Two pyrethroids, deltamethrin (D) and alphacypermethrin (AC), have been used as commercial products for tick control worldwide. However, the effects of sublethal doses of these compounds on various tick species and their developmental stages have not been fully explored yet, although such knowledge could contribute to a more effective application of both pyrethroids and simultaneous reduction of their costs and undesirable side effects, including the harmful impact on other organisms and environmental contamination. This study investigates the effect of sublethal concentrations of D and AC applied to engorged females on the fecundity, development of eggs and larvae, and the number of offspring in the Ixodes ricinus (L.) tick, which is the most important vector of tick-borne diseases of humans in Europe. After detachment from rabbit's skin, fully engorged I. ricinus females were treated with 20 µl of pyrethroid solutions at five concentrations from 0.01562 to 0.25% and kept at 28 °C and 75% relative humidity. The impact of the pyrethroids on maturation and development of eggs as well as development of larvae was assessed based on parameters of the preoviposition and oviposition periods and the course of embryogenesis and egg hatch. The investigations have shown that both tested acaricides reduce the reproductive rate in I. ricinus females by inhibition of egg development and disturb embryonic development and larval hatch. Females did not lay eggs at concentrations higher than 0.0625% AC and 0.125% D. The lower concentrations of both pyrethroids disturbed or inhibited the embryogenesis and egg hatch in I. ricinus.

INTRODUCTION AND OBJECTIVE: Dermacentor reticulatus is one of the tick species of the greatest epidemiological importance in Europe. To date, the Eastern European and Western European populations of this tick species have been separated by an area located in Poland where the species has never been found. In this study, newly discovered D. reticulatus localities in areas transformed by human activities in central-southern Poland are described. MATERIAL AND METHODS: The specimens of the ornate dog tick were identified among ticks collected from companion animals in 2010, 2012, 2013, and 2014. They were examined using PCR methods to detect Borrelia burgdorferi s.l., Rickettsia spp., Anaplasma phagocytophilum, Bartonella spp., Babesia spp., and Toxoplasma gondii. In the case of the positive results, the amplicons were sequenced and examined by a BLAST search. RESULTS: In total, 6 specimens of D. reticulatus were collected (3 females and 3 males). As declared by the owners, animal hosts stayed in the same area throughout the study period and had never travelled outside their place of residence. As many as 3/6 (50%) of D. reticulatus adults removed from dogs were infected with Rickettsia raoultii. CONCLUSIONS: The results expand the available data on the spread of the ornate dog tick and indicate that, since 2010, this tick species and Rickettsia raoultii transmitted by this tick species have probably been present in this area, which has a strongly transformed agricultural structure and and had previously been regarded as a D. reticulatus-free zone. The presence of the ornate dog tick in urban and suburban habitats in central-southern Poland poses new threats to the health of companion animals and humans associated with the transmission of pathogens by this species.

Mutation of the heat shock gene, htpG, causes severe defects of several cellular functions in Escherichia coli. A null htpG mutant constructed by gene replacement was impaired in the biosynthesis and secretion of several enzymes, and in biofilm formation and proteolysis. A significant decrease in the activity of ß-lactamase in the ΔhtpG mutant was observed at 42°C. The alkaline phosphatase activity in sonicates of cells propagated at this raised temperature was lower in the ΔhtpG mutant than in the wild-type strain. The ability of the ΔhtpG mutant to degrade abnormal proteins was also impaired compared with the wild-type, but was increased at 42°C. Assays based on bioluminescence and crystal violet staining demonstrated that biofilm formation was diminished in the ΔhtpG mutant at the elevated temperature. All these defects can be complemented upon introducing htpG wild allele.

INTRODUCTION AND OBJECTIVE: The common tick Ixodes ricinus is one of Europe's most important vectors of tick-borne diseases. The increased risk of attacks by this tick suggests the need for identification of factors contributing to the transmission of tick-borne pathogens, and the routes of pathogen circulation in nature. MATERIAL AND METHODS: Polymerase chain reaction was used to investigate the prevalence of four pathogens, i.e. Borrelia burgdorferi s.l. (Bb), Anaplasma phagocytophilum (Ap), Babesia spp. (Bs), and Bartonella spp. (Ba) in I. ricinus females and males mating on dogs in south-central Poland. RESULTS: The study revealed the presence of three pathogens: Bb, Ap, and Bs in 9.4%, 5.4%, and 5.4% of all I. ricinus adults in copula, respectively. Co-infection with two pathogens was detected in one tick specimen. Borrelia burgdorferi spirochetes were isolated in two females and two males in copula, but the sexual transfer of the spirochetes between these specimens could not be clearly confirmed. CONCLUSIONS: By increasing the feeding dynamics in females, the copulation of I. ricinus males with females attached to the host's skin may stimulate pathogen replication in tick tissues and migration from the gut to the salivary glands. Further investigations of the I. ricinus copulation on the host on female feeding and pathogen transmission may contribute to the elucidation of the eco-epidemiology of tick-borne diseases transmitted by this tick species.

INTRODUCTION AND OBJECTIVE: Nurses are a group of healthcare professionals particularly vulnerable to infestations by Pediculus humanus capitis and Sarcoptes scabiei var. hominis contracted from patients under their medical care. MATERIAL AND METHODS: The cross-sectional study included 322 professionally active nurses within public healthcare units in eastern Poland. The research tool was a questionnaire collecting anonymized data on the occurrence of pediculosis capitis and scabies in nurses, and their patients, concerning the environmental determinants in the period 2001-2013. The study was retrospective and participation of nurses was voluntary. RESULTS: The obtained results indicated that 24.8% and 9.9% of the 322 respondents were infested by head lice and scabies mites, respectively. During their professional work, most nurses contracted head lice once (76.2%) while others (23.8%) were infested twice or more. The respondents did not declare repeated occupational scabies. The risk of contracting pediculosis capitis and scabies was not related to the length of service, but increased with the rise in the number of patients provided with nursing care. In the head lice-infested patients, the majority were aged 6-10 years (31.3%), while in the case of scabietic patients, children aged 0-5 years prevailed (26.4%). CONCLUSIONS: In medical care facilities, regular hygienic checks of both patients and medical staff, covering the condition of the skin and scalp, should be mandatory. The spread of pediculosis capitis and scabies among nurses can be reduced by the implementation not only of protective procedures mitigating occupational risk, but also the improvement of working conditions in medical facilities.

In mountainous regions, diverse ecosystems provide a habitat for numerous species of organisms. In this study, we focused on ixodid ticks and their presence in the Western Carpathians, Poland. Our objectives were to investigate the impact of environmental factors on tick occurrence and activity, the prevalence of vectored pathogens, and tick hosts, and their role as reservoir organisms for tick-borne pathogens (TBPs). To this end, we collected ticks from the vegetation and from animals (Apodemus agrarius, A. flavicollis, Capreolus capreolus, Microtus spp., Myodes glareolus, Ovis aries). In addition, we collected blood samples from rodents. The collected material underwent molecular analysis, utilizing the high-throughput microfluidic real-time PCR technique, to detect the presence of TBPs. Our findings confirmed the occurrence of only two species of ixodid ticks in the study area: the dominant Ixodes ricinus, and Dermacentor reticulatus with very limited abundance. Temperature significantly influenced tick activity, and the number of I. ricinus nymphs varied with altitude. We also observed a circadian pattern of questing activity in I. ricinus ticks. The main hosts for juvenile tick stages were M. glareolus and A. agrarius, while adult stages were frequently found on C. capreolus. I. ricinus ticks collected from the vegetation were often infected with Rickettsia helvetica (up to 35.71%), Borrelia afzelii (up to 28.57%), and Ehrlichia spp. (up to 9.52%). In contrast, juvenile stages frequently carried Bartonella spp. (up to 10.00%), Mycoplasma spp. (up to 16.67%) and R. helvetica (up to 16.67%). Moreover, we detected genetic material of Mycoplasma spp. (up to 100.00%), Ehrlichia spp. (up to 35.71%), Bartonella spp. (up to 25.00%), and Borrelia spp. (up to 6.25%) in rodent blood samples. The obtained results indicate A. agrarius and M. glareolus as reservoir animals for TBPs in the studied region.

Ixodes ricinus and Dermacentor reticulatus ticks are important reservoirs and vectors of pathogens. The aim of the present study was to investigate the dynamic of the prevalence and genetic diversity of microorganisms detected in these tick species collected from two ecologically diverse biotopes undergoing disparate long-term climate condition. High-throughput real time PCR confirmed high prevalence of microorganisms detected in sympatrically occurring ticks species. D. reticulatus specimens were the most often infected with Francisella-like endosymbiont (FLE) (up to 100.0%) and Rickettsia spp. (up to 91.7%), while in case of I. ricinus the prevalence of Borreliaceae spirochetes reached up to 25.0%. Moreover, pathogens belonging to genera of Bartonella, Anaplasma, Ehrlichia and Babesia were detected in both tick species regardless the biotope. On the other hand, Neoehrlichia mikurensis was conformed only in I. ricinus in the forest biotope, while genetic material of Theileria spp. was found only in D. reticulatus collected from the meadow. Our study confirmed significant impact of biotope type on prevalence of representatives of Borreliaceae and Rickettsiaceae families. The most common co-infection detected in D. reticulatus was Rickettsia spp. + FLE, while Borreliaceae + R. helvetica was the most common in I. ricinus. Additionally, we found significant genetic diversity of R. raoultii gltA gene across studied years, however such relationship was not observed in ticks from studied biotopes. Our results suggest that ecological type of biotope undergoing disparate long-term climate conditions have an impact on prevalence of tick-borne pathogens in adult D. reticulatus and I. ricinus.

Ixodes ricinus ticks are one of the most important vectors and reservoirs of infectious diseases in Europe, and tick-borne encephalitis (TBE) is one of the most dangerous human diseases transmitted by these vectors. The aim of the present study was to investigate the TBE incidence in some European countries during the COVID-19 pandemic. To this end, we analyzed the data published by the European Center for Disease Prevention and Control (ECDC) and Eurostat on the number of reported TBE and COVID-19 cases in 2020 and TBE cases in 2015-2019 (reference period). Significant differences in the TBE incidence were found between the analyzed countries. The highest TBE incidence was found in Lithuania (25.45/100,000 inhabitants). A high TBE incidence was also observed in Central European countries. In 12 of the 23 analyzed countries, there was significant increase in TBE incidence during the COVID-19 pandemic during 2020 compared to 2015-2019. There was no correlation between the incidence of COVID-19 and TBE and between the availability of medical personnel and TBE incidence in the studied countries. In conclusion, Central Europe and the Baltic countries are areas with a high risk of TBE infection. Despite the COVID-19 pandemic and imposed restrictions, the incidence of TBE is increasing in more than half of the analyzed countries.

Arthropods of the Hippoboscoidea superfamily are parasites of animals from various systematic groups. Mass appearances of these insects and their attacks on people are increasingly being recorded. Their parasitism has a negative effect on host well-being, as it causes feelings of agitation and irritation as well as skin itching and damage. It may result in weight loss and development of diseases in the long-term perspective. Parasites can be a potential epidemiological threat for their hosts as well. One of such parasites is a cosmopolitan species of the Hippoboscinae subfamily-Hippobosca equina. Studies have confirmed the presence of Corynebacterium pseudotuberculosis equi, Bartonella spp., and Anaplasma spp. in the organism of these insects. The frequency of anaphylactic reactions caused by H. equina attacks has been increasing. The aim of the present paper was to summarize the up-to-date knowledge of Hippobosca equina Linnaeus, 1758 due to its significance in medical and veterinary sciences as a potential vector of pathogens. Given the increasing expansion of ectoparasites, mainly related to climate change, ensuring animal welfare and human health is a priority.

The components of blackfly (Diptera: Simuliidae) saliva secreted during feeding on humans and animals induce various pathological reactions manifested by skin lesions and systemic symptoms. In this study, we describe 43 cases of severe skin lesions induced by blackfly bites and analyze their potential causes. Based on the results of a survey of 418 patients, we identified periods with risk of blackfly attacks and their environmental determinants in the eastern part of the European Union. Especially strong inflammatory reactions after blackfly bites were reported in patients with concomitant cardiovascular diseases (mainly with venous insufficiency), metabolic diseases, and bacterial infections. Enhanced symptoms of simuliosis were also observed in other patients attacked by these insects only once or repeatedly. The greatest number of blackfly attacks in the study area is recorded from May to July, with a peak in June (38.73%) in the afternoon (37.10%) and evening (33.03%), when patients are in fields, forests, or their households. The case analysis indicates interactions of various factors in the development of severe inflammatory reactions in patients bitten by blackflies. Therefore, it is recommended that subjects exposed to the presence of blackflies during their work or rest should limit the length of their stay in a vulnerable environment during the highest seasonal and daily activity of these insects. It is also necessary to take measures to reduce the number of blackflies and popularize methods for prophylaxis of their attacks.

Pediculosis capitis is a current and neglected health issue worldwide. The lack of screening programs contributes to the marginalization of the problem and delays therapeutic measures. Our study aimed to analyze the occurrence of this parasitosis in primary schools in Poland and to determine factors contributing to the persistence of its foci. The research tools were two questionnaires: one for primary school children and the other for school managers. While children answered questions about the epidemiology of pediculosis capitis and expressed their opinion on the hygienic condition of infested persons, the school directors were asked about the occurrence of head lice in schools, preventive measures, and institutions supporting schools in combating the infestation. The survey covered the period 2014-2018. Pediculosis capitis was reported in 87.5% of the schools. The greatest number of cases was reported in the group of 6-9 year-olds (68%). Among 4970 children, 16.7% had no knowledge of head lice; however, 57.1% wanted to increase their awareness of the problem. Campaigns on lice were conducted mainly as a result of emerging pediculosis capitis cases, and most schools could not rely on institutional support. Screening programs and preventive educational campaigns should be part of pediculosis capitis control in Poland.

Migratory birds play an important role in the eco-epidemiology of tick-borne diseases due to their ability to carry ticks for long distances. The aim of the present study was to investigate the prevalence and factors influencing the intensity of tick infestation in migratory birds. The study was conducted in a locality situated in the Vistula River valley, eastern Poland, during autumn, when the high migratory activity of birds is registered in the region. The birds were captured using ornithological nets and identified at the species level. In the next step, they were carefully inspected for attached ticks. Tick infestation was observed in 4.43% of the captured birds. The highest mean intensity of tick infestation was observed in birds foraging on the ground or in low shrubs and by long- and medium-distance migrants, i.e., Turdus merula (2.73), T. philomelos (2.04), and Erithacus rubecula (1.58). Ixodes ricinus was found to infest the birds most frequently. However, other tick species, i.e., I. trianguliceps, I. crenulatus (synonym I. canisuga), and I. apronophorus, rarely found in eastern Poland, were also found parasitizing birds. The occurrence of I. persulcatus, I. frontalis, and I. acuminatus (synonym I. redikorzevi) was confirmed in the region for the first time. The results of the study suggest that captured bird species are susceptible to tick infestation and could play an important role in the circulation of some tick-borne pathogens. They also play a significant role in the spread of ticks. The ecology and ethology of birds, including their foraging styles and migratory habits, are factors determining the risk of exposure of birds to tick attacks.

Colonization of skin by Demodex mites developing as a single facial lesion with a limited range is diagnosed relatively rarely. The presence of this type of skin lesions may hinder and thus delay the diagnostic process and application of appropriate treatment. The persistent cosmetic defect on the face is extremely onerous/embarrassing to the patient and impedes normal functioning. Describe a case of atypical presentation of primary facial demodicosis and its successful treatment. We present a case of a 38-year-old patient who consulted an aesthetic medicine doctor about a skin lesion, that is, erythematous-papular lesion with single pustules persisting for more than half a year around the right eye corner and below the lower eyelid. Previous topical antibiotic treatment has proved ineffective. Diagnosis was targeted at detection of Demodex spp. invasion, which yielded a positive result. Targeted therapy with 1% topical ivermectin was implemented. Complete resolution of the skin lesions was observed after 8.5 weeks of the treatment with no further recurrence. In the case of limited scaly erythematous-papular skin lesions with single pustules, Demodex mites should be considered as an etiological factor or one of the factors in the case of a mixed-etiology lesion. A delayed diagnosis of visible lesions on facial skin has a negative impact on patient's well-being and normal functioning. As diagnosis of primary demodicosis is underestimated, knowledge about this dermatosis and its clinical manifestations should be disseminated among cosmetologists and doctors of various specialties.

PURPOSE: The aim of the study was to examine Demodex survival in makeup cosmetics, i.e., powder cream, mascara, and lipstick, and to determine whether cosmetics shared with others can be a source of D. folliculorum infection. METHODS: Live D. folliculorum adults were placed in cosmetic samples and their motility was observed under a microscope. The mites were fully or partially immersed in the powder cream and lipstick, and only partially immersed in the mascara. Partial immersion means that only the opisthosoma was covered by the cosmetic, whereas the gnathosoma and podosoma had no contact with the cosmetic. Cessation of motility was regarded as a sign of death. RESULTS: In the control (mites placed on a microscope slide with no cosmetics), the survival time was 41.2 h. D. folliculorum that were immersed fully or partially in the lipstick substrate were viable for 38.5 h and 148 h, respectively. The survival time of the mites at full and partial immersion in the powder cream was 0.78 h and 2.16 h, respectively. The average survival time in the mascara was 21 h. CONCLUSIONS: Makeup cosmetics used by different individuals at short intervals (from several hours to several days) can be a source of transmission of Demodex sp. mites.

Ixodes ricinus, a hematophagous arthropod species with great medical importance in the northern hemisphere, is characterized by an ability to survive prolonged periods of starvation, a wide host spectrum, and high vector competence. The aim of the present study was to determine the quantitative parameters of questing I. ricinus ticks collected in eastern Poland during the spring peak of their activity. The study consisted in the determination of quantitative parameters characterizing I. ricinus females and males, i.e., fresh body mass, reduced body mass, lipid-free body mass, water mass, and lipid mass and calculation of the lipid index. A statistically significant difference was observed between the mean values of the lipid index in females collected during the first and last ten days of May, which indicates the progressive utilization of reserve materials in the activity period. Higher activity of I. ricinus female ticks was observed during the last ten days of May despite the less favorable weather conditions, indicating their strong determination in host-seeking behaviors accompanying a decline in the lipid content and the use of the "now or never" strategy.

Dermacentor reticulatus ticks are one of the most important vectors and reservoirs of tick-borne pathogens in Europe. Changes in the abundance and range of this species have been observed in the last decade and these ticks are collected in areas previously considered tick-free. This may be influenced by progressive climate change. Eastern Poland is an area where the local population of D. reticulatus is one of the most numerous among those described so far. At the same time, the region is characterized by a significant increase in the mean air temperature in recent years (by 1.81 °C in 2020) and a decrease in the average number of days with snow cover (by 64 days in 2020) and in the number of days with frost (by 20 days in 2020) on an annual basis compared to the long-term average. The aim of our research was to investigate the rhythms of seasonal activity and the population size of D. reticulatus in the era of progressive climate change. To this end, questing ticks were collected in 2017-2020. Next, the weather conditions in the years of observation were analyzed and compared with multi-year data covering 30 years preceding the study. The research results show that, in eastern Poland, there is a stable population of D. reticulatus with the peak of activity in spring or autumn (up to a maximum of 359 individuals within 30 min of collection) depending on the year of observation. Ticks of this species may also be active in winter months. The activity of D. reticulatus is influenced by a saturation deficit.

Ixodiphagus endoparasitoid wasps are natural tick enemies that can reduce their abundance. In this study, we investigated the presence of Ixodiphagus hookeri in Haemaphysalis concinna and Ixodes ricinus ticks in the Slovak Karst (southern Slovakia) and analysed the ecological and physiological relationships in the parasitoid-host system. Unfed H. concinna and I. ricinus nymphs harvested from vegetation were fed on rabbits. The engorged specimens were kept at 25 °C and 75% RH until the emergence of the adult wasps. For the first time in Europe, we found the presence of I. hookeri in two species of ticks collected in the same locality and compared their development in these tick species. The prevalence of wasps in H. concinna and I. ricinus during their spring activity was estimated at 10.64% and 27.78%, respectively. The presence of the wasps did not affect the duration of nymph feeding. Engorged wasp-infected ticks achieved higher body mass than non-infected specimens. In both tick species, there were no differences in the length of the development period and the number and sex ratio of adult I. hookeri. The analysed indicators and characteristics of the I. hookeri wasp-tick system can be used in research on tick control.

Ixodes ricinus is the most widely distributed tick species in Europe. Mainly deciduous and mixed forests, pastures, and urban parks are habitats preferred by this species. I. ricinus ticks are also one of the most important reservoirs and vectors of human and animal infectious diseases on the continent. Borrelia burgdorferi s.l. spirochetes causing Lyme borreliosis (LB) in humans and tick borne encephalitis virus (TBEV), which is a causative agent of tick-borne encephalitis (TBE), are pathogens with the highest medical importance transmitted by this species. Investigations of the environmental determinants of the occurrence and activity of I. ricinus are crucial for elucidation of the environmental background of tick-borne diseases. In eastern Poland, I. ricinus is a common species with peak activity recorded in May in the entire region. During this period, 49 females, 32 males, and 55 I. ricinus nymphs were collected from an area of 900 m2. The results of the present study show that the occurrence and seasonal activity of this tick species are mainly influenced by microhabitat conditions, and saturation deficit has a significant effect on the activity of the species. Eastern Poland is characterized by a high incidence of LB and TBE. We have shown a correlation between the forest cover and the number of reported cases of tick-borne diseases.

Insects of the genus Lipoptena are parasitic arthropods with a broad host range. Due to the type of parasitism (hematophagy), their potential role as vectors of pathogens, i.e., Bartonella sp., Anaplasma phagocytophilum, Rickettsia spp., and Borrelia burgdorferi is considered. As the range of their occurrence has been changing dynamically in recent years and infestations of humans have increasingly been reported, these organisms are now the subject of numerous studies. Our research aimed to present the molecular characteristics of Bartonella sp. detected in Lipoptena fortisetosa parasitizing wild cervids in south-eastern Poland. Adults of Lipoptena spp. were collected from carcasses of roe deer and red deer between spring and autumn in 2013. The PCR method was used to detect Bartonella sp. in the insects. We report two new haplotypes of the rpoB gene of Bartonella sp. isolated from L. fortisetosa feeding on wild cervids in south-eastern Poland and the presence of this invasive ectoparasitic species in the studied area since 2013. Phylogenetic analyses of newly obtained Bartonella sp. haplotypes confirmed their unique position on the constructed tree and network topology. The rpoB gene sequences found belonging to lineage B support the view that this phylogenetic lineage represents a novel Bartonella species.

Ticks are hematophagous ectoparasites of humans and animals. These arthropods employ different strategies in their host-seeking activity; most often, it is the "nest"- and "pasture-questing" behaviour. Some species, e.g., Dermacentor reticulatus, exhibit both types of activity depending on their developmental stage. The aim of the present study was to investigate the host-seeking activity of adult D. reticulatus ticks in the eastern part of Poland. To this end, ticks were collected with the flagging method during their seasonal activity in three different types of habitat. Active specimens were marked with a permanent marker and then released. This was repeated consistently at 7-day intervals using a different colour of the marker each time, which allowed tracking the questing activity of the specimens. Most frequently, repetitive tick activity (repeated up to seven times) was noted in a locality surrounded by urban developments. In an agriculturally unused open meadow habitat, 69.9% of D. reticulatus ticks were found to undertake questing activity only once. D. reticulatus females proved to be more aggressive and determined to find a host than the males of this species. Adult D. reticulatus ticks are able to stay in the habitat for a long time and undertake multiple host-seeking activities. The greatest threat of attacks on animals, including domestic animals, and sporadically humans, by these ticks occurs in meadow habitats, which are preferred by this species.
