RESUMO

The interaction between 2,2'-bithiophene-5-carboxylic acid (PT2) sublimed under ultra-high vacuum conditions and anatase (101) and rutile (110) TiO2 single crystal surfaces is investigated by studying the electronic spectral density near the Fermi level with synchrotron-based spectroscopy. The experimental results are compared to density functional theory calculations of the isolated PT2 molecule and of the molecule adsorbed on an anatase TiO2 (101) cluster. The relative concentrations of Ti, C, and S atoms indicate that the adsorbed molecule remains intact upon deposition, which is typical of a Stranski-Krastanov growth mode. The analysis of the O1s spectrum suggests a predominant bidentate geometry of the adsorption with both rutile and anatase surfaces, as supported by previous theoretical simulations. It is also theoretically and experimentally demonstrated that the PT2 adsorption causes the appearance of new electronic states in the gap near the TiO2 valence band. A pinning effect of the LUMO level of the dye is also theoretically predicted.

This report presents the first observation of an alkyl radical in supercritical CO(2) by any magnetic resonance technique. Muoniated ethyl radical has been detected in muon-irradiated supercritical CO(2) solutions. In the presence of a low concentration of ethene in supercritical CO(2), it is found that the addition of muonium to ethene is the only reaction channel, and that the yield of this process is enhanced compared to conventional solvents. The temperature dependence of the hyperfine coupling constants of the ethyl radical suggests that at a density of 0.3 g/cm(3) both the rotational motion of the methyl group and the electronic structure of the radical are similar to those in the gas phase, and therefore that the local environment around the ethyl radical is similar to the gas phase under these conditions. At higher densities, however, there is a remarkable and unexpected density dependence of the hyperfine coupling constant of the ethyl radical, which has never been observed in any environment. In this regime, the density dependence suggests that supercritical CO(2) has a significant effect on the electronic structure of the free radical. Thus, changing the density of CO(2) offers a possible means of tuning the radical reactivity. In addition, at a density of close to 0.4 g/cm(3), CO(2) molecules cluster around the ethyl radical, and this increases the local density around the ethyl radical by a factor of ~1.5.

Eukaryotic elongation factor 1 (eEF-1) contains the guanine nucleotide exchange factor eEF-1B that loads the G protein eEF-1A with GTP after each cycle of elongation during protein synthesis. Two features of eEF-1B have not yet been elucidated: (i) the presence of the unique valyl-tRNA synthetase; (ii) the significance of target sites for the cell cycle protein kinase CDK1/cyclin B. The roles of these two features were addressed by elongation measurements in vitro using cell-free extracts. A poly(GUA) template RNA was generated to support both poly(valine) and poly(serine) synthesis and poly(phenylalanine) synthesis was driven by a poly(uridylic acid) template. Elongation rates were in the order phenylalanine > valine > serine. Addition of CDK1/cyclin B decreased the elongation rate for valine whereas the rate for serine and phenylalanine elongation was increased. This effect was correlated with phosphorylation of the eEF-1delta and eEF-1gamma subunits of eEF-1B. Our results demonstrate specific regulation of elongation by CDK1/cyclin B phosphorylation.

Elongation factor 1, a complex involved in protein biosynthesis, contains two guanine-nucleotide-exchange proteins EF-1 beta and EF-1 delta. The sequence of EF-1 delta of Artemia was determined with the purified protein. When compared to EF-1 delta from Xenopus, a high degree of identify (80%) was found in the C-terminal domains of the proteins, which contain the guanine-nucleotide-exchange activity. The N-terminal domains share only 23% of the amino acids at identical positions, and therefore they were further analysed for less obvious types of homology. To this end, a published approach for sequence analysis, which can detect peculiar amino acid patterns in proteins was applied. In this way, a weak albeit unmistakable similarity between the two EF-1 delta proteins was demonstrated in the region of the leucine-zippers, apart from the leucine repeat itself. Apparently, they display a common structural pattern in their N-terminal domains, which so far has been observed mainly in transcription factors.

The biological importance of the various changes in polyamine metabolism that occur during early Xenopus development have been investigated. Incubation of embryos in high salt medium was observed to cause a precocious fall in ornithine decarboxylase activity without affecting development. Similarly, inhibiting ornithine decarboxylase activity with specific inhibitors did not affect development. Injecting spermidine, within physiologically relevant limits, caused a dose-dependent inhibition of mitotic divisions in the injected blastomere. Increasing the intracellular putrescine did not affect cell division or development. Co-injection of both spermidine and putrescine, so that the original molar ratio of these two polyamines was conserved, abrogated the inhibition of cell division observed when spermidine was injected alone. Therefore, in Xenopus embryos the intracellular spermidine concentration must be retained within certain limits relative to that of putrescine to allow normal development.

M-Phase specific protein kinase or cdc2 protein kinase is a component of MPF (M-Phase promoting factor). During meiotic maturation of Xenopus oocytes, cdc2 protein kinase is activated in correlation with MPF activity. A protein phosphorylation cascade takes place involving several protein kinases, among which casein kinase II, and different changes associated with meiosis occur such as germinal vesicle breakdown, chromosome condensation, cytoskeletal reorganization and increase in protein synthesis. Our results provide a biochemical link between cdc2 protein kinase and protein synthesis since they show that the kinase phosphorylates in vitro a p47 protein identified as elongation factor EF1 (gamma subunit) and that the in vitro site of p47 corresponds to the site phosphorylated in vivo. Immunofluorescence showed that the elongation factor (EF1-beta gamma) is localized in the oocyte cortex. Furthermore, they show that cdc2 kinase phosphorylates and activates casein kinase II in vitro, strongly supporting the view that casein kinase II is involved in the phosphorylation cascade originated by cdc2 kinase.

This paper describes the purification of a 47 kDa protein from Xenopus laevis oocytes that becomes phosphorylated when the oocytes undergo meiotic maturation. This protein (p47) is part of a high molecular mass complex containing at least two other proteins of molecular mass 30 and 36 kDa. This complex can be isolated from stage VI oocytes before maturation. We obtained a pattern for phosphopeptides in p47 phosphorylated in vivo very similar to that of the purified protein phosphorylated in vitro by p34cdc2 (a H1 kinase which is a component of the M-phase promoting factor) and [gamma-32P]ATP. Therefore, the purified p47, already described as a marker of MPF activity, is the first reported in vivo substrate for the cell division control kinase.

A polyclonal antibody was raised against bacterially produced catalytic alpha subunit of protein phosphatase 2A (PP2AC) cloned from Xenopus ovarian library. The amount of PP2AC in Xenopus oocytes determined by Western blot analysis was 1 ng/microgram of cytosolic protein. The antibody depleted PP2AC from oocyte extracts in association with 6 components (40, 62, 65, 80, 85 and 90 kDa). Prophase- and metaphase-arrested oocytes contained identical amounts of PP2AC. Metaphase oocytes showed one specific change in the 62 kDa protein associated with PP2AC.

Although surgical wound infections (SWI) following implantation of prosthetic devices can be catastrophic and often require removal of the prosthesis, few studies have identified risk factors for these infections. We conducted a prospective multicenter study to identify risk factors for SWI. Of 561 vascular surgery patients enrolled in the study, 23 (4.1%) developed SWI. Multivariate analysis using logistic regression analyses identified surgery on lower extremities, delayed surgery, diabetes mellitus, past history of vascular surgery, and short antimicrobial prophylaxis (three doses of cefamandole) as independent risk factors for SWI. Consequences of SWI were serious; two (9%) died, 11 (48%) required reoperation, and five (22%) had their prosthesis removed. A risk index was developed using the independent risk factors for SWI identified by logistic regression analyses. When no risk factors were present, no SWI was observed (0 of 100), and the rate of SWI increased from 2.5% when one risk factor was present to 53.8% (7 of 13) when greater than or equal to 4 risk factors were present.

Two experiments explored the effects of speed, accuracy, on speed and accuracy instructions on visual field advantages to lexical decisions in males and females. In Experiment 1, right visual field advantages in reaction time appeared in the groups of males in the accuracy condition and in the speed and accuracy condition. In Experiment 2, feedback consistent with the instructions was given to subjects. Females and males in the accuracy condition showed a right visual field advantage in the second block of trials. These findings suggest that cognitive variables mediate right visual field advantages to lexical decisions in males and females.

The eukaryotic guanine-nucleotide exchange factor commonly called elongation factor-1 betagammadelta (EF-1betagammadelta), comprises four different subunits including valyl-tRNA synthetase (EF-1betagammadelta/ValRS). The factor is multiply-phosphorylated by three different protein kinases, protein kinase C, casein kinase II and cyclin dependent kinase 1 (CDKI). EF-1betagammadelta/ValRS is organized as a macromolecular complex for which we propose a new structural model. Evidence that EF-1betagammadelta/ValRS is a sophisticated supramolecular complex containing many phosphorylation sites, makes it a potential regulator of any of the functions of its partner EF-1alpha, not only involved in protein synthesis elongation, but also in many other cellular functions.

In vivo beta casein phosphorylation was analysed in Xenopus full-grown oocytes arrested in the prophase of the meiotic cell division. The phosphorylation was inhibited by the protein kinase inhibitor (PKI) and also by heparin (3 micrograms/ml; final concentration). beta casein phosphorylation was increased by spermine (2 mM). Therefore, protein kinase A and casein kinase II are both active in vivo in full-grown oocytes and may be involved in the prophase arrest of meiotic cell division.

A componential model capable of representing simple and complex forms of mental addition was proposed and then tested by using chronometric techniques. A sample of 23 undergraduate students responded to 800 addition problems in a true-false reaction time paradigm. The 800 problems comprised 200 problems of each of four types: two single-digit addends, one single- and one double-digit addend, two double-digit addends, and three single-digit addends. The results revealed that the columnwise product of addends, a structural variable consistent with a memory network retrieval process, was the best predictor of mental addition for each of the four types of problem. Importantly, the componential model allowed estimation of effects of several other structural variables, e.g., carrying to the next column and speed of encoding of digits. High levels of explained variance verified the power of the model to represent the reaction time data, and the stability of estimates across types of problem implied consistent component use by subjects. Implications for research on mental addition are discussed.

Environmental conditions can be the driving force behind an epizootic. Environmental changes may favor growth of a particular species, which results in increased contact rates and spread of a disease. We examine this particular phenomenon in SI and SIS models and use it to explain the possible disease outbreaks in nature. Either infected individuals recover from the disease (SIS model) or suffer disease fatalities (SI model). Epizootic models for a single population are examined where contact rate depends on population size. A reproductive number R is defined that depends on environmental carrying capacity. The single-population models are coupled to form three different two-species models with intra- and interspecies contact rates that depend on the population sizes of both populations. The stability results show that it is possible for the disease to drive one of the populations to extinction, the one with disease fatalities. The surviving species serves as a reservoir for the disease. Single- and two-species epizootic models are examined in a particular case where the contact rates are assumed to be constant. This leads to a new definition for the contact rate. A complete global analysis is possible in these latter cases. The results are compared and contrasted with the models with variable contact rates. The prototype for the models is the spread of disease in wildlife populations, which includes such diseases as plague or Lyme disease.

Loss of attachment as measured by a portion of Ramfjord's Index (PDI) was determined in 383, 12-16-year-old schoolchildren in rural Juniata County, PA. Prevalence and distributions were tabulated for sex, last dental visit, and type of treatment received. Dental knowledge test scores, toothbrushing effectiveness periodontal status and oral cleanliness were determined at annual intervals from Spring '75 to Spring'79. Mean scores were contrasted with bone loss measurements. 47.3% had loss of attachment (greater than 1 mm) including 24.5% with 2mm or more of attachment loss per mouth. Females (41.0%) ahd significantly lower prevalence rates than males (52.7%). Students who visited a dentist in the last 6 months and who received scaling had less bone loss, lower GI, PII, PI, and QHI scores. An inverse relationship was found between dental knowledge and the other variables measured. Similar patterns were seen with high, medium and low bone loss groups. Many variables in this study were associated with bone loss, including: plaque, dental knowledge, dental behavior (toothbrushing and visiting a dental clinic), recent treatment received and sex. None appeared more significant than the others.

The present study discusses the development of dental health knowledge tests as part of an integrated dental health curriculum in the public school system of a rural community. It is part of a large project designed to test the effectiveness of a school-based dental health delivery system. Cognitive measures of dental health were designed to study the relationship of dental health knowledge to oral health behaviors. A review of the literature revealed no suitable dental health knowledge tests for grades K-6; new assessment measures were then developed by project staff. The tests consisted of 14 objectively scored tests--two parallel forms at each of the seven grade levels, K-6, and were administered four times to 1,942 students. Consistently, the tests have demonstrated high reliability estimates and low standard errors of measurement and indicate that the instruments are functioning as parallel forms and measuring the same dental concepts with equal precision.

A prospective, randomised study was carried out on 110 patients to find out the effect of giving Cefotetan prophylactically as against no treatment in the prevention of infections after cesarean sections. The cases were selected before or at the beginning of a trial of labour in 72 cases and after a failure of trial of labour in 38 cases. A single dose of 2 gr of Cefotetan was given during the operation, after the cord had been clamped. It caused a significant drop in infection and in particular the number of cases of endometritis and urinary tract infections, which were twice as common in the non-treated as compared with the treated group.

The authors compare the efficiency of two drugs in the treatment of threatened premature labour, the one being the calcium inhibitor (nifedipine) and the other a beta-mimetic drug (ritodrine). 62 patients after random selection were divided into two groups: 32 treated with ritodrine and 30 treated with nifedipine. The treatment was carried out over 7 days. The success rate was similar in both groups -72% for the ritodrine group and 63.33% for the nifedipine group. Women receiving nifedipine had slightly greater gain in weeks, six as against five for ritodrine. The side effects which were often found with nifedipine were; hot flushes (in 10 cases) and headaches (4 cases). These symptoms appeared 15-30 minutes after the first dose and were transitory. No neonatal complications were found. The ease with which a calcium inhibitor can be given suggests that it should be used more frequently in the treatment of threatened premature labour and particularly when there are contra-indications to the use of beta-mimetic drugs.

The purpose of the present study was to document that instruction effects can occur on tasks typically yielding left visual-field advantages. In a lateralized face-recognition task, 40 right-handers (20 men, 20 women) had to memorize pairs of faces and to respond to a probe face displaying positive or negative emotion and presented for 100 msec. 3.8 degrees on the left or right of a fixation cross. Half of the subjects had been instructed to learn the faces through a verbal strategy and the other half through an imagery strategy. Analysis of reaction times yielded significant complex interactions involving gender, coding instruction, visual field, and emotion for both Yes and No responses. A global left visual-field advantage was obtained in the analyses of accuracy and sensitivity data, but no significant effects were recorded in the analysis of decision rule. Thus, the instruction effects on left visual-field advantages were obtained through complex interactions on reaction times only. Further replication of these data would provide a more secure basis for causal interpretations.
