Spermiogenesis is the final process of spermatogenesis in which the spermatid transforms into sperm. During this process, sperm alterations may occur, especially in the acrosome, head morphology, chromatin condensation and formation of nuclear vacuoles. Differentiation between clinical conditions is made with repetitions of andrological exams. The morphological evaluation indicates the status of spermiogenesis in the 4 weeks prior to semen collection. With the results of the breeding soundness examination, it is possible to identify the seminal quality in that period. The present article is a case report of a young 26-month-old semen donor bull, that started semen collection routines at a Semen Collection and Processing Center in southern Brazil. The sperm production of this animal was evaluated during 12 months of collection, always showing very altered sperm morphology and average motility of 54% in the immediate examination. In 13 seminal collections, the ejaculate of this animal presented an average of 86% of major defects, 10% of minor defects and 96% of total defects. Major defects, which have the greatest impact on fertility, are directly linked to spermiogenesis and minor defects, which have less effect on fertility, are mainly caused during transit through the epididymis. The clinical conditions of this animal demonstrated the importance of the morphological examination to evaluate spermatogenesis and especially the process of final differentiation of the spermatid into spermatozoa. Cases like this should be described as altered spermiogenesis and the bull should be withdrawn from breeding, after ruling out morphological changes due to severe testicular degeneration.(AU)

Ejaculation is a complex process of neurophysiological events synchronized in coordination with various systems and organs. Before the advent of intracytoplasmic sperm injection (ICSI), ejaculation was an absolutely essential step in reproduction. Ejaculatory disorders, in turn, are characterized by the nonoccurrence of the ejaculatory process, even if all other parameters related to sexual behavior are normal. The main causes of these failures or changes in ejaculation are related to painful sensitivity during copulation, retrograde ejaculation, psychogenic dysfunction, obstructions of the male reproductive system, urospermia, oligospermia, azoospermia, in addition to failures in the contraction of the smooth muscles of the genital tract or musculoskeletal disorders and neurological. Treatment is dependent on the primary cause of each change. Pain control, management adjustments, increased stimulus before collection associated with pharmacological treatments that act on penile neuromotor synapses tend to resolve the main ejaculatory disorders, taking into account that the responses to these treatments vary according to the individuality of the stallion with regard to dosages, protocols, routes of administration and drug combinations.(AU)

The objective of this work was to evaluate the seminal viability of rabbits after refrigeration at 5°C using three commercial diluents, indicated for refrigeration of semen from other monogastric species. Ten semen collections were performed from each breeder. The ejaculates of each rabbit were mixed, obtaining a pool of semen. Then, the heterospermic pool was divided into three treatments, according to the diluent used: BotuDog (dogs); BotuSemenâ (horses); and Beltsville Thawing Solution - BTS (pigs). The samples were evaluated for macroscopic, microscopic, and functional parameters in four moments: fresh (0 h) and refrigerated (16, 24, and 48 h) semen. The average values of motility, vigor, and total defects of fresh semen were 86.0%, 3.05 and 11.5%, respectively. After the beginning of cooling, a decreasing linear behavior was observed for motility and sperm vigor of semen as a function of time, differing (P < 0.05) between treatments. It is noteworthy that the treatment diluted in BTS® showed a more pronounced reduction in the maintenance of the evaluated parameters (P < 0.05) over time, with the lowest values for sperm motility and vigor within each evaluated time: 16h [20.6 +7.28 and 0.75 (0.375 - 1.5)], 24h [12.0± 6.80 and 0 (0 - 1.0)] and 48h [3.0±2.13 and 0 (0 - 0.125)]. After 48 h of cooling, BotuDog presented the highest value for sperm motility (71.0 ± 2.08) and, together with BotuSemenâ, presented the highest value for sperm vigor (2.5). According to the data obtained, BotuDog and BotuSemenâ diluents were efficient in maintaining adequate sperm parameters, showing good sperm viability in up to 48 hours after refrigeration at 5°C. Through this work, unpublished results were obtained regarding spermatic parameters of refrigerated rabbit semen that can be used as reference values for reproductive management and semen processing of these animals, given the lack of these technical recommendations.

The objective of this study was to evaluate characteristics of the testicular parenchyma and vascular parameters of the pampiniform plexus obtained by ultrasound, semen quality parameters, and sperm freezability in Nellore bulls classified based on residual feed intake (RFI). Twenty-seven bulls (21.82±0.88 months of age) evaluated for feed efficiency were sampled for the study, including 15 with low RFI (−0.592±0.09 kg dry matter/day) and 12 with high RFI (0.792±0.10 kg dry matter/day). In ultrasound and Doppler assessment, the most efficient animals (low RFI) showed higher pulsatility and resistive indexes, as well as a tendency towards greater heterogeneity of the testicular parenchyma (0.625±0.032 vs. 0.508±0.032, 1.012±0.072 vs. 0.802±0.072, and 12.9±0.96 vs. 10.2±0.96, respectively, for low vs. high RFI). However, these animals tended to have lower peak diastolic velocity (5.19±0.50 for low RFI vs. 6.54±0.50 for high RFI). Analysis of fresh semen showed a lower percentage of minor defects in low RFI animals (2.67±1.19%) compared with high RFI animals (8.10±1.19%), without differences in the other parameters in fresh or thawed semen and after thermoresistance testing. Evaluation of flow cytometry parameters showed a higher quality of mitochondrial respiration in semen samples of low RFI animals (22.04±2.50%) compared with high RFI animals (12.29±2.71%). Therefore, although RFI exerts an effect on the Doppler parameters of the pampiniform plexus, it is not sufficient to affect the quality of fresh or thawed semen.(AU)

Mugil curema valenciennes, 1836 (Osteichthyes: Mugilidae) is a cosmopolitan fish popularly known as parati in Southeast and South Brazil and as mullet in Northeast, being found in the open sea and estuaries. This species does not express sexual dimorphism between males and females externally. Males and females have a pair of gonads (testes and ovaries, respectively), which are elongated, fusiform, and symmetrical. This paper describes the occurrence of testicular hypoplasia in a fish of the species Mugil curema, from Babitonga Bay, Santa Catarina, Brazil. Unilateral testicular hypoplasia was identified, which macroscopically showed a decrease in testis volume and, microscopically, the absence of spermatogenic action and a reduction in the number of spermatozoa. It was not possible to determine the factors that may have contributed to the development of such reproductive alteration. However, as the living and reproductive traits of this animal species may be influenced by anthropic action, a continuous study of its life in this environment is important. There are few studies directed at this species, mainly on its reproductive aspects. Thus, this report will contribute to raising the information historic about this species in the region.

Morphological sperm evaluation supported by the morphometry can be used in the determination of the seminal quality and in the investigation of potential extenders. Although there are studies comparing TRIS and ACP extenders, there are no comparative studies between them for the computerized assisted semen analysis (CASA), sperm viability, membrane functionality and sperm morphometry parameters of cryopreserved canine semen. Hence, we aimed to evaluate the effects of ACP-106c and TRIS on post-freezing canine sperm quality. Five dogs were submitted to semen collection twice with one-week interval. The semen was evaluated within the parameters: total motility, vigor, concentration, viability, plasma membrane functionality, morphology and morphometry. In the morphometric evaluation, the morphologically normal sperm was measured as: length, width, area and perimeter of the head and the midpiece, tail length and total length. The parameters of ellipticity, elongation, regularity and roughness were determined. Then, the semen was divided into two aliquots that were diluted in TRIS or ACP-106c, with the addition of egg yolk and glycerol. The diluted semen was refrigerated and frozen. The thawed samples were evaluated. Total motility, viability, sperm membrane functionality and normal morphology reduced after thawing in both extenders (morphology reduced from 89.60 ± 1.3% to 84.40 ± 1.8 and 84.60 ± 1.1% in TRIS and ACP-106c, respectively). However, it did not differ between TRIS and ACP-106c. In the ACP106c the sperm head defects in cryopreserved semen were higher compared to fresh semen (P < 0.05). For all the morphometric parameters evaluated, there were no differences between fresh and cryopreserved samples (3.70 ± 0.4% vs. 2.30 ± 0.5%). In kinetics, with an interval of one week statistical differences between the extenders were found only in the parameters ALH and LIN (P < 0.05). Regardless of the extender, there were no changes in the morphometric parameters of sperm after thawing.(AU)

Waste oil from olive oil extraction industry was used, instead of soybean oil, in heavy roosters diet in order to evaluate birds reproductive parameters. Atotal of forty roosters were housed individually in boxes with 1.2 m². Two experimental diets were used: control diet, based on corn, soybean meal, and soybean oil; and test diet, where soybean oil was totally replaced by waste oil. In order to verify weight gain and feed intake, animals were individually weighed weekly. Seven semen collections were performed with fifteen-day interval. Reproductive variables analyzed sperm volume, motility, concentration, and morphology. No statistical difference (p >0.05) was observed between treatments at the different collection periods for the variables sperm volume, motility, and concentration. There was a statistically significant difference between treatments for body weight in periods three (p =0.04), and seven (p=0.04). Statistical differences (p =0.01) were also observed between treatments for abnormal sperm morphology. Among collection periods, statistical difference was observed for motility (p =0.00), and sperm concentration (p =0.01). Total replacement of soybean oil by waste oil from olive oil extraction in young heavy roosters diets does not affect sperm volume, motility, and concentration; reduces defects in sperm tail, and promotes better weight gain control.(AU)

Sperm routinary fitness evaluation is not sufficient to predict bull reproductive capacity as they present differences in fertility up to 40%. Among the defects which compromise spermatozoa functionality, new approaches consider the study of sperm chromatin, which is the core structure containing paternal genetic information. Sperm chromatin needs to be compacted to maintain the integrity of DNA, which occurs by binding nucleoproteins with high affinity to DNA. In the last stages of sperm maturation, chromatin is hyper-compacted by basic proteins called protamines in a process named protamination. In this review, we summarized intrinsic and extrinsic factors that are suggested to influence protamination in bull spermatozoa, considering old and new evidence from human and murine spermatozoa. Also, the current approaches to evaluate bull protamination and its relationship with fertility were described. Nevertheless, the physiological mechanisms of protamination are still poorly understood.(AU)

Sperm morphology is a seminal trait that is associated with fertility and is therefore an important component of routine quality control in Semen Collection and Processing Centers (SCPC). It is used to characterize and quantify the various types of abnormalities in bull sperm cells, using wet mount preparation for Differential Interference Contrast (DIC), Phase Contrast (PC) and Slides Smear (SS) microscopy. The sperm morphology profile allows decision making on the use or disposal of an ejaculate for cryopreservation and predicts the fertility of a semen sample. Changes in sperm morphology may indicate that fertility may be impacted and therefore it is important to routinely monitor ejaculates. An abnormal spermogram with evidence from a history and physical examination of the bull can lead to the interpretation of abnormal testicular function, and consequently, to the diagnosis, treatment and prognosis, if any. The most common causes of abnormal spermatogenesis in bulls include: abnormal testicular thermoregulation, hormonal imbalances, management, environmental and nutritional stress, effects of toxins or expression of deleterious genes, in addition to defects of genetic origin. Knowledge in the structure of the sperm makes easier the identification of sperm abnormalities, as well as their description and provides a better understanding of their origin.(AU)

The growth, sexual maturity and fertility-related parameters related of young Nellore bulls with divergent residual feed intake (RFI) raised on pasture were evaluated. After classification of 48 young males as low and high RFI (more and less efficient, respectively), the animals were evaluated for growth and reproductive parameters at 28-day intervals from 14.3 to 24.6 months of age. The semen was cryopreserved in the last sampling and fresh and post-thaw semen samples were evaluated. Low RFI bulls exhibited higher initial and final body weight (P < 0.05), but feed intake, body condition score and growth measures evaluated by carcass ultrasound were unaffected by RFI (P > 0.05). The scrotal circumference, sperm concentration, defects, and quality of fresh semen, and ultrasonographic testicular characteristics were unaffected by RFI (P > 0.05). However, velocity parameters such as average path and curvilinear velocities determined by computer-assisted sperm analysis of thawed semen submitted to the rapid thermoresistance test were improved (P < 0.05) in low RFI bulls, but this improvement in quality did not enhance in vitro sperm fertilizing ability. Our results demonstrated significant differences in metabolism and growth performance between bulls of divergent RFI. In addition, there was slight improvement in the semen quality of bulls with low RFI bulls, but this did not enhance in vitro fertilizing ability. Selection of beef bulls for RFI can be performed, which will result in economic benefits by improving the growth performance of the animals without affecting reproductive parameters.(AU)

The objective of this study was to describe and compare the head morphometry of normal and pathological sperm from Saimiri macrodon. In the morphological analysis, 39% of the sperm had normal morphology, and 61% had major or minor defects, with pathology in the tails being the most frequent with 47% (38% curled tail, 7% folded tail and 2% strongly folded tail). Among the evaluated head morphometry parameters, area (A), width (L) and ellipticity (E) showed statistical difference (p>0.05) between normal and pathological sperm. The average head area and width was lower in normal sperm (p=0,01 e p=0,04, respectively), and the mean ellipticity was higher (p=0,038), when compared to pathological sperm. This definition of the sperm morphometric parameters of S. macrodon is important for the samples selection destined to reproduction biotechnologies and for the clarification of taxonomic and evolutionary issues in the genus Saimiri.

Environmental pollution is the unfavorable alteration of our environment, wholly or mainly as a by-product of human actions, by the direct or indirect effects of changes in energy patterns, radiation levels, the chemical and physical constitution, and the abundance of the organisms. Thus, the environment's pollution is one of the most severe problems humanity and other forms of life face today on our planet, and this population makes severe disruption. The xenobiotic substances responsible for this pollution are numerous and diverse due to multiple human activities that can be the source. However, attention mainly focuses on fertilizers, pesticides, heavy metals, and certain petrochemicals of significant consumption, such as solvents. The purpose of this study is to highlight the reprotoxic and hepatotoxic effects of Ethylene Glycol Monoethyl Ether (EGME) on male Wistar rats. Male Wistar rats were exposed to Ethylene Glycol Monomethyl Etheralone (EGME) (500 mg/kg), combined with the aqueous extract of the pollen grains of Sinapis arvensis (P-EGME) (300mg/kg), in addition to the control (T) and positive control (P) groups for a period of 4 weeks by gavage, to estimate the protective role of this plant against the intoxication of EGME. The results show that EGME can induce reprotoxic effects revealed by a reduction in testes and epididymis mass accompanied by decreased male fertility indicators (sperm concentration). The results also show that treatment with EGME caused a reduction in the red blood cell number, hemoglobin and hematocrit rate, white blood cells in the treated groups compared to the control groups. The present study revealed that treatment with Ethylene Glycol Monoethyl Ether (EGME) under the same experimental conditions could affect several biological markers, especially male fertility. A hipertermia testicular compromete a espermatogênese, reduz a concentração seminal, a motilidade progressiva e a viabilidade espermática.
Sheep farming is a relevant activity that provides proteins of high biological value, but its gains can be reduced due to environmental stress. This reinforces the importance of studying the relationship between the thermal environment and the animal, identifying more adapted and fertile animals, and better management practices. Sheep (Ovis aries) are homeothermic animals and thus maintain their body temperature in a state of dynamic balance. Under heat stress, sheep dissipate accumulated heat through sensitive and latent mechanisms, primarily using redirection of blood flow, panting and sweating. The scrotum plays a crucial role in the thermoregulation of the testicles, which need to be maintained up to 6.0oC below the body core temperature. Testicular hyperthermia impairs spermatogenesis, reduces seminal concentration, progressive motility and sperm viability. Furthermore, it leads to an increase in sperm morphological defects, in the production of reactive oxygen species, and in sperm DNA fragmentation, reducing the fertilizing capacity. Disruptive technologies for monitoring the production systems, animal thermoregulation and welfare are already a reality and are expanding, favoring realtime decision making and the reproductive performance of sheep.

The objective of this study was to determine the scrotal thermographic profile and to verify the influence of temperature and humidity of the humid tropical climate on testicular temperature and seminal quality of Mangalarga Marchador stallions. The thermal profiles of the proximal, middle, and distal zones of the testicles and total surface temperature (TSTT) were recorded using an FLIR E60bx thermal imager. The average air temperature (°C) and relative humidity (%) were obtained 1, 5, 9, 33, and 66 days before semen collection and showed a mean value of 26.5±2.4 and 80.4±6.0 respectively. The scrotal surface temperature was close to 34°C and there was no variation with the age of the stallion, reproductive activity, and characteristics of the ejaculate (P>0.05). The only significant correlations obtained were between TSTT and minor defects (R = 0.41; P<0.05), between TSTT and total defects (R = 0.46; P<0.01), and between TSTT and percentage of morphologically normal sperm (R = -0.46; P<0.05). It was concluded that the Mangalarga Marchador stallions maintained the testicular temperature within favorable conditions for spermatogenesis, demonstrating the efficiency of testicular thermoregulation mechanisms in the Atlantic Forest biome.

The objective was to evaluate the sperm morphometry between normal and pathological cells in Saimiri cassiquiarensis aiming at the future use of data in CASA (Computer Assisted Sperm Analysis). Semen was collected from a male (S. cassiquiarensis) by electroejaculation. After collection, a smear in the proportion of 1:1 (semen and eosin-nigrosin stain) was performed, and 100 sperm were measured and morphologically classified as normal and pathological defects (major and minor). Of the total sperm analyzed, 48% were classified as normal and 52% pathological. Of the pathologies, the most frequent was a folded tail (40%), followed by a strongly folded tail (5%), curled tail (2%) and isolated head (5%). The morphometric data showed no statistical difference (p<0.05) between normal and pathological defects. This is the first description for the sperm morphometry in Saimiri cassiquiarensis, therefore, from this database, future assessments at CASA can be conducted for this species.

Awareness of the physiological changes that occur when animals are subjected to climatic changes that are considered stressful is essential to maintain animal welfare and to be able to exploit their reproductive potential efficiently and rationally. The present study was carried out to evaluate climatic variables' influence on physiological parameters, and Murrah buffalo ejaculates reared in a humid tropical climate in the Amazon. The immediate analyzes pertinent to the physical and morphological characteristics of the ejaculates were carried out and corresponded in the rainy season (RS) volume of 3.4±2.0mL; the mass activity of 4.4±0.5; motility of 80.4±5.6%; vigor of 4.4±0.4; concentration of 657,300±237,865.1 x 106sptz/mL; major defects of 9.0±2.6%; minor defects of 11.2±3.9%; total defects 20.2±5.3% and sperm plasma membrane integrity (SPMI) 84.8±5.6%, whereas in the non-rainy season (nRS), the results were 4.0±2.1mL; the mass activity of 3.0±1.0; motility of 56.2±13.4%; vigor of 3.0±1.0; concentration of 586,000±291,925.9 x 106sptz/mL; major defects of 20.8±9.9%; minor defects of 27.5±6.3%; total defects 48.3±9.3% and SPMI of 57.9±12.4%. Furthermore, a statistical difference (P<0.05) was observed for the parameters mass activity, motility, vigor, major defects, minor defects, total defects, and sperm plasma membrane integrity between both periods. The data on heart frequency, superficial temperature (head, back, groin, and scrotal pouch) showed a statistical difference between both periods (P<0.05). To conclude is necessary specific management in the non-rainy season that thermal stress is not a determining factor in reducing the reproductive quality of buffaloes; it is necessary to use means to improve animal welfare; one alternative is to use baths regularly for these animals or provide constant access to areas of rivers or lakes, as well as shading, preventing the buffaloes from being directly exposed to the unfavorable thermal environment.(AU)
