DETECÇÃO E VARIABILIDADE GENÉTICA DE Mycoplasma hyopneumoniae EM AMOSTRAS DE PULMÃO DE SUÍNOS DO ESTADO DE MINAS GERAIS, BRASIL

Resumo

A suinocultura é um setor agropecuário de grande importância para a economia brasileira. Os diversos sistemas de produção enfrentam grandes perdas econômicas causadas pelas doenças respiratórias, sendo a Pneumonia Enzoótica Suína (PES) uma das principais doenças respiratórias. A PES é causada pela bactéria Mycoplasma hyopneumoniae (Mhyo) e a doença se caracteriza por uma tosse seca e crônica que afeta suínos de qualquer faixa etária. Sabemos que Mhyo possui grande diversidade genômica e proteômica, além de diferenças na virulência entre as cinco cepas conhecidas. O objetivo deste trabalho foi avaliar a diversidade genética de Mhyo baseado em genes que codificam proteínas antigênicas. Foram avaliadas 148 amostras de pulmão de suínos provenientes de duas regiões do estado de Minas Gerais, 133 (89.9 %) amostras foram positivas no PCR de identificação de Mhyo. As amostras positivas foram submetidas a PCRs dos genes selecionados: mhp 0043, mhp 0660, mhp 0099, mhp 0360, mhp 0107, mhp 0067, mhp 0272, mhp 0487 e mhp 0580. Além disso, algumas amostras foram selecionadas para realizar o sequenciamento parcial dos genes específicos. Os resultados mostraram variações na frequência de distribuição dos nove genes em amostras de campo. Os genes mhp 0043, mhp 0660, mhp 0107, mhp 0067, mhp 0487 e mhp 0580 apresentaram frequência maior que 90%. Analisando o resultado do sequenciamento parcial de cada gene, observamos que as sequencias destes nove genes são bem conservadas nas cepas analisadas. Foi possível observar que Mhyo possui grande diversidade genética, no que se refere ausência ou presença destes genes. Com base nestes resultados, o estudo sugere que os genes mhp 0043, mhp 0660, mhp 0107, mhp 0067, mhp 0487 e mhp 0580 devem ser investigados como candidatos a produção de vacinas.
Pig farming is an agricultural sector of great importance to Brazilian economy. The systems of production pigs face great economic losses resulting from respiratory diseases, Swine Enzootic Pneumonia (PES) is the major respiratory diseases.PES is caused by Mycoplasma hyopneumoniae (Mhyo) and the disease is characterized by a dry and chronic cough that affects pigs of any age. It is known that Mhyo has genomic and proteomic diversity, as well as differences in virulence among the five known strains. The objective of this work was to evaluate the genetic diversity of Mhyo based on genes encoding antigenic proteins. A total of 148 pig lung samples from two regions of the state of Minas Gerais were evaluated. 133 (89.9 %) samples were positive in PCR of Mhyo. Positive samples were submitted to PCR of nine genes selected: mhp 0043, mhp 0660, mhp 0099, mhp 0360, mhp 0107, mhp 0107, mhp 0067, mhp 0272, mhp 0487 and mhp 0580. In addition, some samples were selected to partial sequencing of specific genes. These results showed variations in the distribution frequency in field samples of nine genes. Genes mhp 0043, mhp 0660, mhp 0107, mhp 0067, mhp 0487 and mhp 0580 showed a frequency greater than 90%. Analyzing the result of the partial sequencing of each gene, we observed that the sequences of these nine genes are well conserved in the analyzed field strains. We observed that Mhyo possesses great genetic diversity, regarding the absence or presence of these genes. Based on these results, our study suggests that the genes mhp 0043, mhp 0660, mhp 0107, mhp 0067, mhp 0487 and mhp 0580 should be investigated as candidates to produce vaccines, however, future studies that determine the antigenicity of the proteins expressed by these genes should be realized.
