Efeito da quercetina na maturação in vitro de oócitos caprinos

Resumo

O objetivo deste estudo foi avaliar o efeito da quercetina como antioxidante alternativo à cisteamina na maturação in vitro. Ovários caprinos foram transportados do matadouro local para laboratório em solução salina a 0,9%, a 30°C, até três horas após o abate. Após a colheita, os oócitos foram avaliados e divididos em três grupos: Grupo CIS, onde os oócitos foram imersos em meio MIV base: TCM-199, suplementado com EGF (10 g/mL), FSH/LH (10 L/mL), soro de ovelha em estro (100 L/mL) e cisteamina (10 L/mL); nos Grupos Q4 ou Q8, os oócitos foram imersos em meio base isento de cisteamina, suplementado com 4 M ou 8 M de quercetina, respectivamente. A MIV dos oócitos foi realizada a 38,5°C em atmosfera umidificada de 5% de CO2 em ar, durante 24 horas. Os grupos CIS e Q4 apresentaram a mesma porcentagem de células cumulus expandidas, mas o grupo Q8 foi significativamente menor que os demais grupos (P<0,05). A taxa de retração de oócitos no grupo Q8 foi maior (P<0,05) do que os tratamentos adicionais. Em relação à expansão das células do cumulus, o tratamento com 8 M de quercetina apresentou menor proporção de oócitos expandidos com grau 1 (Total) do que os tratamentos adicionais (P<0,05). A percentagem de oócitos MII foi maior no grupo Q4 do que no grupo CIS (P<0,05), mas os grupos CIS e Q8 foram semelhantes. A taxa de apoptose foi maior no grupo CIS do que nos grupos adicionais (P<0,05). Os oócitos dos grupos CIS e Q4 apresentaram os mesmos níveis de espécies reativas de oxigênio (ERO) e glutationa (GSH). Além disso, os oócitos maturados com 4 M de quercetina apresentaram maior atividade mitocondrial do que os oócitos maturados nos grupos CIS e Q8 (P<0,05). Em conclusão, 4 M de quercetina pode ser utilizada como alternativa à cisteamina na maturação in vitro de oócitos de caprinos, pois, a mesma resultou em taxas de maturação oocitária de caprinos maiores que àquelas obtidas com cisteamina, mantendo constantes os níveis de expansão das células do cumulus, glutationa, ERO, além, de elevar a atividade mitocondrial. Contudo, a concentração de 8 M, levou à redução dos níveis de ERO, GSH e atividade mitocondrial oocitária, demonstrando menor viabilidade celular.
The objective of this study was to evaluate the effect of quercetin as an alternative antioxidant to cysteamine during in vitro maturation. Ovary goats were transported from the local slaughterhouse to the laboratory in 0.9% saline at 30 ° C until three hours after slaughter. After collection, the oocytes were evaluated and divided into three groups: CIS Group, where the oocytes were immersed in MIV medium: TCM-199, supplemented with EGF (10 g / mL), FSH / LH (10 L / mL), Estrus sheep serum (100 L / mL) and cysteamine (10 L / mL); In Groups Q4 or Q8, oocytes were immersed in cysteamine-free base medium, supplemented with 4 M or 8 M quercetin, respectively. The CIS and Q4 groups presented the same percentage of expanded cumulus cells, but the Q8 group was significantly lower than the other groups (P <0.05). The oocyte retraction rate in the Q8 group was higher (P <0.05) than the additional treatments. In relation to the expansion of cumulus cells, treatment with 8 M quercetin presented a lower proportion of expanded oocytes with grade 1 (Total) than the additional treatments (P <0.05). The percentage of MII oocytes was higher in the Q4 group than in the CIS group (P <0.05), but the CIS and Q8 groups were similar. The rate of apoptosis was higher in the CIS group than in the additional groups (P <0.05). Oocytes from the CIS and Q4 groups showed the same levels of reactive oxygen species (ROS) and glutathione (GSH). In addition, oocytes matured with 4 M quercetin showed higher mitochondrial activity than mature oocytes in the CIS and Q8 groups (P <0.05). In conclusion, 4 M of quercetin can be used as an alternative to cysteamine in the in vitro maturation of goat oocytes, as it resulted in rates of oocyte maturation of goats larger than those obtained with cysteamine, maintaining constant levels of cell expansion Of the cumulus, glutathione, ERO, in addition to elevating mitochondrial activity. However, the concentration of 8 M led to the reduction of ROS levels, GSH and oocyte mitochondrial activity, demonstrating lower cell viability.
