EFEITOS SUBLETAIS DO INSETICIDA BIOLÓGICO XENTARI, SOBRE A TOXICIDADE RENAL E HEPÁTICA EM RATAS

Resumo

No Brasil o número de trabalhadores rurais expostos a inseticidas é bastante expressivo, o que concorre para intoxicações agudas e doenças crônicas. Assim, testou-se a hipótese se a administração do inseticida biológico Xentari® WG (B. thuringiensis subsp. Aizawai), em concentrações subletais, pode interferir na função hepática e renal em ratas. Utilizaram-se 40 ratas albinas, Rattus norvegicus albinus, distribuídas em quatro grupos constituídos por cinco animais: I: Ratas que receberam placebo (Controle); II, III e IV, que receberam 1,0; 10 e 20 mg de Xentari®/100g, respectivamente. Os animais foram pesados aos 20 e 30 dias de administração do inseticida. Foram realizadas dosagens bioquímicas de ALT, AST, HDL, LDL, VLDL, albumina, creatinina e uréia. Para as análises histopatológicas e histoquímica utilizou-se a técnica H.E e P.A.S., respectivamente. Na morfométria do fígado mensurou-se os hepatócitos, capilares sinusoides, ducto bilífero, espaço porta-hepático, veia hepática e veia centro-lobular e no rim mensurou-se diâmetro e volume do glomérulo e da cápsula de Bowman. A eutanásia dos animais ocorreu logo após o término da administração do inseticida, que durou o período de 20 ou 30 dias após aplicação do inseticida. Os resultados mostraram que o Xentari® WG, promoveu uma redução no peso dos animais, além de uma alteração nos níveis de triglicerídeos aos 20 dias, colesterol total aos 30 dias, HDL, LDL e VLDL aos 20 dias. Já a ALT foi alterada com 20 e 30 dias, enquanto que o AST apenas aos 20 dias, também promoveu alterações nos níveis de ureia e albumina aos 30 dias de tratamento. A histopatologia do fígado e do rim mostrou tumefação e vacuolização celular e baixa reação ao P.A.S no fígado, a análise morfométrica do fígado demonstrou um aumento dos hepatócitos e capilares sinusoides em relação ao espaço porta-hepático, veia hepática e centro-lobular, e uma diminuição do volume e diâmetro glomerular, caracterizando atrofia glomerular e glomérulonefrite membranosa no rim para ambos os períodos analisados. Conclui-se que o Xentari® em doses subletais, interfere na função hepática e renal de ratas, sendo mais expressivo no período de aplicação agudo (20 dias). Assim, sugere-se que sejam realizadas mais pesquisas que revelem os efeitos crônicos da exposição a este inseticida biológico.
In Brazil the number of rural workers exposed to pesticides is very expressive, which contributes to acute poisoning and chronic diseases. Thus, we tested the hypothesis that the administration of the biological insecticidal Xentari® WG (B. thuringiensis subsp. Aizawai), at sublethal concentrations may interfere with the hepatic and renal function in rats. They were used 40 albino rats, Rattus norvegicus albinos, divided into four groups consisting of five animals: I: rats who received placebo (control); II, III and IV, which received 1.0; 10 and 20 mg of Xentari® / 100g, respectively. Animals were weighed after 20 and 30 days of administration of the insecticide. biochemical measurements were performed in ALT, AST, HDL, LDL, VLDL, albumin, creatinine and urea. For the histopathologic and histochemical analysis was used to H.E and P.A.S. technique, respectively. Morphometry In the liver measured to hepatocytes, sinusoidal capillaries, bilífero duct, the portal-hepatic space, hepatic vein and central lobular and kidney is measured diameter and volume of the glomerulus and Bowman's capsule. Euthanasia of animals took place shortly after the pesticide administration, which lasted a period of 20 or 30 days after insecticide application. The results showed that the Xentari® WG, promoted a reduction in the weight of animals, as well as a change in triglyceride levels at 20 days, 30 days total cholesterol, HDL, LDL and VLDL at 20 days. Since the ALT was changed to 20 and 30 days, whereas only 20 days AST also promoted changes in urea and albumin levels after 30 days of treatment. The histopathology of liver and kidney showed swelling and cellular vacuolization and low reaction to PAS in the liver, the morphometric analysis of the liver showed an increase in hepatocytes and sinusoidal capillaries relative to the portal-hepatic space, hepatic vein and central lobular, and decreased volume and glomerular diameter, featuring glomerular atrophy and membranous glomerulonephritis kidney for both periods analyzed. It follows that the Xentari® at sublethal doses, interfere with hepatic and renal function of rats, and stronger in the Acute application period (20 days). Thus, it is suggested that further research be conducted to reveal the chronic effects of exposure to this biological insecticide.
