RESUMO

Bioreduction of Cr(VI) is cost-effective and environmentally friendly, however, the slow bioreduction rate limits its application. In this study, the potential synergistic enhancement of Cr(VI) bioreduction by shewanella oneidensis MR-1 (S. oneidensis) with goethite and riboflavin (RF) was investigated. The results showed that the S. oneidensis reaction system reduce 29.2% of 20 mg/L Cr(VI) after 42 h reaction, while the S. oneidensis/goethite/RF reaction system increased the Cr(VI) reduction rate to 87.74%. RF as an efficient electron shuttle and Fe(II) from goethite bioreduction were identified as the crucial components in Cr(VI) reduction. XPS analysis showed that the final precipitates of Cr(VI) reduction were Cr(CH3C(O)CHC(O)CH3)3 and Cr2O3 and adhered to the bacterial cell surface. In this process, the microbial surface functional groups such as hydroxyl and carboxyl groups participated in the adsorption and reduction of Cr(VI). Meanwhile, an increase in cytochrome c led to an increase in electron transfer system activity (ETSA), causing a significant enhancement in extracellular electron transfer efficiency. This study provides insight into the mechanism of Cr(VI) reduction in a complex environment where microorganisms, iron minerals and RF coexist, and the synergistic treatment method of Fe(III) minerals and RF has great potential application for Cr(VI) detoxification in aqueous environment.

Mounting evidence suggests that people may use multiple cues to predict different levels of representation (e.g., semantic, syntactic, and phonological) during language comprehension. One question that has been less investigated is the relationship between general cognitive processing and the efficiency of prediction at various linguistic levels, such as semantic and phonological levels. To address this research gap, the present study investigated how working memory capacity (WMC) modulates different kinds of prediction behavior (i.e., semantic prediction and phonological prediction) in the visual world. Chinese speakers listened to the highly predictable sentences that contained a highly predictable target word, and viewed a visual display of objects. The visual display of objects contained a target object corresponding to the predictable word, a semantic or a phonological competitor that was semantically or phonologically related to the predictable word, and an unrelated object. We conducted a Chinese version of the reading span task to measure verbal WMC and grouped participants into high- and low-span groups. Participants showed semantic and phonological prediction with comparable size in both groups during language comprehension, with earlier semantic prediction in the high-span group, and a similar time course of phonological prediction in both groups. These results suggest that verbal working memory modulates predictive processing in language comprehension.

The utilization of anaerobic hydrolysate from agroforestry wastes is limited by dissolved lignin and aromatics, which have received insufficient attention despite their potential as excellent carbon sources for denitrification. This study aims to investigate the influence of hematite on lignin-derived aromatic compounds and denitrifying carbon sources, as well as to identify iron-reducing bacteria that utilize lignin-derived aromatic compounds as electron donors. The findings revealed that hematite facilitated the anaerobic fermentation of plant biomass, resulting in the production of small molecular organic acids. Moreover, biodegradation of lignin-derived aromatic compounds led to the formation of phenolic acids, while an increased generation of denitrifying carbon sources enhanced nitrogen removal efficiency by 13.84 %. Additionally, due to adsorption by hematite and subsequent microbial degradation, there was a significant improvement (40.32%) in color removal rate within denitrification effluent. Notably, Azonexus strains were hypothesized to be involved in Fe(Ⅲ) reduction coupled with aromatic compounds oxidation.

Until now, no effective method has been found to monitor the Maillard reaction process for complex protein hydrolysates. Dynamic changes in the concentration of α-dicarbonyl compounds, fluorescence intensity, and browning degree were investigated during the Maillard reaction of corn protein hydrolysates. When the fluorescence intensity reached the peak, deoxyosones would continue to be increased by ARP's degradation. However, the reaction node with the highest fluorescence intensity coincided with the turning point of the browning reaction, and the subsequent browning rate remarkably increased. Therefore, the change in fluorescence intensity could be used to monitor the degradation of ARP and the formation of browning melanoidin at different stages of the Maillard reaction of complex systems, thus effectively indicating the process of the Maillard reaction. When Maillard reaction intermediates (MRIs) with maximum fluorescent compounds were heated, the most abundant pyrazines were subsequently achieved. However, furan compounds would be progressively increased during the thermal process of MRIs with continuously enhanced browning.

A cascaded side-polish plastic optical fiber (POF) and FONTEX optical fiber based surface plasmon resonance (SPR) sensor is proposed for simultaneous measurement of refractive index (RI) and temperature. The side-polish POF and FONTEX optical fiber are connected by using the UV glue in a Teflon plastic tube. The SPR phenomenon can be excited at both of the side-polish region and the FONTEX fiber cladding. The polydimethylsiloxane (PDMS) is coated on the side-polish POF to get a temperature sensing channel. Due to the low RI sensitivity of the FONTEX optical fiber, the cascaded fiber sensor can obtain a broader RI measurement range with a low crosstalk. An RI sensitivity of 700 nm/RIU in the RI measurement range of 1.335-1.39 and a temperature sensitivity of -1.02 nm/°C measured in deionized water with a range of 20-60 °C are obtained. In addition, the cascaded POF based SPR sensor has potential application prospects in the field of biochemical sensing.

Basal progenitor cells are crucial for maintaining foregut (the esophagus and forestomach) homeostasis. When their function is dysregulated, it can promote inflammation and tumorigenesis. However, the mechanisms underlying these processes remain largely unclear. Here, we employ genetic mouse models to reveal that Jag1/2 regulate esophageal homeostasis and foregut tumorigenesis by modulating the function of basal progenitor cells. Deletion of Jag1/2 in mice disrupts esophageal and forestomach epithelial homeostasis. Mechanistically, Jag1/2 deficiency impairs activation of Notch signaling, leading to reduced squamous epithelial differentiation and expansion of basal progenitor cells. Moreover, Jag1/2 deficiency exacerbates the deoxycholic acid (DCA)-induced squamous epithelial injury and accelerates the initiation of squamous cell carcinoma (SCC) in the forestomach. Importantly, expression levels of JAG1/2 are lower in the early stages of human esophageal squamous cell carcinoma (ESCC) carcinogenesis. Collectively, our study demonstrates that Jag1/2 are important for maintaining esophageal and forestomach homeostasis and the onset of foregut SCC.

The rational combination of oral antidiabetic agents is more likely to provide better glycemic control than monotherapy. Metformin glibenclamide tablets can be used as second-line therapy for patients with type 2 diabetes mellitus who cannot successfully control their blood glucose levels by diet and exercise plus metformin or sulfonylureas. The aim of this study was to evaluate the bioequivalence and safety of metformin hydrochloride and glibenclamide tablets (500 mg/5 mg) prepared by 2 different vendors in healthy Chinese subjects under fasting and fed conditions. This is an open-label, single-center, randomized, 2-formulation, 2-period crossover study. After screening, 40 subjects were enrolled in the fasting trial, while 40 subjects were enrolled in the fed trial. Qualified subjects were randomly assigned to receive a monotherapy dose of 500 mg/5 mg of the test or reference formulation, and after a 1-week washout period, they took the alternative formulation. Blood samples were collected from 24 blood collection sites per cycle for pharmacokinetic analysis until 36 hours after oral administration. In total, 78 subjects completed the study. Under fasting and fed conditions, the geometric mean ratios of maximum plasma concentration, area under the plasma concentration-time curve (AUC) from time 0 to time of last quantifiable drug level , and AUC from time 0 to infinity between the 2 products, as well as the corresponding 90%CIs, were all within the range of 80%-125%. It was found that exposure (AUC from time 0 to infinity) to metformin is decreased by about 25% in the fed state compared to fasting, whereas glibenclamide exposure is increased by about 30% in the fed state. No severe adverse events were observed in the study.

Online monitoring of antibiotics in the environment attracts more and more attention. Surface-enhanced Raman scattering (SERS) is a promising technique for the detection of trace amounts of antibiotics in the environment, which is fast, non-invasive and sensitive. To investigate the enrichment of trace amounts of antibiotics in water, polyethylene microplastics (PE MPs) were prepared as sorbents to simply concentrate enrofloxacin, ciprofloxacin hydrochloride monohydrate and triclosan in water, followed by the SERS measurement of antibiotics extract washed from MPs on an AgNPs@Si SERS substrate. Limit of detection of Rhodamine 6G is 2.1 × 10-12 M achieved from the AgNPs@Si SERS, indicating a high enhancement. The detection results show that SERS peaks of the antibiotics could be observed from the spectra of the extracts eluted from MPs, indicating MPs could adsorb and desorb antibiotics from water. Besides, for enrofloxacin and triclosan, the intensity of SERS measured from the MPs extracts are higher than that of directly from the spiked water, demonstrating the proposed method could lower the detectable concentration of hydrophobic antibiotics in water. Moreover, the proposed MPs sorbents combined with SERS method was applied to detect the antibiotics in real river water, with minimal detection of 10-10 M, 10-8 M, and 10-8 M achieved for enrofloxacin, ciprofloxacin hydrochloride monohydrate and triclosan, respectively. The proposed method provides a promising simple, rapid and low reagent consuming means for monitoring antibiotics in water.

As a typical disinfectant, the use of benzyl dodecyl dimethyl ammonium bromide (BDAB) has dramatically increased since the emergence of SARS-CoV-2, posing a threat to environmental balance and human health. Screening BDAB co-metabolic degrading bacteria is required for efficient microbial degradation. Conventional methods for screening co-metabolic degrading bacteria are laborious and time-consuming, especially when the number of strains is large. This study aimed to develop a novel method for the rapid screening of BDAB co-metabolic degrading bacteria from the cultured solid medium using near-infrared hyperspectral imaging (NIR-HSI) technology. Based on NIR spectra, the concentration of BDAB in the solid medium can be well predicted by partial least squares regression (PLSR) models, non-destructively and rapidly, with Rc2 > 0.872 and Rcv2 > 0.870. The results show that the predicted BDAB concentrations decrease after degrading bacteria utilization, comparing with the regions where no degrading bacteria grew. The proposed method was applied to directly identify the BDAB co-metabolic degrading bacteria cultured on the solid medium, and two kinds of co-metabolic degrading bacteria RQR-1 and BDAB-1 were correctly identified. This method provides a high-efficiency method for screening BDAB co-metabolic degrading bacteria from a large number of bacteria.

Unlike animals, plants and yeasts only have a class III phosphatidylinositol 3-kinase (PI3KC3). Its lipid product, phosphatidylinositol 3-phosphate (PtdIns-3-P, PI3P), organizes intracellular trafficking routes such as autophagosome formation, multivesicular body (MVB) formation, retro-transport from trans-Golgi network (TGN) to late Golgi, and the fusion events between autophagosomes and MVBs and the vacuole. The catalytic subunit of plant PI3KC3 is encoded by the essential gene Vacuolar Protein Sorting 34 (VPS34). Despite the importance of VPS34 in cellular homeostasis and plant development, a VPS34 interactome is lacking. Here we employed TurboID, an enzyme-catalyzed proximity labelling (PL) method, to describe a proximal interactome of Arabidopsis VPS34. TurboID catalyzed spatially restricted biotinylation and enabled VPS34-specific enrichment of 273 proteins from affinity purification coupled with mass spectrometry. The interactome confirmed known functions of VPS34 in endo-lysosomal trafficking. Intriguingly, carbohydrate metabolism was the most enriched Gene Ontology (GO) term, including glycolytic enzymes in the triose portion and enzymes functioning in chloroplast triose export and sucrose biosynthesis. The interaction between VPS34 and the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH, GAPC1/2) was validated in planta. Also verified was the interaction between VPS34 and the plasma membrane H+-ATPase AHA2, a primary determinant of membrane potential. Our study links PI3KC3 to carbohydrate metabolism and membrane potential, two key processes that maintain cellular homeostasis.

Whereas a growing body of research demonstrates that people may predict conceptual representations during language comprehension, evidence for phonological prediction is less straightforward. Moreover, existing findings of phonological prediction come largely from studies conducted with languages with alphabetic scripts, making it difficult to dissociate the effects of phonology from orthography. In two experiments, we used the visual world paradigm to investigate whether comprehenders predict phonological information during comprehension of Chinese sentences, where phonology and orthography are largely dissociable. Participants listened to sentences containing a highly predictable word while viewing a visual display consisting of a critical object and three distractors (target object, a semantic competitor object, a phonological competitor object, or an unrelated object). We manipulated preview time (i.e., 2 s in Experiment 1 and 1 s in Experiment 2) to investigate how preview time influences the phonological prediction effect. In addition, we used different stimuli to test the robustness of the results. Results showed anticipatory eye movements for semantic competitors: participants fixated more on the semantic competitors than unrelated objects before the onset of predictable target words. Critically, in the two experiments, the results showed anticipatory fixations on phonological competitor objects. Combined antihypertensive drugs have become the basic method of treating hypertension. Olmesartan and amlodipine, as representative drugs of angiotensin receptor blockers and calcium channel blockers, were developed as a compound formulation for antihypertensive treatment. The purpose of this study was to evaluate the bioequivalence of olmesartan medoxomil/amlodipine besylate tablet (20 mg/5 mg) under fasting and fed conditions in healthy Chinese volunteers. A phase 1 randomized, open-label, 2-period, single-dose crossover study (n = 56) was designed, with subjects under fasting (n = 28) or fed (n = 28) conditions. Of the 56 enrolled participants, 55 healthy volunteers completed the study. Blood samples for pharmacokinetic analysis were collected from 1.5 hours before dosing to 168 hours after dosing. The 90%CIs for the geometric mean ratios of maximum plasma drug concentration, area under the plasma concentration-time curve (AUC) from time 0 to the last measurable concentration and AUC from time 0 to infinity of the test/reference were all within the acceptance range for bioequivalence (80%-125%). The data showed that the absorption of amlodipine is not affected by food, but the exposure of olmesartan (both AUC from time 0 to the last measurable concentration and AUC from time 0 to infinity were P < .05) reduced significantly after consuming a high-fat meal, which indicates that the effects of food on olmesartan exposure in healthy Chinese were clinically relevant. During the study, there were no suspected serious adverse reactions or serious adverse events. All adverse events were determined to be mild after Common Terminology Criteria for Adverse Events 5.0 evaluation. These results indicated that both the test and reference formulations were bioequivalent with similar safety profiles.

As a kind of renewable biological resource, leather collagen is the raw material of leather industry. The chrome tanning modified collagen has high humidity and heat resistance, but there are some defects such as environmental pollution, harm to human health and shortage of chromium resources, so it is urgent to research chromium-free tanning to achieve clean modified colloidal. In the chromium-free tanning system, the modification of skin fibrin by bio-based material has the significance of environmental protection. Sodium alginate can be used as crosslinking agent to stabilize collagen. In this study, the polyhedral oligomeric silsesquioxane (POSS) grafted oxidized sodium alginate polymer composite (POSS-OSA-MAA) containing aldehyde group was prepared by two steps. Firstly, POSS grafted sodium alginate polymer composite (POSS-SAG-MAA) was prepared by graft polymerization with POSS, sodium alginate (SAG) and methacrylic acid (MAA) as raw materials. Then POSS-OSA-MAA was obtained by oxidation of POSS-SAG-MAA with sodium periodate. The aldehyde group highest concentration in POSS-OSA-MAA was 3.26 mmol•g-1, and the tanned leather shrinkage temperature was 73.1 °C. The X-ray photoelectron spectroscopy showed that POSS-OSA-MAA could form Schiff base structure with amino-containing substances and form multiple points crosslinking, and POSS-OSA-MAA could improve the shrinkage temperature and thermal stability of skin collagen.

Surface Enhanced Raman Spectroscopy (SERS) is a highly sensitive detection method, however, it is still challenging to directly detect florfenicol due to its low Raman activity. To solve this problem, we present a novel sensor for selective detection of florfenicol residue in milk using gap-enhance Raman tags (GERTs) competitive immunoassay. In this nanosensor, Raman reporters 4-mercaptobenzoic acid (4MBA) were embedded in the 1 nm gap of Au core-gap-Au shell nanoparticles, which can protect 4MBA molecules from unwanted external environments and provide a high enhancement. The parameters affecting the detection sensitivity were discussed and optimized. The limit detection of florfenicol in milk can reach 0.1 µg/kg. To evaluate the performance of GERTs immunoassay, Ag@4MBA tags immunoassay was conducted and the results were compared. Moreover, Raman mapping was investigated to improve the quantitative prediction model with R2 of 0.981 achieved, by eliminating the signal fluctuations caused by the inhomogeneous distribution of GERTs.

Conventional stabilization/solidification materials have defects in the simultaneous treatment of heavy metals (HMs) and phenanthrene (PHe). In order to solve this problem, a new functional material ß-cyclodextrin modified biochar (ß-CD-BC) was prepared by integrating the properties of biochar (BC) and the hydrophilic and hydrophobic properties of the ß-CD surface and combined with Portland cement (PC) to cure and stabilize HMs and PHe. The effect of key parameters on the treatment effect was discussed by response surface method. The results showed that the minimum leaching concentration if HMs was 16.81 mg·L-1, and the leaching concentration of PHe can be as low as 0.059 µg/kg under the conditions of ß-CD-BC and Portland cement ratio of 9.75% and 11.4%, curing for 22.85 d. The weak acid soluble state reduced from 9~13% to 0.5~6%, the residual state was increased from 37~61% to 77~87%. The unconfined compressive strength of sample is more than 50 kPa. The results of this study can provide a new technical scheme for long-term curing and stabilization of HMs and PHe.

High-performance flexible conductive films are highly promising for the development of wearable devices, artificial intelligence, medical care, etc. Herein, a three-step procedure was developed to produce electromagnetic interference (EMI) shielding, Joule heating, and a hydrophobic nanofiber film based on hydrolysate of waste leather scraps (HWLS): (i) electrospinning preparation of the HWLS/polyacrylonitrile (PAN)/zeolitic imidazolate framework-67 (ZIF-67) nanofiber film, (ii) carbonization of the HWLS/PAN/ZIF-67 nanofiber film, and (iii) coating of the carbon nanofiber@cobalt (Co@CNF) nanofiber film with perfluorooctyltriethoxysilane (POTS). The X-ray diffraction results showed that metal nanoparticles and amorphous carbon had obvious peaks. The micromorphology results showed that metal nanoparticles were coated with carbon nanofibers. The conductivity and shielding efficiency of the carbon nanofiber film with 250 µm thickness could reach 45 S/m and 49 dB, respectively, and absorption values (A > 0.5) were higher than reflection (R) values for the Co@CNF nanofiber film, which indicated that the contribution of absorption loss was more significant than that of reflection loss. Ultrafast electrothermal response performances were also achieved, which could guarantee the normal functioning of films in cold conditions. The water contact angle of the Co@CNF@POTS nanofiber film was ∼151.3°, which displayed a self-cleaning property with water-proofing and antifouling. Absorption-dominant and low-reflection EMI shielding and electrothermal films not only showed broad application potential in flexible wearable electronic devices but also provided new avenues for the utilization of leather solid waste.

Background: Molecular typing based on single omics data has its limitations and requires effective integration of multiple omics data for tumor typing of colorectal cancer (CRC). Methods: Transcriptome expression, DNA methylation, somatic mutation, clinicopathological information, and copy number variation were retrieved from TCGA, UCSC Xena, cBioPortal, FireBrowse, or GEO. After pre-processing and calculating the clustering prediction index (CPI) with gap statistics, integrative clustering analysis was conducted via MOVICS. The tumor microenvironment (TME) was deconvolved using several algorithms such as GSVA, MCPcounter, ESTIMATE, and PCA. The metabolism-relevant pathways were extracted through ssGSEA. Differential analysis was based on limma and enrichment analysis was carried out by Enrichr. DNA methylation and transcriptome expression were integrated via ELMER. Finally, nearest template or hemotherapeutic sensitivity prediction was conducted using NTP or pRRophetic. Results: Three molecular subtypes (CS1, CS2, and CS3) were recognized by integrating transcriptome, DNA methylation, and driver mutations. CRC patients in CS3 had the most favorable prognosis. A total of 90 differentially mutated genes among the three CSs were obtained, and CS3 displayed the highest tumor mutation burden (TMB), while significant instability across the entire chromosome was observed in the CS2 group. A total of 30 upregulated mRNAs served as classifiers were identified and the similar diversity in clinical outcomes of CS3 was validated in four external datasets. The heterogeneity in the TME and metabolism-related pathways were also observed in the three CSs. Furthermore, we found CS2 tended to loss methylations while CS3 tended to gain methylations. Univariate and multivariate Cox regression revealed that the subtypes were independent prognostic factors. For the drug sensitivity analysis, we found patients in CS2 were more sensitive to ABT.263, NSC.87877, BIRB.0796, and PAC.1. By Integrating with the DNA mutation and RNA expression in CS3, we identified that SOX9, a specific marker of CS3, was higher in the tumor than tumor adjacent by IHC in the in-house cohort and public cohort. Conclusion: The molecular subtypes based on integrated multi-omics uncovered new insights into the prognosis, mechanisms, and clinical therapeutic targets for CRC.

Short peptide-based hydrogels have attracted extensive research interests in drug delivery because of their responsive properties. So far, most drug molecules have been conjugated with short peptides via an amide bond, restricting the release of the native drug molecules. In this study, we demonstrated the effectiveness of an auxin-based hydrogelator linked by a hydrolysable ester bond. Hydrogel I, formed by the gelator (NAA-G'FFY) linked with an ester bond, was able to release 1-naphthaleneacetic acid (NAA), whereas hydrogel II, formed by the gelator without an ester bond (NAA-GFFY), was not. By mixing NAA-G'FFY with Fmoc-GFFY to form a two-component hydrogel, the spatial and temporal release of NAA was achieved, promoting on-site auxin responses including primary root elongation and lateral root formation in the model plant Arabidopsis thaliana. The strategy of using a hydrolysable ester bond to connect drug molecules and self-assembling peptides could lead to the development of supramolecular hydrogels with more controllable drug release profiles.

Chemical enhancement is an important mechanism in surface-enhanced Raman spectroscopy. It is found that mildly reduced graphene oxide (MR-GO) nanosheets can significantly increase the chemical enhancement of the main peaks by up to 1 order of magnitude for adsorbed Rhodamine B (RhB) molecules, in comparison with the mechanically exfoliated graphene. The observed enhancement factors can be as large as ∼10(3) and show clear dependence on the reduction time of graphene oxide, indicating that the chemical enhancement can be steadily controlled by specific chemical groups. With the help of X-ray photoelectron spectra, these chemical species are identified and the origin of the observed large chemical enhancement can thus be revealed. It is shown that the highly electronegative oxygen species, which can introduce a strong local electric field on the adsorbed molecules, are responsible for the large enhancement. In contrast, the local defects generated by the chemical reduction show no positive correlation with the enhancement. Most importantly, the dramatically enhanced Raman spectra of RhB molecules on MR-GO nanosheets reproduce all important spectral fingerprints of the molecule with a negligible frequency shift. Such a unique noninvasive feature, along with the other intrinsic advantages, such as low cost, light weight, easy availability, and flexibility, makes the MR-GO nanosheets very attractive to a variety of practical applications.
