RESUMO

With the growing global population, abiotic factors have emerged as a formidable threat to agricultural food production. If left unaddressed, these stress factors might reduce food yields by up to 25% by 2050. Plants utilize natural mechanisms, such as reactive oxygen species scavenging, to mitigate the adverse impacts of abiotic stressors. Diverse plants exhibit unique adaptations to abiotic stresses, which are regulated by phytohormones at various levels. Brassinosteroids (BRs) play a crucial role in controlling essential physiological processes in plants, including seed germination, xylem differentiation, and reproduction. The BR cascade serves as the mechanism through which plants respond to environmental stimuli, including drought and extreme temperatures. Despite two decades of research, the complex signaling of BRs under different stress conditions is still being elucidated. Manipulating BR signaling, biosynthesis, or perception holds promise for enhancing crop resilience. This review explores the role of BRs in signaling cascades and summarizes their substantial contribution to plants' ability to withstand abiotic stresses.

We conducted this study to examine the nature of treatment-seeking behavior among carpet weavers in Kashmir. We used a grounded theory approach to gain an in-depth understanding of the phenomenon. Data were collected through face-to-face interviews with 35 research participants recruited through purposive and theoretical sampling techniques. Based on the primary data, our study reveals that carpet weavers approached different agencies and used multiple methods for seeking treatment for their illness(es). The majority of the participants relied on spiritual healers, followed by traditional methods, Unani treatment, compounders and self-medication, while modern medicine was the last option for seeking treatment. We also found that participants base their choices about when and where to seek treatment on multiple socio-economic and cultural factors like financial constraints, perception towards illness, no provision for leave, costly and lengthy medical treatment, side effects of modern medicine, the nature of the disease, lack of infrastructure and insecurity due to conflict. These findings are relevant to employers and various government as well as non-government organizations. Additional implications of these findings for carpet weavers, public health, for practice and research are also discussed.

RESUMO

Using a grounded theory approach, this qualitative study aimed to highlight the evolving technological dimensions of death and mourning practices and its transfer from a set physical setting to virtual and open space. The study was carried on 30 participants from Kashmir. Data was collected using in-depth face-to-face interviews and field notes were also used to capture non-verbal communication. The results of the study revealed that a considerable shift is being witnessed so far as death and mourning practices in Kashmir are concerned. The physical space is shrinking and giving way for virtual spaces to play their role in practices related to death and mourning.

This study examined the changing character of the last honours of those who died of COVID-19 in Kashmir and the life experiences of the families of the deceased. A semi-structured interview schedule was used to collect information from 21 participants. Using qualitative data analysis approaches, five key themes were identified vis-à-vis the impact of COVID-19 on burial rituals and customs; effects on bereaved families, shades of grief, bereavement care, community response, and coping with loss. Based on examining the pandemic-induced changes related to customs and rituals around death, the study found that the bereaved family members were in danger of marginalization, economic burdens, psychological traumas, and overall reduced quality of life. This study would be a credible addition to the existing literature on death practices as there is a shortage of research on funeral rituals during the post-pandemic period in Kashmir.

Brassinosteroids and hydrogen peroxide (H2O2) are extensively used to combat several environmental factors, including heavy metal stress in plants, but their cumulative impact on the maintenance of copper (Cu) homeostasis in plants could not be dissected at elevated level. This study was executed to explore the roles of 24-epibrassinolide (EBL; foliar) and H2O2 (root dipping) in resilience of tomato (Solanum lycopersicum L.) plants to Cu stress. The cumulative effect of EBL and H2O2 in tomato plants grown under Cu stress (10 or 100 mg kg-1 soil) were assessed. Roots of 20 d old plants were submerged in 0.1 mM of H2O2 solution for 4 h and subsequently transplanted in the soil-filled earthen pots and at 30 day after transplantation (DAT), the plants were sprinkled with deionized water (control), and/or 10-8 M EBL and plant performances were evaluated at 40 DAT. High Cu (100 mg kg-1 soil) concentration considerably reduced photosynthetic efficacy, cell viability, and plant growth, and deformed chloroplast ultrastructure and root morphology with altered stomatal behavior, but boosted the activity of antioxidant enzymes, proline content and electrolyte leakage in the leaves of tomato. Moreover, EBL and H2O2 implemented through distinct modes improved photosynthetic efficiency, modified chloroplast ultrastructure, stomatal behavior, root structure, cell viability and production of antioxidants and proline (osmolyte) that augmented resilience of tomato plants to Cu stress. This study revealed the potential of EBL and H2O2 applied through distinct mode could serve as an effective strategy to reduce Cu-toxicity in tomato crop.

Using a phenomenological approach, this study aimed to gain an in-depth understanding of the lived experiences of Kashmiri women suffering from breast cancer within the context of their day-to-day lives. Snowball sampling and theoretical sampling techniques were employed for recruiting participants. Data collection was primarily based on the principles of data saturation. Data saturation occurred after 12 women with breast cancer were interviewed. Data was collected using face-to-face in-depth individual interviews. The interview guide consisted of open-ended questions. To attain the utmost variation, sumptuousness and profundity of responses, interviewer asked probing, specific, and interpreting questions during interviews. Data analysis of the transcripts was based on Graneheim and Lundman's (Qualitative content analysis in nursing research: concepts, procedures and measures to achieve trustworthiness. Nurse Education Today 2004;24:105-12) procedures for taking out themes in qualitative data. Two main themes emerged that explained the lived experiences of women with breast cancer: (i) challenges encountered (altered body image, embarrassment, perceived worries, financial constraints, dealing with negative attitudes and stigma) and (ii) coping strategies (reliance on religion and spirituality, social support, living as usual, optimistic attitude and will to recover, and venting out). The findings revealed that breast cancer diagnosis influences women significantly as they acknowledged that it wasthe most traumatic experience of their life; however, with time, they started to accept the reality and made use of various methods to cope with their illness.

Studies were undertaken across five field locations in Western Australia to determine the relative changes in disease severity and subsequent field pea yield from up to four foliar pathogens associated with a field pea foliar disease complex (viz. genera Didymella, Phoma, Peronospora, and Septoria) across four different pea varieties sown at three different times and at three different densities. Delaying sowing of field pea significantly (P < 0.05) reduced the severity of Ascochyta blight (all five locations) and Septoria blight (one location), increased the severity of downy mildew (four locations), but had no effect on seed yield. In relation to Ascochyta blight severity at 80 days after sowing, at all locations the early time of sowing had significantly (P < 0.05) more severe Ascochyta blight than the mid and late times of sowing. Increasing actual plant density from 20 to 25 plants m-2 to 58 to 78 plants m-2 significantly (P < 0.05) increased the severity of the Ascochyta blight (four locations) and downy mildew (one location), and it increased seed yield at four locations irrespective of sowing date and three locations irrespective of variety. Compared with varieties Dundale, Wirrega, and Pennant, variety Alma showed significantly (P < 0.05) less severe Ascochyta blight, downy mildew, and Septoria blight (one location each). Grain yield was highest for the early time of sowing at three locations. Varieties Alma, Dundale, and Wirrega significantly (P < 0.05) outyielded Pennant at four locations. The percentage of isolations of individual Ascochyta blight pathogens at 80 days after the first time of sowing varied greatly, with genus Didymella ranging from 25 to 93% and genus Phoma ranging from 6 to 23% across the five field locations. This fluctuating nature of individual pathogen types and proportions within the Ascochyta blight complex, along with variation in the occurrence of pathogens Peronospora and Septoria, highlights the challenges to understand and manage the complexities of co-occurring different foliar pathogens of field pea. While the search for more effective host resistance continues, there is a need for and opportunities from further exploring and exploiting cultural management approaches focusing on crop sequence diversification, intercropping, manipulating time of sowing and stand density, and application of improved seed sanitation and residue/inoculum management practices. We discuss the constraints and opportunities toward overcoming the challenges associated with managing foliar disease complexes in field pea.

RESUMO

Melatonin (MEL) is the potential biostimulator molecule, governing multiple range of growth and developmental processes in plants, particularly under different environmental constrains. Mainly, its role is considered as an antioxidant molecule that copes with oxidative stress through scavenging of reactive oxygen species and modulation of stress related genes. It also enhances the antioxidant enzyme activities and thus helps in regulating the redox hemostasis in plants. Apart from its broad range of antioxidant functions, it is involved in the regulation of various physiological processes such as germination, lateral root growth and senescence in plants. Moreover this multifunctional molecule takes much interest due to its recent identification and characterization of receptorCandidate G-protein-Coupled Receptor 2/Phytomelatonin receptor(CAND2/PMTR1) in Arabidopsis thaliana. In this compiled work, different aspects of melatonin in plants such as melatonin biosynthesis and detection in plants, signaling pathway, modulation of stress related genes and physiological role of melatonin under different environmental stresses have been dissected in detail.

While transformation of the major monocot crops is currently possible, the process typically remains confined to one or two genotypes per species, often with poor agronomics, and efficiencies that place these methods beyond the reach of most academic laboratories. Here, we report a transformation approach involving overexpression of the maize (Zea mays) Baby boom (Bbm) and maize Wuschel2 (Wus2) genes, which produced high transformation frequencies in numerous previously nontransformable maize inbred lines. For example, the Pioneer inbred PHH5G is recalcitrant to biolistic and Agrobacterium tumefaciens transformation. However, when Bbm and Wus2 were expressed, transgenic calli were recovered from over 40% of the starting explants, with most producing healthy, fertile plants. Another limitation for many monocots is the intensive labor and greenhouse space required to supply immature embryos for transformation. This problem could be alleviated using alternative target tissues that could be supplied consistently with automated preparation. As a major step toward this objective, we transformed Bbm and Wus2 directly into either embryo slices from mature seed or leaf segments from seedlings in a variety of Pioneer inbred lines, routinely recovering healthy, fertile T0 plants. Finally, we demonstrated that the maize Bbm and Wus2 genes stimulate transformation in sorghum (Sorghum bicolor) immature embryos, sugarcane (Saccharum officinarum) callus, and indica rice (Oryza sativa ssp indica) callus.

Flower and pod production and seed set of chickpea (Cicer arietinum L.) are sensitive to drought stress. A 2-fold range in seed yield was found among a large number of chickpea genotypes grown at three dryland field sites in south-western Australia. Leaf water potential, photosynthetic characteristics, and reproductive development of two chickpea genotypes with contrasting yields in the field were compared when subjected to terminal drought in 106kg containers of soil in a glasshouse. The terminal drought imposed from early podding reduced biomass, reproductive growth, harvest index, and seed yield of both genotypes. Terminal drought at least doubled the percentage of flower abortion, pod abscission, and number of empty pods. Pollen viability and germination decreased when the fraction of transpirable soil water (FTSW) decreased below 0.18 (82% of the plant-available soil water had been transpired); however, at least one pollen tube in each flower reached the ovary. The young pods which developed from flowers produced when the FTSW was 0.50 had viable embryos, but contained higher abscisic acid (ABA) concentrations than those of the well-watered plants; all pods ultimately aborted in the drought treatment. Cessation of seed set at the same soil water content at which stomata began to close and ABA increased strongly suggested a role for ABA signalling in the failure to set seed either directly through abscission of developing pods or seeds or indirectly through the reduction of photosynthesis and assimilate supply to the seeds.

24-Epibrassinolide (EBL) and Selenium (Se) individually confer tolerance to various abiotic stresses, but their interactive effect in the regulation of copper (Cu) homeostasis in plants exposed to toxic levels of Cu is poorly investigated. This study provides an insight into the effects of EBL (foliar) and/or Se (through sand) on Brassica juncea plants exposed to toxic levels of Cu. The combined effect of EBL and Se on compartmentalization of Cu, oxidative stress markers, photosynthetic machinery and biochemical traits in B. juncea were analyzed. Application of EBL and Se through different mode modulated the compartmentalization of Cu in different parts of plants, enhanced the photosynthetic traits, and activities of various antioxidant enzymes and proline accumulation in B. juncea under excess copper levels. These enhanced levels of antioxidant enzymes, proline (osmolyte) accumulation triggered by combination of EBL and Se could have conferred tolerance to the B. juncea plants under toxic level of copper and also maintained Cu homeostasis in various parts of plants. This study indicates that combination of EBL and Se through different mode is an operative approach for Cu detoxification in plants and could be exploited for removal of excess copper from polluted soil.

Black spot, also known as Ascochyta blight, is the most important disease on field pea (Pisum sativum). It is caused by a complex of pathogens, the most important of which in Australia include Didymella pinodes, Phoma pinodella, and P. koolunga. The relative proportions of these and other component pathogens of the complex fluctuate widely across time and geographic locations in Australia, limiting the ability of breeders to develop varieties with effective resistance to black spot. To address this, 40 field pea genotypes were tested under controlled environment conditions for their individual stem and leaf responses against these three pathogens. Disease severity was calculated as area under disease progress curve (AUDPC), and subsequently converted to mean rank (MR). The overall rank (OR) for each pathogen was used to compare response of genotypes under inoculation with each pathogen. The expressions of host resistance across the field pea genotypes were largely dependent upon the individual test pathogen and whether the test was on stem or leaf. Overall, P. koolunga caused most severe stem disease; significantly more severe than either D. pinodes or P. pinodella. This is the first report of the host resistance identified in field pea to P. koolunga; the five genotypes showing highest resistance on stem, viz. 05P778-BSR-701, ATC 5338, ATC 5345, Dundale, and ATC 866, had AUDPC MR values <250.4, while the AUDPC MR values of the 19 genotypes showing the best resistance on leaf was less than 296.8. Two genotypes, ATC 866 and Dundale, showed resistance against P. koolunga on both stem and leaf. Against D. pinodes, the four and 16 most resistant genotypes on stem and leaf had AUDPC MR values <111.2 and <136.6, respectively, with four genotypes showing resistance on both stem and leaf including 05P770-BSR-705, Austrian Winter Pea, 06P822-(F5)-BSR-6, and 98107-62E. Against P. pinodella, four and eight genotypes showing the best resistance on stem and leaf had AUDPC MR values <81.3 and <221.9, respectively; three genotypes, viz. 98107-62E, Dundale, and Austrian Winter Pea showed combined resistance on stem and leaf. A few genotypes identified with resistance against two major pathogens of the complex will be of particular significance to breeding programs. These findings explain why field pea varieties arising from breeding programs in Australia fail to display the level or consistency of resistance required against black spot and why there needs to be a wider focus than D. pinodes in breeding programs.

Black spot (also referred to as Ascochyta blight, Ascochyta foot rot and black stem, and Ascochyta leaf and pod spot) is a devastating disease of pea (Pisum sativum) caused by one or more pathogenic fungi, including Didymella pinodes, Ascochyta pisi, and Phoma pinodella. Surveys were conducted across pea-growing regions of Western Australia in 1984, 1987, 1989, 1996, 2010, and 2012. In total, 1,872 fungal isolates were collected in association with pea black spot disease symptoms. Internal transcribed spacer regions from representative isolates, chosen based on morphology, were sequenced to aid in identification. In most years and locations, D. pinodes was the predominant pathogen in the black spot complex. From 1984 to 2012, four new pathogens associated with black spot symptoms on leaves or stems (P. koolunga, P. herbarum, Boeremia exigua var. exigua, and P. glomerata) were confirmed. This study is the first to confirm P. koolunga in association with pea black spot symptoms in field pea in Western Australia and show that, by 2012, it was widely present in new regions. In 2012, P. koolunga was more prevalent than D. pinodes in Northam and P. pinodella in Esperance. P. herbarum and B. exigua var. exigua were only recorded in 2010. Although A. pisi was reported in Western Australia in 1912 and again in 1968 and is commonly associated with pea black spot in other states of Australia and elsewhere, it was not recorded in Western Australia from 1984 to 2012. It is clear that the pathogen population associated with the pea black spot complex in Western Australia has been dynamic across time and geographic location. This poses a particular challenge to development of effective resistance against the black spot complex, because breeding programs are focused almost exclusively on resistance to D. pinodes, largely ignoring other major pathogens in the disease complex. Furthermore, development and deployment of effective host resistance or fungicides against just one or two of the pathogens in the disease complex could radically shift the make-up of the population toward pathogen species that are least challenged by the host resistance or fungicides, creating an evolving black spot complex that remains ahead of breeding and other management efforts.

The subgingival microbiome has been implicated in oral and systemic diseases such as periodontitis and Alzheimer's disease. However, subgingival sampling is challenging. We developed a novel method of sampling the subgingival microbiome by rotationally swabbing the supragingival area, named subgingival-P (for proxy) samples. We sampled and metatranscriptomically analyzed subgingival and subgingival-P samples of three different teeth in 20 individuals. The subgingival-P samples were comparable to the subgingival samples in the relative abundances of microorganisms and microbial gene expression levels. Our data demonstrate that the novel method of collecting and analyzing the subgingival-P samples can act as a proxy for the subgingiva, paving the way for large and diverse studies investigating the role of the subgingival microbiome in health and disease.

INTRODUCTION: The incidence of pneumonectomy for lung cancer in the UK is continuing to decline in the era of minimally invasive thoracic surgery totalling approximately 3.5% of lung cancer resections annually. Literature is lacking for long-term survival of pneumonectomies. This study updates our previous results. Between 1998 and 2008, 206 patients underwent pneumonectomy compared to 98 patients between 2009 and 2018. METHODS: From January 1998 until December 2018, 304 patients underwent pneumonectomy. This was a retrospective study; data was analysed for age, gender, laterality, histology and time period. RESULTS: Operative mortality was 4.3% overall which was lower than the national average of 5.8%. In the last five years, there were no in-hospital, operative or 30-day mortality. During this period, 90-day mortality was 9.2%. Left-sided pneumonectomies had significantly better overall survival (3.00 vs. 2.03 years; p = 0.0015), squamous cell carcinoma (3.23 vs. 1.54 years; p = 0.00012) as well as those aged less than 70 (2.79 vs. 2.13 years; p = 0.011). There was no significant difference in survival between gender (p = 0.48). Intervention from 1998 to 2008 had significantly greater survival compared to the latter 10 years (2.68 vs. 2.46 years; p = 0.031). The Cox model shows that laterality, age, histology and time period remain significant with multivariate testing. No patient survived after 16 years. DISCUSSION: Our updated retrospective study has built on our previous results by reinforcing the success of pneumonectomies. The incidence of pneumonectomies is likely to decrease with the deployment of nation-wide lung cancer screening in the UK due to earlier detection.

A new Ru(II) arene chlorido organometallic complex [(η6-p-cymene)(L)RuCl]PF6 (named as pCYRuL) using 2-bis(quinolin-2-ylmethylene) hydrazine (L) was developed that exhibits potent anticancer activity against castration-resistant prostate cancer (CRPC) (IC50 = 0.71 µM), and it is 45 times more effective than the standard drug cisplatin (IC50 = 31.3 µM) in a castration-resistant human prostatic adenocarcinoma cell line (PC-3) but non-toxic in normal human kidney cells (HK2) as well as normal breast cells (MCF10A) and found that pCYRuL exerted anticancer activity via apoptosis induction and cell cycle arrest in the G2/M phase of PC-3 cells.

With the growing global population and climate change, achieving food security is a pressing challenge. Vertical farming has the potential to support local food production and security. As a Total Controlled Environment Agriculture (TCEA) system, vertical farming employs LED lighting which offers opportunities to modulate light spectrum and intensity, and thus can be used to influence plant growth and phytochemical composition, including antioxidants beneficial for human health. In this study, we investigated the effect of four red-to-blue light ratios of LEDs (R:B 1, 2.5, 5 and 9) on the growth and antioxidant components in red amaranth microgreens and red lettuce. Plant growth, total phenols, betalains, anthocyanins, vitamin C and antioxidant capacity (ferric reducing antioxidant power assay) were evaluated. A higher proportion of red light resulted in biometric responses, i.e., stem elongation in red amaranth and longer leaves in red lettuce, while the increase in the blue light fraction led to the upregulation of antioxidative components, especially total phenols, betalains (in red amaranth) and anthocyanins (in red lettuce). The antioxidant capacity of both crops was strongly positively correlated with the levels of these phytochemicals. Optimizing the red-to-blue ratio in LED lighting could be effective in promoting antioxidant-rich crops with potential health benefits for consumers.

Salinity stress has detrimental effects on various aspects of plant development. However, our understanding of strategies to mitigate these effects in crop plants remains limited. Recent research has shed light on the potential of sodium acetate as a mitigating component against salinity stress in several plant species. Here, we show the role of acetate sodium in counteracting the adverse effects on oat (Avena sativa) plants subjected to NaCl-induced salinity stress, including its impact on plant morphology, photosynthetic parameters, and gene expression related to photosynthesis and antioxidant capacity, ultimately leading to osmoprotection. The five-week experiment involved subjecting oat plants to four different conditions: water, salt (NaCl), sodium acetate, and a combination of salt and sodium acetate. The presence of NaCl significantly inhibited plant growth and root elongation, disrupted chlorophylls and carotenoids content, impaired chlorophyll fluorescence, and down-regulated genes associated with the plant antioxidant defense system. Furthermore, our findings reveal that when stressed plants were treated with sodium acetate, it partially reversed these adverse effects across all analyzed parameters. This reversal was particularly evident in the increased content of proline, thereby ensuring osmoprotection for oat plants, even under stressful conditions. These results provide compelling evidence regarding the positive impact of sodium acetate on various plant development parameters, with a particular focus on the enhancement of photosynthetic activity.

Urban atmospheric pollution is global problem and and have become increasingly critical in big cities around the world. Issue of toxic emissions has gained significant attention in the scientific community as the release of pollutants into the atmosphere rising continuously. Although, the Pakistani government has started the Pakistan Clean Air Program to control ambient air quality however, the desired air quality levels are yet to be reached. Since the process of mapping the dispersion of atmospheric pollutants in urban areas is intricate due to its dependence on multiple factors, such as urban vegetation and weather conditions. Therefore, present research focuses on two essential items: (1) the relationship between urban vegetation and atmospheric variables (temperature, relative humidity (RH), sound intensity (SI), CO, CO2, and particulate matter (PM0.5, PM1.0, and PM2.5) and (2) the effect of seasonal change on concentration and magnitude of atmospheric variables. A geographic Information System (GIS) was utilized to map urban atmospheric variables dispersion in the residential areas of Faisalabad, Pakistan. Pearson correlation and principal component analyses were performed to establish the relationship between urban atmospheric pollutants, urban vegetation, and seasonal variation. The results showed a positive correlation between urban vegetation, metrological factors, and most of the atmospheric pollutants. Furthermore, PM concentration showed a significant correlation with temperature and urban vegetation cover. GIS distribution maps for PM0.5, PM1.0, PM2.5, and CO2 pollutants showed the highest concentration of pollutants in poorly to the moderated vegetated areas. Therefore, it can be concluded that urban vegetation requires a rigorous design, planning, and cost-benefit analysis to maximize its positive environmental effects.

The rate of progression of Alzheimer's disease (AD) differs dramatically between patients. Identifying the most is critical because when their numbers differ between treated and control groups, it distorts the outcome, making it impossible to tell whether the treatment was beneficial. Much recent effort, then, has gone into identifying RPs. We pooled de-identified placebo-arm data of three randomized controlled trials (RCTs), EXPEDITION, EXPEDITION 2, and EXPEDITION 3, provided by Eli Lilly and Company. After processing, the data included 1603 mild-to-moderate AD patients with 80 weeks of longitudinal observations on neurocognitive health, brain volumes, and amyloid-beta (Aß) levels. RPs were defined by changes in four neurocognitive/functional health measures. We built deep learning models using recurrent neural networks with attention mechanisms to predict RPs by week 80 based on varying observation periods from baseline (e.g., 12, 28 weeks). Feature importance scores for RP prediction were computed and temporal feature trajectories were compared between RPs and non-RPs. Our evaluation and analysis focused on models trained with 28 weeks of observation. The models achieved robust internal validation area under the receiver operating characteristic (AUROCs) ranging from 0.80 (95% CI 0.79-0.82) to 0.82 (0.81-0.83), and the area under the precision-recall curve (AUPRCs) from 0.34 (0.32-0.36) to 0.46 (0.44-0.49). External validation AUROCs ranged from 0.75 (0.70-0.81) to 0.83 (0.82-0.84) and AUPRCs from 0.27 (0.25-0.29) to 0.45 (0.43-0.48). Aß plasma levels, regional brain volumetry, and neurocognitive health emerged as important factors for the model prediction. In addition, the trajectories were stratified between predicted RPs and non-RPs based on factors such as ventricular volumes and neurocognitive domains. Our findings will greatly aid clinical trialists in designing tests for new medications, representing a key step toward identifying effective new AD therapies.
