Variabilidade genética e identificação de QTLs de tíbia e peso corporal em Gallus gallus

Resumo

Neste estudo os objetivos foram estimar parâmetros genéticos e mapear loci associados a características ósseas e peso vivo aos 42 dias de idade (PV42) em Gallus gallus. Estas aves foram oriundas de uma população F2 resultante do cruzamento de uma linhagem de corte e outra de postura, mantidas pela Embrapa Suínos e Aves. Foram estimados parâmetros genéticos e identificados QTLs (loci de características quantitativas) para comprimento, largura e peso da tíbia e PV42, visando a melhor compreensão da arquitetura genética das características estudadas. Os componentes de variância foram estimados pelo método da Máxima Verossimilhança Restrita sob modelo animal multicaracterística. O modelo geral incluiu os efeitos aleatórios genético aditivo direto e residual e o efeito fixo da interação sexo (2 níveis) e incubação (17 níveis), totalizando 34 grupos sexo-incubação. As estimativas de herdabilidade para comprimento, largura e peso da tíbia e PV42, foram, respectivamente, 0,23 ± 0,08, 0,34 ± 0,09, 0,24 ± 0,08 e 0,15 ± 0,05. As correlações genéticas foram positivas e variaram de 0,56 ± 0,18 (entre comprimento da tíbia e PV42) a 0,89 ± 0,06 (entre largura e peso da tíbia). Quando PV42 foi incluído no modelo como covariável para as características ósseas, as estimativas de correlação genética entre as características da tíbia diminuíram e variaram de 0,28 ± 0,23 (entre comprimento e largura da tíbia) a 0,80 ± 0,11 (entre largura e peso da tíbia). Concluiu-se que a seleção favorecendo o aumento no PV42 poderia aumentar também as medidas de comprimento, largura e peso da tíbia. No entanto, como a associação genética entre PV42 e as características estudadas não é máxima, isto poderia em parte justificar os problemas ósseos encontrados em linhagens de frangos de corte, cujo principal critério de seleção é o peso corporal. The objectives of this study were to estimate genetic parameters and to map loci associated with tibia characteristics and body weight at 42 days of age (BW42) in Gallus gallus. An F2 chicken population was developed by crossing a broiler sire line and a layer line maintained by Embrapa Swine and Poultry. To better understand the genetic architecture of tibia length, width and weight and BW42, genetic parameters were estimated and QTL (Quantitative Trait Loci) for these traits were evaluated. Variance components were estimated by Restricted Maximum Likelihood under multi-trait animal model. The general model included the random additive genetic and residual fixed effect of the interaction and sex (2 levels) and incubation (17 levels), totaling 34 groups sex-incubation. Heritability estimates for length, width and weight of the tibia and BW42 were, respectively, 0.23 ± 0.08, 0.34 ± 0.09, 0.24 ± 0.08 and 0.15 ± 0.05. Genetic correlations were positive and ranged from 0.56 ± 0.18 (between tibia length and PV42) to 0.89 ± 0.06 (between width and weight of the tibia). When BW42 was included as a covariate in the model for bone characteristics, estimates of genetic correlations between traits of the tibia decreased and ranged from 0.28 ± 0.23 (between tibia length and tibia width) to 0.80 ± 0.11 (between tibia width and tibia weight). The genetic correlation between tibia traits and PV42 are not maximum, this could in part explain the bone problems found in strains of broilers. QTLs were mapped using 127 microsatellite markers, which covered 2630.30 cM of chromosomes 1-15, 18, 19, 23, 24 and 26-28. The analysis model mapping included the fixed effect of incubation (17 levels), sex (2 levels) and family of mothers, who ranged in different chromosomes.
