Estudos sobre a fisiologia espermática demonstram que padrões de fertilidade e funcionalidade espermática pós-criopreservação possuem relação importante com a eficiência do metabolismo energético destas células, bem como com a sua capacidade de manter a homeostase oxidativa. Os conhecimentos sobre a relação entre perfil fisiológico de espermatozoides e fertilidade foram e, ainda estão sendo, aprimorados, com o uso de análises de perfis moleculares, com destaque para a metabolômica. As análises moleculares permitiram a identificação de classes de metabólitos importantes na fisiologia espermática, bem como de potenciais biomarcadores de fertilidade, inclusive em bovinos. No entanto, ainda não há disponível uma avaliação isolada capaz de estimar o padrão de fertilidade de amostras seminais. Há vários desafios a serem superados para a validação de biomarcadores de fertilidade, principalmente considerando-se as diferenças entre perfis metabólicos de raças distintas de touros e a heterogeneidade dos ejaculados. A superação destes desafios pode ser iniciada com um maior aproveitamento dos resultados já obtidos e futuros, com a aplicação de análises mais avançadas e com eficácia em elevado número de dados. Para tal, podem ser utilizados modelos estatísticos de inteligência artificial, cuja aplicação pode aumentar a acurácia das observações obtidas, bem como aproximá-las da aplicação pelo setor de produção animal.(AU)
Studies on sperm physiology demonstrate that fertility outcomes and sperm post-cryopreservation have an important relation with sperm energy metabolism efficiency and ability to maintain oxidative homeostasis. Knowledge on sperm physiology has been enhanced, continually, with the application of molecular profiling analysis, focusing on metabolomics. Molecular analysis allowed the identification of important classes of metabolites in sperm physiology, as well as potential fertility biomarkers, including in bovine. Despite all developments, there is still no isolated assessment available capable of estimating fertility on sperm samples. There are several challenges to be overcome for the validation of fertility biomarkers, especially considering the metabolic profiles differences between bull breeds and the ejaculate heterogeneity. Overcoming these challenges could start with the application of more advanced and effective data analysis from research database already obtained, as well as future ones. To this end, statistical models of artificial intelligence can be used, whose application can increase the accuracy of the observations obtained, as well as bringing them closer to the application by the animal production sector.(AU)

As leveduras são fungos de importância à medicina veterinária por causarem doenças infecciosas em diferentes hospedeiros animais. A presente revisão de literatura teve como objetivo relatar os principais testes bioquímicos capazes de auxiliar na identificação de fungos leveduriformes de interesse veterinário e zoonótico. Para o levantamento bibliográfico, foram consideradas 48 publicações científicas selecionadas na área e indexadas nas principais bases de dados, entre os anos de 1988 e 2020. Como resultados, observou-se que oito provas são as mais empregadas na rotina micológica. Devido à baixa variabilidade morfológica das espécies leveduriformes, testes bioquímicos complementares são fundamentais na rotina laboratorial. A análise do perfil bioquímico de leveduras contribui na determinação taxonômica dos fungos a partir de reações químicas, visto que o metabolismo varia de acordo com a espécie, resultando em metabólitos distintos, os quais podem ser avaliados por diferentes provas. Conclui-se que a identificação fenotípica das leveduras é imprescindível no diagnóstico, prognóstico, tratamento e controle de doenças fúngicas e contribui para a manutenção da saúde animal.(AU)
Yeasts are fungi of importance to veterinary medicine because they cause infectious diseases in different animal hosts. This literature review aimed to report the main biochemical tests capable of assisting in the identification of yeast-like fungi of veterinary and zoonotic interest. For the bibliographical survey, 48 selected scientific publications in the area and indexed in the main databases, between the years 1988 and 2020, were considered. As a result, it was observed that eight tests are the most used in the mycological routine. Due to the low morphological variability of yeast species, complementary biochemical tests are fundamental in the laboratory routine. The analysis of the biochemical profile of yeast contributes to the taxonomic determination of fungi based on chemical reactions, since the metabolism varies according to the species, resulting in different metabolites, which can be evaluated by different tests. It is concluded that the phenotypic identification of yeasts is essential in the diagnosis, prognosis, treatment and control of fungal diseases and contributes to the maintenance of animal health.(AU)

Para el levantamiento bibliográfico se consideraron 48 publicaciones científicas seleccionadas en el área e indexadas en las principales bases de datos, entre los años 1988 y 2020. Como resultado se observó que ocho pruebas son las más utilizadas en la rutina micológica. Debido a la baja variabilidad morfológica de las especies de levaduras, las pruebas bioquímicas complementarias son fundamentales en la rutina del laboratorio. El análisis del perfil bioquímico de la levadura contribuye a la determinación taxonómica de los hongos en base a reacciones químicas, ya que el metabolismo varía según la especie, dando como resultado diferentes metabolitos, los cuales pueden ser evaluados mediante diferentes pruebas. Se concluye que la identificación fenotípica de levaduras es fundamental en el diagnóstico, pronóstico, tratamiento y control de enfermedades fúngicas y contribuye al mantenimiento de la salud animal.(AU)

This study was conducted to assess the effects of functional oil (FO) blend on performance, blood metabolites, organ biometry and intestinal morphometry in piglets. A total of 128 crossbreed piglets (Landrace × Large White, 64 uncastrated males and 64 females, 21 d of age, and 6.79 ± 1.76 kg BW) were allocated in a randomized complete block design with two dietary treatments: a FO-free (FOF) diet or a diet based on added FO (1,500 mg/kg of diet with castor oil plus cashew nutshell oil). Piglets fed FO showed higher (p ≤ 0.05) average daily feed intake, daily body weight gain and final body weight after 23 d of study. For the total period, piglets fed FO showed greater (p = 0.007) feed conversion ratio. On d 23, higher serum total protein (p = 0.026) and globulin (p = 0.050) concentration, lower liver (p = 0.042) and stomach (p = 0.074) weight, and greater (p = 0.082) villi height (VH) in duodenum were observed in piglets fed FO. Nonetheless, piglets fed FOF showed greater (p = 0.054) ileal VH, but greater (p = 0.004) crypt depth (CD) in jejunum. Piglets fed FO showed higher VH to CD ratio in jejunum (p = 0.068) and duodenum (p = 0.074) on d 23 and 37, respectively. Based on the results, FO blend improved the performance of weaned piglets; however, it negatively affected the feed conversion ratio in the total period. Moreover, FO blend promoted changes in total protein concentrations and improvements in digestive and absorptive capacity assessed through VH to CD ratio, with a significant reduction in organs.
Este estudo foi conduzido para avaliar os efeitos de uma mistura de óleo funcional (OF) no desempenho zootécnico, nos metabólitos sanguíneos, na biometria de órgãos e na morfometria intestinal de leitões. Um total de 128 leitões mestiços (Landrace × Large White, 64 machos inteiros e 64 fêmeas; 21 dias de idade e peso corporal de 6,79 ± 1,76 kg) foram alocados em um delineamento de blocos casualizados completos, com dois tratamentos dietéticos: uma dieta sem OF (SOF) ou uma dieta baseada na adição de OF (1.500 mg/kg de dieta com OF de mamona e castanha de caju). Os leitões alimentados com OF apresentaram maior (p ≤ 0,05) consumo de ração diário médio, ganho de peso corporal diário e peso corporal aos 23 dias de experimento. Entretanto, os leitões que consumiram dietas com OF tiveram conversão alimentar superior (p = 0,007) no período total. No dia 23, houve aumento na concentração de proteína total (p = 0,026) e globulina (p = 0,050), menor peso de fígado (p = 0,042) e estômago (p = 0,074), e maior (p = 0,082) altura de vilosidade (AV) no duodeno em leitões que consumiram OF; entretanto, os leitões SOF tiveram (p = 0,054) AV superior no íleo, mas apresentaram (p = 0,004) profundidade de cripta (PC) superior no jejuno. Uma maior relação AV:PC no jejuno (p = 0,068) e duodeno (p = 0,074) foi observada em leitões com OF nos dias 23 e 37, respectivamente. Com base nos resultados, a mistura de OF melhorou o desempenho dos leitões desmamados; no entanto, afetou negativamente a taxa de conversão alimentar no período total. Além disso, a mistura de OF promoveu alterações nas concentrações proteínas totais e melhorias na capacidade digestiva e absortiva avaliadas através da relação AV:PC, com uma redução significativa nos órgãos.

This study aimed to evaluate the influence of gestation and lactation order on serum concentrations of proteins, metabolites, minerals, and enzymes in first-, second-, and third-lactation order Bonsmara cows. The Bonsmara breed, originated in South Africa, stands out for its tolerance to heat and productive characteristics, such as meat quality, high fertility, sexual precocity, ease of calving, and good maternal ability. These characteristics make the breed a genetic alternative for crossbreeding with zebu breeds. Blood samples were collected from 34, 29, and 30 first-, second-, and third-lactation order cows, respectively, totaling 93 cows. The samples were processed in an automatic multichannel analyzer using Labtest Diagnóstica® kits. The Kruskal-Wallis test was chosen to compare the values between lactation orders. The Mann-Whitney test was used to compare pregnant and non-pregnant cows within lactation orders. Among the analyzed constituents, the lactation order significantly influenced only the serum albumin concentration (ALB) (p<0.0001). Gestation significantly influenced the A:G ratio (p=0.034) in third-lactation cows, and cholesterol (Chol) (p=0.004), triglycerides (TRI) (p<0.0001), inorganic phosphorus (iP) (p=0.033), iron (Fe) (p=0.001), aspartate aminotransferase (AST) (p=0.018), and alkaline phosphatase (ALP) (p=0.039) in second-lactation cows and the general group. Creatinine (Crea) (p<0.0001) was influenced only in the general group. Therefore, gestation and lactation order significantly influenced the concentration of several biochemical serum constituents in Bonsmara cows, especially second-lactation order cows.
Objetivou-se avaliar a influência da gestação e ordem de lactação nas concentrações séricas de proteínas, metabólitos, minerais e enzimas de vacas da raça Bonsmara de primeira, segunda e terceira ordem de lactação. A raça Bonsmara, originada na África do Sul, se destaca por sua tolerância ao calor, e caracteristicas produtivas, como a qualidade da carne, a alta fertilidade, precocidade sexual, facilidade ao parto e boa habilidade materna. Essas características tornam a raça uma alternativa genética para realização de cruzamentos com raças zebuínas. Foram colhidas amostras de sangue de 93 vacas, sendo 34 de primeira ordem de lactação, 29 de segunda ordem de lactação e 30 de terceira ordem de lactação. As amostras foram processadas em analisador automático multicanal, utilizando kits da Labtest Diagnóstica®. Para confrontar os valores entre as ordens de lactação optou-se pelo teste Kruskall-Wallis. Para comparar vacas gestantes e não gestantes dentro das ordens de lactação, foi utilizado o teste de Mann-Whitney. Dos constituintes analisados, a ordem de lactação influenciou significativamente apenas a concentração sérica de albumina (ALB) (p<0,0001). A gestação influenciou significativamente na relação A:G (p=0,034), nas vacas de terceira lactação, no colesterol (COL) (p=0,004), triglicérides (TRI) (p<0,0001), fósforo inorgânico (Pi) (p=0,033), ferro (Fe) (p=0,001), aspartato aminotransferase (AST) (p=0,018) e fosfatase alcalina (FAL) (p=0,039) nas de segunda ordem e no valor do grupo geral. A creatinina (Crea) (p<0,0001) foi influenciada somente no grupo geral. Conclui-se que a gestação e ordem de lactação influenciou significativamente na concentração de vários constituintes bioquímicos séricos de vacas da raça Bonsmara, em especial nas de segunda ordem de lactação.

Identifying the metabolic profile of farm animals constitutes an important tool for the clinical diagnosis of metabolic diseases that can affect animal production performance. This study investigates the prepartum metabolic profile of ewes reared in two feeding systems native pasture and cultivated pasture of black oat and ryegrass and their effects on lamb development. Forty adult Texel ewes and their lambs were used and evaluated for metabolites representative of energy, protein, and mineral metabolism in the prepartum period. Ewes kept on cultivated pasture exhibited higher glucose levels than those kept on native pasture (59.67 vs. 31.98 mg/dL, respectively), whereas those kept on native pasture had higher serum Ca and P levels (7.62 and 4.58 mg/dL, respectively) than the ewes on cultivated pasture (6.21 and 3.73 mg/dL, respectively). Albumin was higher in the ewes with single pregnancy (2.92 vs. 2.76 g/dL), while urea levels were affected by the interaction between feeding system and type of pregnancy. Prepartum levels of glucose, phosphorus, and calcium in the blood of the ewes were correlated with lamb weight at 30 days. The feeding system influenced the metabolic profile of the ewes in the prepartum period, with lamb weight at 30 days of age being mainly associated with the mother's glucose level at the end of the gestation period.(AU)
A identificação do perfil metabólico em animais de produção apresenta-se como uma importante ferramenta no diagnóstico clínico de doenças do metabolismo que podem afetar no desempenho produtivo dos animais. Avaliou-se o perfil metabólico no pré-parto de ovelhas criadas em dois sistemas alimentares, a pasto (pastagem nativa) e pastagem cultivada de aveia preta e azevém e seus efeitos sobre o desenvolvimento dos cordeiros. Foram utilizadas 40 ovelhas adultas, da raça Texel, e seus respectivos cordeiros, nas quais foram mensurados no pré-parto os metabólitos representativos do metabolismo energético, proteico e mineral. Animais mantidos em pastagem cultivada apresentaram maiores valores de glicose em relação aos mantidos em pastagem nativa (59,67 vs 31,98 mg/dL, respectivamente), e os animais mantidos em pastagem nativa apresentaram maiores níveis séricos de Ca e P (7,62 e 4,58 mg/dL, respectivamente) do que os em pastagem cultivada (6,21e 3,73 mg/dL, respectivamente). A albumina foi maior nas ovelhas com gestação simples (2,92 vs 2,76 g/dL) enquanto os níveis de ureia sofreram efeito da interação sistema alimentar vs tipo de gestação. Os teores de glicose, fósforo e cálcio no sangue das ovelhas no pré-parto se mostraram correlacionados com o peso dos cordeiros aos 30 dias. O sistema alimentar influenciou o perfil metabólico das ovelhas no pré-parto sendo o peso dos cordeiros aos 30 dias de idade principalmente associado com o nível de glicose da mãe no final do período gestacional.(AU)

This study aimed to evaluate the effects of animal age and sex on serum concentrations of proteins, metabolites, minerals, and enzymes in Bonsmara cattle up to two years of age. Blood samples were collected from 179 animals (92 males and 87 females), aged between 15 days and 24 months. The animals were divided into four age groups: G1: 15 days â¤2 months, G2: 2 â¤6 months, G3: 6 ⤠12 months, and G4: 12 â¤24 months of age. The samples were processed in an automatic multichannel analyzer using Labtest Diagnóstica™ kits. Animal age had a significant influence on most of the serum biochemical constituents except for magnesium (Mg) and the Ca+:iP ratio. As age increased, G3 showed the highest concentrations of total proteins (TP), globulins (Glob), urea, and cholesterol (Chol); while G4 had a gradual increase and higher values of creatinine (Crea), aspartate aminotransferase (AST), and alanine aminotransferase (ALT). Conversely, albumin:globulin (A:G) ratio, calcium (Ca+), inorganic phosphorus (iP), alkaline phosphatase (ALP), and gamma-glutamyltransferase (GGT) decreased from G1 to G4. Moreover, animal sex influenced serum values of albumin (ALB), Glob, A:G ratio, Crea, urea, triglycerides (TRI), Ca+, iP, Ca+:iP ratio and Mg. In conclusion, the factors of age group and sex significantly influenced the concentrations of serum biochemical constituents in Bonsmara cattle in the growth phase.
Objetivou-se avaliar a infuência da idade e sexo nas concentrações séricas de proteínas, metabólitos, minerais e enzimas em bovinos da raça Bonsmara com até dois anos de idade. Foram colhidas amostras de sangue de 179 animais (92 machos e 87 fêmeas), de 15 dias a 24 meses de idade, distribuídos em quatro grupos etários: G1: 15 dias â¤2 meses, G2: 2 â¤6 meses, G3: 6 â¤12 meses e G4: 12 â¤24 meses de idade. As amostras foram processadas em analisador automático multicanal, utilizando kits da Labtest Diagnóstica®. A variável idade dos animais influenciou significativamente nos valores da maioria dos constituintes bioquímicos séricos avaliados, exceto no magnésio (Mg) e na relação Ca+:Pi. O aumento da idade culminou com a maior concentração das proteínas totais (PT), globulinas (Glob), ureia e colesterol (COL) no G3; aumento gradual e maior valor de creatinina (Crea), aspartato aminotransferase (AST) e alanina aminotransferase (ALT) no G4. Em contrapartida, houve diminuição da relação albumina:globulina (A:G), cálcio (Ca+), fósforo inorgânico (Pi), fosfatase alcalina (FAL) e gama glutamiltransferase (GGT) do G1 ao G4. O fator sexo influenciou nos valores séricos da albumina (ALB), Glob, relação A:G, Crea, ureia, triglicérides (TRI), Ca+, Pi, relação Ca+:Pi e Mg. Conclui-se que o fator idade e sexo influenciaram significativamente nos valores dos constituintes bioquímicos séricos analisados de bovinos da raça Bonsmara em fase de crescimento.

O melaço de soja é um subproduto da indústria da soja, com baixo custo comercial e rico em carboidratos e proteínas. Apesar da grande variedade de subprodutos e de serem potencialmente benéÞ cos, ainda não está totalmente elucidado como ocorre a interação entre os nutrientes deste produto e as mudanças Þ siológicas em ovinos. Dessa forma, objetivou-se avaliar os efeitos de níveis de suplementação com melaço de soja sob os metabólitos energéticos e parâmetros hematológicos de ovinos. Os tratamentos consistiram em 0, 3, 6, 9 e 12% de inclusão de melaço de soja na base da matéria seca para uma dieta basal de silagem de milho, que atendia as necessidades nutricionais das ovelhas em mantença, distribuídas em delineamento em quadrado latino 5 × 5. Foram avaliados os metabolitos energéticos: colesterol, triglicérides, lipoproteínas de muito baixa densidade (VLDL) e frutosamina. Já os parâmetros hematológicos foram: hemácias, hemoglobinas, hematócritos e plaquetas, além de leucócitos, bastonetes, neutróÞ los, eosinóÞ los, monócitos e linfócitos. A suplementação com melaço de soja não alterou signiÞ cativamente (P>0,05) as concentrações séricas de colesterol, triglicérides, VLDL e frutosamina. Não houve diferença signiÞ cativa nas concentrações de hemácias, hemoglobinas, hematócritos e plaquetas das ovelhas com suplementação de melaço de soja (P>0,05), podendo ser indício que o nível de inclusão foi suÞ ciente para atender as necessidades nutricionais dos animais, sem causar balanço energético positivo. As concentrações de leucócitos, bastonetes, neutróÞ los, eosinóÞ los, monócitos e linfócitos não alteraram (P>0,05) com a inclusão de melaço de soja na dieta das ovelhas, não indicando a presença de intoxicação, doenças e infecções. Conclui-se que a suplementação com melaço de soja não altera os metabólitos energéticos e parâmetros hematológicos de ovinos em mantença.(AU)

Background: Ketosis is the most important metabolic disease with prevalence from 15 to 45%. Ketosis is diagnosed using a metabolic profile. Due to the high prevalence, it is necessary to determine a large number of metabolic profiles within farm, which represents an additional cost, so the implementation of pooled serum in assessing the metabolic status of cows was examined. The aim of this study was to validate and evaluate the influence of the relative position (Z-score) of the value of pooled sample metabolic parameters within the known reference value of healthy cows in the detection of ketosis in herd during early lactation. Materials, Methods & Results: The experiment has been carried out using blood samples collected by puncture of coccygeal vein from 50 ketotic and 50 healthy cows. Laboratory analysis includes determination of beta-hydroxybutyrate-BHB, non-esterified fatty acids-NEFA, cholesterol-CHOL, triglycerides-TGC, glucose-GLU, albumin-ALB, total protein-TPROT, UREA, Ca, P, total bilirubin-TBIL and aspartat aminotransferase-AST. The pooled serum was made from 10 individual samples originating from 10 different cows. A serum aliquot of 0.1 mL was taken from each sample, and a 1 mL volume of pooled serum was finally formed. Three types of serum pools were made: 1) 30 pooled sample were from ketosis; 2) 30 pooled sample were from healthy cows and 3) 60 pooled samples containing mixed sera of healthy cows and cows with ketosis were made as follows: 10 pools contain 10% to 60% of ketotic cows (1/10 to 6/10 samples). Statistical analysis includes: a) difference in metabolite concentration and Z-score in pooled sample and arithmetic mean individual sample in healthy and ketotic cow, b) correlation between Z-score of pooled sample and arithmetic mean of individual sample, c) ability of Z-score of metabolite to divide ketotic from healthy cow, d) correlation between Z-score and % of ketotic cow in pooled sample; and e) calculation of 95%CI of pooled sample Z-scores for each % of ketotic cow in pools. Z-score and all analysis were calculated for each metabolic parameter. The results of the study show that the mean values and Zscores of the pool and the calculated average value of the individual samples participating in that pool differ significantly in healthy cows and cows in ketosis, except for TPROT and Ca. A higher value and a higher Z-score were found for BHB, NEFA, UREA, TBIL and AST, and a lower value and a lower Z-score for TGC, CHOL, GLU, ALB and P in ketotic cows compared to healthy cows. The value of the Z-score of the pooled sample and the calculated mean values of individual samples participating in the pool are highly correlated with each other (coefficient of determination over 99%). Z-score of metabolites in the pooled sample can be used to distinguish healthy from ketotic cows (ROC AUC= 0.711 to 0.989), except for TPROT and Ca. The Z-score value of the pooled sample shows a linear correlation with the percentage of ketotic cows in the pool and the reference ranges of Z-scores change significantly as a function of the percentage of ketosis cows. Discussion: Modern research on the metabolic profile in cows requires obtaining a large amount of information from as few samples as possible. The advantages of using the Z-score are reflected in the following: this score does not depend on the absolute value of the metabolite, but on the position within the known population reference value, Z-score of sample and the arithmetic mean of individual samples included in the same pool are almost identical, the Z-score of these 2 groups of results is ideally correlated, the Z-score significantly correlates with the % of ketosis samples in the pooled sample. The use of pooled sample Z-score can be a useful in a herd level assessment of metabolic status and detection of ketosis as most important metabolic disease in dairy cows.

The objective of this study was to evaluate the effects of dietary supplementation of purified lignin on the performance, relative organ weights, serum metabolites, and gene expression profiles of broiler chickens subjected to cyclic heat stress (HS). At 22 days old, 280 broilers were distributed in a completely randomized design with four treatments, ten repetitions, and seven birds per experimental unit. The birds were subjected to daily cyclic HS. A high temperature of 32±1 °C was maintained for 10 h/day (08:0018:00 h), while a temperature of 22±1 °C was maintained for the remaining time. Treatments were a basal diet or basal diet with the addition of 5, 10, or 15 g of purified lignin/kg of diet. Data were analyzed using one-way ANOVA and means were compared by Tukey's test at 0.05 significance. There was no effect of lignin supplementation on performance, carcass yield, relative weights of the bursa, spleen, and liver, or serum levels of glucose, triglycerides, uric acid, malondialdehyde, triiodothyronine, or tetraiodothyronine. The abundance of mRNA of heat shock protein 70, nuclear factor-κB, glutathione peroxidase, and Cu,Zn-superoxide dismutase in the liver was similarly unaffected by treatments. Purified lignin supplementation does not improve performance or the antioxidant response of broiler chickens subjected to HS.(AU)

This study was conducted to evaluate the effects of dietary supplementation with capsaicinoids on the performance and gene expression of broilers. At 18 days of age, 120 male broilers chickens (Cobb 500) were distributed in a completely randomized design with three treatments, eight replicates, and five birds per experimental unit. The treatments were a basal diet or basal diet with the addition of 1 or 2 mg of capsaicinoids/kg of diet. The birds had free access to water and feed throughout the experimental period (18 to 26 days of age). Broiler performance was evaluated at 26 days of age, and one bird per experimental unit was selected to collect serum and jejunum samples. Jejunum samples were used to analyze the mRNA content. Data were analyzed using one-way ANOVA, and the means were compared with Tukey's test at a significance of 0.05. There was no effect of capsaicinoid supplementation on performance, serum metabolites, or the expression of glutathione peroxidase mRNA in the jejunum of broilers. However, broilers supplemented with capsaicinoids showed a higher mRNA expression of Cu, Zn-superoxide dismutase and a reduced mRNA expression of nuclear factor-κB in the jejunum. Supplementation with 1 and 2 mg/kg capsaicinoids did not improve the performance of broilers from 18 to 26 days of age but increased the mRNA expression of Cu, Zn-superoxide dismutase and reduced the mRNA expression of nuclear factor-κB in the jejunum of broilers.(AU)

Pheochromocytoma is a functional neoplasm that produces catecholamines, located in the medullary region of the adrenal glands. As it presents nonspeciÀ c signs, scarcity of diagnostic tools and is still associated with concomitant diseases, it may not be easily considered as a differential condition among veterinarians. This is an uncommon neoplasm that usually affects dogs, with medium to advanced age and without racial predilection, being very rare in cats. Clinical manifestations vary according to the effects of catecholamines and/or circulatory impairment due to the local invasion of the tumor into adjacent structures. In this way, systemic arterial hypertension and consequences in target organs such as the central nervous system and kidneys can be observed, in addition to ocular changes. The deÀ nitive diagnosis is given by the histopathological analysis of the excised adrenal gland. However, the increase in the serum concentration of catecholamine metabolites may contribute to the conÀ rmation of the disease. Adrenalectomy is the therapy of choice, but if this is not feasible, α and ß adrenergic blocking drugs should be used. The prognosis varies from reserved to favorable, based on surgical success and the presence of metastases.(AU)

The study aimed to determine the biochemical reference ranges for lambs from birth to 1 year of age in the tropics. Data were obtained from experiments performed in several institutions and commercial farms using Santa Inês, Dorper, Lacaune, Morada Nova, Bergamacia and Suffolk lambs reared under different conditions (grazing, feedlot, semi-feedlot) from 2006 to 2017. Serum energy-, protein-, mineral-, and enzyme-related metabolites were evaluated. The metabolic energy profile included data from glucose, cholesterol, triglycerides, fructosamine, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol; the metabolic protein profile included total protein, uric acid, urea, albumin, and creatinine; the metabolic mineral profile included data on calcium, phosphorus, and magnesium; and the metabolic enzymatic profile included the enzymes aspartate aminotransferase, gamma glutamyl transferase, alkaline phosphatase, and creatine kinase. The reference ranges were estimated using confidence intervals with a 95% confidence level, and percentiles were estimated or bootstrapped nonparametrically when the data were not normally distributed. The serum biochemical reference ranges determined for lambs are strongly divergent from those established by one of the most cited books on the topic, especially considering the high serum urea and cholesterol concentrations and low levels of blood glucose observed. Therefore, the serum biochemical reference ranges for lambs from birth to 1 year of age in the tropics differ from international data, which consider adult sheep in temperate climate zones.
Objetivou-se determinar as faixas de referência dos parâmetros bioquímicos para cordeiros desde o nascimento até a idade de um ano nos trópicos. Os dados foram obtidos a partir de experimentos realizados em diversas instituições e fazendas comerciais, utilizando cordeiros Santa Inês, Dorper, Lacaune, Morada Nova, Bergamacia e Suffolk criados em diferentes condições (pastejo, confinamento, semi-confinamento) de 2006 a 2017. Foram avaliados os metabólitos séricos relacionados à energia, proteína, minerais e vitaminas. The objective of this study was to evaluate the metabolic status of beef cows during the final third of gestation and thereafter until the end of the subsequent breeding season. The study was conducted using 30 beef cows, aged between 4 and 11 years, derived from crosses between Charolais and Nellore cattle. To assess the metabolic status of cows, we obtained measurements of body weight and condition (BW and BCS), and complemented these by analyzing the following blood metabolites: total proteins, globulins, albumin, glucose, cholesterol, and triglycerides. Assessments were performed at 95 days pre-calving, and again at 7, 21, 63, and 110 days post-calving. The experimental design was completely randomized with five collection periods and 30 repetitions (cows). The BW of cows was found to be highest at 110 days postpartum (489.1 kg), and loss of BW was observed between 21 and 63 days postpartum (-3.72 kg). The lowest BCS value was recorded after 63 days of lactation (2.85 points). With respect to blood metabolites, concentrations of total proteins were higher at 7 and 110 days postpartum relative to the levels recorded during gestation (8.3 and 8.3 vs. 7.7 g dL-1, respectively), whereas serum globulin levels were higher at 7, 21, and 110 days postpartum relative to those at gestation (5.2, 5.2, and 5.1 vs. 4.8 g dL-1, respectively). Compared with samples obtained during gestation, the highest levels of albumin (P < 0.05) were observed at 7, 63, and 110 days postpartum (3.1, 3.1, and 3.2 vs. 2.9 g dL-1, respectively). Serum glucose was higher during pregnancy and in the first week of lactation relative to the values recorded after 21 and 63 days of lactation (80.9 and 76.7 vs 71.5 and 72.3 mg dL-1, respectively). Moreover, cholesterol concentrations increased with the progression of lactation, with the highest value being recorded at 110 days postpartum (222.1 mg dL-1). Collectively, the findings of this study, indicate that Charolais × Nellore cows raised exclusively on native pasture mobilize body reserves, lose weight, and show a negative energy balance during the first weeks of lactation, with detrimental consequences for subsequent reproductive activity.(AU)
Avaliar o estado metabólico de vacas de corte do terço final de gestação até o final da estação reprodutiva subsequente foi o objetivo do presente estudo. Foram utilizadas trinta vacas de corte, oriundas do cruzamento entre as raças Charolês e Nelore, com idade entre 4 a 11 anos. Para avaliação do estado metabólico das vacas, foram tomadas as medidas de peso e escore de condição corporal (PC e ECC), sendo estas variáveis complementadas pela análise dos metabólitos sanguíneos: proteínas totais, globulinas, albumina, glicose, e colesterol e triglicerídeos. Estas avaliações foram realizadas aos 95 dias pré-parto e aos sete, 21, 63 e 110 dias após a parição. O delineamento experimental foi o inteiramente casualizado com cinco períodos de coleta e 30 repetições (vacas). As vacas apresentaram o maior PC aos 110 dias pós-parto (489,1 kg). Foi observado perda de PC entre 21 e 63 dias pós-parto (-3,72 kg). Menor valor de ECC foi obtido aos 63 dias de lactação (2,85 pontos). A concentração das proteínas totais foi maior aos sete e 110 dias pósparto em relação aos níveis obtidos durante a gestação (8,3 e 8,3 vs. 7,7 g dL-1, respectivamente). Níveis séricos de globulinas foram maiores aos sete, 21 e 110 dias pós-parto em relação à gestação (5,2; 5,2 e 5,1 vs. 4,8 g dL-1, respectivamente). Os maiores níveis de albumina foram observados aos sete, 63 e aos 110 dias pós-parto quando comparados à amostragem durante a gestação (3,1; 3,1 e, 3,2 vs. 2,9 g dL-1, respectivamente). A glicose sérica foi maior durante a gestação e na primeira semana de lactação em relação às coletas realizadas aos 21 e 63 dias de lactação (80,9 e 76,7 vs 71,5 e 72,3 mg dL-1, respectivamente). A concentração de colesterol aumentou com o avanço da lactação, com maior valor aos 110 dias pós-parto (222,1 mg dL-1). Vacas Charolês × Nelore criadas exclusivamente em pastagem nativa mobilizam reservas corporais, perdem peso e apresentam quadro de balanço energético negativo nas primeiras semanas de lactação, com consequências na atividade reprodutiva subsequente.(AU)

This study aims to evaluate the use of active, inactive plus active, and inactive yeasts on the consumption of dry matter, water, and the apparent digestibility of dry matter, urinary parameters, and serum metabolites of lambs. Twenty crossbred lambs (Dorper x Santa Inês) were used, with an initial average body weight of 31.89 kg and seven months of age, distributed in a completely randomized design. Treatments were Control group (without yeasts); Active Flora® (ICC), live yeast plus inactivated yeast (Saccharomyces cerevisiae, 2.0 x 1010 UFC g-1) in the dose of 0.003 kg of animal dry matter -1 day-1,; Milk Sacc X® (Alltech®), active yeast - Saccharomyces cerevisiae strain 1026, 5.0 x 108 UFC g-1 - at the dose of 0.0015 kg of animal dry matter-1 day-1; and Rumen Yeast® (York Ag Products INC.), inactive yeast - Saccharomyces cerevisiae,1.5 x 104 UFC g-1 - in the dose of 0.0045 kg of animal dry matter-1 day-1. Variance analysis and the SNK (Student-Newman-Keuls) test were performed considering 5% significance. For glycemic concentrations over time, regression analysis at 5% significance were performed. The fecal score, as a non-parametric variable, was assessed by the Kruskal and Wallis test at a 5% significance level. There was a statistical difference (P< 0.05) in fecal dry matter (FDM), where the Active Flora® treatment was inferior to the others. The use of different yeasts did not change the intake and digestibility of dry matter, water, urinary parameters, and serum concentrations of energy, protein, and liver metabolites (P> 0.05). The average dry matter intake was 1.16 ± 0.16 kg day-1, whereas the dry matter digestibility was 85.40 ± 2.73%. It was concluded that active and/or inactive yeasts can be used as additives in the diet of lambs, maintaining the intake and digestibility of dry matter without causing metabolic disturbances.(AU)

Os tratamentos consistiram em Controle, sem uso de enzimas, e no uso das leveduras Active Flora® (levedura viva junto a levedura inativada - Saccharomyces Cerevisiae, com 2,0 x 1010 UFC g-1), Milk Sacc X® (levedura ativa - Saccharomyces C. 1026, 5,0 x 108 UFC g-1) e Rúmen Yeast® (levedura inativa - Saccharomyces C. com 1,5 x 104 UFC g-1). Foi realizado uma análise de variância e um teste SNK considerando 5% de significância. Para as concentrações glicêmicas ao longo do tempo foi realizada uma análise de regressão a 5% de significância. O escore fecal, por ser uma variável não paramétrica, foi avaliado pelo teste Kruskal e Wallis à significância de 5%. Observou-se a diferença estatística (P< 0,05) na matéria seca fecal (MSF), onde o tratamento Active Flora® se mostrou inferior aos demais. O uso de diferentes leveduras não modificou o consumo e a digestibilidade da matéria seca, de água, dos parâmetros urinários e das concentrações séricas dos metabólitos energéticos, proteicos e hepáticos (P> 0,05). A média de consumo da matéria seca foi de 1,16±0,16 kg dia-1, já a digestibilidade da matéria seca foi de 85,40 ±2,73%. Concluiu-se que as leveduras ativas e/ou inativas podem ser utilizadas como aditivos na dieta de borregas, mantendo o consumo e a digestibilidade da matéria seca sem causar distúrbios metabólicos.(AU)

Purpose To analyze the plasma lipid spectrum between healthy control and patients with pancreatic cancer and to select differentially expressed tumor markers for early diagnosis. Methods In total, 20 patents were divided into case group and healthy control group according to surgical pathology. Of almost 1206 plasma lipid molecules harvested from 20 patients were measured by HILIC using the normal phase LC/MS. Heat map presented the relative levels of metabolites and lipids in the healthy control group and patients with pancreatic cancer. The PCA model was constructed to find out the difference in lipid metabolites. The principal components were drawn in a score plot and any clustering tendency could be observed. PLS-DA were performed to distinguish the healthy control group and pancreatic cancer according to the identified lipid profiling datasets. The volcano plot was used to visualize all variables with VIP>1 and presented the important variables with P 0.01 and ,FC,>2. Results The upregulated lipid metabolites in patients with pancreatic cancer contained 9 lipids; however, the downregulated lipid metabolites contained 79 lipids. Conclusion There were lipid metabolomic differences in patients with pancreatic cancer, which could serve as potential tumor markers for pancreatic cancer.(AU)

This study aimed both to evaluate the metabolic profile of lambs fed with extruded forage and concentrate containing different roughageconcentrate ratios (R:C) and to evaluate in-vitro ruminal fermentation kinetics of these rations. An experiment was carried out with 20 weanling, female crossbred lambs (½Dorper x ½Santa Inês) aged 120 ± 8 days. The treatments were four R:C ratios of extruded forage (Foragge®) and extruded concentrate (Beef Total®) 30%F:70%C, 40%F:60%C, 50%F:50%C, and 60%F:40%C distributed randomly with five replicates. The analysis targeted energy metabolites (VLDL, triglycerides, and cholesterol), hepatic metabolites (alkaline phosphatase, gamma glutamyltransferase [GGT], and aspartate aminotransferase [AST]), and protein metabolites (total protein, uric acid, urea, creatinine, albumin, and globulin). In-vitro gas production was quantified by inoculating the extruded rations in bovine ruminal fluid with the following forage:concentrate proportions evaluated: 30%F:70%C, 40%F:60%C, 50%F:50%C, 60%F:40%C and 70%F:30%C. Fermentation kinetic values were compared using parallelism test and curve identity at the error probability level of 5%. A regression study was used to compare the means of the metabolites at the error probability level of 5%. Average dry matter intake data are provided descriptively. The extruded ration increased the hepatic metabolites (GGT and AST) without compromising the liver and kept the energy metabolites stable. Similarly, the serum concentration of protein metabolites remained at normal levels recommended to the ovine species. In-vitro fermentation pattern displayed high gas production associated with high ration digestibility. The extruded ration provides protein and energy supply to meet the animals nutritional requirements without compromising their health.
Objetivou-se avaliar (i) o perfil metabólico de borregas alimentadas com volumoso e concentrado extrusados em diferentes proporções de volumoso:concentrado (V:C) e (ii) a cinética de fermentação ruminal in vitro dessas rações. Foram utilizadas 20 fêmeas mestiças ½Dorper x ½Santa Inês recém-desmamadas com 120 ± 8 dias de idade. Os tratamentos foram quatro relações de volumoso extrusado (Foragge® ) e concentrado extrusado (Beef Total® ): 30%V:70%C, 40%V:60%C, 50%V:50%C e 60%V:40%C, distribuídos inteiramente ao acaso com cinco repetições. Os metabólitos energéticos analisados foram: VLDL, triglicerídeos e colesterol. Os metabólitos hepáticos foram: fosfatase alcalina, gama glutamiltransferase (GGT) e aspartatoaminotransferase (AST). Os metabólitos proteicos avaliados foram: proteínas totais, ácido úrico, ureia, creatinina, albumina e globulina. A quantificação da produção in vitro de gás foi feita com as rações extrusadas inoculadas em líquido ruminal de bovinos, avaliando as seguintes proporções volumoso:concentrado: 30%V:70%C, 40%V:60%C, 50%V:50%C, 60%V:40%C e 70%V:30%C. Os valores de cinética de fermentação foram comparados por meio do teste de paralelismo e identidade de curvas ao nível de 5% de probabilidade de erro. Para comparação das médias dos metabólitos, foi adotado estudo de regressão a 5 % de probabilidade de erro. Os dados médios de consumo de matéria seca são apresentados de forma descritiva. A ração extrusada provocou aumento nas enzimas hepáticas (GGT e AST), sem provocar lesões hepáticas e manteve estáveis os metabólitos energéticos. Similarmente, os níveis séricos proteicos permaneceram nos níveis normais recomendados para a espécie. O padrão de fermentação in vitro mostrou alta produção de gases associadas à alta digestibilidade da ração. A ração extrusada fornece aporte energético e proteico para atender às demandas nutricionais dos animais sem comprometer a sua saúde.
