BACKGROUND: The prevalence of unprotected anal intercourse (UAI) is high among Chinese men who have sex with men (MSM). As important aspects of quality of life (QOL), psychological health and social relationships have been found to be associated with UAI among MSM, which was of great concern for intervening on UAI. METHODS: We conducted a cross-sectional study in Zhengzhou, Henan province, and in Huludao, Liaoning province, China, to measure quality of life (QOL) using the brief version of the World Health Organization Quality of Life (WHOQOL-BREF). Cronbach's α coefficient was used to test the internal consistency of the questionnaire items, and construction validity was assessed by exploratory factor analysis. T-test, chi-square test and multivariate logistic analysis were carried out to test the relationship between quality of life and unprotected anal intercourse. RESULTS: The questionnaires of 320 of the 373 men who have sex with men that were surveyed met our validity criteria (response rate: 85.8 %). A total of 161 subjects (50.3 %) reported having unprotected anal sex in the last 6 months. The results of univariate analysis indicated that having unprotected anal sex was significantly associated with psychological health (PSYCH), social relationships (SOCIL), age, and marital status (P-value <0.05). The 3 items ("Negative feelings", "Hopeness on your life", and "Be able to concentrate") in the PSYCH subscale were associated with UAI significantly (P-value <0.05). Multivariate analysis showed that psychological health (adjusted odd ratio (AOR) 0.979, 95 % confidence interval (CI) 0.961-0.998) was independent factor affecting the likelihood of having UAI in the population of Chinese MSM, and participants who aged 45 or more had higher odds of UAI (AOR 3.986, 95 % CI 1.199-13.255). CONCLUSIONS: WHOQOL-BREF was acceptable for evaluating the quality of life of MSM. Psychological health, as one important aspect of quality of life, was negatively associated with unprotected anal intercourse. The finding underscored the needs to incorporate the strategies on improving psychological health into the UAI intervention to reduce the HIV transmission among MSM.

Cellulose material holds considerable promise for effective surface-enhanced Raman scattering (SERS) substrate construction due to its extensive availability, chemically modifying capacity, ease of manufacture, high flexibility and low optical activity. A large-area, high-sensitivity, stable and uniform Au@Ag nanorods (NRs)-CMC substrate was successfully developed via electrostatic repulsion by using negatively-charged core-shell Au@Ag NRs as SERS active plasmonic nanomaterial, combined with negatively-charged carboxymethylcellulose (CMC) hydrogel for nanoparticles stabilization, homodisperse and protection. The obtained Au@Ag NRs-CMC substrate showed excellent sensitivity for the detection of thiram residues in fruits containing low and abundant pigment interferents, such as apples and blueberries, with detection limits of 58 and 78 ppb, respectively. Additionally, it retained more than 80% SERS performance after storage for 9 months under ambient conditions, demonstrating its great potential in facilitating the commercialization of cellulose-based SERS technology for cost-effective detection of food contaminants with advantages of facile preparation procedure, uniformity, reproducibility and long-term stability.

In this study, a high-performance, stable and homogeneous Au@AgNRs/CMC/qPCR flexible film surface-enhanced Raman scattering (SERS) substrate was constructed by synergistically stabilizing and protecting bimetallic core-shell Au@Ag nanorods (Au@AgNRs) with carboxymethylcellulose (CMC) and fluorescent-quantitative-polymerase-chain-reaction (qPCR) film. The network structure of CMC immobilized and aligned Au@AgNRs through coordination of carboxyl groups with surface Ag atoms to provide intensive and stable 'hot spots', and the qPCR bilayer film performed as carrier and barrier to protect Au@AgNRs from oxidation, humidity and optical damage and improved the robustness and stability. The Au@AgNRs/CMC/qPCR film was used for in-situ extraction and SERS detection of thiabendazole on nectarine (0.24 ppm) and lemon (0.27 ppm) with low detection of limits. Furthermore, it retained 98.6% SERS performance after storage for 90 days under ambient conditions, revealing the great potential in promoting the commercialization of the SERS technique for sensitive contaminants sensing with simple fabrication procedures, homogeneity, reproducibility and long-term stability.

Background: Lower respiratory tract infections are the leading cause of morbidity and mortality in children worldwide. Timely and accurate pathogen detection is crucial for proper clinical diagnosis and therapeutic strategies. The low detection efficiency of conventional methods and low specificity using respiratory samples seriously hindered the accurate detection of pathogens. Methods: In this study, we retrospectively enrolled 1,032 children to evaluate the performance of metagenomics next-generation sequencing (mNGS) using bronchoalveolar lavage fluid (BALF) sample and protected bronchial brushing (BB) sample in diagnosing pneumonia in children. In addition, conventional tests (CTs) were also performed. Results: The specificity of BB mNGS [67.3% (95% CI 58.6%-75.9%)] was significantly higher than that of BALF mNGS [38.5% (95% CI 12.0%-64.9%)]. The total coincidence rate of BB mNGS [77.6% (95% CI 74.8%-80.5%)] was slightly higher than that of BALF mNGS [76.5% (95% CI 68.8%-84.1%)] and CTs [38.5% (95% CI 35.2%-41.9%)]. During the epidemics of Mycoplasma pneumoniae, the detection rate of M. pneumoniae in the >6-year group (81.8%) was higher than that in the 3-6-year (78.9%) and <3-year groups (21.5%). The highest detection rates of bacteria, fungi, and viruses were found in the <3-year, >6-year, and 3-6-year groups, respectively. mNGS detection should be performed at the duration of 5-7 days after the start of continuous anti-microbial therapy or at the duration of 6-9 days from onset to mNGS test. Conclusions: This is the first report to evaluate the performance of BB mNGS in diagnosing pulmonary infections in children on a large scale. Based on our findings, extensive application of BB mNGS could be expected.

Introduction: Early and effective application of antimicrobial medication has been evidenced to improve outcomes of patients with bloodstream infection (BSI). However, conventional microbiological tests (CMTs) have a number of limitations that hamper a rapid diagnosis. Methods: We retrospectively collected 162 cases suspected BSI from intensive care unit with blood metagenomics next-generation sequencing (mNGS) results, to comparatively evaluate the diagnostic performance and the clinical impact on antibiotics usage of mNGS. Results and discussion: Results showed that compared with blood culture, mNGS detected a greater number of pathogens, especially for Aspergillus spp, and yielded a significantly higher positive rate. With the final clinical diagnosis as the standard, the sensitivity of mNGS (excluding viruses) was 58.06%, significantly higher than that of blood culture (34.68%, P<0.001). Combing blood mNGS and culture results, the sensitivity improved to 72.58%. Forty-six patients had infected by mixed pathogens, among which Klebsiella pneumoniae and Acinetobacter baumannii contributed most. Compared to monomicrobial, cases with polymicrobial BSI exhibited dramatically higher level of SOFA, AST, hospitalized mortality and 90-day mortality (P<0.05). A total of 101 patients underwent antibiotics adjustment, among which 85 were adjusted according to microbiological results, including 45 cases based on the mNGS results (40 cases escalation and 5 cases de-escalation) and 32 cases on blood culture. Collectively, for patients suspected BSI in critical condition, mNGS results can provide valuable diagnostic information and contribute to the optimizing of antibiotic treatment. Combining conventional tests with mNGS may significantly improve the detection rate for pathogens and optimize antibiotic treatment in critically ill patients with BSI.

Transmembrane protein 16A (TMEM16A) localizes at plasma membrane and controls chloride influx in various type of cells. We here showed an intracellular localization pattern of TMEM16A molecules. In myoblasts, TMEM16A was primarily localized to the cytosolic compartment and partially co-localized with intracellular organelles. The global deletion of TMEM16A led to severe skeletal muscle developmental defect. In vitro observation showed that the proliferation of Tmem16a-/- myoblasts was significantly promoted along with activated ERK1/2 and Cyclin D expression; the myogenic differentiation was impaired accompanied by the enhanced caspase 12/3 activation, implying enhanced endoplasmic reticulum (ER) stress. Interestingly, the bradykinin-induced Ca2+ release from ER calcium store was significantly enhanced after TMEM16A deletion. This suggested a suppressing role of intracellular TMEM16A in ER calcium release whereby regulating the flux of chloride ion across the ER membrane. Our findings reveal a unique location pattern of TMEM16A in undifferentiated myoblasts and its role in myogenesis.

Surface-enhanced Raman spectroscopy based on thioglycolic acid (TGA) functionalized silver-coated gold nanoparticles (Au@Ag-TGANPs) was developed for the facile screening of thiabendazole (TBZ) and ferbam (0.025-10 ppm) in liquid milk for the first time. Results showed that silver-coated gold nanoparticles (Au@AgNPs) with a core size of 32 nm and a shell thickness of 5 nm was successfully modified with 3 nm TGA. The sensitive Au@Ag-TGANPs could enhance TBZ and ferbam signals by factors of 6.4 × 104 and 9.8 × 104, respectively, and achieved the detection of TBZ and ferbam with limits of detection of 0.12 and 0.003 ppm, R2 of 0.988 and 0.9821, percent recoveries of 88-103% and of 87.2-103.5%, and relative standard deviations of 4.1-9.2% and 3.5-8.3%, respectively. The current simple and green method could thus be used to detect other unsafe chemicals in future studies.

A universal surface-enhanced Raman scattering (SERS) substrate with tuneable plasmonic behaviour and long-term stability was synthesized and characterized, and the substrate was applied in the sensitive detection of fungicide malachite green (MG) in fish fillets. Temperature-responsive poly (N-isopropyl acrylamide) (pNIPAM) template and anisotropic Au nanorods (NRs) were synthesized to prepare pNIPAM @ Au NRs nano-hybrids with dynamically optical and SERS properties and high stability. Results showed that excitation wavelength-dependent SERS efficiency could be tailored by temperature-induced swelling and collapse of the nano-hybrids, thus anticipated optical and SERS properties of the composites could be obtained. Furthermore, the SERS performance, uniformity, long-term stability analysis, and MG in fish evaluation showed that the detection of MG in fish tissues was realized with a limit of detection of 1.58 × 10-9 M (0.73 ng/g), and the pNIPAM @ Au NRs composites could serve as effective SERS substrates allowing trace level detection of contaminants for the food industry.

Mixed pesticides are usually applied to agricultural products to enhance agricultural production. However, their presence in the environment and food poses serious health risks to humans. Therefore, rapid and reliable detection methods for mixed pesticides residues are in need to minimize potential health hazards. Herein, a nondestructive and sensitive strategy was developed to determine thiram and thiabendazole (TBZ) mixture on fruit surface using surface-enhanced Raman spectroscopy (SERS) technology coupled with interfacial self-assembly gold nanorods (Au NRs) array substrates together with self-modelling mixture analysis (SMA) method. Firstly, a large area high-density Au NRs array was fabricated by organic-aqueous interfacial self-assembly to serve as a sensitive SERS substrate for simultaneously screening of thiram and TBZ on the fruit surface. Then, an SMA method was employed to identify and separate the Raman spectrum for each pesticide from the Raman spectra of the pesticides mixture on the contaminated fruit surface. Results showed that using SERS technique with the SMA method, qualitative and quantitative analyses of a single component from the spectra of the mixture were simultaneously realized, and the resolved pure spectrum of each pesticide was presented. The limits of detection (LOD) of pesticides on the surface of apple, tomato and pear were 0.041, 0.029 and 0.047 ng/cm2 for thiram, and 0.79, 0.76 and 0.80 ng/cm2 for TBZ, respectively. It was anticipated that the proposed SERS detection approach combined with SMA methods should pave the way for detecting multi-analytes in practical applications for agriculture and food safety inspection.

FHND004 is a newly synthesized epidermal growth factor receptor (EGFR) inhibitor for the treatment of non-small cell lung cancer (NSCLC). The aim of the present study was to investigate the impacts of FHND004 on human ether-à-go-go-related gene (hERG) K+ channels and the molecular mechanisms underlying of its action. Whole-cell patch clamp recording was performed on wild type (WT), mutant hERG channels heterologously expressed in human embryonic kidney (HEK) 293 cells or IKr endogenously expressed in HL-1 cells, respectively. FHND004 inhibited hERG K+ currents in a concentration-dependent manner with IC50 values of 8.46 ± 0.33 µM in HEK293 cells and 7.52 ± 1.27 µM in HL-1 cells, respectively. However, the inhibitory potency of FHND004 on hERG channels was significantly less than its precursor AZD9291. FHND004-induced inhibition was state-dependent with a preference within open state, but did not alter other kinetics including activation, inactivation, and recovery from inactivation or deactivation. In addition, FHND004 exhibited more potent inhibitory effects on WT/A422T and WT/H562P-hERG, two known long QT syndrome (LQTS) associated KCNH2 mutations, than WT alone. Mutations of the residues at pore regions (F656C, Y652A, S624A, and F557L) in hERG channels attenuated block effects of FHND004. Taken together, our results demonstrate the evidence that FHND004 is a less potent hERG blocker than its precursor AZD9291. There is, however, a need for caution in the potential use of FHND004 for treating NSCLC patients, especially in those with other concurrent triggering factors.

A novel enzymatic method for glyceryl monocaffeate (GMC) preparation by the transesterification of ethyl caffeate (EC) was investigated. The effects of reaction variables (reaction pressure, temperature, reaction time, enzyme load, and substrate ratio) on the enzymatic transesterification were studied and optimized using response surface methodology. HPLC-ESI-MS and HPLC-UV were used to monitor the transesterification. Thermodynamics, kinetic analyses and reaction mechanism were also evaluated. Results showed that, GMC can be successfully prepared by the enzymatic transesterification of EC with glycerol. Under the optimal conditions (enzyme load 22.54%, EC:glycerol=1:12.75 (mol/mol), 72.5°C, and 10.5h), EC conversion and GMC yield were 97.9±0.7% and 95.8±1.0%, respectively. The activation energies (Ea) for EC conversion and GMC formation were 44.23 and 46.51kJ/mol, respectively. The kinetic values for Vmax, Km(') and KIA were 2.18×10(-3)mol/(Lmin), 0.086mol/L, and 0.52mol/L, respectively. The transesterification mechanism with EC inhibition was also proposed.

BACKGROUND AND OBJECTIVES: Substantial climate changes have led to the emergence and re-emergence of various infectious diseases worldwide, presenting an imperative need to explore the effects of meteorological factors on serious contagious disease incidences such as that of meningococcal meningitis (MCM). METHODS: The incidences of MCM and meteorology data between 1981 and 2010 were obtained from Chaoyang city. Structure Equation Modeling was used to analyze the relationships between meteorological factors and the incidence of MCM, using the LISREL software. RESULTS: The SEM results showed that Adjusted Goodness of Fit Index (AGFI) = 0.30, Goodness of Fit Index (GFI) = 0.63, and Root Mean Square Error of Approximation (RMSEA) = 0.31. Humidity and temperature both had negative correlations with MCM incidence, with factor loads of -0.32 and -0.43, while sunshine was positively correlated with a factor load of 0.42. For specific observable variables, average air pressure, average evaporation, average air temperature, and average ground temperature exerted stronger influence, with item loads between observable variables and MCM incidence being -0.42, 0.34, -0.32, and -0.32 respectively. CONCLUSION: Public health institutions should pay more attention to the meteorological variables of humidity, sunshine, and temperature in prospective MCM control and prevention.

Feruloylated acylglycerols (FAGs) are the lipophilic derivatives of ferulic acid. In this work, soybean oil (SBO) and the mixed fatty acids (MFA) were selected as fatty acyl donors, and reacted with glyceryl monoferulate (GMF) to prepare FAGs in ionic liquids (ILs). Effect of various reaction parameters (time, temperature, enzyme concentration, and substrate ratio) and ILs on the GMF conversion and the reaction selectivity for FAGs formation were investigated. Response surface methodology (RSM) based on a 3-level-4-factor Box-Behnken experimental design was employed to evaluate the inactive effect of reaction parameters. For the esterification of GMF with MFA, the maximum GMF conversion (98.9 ± 0.9%) and FAG yield (88.9 ± 0.6%) were achieved in [C10mim]PF6. However, for the transesterification of GMF with SBO, the maximum GMF conversion (94.3 ± 0.7%) and FAG yield (83.8 ± 1.0%) were obtained in [C12mim]PF6. High FAG selectivities (∼0.90) were also obtained using SBO or MFA as acyl donors.

The quality of life (QOL) of men who have sex with men (MSM) has received increasing attention in recent years. Our study surveyed the QOL and explored the influence of social support on QOL in Chinese MSM. A cross-sectional survey was conducted from September 2013 to March 2014 of 438 MSM in Huludao and Zhengzhou City, China. The results of univariate analysis showed that higher QOL scores were associated with receiving psychosocial counseling, higher health education, younger age and marital status of being single or unmarried p < 0.05). The structural equation model fitted well, with χ2 = 2083.47 (p < 0.05), RMSEA = 0.07, and GFI = 0.88. Among the latent factors, social support, with a factor load of 0.47, had greater impact on QOL than demographic characteristics. Within social support, the item loads for psychosocial counseling and health education were 0.17 and 0.29, respectively. Basic demographic characteristics also influenced social support, with a factor load of -0.65. For demographic characteristics, the greatest item loads were for marital status and age (0.77 and 0.71, respectively). These findings suggest that strengthening social support, especially for older and married individuals, would improve QOL in MSM in China.

OBJECTIVE: The aim of the study was to assess the psychometric properties of the 36-Item Short Form Health Survey (SF-36) in the men who have sex with men (MSM) population in China. METHODS: A cross-sectional survey was conducted among 373 MSM from September to December, 2012, in Zhengzhou and Huludao City, China. Internal reliability of the questionnaire was calculated by Cronbach's α coefficient. Validity was analyzed through construct validity, divisional validity, and collective validity testing. RESULTS: The overall Cronbach's α coefficient of the SF-36 questionnaire was 0.943, while the Cronbach's α coefficients for each of the dimensions were all > 0.70. Results showed that the SF-36 questionnaire was reliable and valid. CONCLUSIONS: This study provided evidence that the SF-36 is an acceptable, valid and reliable instrument in evaluating the quality of life of MSM in Mainland China.
