EXPRESSÃO DE GENES E FATORES DE TRANSCRIÇÃO LIPOGÊNICOS EM EXPLANTES MAMÁRIOS DE OVELHAS LACTANTES CULTIVADOS IN VITRO COM ÁCIDO PALMÍTICO

Resumo

A suplementação lipídica com ácido palmítico (C16:0), tem se mostrado eficiente em dar suporte à produção e aumentar o teor de gordura do leite. Assim, o objetivo deste estudo foi avaliar os efeitos do C16:0 na expressão de genes envolvidos na síntese de novo (ACC- PII, ACC- PIII e FASN), internalização (CD36), transporte intracelular (FABP 3 e 4) e dessaturação (SCD) de ácidos graxos, e também em um dos fatores de transcrição (PPAR-) envolvidos na regulação de enzimas envolvidas na síntese da gordura do leite. Foram realizados cultivos de explantes mamários de três ovelhas da raça Lacaune com 40 dias em lactação, obtidos através de biópsia. Os explantes foram cultivados por 24 horas, com os tratamentos: controle [meio de cultivo + albumina bovina (98%; BSA) a 75 M] e ácido palmítico [meio de cultivo + C16:0 (99%) a 75 M + albumina bovina (98%; BSA)]. Posteriormente foi extraído o RNA total, sintetizado o DNA complementar (cDNA) e realizada a análise quantitativa da reação em cadeia da polimerase em tempo real (RT-qPCR). Os dados foram analisados pelo programa estatístico SAS, utilizando o procedimento MIXED. O C16:0 aumentou, respectivamente, a expressão gênica da ACC- PIII, ACC- PII e FASN em 1,5, 1,3 e 1,3 vezes, quando comparado ao controle. A expressão gênica da CD36, FABP3, FABP4, SCD e PPAR- não sofreu efeito de tratamento. O aumento de ácidos graxos de cadeia curta e média observados na literatura pode justificar o aumento da expressão gênica das enzimas envolvidas na síntese de novo no presente estudo. O motivo pelo qual os genes envolvidos na via da captação de AG da circulação sanguínea não foram afetados pelo ácido palmítico não está claro.
Lipid supplementation with palmitic acid (C16:0) has been shown to increase milk production and milk fat content. The objective of this study was to evaluate the effect of C16:0 in the expression of genes involved in de novo synthesis (ACC- PII, ACC- PIII and FASN), internalization (CD36), intracellular transport (FABP 3 and 4) and fatty acids desaturation (SCD), and also in the transcription factor (PPAR-) involved in the regulation of genes involved in the milk fat synthesis. Mammary explants from three Lacaune ewes with 40 days in milk, obtained through biopsy were cultured for 24 hours with one of the following treatments: control [culture medium + bovine albumin (98%; BSA) at 75 M] and palmitic acid [culture medium + C16:0 (99%) at 75 M]. Subsequently, total RNA was extracted, complementary DNA (cDNA) was synthesized and carried out the real-time quantitative polymerase chain reaction analysis (RT-qPCR). The data were analyzed by the SAS statistical software, using the MIXED procedure. C16:0 increased, respectively, ACC- PIII, ACC- PII and FASN gene expression in 1.5, 1.3 and 1.3 fold, when compared to the control. The CD36, FABP3, FABP4, SCD and PPAR- gene expression had no treatment effect. The increase in short and medium-chain fatty acids observed in the literature may justify the increase in gene expression of the enzymes involved in de novo synthesis in the present study. The reason the genes involved in the FA uptake pathway from the bloodstream were not affected by palmitic acid is unclear.
