Transfecção de anticorpos como terapia anti-viral para a raiva

Resumo

A raiva é uma zoonose conhecida desde a Mesopotâmia, temida por milênios em todo omundo por seu rápido e progressivo número de casos fatais, e agora é endêmicaem países em desenvolvimento, com uma estimativa de 6.000.000 de mortes humanas por ano. O pequeno número de pacientes que sobreviveram a raiva ainda é motivo de debate, em meio a tentativas de estabelecer um tratamento eficaz para pacientes humanos. O objetivo deste estudo foi avaliar a capacidade de tranfecção de anticorpos F (ab ') anti-RABV com uso de dois agentes de transfecção catiônicos (Pulsin / Bioporter) in vivo, bem como avaliar a eficácia deste método como terapia antiviral. Camundongos foram inoculados pela via intracerebral com 30 l de um isolado de RABV (AgV2), contendo 10 LD50%. Após 48h, dois tratamentos foram realizados: Para o Tratamento-1, 180 l de F (ab ') anti-RABV foram combinados com 360 l de Hepes 20 mM pH 7,4 e adicionados 60 l de Pulsin ® (Polyplus). Para otratamento 2, o reagente Bioporter ® foi ressuspendido com a F (ab ') anti-RABVna concentração final de 250 g / ml. Os grupos controles foram tratados com os agentes de transfecção sem anticorpos. As soluções foram então inoculadas por via intracerebral em volume de (30 l) em cada um dos quinze animais. No tratamento-1, comparando os grupos tratado e controle, não houve diferença significativa (p = 1). No tratamento-2, a taxa de sobrevivência foi de 70% para o grupo tratado, enquanto a mortalidade nos animais do grupo controle foi de 90% (p <0,0198). De modo a diferenciar entre o efeito antiviral pela transfecção da F (ab ') anti-RABV e um possível efeito antiviral do próprio agente de transfecção Bioporter ®, os camundongos foram sujeitos a infecção com 30 l do isolado RABV acima mencionado, contendo 10 LD50% . Após 48 horas, 30 L de reagente Bioporter ressuspenso em Hepes 20 mM, pH 7, foram inoculados pela via intracerebral em 10 camundongos, enquanto para o grupo controle (n = 10), foi utilizado somente Hepes 20 mM, pH 7,4. Houve uma taxa de sobrevivência de 50 e 20%, para os grupos Bioporter e controle, respectivamente, sem diferença significativa (p = 0,4949), mostrando que o efeito antiviral observado no primeiro experimento foi devido ao anticorpo transfectado. A inibição da replicação de RABV pode ser obtida pela técnica de transfecção de anticorpos, uma ferramenta valiosa a ser adicionada como uma terapia antiviral para o tratamento da raiva em humanos.
Rabies is a zoonosis known since Mesopotamia, which has been feared for millennia throughout the world for its rapid and progressive number of fatal cases, and now itss endemic in developing countries, with an estimated 6,000,000 human deaths per year. The small number of patients who survived the disease is still a subject of debate, amid attempts to establish an effective treatment for human patients. The objective of this study was to evaluate the transfection capacity of anti-RABV F (ab ') antibodies with the use of two cationic reagents (Pulsin / Bioporter) in vivo, as well as to evaluate the effectiveness of this method as an antiviral therapy. Mice were inoculated by the intracerebral route with 30 l of a dog RABV isolate (AgV2), containing 10 LD50%. After 48h, two treatments were performed: For Treatment 1, 180 l of anti-RABV F (ab ') were combined with 360 l of 20 mM Hepes pH 7.4 and added 60 l of Pulsin ® (Polyplus). For Treatment-2, the Bioporter ® reagent was resuspended with the anti-RABV F (ab ') in final concentration of 250 g / ml. Control groups were mock-treated with the transfection agents devoid of antibobies. The solutions were then inoculated (30 l) by intracerebral route into fifteen animals each. In treatment-1, comparing the treated and control groups, there was no significant difference (p = 1). In treatment-2, survival rate was 70% for the treated group, while mortality in animals of the control group was 90% ((p<0,0198).). In order to differentiate between the antiviral effect of transfected anti-RABV F (ab ') and a possible antiviral effect of the Bioporter ® transfection agent itself, mice were subjected to infection with 30 l of the abovementioned RABV isolate, containing 10 LD50%. After 48hr, 30 L Bioporter reagent resuspended in 20 mM Hepes,pH 7 were inoculated by the intracerebral route in 10 mice, while, for the control group (n=10), 20 mM Hepes, pH 7.4 was used instead. There was a 50 and 20% survival rate for the Bioporter and control groups, respectively, with no significant difference (p = 0.4949), showing that the antiviral effect observed in the first experiment was due to the transfected antibody. Inhibition of RABV replication can be achieved by the antibody transfection technique, a valuable tool to be added as an antiviral therapy for the treatment of rabies in humans.
