Caracterização histológica da glândula de sal da tartaruga-verde (Chelonia mydas) LINNAEUS, 1758

Resumo

Este trabalho tem por objetivo a atualização da anatomia microscópica, através da microscopia óptica e eletrônica de varredura, da Glândula de Sal de Tartarugasverdes (Chelonia mydas). Para isso foram utilizadas glândulas de sal de 10 exemplares (5 machos e 5 fêmeas) de C. mydas extraídas do acervo do Setor de Morfologia e Anatomia Patológica (SMAP) do Laboratório de Morfologia e Patologia Animal (LMPA). As amostras foram processadas para avaliação microscópica óptica e eletrônica de varredura. Para a avaliação na microscopia óptica, além do corante de rotina, foram utilizadas técnicas de histoquímica para diferenciação e identificação de tecidos específicos. A Glândula de Sal foi caracterizada sendo uma glândula multilobulada, constituída por inúmeras unidades secretoras (US) distribuídas por toda a extensão dos lóbulos. Cada lóbulo é individualizado por um septo de tecido conjuntivo frouxo com a presença das vênulas e arteríolas interlobulares que fazem a irrigação do órgão. Há a presença de unidades secretoras, que são constituídas por um epitélio glandular composto tubular ramificado, formado por células serosas de formato triangular. Estas células assumem a função do órgão, de retirada do excesso de sais do organismo para excretá-los por uma secreção serosa para fora do organismo. Na região periférica observa-se uma maior concentração celular, com células de formato mais cuboide, citoplasma escasso, núcleos com a cromatina mais densa e citoplasma mais basofílico, características que indicam funções de células germinativas, possuindo a função de reposição celular. Na região central do lóbulo e na região interlobular foi observada a presença de tecido conjuntivo rico em fibras colágenas, sendo identificada, através de teste histoquímico, a presença de colágeno do tipo I na região ao redor do ducto central, no tecido conjuntivo intralobular. No tecido conjuntivo interlobular, observou-se uma variação em relação ao tipo do colágeno encontrado, no qual foi encontrado, em proporções relativamente iguais, a presença de colágeno do tipo I e III. Auxiliando no processo de escoamento da secreção produzida pelas US, uma rede de túbulos excretores, que se iniciam a partir do lúmen de cada US, vai confluindo e se agrupando em direção ao ducto central. Estes túbulos, assim como os ductos centrais, são revestidos por dois tipos celulares: um epitélio cilíndrico pseudoestratificado, com citoplasma escasso e basofílico na coloração de Hematoxilina-eosina, e núcleos esféricos e centrais, caracterizando células epiteliais de função germinativa; e uma camada de células apicais, colunares, com citoplasma fracamente corado pela HE, com o núcleo esférico em seu terço basal, que ao teste histoquímico específico foi possível a observação de um conteúdo de mucopolissacarídeos, insinuando uma função protetora do epitélio dos túbulos excretores e do canal central à ação lesiva da alta osmolaridade da secreção glandular. Conclui-se que este trabalho confirmou o que era descrito sobre o tema na escassa literatura e trouxe novas informações, e um banco de fotomicrografias de qualidade, que servirão de referência para demais pesquisas sobre o órgão e apoio para identificação de alterações morfofuncionais.
This work aims to update the microscopic anatomy, through scanning electron microscopy and scanning of the Salt Gland of Green Turtles (Chelonia mydas). For this, salt glands of 10 specimens (5 males and 5 females) of C. mydas extracted from the Morphology and Pathological Anatomy Sector (SMAP) of the Laboratory of Morphology and Animal Pathology (LMPA) were used. The samples were processed for optical microscopy and scanning electron microscopy. For optical microscopy evaluation, in addition to routine dye, histochemical techniques were used to differentiate and identify specific tissues. The salt gland was characterized as a multilobulated gland, consisting of innumerable secretory units (US) distributed throughout the lobes. Each lobe is individualized by a loosely connected connective tissue septum with the presence of interlobular venules and arterioles that irrigate the organ. There are the presence of secretory units, which are constituted by a branched tubular glandular epithelium composed of serous cells of triangular shape. These cells assume the function of the organ, the removal of excess salts from the body to excrete them by a serous secretion out of the body. In the peripheral region there is a higher cell concentration, with cells with a more cuboid shape, scarce cytoplasm, nuclei with more dense chromatin and more basophilic cytoplasm, characteristics that indicate germ cell functions, having the cell replacement function. In the central region of the lobe and in the interlobular region the presence of connective tissue rich in collagen fibers was observed. The histochemical test identified the presence of type I collagen in the region around the central duct in the intralobular connective tissue. In the interlobular connective tissue, a variation was observed in relation to the type of collagen found, in which the presence of type I and III collagen was found in relatively equal proportions.By aiding in the process of flow of the secretion produced by the US, a network of excretory tubules, which start from the lumen of each US, converge and clump towards the central duct. These tubules, as well as the central ducts, are covered by two cell types: a pseudostratified cylindrical epithelium with scarce and basophilic cytoplasm in Hematoxylin-eosin staining, and spherical and central nuclei, characterizing epithelial cells of germinal function; and a layer of apical cells, columnar, with cytoplasm weakly stained by HE, with the spherical nucleus in its basal third, that to the specific histochemical test it was possible to observe a content of mucopolysaccharides, insinuating a protective function of the epithelium of the excretory tubules and of the central canal to the action of the high osmolarity of the glandular secretion. It is concluded that this work confirmed what was described on the subject in the scarce literature and brought new information, and a bank of quality photomicrographs, which will serve as reference for other research on the organ and support for identification of morphofunctional changes.
