RESUMO

In urban areas around the world, air pollution introduced by vehicular movement is a key concern. However, restricting vehicular traffic during the COVID-19 shutdown improved air quality to some extent. This study was conducted out in the smart city of Bhubaneswar, which is also the state capital of Odisha, India. The study has tried to map Bhubaneswar by collecting the air quality data before, during, and after the COVID lockdown of six air quality monitoring stations present in Bhubaneswar established under "National Ambient Air Monitoring Program" (NAMP). Furthermore, plants, which are the most vulnerable to air pollution, can show a variety of visible changes depending on their level of sensitivity. Moreover, leaves of Mangifera indica, Monoon longifolium, Azadirachta indica, Millettia pinnata, Aegle marmelos were collected from nearby of six air monitoring stations to assess the "Air Pollution Tolerance Index." M. indica was found to be intermediately tolerant, and all of the other species were found to be sensitive. The structural equation modeling results also revealed a significant relationship between total chlorophyll content, relative water content, ascorbic acid content, leaf extract pH, APTI with species, air quality index, and PM10.

Lipoprotein has been reported as a vaccine candidate against many pathogenic bacteria, it plays direct role as a virulence-associated function. Here the approach is toward the expression of recombinant lipoprotein of Salmonella typhi in prokaryotic host and its evaluation as a vaccine candidate. Lipoprotein gene (lp1) was cloned in pET32a expression vector in addition to Bam HI and Hind III restriction sites, and BL21(pLysS) was used as prokaryotic expression host for transformation. Lipoprotein induction was performed by IPTG and 55 kDa (31 kDa of Gene +24 kDa of vector additional protein with His-tag) was analyzed by 12% SDS-PAGE. The recombinant lipoprotein was purified by Ni-NTA affinity chromatography due to the addition of 6X His-tag in recombinant lipoprotein. Western blot analysis using anti-His tag polyclonal antibodies confirmed the specificity of recombinant lipoprotein. Immunogenicity and protection study of recombinant lipoprotein against S. Typhi was performed in BALB/c mice. Adjuvants IFA and alum salts were used to enhance the immune response. ELISA results proved that biologically active truncated recombinant lipoprotein (31 kDa) is a suitable immunogen. Alum salts used as adjuvant was effective for long-lasting immunity.

Asian tiger shrimp Penaeus monodon (P. monodon) of Chilika lagoon, India was studied regarding the metal accumulation and its associated human health risks. It showed a tendency of metal accumulation in the following order: Zn > Ni > Cu > Co > Cr > Pb > Cd. A two-way ANOVA indicated the metal accumulation was insignificant with respect to season (n = 421, p = 0.59) and sector (n = 32, p = 0.61). The estimated daily intake (EDI), targeted hazard quotient (THQ) and hazard index (HI), and carcinogenic risks (CR) revealed no potential human health risks and were safe for consumption. The pollution load index (PLI) of <1, Geo-accumulation index (Igeo,) and contamination factor (CF) indicated that the study area was unpolluted. This pioneering study highlighted that P. monodon was nurtured well in the healthy habitat of Chilika lagoon and the fair level of metal content made it an excellent source of dietary components.

India successfully executed one of the strictest lockdowns in the world during the height of the COVID-19 pandemic in early 2020, which provided unique opportunities to analyze the second-largest populous country's anthropogenic footprint on its natural systems. India's first Ramsar site and the world's second-largest brackish water system Chilika lagoon experienced a substantial decline (64%) in the total petroleum hydrocarbon (TPHC) level in water, which was attributed to the massive declines or, at times, an abrupt complete halt of motorized boat operations for fishing and tourism. Using the TPHC values during the lockdown period, our study recommends a TPHC baseline threshold of 2.02 µg L-1 and 0.91 µg g-1 for Chilika waters and sediment, respectively. These baseline values can be used to quantify oil pollution and to formulate policy and management action plans for Chilika lagoon as well as for other similar ecosystems by local environmental agencies.

Design and facile fabrication of a magnetically separable hetero-structure photocatalyst as well as an adsorbent having dual green benefits towards energy conversion and pollutant remediation are quite indispensable in the current scenario. In this regard, a composite of citrate capped Fe3O4 and UiO-66-NH2 has been designed to remediate Cr (VI) by adsorption and harvest photons from visible light for clean energy (H2) conversion. The material was prepared by the union of citrate capped Fe3O4 (CCM) and versatile aqueous stable Zr-based MOF (UiO-66-NH2) through in-situ solvothermal method. The composite of CCM with MOF (MU-2) was studied through sophisticated analysis techniques; PXRD, FT-IR, BET, UV-Visible DRS, PL, TG, HRTEM and XPS etc. to reveal the inherent characteristics of the material. BET surface analysis revealed high specific surface area (572.13 m2 g-1) of MU-2 in comparison to its pristine MOF. Furthermore, the dual function composite MU-2's VSM studies showed that its magnetic saturation is 3.07 emu g-1 that is suitable for magnetic separation after desired reaction from aqueous media. The Cr (VI) sorption studies revealed that the composite adsorbent (MU-2) showed maximum monolayer adsorption capacity (Qm) of 743 mg g-1 which followed pseudo second order kinetics. Moreover, the sorption thermodynamics revealed that the process was spontaneous and endothermic in nature. In addition to it, the synthesized composite material displayed enhanced activity towards photocatalytic H2 evolution with a maximum evolution rate of 417 µmole h-1 with an apparent conversion efficiency (ACE) of 3.12 %. Typically, MU-2 displays high adsorptions of Cr (VI) as well as some extent of Cr (VI) reduction owning to its populous active sites and free carboxylate groups respectively. Moreover, the synergistic effect of CCM and UNH in the composite resulted in Z scheme mediated charge transfer mechanism that showed enhanced H2 photo-evolution rates. Hence, MU-2 can be readily utilized as magnetically retrievable dual function composite for Cr (VI) adsorption and photocatalytic H2 evolution.

 In present study, Taguchi's design of experiment L9 orthogonal array was created using Qualitek-4 software with four most critical factors namely, K2HPO4, MgSO4, CaCl2 and culture pH. Production of a new intracellular antifungal protein in submerged fermentation was optimized with yield of 0.98±0.1 mg/gram dry cell weight mycelia from Aspergillus giganteus MTCC 8408. The average molecular mass of the purified protein was figured as 5.122 kDa using Electro Spray Ionization-Mass Spectrometry. Scanning electron microscopy was used to correlate the effect of selected factors on fungal cell morphology and its metabolite production. In vitro antifungal susceptibility assay was profiled against Aspergillus niger and minimum inhibitory concentrations were in the range 0.3±0.06 µg/ml. The stronger influencing factors on afp production and mycelial biomass were noted with CaCl2 and K2HPO4 respectively. The validation experiments using optimized conditions confirmed an improvement in afp by 3.86 times with mycelial biomass by 1.52 times, compared to the basal medium. The present statistical optimization study revealed an opportunity to promote economical design at the industrial level for future scale up of effective antifungal agent against systemic aspergillosis as well as possible post harvest loss.

A new intracellular antifungal protein (afp) production with average molecular weight 24.3 kDa and yield of 0.65 ± 0.1 mg/gram dry cell weight (gdcw) of mycelia in submerged fermentation of Aspergillus giganteus MTCC 8408 was optimized. Taguchi's DOE (design of experiment) L27 orthogonal array (OA) was constructed using Qualitek-4 software with 8 most influensive factors namely, culture pH, temperature, slant age, inoculum volume, agitation and KH2PO4. Scanning electron microscopy (SEM) was used to correlate the effect of selected factors on fungal cell morphology and afp production. The crude protein purification was accomplished using pure ammonium sulfate fractionation followed by carboxymethyl cellulose (CMC) ion-exchange chromatography and sephadex G-100 gel filtration. The average molecular mass of the purified protein was figured by silver stained SDS (sodium dodecylsulphate)-PAGE (poly-acryl amide gel electrophoresis). In vitro antifungal susceptibility assay was profiled against Candida albicans NCIM 3471 and minimum inhibitory concentrations (MICs) were in the range 3 to 4 µg/ml. Characterization of protein was observed with FTIR (Fourier transform infrared spectroscopy) analysis. The optimal production condition for crude afp was obtained as follows: soluble starch: 20 g/l; Corn steep liquor (CSL, 2%) + proteose peptone (PP, 1%): 30 g/l; pH: 5.8; temperature: 25°C; slant age: 3 d; inoculum size: 5% (v/v); agitation: 180 rpm; KH2PO4: 0.1 g/l. The validation experiments using optimized conditions confirmed an improvement in afp production by 59.4% against the expected enhancement of afp production by 61.22%. The present statistical optimization study revealed an opportunity to promote economical design at the industrial level for future scale up of effective antifungal agent against opportunistic oral and vaginal infection.

This paper is concerned with the effect of individual Na+, K+, Mg2+ and Ca2+ ions upon the nutrient dynamics and cell morphology during submerged fermentation of Aspergillus giganteus MTCC 8408 in a medium containing ample soluble starch, corn steep liquor and proteose peptone supply. Nutrient dynamics was elucidated using Taguchi's DOE (design of experiment) L8 orthogonal array (OA) with carbon, nitrogen and four most influensive microelements at their assigned ratio namely; K+/Ca2+ and Mg2+/Na+. Scanning electron microscopy (SEM) was used to correlate the effect of selected factors on morphological changes and nutrient dynamics during submerged fermentation. At higher K+/Ca2+ ratio 4.78:1; the maximum growth was figured out in terms of dry weight, while the inhibition of dry weight production was observed at higher Mg2+/Na+ ratio 0.94:1 with enhanced intracellular afp production. At lower C/N ratio 27.4:1; filamentous form of growth was maintained with the hyphae being scarcely branched without bulbous cells. Membrane perturbation due to the induction of intracellular oxidative stress was noticed at higher C/N ration. Taguchi DOE resulted in afp yield per substrate utilized, Y p/s of 1.08 mg g-1 soluble starch utilized with gram dry cell weight (gdcw) of 23.9 g L-1 while afp yield per biomass utilized resulted, Y p/x of 1.12 mg afp gdcw-1 L-1 and biomass yield per substrate utilized, Y x/s of 1.195 gdcw g-1 L-1. The present study revealed an opportunity to develop a cost effective methods for cell propagation so as to yield effective inoculum levels towards mass production and formulation studies.

A study was conducted by house-to-house visit during 2nd and 3rd round of IPPI in Nov and Dec 2000 in two slums adjacent to Medical College Kolkata. Total houses visited in these two rounds were 2836 and 611 children were immunised at home. Mothers of 503 children (who did not receive IPPI dose at booth on NIDs) were interviewed. For rest 104 children, mothers were not available during the time of visit. The result showed that infants were the largest group (30.6%) to receive vaccine at home. Majority (84.1%) was permanent residents of the locality. Only 4.2% were Bengali-speaking Hindus while most other were Hindi or Urdu speaking Muslims. "Health workers will bring vaccine at door steps" was the reason of non-attendance at booth by 30.2% parents. No routine immunisation was received by 14.9% children. The source of information regarding IPPI was, 45.4% from medical students, 28.5% from health workers, 15.1% from TV, 5.6% from relatives and neighbours, 3.6% from miking and 1.8% from newspapers respectively.

The present study reports the photo-Fenton degradation of phenolic compounds (phenol, 2-chloro-4-nitrophenol and 4-chloro-2-nitrophenol) in aqueous solution using mesoporous Cu/Al(2)O(3)-MCM-41 nanocomposite as a heterogeneous photo-Fenton-like catalyst. The in situ incorporation of mesoporous Al(2)O(3) (MA) into the framework of MCM-41 (sol-gel method) forms Al(2)O(3)-MCM-41 and wetness impregnation of Cu(II) on Al(2)O(3)-MCM-41 generates mesoporous Cu/Al(2)O(3)-MCM-41 composite. The effects of pH and H(2)O(2) concentration on degradation of phenol, 2-chloro-4-nitrophenol and 4-chloro-2-nitrophenol are studied. Kinetics analysis shows that the photocatalytic degradation reaction follows a first-order rate equation. Mesoporous 5 Cu/Al(2)O(3)-MCM-41 is found to be an efficient photo-Fenton-like catalyst for the degradation of phenolic compounds. It shows nearly 100% degradation in 45 min at pH 4. The combined effect of small particle size, stabilization of Cu(2+) on the support Al(2)O(3)-MCM-41, ease reducibility of Cu(2+) and visible light activeness are the key factors for quick degradation of phenolic compounds by Cu/Al(2)O(3)-MCM-41.

The aim of the present research was to study the anticancer effects of Aspergillus niger (A.niger) RNase. We found that RNase (A.niger RNase) significantly and dose dependently inhibited invasiveness of breast cancer cell line MDA MB 231 by 55 % (P<0.01) at 1 µM concentration. At a concentration of 2 µM, the anti invasive effect of the enzyme increased to 90 % (P<0.002). Keeping the aim to determine molecular level interactions (molecular simulations and protein docking) of human actin with A.niger RNase we extended our work in-vitro to in-silico studies. To gain better relaxation and accurate arrangement of atoms, refinement was done on the human actin and A.niger RNase by energy minimization (EM) and molecular dynamics (MD) simulations using 43A(2) force field of Gromacs96 implemented in the Gromacs 4.0.5 package, finally the interaction energies were calculated by protein-protein docking using the HEX. These in vitro and in-silico structural studies prove the effective inhibition of actin activity by A.niger RNase in neoplastic cells and thereby provide new insights for the development of novel anti cancer drugs.

The 3D models of human actin protein and A.niger RNase were designed using the templates ACTBIND (PDB ID: 3D3Z) and crystalline profilin-beta-actin (PDB ID: 2BTF), respectively in Modeller9v5. These models are testified using several validation methods including PROCHECK, ERRAT, WHAT-IF, PROSA2003 and VERIFY-3D. The stereo-chemical quality of the models was judged by Ramachandran plot with PROCHECK. The total quality G-factor -0.2, shows a good quality model. The ERRAT score for the human actin and A.niger RNase models are 86.104 and 84.615, respectively, fit well within the range of a high quality model. The ERRAT score for the templates 2BTF and 3D3Z are 91.111 and 97.391, respectively. The WHAT-IF evaluation justifies a reasonable homology model structure as none of the scores for each residue in the homology model is lower than -5.0. The energy-minimized model of human actin with PROSA reveals the Z-score value -10.52 between native conformations of the crystal structures. The VERIFY 3D average score is 0.36. All evidence suggests that the geometric quality of the backbone conformation, the residue interaction, the residue contact and the energy profile of the structures were well within the limits of reliable structures. The interaction energy of docking was calculated using the HEX server. The Etotal, lowest docked energy, and calculated RMSD values were -1.608 kcal mol(-1), -8.369 kcal mol(-1) and 0.617 Å, respectively. The study presented in the current project may be useful to design molecules that may have anticancer activity.
