Estudo de diferentes sistemas e curvas de congelação na eficiência da congelabilidade e fertilidade de sêmen equino

Resumo

O presente estudo objetivou comparar diferentes curvas de congelação utilizando-se uma máquina com curvas programadas (TK3000®) com congelação em vapor de nitrogênio liquido, utilizando sêmen de garanhões de alta e baixa congelabilidade. Comparou-se a eficácia do período de estabilização do sêmen em diferentes sistemas de transporte refrigerado (Botutainer® e Equitainer®) em relação à estabilização feita em refrigerador Minitüb® previamente à congelação. No experimento I e II foram utilizados 4 garanhões com sêmen de alta congelabilidade. As amostras foram refrigeradas durante 20 minutos previamente à congelação em refrigerador Minitüb® (GM), Botutainer® (GB) e Equitainer® (GE). Não houve diferença entre os grupos quanto a motilidade total (MT) e motilidade progressiva (MP): GM (67,1;32,1%); GB (65,2;31,9%); GE (63,4;30,4%) demonstrando que os dispositivos podem ser utilizados como uma alternativa aos refrigeradores. Os valores de integridade de membrana plasmática (IMP%) foram GM(49,43a); GB(48,70a); GE(44,90b). No experimento II foram utilizadas 3 curvas programadas na TK3000® (C1, C2, C3) e a metodologia em caixa de isopor (GC). As comparações foram realizadas em relação ao GC e não apresentaram diferença significativa. Valores em porcentagem de MT, MP e IMP: GCxC1 (70 x 67,5; 35,7 x 32; 48,3 x 46); GCxC2 (63,3 x 67,66; 33,3 x 34,8; 45,4 x 48,3); GCxC3 (63,2 x 65; 32,2 x 32,4; 48,09 x 49,3). No teste de fertilidade, as inseminações foram realizadas com amostras congeladas com C2 e GC apresentando índices de prenhez de 65 e 60% respectivamente, sem diferença significativa. A TK3000® foi tão eficaz quanto à metodologia de congelação em caixa de isopor em relação à congelabilidade e fertilidade. Todas as curvas testadas foram eficientes na manutenção da viabilidade
This present study compared freezing ratios in a programmable freezer (TK3000) performed by usual methodology, using good freezers and poor freezers stallions. The semen stabilization period efficiency was compared in cooled semen transport systems (Botutainer® e Equitainer®) in relation with Minitüb® fridge before the cryopreservation. Four good freezers stallions was used in experiments I and II. Samples were cooled during 20 minutes before the cryopreservation in a Minitüb® fridge (GM), Botutainer® (GB) and Equitainer® (GE). There was not difference between groups in relation with total motility (TM) and progressive motility (PM): CM (67,1;32,1%); GB (65,2;31,9%); GE (63,4;30,4%) showing that transport systems can be used as an alternative instead of fridges. Plasma membrane integrity (MPI%) values were CM (49,43a); GB (48,70a); GE (44,90b). Three frozen rations were used in experiment II in the TK3000® (C1, C2, C3) and the isothermic box (CG). Results from each freezing ratio were compared to the control group and no difference was observed. Percentage values from TM, PM and MPI were the following: CGxC1(70x67,5; 35,7x32; 48,3x46); CGxC2 (63,3x67,66; 33,3x34,8; 45,4x48,3); CGxC3 (63,2x65; 32,2x32,4; 48,09x49,3). In the fertility test, artificial inseminations were performed with C2 and GC frozen samples showing pregnancy rates of 65 and 60%, respectively, without significant difference. The TK3000® was as efficient as the isothermic box frozen methodology in relation to freezability and fertility. All freezing ratios were efficient in semen viability maintenance. Ten Mangalarga Marchador breed stallions were used in the experiment III.
