Resumo

O sêmen de oito reprodutores foi coletado utilizando a técnica da mão enluvada sendo utilizado o ejaculado total. De cada ejaculado foi retirado 10,2 x109 sptz para cada tratamento. A qualidade do sêmen foi avaliada pela concentração (x106 sptz/mL), volume (mL), total de espermatozoides (x109 sptz), vigor espermático (0 a 5), porcentagem de células móveis (%), morfologia espermática (%) e integridade de membrana (%). O sêmen foi diluído no Beltsville Thawing Solution (BTS) e na água de coco em pó (ACP). O ejaculado foi diluído em partes iguais (1:1) e incubado entre 30 oC e 15 oC. Os exames da integridade do acrossoma e vitalidade foram feitos no sêmen puro e diluído pela técnica do azul de bromo-fenol em aumento de 1000x. A integridade da membrana plasmática foi avaliada pela coloração fluorescente em microscopia óptica de campo escuro. O delineamento experimental utilizado foi o de blocos ao acaso, utilizando-se os testes de Mann Whitney, Tukey e Student, com análise das diferenças entre médias feita por variância multifatorial com o General Linear Models do programa Statistical Analysis System (SAS 6.03), a um intervalo de confiança de p<0,05. As avaliações do sêmen in natura, apresentaram valores médios de vigor espermático (4,2) e de porcentagem de espermatozoides móveis (92%) acima dos parâmetros mínimos exigidos (3,0 e 70%) para sua utilização em programas de inseminação artificial. Na característica porcentagem de células móveis, o ACP (74%) foi significativamente melhor que o BTS (67%) a 15 oC. Os melhores resultados foram obtidos com o uso do diluente ACP: acrossoma intacto (49,2%); células vivas (62,5%) e membrana íntegra (65,6%). O diluente ACP proporcionou um meio de conservação que manteve maior número de espermatozoides vivos com membranas íntegras, podendo ser recomendado para uso(...)(AU)
The semen of eight swine males was collected by the glove handled technique, being used the total ejaculated. It was retired from each ejaculated a total of 10.2 x109 sptz, distributed among the treatments. The semen quality was evaluated by the concentration (x106 sptz/mL), volume (mL), total of spermatozoa (x109 sptz), spermatic vigour (0 to 5), movable cells (%), spermatic morphology (%) and membrane integrity (%). The semen was diluted in Beltsville Thawing Solution (BTS) and the coconut water powdered extender (ACP), in the same parts (1:1) and incubated between 30 oC and 15 oC. The exams of acrossome integrity and vitality, were made in the pure and diluted semen, by the bromophenol blue technique in increase at 1000x. The integrity of plasmatic membranes was evaluated by the fluorescent coloration in optical microscopy of dark field. The experimental statistic test used was the maybe blocks, with the Mann Whitney, Tukey and Student analysis, in the Lineal General Models of Statistical Analysis System Program (SAS 6.03), with a trust interval of p<0.05. The in nature semen evaluations, presented medium values of spermatic vigour (4.2) and the movable spermatozoa (92%) above the minimum demanded parameters (3.0 and 70%) for your use in artificial insemination programs. In the percentage of movable cells characteristic, the ACP extender (74%) it was significantly better than BTS (67%) at 15 oC. The best results were obtained with use of the ACP extender: intact acrossome (49.2%); alive cells (62.5%) and complete membrane (65.6%). The coconut water extender provided a conservation middle that maintained a larger number of alive spermatozoids with integrate membranes and could be recommended for routine use in reproduction laboratories. The ACP extender demonstrated to be a good middle of swine spermatozoa conservation in temperatures between 17 and 15 °C, and may be indicated for use in artificial insemination programs.(AU)
