DETERMINAÇÃO DO INTERVALO DE TEMPO IDEAL PARA ESTABELECIMENTO DE CULTURA DE CÉLULAS TRONCO MESENQUIMAIS DE CADELAS

Resumo

As células tronco mesenquimais (CTM) são células tronco adultas, também conhecidas como células tronco somáticas mesenquimal. Estas células encontramse nas regiões perivasculares de todos os tecidos adultos, com a medula óssea, tecido adiposo, muscular, periósteo e órgãos parenquimatosos. Na ocasião dos primeiros relatos deste tipo celular, a fonte que era mais utilizada para isolamento e cultivo destas células era a medula óssea, entretanto, após o ano de 2010, muitas pesquisas vem difundindo o uso de células oriundas do tecido adiposo, defendendo a facilidade de coleta do mesmo e abundância no organismo, a forma de coleta pouco invasiva, com pouco desconforto para o paciente, humano ou animal e quantidade de células mononucleadas presente no tecido. Recentemente o grupo de pesquisa em terapia celular da Universidade Vila Velha, descreveu, de forma pioneira, a utilização do ligamento largo do útero como uma potencial fonte de células tronco mesenquimais. O interesse terapêutico pelas células tronco, tanto para pesquisa quanto para uso clínico, fez surgir novos grupos de pesquisas vinculados à universidades e vários laboratórios de cultivo na rede privada, que disponibilizam células tronco mesenquimais autógenas e/ou alógenas para o tratamento de várias afecções animais. Para tal, observa-se por vezes, que o tecido é coletado do paciente, enviado ao laboratório de cultivo, processado e posteriormente encaminhado ao responsável pela terapia celular no paciente. Todavia, não encontramos referências técnico-científicas que nos descrevam qual seria o tempo ideal entre a coleta do tecido e o início do estabelecimento da cultura primária. Com esse intuito, o objetivo do presente estudo foi avaliar se existe diferença na quantidade de células e na qualidade das culturas de tecidos coletados para o estabelecimento de culturas de células tronco mesenquimais, cultivados em diferentes tempos. Utilizou-se dez cadelas hígidas, submetidas à ovariohisterectomia eletiva, das quais foram coletados fragmentos de tecido adiposo do subcutâneo e ligamento largo do útero, cada fragmento foi dividido em três amostras de 1,0 grama de tecido, acondicionado em tubos contendo 10,0ml de meio de cultivo, identificados e processados no laboratório de cultivo celular , em três momentos distintos: imediatamente após a coleta, 24 horas e 48 horas após a coleta. Em todos os momentos de processamento encontrou-se quantidade inicial de células mononucleares maior que 107 cel/ml. As primeiras células aderentes foram visualizadas a partir de 12 horas e a taxa de expansão celular e confluência nas garrafas foi maior nos tecidos processados imediatamente após a coleta e com 24 horas. Nesse estudo foi possível perceber que a variável tempo não interferiu na concentração de células encontradas no momento do processamento, entretanto, que houve um melhor estabelecimento e expansão nas culturas processadas imediatamente após a coleta e com 24 horas.
Mesenchymal stem cells (MSCs) are adult stem cells, also known as mesenchymal somatic stem cells. Recently, the cell therapy research group at Vila Velha University has pioneered the use of the broad ligament of the uterus as a potential source of mesenchymal stem cells. The therapeutic interest in stem cells, both for research and for clinical use, has led to the emergence of new research groups linked to universities and several laboratories in the private network that provide autologous and / or allogeneic mesenchymal stem cells for the treatment of various animal diseases . For this, it is sometimes observed that the tissue is collected from the patient, sent to the culture laboratory, processed and then referred to the person responsible for the cell therapy in the patient. However, we do not find technical-scientific references that describe the ideal time between tissue collection and the beginning of the establishment of the primary culture. The aim of the present study was to evaluate if there is a difference in the number of cells and in the quality of tissue cultures collected for the establishment of cultures of mesenchymal stem cells, grown at different times. Ten healthy bitches were submitted to elective ovariohysterectomy, from which fragments of subcutaneous adipose tissue and broad ligament of the uterus were collected, each fragment was divided into three 1.0 gram samples of tissue, packed in tubes containing 10.0 ml Of culture medium, identified and processed in the cell culture laboratory, in three distinct moments: immediately after collection, 24 hours and 48 hours after collection. At all times of processing an initial amount of mononuclear cells greater than 107 cells / ml was found. The first adherent cells were visualized after 12 hours and the rate of cell expansion and confluence in the bottles was higher in the processed tissues immediately after collection and at 24 hours. In this study it was possible to notice that the time variable did not interfere in the concentration of cells found at the moment of processing, however, that there was a better establishment and expansion in the cultures processed immediately after the collection and with 24 hours.
