Ambient GABA modulates firing patterns in adult neural circuits by tonically activating extrasynaptic GABA(A) receptors. Here, we demonstrate that during a developmental period when activation of GABA(A) receptors causes membrane depolarization, tonic activation of GABA(A) receptors blocks all spontaneous activity recorded in retinal ganglion cells (RGCs) and starburst amacrine cells (SACs). Bath application of the GABA(A) receptor agonist muscimol blocked spontaneous correlated increases in intracellular calcium concentration and compound postsynaptic currents in RGCs associated with retinal waves. In addition, GABA(A) receptor agonists activated a tonic current in RGCs that significantly reduced their excitability. Using a transgenic mouse in which green fluorescent protein is expressed under the metabotropic glutamate receptor subtype 2 promoter to target recordings from SACs, we found that GABA(A) receptor agonists blocked compound postsynaptic currents and also activated a tonic current. GABA(A) receptor antagonists reduced the holding current in SACs but not RGCs, indicating that ambient levels of GABA tonically activate GABA(A) receptors in SACs. GABA(A) receptor antagonists did not block retinal waves but did alter the frequency and correlation structure of spontaneous RGC firing. Interestingly, the drug aminophylline, a general adenosine receptor antagonist used to block retinal waves, induced a tonic GABA(A) receptor antagonist-sensitive current in outside-out patches excised from RGCs, indicating that aminophylline exerts its action on retinal waves by direct activation of GABA(A) receptors. These findings have implications for how various neuroactive drugs and neurohormones known to modulate extrasynaptic GABA(A) receptors may influence spontaneous firing patterns that are critical for the establishment of adult neural circuits.

PURPOSE: To describe multicolor scanning laser imaging (MSLI) and conventional fundus photography findings in lipemia retinalis (LR). METHODS: We report two LR cases. The first case is a 47-year-old diabetic woman with LR secondary to familial hypertriglyceridemia examined with MSLI, conventional fundus photography, and optical coherence tomography (OCT). The second case is a 39-year-old diabetic man with hypertriglyceridemia and LR. He was followed over time with conventional fundus photography of his retina until metabolic control was achieved. RESULTS: In the first case, MSLI showed retinal arteries with an intense yellow and retinal veins with a pale yellow color. Fundus photography disclosed only mild pale red vessels. Optical coherence tomography detected macular edema and hyperreflective changes of retinal vessels. In the second case, fundus photography showed vessels with a pale red appearance that normalized after metabolic control of triglycerides. CONCLUSION: We described the retinal findings in patients with LR using different image modalities. Specifically, we report the findings with MSLI not previously described. While regular fundus photography may show mild changes in LR, the MSLI modality may show more significant findings assisting in the diagnosis and follow-up of this disease.

PURPOSE: Implantation of intraocular devices may become critical as they decrease in size in the future. Therefore, it is desirable to evaluate the relationship between radial location and Schwalbe's line (smooth zone) by examining its width with scanning electron microscopy (SEM) and to correlate this with observations by optical coherence tomography (OCT). METHODS: Full corneoscleral rings were obtained from twenty-six formalin-fixed human phakic donor eyes. SEM of each eye yielded a complete montage of the smooth zone, from which the area was measured, and width was determined in each quadrant. In three different eyes, time domain anterior segment OCT (Visante, Carl Zeiss Meditec Inc., Dublin, CA, USA) and spectral domain OCT (Cirrus 4.0, Carl Zeiss Meditec Inc., Dublin, CA, USA) were used to further characterize Schwalbe's line. RESULTS: The overall smooth zone width was 79±22 µm, (n=15) ranging from 43 to 115 µm. The superior quadrant (103±8 µm, n=19), demonstrated significantly wider smooth zone than both the nasal (71±5 µm, n=19, P<0.001), and inferior (64±5 µm, n=18, P<0.0001) quadrants but not the temporal quadrant (81±7 µm, n=17, P>0.05). SEM findings of the smooth zone were correlated with visualization of Schwalbe's line by Cirrus and Visante OCT imaging. CONCLUSION: The smooth zone appears widest superiorly and thinnest inferonasally, suggesting that as glaucoma surgical devices become smaller, their placement could be argeted clinically by using OCT with preference to the superior quadrant, to minimize damage to the corneal endothelium.
