Caracterização microbiológica da resina de Amburana cearensis como potencial para terapia alternativa da mastite caprina por Staphylococcus aureus

Resumo

A mastite é uma das afecções de maior importância na caprinocultura leiteira. Esta afeta a qualidade e a quantidade de leite produzido, acarretando em grandes prejuízos econômicos. Na maioria das vezes, a mastite é causada por bactérias, com destaque para Staphylococcus aureus. O uso da fitoterapia no tratamento oferece como vantagens a possível redução nas reações adversas durante o tratamento, bem como diminuição na pressão de seleção sobre cepas resistentes associadas ao uso excessivo dos antibióticos. Desse modo, o objetivo do presente estudo foi a avaliação in vitro do potencial da resina de Amburana cearensis para o tratamento da mastite caprina causada por Staphylococcus aureus. A atividade antimicrobiana da resina foi determinada por meio do teste de microdiulição em caldo para determinação da Concentração Inibitória Mínima (CIM) e Concentração Bactericida Mínima (CBM). A atividade anti-biofilme foi realizada pelo teste de aderência em microplacas. Para avaliação da sinergia, realizou-se o método Checkerboard. A inibição da bomba de efluxo foi avaliada por redução da CIM do brometo de etídeo e para avaliar a interferência da resina na adesão e invasão dos isolados foram utilizadas células epiteliais mamária bovina (MAC-T). O efeito citotóxico da resina foi medido através dos ensaios do MTT sobre células MAC-T. Foi verificado que a resina apresentou atividade antibacteriana com valores variando entre 195,3 g/mL a 1.562,5 g/mL. Da mesma forma a resina interferiu com a formação de biofilme em cinco isolados, bem como quando esta estrutura já estava consolidada (%) em três isolados. O sinergismo da resina com a oxacilina no isolado MRSA, reduziu em quatro vezes a concentração bactericida do antibiótico, porém mostrou-se apenas aditivo quando combinado com a ampicilina. A resina não apresentou efeito como inibidor de bomba de efluxo, porém observou-se que a ação antimicrobiana da resina não foi influenciada pela presença desta estrutura em S. aureus. Foi observada uma redução significativa (p<0,01) na adesão e invasão dos isolados testados frente as células MAC-T. Na avaliação da citotoxicidade observou-se que houve uma pequena redução da viabilidade celular, sendo 195,3 g/mL a melhor concentração da CIM da resina, reduzindo apenas 1, 4% da viabilidade da MAC-T. Com isso, o presente trabalho mostrou que a resina representa uma possível alternativa para utilização como futuro medicamento fitoterápico com ação antimicrobiana.
Mastitis is one of the most important diseases in dairy goat, which affects the quality and quantity of milk produced, leading to great economic losses. Most of the time, mastitis is caused by bacteria, especially Staphylococcus aureus. The use of phytotherapy in the treatment offers as advantages the possible reduction in the adverse reactions during the treatment, as well as decrease in the pressure of selection on resistant strains associated with the excessive use of the antibiotics. Thus, the objective of the present study was the evaluation in vitro of the potential of Amburana cearensis resin for the treatment of caprine mastitis caused by Staphylococcus aureus. The antimicrobial activity of the resin was determined by the determination of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (CBM), the anti-biofilm activity was performed by the microplate adhesion test. To evaluate the synergy, the Checkerboard method was used. Inhibition of the efflux pump was evaluated by reduction of the MIC, to evaluate resin interference at adhesion and invasion of the isolates using bovine mammary epithelial cells (MAC-T), the cytotoxic effect of the resin was measured by the MTT assays on MAC-T cells. It was verified that the resin presented antibacterial activity with values ranging from 195.3 g/mL to 1,562.5 g/mL and anti-biofilm action against the isolates interfering with the formation of the biofilm, as well as when it was already consolidated. The synergism of the resin with oxacillin in the MRSA isolate reduced the bactericidal concentration of the antibiotic by four times, thus reversing the resistance phenotype of the isolate, but was only additive when combined with ampicillin. The resin showed no efflux pump inhibitor effect, however, it was observed that the antimicrobial action of the resin was not influenced by the presence of this structure in S. aureus. There was a significant (p <0.01) reduction in adhesion and invasion of the isolates tested against MAC-T cells. In the evaluation of cytotoxicity, it was observed that there was a small reduction in cell viability, with 195.3 g/mL being the best concentration of the resin MIC, reducing only 1.4% of viability of MAC-T. With this, the present work showed that the resin represents a possible alternative for use as a future herbal medicine with antimicrobial action.
