Fish germ cell transplantation presents several important potential applications for aquaculture, including the preservation of germplasm from endangered fish species with high genetic and commercial values. Using this technique in studies developed in our laboratory with adult male Nile tilapias (Oreochromis niloticus), all the necessary procedures were successfully established, allowing the production of functional sperm and healthy progeny approximately 2months after allogeneic transplantation. In the present study, we evaluated the viability of the adult Nile tilapia testis to generate sperm after xenogeneic transplant of germ cells from sexually mature Jundia catfish (Rhamdia quelen) that belong to a different taxonomic order. Therefore, in order to investigate at different time-periods post-transplantation, the presence and development of donor PKH26 labeled catfish germ cells were followed in the tilapia seminiferous tubules. From 7 to 20days post-transplantation, only PKH26 labeled spermatogonia were observed, whereas spermatocytes at different stages of development were found at 70days. Germ cell transplantation success and progression of spermatogenesis were indicated by the presence of labeled PKH26 spermatids and sperm on days 90 and 120 post-transplantation, respectively. Confirming the presence of the catfish genetic material in the tilapia testis, all recipient tilapias evaluated (n=8) showed the genetic markers evaluated. Therefore, we demonstrated for the first time that the adult Nile tilapia testis offers the functional conditions for development of spermatogenesis with sperm production from a fish species belonging to a different order, which provides an important new venue for aquaculture advancement.

Alzheimer's disease (AD) is a neurodegenerative disorder in which there is a decline of cholinergic function. The symptomatic AD treatment involves the use of ChEIs (cholinesterase inhibitors) as rivastigimine, a dual inhibitor. The human butyrylcholinesterase (BChE) is an enzyme that has specific roles in cholinergic neurotransmission and it has been associated with AD. In the serum, BChE is found in four main molecular forms: G1 (monomer); G1-ALB (monomer linked to albumin); G2 (dimer); and G4 (tetramer). The interaction between the products of BCHE gene and CHE2 locus results in CHE2 C5+ and CHE2 C5- phenotypes. CHE2 C5+ phenotype and BChE-K are factors that influence on BChE activity. This work aimed to verify the proportions of BChE molecular forms, total and relative activity in 139 AD patients and 139 elderly controls, taking into account K variant, CHE2 locus, rivastigmine treatment and clinical dementia rating (CDR) of AD patients. Phenotypic frequencies of CHE2 C5+ and frequency of the carriers of the K allele were similar between groups. Total BChE activity in plasma was significantly lower in AD patients than in elderly controls. Furthermore, we found that reduction on plasma BChE activity is associated directly with AD progression in AD patients and that rivastigmine treatment has a stronger effect on BChE activity within the CDR2 group. The reduction in BChE activity did not occur proportionally in all molecular forms. Multiple regression analysis results confirmed that AD acts as the main factor in plasma BChE activity reduction and that severe stages are related with an even greater reduction. These findings suggest that the reduction of total plasma BChE and relative BChE molecular forms activity in AD patients is probably associated with a feedback mechanism and provides a future perspective of using this enzyme as a possible plasmatic secondary marker for AD.

Aspects concerned with morphogenesis of Gallus gallus domesticus, avail studies related to the action of the insulin in the topography and embryonic structures. At the temperature of 37.5 degrees C, eggs were incubated during 24 h, injected with 5 ml of swine insulin in three concentrations and reincubated for more 72 h. The morphological characteristics of 80 embryos were evaluated and, according to the presented organization, classified in 5 morphogenetic levels. It was registered generalized dysmorphism (4th level) in 21 embryos that went through the tests with insulin. Standard morphogenesis (1st level) and located dysmorphism (3rd level) were verified among those from the control experiments. Those individuals concerned with the 4th level, showed reduced dimension of the body and were characterized by anterior-dorsal limits organized in a cephalic projection, and also presented alterations in the posterior-ventral region. These features evidence a pattern of abnormality in the determination of the cephalic-caudal axis and indicate a specific action of the insulin in the embryonic morphogenesis, in the period of 96 hours of incubation.

Aspectos concernentes à morfogênese de Gallus gallus domesticus viabilizam estudos relacionados à ação da insulina sobre estruturas e topografia embrionárias. Na temperatura de 37,5ºC, ovos foram incubados por 24 h, injetados com 5 ml de insulina de suínos em 3 concentrações e reincubados por mais 72 h. As características morfológicas de 80 embriões foram avaliadas e, de acordo com a organização apresentada, classificados em 5 níveis de morfogênese. Em 21 embriões submetidos aos testes com a insulina registrou-se dismorfismo generalizado (4o nível), enquanto nos de experimento de controle foi verificado morfogênese-padrão (1o nível) e dismorfismo localizado (3o nível). Aqueles espécimes mostraram corpo com dimensões reduzidas, caracterizado por limites ântero-dorsal organizados em uma projeção cefálica e regiões posterior-ventral alteradas, evidenciando um padrão de anormalidades na determinação do eixo ântero-posterior, que indica a ação específica da insulina na morfogênese embrionária no período de 96 horas de incubação.

Objetivando avaliar o desenvolvimento embrionário de Gallus gallus domesticus, foram incubados ovos nas temperaturas de 34,0ºC, 37,5ºC e 40,0ºC, nos períodos de 48, 72 e 96 horas. Avaliaram-se em 81 embriöes o comprimento cefalocaudal, o peso total e 6 características morfológicas, utilizando-se um protocolo referindo o desenvolvimento como padräo, acelerado ou defasado. Os dados relativos ao nível de diferenciaçäo das estruturas embrionárias demonstram que os fatores temperatura e tempo de incubaçäo interagem nos processos de morfogênese
