lymph nodes or other solid organs and comprises90% of all hematopoietic tumors in dogs. However, primary kidney lymphoma is rare and is associated with nonspecificclinical signs. Tumor invasion in both kidneys can cause severe clinical signs due to renal failure, complicating the patient’streatment and prognosis. The aim of this case was to report the case of a dog affected by bilateral primary kidney lymphoma. In addition, to characterize the clinical and histopathological presentation due to the intense morphological changes.Case: A 5-year-old male Poodle dog was admitted showing apathy and emesis for 5 days. On physical examination, thedog showed 10% of dehydration, reddish oral mucous membranes, poor body condition (score 1/5), uremic breath, andpain in the kidney area. Complementary tests revealed severe low white blood cells count, high BUN levels, high levels ofpotassium, calcium, and phosphorus (serum biochemistry). Abdominal ultrasound showed bilateral kidney enlargement.Fine needle aspiration of the mass (guided by ultrasound) revealed round cell tumor. Radiographs showed no alterations.The dog died due to his poor condition and necropsy was performed. On post-mortem examination, the kidneys were bothenlarged, pale, and with an irregular subcapsular surface. The histopathological diagnostic was primary renal lymphoma.Immunohistochemical staining revealed that neoplastic cells were strongly positive for anti CD20 and PAX5, while negative for CD3, supporting the diagnosis of B-cell lymphoma.Discussion: The diagnosis was based on clinical, complementary tests, fine needle aspiration, histopathological andimmunohistochemical findings. In dogs, primary kidney tumors are uncommon and usually malignant. Background: Lymphoma is a malignant lymphoid tumor originating in the lymph nodes or other solid organs and comprises90% of all hematopoietic tumors in dogs. However, primary kidney lymphoma is rare and is associated with nonspecificclinical signs. Tumor invasion in both kidneys can cause severe clinical signs due to renal failure, complicating the patientstreatment and prognosis. The aim of this case was to report the case of a dog affected by bilateral primary kidney lymphoma. In addition, to characterize the clinical and histopathological presentation due to the intense morphological changes.Case: A 5-year-old male Poodle dog was admitted showing apathy and emesis for 5 days. On physical examination, thedog showed 10% of dehydration, reddish oral mucous membranes, poor body condition (score 1/5), uremic breath, andpain in the kidney area. Complementary tests revealed severe low white blood cells count, high BUN levels, high levels ofpotassium, calcium, and phosphorus (serum biochemistry). Abdominal ultrasound showed bilateral kidney enlargement.Fine needle aspiration of the mass (guided by ultrasound) revealed round cell tumor. Radiographs showed no alterations.The dog died due to his poor condition and necropsy was performed. On post-mortem examination, the kidneys were bothenlarged, pale, and with an irregular subcapsular surface. The histopathological diagnostic was primary renal lymphoma.Immunohistochemical staining revealed that neoplastic cells were strongly positive for anti CD20 and PAX5, while negative for CD3, supporting the diagnosis of B-cell lymphoma.Discussion: The diagnosis was based on clinical, complementary tests, fine needle aspiration, histopathological andimmunohistochemical findings. In dogs, primary kidney tumors are uncommon and usually malignant. It is estimated that a quarter of all feline neoplasms affect the integumentary system. Cytological examination is of admirable importance for detection of neoplasms in companion animals, being considered a simple and non-invasive method. To date, there are limited information regarding the prevalence of feline skin cancer as well as the use of cytopathology as a diagnostic tool. In this sense, the study aimed to characterize cutaneous and subcutaneous tumors in 192 cats from the archives of a private clinic in São Paulo-SP, Brazil, specialized in the care of felines. Data regarding the animal (breed definition, sex, and age group) and tumor characteristics (biological behavior, cytomorphological category of neoplasm, and cytological suggestion for neoplasm) were obtained in a five-year period. A 95% confidence interval (CI) was estimated to verify the statistical difference between the animal and tumor characteristics. The chi-square test was carried out to verify the association between the tumor biological behavior and animal variables, as well as other tumor characteristics. Most cats were mixed breed and aged 11 to 15 years. Malignant tumors were predominant. A statistical association was observed between cytomorphological category of neoplasm and biological behavior (p<0.001). Mesenchymal (24.5%) and epithelial tumors (25%) presented the highest frequency among those benign and malignant, respectively. The cytological suggestion for neoplasm was also statistically associated with biological behavior (p<0.001). The highest percentage of occurrence of benign and malignant tumors consisted of lipomas (22.9%) and sarcomas (19.3%), respectively.(AU)


Estima-se que um quarto de todas as neoplasias felinas afetam o sistema tegumentar. O exame citológico é impor-tante para detecção de neoplasias em animais de companhia, sendo considerado um método simples e não invasivo. Até o momento, há informações limitadas sobre a prevalência das neoplasias cutâneas felinas, bem como o uso da citopatologia como ferramenta diagnóstica. Nesse sentido, o estudo teve como objetivo caracterizar neoplasias cutâneas e subcutâneas em 192 gatos do arquivo de uma clínica veterinária privada da cidade de São Paulo-SP, Brasil, especializada no atendimento de feli-nos. Dados referentes ao animal (raça, sexo e faixa etária) e características das neoplasias (comportamento biológico, categoria citomorfológica e sugestão citológica) foram obtidos em um período de cinco anos. Foi estimado um intervalo de confiança (IC) de 95% para verificar a diferença estatística entre as características do animal e da neoplasia. O teste do qui-quadrado foi realizado para verificar a associação entre o comportamento biológico da neoplasia e as variáveis dos animais, além de outras características neoplásicas. A maioria dos gatos era sem definição racial e com idade entre 11 e 15 anos. As neoplasias malignas foram predominantes. Foi observada associação estatística entre a categoria citomorfológica e o comportamento biológico da neoplasia (p<0,001). Neoplasias mesenquimais (24,5%) e epiteliais (25%) apresentaram a maior frequência entre os benignos e malignos, respectivamente. A sugestão citológica para neoplasia também foi estatisticamente associada ao comportamento biológico (p<0,001). O maior percentual de ocorrência de tumores benignos e malignos consistiu em lipomas (22,9%) e sar-comas (19,3%), respectivamente.(AU)

L-Asparaginase catalysing the breakdown of L-Asparagine to L-Aspartate and ammonia is an enzyme of therapeutic importance in the treatment of cancer, especially the lymphomas and leukaemia. The present study describes the recombinant production, properties and anticancer potential of enzyme from a hyperthermophilic archaeon Pyrococcus abyssi. There are two genes coding for asparaginase in the genome of this organism. A 918 bp gene encoding 305 amino acids was PCR amplified and cloned in BL21 (DE3) strain of E. coli using pET28a (+) plasmid. The production of recombinant enzyme was induced under 0.5mM IPTG, purified by selective heat denaturation and ion exchange chromatography. Purified enzyme was analyzed for kinetics, in silico structure and anticancer properties. The recombinant enzyme has shown a molecular weight of 33 kDa, specific activity of 1175 U/mg, KM value 2.05mM, optimum temperature and pH 80°C and 8 respectively. No detectable enzyme activity found when L-Glutamine was used as the substrate. In silico studies have shown that the enzyme exists as a homodimer having Arg11, Ala87, Thr110, His112, Gln142, Leu172, and Lys232 being the putative active site residues. The free energy change calculated by molecular docking studies of enzyme and substrate was found as ∆G – 4.5 kJ/mole indicating the affinity of enzyme with the substrate. IC50 values of 5U/mL to 7.5U/mL were determined for FB, caco2 cells and HepG2 cells. A calculated amount of enzyme (5U/mL) exhibited 78% to 55% growth inhibition of caco2 and HepG2 cells. In conclusion, the recombinant enzyme produced and characterized in the present study offers a good candidate for the treatment of cancer. The procedures adopted in the present study can be prolonged for in vivo studies.


A L-asparaginase, que catalisa a degradação da L-asparagina em L-aspartato e amônia, é uma enzima de importância terapêutica no tratamento do câncer, especialmente dos linfomas e da leucemia. O presente estudo descreve a produção recombinante, propriedades e potencial anticancerígeno da enzima de Pyrococcus abyssi, um archaeon hipertermofílico. Existem dois genes que codificam para a asparaginase no genoma desse organismo. Um gene de 918 bp, que codifica 305 aminoácidos, foi amplificado por PCR e clonado na cepa BL21 (DE3) de E. coli usando o plasmídeo pET28a (+). A produção da enzima recombinante foi induzida sob 0,5mM de IPTG, purificada por desnaturação seletiva por calor e cromatografia de troca iônica. A enzima purificada foi analisada quanto à cinética, estrutura in silico e propriedades anticancerígenas. A enzima recombinante apresentou peso molecular de 33 kDa, atividade específica de 1.175 U / mg, valor de KM 2,05 mM, temperatura ótima de 80º C e pH 8. Nenhuma atividade enzimática detectável foi encontrada quando a L-glutamina foi usada como substrato. Estudos in silico mostraram que a enzima existe como um homodímero, com Arg11, Ala87, Thr110, His112, Gln142, Leu172 e Lys232 sendo os resíduos do local ativo putativo. A mudança de energia livre calculada por estudos de docking molecular da enzima e do substrato foi encontrada como ∆G – 4,5 kJ / mol, indicando a afinidade da enzima com o substrato. Valores de IC50 de 5U / mL a 7,5U / mL foram determinados para células FB, células caco2 e células HepG2. Caracterizado como uma neoplasia maligna, o linfoma avança inicialmente em tecidos linfoides, mas pode apresentar-se em tecidos distintos. Os cães em comparação as outras espécies, tendem a manifestar o linfoma de maneira mais agressiva, independente do seu local de expressão. O presente trabalho objetivou descrever um caso de um cão da raça Fox Americano que apresentou aumento de volume da região periorbital e terceira pálpebra, sendo diagnosticado com linfoma, após exames ultrassonográfico e histopatológico. Após o diagnóstico definitivo de linfoma, iniciou-se o tratamento quimioterápico que teve duração de 19 semanas. Ao término do protocolo, o paciente apresentou remissão completa das manifestações oftálmicas, porém, foi orientado o acompanhamento mensal, e durante 3 meses consecutivos nas avalições o paciente não apresentou nenhuma alteração clínica. Conclui-se que quadros de linfoma podem apresentar manifestações que acometam a região periorbital, sendo de suma importância a avaliação clínica e realização de exames complementares para exclusão de outras doenças oftálmicas, e adoção de medidas terapêuticas efetivas.(AU)


Characterized as a malignant neoplasm, lymphoma advances initially in lymphoid tissues, but can present itself in different tissues. Dogs, in comparison to other species, tend to manifest lymphoma in a more aggressive way, regardless of their place of expression. The present study aimed to describe a case of a dog of the Fox American breed that presented an increase in the volume of the periorbital region and third eyelid, being diagnosed with lymphoma, after ultrasound and histopathological exams. After the definitive diagnosis of lymphoma, chemotherapy treatment was started, which lasted 19 weeks. At the end of the protocol, the patient showed complete remission of the ophthalmic manifestations, however, monthly follow-up was advised, and for three consecutive months in the evaluations, the patient did not present any clinical changes. It is concluded that lymphoma pictures can present manifestations that affect the periorbital region, being extremely important the clinical evaluation and accomplishment of complementary exams for exclusion of other ophthalmic diseases, and adoption of effective therapeutic measures.(AU)

Los perros, en comparación con otras especies, tienden a manifestar el linfoma de forma más agresiva, independientemente de su lugar de expresión. El presente estudio tuvo como objetivo describir un caso de un perro de la raza Fox American que presentó aumento de volumen de la región periorbitaria y tercer párpado, siendo diagnosticado de linfoma, luego de exámenes ecográficos e histopatológicos. Tras el diagnóstico definitivo de linfoma se inició tratamiento de quimioterapia, que duró 19 semanas. Al final del protocolo, el paciente presentó remisión completa de las manifestaciones oftálmicas, sin embargo, se recomendó un seguimiento mensual y durante 3 meses consecutivos en las evaluaciones, el paciente no presentó cambios clínicos. Se concluye que los cuadros de linfoma pueden presentar manifestaciones que afecten a la región periorbitaria, siendo de suma importancia la evaluación clínica y la realización de exámenes complementarios para la exclusión de otras enfermedades oftálmicas y la adopción de medidas terapéuticas efectivas.(AU)

O linfoma é a neoplasia linfo-hematopoiética mais frequente em cães. A forma alimentar é pouco comum em cães, representando cerca de 5 a 7% de todos os tipos de linfomas caninos. Apenas 20% dos cães acometidos apresentam idade inferior a quatro anos. Ela caracteriza-se pela localização no trato gastrointestinal e/ou linfonodos mesentéricos e aqueles não tratados vivem entre quatro a seis semanas após o início dos sinais clínicos ou diagnóstico. A quimioterapia é a modalidade terapêutica de escolha para cães com linfoma. Este trabalho tem como objetivo descrever um caso de linfoma alimentar em um cão de três anos de idade, atendido no Hospital Veterinário da Ulbra - Canoas, com quadro de vômito, hematoquezia, polidipsia, anorexia e perda de peso progressiva. Com presença de espessamento grave e difuso de segmentos intestinais e linfonodomegalia de linfonodo pancreático e jejunal ao exame ultrassonográfico. O diagnóstico foi confirmado posteriormente por necropsia e exame histopatológico, após o óbito do paciente.(AU)


Lymphoma is the most frequent lympho-hematopoietic neoplasm in dogs. The alimentary form is uncommon in dogs, representing 5 to 7% of all types of canine lymphomas. Only 20% of affected dogs are under the age of four. This disease is characterized by the location in the gastrointestinal tract and/or mesenteric lymph nodes and those untreated live four to six weeks after the onset of clinical signs or diagnosis. Chemotherapy is the therapeutic modality of choice for dogs with lymphoma. This report aims to describe a case of alimentary lymphoma in a three year old dog treated at the Hospital Veterinário da Ulbra - Canoas, presenting vomiting, hematochezia, polydipsia, anorexia and progressive weight loss. With the presence of severe and diffuse thickening of intestinal segments and lymph node enlargement of pancreatic and jejunal lymph nodes on abdominal ultrasound exam. The diagnosis was later confirmed by necropsy and histopathological examination after the patient's death.(AU)


El linfoma es la neoplasia linfohematopoyética más frecuente en perros, La forma alimentaria es poco común en los perros y representa aproximadamente 5 al 7% de todos los tipos de linfomas caninos. Solo el 20% de los perros afectados tienen menos de cuatro años. Se caracteriza por su localización en el tracto gastrointestinal y/o en los ganglios linfáticos mesentéricos y los que no reciben tratamiento viven entre cuatro y seis semanas después del inicio de los signos clínicos o del diagnóstico. La quimioterapia es la modalidad terapéutica de elección para perros con linfoma. Este artículo tiene como objetivo describir un caso de linfoma alimentario en un perro de tres años atendido en el Hospital Veterinário da Ulbra - Canoas, con vómitos, hematoquecia, polidipsiaa, anorexia y pérdida progresiva de peso. Con la presencia de engrosamiento severo y difuso de los segmentos intestinales y agrandamiento de los ganglios linfáticos pancreáticos y yeyunales en el examen de ultrasonido. El diagnóstico se confirmó posteriormente mediante necropsia y examen histopatológico tras la muerte del paciente.(AU)

Osteosarcoma is a mesenchymal cancer associated with high mortality in dogs and in humans. The biodiversity-rich Cerrado, the predominant biome in the Midwest region of Brazil, is native to locally appreciated fruits such as pequi (Caryocar brasiliense, Camb). Although this plant has been frequently used in folk medicine, the pharmacological properties of pequi fruit shells have not been fully evaluated. Thus, this study aimed to determine the cytotoxic activity of ethanol extract of recycled pequi shells on canine osteosarcoma cells in vitro. Cells were cultured and treated with final extract concentrations of 0, 0.029 µg/µL, 0.29 µg/µL, and 2.91 µg/µL for 24, 48, or 72 hours. Cell viability assay using trypan blue exclusion method and tetrazolium reduction method, cell survival assay, and double labeling with annexin V and propidium iodide were performed in the treated osteosarcoma cells. These allowed the determination of IC50, survival fraction, and type of cell death, respectively. Pequi shell ethanol extract at a concentration of 2.91 µg/µL showed the greatest inhibition of osteosarcoma cell growth in vitro, resulting in a 71.80% decrease in growth compared to the control. The mean IC50 was 155.2 µg/mL at 72 hours. The calculated survival fractions showed that cell growth at 72 hours was 3.33% lower in cells treated with 2.91 µg/µL extract. Results from the double labeling experiment suggest that apoptosis was the predominant type of cell death in cells treated with 2.91 µg/µL extract. These results demonstrate that ethanol extract of recycled pequi shells promotes apoptosis in canine osteosarcoma cells.


O osteossarcoma é uma neoplasia mesenquimal associada à alta mortalidade em cães e humanos. O pequi (Caryocar brasiliense, Camb.) é nativo do Cerrado, bioma rico em biodiversidade predominante na região Centro-Oeste do Brasil. Embora esta planta seja frequentemente utilizada na medicina popular, as propriedades farmacológicas da casca do pequi ainda são pouco conhecidas. Este estudo teve como objetivo determinar a atividade citotóxica do extrato etanólico de cascas de pequi recicladas em células de osteossarcoma canino. As células foram cultivadas e tratadas com concentrações finais de extrato de zero, 0,029 µg/µL, 0,29 µg/µL e 2,91 µg/µL por 24, 48 ou 72 horas. As células foram submetidas a ensaio de viabilidade celular pelos métodos de exclusão de azul de tripano da redução de tetrazólio para determinar a IC50, ao ensaio de sobrevivência celular para calcular a fração de sobrevivência e à dupla marcação com anexina V e iodeto de propídio para determinar o tipo de morte celular. O extrato etanólico de casca de pequi na concentração de 2,91 µg/µL apresentou a maior inibição do crescimento de células de osteossarcoma, com redução de 71,80% no crescimento em relação ao controle. A IC50 média foi de 155,2 µg/mL em 72 horas. O crescimento celular em 72 horas foi 3,33% menor nas células tratadas com 2,91 µg/µL de extrato. A apoptose foi o tipo predominante de morte celular em células tratadas com 2,91 µg/µL de extrato. Esses resultados demonstram que o extrato etanólico de cascas de pequi recicladas promove apoptose em células de osteossarcoma canino.

Background: Mediastinal lymphoma occurs at a high incidence in cats positive for feline leukemia virus (FeLV). It is a malignant lymphocytic neoplasm that may trigger clinical signs such as dyspnea, apathy, regurgitation, and weight loss. The objective of this work is to report a case of mediastinal lymphoma associated with FeLV in a cat, and describe the clinical, pathological, diagnostic, and therapeutic aspects that can help the diagnosis and treatment of this disease. Case: A 3-year-old male neutered cat weighing 4.6 kg, positive for FeLV, and with a history of dyspnea and hyporexia was referred to a private veterinary clinic in the city of Caxias do Sul, RS, Brazil. The only alteration found at the clinical examination was a muffled sound during lung auscultation. Radiographs of the thorax revealed the presence of pleural effusion; after drainage of the fluid, a mass located in the mediastinal area became radiographically observable. An analysis of the effusion fluid showed high cellularity characterized by a markedly pleomorphic population of individual round cells consistent with lymphocytes. Small lymphocytes with a high nucleus:cytoplasm ratio, round nucleus, condensed chromatin, and inconspicuous nucleoli were observed, along with medium and large lymphocytes. The medium and large lymphocytes were characterized by a variably increased nucleus:cytoplasm ratio and a scant to moderate cytoplasm exhibiting moderate to intense basophilia and, occasionally, vacuoles. The nuclei were round, idented, or irregular; most of them were located eccentrically and contained coarse to finely granular chromatin. Nucleoli varied from single to multiple, round to angular, and central to peripheral and prominent. Macronucleoli and marked anisonucleosis were also observed, as well as binucleated cells and rare multinucleated cells. The conclusion was that it was a case of neoplastic effusion caused by a lymphoma. After an appointment with an oncologist, a chemotherapy protocol was established. The treatment of choice was CHOP, a combination of cyclophosphamide (20 mg/kg orally), doxorubicin (1 mg/kg intravenously), vincristine (0.5 mg/m2 intravenously), and prednisolone with a regressive dose starting at 2 mg/kg. The patient underwent 4 rounds of chemotherapy, and received a maintenance treatment thereafter. The patient's condition remained stable for 390 days without relevant clinical or hematological alterations. Discussion: The diagnosis of mediastinal lymphoma was established by associating clinical finings, laboratory exams, and radiographic findings. Exams of utmost importance to reach this diagnosis were the thoracic radiography, which revealed the presence of a mass in the thorax, and the cytopathological analysis of the effusion fluid, as round cell neoplasms are easily dissociated. Young cats are the group most affected by FeLV, and mediastinal lymphoma is considered the most prevalent type of lymphoma in this species. The most common clinical sign is dyspnea; however, other signs such as apathy, weight loss, regurgitation (due to pressure on the esophagus), and Horner's syndrome (owing to pressure on the thoracic sympathetic innervation) may also occur. This disease is treated with chemotherapy, with CHOP frequently used as the chemotherapeutic protocol. The good therapeutic response for a longer time than the average estimated by studies on the prognostic of this disease indicates that a correct diagnosis along with an assertive approach and the cooperation of the tutor are essential in cases of mediastinal lymphoma.

Resumo

O osteossarcoma é uma das neoplasias ósseas mais comumente relatadas na medicina veterinária, principalmente em cães de raça grande e gigante, sendo que a forma vertebral possui importante cenário clínico apresentando 5% do total de relatos. Este possui um prognóstico desfavorável e tempo de sobrevida pequeno. A apresentação clínica é variável e podem ser observados sinais de dor intensa a déficits neurológicos em decorrência do envolvimento medular progressivo. O diagnóstico pode ser realizado por imagens através de radiografia ou tomografia computadorizada, e por análises laboratoriais de citopatologia ou histopatologia, sendo o último preferível para classificação de tumores. A ressecção tumoral parcial é uma forma paliativa de tratamento, indicada em alguns casos para aliviar temporariamente os sinais clínicos relacionados a dor e déficits neurológicos. Contudo, quando há possibilidade terapêutica, a excisão completa fornece melhor prognóstico e tempo de sobrevida, principalmente quando associada a quimioterapia ou radioterapia. Mediante procedimento de vertebrectomia, a estabilização cirúrgica é necessária, podendo ser realizada respeitando os princípios para estabilização vertebral por trauma, sendo necessária a colocação de um espaçador vertebral para evitar o colabamento medular. Diversos materiais foram propostos para a confecção do espaçador vertebral. A manufatura aditiva, também conhecida como impressão 3D, tem transformado o conceito de prototipagem rápida em realidade, devido a habilidade de fabricar peças geométricas específicas de alta complexidade e de forma rápida, permitindo elaborar protótipos para uso pré ou transoperatórios em cirurgias ortopédicas de alta complexidade. O polietileno tereftalato glicol (PETG), por ser um termoplástico de alta resistência, não-biodegradável, biocompatível e de baixo custo é foco desse trabalho no desenvolvimento do espaçador vertebral como estabilizador em um procedimento de vertebrectomia em cão.


Osteosarcoma is one of the most commonly reported bone neoplasms in veterinary medicine, especially in large and giant breed dogs, and the vertebral form has an important clinical scenario presenting 5% of the total reports. This has an unfavorable prognosis and a short survival time. The clinical presentation is variable and signs of severe pain to neurological deficits can be observed as a result of progressive spinal cord involvement. The diagnosis can be made by imaging with radiography or computed tomography, and by laboratory analysis of cytopathology or histopathology, the latter being preferable for tumor classification. Partial tumor resection is a palliative form of treatment, indicated in some cases to temporarily relieve clinical signs related to pain and neurological deficits. However, when there is therapeutic possibility, complete excision provides better prognosis and survival time, especially when associated with chemotherapy or radiotherapy. Through the vertebrectomy procedure, surgical stabilization is necessary and can be performed following the principles for vertebral stabilization due to trauma, with the placement of a vertebral spacer being necessary to prevent spinal collapse. Several materials have been proposed for the manufacture of the spinal spacer. Additive manufacturing, also known as 3D printing, has transformed the concept of rapid prototyping into reality, due to its ability to quickly manufacture specific geometric parts of high complexity, allowing the elaboration of prototypes for pre- or trans-operative use in highly complex orthopedic surgeries. Polyethylene terephthalate glycol (PETG), being a high-strength, non-biodegradable, biocompatible and low-cost thermoplastic, is the focus of this work in the development of the vertebral spacer as a stabilizer in a vertebrectomy procedure in a dog.


El osteosarcoma es una de las neoplasias óseas más comúnmente reportadas en medicina veterinaria, principalmente en perros de raza grande y gigante, siendo que la forma vertebral tiene importante escenario clínico presentando el 5% del total de reportes. Tiene un pronóstico desfavorable y un tiempo de supervivencia corto. La presentación clínica es variable y pueden observarse desde signos de dolor intenso hasta déficits neurológicos como resultado de la afectación medular progresiva. El diagnóstico puede realizarse mediante imágenes con radiografía o tomografía computarizada, y mediante análisis de laboratorio de citopatología o histopatología, siendo este último preferible para la clasificación del tumor. La resección parcial del tumor es una forma de tratamiento paliativo, indicada en algunos casos para aliviar temporalmente los signos clínicos relacionados con el dolor y los déficits neurológicos. Sin embargo, cuando hay posibilidad terapéutica, la escisión completa proporciona un mejor pronóstico y tiempo de supervivencia, principalmente cuando se asocia con quimioterapia o radioterapia. Mediante el procedimiento de la vertebrectomía, es necesaria la estabilización quirúrgica, que puede realizarse respetando los principios para la estabilización vertebral por traumatismo, siendo necesaria la colocación de un espaciador vertebral para evitar el colapso medular. Se han propuesto varios materiales para la fabricación del espaciador vertebral. La fabricación aditiva, también conocida como impresión 3D, ha transformado el concepto de prototipado rápido en una realidad, debido a la capacidad de fabricar piezas geométricas específicas de gran complejidad y con rapidez, lo que permite la elaboración de prototipos para su uso pre o trans-operatorio en cirugías ortopédicas de gran complejidad. El polietileno tereftalato glicol (PETG), por ser un termoplástico de alta resistencia, no biodegradable, biocompatible y de bajo costo es el foco de este trabajo en el desarrollo del espaciador vertebral como estabilizador en un procedimiento de vertebrectomía en un perro.

Curcuma longa L., also known as turmeric, has been widely studied for its various therapeutic properties, including antineoplastic action. The ethanolic extract of the plant contains several phenolic compounds, especially curcumin. Osteosarcoma is a predominant bone tumor in dogs and humans, characterized by high metastatic potential and an unfavorable prognosis. The aim of this study was to investigate the effects of turmeric ethanol extract on canine osteosarcoma cells from established culture. The cells were cultured and treated with different curcumin concentrations (0, 10 µM, 20 µM, 50 µM, 100 µM, and 1000 µM) and exposure times (24h, 48h, and 72h). We first performed tetrazolium reduction technique (MTT) assay and calculated IC50. An immunocytochemistry assay was performed after extract treatment to verify the expression of mutated p53 and therefore study the proliferative potential of malignant cells; Bcl-2 and Ki-67 were used to assess apoptosis and the degree of malignancy, respectively. The extract enhanced the proliferation of canine osteosarcoma cells, reaching 3,819.74% at 50 µM of curcumin. The extract also significantly altered the expression of mutated p53 and Ki-67 proteins but not that of Bcl-2, suggesting that it did not induce this antiapoptotic pathway. Overall, these results are prerequisite to better understanding how natural compounds such as turmeric ethanolic extract affect cell proliferation and could be used to treat various diseases.


A Curcuma longa L., planta conhecida popularmente como açafrão, tem sido amplamente estudada por suas diversas propriedades terapêuticas, incluindo a ação antineoplásica. O extrato etanólico da planta contém diversos compostos fenólicos, com destaque para a curcumina. O osteossarcoma é um tumor ósseo predominante em cães e humanos, caracterizado por apresentar alto potencial metastático e prognóstico desfavorável. Procurou-se investigar os efeitos de diferentes concentrações de curcumina do extrato etanólico de açafrão sobre células de osteossarcoma canino de cultura estabelecida. As células foram cultivadas e submetidas ao tratamento com extrato com diferentes concentrações de curcumina (0, 10 µM, 20 µM, 50 µM, 100 µM e 1000 µM) e tempos de exposição (24h, 48h e 72h) pelo EEA. Inicialmente, foram realizados: técnica de redução do tetrazólio (MTT) e cálculo da IC50. Posteriormente, após o tratamento com o extrato, realizou-se o ensaio de imunocitoquímica para verificar a expressão de p53 mutada e estudar o potencial proliferativo das células malignas; Bcl-2, com intuito de averiguar o estímulo de via antiapoptótica; e o marcador Ki-67, que sinaliza aumento no grau de malignidade. O extrato promoveu proliferação de células de osteossarcoma canino, com incremento de até 3819,74% na concentração de 50µM de curcumina. O composto também alterou a expressão das proteínas p53 mutante e Ki-67 significativamente, mas não alterou a expressão de Bcl-2, mostrando que não induziu a via antiapoptótica mediada por esta. Estes resultados demonstram que o extrato etanólico do açafrão apresenta potencial proliferativo sobre células de osteossarcoma canino, sugerindo a necessidade de conscientização e conhecimento dos reais efeitos de determinados compostos naturais, considerados seguros ao serem utilizados como tratamento de diversas enfermidades.
