Placental abnormalities more frequently occur during pregnancy of somatic cell clones and may lead to pregnancy loss or dystocia. Adventitious placentation, or diffuse semi-placenta, is determined by the development of areas of accessory placentation between the cotyledons due to the abnormal growth of placentomes.After a full-term pregnancy, a 3-year-old Jersey heifer was referred for dystocia which resulted in the delivery of a dead calf. The cause of dystocia was found to be foetal malposition, while the placenta was physiologically expelled after dystocia resolution.Grossly, cotyledons appeared reduced in size and number in one placental horn, while the surface of the other horn was covered with microplacentomes. Numerous villous structures without trophoblastic coating were highlighted after histopathology. The dominant sign was an inflammatory reaction. The findings were consistent with inter-cotyledonal placentitis, which led to adventitial placentation.Diffuse semi-placenta compensates for the inadequate development of placentomes and may occur as a congenital or acquired defect. The outcome depends on its severity: in the worst scenario, pregnancy may not proceed beyond midterm and may be complicated by hydrallantois. In the case under examination, the dimensions of the cotyledons (from 2 to 10 cm) allowed for the natural course of pregnancy.

To assess the effects of dry period (DP) length on metabolic, reproductive, and productive parameters, second- (SP) and third- (TP) parity cows were assigned to a traditional (9 weeks, T) or short (5 weeks, S) DP, obtaining four subgroups: second-parity cows with traditional (SPT = 8) and short (SPS = 8) DP, third-parity cows with traditional (TPT = 8) and short (TPS = 10) DP. Plasma insulin-like growth factor-I (IGF-I) and non-esterified fatty acid (NEFA) levels were assessed from 5 weeks before to 14 weeks after parturition. IGF-I concentrations were affected by parity (P < 0.05) and by the interaction of time and DP length (P < 0.01). NEFA levels were affected only by time (P < 0.01). S DP cows showed a shorter interval between calving and ovarian cyclicity resumption (P < 0.01) and a higher milk yield (P < 0.01) and fat and protein corrected milk (P < 0.01) compared with T DP cows. Decreased milk protein content was found in the SPS group compared to the SPT (P < 0.05) and the TPS (P < 0.05) group. In conclusion, a short DP length does not affect reproductive performances, except for hastening the resumption of ovarian cyclicity. A short DP appears to increase milk production and is associated with higher IGF-I levels both in the prepartum and the postpartum period.

The aim of this study was to evaluate the prevalence of uterine prolapse in cows and assess its effects on survival and subsequent fertility. Of 33,450 calving cows considered retrospectively, 216 (0.6%) developed uterine prolapse. A higher prevalence was found in beef cows (n = 57/5,700 cows, 1%) compared to dairy cows (n = 157/27,750 cows, 0.6%). Treatment consisted of cleaning and replacing the uterus with local administration of antibiotics, and applying a harness for uterine containment. The recovery rate was 81.9% (n = 177), similar in dairy (n = 129; 81.1%) and beef (n = 48; 84.2%) cows. Of the 216 cows with uterine prolapse, 18 (8.3%) died before or immediately after treatment; 21 cows (9.7%) were voluntarily culled for economic reasons (low milk yield, low fertility, insufficient weight gain). All recovered dairy cows were artificially inseminated with semen of proven fertility after a voluntary waiting period of 50 days; the beef cows were naturally mated. Among the 172 inseminated/mated cows, 84.7% (n = 150) became pregnant (83.7% dairy cows, 87.5% beef cows), while 15.2% (n = 27) did not conceive. Recurrence of uterine prolapse at subsequent calvings was recorded in one dairy cow. Based upon the data presented here, treated cows with uterine prolapse showed high chances of survival and conception, and a low risk of recurrence.

The Martina Franca (MF) donkey breed (Equus asinus) primarily inhabits the rural areas surrounding the homonymous municipality, as well as neighboring municipalities in the provinces of Bari and Brindisi, all located in the Apulia region of Italy. The objective of this study was to assess the current phenotype through the evaluation of 27 morphologic measurements. The study was conducted on 73 female and 18 male breeding animals from eight different herds located in central and southern Italy. Statistical analysis was performed in order to demonstrate statistical differences between males and females, as well as sexual dimorphism and uniformity of all measurements in both genders. The results demonstrated that the mean of three parameters used for the evaluation of an MF donkey (height at withers, circumference of thorax, and shin circumference) are highly representative of the population, with minimal dispersion Cv^ = 0.05-0.06 up to a maximum of 10%, as evidenced by relatively low standard deviations across observed measurements in both genders. Regarding sexual dimorphism, a statistically significant difference was found between males and females. This dimorphism is linked to reproductive activity and is useful during gestation. Overall, our findings suggest that the MF donkey phenotype has been largely preserved over time with high uniformity in males, slight inhomogeneity in the female population, and less variability in both genders.

Good sperm production is a notable requirement for subjects intended for reproduction, particularly in endangered species, and it has been demonstrated that in horse stallions, this is correlated to testicular volume. The present study, which involved Martina Franca jacks, aimed to determine whether, also in this endangered breed, there is a correlation between the total sperm number (TSN) and testicular volume. Testes were measured with both ultrasound and a caliper. Testicular volume was calculated using two different formulas: one representing the volume of an ellipsoid and one developed to describe round-shaped testicles. The average sperm concentration was 380.14 ± 254.58 × 106/mL, while the average TSN was 16.34 ± 7.76 × 109. Our findings evidenced a significant correlation (r > 0.75; p < 0.05) only between sperm production and the volume calculated with the formula V (cm3) = 33.57 × H - 56.57 for round-shaped testes. Moreover, significance was evidenced only for data obtained with ultrasound (VTs-us 315.03 ± 25.83 cm3) but not with caliper. In conclusion, testicular volume can be suggestive of good fertility in Martina Franca jacks; thus, this parameter could be considered when selecting breeding animals.

Fatty acid-based lipidomic analysis has been widely used to evaluate health status in human medicine as well as in the veterinary field. In equine species, there has been a developing interest in fertility and sperm quality. Fatty acids, being the principal components of the membranes, play an active role in the regulation of the metabolic activities, and their role on spermiogenesis seems to be of great importance for the resulting quality of the sperm and, thus, fertility. With the application of widely used lipidomic techniques, the aim of this study was to evaluate: (a) the fatty acid content of the spermatozoa's membranes of 26 healthy male Martina Franca donkeys and its possible correlation with sperm parameters, and (b) the evaluation of the composition of the red blood cells' membrane. PUFA omega-6 are the principal components (40.38%) of the total PUFA content (47.79%) in both types of cells; however, DPA is the predominant one on the spermatozoa's membrane (27.57%) but is not present in the erythrocyte's membrane. Spermatozoa's motility (%) is positively correlated with stearic acid and EPA, and progressive motility (%), with oleic acid. These findings offer information on the composition of both types of cells' membranes in healthy male MF donkeys and reflect the metabolic transformations of the spermatozoa's membrane during the maturation period, providing a better perception of the role of fatty acids in sperm parameters and fertility.

Castration is one of the most frequently performed surgical procedures in livestock. All methods of castration are associated with a significant degree of pain, affecting the patients' welfare. Limited information on species-specific protocols in buffaloes is available. Considering the importance of this species and the scarcity of reports available, the objective of this study was to evaluate the effects of two different intramuscular doses of xylazine for premedication in water buffalo calves undergoing isoflurane maintenance anaesthesia for routine castration surgery. Twenty clinically healthy male water buffaloes undergoing bilateral orchiectomy were randomly assigned to two groups of 10 animals each: Group 1 was premedicated with xylazine 0.1 mg/kg bw i.m. and Group 2 was premedicated with xylazine 0.15 mg/kg bw i.m. Anaesthesia was induced by ketamine (2 mg/kg bw i.v.) and maintained with isoflurane-oxygen-air in both groups. The HR, RR, presence/absence of ataxia, sedation, depth of anaesthesia, muscle relaxation status, response to acoustic and tactile stimuli, eyelid reflex, extent of salivation and stimulus were evaluated every 5 min. Both protocols provided consistent sedation and were safe. Faster and more satisfactory sedation was recorded when xylazine was administered at 0.15 mg/kg bw i.m., leading us to recommend this dose for buffalo orchiectomy.

This study investigated the postpartum (PP) uterine involution in nine multiparous Martina Franca jennies with at term, normal, and singleton foaling. Transrectal ultrasonography performed at Days 1, 3, 7, 14, 21, and 28 showed that the uterine tip diameters did not differ between the post-pregnant (PPH) and non-post-pregnant uterine horns (NPPH), whereas the diameter of the middle PPH was larger than the NPPH until Day 7 (p < 0.05). The diameter of the corpora-cornual junction resulted larger in the PPH than NPPH at Day 7 (p < 0.05). At Day 3, the microcaruncolae were not detected. Endometrial glands (GL) number increased, with the highest value on Day 28. Endometrial GL area and perimeter decreased (p < 0.001) from Day 1 to Day 28. Epithelial thickness increased from Days 1-3 to Day 7 (p < 0.001), and concurrently with the foal heat to Day 14 (p < 0.001), with a decrease at Days 21-28. A marked neutrophils reduction on Day 7 and eosinophil increase from the first three days to Days 7-14 was observed. The results suggest that, in Martina Franca jennies with normal foaling, the PP uterine involution can be considered complete on Day 14.

The Martina Franca donkey (MFd) is one of the largest Italian donkey breeds, considered as endangered breed. To support the conservation strategies, knowledge about the physiologic hematological parameters of MFds is needed. The aims of the study were to determine reference value for hematological and major serum parameters in a population of healthy MFds and to estimate the influence of age on these parameters. Eighty-one clinically healthy MFds (17 males and 64 females) in different ages were enrolled: group A (foals, n° 16, animals < 1 year old) group B (young animals, n° 36, from 1 to 3 years old), and group C (adult animals, n° 29, over 3 years old). Red blood cell count (RBC); hematocrit value (HCT); hemoglobin concentration (HGB); mean corpuscular volume (MCV); mean corpuscular hemoglobin (MCH); hemoglobin concentration distribution width (HDW); RBC distribution width (RDW); total white blood cell (WBC); WBC differential count for neutrophils, lymphocytes, monocytes, eosinophils and basophils, and platelets (PLT); mean platelet volume (MPV); platelet volume distribution width; and plateletcrit (PCT) were analyzed. The biochemistry panel included aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), gamma glutamyl transferase (GGT), total serum protein (TP), albumin (ALB), cholesterol (CHOL), triglyceride (TGL), blood urea nitrogen (BUN), creatinine (CREA), glucose (GLU), Ca2+, phosphorus, Na+, Cl-, K+, and Mg2+. The effect of age on hematological parameters was investigated using one-way ANOVA test. Age of donkeys does not influence total WBC, HGB, HCT, platelet count and MPV, and PCT (P > 0.05). Some leukocyte populations such as eosinophils, monocytes, and basophils showed age-linked variations (P < 0.05). RBC count, RDW, and HDW decrease with age whereas MCV and MCH increase. Na+, K+, Cl-, Ca2+, phosphorus, ALP, GGT, CREA, GLUC, and CHOL decrease with age (P < 0.05), while AST and TP showed an increase with aging (P < 0.05). ALB reaches the lowest values in young donkeys and returns to values of foals in older animals (P < 0.05). Finally, a difference among groups for BUN and TGL was not found (P < 0.05). The results suggest how even for the MFd breed, age is a variable that affects different hematological and biochemical parameters. Compared to other donkey and horses, the MFd breed showed some differences that clinicians involved during conservation strategies need to be consider.

Mules are hybrids bred from the mating of a jack donkey and a horse mare, known for their strength and resistance and still used to work in agriculture. Although they have been for long considered sterile, evidence of estrus cycle has been demonstrated together with abnormal behavior related to ovarian activity. In this study, a bilateral standing laparoscopic ovariectomy technique using the LigaSure technology was applied in 10 mare mules for treating unwanted behavioral patterns. The technique was effectively performed on these animals avoiding the risk of general anesthesia, and the use of the LigaSure technology allowed good hemostasis and reduced surgical time. Owners declared to be satisfied with the resolution of the behavior.

Eight Martina Franca pregnant jennies were selected in order to evaluate the transfer of colostral antibodies against equine herpesvirus type 1 in their relative foals after immunization with a commercial inactivated vaccine, compared with an unvaccinated group. Samples of serum and colostrums/milk were collected from jennies and foals under study starting from 10 min before and up to 21 days after the foaling. Specific anti-EHV-1 antibody titers were evaluated by means of a serum neutralization test, and the results obtained from both groups were analyzed. The serological titers in the vaccinated jennies was significantly higher (p < 0.01). No significant differences were found in the specific time-point intervals in both groups examined (p > 0.05). The antibody titers in milk at the time of delivery and subsequent withdrawal (T0 and T1) were very high in both groups, but no significant differences were found between the two groups (p > 0.05). In the foal sera, a significant difference was found between foals in the vaccinated group compared with those in the unvaccinated group (p < 0.05). Finally, a significant correlation (p < 0.05) was observed between the antibody titers found in serum and colostrum of jennies and the foal titers in the first time-point sampling (up to 12 h after foaling). The results confirm a substantial homology in the antibody production compared with other most investigated equids, highlighting the efficacy of the vaccination against EHV-1 of the jennies to ensure the protective immunity to their foals during the first weeks after delivery.

Introduction: Persistent Müllerian duct syndrome (PMDS), or uterus masculinus, is a rare autosomal recessive form of male pseudohermaphroditism due to the failure of paracrine anti-Müllerian hormone (AMH) secretion by Sertoli cells or failure of the Müllerian ducts to respond to AMH secretion. The malignant degeneration of persistent Müllerian remnants is rare. In human medicine, few related reports exist. In veterinary medicine, this is the first report describing adenocarcinoma of the uterus masculinus involving the prostate in a dog. Clinical history: An 11-year-old, male, neutered Pomeranian dog was referred for computed tomography due to the suspicion of prostatic carcinoma based on ultrasound and cytological examinations. The computed tomography findings were consistent with a uterus masculinus mass with possible prostatic infiltration. Uterus masculinus removal and total prostatectomy were performed; termino-terminal urethral anastomosis was carried out. Dehiscence of the anastomosis was observed 3 days after surgery. The owner declined any further procedures, and the dog was euthanized 5 days after surgery. Histopathological evaluation revealed adenocarcinoma of the uterus masculinus. Conclusion: Adenocarcinoma of the uterus masculinus may occur, suggesting that patients with PMDS should be evaluated for malignant changes of Müllerian remnants.

Orchitis and epididymo-orchitis are inflammatory lesions of the testicle. We herein describe a case of monolateral chronic orchitis which occurred in a Tiro Pesante Rapido (TPR) stallion, born in 2002, with a history of good fertility. The stallion was healthy and asymptomatic although the left testis was found to be smaller as compared with the right one and was hard in consistency. Histopathology examination revealed tubular atrophy and parenchymal sclerosis. Scattered foci of calcification and chronic inflammation, the latter dominated by macrophages and lymphocytes, were also observed. Although lesions were clearly present, the semen was demonstraed to be of good quality. This study highlights the need for periodic clinical and ultrasound evaluation of stallions, in order to preserve their reproductive performance.

An assessment of the natural ochratoxin A (OTA) exposure of seven Martina Franca jennies was carried out by analyzing blood and milk samples collected close to and after delivery. A total of 41 and 34 blood samples were collected from jennies and foals, respectively, and analyzed by ELISA. A total of 33 milk samples were collected from jennies and analyzed by the HPLC/FLD method based on IAC clean-up. Furthermore, 53 feed samples were collected from January to September and analyzed by a reference method (AOAC Official Method No. 2000.03) for OTA content. Feed samples showed OTA levels up to 2.7 ng/g with an incidence of 32%, while the OTA incidence rate in jennies' blood samples was 73%, with a median value of 97 ng/L and concentrations ranging from

In avian species, cryopreservation of semen is necessary for developing sperm cryobanks. It is very difficult, however to cryopreserve turkey sperm and have sperm be viable after thawing. Glycerol, the commonly used sperm cryoprotectant in many species, is toxic to sperm of avian species. The aim of this study was to evaluate whether the non-permeating dextran was effective for the cryopreservation and maintenance of turkey spermatozoa viability after thawing, avoiding the use of permeating cryoprotectants. Turkey sperm were diluted with a medium supplemented with 11% glycerol or dextran with a 1,000 molecular weight (MW), dextran with a 10,000 MW, or dextran with a 20,000 MW each at a 2%, 5%, or 10% concentration. Sperm kinetic characteristics, membrane and acrosome integrity (AI), and the capacity of spermatozoa to interact with the autologous perivitelline layer were evaluated after equilibration and cryopreservation. Results indicate that with use of glycerol and the 1,000 MW dextran there was lesser sperm viability after both equilibration and cryopreservation, compared with use of the 10,000 or 20,000 MW dextran compounds. There was a greater cryoprotective effect with the 10,000 and 20,000 MW dextran compounds at the 10% concentration with spermatozoa maintaining a greater functionality and capacity to interact with the autologous perivitelline layer. In conclusion, the results of this study indicate turkey spermatozoa could be effectively cryopreserved in extender without the use of glycerol as a penetrating cryoprotectant but with the use of the 10,000 or 20,000 MW dextran compounds at a 10% extender concentration.

The aim of this study was to report prevalences and causes of dystocias in dairy and beef cattle, in primiparous and multiparous cows, as well as the mortality rate of calves and cows, obtained after 11 years of records across various farms in Italy. On a total of 14,575 records from dairy Italian Friesian cows, beef Romagnola and Marchigiana cows, a prevalence of 5.6% was observed, with a significant higher prevalence in primiparous (p < 0.0001), and dairy cows (p < 0.0001). Dystocias of fetal origin were higher than the ones of maternal origin (p < 0.0001). Dystocia management, performed with manual correction in 96% of the cases, was associated with the 25% of calf mortality and the 11% of maternal mortality. When the combined effects of attitude and parity were assessed in relation to each fetal or maternal dystocia cause, dystocia resolution method and on calf, cow and calf-and-cow mortality, results showed a stronger association of dairy primiparous and multiparous cows than beef cows to several dystocia causes and calf-and-cow mortality. Taken together the results from the present study highlighted, once more, the importance of a correct breeding herd management and genetic selection programmes, especially in dairy cows, as well as the prompt diagnosis and correction of difficult calvings, for the effective management of dystocias aimed to reduce calf mortality.

The availability of biochemical blood data specific for the pregnant condition are essential for the correct management of both normal pregnancies and also for the prompt recognition of every abnormality. Because of the lacking knowledge about biochemical blood analysis in the donkey along the entire pregnancy, the study was designed to provide first preliminary data about the values and possible changes of blood alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), alkaline phosphatase (ALP), creatine-kinase (CK), blood urea nitrogen (BUN), creatinine (CREA), uric acid (UA), amylase (AMY), gamma-glutamyl transferase (γ-GT), triglycerides (TRI), cholesterol (CHOL), total protein (TP), albumin (ALB), glucose (GLU), phosphorus (P), calcium (Ca), occurring from the beginning of pregnancy until parturition. The study was performed on 10 Martina Franca healthy jennies with normal pregnancy course and giving birth to mature, healthy and viable foals. Blood samples were collected monthly from the 1st to the 6th month of pregnancy, then twice a month from the 6th to the 9th month and afterwards weekly until parturition. The results showed a significant slight increase of glucose and creatinine in the second quarter of pregnancy and a minor decrease of cholesterol near to parturition, while all the other parameters did not significantly change along pregnancy.

Equine cutaneous habronemosis, also known as "summer sores", is a parasitic infection caused by larvae of Habronema microstoma and Habronema muscae (Nematoda, Spirurida) released by dung-inhabiting fly vectors on abraded skin, skin wounds or muco-cutaneous transition sites. Larvae induce a local inflammatory reaction characterised by itching, granulomatous, ulcerated and, often non-healing, lesions. The diagnosis of summer sores may be unreliable mainly because of the limits of clinical and microscopic examination. The applicability of a semi-nested PCR assay developed for the diagnosis of gastric habronemosis has been herein demonstrated for the detection of the cutaneous infection. The potential applicability of this diagnostic tool may have for clinical and epidemiological studies of cutaneous habronemosis in equids is discussed.

The chamois of Abruzzi (Rupicapra pyrenaica ornata) has been classified as endangered by the World Union for Conservation. The objective of this study was to analyze seasonal differences in the characteristics of various male reproductive organs and in semen quality. The study was conduced during 2004 in the reserve of Lama dei Peligni (Italy) on three chamois males aged between 2 and 5 years. Males were captured during March-May months and October-December months. Various testicular and scrotal measurements were taken and semen was collected using an electroejaculator. Sperm motility pattern was evaluated using computer assisted sperm analyzer, membrane integrity using differential staining and morphology with phase contrast microscopy. Testicular size, sperm motility membrane integrity and the percent of morphological normal spermatozoa were greater during October-December. The authors concluded that sperm characteristics are strongly influenced by season and that semen collected in this period (breeding season) has suitable quality for manipulation and long-term preservation.

OBJECTIVE: To determine survival rate, complications, and short-term fertility rate after fetotomy in mares. DESIGN: Retrospective study. ANIMALS: 72 mares with severe dystocia. PROCEDURES: Records from 1991 to 2005 were searched for mares with dystocia in which a fetotomy was performed. Data relating to presentation and position of foals; survival rate, complications, and short-term fertility rate in mares; and 45-day pregnancy rate in mares bred 2 to 3 months after fetotomy were recorded. RESULTS: Anterior fetal presentation was detected for 54 of 72 (75%) mares, posterior presentation was detected for 13 (18.1%), and transverse presentation was detected for 5 (6.9%). One fetus in anterior presentation was hydrocephalic. Survival rate after fetotomy was 95.8%. Complications included retained fetal membranes (5.5%), laminitis (6.9%), vaginal and cervical lacerations (2.8%), and delayed uterine involution (2.8%). Mares bred 2 to 3 months after fetotomy had good short-term fertility, with a mean pregnancy rate of 79.4% at 45 days after breeding. CONCLUSIONS AND CLINICAL RELEVANCE: The survival rate was high, compared with rates reported after cesarean section, and short-term fertility rate was similar to those reported for mares that had a controlled vaginal delivery or cesarean section. Fetotomy performed by a skilled veterinarian on a nonviable fetus should be considered as a means of quick and safe correction of dystocia that does not necessarily impair short-term fertility in affected mares.
