Estudo da atividade cicatrizante da Sphagneticola trilobata em ferimentos de pele em ratos wistar: análises anatomohistopatológicas

Resumo

A Sphagneticola trilobata (L.) Pruski (Vedélia) é uma planta muito utilizada na medicina popular. Ela é uma herbácea pertencente à família das Asteraceae com ação antioxidante, anti-inflamatória, antimicrobiana e cicatrizante, características essas que impulsionaram a realização dessa pesquisa. Este estudo teve como objetivo testar a atividade antimicrobiana e cicatrizante do extrato hidroalcoólico bruto das folhas de Sphagneticola trilobata. Para a produção do extrato hidroalcoólico bruto, o material vegetal (folhas) foi desidratado em estufa a 40 ºC, posteriormente triturado e macerado em etanol 70% por 72 horas. A proporção extrato/planta empregado foi de 2 ml de solvente, etanol 70%, para 1 g do material vegetal. A solução extrativa foi filtrada e posteriormente concentrada em evaporador rotatório sobre pressão reduzida, a 45 ºC, resultando no extrato hidroalcoólico bruto com uma concentração de 140 mg/ml. Na determinação da quantidade de fenóis e flavonoides totais, foram usadas as metodologias de espectrofotometria e método calorimétrico por complexação metálica, respectivamente, e a triagem fitoquímica foi realizada por meio da cromatografia em camada delgada analítica. Para avaliar a atividade antimicrobiana foram testadas culturas de Staphylococcus spp., Escherichia coli, Serratia marcescens, Enterococcus faecalis, Pseudomonas aeruginosa, Salmonella enterica, Salmonella Typhimurium, Klebsiella pneumoniae isoladas de pele humana e culturas de Staphylococcus spp. isoladas de pele de cães pelo o método de microdiluição em caldo. E para o estudo da atividade cicatrizante foram utilizados 60 ratos, nos quais foram realizadas quatro feridas cutâneas. Eles foram distribuídos em três grupos experimentais: grupo tratado (GT), grupo controle (GC) e grupo controle absoluto (GCA). As feridas foram avaliadas em três, sete, 14 e 21 dias de pós operatório (PO) quanto à contração, avaliação macroscópica, morfohistológica e morfohistométrica. Como resultados foram verificadas na análise fitoquímica a presença de fenóis e flavonoides totais nas concentrações de 21,7 mg e 0,23 mg por grama de extrato, respectivamente, e na triagem fitoquímica foi verificada a presença de derivados antracênicos, mono, sesqui e diterpenos. Na análise microbiológica, o extrato hidroalcoólico das folhas de Sphagneticola trilobata apresentou atividade antimicrobiana frente às culturas de Staphylococcus spp., Escherichia coli, Serratia marcescens e Enterococcus faecalis e as culturas de Pseudomonas aeruginosa, Salmonella enterica, Salmonella typhimurium, Klebsiella pneumoniae apresentaram resistência ao extrato. Todas as culturas de Staphylococcus spp. isoladas de pele de cães foram sensíveis ao extrato. Com relação a atividade cicatrizante, na análise macroscópica foi observada menor área de cicatrização aos 21 dias PO (p<0,05) no GT. Na avaliação morfohistológica o GT apresentou maior preenchimento do tecido de granulação pelas fibras colágenas e menor inflamação. Na morfohistometria, o comprimento da língua epitelial não teve diferença entre os grupos (p<0,05) e na contagem dos vasos sanguíneos a média dos valores foi maior no GT com três e sete dias PO (p<0,05). Na densidade das fibras colágenas verificou-se que o GT apresentou as maiores médias em todos os períodos (p<0,05). Conclui-se que o extrato hidroalcoólico bruto das folhas de Sphagneticola trilobata possui compostos secundários que favorecem a atividade antimicrobiana frente a bactérias isoladas de pele de diferentes origens e que o creme à base do extrato contribui positivamente no processo de cicatrização das feridas em pele de ratos.
Sphagneticola trilobata (L.) Pruski (Vedelia) is a plant widely used in folk medicine. It is an herbaceous belonging to the family of the Asteraceae with antioxidant, anti-inflammatory, antimicrobial and cicatrizante action, characteristics that propelled the realization of this research. This study aimed to test the antimicrobial and cicatrizant activity of the crude hydroalcoholic extract of the leaves of Sphagneticola trilobata. For the production of the crude hydroalcoholic extract, the plant material (leaves) was dehydrated in an oven at 40 ºC, later crushed and macerated in 70% ethanol for 72 hours. The extract/plant ratio used was 2 ml solvent, 70% ethanol, to 1 g of the plant material. The extractive solution was filtered and then concentrated on rotary evaporator under reduced pressure at 45 °C, resulting in the crude hydroalcohol extract having a concentration of 140 mg/ml. In the determination of the amount of total phenols and flavonoids, spectrophotometry and calorimetric methodologies were used by metal complexation, respectively, and phytochemical screening was performed by means of analytical thin layer chromatography. To evaluate the antimicrobial activity cultures of Staphylococcus spp., Escherichia coli, Serratia marcescens, Enterococcus faecalis, Pseudomonas aeruginosa, Salmonella enterica, Salmonella Typhimurium, Klebsiella pneumoniae isolated from human skin and cultures of Staphylococcus spp. isolated from dogs skin by the broth microdilution method. For the study of cicatrizant activity, 60 rats were used, in which four cutaneous wounds were performed. They were divided into three experimental groups: treated group (GT), control group (CG) and absolute control group (GCA). Wounds were evaluated at three, seven, 14 and 21 postoperative days (PO) for contraction, macroscopic, morphohistological and morphohistometric evaluation. Phytochemical analysis showed the presence of total phenolics and flavonoids in the concentrations of 21.7 mg and 0.23 mg per gram of extract, respectively, and in the phytochemical evaluation the presence of anthracenic, mono, sesqui and diterpene derivatives. In the microbiological analysis, the hydroalcoholic extract of the leaves of Sphagneticola trilobata presented antimicrobial activity against the cultures of Staphylococcus spp., Escherichia coli, Serratia marcescens and Enterococcus faecalis, and the cultures of Pseudomonas aeruginosa, Salmonella enterica, Salmonella Typhimurium and Klebsiella pneumoniae showed resistance to the extract. All cultures of Staphylococcus spp. isolated from dogs' skin were sensitive to the extract. Regarding the cicatrizing activity, in the macroscopic analysis a smaller area of healing was observed at 21 days PO (p<0.05) in GT. In the morphohistological evaluation, the GT showed greater filling of the granulation tissue by the collagen fibers and less inflammation. In morphohistometry, the length of the epithelial tongue had no difference between the groups (p<0.05) and in the blood vessel count the mean values were higher in the GT with three and seven days PO (p<0.05). In the density of the collagen fibers, it was found that the GT presented the highest averages in all periods (p<0.05). It is concluded that the crude hydroalcoholic extract of the leaves of Sphagneticola trilobata has secondary compounds that favor the antimicrobial activity against isolated bacteria of skin of different origins and that the cream based on the extract contributes positively in the process of cicatrization of wounds in rats skin.
