ATIVIDADE IN VITRO DE Carapa guianensis Aublet, Copaifera offinalis Jacquin Linnaeus e Psidium guajava Linnaeus SOBRE Haemonchus Contortus

Resumo

O parasitismo por Haemonchus contortus constitui-se num dos fatores limitantes para a produção de caprinos e ovinos em todo o mundo, principalmente nas regiões tropicais, onde os prejuízos econômicos são mais acentuados. Os estudos de plantas medicinais estão sendo realizados para encontrar um fitoterápico efetivo no controle das populações deste nematóide e assim minimizar os prejuízos na saúde e produção de seus hospedeiros. Desta forma, avaliouse a atividade anti-helmíntica in vitro do óleo fixo da semente de Carapa guianensis (andiroba) e Copaifera officinalis (copaíba) e do óleo essencial de Psidium guajava (goiaba), sobre ovos e larvas de H. contortus de caprinos. Foram utilizados testes in vitro do óleo das três plantas, avaliando-se a inibição da eclodibilidade dos ovos e do desembainhamento larvar. Para a primeira avaliação, uma alíquota de suspensão de ovos de H. contortus foi incubada com os óleos e mantida em estufa BOD a 27 ° C. por 48 horas. O óleo de C. guianensis foi o que apresentou melhor resultado, com maior média de inibição de eclodibilidade larvar (54,60%) obtida na maior concentração (20 mg mL-1), que inclusive diferiu das demais concentrações (p0,05). Nesta concentração, os óleos da C. officinalis e da P. guajava apresentaram um percentual de apenas 6,69 e 10,33% dos ovos. Para a C. guianensis, a CE50 encontrada foi de 17,92 (16,92 - 18,93) mg mL-1. Na segunda avaliação, alíquotas de larvas de terceiro estágio foram incubadas com o óleo das plantas e solução de hipoclorito em placas estéreis, observando-se os índices de desembainhamento em intervalos de 20 minutos, finalizando com 60 minutos. Durante as quatro observações (0, 20, 40 e 60 minutos) os percentuais de larvas de H. contortus sem bainhas encontrados nas placas tratadas com os óleos, independente da concentração (0,6 mg mL-1 e 1,2 mg mL-1), não diferiram daqueles percentuais nos respectivos grupos controles (p>0,05), cujos percentuais obtidos no final do experimento (60 minutos) foram 98,09 (C. guianensis e C. guianensis) e 93,86 (P. guajava). Adicionalmente, o óleo de C. guianensis foi submetido a análise por cromatografia gasosa, para caracterizar os ácidos graxos presentes na composição, sendo isolados os ácidos palmitoléico, araquídico e behênico que não estão descritos na literatura. In vitro, o óleo fixo de C. guianensis e C.officinalis e o óleo essencial de P. guajava não apresentaram índices de inibição da eclodibilidade de ovos e do desembainhamento larvar de H. contortus de caprinos.
The parasitism by Haemonchus contortus constitutes a limiting factor for the production of goats and sheep in the world, especially in tropical regions, where economic losses are more pronounced. The studies of medicinal plants are being conducted to find an effective herbal medicine in controlling populations of this nematode and so minimize the losses in health and production of their hosts. Thus, we evaluated the anthelmintic activity in vitro of fixed oil of Carapa guianensis seed (andiroba) and Copaifera officinalis (copaiba) and the essential oil of Psidium guajava (goiaba), on eggs and larvae of H. contortus goats. The three plants Oil In vitro tests were used, evaluating the inhibition hatchability of eggs and larval unsheathing. For the first assessment, H. contortus egg suspension rate was incubated with oils and maintained in an environmental chamber at 27 ° C. for 48 hours. C. guianensis oil showed the best result, with higher mean inhibition of larval hatchability (54,60 %) obtained in the highest concentration (20 mg mL -1 ), which also differ from the other concentrations (p<0,05). At this concentration, the oils of C. officinalis and P. guajava showed a percentage of only 6,69 and 10,33 % of the eggs. To C. guianensis, EC50 was found to be 17,92 (16,92 - 18,93) mg mL-1 . In the second evaluation of third stage larvae were incubated with aliquots of oil plants and hypochlorite solution into sterile plates observing the unsheathing indices at 20 minute intervals ending with 60 minutes. During the four points (0, 20, 40 and 60 minutes) the percentage of H. contortus larvae without sheaths found in the plates treated with oils, independent of concentration (0,6 mg mL -1 and 1,2 mg mL- 1) did not differ from those percentages in the respective control groups (p> 0,05), the percentage obtained at the end of the experiment (60 minutes) was 98,09 (C. guianensis and C. guianensis) and 93,86 (P. guajava). Additionally, C. guianensis oil was subjected to gas chromatographic analysis to characterize the fatty acids present in the composition, and the isolated palmitoleic acid, arachidic and behenic that are not described in the literature. In vitro, fixed oil of C. guianensis and C.officinalis and the essential oil of P. guajava indices showed no inhibition of hatchability of eggs and larvae of H. contortus unsheathing goats.
