This study aims to determine the phenological characteristics of thermal responses in the larvae of a Culex pipiens complex field population at the individual level under the influence of thermal regime of its habitat. The analysis is based on a structured population model quantifying the thermal responses of development time and survival under variable conditions and characterising the variety between the larvae (interindividual variety). During the study performed in Turkish Thrace on a monthly basis between May 2021 and June 2022, a total of 3744 larvae were reared as peer larval cohorts and 2330 larvae as siblings in artificial containers to be fully exposed to the natural thermal condition that was recorded hourly. The development process of larvae was monitored daily from egg to adult. As a result, a total of 4788 adult mosquitoes emerged, with a development period ranging from 8 to 52 days in the females and 7 to 50 days in the males, and the survival rate was found to range from 0% to 100%. Both parameters varied by month and individuals, and the variations manifested itself, particularly in the colder periods. The results indicate that the variation between the individuals in terms of thermal response in the larvae of C. pipiens, along with the thermal acclimation ability, appears to be fate determinant in resisting fluctuating thermal regimes, surviving in concurrent climate change and adapting to new conditions with modifications in the seasonal phenology, such as maintaining reproductive dynamics throughout the winter thanks to global warming.

Endoparasitoid species devoid of symbiotic viruses inject secretions derived from their reproductive glands into their hosts during parasitism in order to avoid various immune responses of their hosts. Pimpla turionellae L. (Hymenoptera: Ichneumonidae) is an endoparasitoid that lacks polydnaviruses, and its venom has previously been shown to paralyze the host Galleria mellonella (Lepidoptera: Pyralidae) and suppress its immune reactions to ensure the egg survival. The present study demonstrates that another female-injected factor calyx fluid extracted from the P. turionellae ovary is also responsible for the suppression of G. mellonella immunity. The total hemocyte counts of G. mellonella decrease after treatment with calyx fluid in a concentration-dependent manner. Significant reductions in cell viability are also observed at all calyx fluid doses both in vivo and in vitro. The analyses of the beads injected into the insects as encapsulation targets revealed that the number of encapsulated beads reduced significantly compared to controls post-calyx fluid injection. The injection of the highest calyx fluid dose (1 female equivalent calyx) is sufficient to completely inhibit the strong encapsulation and melanization reactions of the last instar larvae 24 h post-injection. These results demonstrate that P. turionellae calyx fluid is required to regulate host immunity for successful parasitization.

The greater wax moth, Galleria mellonella, is one of the most ruinous pests of honeycomb in the world. Beta-glucosidases are a type of digestive enzymes that hydrolytically catalyzes the beta-glycosidic linkage of glycosides. Characterization of the beta-glucosidase in G. mellonella could be a significant stage for a better comprehending of its role and establishing a safe and effective control procedure primarily against G. mellonella and also some other insect pests. Laboratory reared final instar stage larvae were randomly selected and homogenized for beta-glucosidase activity assay and subsequent analysis. The enzyme was purified to apparent homogeneity by salting out with ammonium sulfate and using sepharose-4B-l-tyrosine-1-naphthylamine hydrophobic interaction chromatography. The purification was 58-fold with an overall enzyme yield of 29%. The molecular mass of the protein was estimated as ca. 42 kDa. The purified beta-glucosidase was effectively active on para/ortho-nitrophenyl-beta-d-glucopyranosides (p-/o-NPG) with Km values of 0.37 and 1.9 mM and Vmax values of 625 and 189 U/mg, respectively. It also exhibits different levels of activity against para-nitrophenyl-ß-d-fucopyranoside (p-NPF), para/ortho-nitrophenyl ß-d-galactopyranosides (p-/o-NPGal) and p-nitrophenyl 1-thio-ß-d-glucopyranoside. The enzyme was competitively inhibited by beta-gluconolactone and also was very tolerant to glucose against p-NPG as substrate. The Ki and IC50 values of δ-gluconolactone were determined as 0.021 and 0.08 mM while the enzyme was more tolerant to glucose inhibition with IC50 value of 213.13 mM for p-NPG.

Prophenoloxidase (PPO) was purified from Galleria mellonella L. A 67-fold purification of the proenzyme with 352% yield was achieved by using a Sepharose 4B-L-tyrosine-p-amino benzoic acid affinity column. The purified enzyme was migrated as a single band on SDS-polyacrylamide gel electrophoresis. K(m) and V(max) values were 0.017 M and 1430.45 EU for catechol. Inhibition of PPO was investigated with inhibitors such as p-aminobenzoic acid, etyleneglycol, and ascorbic acid. Among them, ascorbic acid showed the strongest inhibitory activity with IC(50) value of 2.94 µM. The current paper represents new strategies for the biological control of the Galleria mellonella L. insect.
