Endemic medicinal plants deserve immediate research priorities as they typically show a limited distribution range, represent few and fragmented populations in the wild and are currently facing anthropogenic threats like overharvesting and habitat degradation. One of the important aspects of ensuring their successful conservation and sustainable utilization lies in comprehending the fundamental seed biology, particularly the dormancy status and seed germination requirements of these plants. Here, we studied the seed eco-physiology and regeneration potential of Swertia thomsonii-an endemic medicinal plant of western Himalaya. We investigated the effect of different pre-sowing treatments, sowing media and sowing depth on seed germination parameters of S. thomsonii. Seeds of S. thomsonii exhibit morphophysiological dormancy (MPD), i.e. when the embryo of the seed is morphologically and/or physiologically immature. Wet stratification at 4 °C for 20 days, pre-sowing treatment with 50 ppm GA3 and pre-sowing treatment with 50 ppm KNO3 were found ideal for overcoming dormancy and enhancing the seed germination of S. thomsonii. Furthermore, seed germination and seedling survival were significantly influenced by pre-sowing treatments, sowing media and sowing depth. The percentage of seed germination and seedling survival got enhanced up to 84-86% and 73-75% respectively when seeds were pre-treated with GA3 or KNO3 and then sown in cocopeat + perlite (1:1) at a depth of 1 cm. The information obtained in the present study outlines an efficient protocol for large-scale cultivation of S. thomsonii thereby limiting the pressure of overexploitation from its natural habitats and may also help in the restoration and conservation of this valuable plant species.

Phytochemicals are broadly acknowledged for their health-promoting effects owing to the fact of their capacity to counteract free radicals (e.g., superoxide anion radical, hydroxyl radical, hydroperoxyl radical, singlet oxygen, hypochlorite, and nitric oxide) and shield against oxidative stress induced by environmental factors. This study aimed to investigate the relationship between altitude, morphology, soil parameters, in vitro antioxidant potential and phytochemical composition of Phlomis cashmeriana collected from four different locations of Kashmir Himalaya characterized by diverse habitats and elevations. Various factors, such as extraction method, solvent polarity, and habitat conditions, can impact the quantity and efficacy of phytochemicals in plants. The aim of current study was to analyze phytochemical composition and antioxidant activity of P. cashmeriana, an important medicinal plant found in the Kashmir Himalaya region. The antioxidant activity was accessed using several assays and the plant populations were selected based on their diverse habitat features and altitudes. HR-LCMS was conducted for both below-ground and above-ground parts. Some important compounds such as, catechin, vinainsenoside, acutilobin, and kaempferol were reported for the first time from P. cashmeriana. Results showed that methanol was the most efficient solvent for extracting phytochemicals. During the current study, it was also found that the below-ground parts exhibited superior antioxidant activity compared to the above-ground parts. Notably, Site IV demonstrated the highest antioxidant potential; a positive correlation between altitude and antioxidant activity was also found. In conclusion, present research identified specific elite populations having highest antioxidant potential and are well-suited for large-scale cultivation of P. cashmeriana.

The ethnic communities living near protected areas are critically dependent on medicinal plants for their healthcare. Although several ethnomedicinal studies are available from the Himalaya, quantitative studies focusing on protected areas in this region are largely overlooked. In this study, we aimed to document the ethnomedicinal plants of the Overa-Aru wildlife sanctuary in Kashmir Himalaya. We conducted a systematic field survey in the study area during 2020 - 2021 to collect primary data from 110 participants through semi-structured interviews and questionnaires. The data was analyzed using quantitative indices and Pearson's correlation. We recorded 64 plant species used against 8 human disease categories, including some first-time reports from the region. Asteraceae and Lamiaceae were the most dominant families and herbs were the dominant growth form. The most common plant part used were leaves, and the main method of preparation was decoction. Plant species with the highest relative frequency of citation was Achillea millifolium (0.83). Informant consensus factor for each disease category ranged from 0.94 to 0.97. We found a strong positive correlation between participants' age and citations (r = 0.85), but a negative correlation between the participants' education status and citations (r = - 0.11). Our results revealed that the younger generation possesses the least ethnomedicinal information. We recommend that the species with high use values and fidelity level merit priority in phytochemical and pharmaceutical studies. Our findings highlight that protected areas, besides their prime importance in biodiversity conservation, play a pivotal role in providing the healthcare provisioning ecosystem services to the ethnic communities.

In the Himalaya, collection of fodder from protected areas by local communities for sustenance of their livestock is an integral part of mountain farming systems. Here, we assess the diversity and utilization patterns of fodder resources collected by local communities from Overa-Aru wildlife sanctuary in Kashmir Himalaya. A multi-stage random sampling was employed to select sample villages (5) and households (81) for the survey. Data were collected through personal interviews of household heads administering an interview schedule and then analyzed using descriptive statistics and linear regression model. The results revealed that 74 fodder species were collected by the local communities from the sanctuary. The diversity and magnitude of fodder resource utilization varied across the study area. The fodder collection and utilization on per day and annual basis ranged from 25 to 90 and 2760 to 13,770 kg/household, respectively. Across the surveyed villages, the fodder was mainly collected by the females (60%). A positive but non-significant relationship was found between the number of persons collecting fodder and quantity of fodder collection/household/day, while a positive and significant relationship was found between the herd size and quantity of fodder collection/household/day. We also found a negative but non-significant relationship between the education status of respondents and the number of livestock holding. Based on our findings, we suggest policy and management interventions such as regulating livestock grazing, promoting plantation of frequently collected fodder species, and better use of agriculture byproducts to guide the sustainable management of fodder resources in this Himalayan protected area, with learning for elsewhere.

Studying functional trait diversity can provide crucial clues about the adaptive survival strategies of regional plant species pool. Despite large-scale trait datasets available worldwide, the plant trait data from many biodiversity hotpot regions, like the Himalaya is still scarce. In this study, we aimed to investigate the plant functional traits and aboveground biomass of understory herbaceous vegetation in temperate forests of Overa-Aru wildlife sanctuary of Kashmir Himalaya. We also investigate how these functional traits correlate and what is the magnitude of trait-biomass relationship across the herbaceous species pool. For this, we conducted field sampling and measured leaf functional traits and aboveground biomass of 38 plant species in the study region during peak growing season (July-August) in the year 2021. The results revealed a significant interspecific trait variability among the species studied. We observed a high variability in leaf size and type spectra of the species, with nanophyll and simple leaf lamina, respectively, the most common types among the species studied. The correlation analysis revealed that plant height was positively correlated with aboveground biomass. The variation partitioning analysis revealed that the plant height explained the maximum fraction of variation in aboveground biomass, while least by specific leaf area. Overall, the findings from the present study provide useful insights in understanding trait-trait relationship and trait-environment interaction at the regional scale and can also help in recognizing adaptive functional traits of plant species that determine plant survival under the changing climate in this Himalayan region.

Development of eco-friendly synthetic methods has resulted in the production of biocompatible Ag NPs for applications in medical sector. To overcome the prevailing antibiotic resistance in bacteria, Ag NPs are being extensively researched over the past few years due to their broad spectrum and robust antimicrobial properties. Silver nanoparticles are also being studied widely in advanced anticancer therapy as an alternative anticancer agent to combat cancer in an effective manner. Keeping this backdrop in consideration, this review aims to provide an extensive coverage of the recent progresses in the green synthesis of Ag NPs specifically using plant derived reducing agents such phytochemicals and numerous other biopolymers. Current development in antimicrobial activity of Ag NPs against various pathogens has been deliberated at length. Recent advances in potent anticancer activity of the biogenic Ag NPs against various cancerous cell lines has also been discussed in detail. Mechanistic details of the synthesis of Ag NPs, their anticancer and antimicrobial action has also been highlighted.

Two-dimensional (2D) magnetic layered materials have attracted considerable attention in memory storage devices due to their exciting magnetic ordering. Herein, the electronic and magnetic properties of high-quality single crystals zirconium diselenide and copper (Cu)-doped zirconium diselenide as grown via chemical vapor transport technique combined with first principle density functional theory calculations were investigated. A semimetallic state is recognized for Cu0.052Zr0.93Se2 as measured through resistance versus temperature measurements and angle resolved photoemission spectroscopy (ARPES). The magnetic measurement shows diamagnetic semiconducting behaviour for ZrSe2, whereas Cu0.052Zr0.93Se2 exhibits ferromagnetic character via applying perpendicular magnetic field. Cu0.052Zr0.93Se2 reveals the room temperature magnetic moment ∼0.0125 emu g-1, while the Curie temperature is ∼363.49 K. Furthermore, first principle density functional theory (DFT) calculations show energetically long range ferromagnetic ordering in a half-metallic Cu-doped ZrSe2, while a diamagnetic state in case of ZrSe2 agrees well with experiment results. These results suggest that due to strong interaction elements at the octahedral site of zirconium atoms when replaced by copper atoms, which can change the spin ordering of electrons and make zirconium vacancy, while their magnetic moment is increased. Very importantly the half-metallic character of Cu0.052Zr0.93Se2 promotes much spin polarized electrons around the Fermi level, suggesting significant potential in future memory devices and spintronic applications.

"Stay-green" crop phenotypes have been shown to impact drought tolerance and nutritional content of several crops. We aimed to genetically describe and functionally dissect the particular stay-green phenomenon found in chickpeas with a green cotyledon color of mature dry seed and investigate its potential use for improvement of chickpea environmental adaptations and nutritional value. We examined 40 stay-green accessions and a set of 29 BC2F4-5 stay-green introgression lines using a stay-green donor parent ICC 16340 and two Indian elite cultivars (KAK2, JGK1) as recurrent parents. Genetic studies of segregating populations indicated that the green cotyledon trait is controlled by a single recessive gene that is invariantly associated with the delayed degreening (extended chlorophyll retention). We found that the chickpea ortholog of Mendel's I locus of garden pea, encoding a SGR protein as very likely to underlie the persistently green cotyledon color phenotype of chickpea. Further sequence characterization of this chickpea ortholog CaStGR1 (CaStGR1, for carietinum stay-green gene 1) revealed the presence of five different molecular variants (alleles), each of which is likely a loss-of-function of the chickpea protein (CaStGR1) involved in chlorophyll catabolism. We tested the wild type and green cotyledon lines for components of adaptations to dry environments and traits linked to agronomic performance in different experimental systems and different levels of water availability. We found that the plant processes linked to disrupted CaStGR1 gene did not functionality affect transpiration efficiency or water usage. Photosynthetic pigments in grains, including provitaminogenic carotenoids important for human nutrition, were 2-3-fold higher in the stay-green type. Agronomic performance did not appear to be correlated with the presence/absence of the stay-green allele. We conclude that allelic variation in chickpea CaStGR1 does not compromise traits linked to environmental adaptation and agronomic performance, and is a promising genetic technology for biofortification of provitaminogenic carotenoids in chickpea.

M. charantia extract partitioned in different solvents was assessed for antioxidant (2, 2-diphenyl l-picrylhydrazyl), total phenolic contents (TPC), total flavonoid contents (TFC), antiglycation, alpha amylase and acetylcholinesterase inhibitory activities along with cytotoxic, thrombolytic and antibiofilm potentials. Most effective antioxidant fraction was n-hexane with TPC and TFC, highest in n-butanol and ethanol fractions, respectively. Ethyl acetate fraction showed maximum glycation and alpha amylase inhibitions and optimum acetylcholinesterase inhibition was by ethanol fraction. Fractions exhibited significant hemolytic and thrombolytic efficacies and bacterial growth restraint. The present research reveals some medicinal potency of M. charantia.

The global demand for good quality food indicates that consumers are more concerned about a particular diet associated with good health and lower risk for certain ailments. Mushrooms are widely used as healthy nutritious food. In the present study, the nutritional composition of four different Pleurotus sp. was determined. Prior to extraction, all the selected mushrooms were subjected for proximate composition analysis. The protein, fat, ash, total carbohydrate, fiber and energy contents were in the range of (16.07- 25.15%), (0.64-2.02%), (2.1-9.14%), (65.66-82.47%), (6.21-54.12%) and (342.20-394.30Kcal/100g), respectively. The spectrophotometric analysis showed that the concentration of protein was in the range of 45.78-33.47 mg/g in all Pleurotus sp. High performance liquid chromatographic analysis of sugars showed six different mono and disaccharides in all the selected mushrooms. The fatty acid profile by gas chromatography-mass spectrometry revealed that the main fatty acids in selected mushrooms were present in the order linoliec acid> oleic acid> palmitic acid. The results suggested that all the Pleurotus sp. could be considered as a rich source of nutrients.

TThe efficacy of the two commonly used commercial vaccines for Newcastle disease (ND) and low path avian influenza (LPAI) H9N2 were evaluated against field virus in broiler chicks. One hundred one-dayold commercial broiler chicks were divided into four groups (A to D) with an equal number of birds per group. Group A and B were vaccinated against H9N2 and NDV, respectively, at day 7 of age while group C served as positive infected control for H9N2 and group D for NDV. Serum samples from birds in all groups were tested for presence of antibodies against H9N2 and NDV at day 21 of age. Subsequently, on day 28 of age, groups A and C were challenged with the field strain of H9N2 virus, and Group B and D with NDV. Birds were monitored for a period of 2 weeks for development of any clinical signs and mortality. The geometric mean titer were high in groups A (4.90) and B (7.3), and low in the unvaccinated groups C (0.7) and D (1.1). The highest and lowest value of H9N2 antibody titer detected through ELISA were 1.498 and 0.502, respectively. The S/P ratios greater than 0.5 were considered positive. The highest and lowest value for NDV antibody titer detected through ELISA were 783 and 882, respectively. Serum samples with titer greater than 396 were considered positive and indicated vaccination or other exposure to NDV. On histological examination severe congestion, necrosis, degeneration, hemorrhages and leukocyte infiltration were observed in intestine, lungs, trachea and bursa of Fabricius of the non-vaccinated group post-infection. Mild tissues changes were observed in the vaccinated group. It can be concluded from the findings that the commonly used commercial vaccines may provide effective protection against the circulating H9N2 and ND virus in broiler birds by producing protective antibody titer.

Mushrooms can be used as nutraceutical or functional foods to maintain and promote good health. In the present study, wild Ganoderma lucidum and four commercial mushrooms, Pleurotus ostreatus, Volvariella volvacea, Hericium erinaceus and Lentinus edodes, collected from Pakistan were screened for phenolics, tocopherols and fatty acid contents. High performance liquid chromatography analysis of phenolic acids showed that chlorogenic acid, ferulic acid, gallic acid, p-Coumaric and caffeic acids were observed in selected mushrooms. H. erinaceus contained high amounts of chlorogenic acid (11.49±0.1 µ/g of dry weight) and ferulic acid (7.84±0.7 µg/g of dry weight). γ-tocopherol and lutein were present in all studied mushrooms. Lutein contents were higher in H. erinaceus (2.42±0.087 µg/g of DW) followed by V. volvacea> P. ostreatus> L. edodes. γ-tocopherol was observed in the range of 74.25±3.01 to 29.65±1.2 µg/g of dry weight. GC/MS analysis of fatty acids showed that linoleic acid (18’:2n6c), oleic acid (18’:1n9c), palmitic acid (C16:0), stearic acid (C18:0), linolenic acid (18’:3n3) and nonadecanoic acid (C19-0), were the main fatty acids found in selected mushrooms. The unsaturated fatty acids were predominated over saturated fatty acids. It is concluded that selected mushrooms are good sources of antioxidant compounds and unsaturated fatty acids.

Bacterial ghosts (BG) are empty cell envelopes derived from Gram-negative bacteria. They contain many innate immunostimulatory agonists, and are potent activators of a broad range of cell types involved in innate and adaptive immunity. Several considerable studies have demonstrated the effectiveness of BG as adjuvants as well as their ability to induce proinflammatory cytokine production by a range of immune and non-immune cell types. These proinflammatory cytokines trigger a generalized recruitment of T and B lymphocytes to lymph nodes that maximize the chances of encounter with their cognate antigen, and subsequent elicitation of potent immune responses. The plasticity of BG has allowed for the generation of envelope-bound foreign antigens in immunologically active forms that have proven to be effective vaccines in animal models. Besides their adjuvant property, BG also effectively deliver DNA-encoded antigens to dendritic cells, thereby leading to high transfection efficiencies, which subsequently result in higher gene expressions and improved immunogenicity of DNA-based vaccines. In this review, we summarize our understanding of BG interactions with the host immune system, their exploitation as an adjuvant and a delivery system, and address important areas of future research interest.

The present study was conducted to evaluate the antioxidant activity of ethanol, n-hexane, dichloromethane, ethyl acetate and water extracts of four different Pleurotus spps. P. ostreatus, P. sajor-caju, P. sapidus and P. columbinus. The extraction was performed by classical organic solvent extraction (COSE). The extracts of Pleurotus spps. contained appreciable levels of total phenolic contents (TPC) (0.95-19.49 GAE, mg/g) and total flavonoid contents (0.85-3.73 CE, mg/g). All Pleurotus spps. also contained considerable DPPH radical scavenging activity, showing IC50 (19.15-54.50%) and reducing power (0.50-2.94 nm), respectively. The ascorbic acid content was in the range of (2.90-5.97 mg/g) for all Pleurotus spps. All studied Pleurotus spps. showed potential antioxidant activity. The results of four different Pleurotus spps. extracts showed that they can be used as a good food ingredient, and as a medicinal mushroom for digestive ailments, as well as in the pharmaceutical industry and in cosmetics.

The interest in analysing antigen-specific cytokine responses has substantially increased in recent years, in part due to their use in assessing vaccine efficacy. In the present study, the kinetics of IL-2, IL-4 and IFN-γ expression was determined in bovine PBMCs by real-time PCR and in whole blood by cytokine-release assay after in vitro stimulation with recall foot-and-mouth disease virus (FMDV) antigen. The results showed that the cytokine mRNA of IL-2 and IFN-γ in PBMCs were induced early (peak induction at 6 h), whereas the IL-4 mRNA showed delayed induction (peaked at 24 h). In contrast, the kinetics of cytokine proteins in whole blood was different and required the accumulation of the proteins before being optimally detected. The peak accumulation of cytokine protein in whole blood was recorded at 72 h for IL-2 and IL-4, and 96 h for IFN-γ. The findings of this study are of importance when selecting an optimal time points for measuring antigen-specific cytokine expression in cattle.

Endophytic fungi are those that live internally in apparently healthy and asymptomatic hosts. Endophytic fungi appear to be ubiquitous; indeed, no study has yet shown the existence of a plant species without endophytes. High species diversity is another characteristic of endophytic mycobiota which is depicted by the fact that it is quite common for endophyte surveys to find assemblages consisting of more than 30 fungal species per host plant species. Medicinal plants had been used to isolate and characterize directly the bioactive metabolites. However, the discovery of fungal endophytes inside these plants with capacity to produce the same compounds shifted the focus of new drug sources from plants to fungi. Bioactive natural products from endophytic fungi, isolated from different plant species, are attracting considerable attention from natural product chemists and biologists alike which is clearly depicted by the steady increase of publications devoted to this topic during the recent years. This review will highlight the chemical potential of endophytic fungi with focus on the detection of pharmaceutically valuable plant constituents as products of fungal biosynthesis. In addition, it will cover newly discovered endophytic fungi and also new bioactive metabolites reported in recent years from fungal endophytes. It summarizes the up-to-date and comprehensive information on bioactive compounds from endophytic fungi by having done a thorough survey of literature.

Biofuel tree species are recognized as a promising alternative source of fuel to conventional forms. Additionally, these tree species are also effective in accumulating toxic heavy metals present in some industrial effluents. In developing countries such as Pakistan, the use of biofuel tree species is gaining popularity not only for harvesting economical and environmentally friendly biofuel, but also to sequester poisonous heavy metals from industrial wastewater. This study was aimed at evaluating the genetic potential of two biofuel species, namely, Jatropha curcas and Pongamia pinnata, to grow when irrigated with industrial effluent from the Pak-Arab Fertilizer Factory Multan, Southern Punjab, Pakistan. The growth performances of one-year-old seedlings of both species were compared in soil with adverse physiochemical properties. It was found that J. curcas was better able to withstand the toxicity of the heavy metals present in the fertilizer factory effluent. J. curcas showed maximum gain in height, diameter, and biomass production in soil irrigated with 75% concentrated industrial effluent. In contrast, P. pinnata showed a significant reduction in growth in soil irrigated with more than 50% concentrated industrial effluent, indicating that this species is less tolerant to higher toxicity levels of industrial effluent. This study identifies J. curcas as a promising biofuel tree species that can be grown using industrial wastewater.

The rainfed rice-growing environment is perhaps one of the most vulnerable to water stress such as drought and floods. It is important to determine the spatial extent of the stress-prone areas to effectively and efficiently promote proper technologies (e.g., stress-tolerant varieties) to tackle the problem of sustainable food production. This study was conducted in Odisha state located in eastern India. Odisha is predominantly a rainfed rice ecosystem (71% rainfed and 29% canal irrigated during kharif-monsoon season), where rice is the major crop and staple food of the people. However, rice productivity in Odisha is one of the lowest in India and a significant decline (9%) in rice cultivated area was observed in 2002 (a drought year). The present study analyzed the temporal rice cropping pattern in various ecosystems and identified the stress-prone areas due to submergence (flooding) and water shortage. The spatial distribution of rice areas was mapped using MODIS (MOD09Q1) 250-m 8-day time-series data (2000-2010) and spectral matching techniques. The mapped rice areas were strongly correlated (R(2) = 90%) with district-level statistics. Also the class accuracy based on field-plot data was 84.8%. The area under the rainfed rice ecosystem continues to dominate, recording the largest share among rice classes across all the years. The use of remote-sensing techniques is rapid, cost-effective, and reliable to monitor changes in rice cultivated area over long periods of time and estimate the reduction in area cultivated due to abiotic stress such as water stress and submergence. Agricultural research institutes and line departments in the government can use these techniques for better planning, regular monitoring of land-use changes, and dissemination of appropriate technologies.

Interannual variation in rainfall throughout Tamil Nadu has been causing frequent and noticeable land use changes despite the rapid development in groundwater irrigation. Identifying periodically water-stressed areas is the first and crucial step to minimizing negative effects on crop production. Such analysis must be conducted at the basin level as it is an independent water accounting unit. This paper investigates the temporal variation in irrigated area between 2000-2001 and 2010-2011 due to rainfall variation at the state and sub-basin level by mapping and classifying Moderate Resolution Imaging Spectroradiometer (MODIS) 8-day composite satellite imagery using spectral matching techniques. A land use/land cover map was drawn with an overall classification accuracy of 87.2%. Area estimates between the MODIS-derived net irrigated area and district-level statistics (2000-2001 to 2007-2008) were in 95% agreement. A significant decrease in irrigated area (30-40%) was observed during the water-stressed years of 2002-2003, 2003-2004, and 2009-2010. Major land use changes occurred three times during 2000 to 2010. This study demonstrates how remote sensing can identify areas that are prone to repeated land use changes and pin-point key target areas for the promotion of drought-tolerant varieties, alternative water management practices, and new cropping patterns to ensure sustainable agriculture for food security and livelihoods.
