INFLUÊNCIA DO CONSUMO PROLONGADO DE SEMENTE DA LINHAÇA (Linum usitatissimum) SOBRE A HISTOARQUITETURA DO TESTÍCULO E EPIDÍDIMO DO RATO Niterói

Resumo

A semente de linhaça é considerada um alimento funcional, exercendo vários efeitos benéficos sobre a saúde. Entretanto, devido ao seu elevado teor de fitoestrógenos, são capazes de influenciar o metabolismo hormonal e interferir na biomorfologia gonadal. O objetivo deste trabalho é avaliar através de histomorfometria computadorizada os túbulos seminíferos e epididimários, considerando as diferentes regiões do epidídimo (cabeça, corpo e cauda) em ratos submetidos a uma dieta prolongada de semente de linhaça. Foram utilizados ratos Wistar machos adultos jovens (n=20) divididos em 2 grupos, durante o período de lactação: Grupo Controle (GC) a base de caseína e Grupo Linhaça (GL) com adição de 25% de semente de linhaça. Ao final de 250 dias de ingestão contínua, os animais foram sacrificados, seus testículos/epidídimos retirados e fixados em formol tamponado. As amostras foram submetidas ao processamento histológico de rotina para parafina e coradas em HE. Foi feita a imunomarcação com anticorpo anti-vimentina para identificação das células de Sertoli. Para morfometria, as imagens das lâminas foram digitalizadas e analisadas pelo software ImageJ para obtenção dos dados de altura epitelial, diâmetro luminal, diâmetro tubular e área tubular. As análises estatísticas foram realizadas pelo programa GraphPad InStat através do Teste t Student. Na avaliação hormonal, o GL teve maior concentração de estrógeno sérico (p=0,001), mas não apresentou alteração significativa na concentração de testosterona. Em relação aos parâmetros morfométricos dos túbulos seminíferos e das regiões epididimárias, não houve diferenças significativas entre os grupos analisados. Na quantificação das células de Sertoli também não houve diferença significativa no GL (p=0,98). Os resultados mostraram que o consumo contínuo e prolongado de 25% de semente de linhaça desde período gestacional até 250 dias de idade não acarreta efeitos adversos sobre a estrutura testicular e epididimária, nos níveis séricos de testosterona assim como nas células participantes da manutenção espermatogênica em ratos.
Flaxseed is considered a functional food, exercising several beneficial effects on health. However, due to its high content of phytoestrogens, they are capable of influencing the hormonal metabolism and interfere with gonadal biomorphology. The aim of this study is to evaluate, using computerized histomorphometry, seminiferous and epididymal tubules, considering the different regions of the epididymis (head, body and tail) in rats submitted to a prolonged diet of flaxseed. Young adult male Wistar rats (n=20) were used and divided into two groups during their lactation period: Control Group (CG) fed casein-based meals and the Flax Group (FG) with addition of 25% flaxseed meal. At the end of 250 days of continuous ingestion, the animals were euthanized, their testicles/epididymis removed and fixated in buffered formalin solution. The samples were submitted to routine histological paraffin techniques and stained with HE. Immunostaining was performed using an anti-vimentin antibody for Sertoli cell identification. For morphometry, images of the slides scanned and analyzed using the ImageJ software to obtain the data of height epithelial, luminal diameter, tubule diameter and tubule area. Statistical analyzes were performed using GraphPad Stat program through the Student t test. In hormone evaluation, the GL had higher concentrations of serum estrogen (p = 0.001), but no significant change in the concentration of testosterone. Regarding the morphometric parametrers of seminiferous tubules and epididymal regions, no significant differences among the analyzed groups were found. In the Sertoli cell quantification, no significant differences in the FG (p=0.98) were found either. The results showed that the continuous and prolonged intake of 25% flaxseed meals from gestation to 250 days of age does not lead to adverse effects on testicular and epididymal structure, in testosterone serum levels as well as as cells that participate in the spermatogenic maintenance mice.
