DETECÇÃO E DISTRIBUIÇÃO DE Mycoplasma spp. EM CÃES DE DIFERENTES LOCAIS DE CRIAÇÃO

Resumo

Os micoplasmas podem habitar as mucosas dos cães e são detectados em mucosas genital, respiratória e conduto auditivo desses animais. Quando patogênicos, causam doenças respiratória e urogenitais de curso crônico. O diagnóstico laboratorial das micoplasmoses é realizado pelo cultivo, técnicas sorológicas, imunoenzimáticas e moleculares. O objetivo geral deste trabalho foi detectar e investigar a distribuição de micoplasmas em cães procedentes de canis, centros de treinamento e acautelados em abrigos, em um estudo observacional descritivo. Foram coletados suabes de orofaringe, conduto auditivo e genital de 98 cães, totalizando 294 amostras, provenientes de oito canis com três diferentes tipos de manejo (A: centros de treinamento; B: abrigos; C: canis comerciais). As amostras brutas dos diferentes sítios anatômicos foram submetidas ao isolamento de micoplasma e a reação em cadeia da polimerase (PCR) para Mycoplasma spp., M. canis, M. edwardii, M. molare e M. cynos. Amostras positivas no isolamento foram encaminhadas para realização da imunoperoxidase indireta e PCR genérica e das espécies supra-citadas no intuito de tipificar os isolados. Em relação às espécies detectadas, M. edwardii foi a espécie mais encontrada nos cães com frequência de 15,31% (45/294), seguido de 11,22% (33/294) de M. canis e 1,02% (3/294) M. molare. Já M. cynos não foi detectado. Dos 98 animais testados, 74,5% (73/98) foram positivos no cultivo e/ou PCR. A orofaringe foi o sítio de maior detecção, com 59,2% (58/98) animais positivos. Houve maior detecção de micoplasma nos locais onde os cães tinham maior contato por compartilharem os mesmos ambientes e fômites, e menor cuidado sanitário. Características individuais dos cães como idade, sexo, uso prévio de antibióticos, doenças crônicas e o tamanho do plantel não apresentaram associação com a presença de micoplasma. A partir dos resultados encontrados, pode-se concluir que a técnica de cultivo mostrou-se mais eficiente que a PCR na detecção de micoplasmas em lavados de suabes provenientes de orofaringe, conduto auditivo e genital. Do ponto de vista epidemiológico, o principal fator que permitiu a maior frequência de micoplasma dentro das criações foi o manejo aplicado em cada estabelecimento, destacando-se uma alta frequência em cães que compartilham o mesmo ambiente por longo tempo, como em canis comerciais. Características inerentes aos animais pareceram não influenciar na ocorrência de micoplasma em cães.
Mycoplasmas can inhabit the mucosa of dogs, having already been detected in the genital, respiratory and ear canal of these animals. When pathogenic, they cause respiratory and urogenital diseases of chronic course. The laboratory diagnosis of mycoplasmosis is performed by culture, serological, immunoenzymatic and molecular techniques. The objective of this work was to detect and investigate the distribution of mycoplasmas in dogs from kennels, training centers and shelters, in a descriptive observational study. Swabs from the mucosa of oropharynx, genital and ear canal were collected from 98 asymptomatic dogs, totaling 294 samples, from eight kennels with three different types of management. Crude samples from different anatomical sites were subjected to mycoplasma culture and polymerase chain reaction (PCR) for Mycoplasma spp., M. canis, M. edwardii, M. molare and M. cynos. Positive samples in culture were sent to indirect immunoperoxidase and generic PCR and the species mentioned above in order to typify the isolates. Of the 98 animals tested, 74.5% (73/98) were positive through the analysis of the association of the tests, with the oropharynx being the location of more detection with 59.2% (58/98). M. edwardii was the species most found in dogs with frequency of 40.5% (45/294), followed by M. canis with 11.2% (33/294) and M. molare with 1,02% (3/294). M. cynos was not detected in any of the animals evaluated. There was a more detection of mycoplasma in the places where the dogs had greater contact and less health care. Individual characteristics of dogs such as age, sex, previous use of antibiotics, chronic diseases and the size of the herd were not associated with the presence of mycoplasma. From the results found, it can be concluded that the culture technique proved to be more efficient than PCR in detecting mycoplasmas in swabs from the oropharynx, genital and ear canal. Mycoplasma spp. was detected in the three anatomical sites studied, with a more frequency in the oropharynx. Mycoplasma edwardii was the most prevalent species in the studied dogs. From the epidemiological point of view, the main factor that allowed the highest frequency of mycoplasma within the breeding was the management applied in each establishment, with a high frequency in dogs that share the same environment for a long time, as in commercial kennels. Characteristics inherent to the animals did not seem to influence the occurrence of mycoplasma in dogs.
